Converging interactions between ascending proprioceptive afferents and descending corticospinal tract projections are critical in the modulation and coordination of skilled motor behaviors. Fundamental to these processes are the functional inputs and the mechanisms of integration in the brain and spinal cord between proprioceptive and corticospinal tract information. In this review, we first highlight key connections between corticospinal tract motor circuit and spinal interneurons that receive proprioceptive inputs. We will also address corticospinal tract access to the presynaptic inhibitory system in the spinal cord and its role in modulating proprioceptive stimuli. Lastly, we will focus on the corticospinal neuron influences on the dorsal column nuclei complex, an integration hub for processing ascending somatosensory information.

Objective This is a comprehensive, structured review synthesizing and summarizing the current experimental data and knowledge about the mechanisms of action (MOA) underlying spinal cord stimulation with the burst waveform (as defined by De Ridder) in chronic pain treatment. Methods Multiple database queries and article back-searches were conducted to identify the relevant literature and experimental findings for results integration and interpretation. Data from recent peer-reviewed conference presentations were also included for completeness and to ensure that the most up-to-date scientific information was incorporated. Both human and animal data were targeted in the search to provide a translational approach in understanding the clinical relevance of the basic science findings. Results/Conclusions Burst spinal cord stimulation likely provides pain relief via multiple mechanisms at the level of both the spinal cord and the brain. The specific waveforms and temporal patterns of stimulation both play a role in the responses observed. Differential modulation of neurons in the dorsal horn and dorsal column nuclei are the spinal underpinnings of paresthesia-free analgesia. The burst stimulation pattern also produces different patterns of activation within the brain when compared with tonic stimulation. The latter may have implications for not only the somatic components of chronic pain but also the lateral and affective pathway dimensions as well.

Neural prostheses enable users to effect movement through a variety of actuators by translating brain signals into movement control signals. However, to achieve more natural limb movements from these devices, the restoration of somatosensory feedback is required. We used feature-learnability, a machine-learning approach, to assess signal features for their capacity to enhance decoding performance of neural signals evoked by natural tactile and proprioceptive somatosensory stimuli, recorded from the surface of the dorsal column nuclei (DCN) in urethane-anesthetized rats. The highest performing individual feature, spike amplitude, classified somatosensory DCN signals with 70% accuracy. The highest accuracy achieved was 87% using 13 features that were extracted from both high and low-frequency (LF) bands of DCN signals. In general, high-frequency (HF) features contained the most information about peripheral somatosensory events, but when features were acquired from short time-windows, classification accuracy was significantly improved by adding LF features to the feature set. We found that proprioception-dominated stimuli generalize across animals better than tactile-dominated stimuli, and we demonstrate how information that signal features contribute to neural decoding changes over the time-course of dynamic somatosensory events. These findings may inform the biomimetic design of artificial stimuli that can activate the DCN to substitute somatosensory feedback. Although, we investigated somatosensory structures, the feature set we investigated may also prove useful for decoding other (e.g., motor) neural signals.

The dorsal column nuclei complex (DCN-complex) includes the dorsal column nuclei (DCN, referring to the gracile and cuneate nuclei collectively), external cuneate, X, and Z nuclei, and the median accessory nucleus. The DCN are organised by both somatotopy and modality, and have a diverse range of afferent inputs and projection targets. The functional organisation and connectivity of the DCN implicate them in a variety of sensorimotor functions, beyond their commonly accepted role in processing and transmitting somatosensory information to the thalamus, yet this is largely underappreciated in the literature. To consolidate insights into their sensorimotor functions, this review examines the morphology, organisation, and connectivity of the DCN and their associated nuclei. First, we briefly discuss the receptors, afferent fibres, and pathways involved in conveying tactile and proprioceptive information to the DCN. Next, we review the modality and somatotopic arrangements of the remaining constituents of the DCN-complex. Finally, we examine and discuss the functional implications of the myriad of DCN-complex projection targets throughout the diencephalon, midbrain, and hindbrain, in addition to their modulatory inputs from the cortex. The organisation and connectivity of the DCN-complex suggest that these nuclei should be considered a complex integration and distribution hub for sensorimotor information.

KEY POINTS The brainstem dorsal column nuclei (DCN) process sensory information arising from the body before it reaches the brain and becomes conscious. Despite significant investigations into sensory coding in peripheral nerves and the somatosensory cortex, little is known about how sensory information arising from the periphery is represented in the DCN. Following stimulation of hind-limb nerves, we mapped and characterised the evoked electrical signatures across the DCN surface. We show that evoked responses recorded from the DCN surface are highly reproducible and are unique to nerves carrying specific sensory information. Current neural prostheses can restore limb movement to tetraplegic patients by translating brain signals coding movements to control a variety of actuators. Fast and accurate somatosensory feedback is essential for normal movement, particularly dexterous tasks, but is currently lacking in motor neural prostheses. Attempts to restore somatosensory feedback have largely focused on cortical stimulation which, thus far, have succeeded in eliciting minimal naturalistic sensations. Yet, a question that deserves more attention is whether the cortex is the best place to activate the central nervous system to restore somatosensation. Here, we propose that the brainstem dorsal column nuclei are an ideal alternative target to restore somatosensation. We review some of the recent literature investigating the dorsal column nuclei functional organization and neurophysiology and highlight some of the advantages and limitations of the dorsal column nuclei as a future neural prosthetic target. Recent evidence supports the dorsal column nuclei as a potential neural prosthetic target, but also identifies several gaps in our knowledge as well as potential limitations which need to be addressed before such a goal can become reality.

The brainstem dorsal column nuclei (DCN) are essential to inform the brain of tactile and proprioceptive events experienced by the body. However, little is known about how ascending somatosensory information is represented in the DCN. Our objective was to investigate the usefulness of high-frequency (HF) and low-frequency (LF) DCN signal features (SFs) in predicting the nerve from which signals were evoked. We also aimed to explore the robustness of DCN SFs and map their relative information content across the brainstem surface. DCN surface potentials were recorded from urethane-anesthetized Wistar rats during sural and peroneal nerve electrical stimulation. Five salient SFs were extracted from each recording electrode of a seven-electrode array. We used a machine learning approach to quantify and rank information content contained within DCN surface-potential signals following peripheral nerve activation. Machine-learning of SF and electrode position combinations was quantified to determine a hierarchy of information importance for resolving the peripheral origin of nerve activation. A supervised back-propagation artificial neural network (ANN) could predict the nerve from which a response was evoked with up to 96.8 ± 0.8% accuracy. Guided by feature-learnability, we maintained high prediction accuracy after reducing ANN algorithm inputs from 35 (5 SFs from 7 electrodes) to 6 (4 SFs from one electrode and 2 SFs from a second electrode). When the number of input features were reduced, the best performing input combinations included HF and LF features. Feature-learnability also revealed that signals recorded from the same midline electrode can be accurately classified when evoked from bilateral nerve pairs, suggesting DCN surface activity asymmetry. Here we demonstrate a novel method for mapping the information content of signal patterns across the DCN surface and show that DCN SFs are robust across a population. Finally, we also show that the DCN is functionally asymmetrically organized, which challenges our current understanding of somatotopic symmetry across the midline at sub-cortical levels.

The lateral cortex of the inferior colliculus (LCIC) is parcellated into two neurochemical compartments: one that comprises periodic neurochemical modules rich in GABAergic and cholinergic terminals and an extramodular matrix rich in calretinin neurons. We recently found that projections from auditory structures (auditory cortex and central nucleus of the IC) target the extramodular matrix, while somatosensory structures (somatosensory cortex and dorsal column nuclei) target the modules. What is peculiar about this finding of segregated inputs is that previous work has found that many LCIC neurons respond to both auditory and somatosensory stimuli. To investigate how these pathways interact, here we use laser photostimulation of caged glutamate to interrogate local LCIC circuits in brain slices from mouse. We found that most cell types in the LCIC receive inputs only from their home domain, but that GABAergic neurons in the modules serve as a bridge between modules and extramodular space. Further, we found that residence in- or out-of a module strongly predicted the output connectivity of that cell. These data suggest that distinct processing streams are seen in the LCIC and that GABAergic cells in modules serve to link these streams.

A wealth of mechanical information from the body generates various forms of sensory experience during touch or kinesthesia. Dorsal column nuclei (DCN) in the medulla are the first relay station for somatosensory inputs from peripheral receptors. These nuclei integrate somatosensory information and send the output to higher-order centers; therefore, investigating the firing patterns of DCN neurons should elucidate coding principles within the somatosensory system. In this study, we quantified the firing patterns of DCN neurons and examined whether the firing patterns of particular neurons are altered when moving tactile stimuli are applied in different directions. The activities of 17 neurons in the DCN of anesthetized rats were selected and their firing patterns were analyzed using LvR, which refers to the local variation of intervals of action potentials (i.e., the cross-correlation between consecutive intervals of action potentials) compensated by the refractoriness constant, R. The LvR of the 17 neurons ranged widely from 0.35 to 2.28. Of the 17 neurons, 12 responded to hair deflection (hair neurons), whereas five responded specifically to movement of forelimb joints. In 11 of 12 hair neurons, moving stimuli were applied in two to four different directions, which yielded 25 pairs of comparisons. Of these, 14 pairs (56%) showed significant differences in LvR. Among these 14 pairs, the range of LvR fluctuation was 0.13 ± 0.06 (mean ± standard deviation) and its effect size (Cohen's d) was 0.6 ± 0.2. These results suggest that the firing pattern of individual DCN neurons may contribute to somatosensory discrimination.

The dorsal column nuclei (DCN) of the brain stem contain secondary afferent neurons, which process ascending somatosensory information. Most of the known physiology of the DCN in primates has been acquired in acute experiments with anesthetized animals. Here, we developed a technique to implant a multielectrode array (MEA) chronically in the DCN of macaque monkeys to enable experiments with the animals awake. Two monkeys were implanted with brain-stem MEAs for 2–5 mo with no major adverse effects. Responses of the cuneate and gracile nuclei were quantified at the level of both field potentials and single units. Tactile receptive fields (RFs) were identified for 315 single units. A subset of these units had very regular spiking patterns with spike frequencies predominantly in the alpha band (8–14 Hz). The stability of the neuronal recordings was assessed with a novel analysis that identified units by their mean spike waveform and by the spike-triggered average of activity on all other electrodes in the array. Fifty-six identified neurons were observed over two or more sessions and in a few cases for as long as 1 mo. RFs of stable neurons were largely consistent across days. The results demonstrate that a chronic DCN implant in a macaque can be safe and effective, yielding high-quality unit recording for several months. The unprecedented access to these nuclei in awake primates should lead to a better understanding of their role in sensorimotor behavior.

Summary Tactile stimuli are integrated and processed by neuronal circuits in the deep dorsal horn of the spinal cord. Several spinal interneuron populations have been implicated in tactile information processing. However, dorsal horn projection neurons that contribute to the postsynaptic dorsal column (PSDC) pathway transmitting tactile information to the brain are poorly characterized. Here, we show that spinal neurons marked by the expression of Zic2creER mediate light touch sensitivity and textural discrimination. A subset of Zic2creER neurons are PSDC neurons that project to brainstem dorsal column nuclei, and chemogenetic activation of Zic2 PSDC neurons increases sensitivity to light touch stimuli. Zic2 neurons receive direct input from the cortex and brainstem motor nuclei and are required for corrective motor movements. These results suggest that Zic2 neurons integrate sensory input from cutaneous afferents with descending signals from the brain to promote corrective movements and transmit processed touch information back to the brain. Pinnipeds (sea lions, seals, and walruses) are notable for many reasons, including their ape-sized brains, their adaptation to a coastal niche that combines mastery of the sea with strong ties to land, and the remarkable abilities of their trigeminal whisker system. However, little is known about the central nervous system of pinnipeds. Here we report on the somatosensory areas of the nervous system of the California sea lion (Zalophus californianus). Using stains for Nissl, cytochrome oxidase, and vesicular glutamate transporters, we investigated the primary somatosensory areas in the brainstem, thalamus, and cortex in one sea lion pup and the external anatomy of the brain in a second pup. We find that the sea lion's impressive array of whiskers is matched by a large trigeminal representation in the brainstem with well-defined parcellation that resembles the barrelettes found in rodents but scaled upward in size. The dorsal column nuclei are large and distinct. The ventral posterior nucleus of the thalamus has divisions, with a large area for the presumptive head representation. Primary somatosensory cortex is located in the neocortex just anterior to the main vertical fissure, and precisely locating it as we do here is useful for comparing the highly gyrified pinniped cortex with that of other carnivores. To our knowledge this work is the first comprehensive report on the central nervous system areas for any sensory system in a pinniped. The results may be useful both in the veterinary setting and for comparative studies related to brain evolution.

Plastic changes in the CNS in response to peripheral sensory nerve injury are a series of complex processes, ranging from local circuit remodeling to somatotopic reorganization. However, the link between circuit remodeling and somatotopic reorganization remains unclear. We have previously reported that transection of the primary whisker sensory nerve causes the abnormal rewiring of lemniscal fibers (sensory afferents) on a neuron in the mouse whisker sensory thalamus (V2 VPM). In the present study, using transgenic mice whose lemniscal fibers originate from the whisker sensory principle trigeminal nucleus (PrV2) are specifically labeled, we identified that the transection induced retraction of PrV2-originating lemniscal fibers and invasion of those not originating from PrV2 in the V2 VPM. This anatomical remodeling with somatotopic reorganization was highly correlated with the rewiring of lemniscal fibers. Origins of the non-PrV2-origin lemniscal fibers in the V2 VPM included the mandibular subregion of trigeminal nuclei and the dorsal column nuclei (DCNs), which normally represent body parts other than whiskers. The transection also resulted in ectopic receptive fields of V2 VPM neurons and extraterritorial pain behavior on the uninjured mandibular region of the face. The anatomical remodeling, emergence of ectopic receptive fields, and extraterritorial pain behavior all concomitantly developed within a week and lasted more than three months after the transection. Our findings, thus, indicate a strong linkage between these plastic changes after peripheral sensory nerve injury, which may provide a neural circuit basis underlying large-scale reorganization of somatotopic representation and abnormal ectopic sensations.

In this study, we examined the role of the ventral posterior lateral nucleus (VPL) as a possible substrate for large-scale cortical reorganization in the forepaw barrel subfield (FBS) of primary somatosensory cortex (SI) that follows forelimb amputation. Previously, we reported that, 6 weeks after forelimb amputation in young adult rats, new input from the shoulder becomes expressed throughout the FBS that quite likely has a subcortical origin. Subsequent examination of the cuneate nucleus (CN) 1 to 30 weeks following forelimb amputation showed that CN played an insignificant role in cortical reorganization and led to the present investigation of VPL. As a first step, we used electrophysiological recordings in forelimb intact adult rats (n=8) to map the body representation in VPL with particular emphasis on the forepaw and shoulder representations and showed that VPL was somatotopically organized. We next used stimulation and recording techniques in forelimb intact rats (n=5) and examined the pattern of projection (a) from the forelimb and shoulder to SI, (b) from the forepaw and shoulder to VPL, and (c) from sites in the forepaw and shoulder representation in VPL to forelimb and shoulder sites in SI. The results showed that the projections were narrowly focused and homotopic. Electrophysiological recordings were then used to map the former forepaw representation in forelimb amputated young adult rats (n=5) at 7 to 24 weeks after amputation. At each time period, new input from the shoulder was observed in the deafferented forepaw region in VPL. To determine whether the new shoulder input in the deafferented forepaw VPL projected to a new shoulder site in the deafferented FBS, we examined the thalamocortical pathway in 2 forelimb-amputated rats. Stimulation of a new shoulder site in deafferented FBS antidromically-activated a cell in the former forepaw territory in VPL; however, similar stimulation from a site in the original shoulder representation, outside the deafferented region, in SI did not activate cells in the former forepaw VPL. These results suggest that the new shoulder input in deafferented FBS is relayed from cells in the former forepaw region in VPL. In the last step, we used anatomical tracing and stimulation and recording techniques in forelimb intact rats (n=9) to examine the cuneothalamic pathway from shoulder and forepaw receptive field zones in CN to determine whether projections from the shoulder zone might provide a possible source of shoulder input to forepaw VPL. Injection of biotinylated dextran amine (BDA) into physiologically identified shoulder responsive sites in CN densely labeled axon terminals in the shoulder representation in VPL, but also gave off small collateral branches into forepaw VPL. In addition, microstimulation delivered to forepaw VPL antidromically-activated cells in shoulder receptive field sites in CN. These results suggest that forepaw VPL also receives input from shoulder receptive sites in CN that are latent or subthreshold in forelimb intact rats. However, we speculate that following amputation these latent shoulder inputs become expressed, possibly as a down-regulation of GABA inhibition from the reticular nucleus (RTN). These results, taken together, suggest that VPL provides a substrate for large-scale cortical reorganization that follows forelimb amputation.

The dorsal fasciculi of the spinal cord are known as the chief pathways for the conduction of impulses underlying deep sensibility and tactile discrimination from limbs which reach the thalamus and ultimately the cerebral cortex after a relay at a bulbar level, the dorsal column nuclei. It is believed that the development of the dorsal fasciculi and their nuclei in the mammalian and primate series is correlated with increasing sensory discrimination in the skin and the increased development of proprioceptive sense in the limb musculature. In this review we will discuss the reflection of the development of limbs and sensory discrimination in lower animals and mammals on the organization and some functional aspects of these nuclei.

The current study examined the organization of the diencephalon and brainstem of the African elephant (Loxodonta africana) - a region of the elephant brain that has not been examined for at least 50 years. The current description, employing material amenable for use with modern neuroanatomical methods, shows that, for the most part, the elephant diencephalon and brainstem are what could be considered typically mammalian, with subtle differences in proportions and topology. The variations from these previous descriptions, where they occurred, were related to four specific aspects of neural information processing: (1) the motor systems, (2) the auditory and vocalization systems, (3) the orexinergic satiety/wakefulness centre of the hypothalamus and the locus coeruleus, and (4) the potential neurogenic lining of the brainstem. For the motor systems, three specific structures exhibited interesting differences in organization - the pars compacta of the substantia nigra, the facial motor nerve nucleus, and the inferior olivary nuclear complex, all related to the timing and learning of movements and likely related to the control of the trunk. The dopaminergic neurons of the substantia nigra appear to form distinct islands separated from each other by large fibre pathways, an appearance unique to the elephant. Each island may send topologically organized projections to the striatum forming a dopaminergic innervation mosaic that may relate to trunk movements. At least five regions of the combined vocalization production and auditory/seismic reception system were specialized, including the large and distinct nucleus ellipticus of the periaqueductal grey matter, the enlarged lateral superior olivary nucleus, the novel transverse infrageniculate nucleus of the dorsal thalamus, the enlarged dorsal column nuclei and the ventral posterior inferior nucleus of the dorsal thalamus. These specializations, related to production and reception of infrasound, allow the proposal of a novel concept regarding the reception and localization of infrasonic sources. The orexinergic system of the hypothalamus displayed a medial hypothalamic parvocellular cluster of neurons in addition to the magnocellular clusters typical of mammals located in the lateral hypothalamus, and a novel medial division of the locus coeruleus was observed in the pons. These systems are related to appetitive drive and promotion of wakefulness, two aspects of elephant behaviour that appear to be inextricably linked. Lastly, we observed an extensive potential neurogenic lining of the ventricles throughout the brainstem that is present in even quite old elephants, although the function of these cells remains elusive. These observations combined demonstrate that, while much of the elephant brainstem is typically mammalian, certain aspects of the anatomy related to specialized behaviour of elephants are present and instructive in understanding elephant behaviour.

Mitochondrial ferritin (FtMt), a recently-studied iron storage protein, which we suspect is an important defense against oxidative stress in neurons and elsewhere. The 242-amino acid FtMt precursor protein is cleaved to mature protein (of molecular weight about 22-kDa) in the mitochondrial matrix. Compared with the ubiquitously expressed traditional ferritin (H-ferritin and L-ferritin), FtMt has been found in fewer locations including the testis, heart and brain. Previous studies have reported that the expression of FtMt in mouse and human brain is predominantly localized to neurons and partly to glial cells, and FtMt exerts protective effects on neurons by maintaining normal function and regulates apoptosis in Alzheimer’s disease and Parkinson’s disease. To find out the function of FtMt in neurodegenerative disease, we had a novel antibody made against human FtMt and characterized it via Western blot analysis, immunoabsorption testing, and double immunofluorescence histochemistry. Then we used this new FtMt antibody to map the distribution of FtMt in the monkey brainstem. We demonstrated widespread distribution of FtMt immunoreactivity throughout the monkey brainstem, with variable staining intensity. FtMt immunoreactivity was observed in the extrapyramidal system, sensory trigeminal nerve nuclei, some motor nuclei including ambiguous nucleus, dorsal motor nucleus of the vagus and hypoglossal nucleus, and some dorsal column nuclei such as the gracile nucleus and cuneate nucleus. In addition, double immunohistochemical stainings confirmed that FtMt immunoreactivity was co-localized with catecholaminergic neurons in the locus coeruleus (63.64%), substantia nigra pars compacta (69.18%), and ventral tegmental area (56.89%). The distribution of FtMt which we found in the brainstem implies possible involvement of FtMt in several physiological mechanisms, especially in the catecholaminergic neurons, and the possibility of significant involvement in neurodegenerative disease.

The trigeminothalamic projections (a.k.a. trigeminal lemnniscus) arising from the principal sensory nucleus of the trigeminal nerve (PrV) form the orofacial component of the major ascending somatosensory pathway, the medial lemniscus. This pathway is entirely contralateral and terminates in the ventroposteromedial (VPM) nucleus of the thalamus (Erzurumlu et al., 1980; Ding et al., 2003). The somatosensory map of the body is also conveyed contralateral to the ventroposterolateral (VPL) nucleus of the thalamus via the dorsal column nuclei (reviewed in Tracey and Waite, 1995). The somatotopic face and body maps formed in these two thalamic nuclei are then conveyed to the primary somatosensory cortex via the thalamocortical projections. In rodents, relatively large volume of neural substrate is devoted to the point-to-point representation of the whiskers on the snout in the VPM (“barreloids”) and in layer IV (“barrels”) of the primary somatosensory cortex; these whisker-specific patterns are first initiated in the PrV and then sequentially relayed to the thalamus and cortex (reviewed in Sehara and Kawasaki, 2011). Numerous studies have focused on the development of the thalamocortical pathway and formation of the barrels (Erzurumlu and Kind, 2001; Inan and Crair, 2007; Pinon et al., 2009; Wu et al., 2011). In particular studies in mice with targeted gene mutations have revealed many of the underlying molecular mechanisms in thalamocortical axon guidance (Garel and Rubenstein, 2004). However, very little is known about the development of the trigeminal lemniscal pathway and the molecular guidance these axons use as they form a ribbon-shaped (lemniscus) pathway heading towards the contralateral thalamus. In the preceding paper we describe the normal development of the mouse trigeminothalamic pathway and here we address the molecular mechanisms of axon guidance as this tract acquires a contralateral course. Specialized midline cells of the vertebrate central nervous system secrete a variety of molecular signals, which attract or repel growing axons (Kennedy et al., 1994; Kidd et al., 1999). Commissural pathways and long axonal tracts such as the corticospinal and medial lemniscal pathways all interact with the neuroaxial midline during their establishment. Developmental regulation of the midline cues and specific receptors for them on the growing tips of axons play an essential role in contralateralization of these pathways. Slit and netrin proteins are abundant along the developing hindbrain and spinal cord midline, and the ascending somatosensory axons are endowed with cognate receptors. Three types of netrins (netrin-1, -3 and -4) have been identified in mammals (Moore et al., 2007). Netrin-1 is essential for proper extension of spinal commissural axons, corpus callosum and hippocampal commissure (Serafini et al., 1996). In the spinal cord, floor plate glial cells secrete netrin-1 and attract commissural axons towards the floor plate (Kennedy et al., 1994; Kennedy et al., 2006). In vertebrates netrin receptors are “deleted in colorectal cancer”(dcc), neogenin and four “uncoordinated” (unc) 5 proteins (unc5A-D). Axon attraction occurs when netrin-1 binds dcc (Moore et al., 2007). In Drosophila, “frazzled” is the name given to the member of the dcc gene family (Kolodziej et al., 1996) and mediates axon attraction upon binding netrin. Genetic and functional blockade of dcc results in loss of attraction towards the midline by commissural axons (Keino-Masu et al., 1996; Fazeli et al., 1997). Similarly, failure in midline crossing of internal arcuate fibers arising from the dorsal column nuclei has been reported for netrin-1 knockout mice (Kubota et al., 2004). Slit is an axon repellent protein, also secreted by the midline glia (Kidd et al., 1999). Three slit proteins (slit1-3) have been identified in mammals. They bind with roundabout or robo (robo1-4) receptors. Slit2 and slit1/2 double knockout mice display axon pathfinding defects in the corpus callosum, hippocampal commissure and thalamocortical projections (Bagri et al., 2002). Axonal pathfinding defects were also reported in robo1 knockout mice (Andrews et al., 2006; Jaworski et al., 2010). Our in situ hybridization assays indicated that the message for the slit and netrin proteins and their receptors are found at the right time and place during the development of the mouse trigeminal lemniscal pathway. We used dcc and slit 1, slit2, slit3 single or double knockout mice and observed marked path finding errors in trigeminothalamic projection axons of the PrV. Collectively, our results indicate that a cooperative signaling of attractant (netrin1) and repellent (slits) midline cues plays a fundamental role in guiding developing trigeminal lemniscal axons across the midline and contralateralization of the facial somatosensory inputs.

Somatosensory signals undergo substantial modulation in the dorsal column nuclei. We examined transmission of signals from forelimb afferents in primate cuneate and external cuneate nuclei. In anesthetized macaque monkeys, the median, ulnar, deep radial, and superficial radial nerves were electrically stimulated at 1.5–2× motor threshold with independent Poisson trains whereas extracellular recordings were made from 317 cells. Responses to peripheral stimulation included instances of both brief facilitation and long lasting suppression. A high proportion of cells (87%) responded to stimulation of two or more peripheral nerves, suggesting a large amount of convergence. Facilitated cells showed coherence with the peripheral stimulation across a broad frequency range; coherence was especially high in cells that responded with a burst of action potentials. Cells that responded with suppression also showed significant coherence, but this fell rapidly for frequencies above 25 Hz. Similar results were seen in both the main and external cuneate. When stimulation of one nerve was conditioned by a preceding nerve stimulus, the response to the second stimulus was attenuated for around 40 ms. This occurred independently of whether the first stimulus produced an initial facilitation or suppression or whether the same or a different nerve served as a conditioning stimulus. Mechanical stimulation of a receptive field suppressed responses to a second identical mechanical stimulus over a similar timescale. We conclude that the primate cuneate nucleus is capable of transmitting temporal information about stimuli with high fidelity; stimuli interact both temporally and spatially to modulate the onward transmission of information.

Hindbrain networks important for sensation and arousal contain diverse neuronal populations with distinct projections, yet share specific characteristics such as neurotransmitter expression. The relationship between the function of these neurons, their developmental origin, and the timing of their migration remains unclear. Mice lacking the proneural transcription factor Math1 ( Atoh1 ) lose neurons essential for hearing, balance, and unconscious proprioception. By using a new, inducible Math1 Cre * PR allele, we found that Math1 is also required for the conscious proprioceptive system, including excitatory projection neurons of the dorsal column nuclei and for vital components of the interoceptive system, such as Barrington's nucleus, that is closely associated with arousal. In addition to specific networks, Math1 lineages shared specific neurotransmitter expression, including glutamate, acetylcholine, somatostatin, corticotropin releasing hormone, and nitric oxide. These findings identify twenty novel Math1 lineages and indicate that the Math1 network functions partly as an interface for conscious (early-born) and unconscious (late-born) proprioceptive inputs to the cortex and cerebellum, respectively. In addition, these data provide previously unsuspected genetic and developmental links between proprioception, interoception, hearing, and arousal.

Clinical human and animal studies show that upper cervical spinal cord stimulation (cSCS) has beneficial effects in treatment of some cerebral disorders, including those due to deficient cerebral circulation. However, the underlying mechanisms and neural pathways activated by cSCS using clinical parameters remain unclear. We have shown that a cSCS-induced increase in cerebral blood flow is mediated via rostral spinal dorsal column fibers implying that the dorsal column nuclei (DCNs) are involved. The aim of this study was to examine how cSCS modulated neuronal activity of DCNs.. A spring-loaded unipolar ball electrode was placed on the left dorsal column at cervical (C2) spinal cord in pentobarbital anesthetized, ventilated and paralyzed male rats. Stimulation with frequencies of 1, 10, 20, 50 Hz (0.2 ms, 10 s) and an intensity of 90% of motor threshold was applied. Extracellular potentials of single neurons in DCNs were recorded and examined for effects of cSCS. In total, 109 neurons in DCNs were isolated and tested for effects of cSCS. Out of these, 56 neurons were recorded from the cuneate nucleus and 53 from the gracile nucleus. Mechanical somatic stimuli altered activity of 87/109 (83.2%) examined neurons. Of the neurons receiving somatic input, 62 were classified as low-threshold and 25 as wide dynamic range. The cSCS at 1 Hz changed the activity of 96/109 (88.1%) of the neurons. Neuronal responses to cSCS exhibited multiple patterns of excitation and/or inhibition: excitation (E, n=21), inhibition (I, n=19), E-I (n=37), I-E (n=8) and E-I-E (n=11). Furthermore, cSCS with high-frequency (50 Hz) altered the activity of 92.7% (51/55) of tested neurons, including 30 E, 24 I, and 2 I-E responses to cSCS. These data suggested that cSCS significantly modulates neuronal activity in dorsal column nuclei. These nuclei might serve as a neural relay for cSCS-induced effects on cerebral dysfunction and diseases.

We have investigated the alterations that the long-term implant of sieve electrodes to the peripheral nerves could evoke in the central nervous system by studying the neural activity at various levels of the somatosensory system [the implanted nerves, the dorsal column nuclei (DCN), and the primary somatosensory cortex (SI)] up to 30 months after implantation of the electrode in the distal median nerve in adult cats. This long survival period, which could correspond to 15 years within a human lifespan, enabled us to confirm the biocompatibility of the electrode. We also performed histological and molecular studies on sections of the spinal cord, DCN, and SI and electrophysiological recordings on of DCN and SI. Although in the ldquoimplantedrdquo regions (ipsilateral nerve, ipsilateral DCN, and contralateral SI) there was apparently a good recovery of a number of molecular markers, mostly related to local metabolism and neurotransmission in central relay structures, the expression of calcium-binding proteins in the cortex, which identifies inhibitory interneurons, remained clearly abnormal, although they were never as altered as in case of irreversible, chronic denervations. Also, prominent anatomical disorganization was detected in the normal spatial arrangement of neural clusters within the DCN. With respect to the electrical activity the ldquoimplanted siderdquo showed minor changes in response latency, intensity, and somatotopy, compared to control recordings. These findings show that central sensory-processing structures achieve fair, but not complete, levels of structural and functional reorganization following chronic intraneural implants of sieve electrodes.

The voltage-gated potassium channel subunit Kv3.1 confers fast firing characteristics to neurones. Kv3.1b subunit immunoreactivity (Kv3.1b-IR) was widespread throughout the medulla oblongata, with labelled neurones in the gracile, cuneate and spinal trigeminal nuclei. In the nucleus of the solitary tract (NTS), Kv3.1b-IR neurones were predominantly located close to the tractus solitarius (TS) and could be GABAergic or glutamatergic. Ultrastructurally, Kv3.1b-IR was detected in NTS terminals, some of which were vagal afferents. Whole-cell current-clamp recordings from neurones near the TS revealed electrophysiological characteristics consistent with the presence of Kv3.1b subunits: short duration action potentials (4.2 ± 1.4 ms) and high firing frequencies (68.9 ± 5.3 Hz), both sensitive to application of TEA (0.5 mm) and 4-aminopyridine (4-AP; 30 μm). Intracellular dialysis of an anti-Kv3.1b antibody mimicked and occluded the effects of TEA and 4-AP in NTS and dorsal column nuclei neurones, but not in dorsal vagal nucleus or cerebellar Purkinje cells (which express other Kv3 subunits, but not Kv3.1b). Voltage-clamp recordings from outside-out patches from NTS neurones revealed an outward K+ current with the basic characteristics of that carried by Kv3 channels. In NTS neurones, electrical stimulation of the TS evoked EPSPs and IPSPs, and TEA and 4-AP increased the average amplitude and decreased the paired pulse ratio, consistent with a presynaptic site of action. Synaptic inputs evoked by stimulation of a region lacking Kv3.1b-IR neurones were not affected, correlating the presence of Kv3.1b in the TS with the pharmacological effects.

The dorsal column nuclei, cuneatus and gracilis, play a fundamental role in the processing and integration of somesthetic ascending information. Intracellular and patch-clamp recordings obtained in cat in vivo have shown that cuneothalamic projection neurons present two modes of activity: oscillatory and tonic (Canedo et al 1998 Neuroscience 84 603-17). The former is the basis of generating, in sleep and anaesthetized states, slow, delta and spindle rhythms under the control of the cerebral cortex (Mariño et al 2000 Neuroscience 95 657-73). The latter is needed, during wakefulness, to process somesthetic information in real time. To study this behaviour we have developed the first realistic computational model of the cuneothalamic projection neurons. The modelling was guided by experimental recordings, which suggest the existence of hyperpolarization-activated inward currents, transient low- and high-threshold calcium currents, and calcium-activated potassium currents. The neuronal responses were simulated during (1) sleep, (2) transition from sleep to wakefulness and (3) wakefulness under both excitatory and inhibitory synaptic input. In wakefulness the model predicts a set of synaptically driven firing modes that could be associated with information processing strategies in the middle cuneate nucleus.

Prevention of nerve injury-induced tactile, but not thermal, hypersensitivity is achieved by ipsilateral lesions of the dorsal columns or lidocaine microinjection into the nucleus gracilis (n. gracilis). These and other data support the possibility that tactile hyperresponsiveness after nerve injury may be selectively mediated by a low-threshold myelinated fiber pathway to the n. gracilis. Here we identify a transmitter that might selectively mediate such injury-induced tactile hypersensitivity. Neuropeptide Y (NPY), normally not detected in the dorsal root ganglion (DRG) or in the n. gracilis of rats, became markedly upregulated at both sites and in the spinal cord after spinal nerve injury. Injury-induced NPY-IR occurred predominately in large-diameter DRG cells, and the NPY-IR in the n. gracilis was blocked by dorsal rhizotomy or dorsal column lesion. NPY microinjection into the n. gracilis of uninjured rats elicited reversible tactile, but not thermal, hypersensitivity only in the ipsilateral hindpaw. Administration of anti-NPY antiserum, but not control serum or preabsorbed serum, into the n. gracilis ipsilateral to nerve injury reversed tactile, but not thermal, hypersensitivity. Similarly, microinjection of the NPY antagonists NPY18–36 and ( R )- N -[[4-(aminocarbonylaminomethyl)-phenyl]methyl]- N 2-(diphenylacetyl)-argininamide trifluoroacetate, into the n. gracilis ipsilateral to the injury reversed tactile, but not thermal, hypersensitivity. Antagonist administration into the contralateral n. gracilis had no effect on injury-induced hypersensitivity. These data suggest the selective mediation of nerve injury-induced tactile hypersensitivity by upregulated NPY via large fiber input to n. gracilis. Selective reversal of injury-induced tactile allodynia by NPY receptor antagonists would have significant implications for human neuropathic conditions.

In this study, sensory maps in the thalamus were investigated by examining their volume and shape. We determined the forelimb representation in adult rats after the removal of hindlimb input by nucleus gracilis lesions. Three-dimensional reconstructions of thalamic sensory maps were obtained from a grid of electrode penetrations. We found that the volume of the shoulder sensory map contracted >50% at an acute time interval (n= 6), followed by a robust volumetric sensory map expansion of 25% at 1 week (n= 8) and 1 month (n= 8) after lesion relative to controls (n= 8). The topology of the volumetric increase was scrutinized by slicing functional maps in the coronal, sagittal, and horizontal planes. The equivalence of such slices from each animal was established by virtue of their distance from either a functional or neuroanatomical landmark. Surprisingly, all of the volumetric increase unequivocally occurred in a circumscribed coronal slice 300 μm thick. This focal zone was located toward the rostral pole of the thalamic tactile relay, the ventroposterolateral nucleus. Analysis in the sagittal plane revealed that, unexpectedly, the shoulder map volume expanded by superimposing its representation on that of the forepaw, via an advancement of the shoulder representation by 0.6 mm medially. We propose a “hot spot” hypothesis in which focal zones of plasticity may not be specific to the thalamus but may have manifestations elsewhere in the nervous system, such as the cerebral cortex or dorsal column nuclei.

1. Partial deafferentation, based on peripheral nerve section or local anaesthetic blockade, has been reported to induce both immediate loss of responsiveness and/or immediate reorganization in receptive fields of neurones in the somatosensory system. In the present study, in anaesthetized cats, we have used a rapid, reversible deafferentation procedure based on cold block of the median nerve in order to evaluate quantitatively the response characteristics of cuneate neurones (n = 39) before, during and after partial deafferentation. 2. The first hypothesis tested was that cuneate neurones with input from ulnar or superficial radial nerve fields in the vicinity of the median nerve field should undergo, in association with median nerve blockade, an increased level of responsiveness to tactile stimuli within the ulnar or radial nerve zone, and an expansion of their cutaneous receptive fields. However, among eighteen cuneate neurones of this type, there was no evidence for any systematic enhancement of responsiveness nor, in at least sixteen of the eighteen neurones, any evidence for receptive field expansion. 3. The second hypothesis tested was that cuneate neurones whose input came from both the median nerve and another peripheral nerve source should undergo, in association with median nerve blockade, an increase in responsiveness to the remaining input and an expansion of the receptive field into the field of that remaining nerve source. However, in none of thirteen neurones of this type tested was there evidence of such a change. 4. The third hypothesis was that cuneate neurones whose control' receptive fields were within the median nerve zone of deafferentation should show an emergence of novel receptive fields and responsiveness from areas around the field of innervation of the median nerve. However, in none of eight neurones of this type was there evidence for such changes in adjacent skin areas. 5. In conclusion, with the use of cold block of the median nerve for partial deafferentation, the present study has confirmed previous findings of denervation-related loss of responsiveness in dorsal column nuclei neurones. The conflicting findings in studies of central nervous system plasticity indicate the need to understand better factors that do and do not lead to acute central changes.

The present paper is aimed at defining distinctive subdivisions of the human cuneate nucleus (Cu), evident from prenatal to old life, whose occurrence has never been clearly formalized in the human brain, or described in other species so far. It extends our early observations on the presence of gray matter areas that host strong substance P (SP) immunoreactivity in the territory of the human Cu and adjacent cuneate fascicle. Here we provide a three-dimensional reconstruction of the Cu fields rich in SP and further identify those areas by means of their immunoreactivity to the neuropeptides SP, calcitonin gene-related peptide, methionine- and leucine-enkephalin, peptide histidine-isoleucine, somatostatin and galanin, to the trophins glial cell line-derived neurotrophic factor and brain-derived neurotrophic factor, and to the neuroplasticity proteins polysialylated neural cell adhesion molecule and growth-associated protein-43. The presence, density and distribution of immunoreactivity for each of these molecules closely resemble those occurring in the superficial layers of the caudal spinal trigeminal nucleus (Sp5C). Myelin and Nissl stainings suggest that those Cu subregions and the Sp5C superficial layers share a similar histological aspect. This work establishes the existence of definite subregions, localized within the Cu territory, that bear the neurochemical and histological features of sensory nuclei committed to the neurotransmission of protopathic stimuli, including pain. These findings appear of particular interest when considering that functional, preclinical and clinical studies show that the dorsal column nuclei, classical relay station of fine somatic tactile and proprioceptive sensory stimuli, are also involved in pain neurotransmission.

Information about bodily events is conveyed by primary sensory fibres to higher brain centres through neurons in the dorsal column nuclei (DCN) and spinal dorsal horn. The DCN route is commonly considered a ‘touch pathway’, separate from the spinal ‘pain pathway’, in part because DCN neurons respond to gentle tactile stimulation of small skin areas. Here we report that DCN neurons can additionally respond to gentle and noxious stimulation of viscera and widespread skin regions. These and other experimental and clinical data suggest that the DCN and spinal routes cooperate, rather than operate separately, to produce the many perceptions of touch and pain, an ensemble view that encourages novel approaches to health care and research.

The rostral ventrolateral medulla (RVLM) primarily regulates respiration and the autonomic nervous system. Its medial portion (mRVLM) contains many choline acetyltransferase (ChAT)-immunoreactive (ir) neurons of unknown function. We sought to clarify the role of these cholinergic cells by tracing their axonal projections. We first established that these neurons are neither parasympathetic preganglionic neurons nor motor neurons because they did not accumulate intraperitoneally administered Fluorogold. We traced their axonal projections by injecting a Cre-dependent vector (floxed-AAV2) expressing either GFP or mCherrry into the mRVLM of ChAT-Cre mice. Transduced neurons expressing GFP or mCherry were confined to the injection site and were exclusively ChAT-ir. Their axonal projections included the dorsal column nuclei, medullary trigeminal complex, cochlear nuclei, superior olivary complex and spinal cord lamina III. For control experiments, the floxed-AAV2 (mCherry) was injected into the RVLM of dopamine beta-hydroxylase-Cre mice. In these mice, mCherry was exclusively expressed by RVLM catecholaminergic neurons. Consistent with data from rats, these catecholaminergic neurons targeted brain regions involved in autonomic and endocrine regulation. These regions were almost totally different from those innervated by the intermingled mRVLM-ChAT neurons. This study emphasizes the advantages of using Cre-driver mouse strains in combination with floxed-AAV2 to trace the axonal projections of chemically defined neuronal groups. Using this technique, we revealed previously unknown projections of mRVLM-ChAT neurons and showed that despite their close proximity to the cardiorespiratory region of the RVLM, these cholinergic neurons regulate sensory afferent information selectively and presumably have little to do with respiration or circulatory control.

This study aims to determine the dysfunction caused by existing pathological condition in structures involved in the transfer of sensory functions of the neural system in cervical disc herniation, and to establish whether or not the level and degree of this anatomical damage can be anticipated by SEP (Somatosensorial Evoked Potentials). We compared the obtained SEP values for statistical significance using the Friedman Variation Analysis. In parameters with statistical significance, the Wilcoxon Signed Rank test was used to identify when significant improvements occurred. The study found that the statistical data of the latency of the N14 wave originating from the dorsal column nuclei of the medulla spinalis and dorsal column gray matter improved (p < 0.05) in the postoperative period compared with the preoperative values. Using the Wilcoxon Signed Rank test, we studied postoperative months separately in regard to the difference in the latency of the N14 wave, and found the statistically significant improvement to be marked particularly in months 3 and 6 postoperatively (p < 0.05). In conclusion, we suggest that SEP is a useful tool to check the functional condition of the dorsal spinal column. The benefit of the SEP utilization is the ability to determine the severity of the pathological condition preoperatively and follow the patient's functional postoperative improvement.

The synaptic relationships formed by medial lemniscal (ML) or spinothalamic tract (STT) axon terminals with neurons of the somatosensory ventroposterolateral thalamic nucleus of the macaque monkey have been examined quantitatively by electron microscopy. ML and STT axons were labeled by the anterograde axon transport of WGA-HRP following injection of the tracer into the contralateral dorsal column nuclei, or the dorsal horn of the spinal cord, respectively. Thalamic tissue was histochemically reacted for the presence of HRP. Serial thin sections were stained with a gold-labeled antibody to GABA, to determine which neuronal elements exhibited GABA immunoreactivity (GABA- ir). Serially sectioned thalamic structures were recorded in electron micrographs and reconstructed in three dimensions by computer. Individual ML axon terminals form multiple synaptic contacts with segments of the proximal dendritic trees of thalamocortical relay neurons and also synapse upon the dendritic appendages of GABA-ir interneurons (local circuit neurons). These GABA-ir dendritic appendages contain synaptic vesicles and are presynaptic (presynaptic dendrites) to the same segments of relay neuron dendrites that receive ML contacts. When analyzed in serial sections and reconstructed by computer, the ML terminals form triadic relationships (ML, GABA appendage, and relay neuron dendrite) or more complex glomerular arrangements involving multiple appendages, all of which then contact the relay neuron dendritic segment. In contrast, multiple STT terminals make synaptic contacts along segments of projection neuron dendrites and are usually the only type of profile to contact that segment of dendrite. More than 85% of the spinal afferents form simple axodendritic synapses with relay cells and do not contact GABA-ir appendages. The thalamic synaptic relationships of ML terminals are fundamentally different from those formed by the STT. Because STT neurons predominatly transmit information about noxious stimuli, the simple axodendritic circuitry of the majority of these spinal afferents suggests that the transmission of noxious information is probably not subject to GABAergic modulation by thalamic interneurons, in contrast to the GABAergic processing of non-noxious information carried by the ML afferents. The differences in the GABAergic circuits of the thalamus that mediate ML and STT afferent information are believed to underlie differential somatosensory processing in the forebrain. We suggest that changes in thalamic GABAergic dendritic appendages and GABA receptors following CNS injury may play a role in the genesis of some central pain states.

Transplantation of neural progenitor cells (NPC) is a promising therapeutic strategy for replacing neurons lost after spinal cord injury, but significant challenges remain regarding neuronal integration and functional connectivity. Here we tested the ability of graft-derived neurons to reestablish connectivity by forming neuronal relays between injured dorsal column (DC) sensory axons and the denervated dorsal column nuclei (DCN). A mixed population of neuronal and glial restricted precursors (NRP/GRP) derived from the embryonic spinal cord of alkaline phosphatase (AP) transgenic rats were grafted acutely into a DC lesion at C1. One week later, BDNF-expressing lentivirus was injected into the DCN to guide graft axons to the intended target. Six weeks later, we observed anterogradely traced sensory axons regenerating into the graft and robust growth of graft-derived AP-positive axons along the neurotrophin gradient into the DCN. Immunoelectron microscopy revealed excitatory synaptic connections between regenerating host axons and graft-derived neurons at C1 as well as between graft axons and DCN neurons in the brainstem. Functional analysis by stimulus-evoked c-Fos expression and electrophysiological recording showed that host axons formed active synapses with graft neurons at the injury site with the signal propagating by graft axons to the DCN. We observed reproducible electrophysiological activity at the DCN with a temporal delay predicted by our relay model. These findings provide the first evidence for the ability of NPC to form a neuronal relay by extending active axons across the injured spinal cord to the intended target establishing a critical step for neural repair with stem cells.
