Abstract: The European Spallation Source, currently under construction in Lund, Sweden, is a multidisciplinary international laboratory. Once completed to full specifications, it will operate the world's most powerful pulsed neutron source. Supported by a 3 million Euro Research and Innovation Action within the EU Horizon 2020 program, a design study (HighNESS) has been completed to develop a second neutron source located below the spallation target. Compared to the first source, designed for high cold and thermal brightness, the new source has been optimized to deliver higher intensity, and a shift to longer wavelengths in the spectral regions of cold (CN, 2--20\,\AA), very cold (VCN, 10--120\,\AA), and ultracold (UCN, ${>}\,{500}$\,\AA) neutrons. The second source comprises a large liquid deuterium moderator designed to produce CN and support secondary VCN and UCN sources. Various options have been explored in the proposed designs, aiming for world-leading performance in neutronics. These designs will enable the development of several new instrument concepts and facilitate the implementation of a high-sensitivity neutron-antineutron oscillation experiment (NNBAR). Abstract: In this work we report on measurements of neutron absorption in supermirror coatings. The measurements were carried out using the SuperADAM instrument at the Institut Laue-Langevin and by measuring the gamma-ray production from m = 3 and m = 4 neutron supermirrors when illuminated by a beam of neutrons. The results provide a valuable validation for recent computational and theoretical work that can be used as input to Monte-Carlo radiation transport calculations for the design of the shielding of neutron scattering instruments.

Abstract: The HIBEAM/NNBAR program is a proposed two-stage experiment at the European Spallation Source focusing on searches for baryon number violation via processes in which neutrons convert to antineutrons. This paper outlines the computing and detector simulation framework for the HIBEAM/NNBAR program. The simulation is based on predictions of neutron flux and neutronics together with signal and background generation. A range of diverse simulation packages are incorporated, including Monte Carlo transport codes, neutron ray-tracing simulation packages, and detector simulation software. The common simulation package in which these elements are interfaced together is discussed. Data management plans and triggers are also described., Comment: Contribution to CHEP2021. Abstract: The European Spallation Source being constructed in Lund, Sweden will provide the user community with a neutron source of unprecedented brightness. By 2025, a suite of 15 instruments will be served by a high-brightness moderator system placed above the spallation target. The ESS infrastructure, consisting of the proton linac, the target station, and the instrument halls, allows for implementation of a second source below the spallation target. We propose to develop a second neutron source with a high-intensity moderator able to (1) deliver a larger total cold neutron flux, (2) provide high intensities at longer wavelengths in the spectral regions of Cold (4-10 \AA ), Very Cold (10-40 \AA ), and Ultra Cold (several 100 \AA ) neutrons, as opposed to Thermal and Cold neutrons delivered by the top moderator. Offering both unprecedented brilliance, flux, and spectral range in a single facility, this upgrade will make ESS the most versatile neutron source in the world and will further strengthen the leadership of Europe in neutron science. The new source will boost several areas of condensed matter research such as imaging and spin-echo, and will provide outstanding opportunities in fundamental physics investigations of the laws of nature at a precision unattainable anywhere else. At the heart of the proposed system is a volumetric liquid deuterium moderator. Based on proven technology, its performance will be optimized in a detailed engineering study. 5. Population-based metaheuristic optimization in neutron optics and shielding design

Abstract: Population-based metaheuristic algorithms are powerful tools in the design of neutron scattering instruments and the use of these types of algorithms for this purpose is becoming more and more commonplace. Today there exists a wide range of algorithms to choose from when designing an instrument and it is not always initially clear which may provide the best performance. Furthermore, due to the nature of these types of algorithms, the final solution found for a specific design scenario cannot always be guaranteed to be the global optimum. Therefore, to explore the potential benefits and differences between the varieties of these algorithms available, when applied to such design scenarios, we have carried out a detailed study of some commonly used algorithms. For this purpose, we have developed a new general optimization software package which combines a number of common metaheuristic algorithms within a single user interface and is designed specifically with neutronic calculations in mind. The algorithms included in the software are implementations of Particle-Swarm Optimization (PSO), Differential Evolution (DE), Artificial Bee Colony (ABC), and a Genetic Algorithm (GA). The software has been used to optimize the design of several problems in neutron optics and shielding, coupled with Monte-Carlo simulations, in order to evaluate the performance of the various algorithms. Abstract: A key aim of the HighNESS project for the European Spallation Source is to enable cutting-edge particle physics experiments. This volume presents a conceptual design report for the NNBAR experiment. NNBAR would exploit a new cold lower moderator to make the first search in over thirty years for free neutrons converting to anti-neutrons. The observation of such a baryon-number-violating signature would be of fundamental significance and tackle open questions in modern physics, including the origin of the matter-antimatter asymmetry. This report shows the design of the beamline, supermirror focusing system, magnetic and radiation shielding, and anti-neutron detector necessary for the experiment. A range of simulation programs are employed to quantify the performance of the experiment and show how background can be suppressed. For a search with full background suppression, a sensitivity improvement of three orders of magnitude is expected, as compared with the previous search. Civil engineering studies for the NNBAR beamline are also shown, as is a costing model for the experiment.

7. Study of neutron shielding collimators for curved beamlines at the European Spallation Source

Abstract: The European Spallation Source is being constructed in Lund, Sweden and is planned to be the world's brightest pulsed spallation neutron source for cold and thermal neutron beams ($\le$ 1 eV). The facility uses a 2 GeV proton beam to produce neutrons from a tungsten target. The neutrons are then moderated in a moderator assembly consisting of both liquid hydrogen and water compartments. Surrounding the moderator are 22 beamports, which view the moderator's outside surfaces. The beamports are connected to long neutron guides that transport the moderated neutrons to the sample position via reflections. As well as the desired moderated neutrons, fast neutrons coming directly from the target can find their way down the beamlines. These can create unwanted sources of background for the instruments. To mitigate such a kind of background, several instruments will use curved guides to lose direct line-of-sight (LoS) to the moderator and the target. In addition instruments can also use shielding collimators to reduce the amount of fast neutrons further traveling down the guide due to albedo reflections or streaming. Several different materials have been proposed for this purpose. Abstract: Over the last decades, the field of thermal neutron detection has overwhelmingly employed He-3-based technologies. The He-3 crisis together with the forthcoming establishment of the European Spallation Source have necessitated the development of new technologies for neutron detection. Today, several promising He-3-free candidates are under detailed study and need to be validated. This validation process is in general long and expensive. The study of detector prototypes using neutron-emitting radioactive sources is a cost-effective solution, especially for preliminary investigations. That said, neutron-emitting sources have the general disadvantage of broad, structured, emitted-neutron energy ranges. Further, the emitted neutrons often compete with unwanted backgrounds of gamma-rays, alpha-particles, and fission-fragments. By blending experimental infrastructure such as shielding to provide particle beams with neutron-detection techniques such as tagging, disadvantages may be converted into advantages. In particular, a technique known as tagging involves exploiting the mixed-field generally associated with a neutron-emitting source to determine neutron time-of-flight and thus energy on an event-by-event basis. This allows for the definition of low-cost, precision neutron beams. The Source-Testing Facility, located at Lund University in Sweden and operated by the SONNIG Group of the Division of Nuclear Physics, was developed for just such low-cost studies. Precision tagged-neutron beams derived from radioactive sources are available around-the-clock for advanced detector diagnostic studies. Neutron measurements performed at the Source Testing Facility are thus cost-effective and have a very low barrier for entry. 9. Measurements and Monte-Carlo simulations of the particle self-shielding effect of B4C grains in neutron shielding concrete

Abstract: A combined measurement and Monte-Carlo simulation study was carried out in order to characterize the particle self-shielding effect of B4C grains in neutron shielding concrete. Several batches of a specialized neutron shielding concrete, with varying B4C grain sizes, were exposed to a 2 {\AA} neutron beam at the R2D2 test beamline at the Institute for Energy Technology located in Kjeller, Norway. The direct and scattered neutrons were detected with a neutron detector placed behind the concrete blocks and the results were compared to Geant4 simulations. The particle self-shielding effect was included in the Geant4 simulations by calculating effective neutron cross-sections during the Monte-Carlo simulation process. It is shown that this method well reproduces the measured results. 10. A Polyethylene-B4C based concrete for enhanced neutron shielding at neutron research facilities

Abstract: We present the development of a specialized concrete for neutron shielding at neutron research facilities, based on the addition of hydrogen atoms in the form of polyethylene and also B$_{4}$C for enhancing the neutron capture properties of the concrete. We show information on the mechanical properties of the concrete and the neutronics, in particular it's relevance to modern spallation neutron sources, such as the European Spallation Source (ESS), currently under construction in Lund, Sweden. The new concrete exhibits a 15% lower mass density, a compressible strength of 50% relative to a standard concrete and a significant increase in performance of shielding against MeV neutrons and lower energies. The concrete could find application at the ESS in for example common shielding components, individual beamline shielding and instrument caves. Abstract: A key aim of the HighNESS project for the European Spallation Source is to enable cutting-edge particle physics experiments. This volume presents a conceptual design report for the NNBAR experiment. NNBAR would exploit a new cold lower moderator to make the first search in over thirty years for free neutrons converting to anti-neutrons. The observation of such a baryon-number-violating signature would be of fundamental significance and tackle open questions in modern physics, including the origin of the matter-antimatter asymmetry. This report shows the design of the beamline, supermirror focusing system, magnetic and radiation shielding, and anti-neutron detector necessary for the experiment. A range of simulation programs are employed to quantify the performance of the experiment and show how background can be suppressed. For a search with full background suppression, a sensitivity improvement of three orders of magnitude is expected, as compared with the previous search. Civil engineering studies for the NNBAR beamline are also shown, as is a costing model for the experiment.

Abstract: The long (3ms) proton pulse of the European Spallation Source (ESS) gives rise to unique and potentially high backgrounds for the instrument suite. In such a source an instrument capabilities will be limited by it's Signal to Noise (S/N) ratio. The instruments with a direct view of the moderator, which do not use a bender to help mitigate the fast neutron background, are the most challenging. For these beam lines we propose the innovative shielding of placing blocks of material directly into the guide system, which allow a minimum attenuation of the cold and thermal fluxes relative to the background suppression. This shielding configuration has been worked into a beam line model using Geant4. Abstract: Sub-barrier Coulomb-excitation was performed on a mixed beam of $^{62}$Mn and $^{62}$Fe, following in-trap $\beta^{-}$ decay of $^{62}$Mn at REX-ISOLDE, CERN. The trapping and charge breeding times were varied in order to alter the composition of the beam, which was measured by means of an ionisation chamber at the zero-angle position of the Miniball array. A new transition was observed at 418~keV, which has been tentatively associated to a $(2^{+},3^{+})\rightarrow1^{+}_{g.s.}$ transition. This fixes the relative positions of the $\beta$-decaying $4^{+}$ and $1^{+}$ states in $^{62}$Mn for the first time. Population of the $2^{+}_{1}$ state was observed in $^{62}$Fe and the cross-section determined by normalisation to the $^{109}$Ag target excitation, confirming the $B(E2)$ value measured in recoil-distance lifetime experiments., Comment: 9 pages, 10 figures

Abstract: Background: Shape coexistence in heavy nuclei poses a strong challenge to state-of-the-art nuclear models, where several competing shape minima are found close to the ground state. A classic region for investigating this phenomenon is in the region around $Z=82$ and the neutron mid-shell at $N=104$. Purpose: Evidence for shape coexistence has been inferred from $\alpha$-decay measurements, laser spectroscopy and in-beam measurements. While the latter allow the pattern of excited states and rotational band structures to be mapped out, a detailed understanding of shape coexistence can only come from measurements of electromagnetic matrix elements. Method: Secondary, radioactive ion beams of $^{202}$Rn and $^{204}$Rn were studied by means of low-energy Coulomb excitation at the REX-ISOLDE facility in CERN. Results: The electric-quadrupole ($E2$) matrix element connecting the ground state and first-excited $2^{+}_{1}$ state was extracted for both $^{202}$Rn and $^{204}$Rn, corresponding to ${B(E2;2^{+}_{1} \to 2^{+}_{1})=29^{+8}_{-8}}$ W.u. and $43^{+17}_{-12}$ W.u., respectively. Additionally, $E2$ matrix elements connecting the $2^{+}_{1}$ state with the $4^{+}_{1}$ and $2^{+}_{2}$ states were determined in $^{202}$Rn. No excited $0^{+}$ states were observed in the current data set, possibly due to a limited population of second-order processes at the currently-available beam energies. Conclusions: The results are discussed in terms of collectivity and the deformation of both nuclei studied is deduced to be weak, as expected from the low-lying level-energy schemes. Comparisons are also made to state-of-the-art beyond-mean-field model calculations and the magnitude of the transitional quadrupole moments are well reproduced., Comment: 12 pages, 14 figures

Abstract: The nucleosynthesis of elements beyond iron is dominated by neutron captures in the s and r processes. However, 32 stable, proton-rich isotopes cannot be formed during those processes, because they are shielded from the s-process flow and r-process beta-decay chains. These nuclei are attributed to the p and rp process. For all those processes, current research in nuclear astrophysics addresses the need for more precise reaction data involving radioactive isotopes. Depending on the particular reaction, direct or inverse kinematics, forward or time-reversed direction are investigated to determine or at least to constrain the desired reaction cross sections. The Facility for Antiproton and Ion Research (FAIR) will offer unique, unprecedented opportunities to investigate many of the important reactions. Abstract: We present recent developments of improved modelling methods for simulating neutron transport in reflector materials of interest for neutron source applications. These include materials to be used as traditional reflectors around the neutron moderator, such as beryllium, and also novel materials, such as nanodiamond particles, to be used as a reflector for very-cold neutrons in the neutron beam extraction area of a neutron scattering instrument. Of particular interest is the inclusion of physical effects that are not modelled in standard thermal scattering libraries used for Monte-Carlo simulations, such as extinction in beryllium reflectors and effects of small-angle neutron scattering from nanodiamond particles.

Abstract: In this work we report on measurements of neutron absorption in supermirror coatings. The measurements were carried out using the SuperADAM instrument at the Institut Laue-Langevin and by measuring the gamma-ray production from m = 3 and m = 4 neutron supermirrors when illuminated by a beam of neutrons. The results provide a valuable validation for recent computational and theoretical work that can be used as input to Monte-Carlo radiation transport calculations for the design of the shielding of neutron scattering instruments.

Abstract: The violation of baryon number, , is an essential ingredient for the preferential creation of matter over antimatter needed to account for the observed baryon asymmetry in the Universe. However, such a process has yet to be experimentally observed. The HIBEAM/NNBAR program is a proposed two-stage experiment at the European Spallation Source to search for baryon number violation. The program will include high-sensitivity searches for processes that violate baryon number by one or two units: free neutron–antineutron oscillation () via mixing, neutron–antineutron oscillation via regeneration from a sterile neutron state (), and neutron disappearance (n → n'); the effective process of neutron regeneration () is also possible. The ESS infrastructure, consisting of the proton linac, the target station, and the instrument halls, allows for implementation of a second source below the spallation target. We propose to develop a second neutron source with a high-intensity moderator able to (1) deliver a larger total cold neutron flux, (2) provide high intensities at longer wavelengths in the spectral regions of Cold (4–10 Å), Very Cold (10–40 Å), and Ultra Cold (several 100 Å) neutrons, as opposed to Thermal and Cold neutrons delivered by the top moderator. Offering both unprecedented brilliance, flux, and spectral range in a single facility, this upgrade will make ESS the most versatile neutron source in the world and will further strengthen the leadership of Europe in neutron science. The new source will boost several areas of condensed matter research such as imaging and spin-echo, and will provide outstanding opportunities in fundamental physics investigations of the laws of nature at a precision unattainable anywhere else. At the heart of the proposed system is a volumetric liquid deuterium moderator. Based on proven technology, its performance will be optimized in a detailed engineering study. This moderator will be complemented by secondary sources to provide intense beams of Very- and Ultra-Cold Neutrons.

Abstract: A combined measurement and Monte-Carlo simulation study was carried out in order to characterize the particle self-shielding effect of B4C grains in neutron shielding concrete. Several batches of a specialized neutron shielding concrete, with varying B4C grain sizes, were exposed to a 2 angstrom neutron beam at the R2D2 test beamline at the Institute for Energy Technology located in Kjeller, Norway. The direct and scattered neutrons were detected with a neutron detector placed behind the concrete blocks and the results were compared to Geant4 simulations. The particle self-shielding effect was included in the Geant4 simulations by calculating effective neutron cross-sections during the Monte-Carlo simulation process. It is shown that this method well reproduces the measured results. Our results show that shielding calculations for low-energy neutrons using such materials would lead to an underestimate of the shielding required for a certain design scenario if the particle self shielding effect is not included in the calculations.

Abstract: The nucleosynthesis of elements beyond iron is dominated by neutron captures in the s and r processes. However, 32 stable, proton-rich isotopes cannot be formed during those processes, because they are shielded from the s-process flow and r-process, β-decay chains. These nuclei are attributed to the p and rp process. For all those processes, current research in nuclear astrophysics addresses the need for more precise reaction data involving radioactive isotopes. Depending on the particular reaction, direct or inverse kinematics, forward or time-reversed direction are investigated to determine or at least to constrain the desired reaction cross sections. The Facility for Antiproton and Ion Research (FAIR) will offer unique, unprecedented opportunities to investigate many of the important reactions. 26. Systematic study on the performance of elliptic focusing neutron guides

Abstract: In neutron scattering experiments there is an increasing trend towards the study of smaller volume samples, which make the use of focusing optics more important. Focusing guide geometries based on conic-sections, such as those with parabolic and elliptic shapes, have been extensively used in both recently built neutron instruments and upgrades of existing hardware. A large fraction of proposed instruments at the European Spallation Source feature the requirement of good performance when measuring on small samples. The optimised design of a focusing system comes after time consuming Monte-Carlo (MC) simulations. Therefore, in order to help reduce the time needed to design such focusing systems, it is necessary to study systematically the performance of focusing guides. In the present work, we perform a theoretical analysis of the focusing properties of neutron beams, and validate them using a combination of Monte-Carlo simulations and Particle Swarm Optimisations (PS0s), where there is a close correspondence between the maximum divergence of the beam and the shape of the guide. The analytical results show that two limits can be considered, which bound a range of conic section shapes that provide optimum performance. Finally, we analyse a more realistic guide example and we give an assessment of the importance of the contribution from multiple reflections in different systems.

27. High-energy in-beam neutron measurements of metal-based shielding for accelerator-driven spallation neutron sources

Abstract: Metal-based shielding plays an important role in the attenuation of harmful and unwanted radiation at an accelerator-driven spallation neutron source. At the European Spallation Source, currently under construction in Lund, Sweden, metal-based materials are planned to be used extensively as neutron guide substrates in addition to other shielding structures around neutron guides. The usage of metal-based materials in the vicinity of neutron guides however requires careful consideration in order to minimize potential background effects in a neutron instrument at the facility. Therefore, we have carried out a combined study involving high-energy neutron measurements and Monte Carlo simulations of metal-based shielding, both to validate the simulation methodology and also to investigate the benefits and drawbacks of different metal-based solutions. The measurements were carried out at The Svedberg Laboratory in Uppsala, Sweden, using a 174.1 MeV neutron beam and various thicknesses of aluminum-, iron-, and copper-based shielding blocks. The results were compared to Geant4 simulations and revealed excellent agreement. Abstract: Population-based metaheuristic algorithms are powerful tools in the design of neutron scattering instruments and the use of these types of algorithms for this purpose is becoming more and more commonplace. Today there exists a wide range of algorithms to choose from when designing an instrument and it is not always initially clear which may provide the best performance. Furthermore, due to the nature of these types of algorithms, the final solution found for a specific design scenario cannot always be guaranteed to be the global optimum. Therefore, to explore the potential benefits and differences between the varieties of these algorithms available, when applied to such design scenarios, we have carried out a detailed study of some commonly used algorithms. For this purpose, we have developed a new general optimization software package which combines a number of common metaheuristic algorithms within a single user interface and is designed specifically with neutronic calculations in mind. The algorithms included in the software are implementations of Particle-Swarm Optimization (PSO), Differential Evolution (DE), Artificial Bee Colony (ABC), and a Genetic Algorithm (GA). The software has been used to optimize the design of several problems in neutron optics and shielding, coupled with Monte Carlo simulations, in order to evaluate the performance of the various algorithms. Generally, the performance of the algorithms depended on the specific scenarios, however it was found that DE provided the best average solutions in all scenarios investigated in this work.

Abstract: We present a survey of the radiation background at the Spallation Neutron Source (SNS) at Oak Ridge National Laboratory, TN, USA during routine daily operation. A broad range of detectors was used to characterize primarily the neutron and photon fields throughout the facility. These include a WENDI-2 extended range dosimeter, a thermoscientific NRD, an Arktis 4He detector, and a standard NaI photon detector. The information gathered from the detectors was used to map out the neutron dose rates throughout the facility and also the neutron dose rate and flux profiles of several different beamlines. The survey provides detailed information useful for developing future shielding concepts at spallation neutron sources, such as the European Spallation Source (ESS), currently under construction in Lund, Sweden.

Abstract: To investigate the systematics of mixed-symmetry states in N = 52 isotones, a relativistic Coulomb excitation experiment was performed during the PreSPEC campaign at the GSI Helmholtzzentrum fur Schwerionenforschung to determine E2 transition strengths to 2(+) states of the radioactive nucleus Kr-88. Absolute transition rates could be measured towards the first and third 2+ states. For the latter a mixed-symmetry character is suggested on the basis of the indication for a strong M1 transition to the fully symmetric 2(1)(+) state, extending the knowledge of the N = 52 isotones below Z = 40. A comparison with the proton-neutron interacting boson model and shell-model predictions is made and supports the assignment.

Abstract: Sub-barrier Coulomb excitation was performed on a mixed beam of Mn-62 and Fe-62, following in-trap beta(-) decay of Mn-62 at REX-ISOLDE, CERN. The trapping and charge breeding times were varied in order to alter the composition of the beam, which was measured by means of an ionisation chamber at the zero-angle position of the Miniball array. A new transition was observed at 418 keV, which has been tentatively associated to a (2(+),3(+)) -> 1(g.s.)(+) transition. This fixes the relative positions of the beta-decaying 4(+) and 1(+) states in Mn-62 for the first time. Population of the 2 (+)(1) state was observed in Fe-62 and the cross-section determined by normalisation to the Ag-109 target excitation, confirming the B(E2) value measured in recoil-distance lifetime experiments.

Sn104 is the lightest isotope in the Sn chain for which this quantity has been measured. The result is a key point in the discussion of the evolution of nuclear structure in the proximity of the doubly magic nucleus Sn100. The value B(E2;0+→2 +)=0.10(4) e2b2 is significantly lower than earlier results for Sn106 and heavier isotopes. Abstract: A set of B(E2) values for the low-lying excited states in the radioactive isotope 109Sn were deduced from a Coulomb excitation experiment. The 2.87-MeV/u radioactive beam was produced at the REX-ISOLDE facility at CERN and was incident on a secondary 58Ni target. The B(E2) values were determined using the known 2+ → 0+ reduced transition probability in 58Ni as normalization with the semi-classical Coulomb excitation code GOSIA2. The transition probabilities are compared to shell-model calculations based on a realistic nucleon-nucleon interaction and the predictions of a simple core-excitation model. This measurement represents the first determination of multiple B(E2) values in a light Sn nucleus using the Coulomb excitation technique with low-energy radioactive beams. Abstract: Nuclear shell evolution in neutron-rich Na nuclei around N = 20 was studied by determining reduced transition probabilities, i.e., B(E2) and B(M1) values, in order to map the border of the island of inversion. To this end Coulomb-excitation experiments, employing radioactive Na-29,Na-30 beams with a final beam energy of 2.85 MeV/nucleon, were performed at REX-ISOLDE, CERN. De-excitation gamma rays were detected by the MINIBALL gamma-ray spectrometer in coincidence with scattered particles in a segmented Si detector. Transition probabilities to excited states were deduced. The measured B(E2) values agree well with shell-model predictions, supporting the idea that in the Na isotopic chain the ground-state wave function contains significant intruder admixture already at N = 18, with N = 19 having an almost pure two-particle-two-hole deformed ground-state configuration.

Abstract: The analysis of the Coulomb excitation experiment conducted on 104 Sn required a strict selection of the data in order to reduce the large background present in the Î³-ray energy spectra and identify the Î³-ray peak corresponding to the Coulomb excitation events. As a result the B(E2; 0 + â 2 + ) value could be extracted, which established the downward trend towards 100 Sn and therefore the robustness of the N=Z=50 core against quadrupole excitations.

Abstract: Reduced transition strengths of the deexciting transitions from the first two excited states in 33Ar were measured in a relativistic Coulomb excitation experiment at the GSI Helmholtz center. The radioactive ion beam was produced by fragmentation of a primary 36Ar beam on a 9Be target followed by the selection of the reaction product of interest via the GSI Fragment Separator. The 33Ar beam hit a secondary 197Au target with an energy of approximately 145 MeV/nucleon. An array of high-purity germanium cluster detectors and large-volume BaF2 scintillator detectors were employed for γ -ray spectroscopy at the secondary target position. The Lund-York-Cologne Calorimeter was used to track the outgoing ions and to identify the nuclear reaction channels. For the two lowest energy excited states of 33Ar the reduced transition strengths have been determined. With these first results the Tz = −3/2 nucleus 33Ar is now, together with 21Na (Tz = −1/2), the only neutron-deficient odd-A sd shell nucleus in which experimental transition strengths are available. The experimental values are compared to results of shell-model calculations which describe simultaneously mirror-energy differences and transition-strength values of mirror pairs in the sd shell in a consistent way

Abstract: Excited states of the nucleus Er-170 have been studied by Coulomb excitation using the GASP gamma-ray detector system at the Laboratori Nazionali di Legnaro. The ground-state band along with a low-lying K-pi = 0(+) band and gamma-vibrational band were populated during the experiment. Based on the measured gamma-ray yields, a set of interband and intraband matrix elements has been extracted using the Coulomb excitation code GOSIA. The resulting E2 matrix elements are compared to collective model predictions.

49. The type 8 adenylyl cyclase is critical for Ca2+ stimulation of cAMP accumulation in mouse parotid acini.

Abstract: Capacitative Ca(2+) entry stimulates cAMP synthesis in mouse parotid acini, suggesting that one of the Ca(2+)-sensitive adenylyl cyclases (AC1 or AC8) may play an important role in the regulation of parotid function (Watson, E. L., Wu, Z., Jacobson, K. L., Storm, D. R., Singh, J. C., and Ott, S. M. (1998) Am. J. Physiol. 274, C557-C565). To evaluate the role of AC1 and AC8 in Ca(2+) stimulation of cAMP synthesis in parotid cells, acini were isolated from AC1 mutant (AC1-KO) and AC8 mutant (AC8-KO) mice and analyzed for Ca(2+) stimulation of intracellular cAMP levels. Although Ca(2+) stimulation of intracellular cAMP levels in acini from AC1-KO mice was indistinguishable from wild type mice, acini from AC8-KO mice showed no Ca(2+)-stimulated cAMP accumulation. This indicates that AC8, but not AC1, plays a major role in coupling Ca(2+) signals to cAMP synthesis in parotid acini. Interestingly, treatment of acini from AC8-KO mice with agents, i.e. carbachol and thapsigargin that increase intracellular Ca(2+), lowered cAMP levels. This decrease was dependent upon Ca(2+) influx and independent of phosphodiesterase activation. Immunoblot analysis revealed that AC5/6 and AC3 are expressed in parotid glands. Inhibition of calmodulin (CaM) kinase II with KN-62, or inclusion of the CaM inhibitor, calmidazolium, did not prevent agonist-induced inhibition of stimulated cAMP accumulation. In vitro studies revealed that Ca(2+), independently of CaM, inhibited isoproterenol-stimulated AC. Data suggest that agonist augmentation of stimulated cAMP levels is due to activation of AC8 in mouse parotid acini, and strongly support a role for AC5/6 in the inhibition of stimulated cAMP levels.

50. Ryanodine receptor type III (Ry3R) identification in mouse parotid acini. Properties and modulation of [3H]ryanodine-binding sites.

Abstract: Immunoblot analysis and [3H]ryanodine binding were used to characterize and identify ryanodine receptors (RyRs) in nonexcitable mouse parotid acini. Western analysis revealed ryanodine receptor type III (Ry3R) to be the only detectable isoform in parotid microsomal membranes. Binding of [3H]ryanodine to microsomal fractions was dependent on Ca2+, salt, pH, and temperature. At 23 degrees C, and in the presence of 0.5 M KCl and 100 microM Ca2+, [3H]ryanodine bound specifically to membranes with high affinity (Kd = 6 nM); maximum binding capacity (Bmax) was 275 fmol/mg protein. Mg2+ and ruthenium red inhibited [3H]ryanodine binding (IC50 = 1.4 mM and 0.5 microM, respectively). 4-Chloro-3-ethylphenol enhanced the binding of [3H]ryanodine 2.5-fold; whereas ATP and caffeine were much less efficacious toward activating Ry3R (56% and 18% maximal enhancement, respectively). Bastadin, a novel modulator of the 12-kDa FK506 binding protein.RyR complex, increased [3H]ryanodine binding 3-4-fold by enhancing Kd. The immunosuppressant FK506 enhanced [3H]ryanodine receptor occupancy at >100 microM and antagonized the action of bastadin, suggesting that an immunophilin modulates Ry3R in parotid acini. These results suggest that Ry3R may play an important role in Ca2+ homeostasis in mouse parotid acini.
