The carotid body (CB) is a polymodal chemosensory organ that plays an essential role in initiating respiratory and cardiovascular adjustments to maintain blood gas homeostasis. It is strategically located at the carotid bifurcation. The CB works in concert with the apposing afferent nerve endings of the petrosal ganglion (PG) cells and together they form a functional unit, the CB chemosensory system. The organ consists of small clusters called glomeruli composed of two cell types, glomus and sustentacular cells, interspersed by blood vessels and nerve bundles, and separated by connective tissue. During chemotransduction glomus cells release a variety of neurotransmitters which activate chemoafferent nerve endings of PG neurons. Much of the available evidence suggests that the CB dysfunction and altered oxygen homeostasis are involved in the pathophysiology of several diseases including systemic hypertension. Our recent data show that in glomus cells of hypertensive animals the production of nitric oxide is impaired and components of the neurotrophin signaling system display enhanced expression. These results suggest that a heightened chemosensory discharge may contribute to sympathetic hyperactivity leading to hypertension. Knowledge of the morphofunctional and neurochemical aspects of the CB would improve our current understanding of respiratory and cardiovascular homeostasis in health and disease.

Changes occurring in acupuncture and sublingually administration of medicines also have a morphological basis, which can be objectively assessed. Objective: Refine of the methodology for evaluating morphological changes associated with tongue acupuncture. Material and methods: Selection of experimental animals and appropriate protocol for material collection, fixation, staining and counting of selected morphological structures and elements. Results: The samples fixed with formalin 10% were fixed well, the consistency of the tissues remained hard and the cuts were obtained well in all thicknesses (from 5 to 15 μm). Cuts from 5 to 7 μm are suitable for visualizing the structure of the tongue. The sagittal cuts provide the largest area for single-cut exploration, as well as are more suitable than others to track the acupuncture channel in the chosen for our study point. Conclusions: The study can be carried out in rats, but if possible, it is preferable to take the material from pigs. The use of formalin 10% gives a better texture to the material and facilitates the cutting with the microtome. It is necessary to use a variety of colorings due to the wide range of structures that are involved with the acupuncture needle tract.

PURPOSE: The aim is to find out the mast cells (MCs) reaction in tongue after experimental acupuncture. METHODS: For experiments were carried 10 adults rats (28 months age). The needles used for the acupuncture is 0.22x13mm, and were placed for 10 minutes into standard acupuncture point Ex-HN-10 (Juquan) corresponding to that of humans. This point is located on the upper surface in the sagittal plane of the tongue and is close to the center of the tongue body. As normal consequence of every acupuncture is the forming of a needle tract and also here in the tissues of the rats tongue we could demonstrate this. This was done with a visualization method for the needle tract that we developed for tongue. The proximity of the needle tract was examined for MCs. Two stains were used for proper visualization: Toluidine blue and Bismarck brown staining. RESULTS: In close proximity of the needle tract we observed degranulation of MCs that was massive and few destroyed MCs in the needle tract itself. At a considerable distance from the MCs some discharged granules from them was found. CONCLUSIONS: There is a MCs reaction on the acupuncture of tongue that includes a degranulation of the MCs that was massive in proximity of the acupuncture needle tract. Some of the effects ot acupuncture could be due to the demonstrated MCs degranulation.

Relics are a special type of human body remains, which are venerated by the faithful due to the prominence the person had achieved during lifetime. Even if no such claims for incorruptibility are made, body relics have to be preserved in some fashion, in order to allow for their long-term viewing and veneration. The present report summarizes the available data regarding the impregnation as a method for body preservation, used by the Christian Church to conserve body relics. Impregnation is a widely used method, both by the Orthodox as well as by the Roman Catholic Church. It is known that bodies are treated with different protective mixtures on variable intervals after death. Public worship historically involved pouring of oil over relics. Moreover, small tissue fragments are embedded in wax blocks or are sealed with a mixture of molten wax and other resins. All of those methods are in fact impregnation with compounds with high molecular mass as a final stage of a preservation procedure. This impregnation of a tissue already reached some degree of dehydration or other preservation ensures longevity and protection of the relics by greatly slowing down the natural processes of decomposition and disintegration.

Abstract. In the field and boundary of certificated field for organic farming in the Institute of Agriculture – Karnobat have been observed and described diseases, pests and weeds in wild and cultivated plants and dependencies between them are reported. The cultural plant diversity at the certified field for organic farming is significant and amounts to 32 species and varieties of cereals. The weeds in crops and in the free areas significantly differed. In crops, the annual broadleaf weeds had greater diversity of species (Anthemis arvensis L., Lathyrus aphaca L., Papaver phoeas L., Polygonum aviculare L., Polygonum convolvulus L.) and at the boundaries – the cereal weeds (Аpera spica-venti L., Hordeum murinum L., Bromus tectorum L., Poa pratensis L.). At the adjacent areas the perennial broadleaf weeds – Cirsium arvense (L.) Scop. and Convolvulus arvensis L. were not found. At the boundaries less pest species and types of diseases were registered than in the crop fields. In cereals the most common pests are Sitobion avenae L., Aelia acuminate L., Eurygaster austriaca Schr. Beneficial insects are Coccinella septempunctata L. The most common diseases are Erysiphe graminis and Rhynchosporium secalis (Oudem.) Davis

Abstract. On a certified organic field a trial of the hull-less oat variety Mina was conducted, sown after peas-sunflower mixture, maize for silage, and fallow. The aim of this experiment was to evaluate the effect of predecessors on the productivity and phytosanitary condition of hull-less oats in organic farming. The study was conducted in the period 2012 – 2014 under field conditions on leached, moderately loamy to slightly clayey smolniza at the Institute of Agriculture in Karnobat, Bulgaria, on a field certified for organic farming. The trial was conducted with spring hull-less oat, variety Mina. Predecessors – peas-sunflower mixture, maize for silage and fallow. Phytosanitary monitoring was conducted. The insect reporting was performed through direct investigation for plant pests. The weed reporting – species composition and density of weed infestation by a quantity-weight method, at the end of tillering and beginning of ear formation stage. Disease infestation was determined by walking through the plots during crop vegetation, by the route method, by inspection of the plants. The best predecessor for hull-less oats was fallow, followed by peas-sunflower mixture. The yield was high and the level of weed infestation was comparatively lower than after maize for silage. During the studied period a tendency for the effect of predecessor on disease infestation was not formed. The predecessors did not affect the species composition but mainly their density

Abstract. During the period of 2013 – 2015 the influence of farming types (organic and biodinamic) on the phytosanitary status and yield of kamut was studied. Kamut is resistant to diseases and they are not a problem for growing it in an organic and biodynamic way. In both kinds of farming single numbers of five types of pests on kamut were recorded. Two of them were from Order Homoptera (Sitobion avenae L. and Rhopalosiphum maidis (Fitch)) and three from Hemiptera (Philaenus spumarius (L.), Eurygaster integriceps Put. and Aelia acuminata L.).The species diversity of the weeds was greater in biodynamic farming. Kamut yield from biodynamic farming was proven to exceed the organic one. The strength of effect of the type of farming factor was 83.57%.

Abstract. Until recently, Penthaleus major (Dugès) has not been recognized as an economically significant pest for the cereal crops. After climatic changes, its population began to grow and inflict damages around the world. The aim of this study was to investigate its distribution in Bulgaria and establish whether it presents a danger to the cereal crops. In the autumn of 2015 and the spring of 2016, a monitoring survey was conducted to establish Penthaleus major (Dugès) with the cereal crops in Bulgaria. Over 60 sowed fields were investigated from all around the country. Samples were taken to identify the pest. It was established that Penthaleus major (Dugès) inflicted harm to the wheat in north-eastern (12-14 mites per stem) and south-eastern Bulgaria (6-8 mites per stem). Its density was under the threshold of economic harm. There was not found infestation of barley, rye, oat and triticale. On the field boundaries bordering the areas attacked by the mite were reported the following weeds: Capsella bursa pastoris (L.) Medic, Descurania sophia (L.) Welb. et Berth, Senecio spp., Sisymbrium orientale Torn., Taraxsacum officinale Weber, Anthemis spp., Bromus arvensis L., Eragrostis pilosa (L.) P.B. Lolium temulentum L., which can be habitat for Penthaleus major (Dugès).

Preoperative radiotherapy (RT) for non-metastatic rectal cancer reduces local recurrence rates but can cause pelvic insufficiency fractures. Despite the high morbidity from RT-induced skeletal injuries, predictive and preventive measures are lacking. How these injuries are reflected by bone biomarkers are largely unknown. The aim was to assess longitudinal changes in bone biomarkers and their relation to RT-related bone injuries in women with rectal cancer. This longitudinal cohort study includes 47 women with non-metastatic rectal cancer treated with surgery ± preoperative RT with or without chemotherapy. Sclerostin, bioactive sclerostin, C-terminal telopeptide cross-links of collagen type I (CTX), bone-specific alkaline phosphatase (BALP), and type I procollagen intact N-terminal propeptide (PINP) were measured at baseline, after RT, and 1 yr postoperatively. Pelvic magnetic resonance imaging was used for detection of skeletal injury. Sixteen of 36 (44%) irradiated women had radiation-induced bone injuries and were compared to 11 women (RT-) and 20 women (RT+) without bone injuries. Serum CTX, BALP, and PINP increased during the first year after RT in women with radiation-induced bone injuries. The difference in mean change of CTX ( p =.037) and BALP ( p =.042) was conferred by longitudinal regression analyses adjusted for serum estradiol. Serum sclerostin and bioactive sclerostin remained stable over time. Taken together, bone markers may be of interest for future research on fracture prediction or preventive measures in women susceptible to radiation-induced bone injury. The pivotal role of the basolateral amygdala (BLA) in the emotional modulation of hippocampal plasticity and memory consolidation is well-established. Specifically, multiple studies have demonstrated that the activation of the noradrenergic (NA) system within the BLA governs these modulatory effects. However, most current evidence has been obtained by direct infusion of synthetic NA or beta-adrenergic agonists. In the present study, we aimed to investigate the effect of endogenous NA release in the BLA, induced by a natural aversive stimulus (coyote urine), on memory consolidation for a low-arousing, hippocampal-dependent task. Our experiments combined a weak object location task (OLT) version with subsequent mild predator odor exposure (POE). To investigate the role of endogenous NA in the BLA in memory modulation, a subset of the animals (Wistar rats) was treated with the non-selective beta-blocker propranolol at the end of the behavioral procedures. Hippocampal tissue was collected 90 min after drug infusion or after the OLT test, which was performed 24 h later. We used the obtained samples to estimate the levels of phosphorylated CREB (pCREB) and activity-regulated cytoskeleton-associated protein (Arc)-two molecular markers of experience-dependent changes in neuronal activity. The result suggests that POE has the potential to become a valuable behavioral paradigm for studying the interaction between BLA and the hippocampus in memory prioritization and selectivity.

A plethora of pathophysiological events have been shown to play a synergistic role in neurodegeneration, revealing multiple potential targets for the pharmacological modulation of Alzheimer's disease (AD). In continuation to our previous work on new indole- and/or donepezil-based hybrids as neuroprotective agents, the present study reports on the beneficial effects of lead compounds of the series on key pathognomonic features of AD in both cellular and in vivo models. An enzyme-linked immunosorbent assay (ELISA) was used to evaluate the anti-fibrillogenic properties of 15 selected derivatives and identify quantitative changes in the formation of neurotoxic β-amyloid (Aβ42) species in human neuronal cells in response to treatment. Among the most promising compounds were 3a and 3c , which have recently shown excellent antioxidant and anticholinesterase activities, and, therefore, have been subjected to further in vivo investigation in mice. An acute toxicity study was performed after intraperitoneal (i.p.) administration of both compounds, and 1/10 of the LD 50 (35 mg/kg) was selected for subacute treatment (14 days) with scopolamine in mice. Donepezil (DNPZ) and/or galantamine (GAL) were used as reference drugs, aiming to establish any pharmacological superiority of the multifaceted approach in battling hallmark features of neurodegeneration. Children with hemato-oncological diseases may have significant skeletal morbidity, not only during and after treatment but also at the time of diagnosis before cancer treatment. This study was designed to evaluate the vitamin D status and circulating bone metabolic markers and their determinants in children at the time of diagnostic evaluation for hemato-oncological disease. This cross-sectional study included 165 children (91 males, median age 6.9 yr range 0.2-17.7 yr). Of them, 76 patients were diagnosed with extracranial or intracranial solid tumors, 83 with leukemia, and 6 with bone marrow failure. Bone metabolism was assessed by measuring serum 25OHD, PTH, bone alkaline phosphatase, intact N-terminal propeptide of type I procollagen, and C-terminal cross-linked telopeptide of type I collagen. Vitamin D deficiency was found in 30.9% of children. Lower 25OHD levels were associated with older age, lack of vitamin D supplementation, season outside summer, and a country of parental origin located between latitudes -45° and 45°. Children diagnosed with leukemia had lower levels of markers of bone formation and bone resorption than those who had solid tumors or bone marrow failure. In conclusion, vitamin D deficiency was observed in one-third of children with newly diagnosed cancer. Bone turnover markers were decreased in children with leukemia, possibly because of the suppression of osteoblasts and osteoclasts by leukemic cells. The identification of patients with suboptimal vitamin D status and compromised bone remodeling at cancer diagnosis may aid in the development of supportive treatment to reduce the adverse effects of cancer and its treatment., Competing Interests: None declared., (© The Author(s) 2024. The aims of this study were to (1) establish the intrapopulation and seasonal variation of Juniperus excelsа essential oil (EO); (2) compare the J. excelsa concrete and resinoid composition with its EO composition; and (3) investigate the potential herbicidal activity of J. excelsa EO against seeds of Papaver rhoeas L., Consolida orientalis (J.Gay) Schrödinger, Anthemis arvensis L., Avena fatua L., and Agrostemma githago L. Four independent studies were performed to meet these objectives. Twenty-eight individual trees were analyzed from two populations to establish intrapopulation and interpopulation variability of EOs yield and composition. In the seasonal dynamic study of leaf EO, samples from the same three trees and in the same population were collected in January, March, May, July, October, and December and their EO yield and composition determined. The EOs (intrapopulation and seasonal) were extracted by hydrodistillation, while the EO for the herbicidal test was obtained by steam distillation in a semi-commercial (SCom) apparatus. Overall, the EO yield varied significantly from 0.93% to 2.57%. α-Pinene (8.85-35.94%), limonene (11.81-50.08%), and cedrol (3.41-34.29%) were the predominant EO compounds in all samples (intrapopulation variability); however, trans-2,4-decadienol and β-caryophyllene were predominant in some individual trees. Four chemical groups were identified in the samples collected from two natural populations (intrapopulation). This is the first report on the compositions of J. excelsa concrete and resinoid. Cedrol (15.39%), 7-hydroxy-4-methyl-coumarin (17.63%), 1-octacosanol (36.85%), tritriacontane (16.08%), and tiacontanoic acid were the main compounds in the concrete and resinoid. Juniperus excelsa EO suppressed seed germination and seedling growth of P. rhoeas, C. orientalis, A. arvensis, A. fatua, and A. githago, demonstrating its potential to be used for the development of new biopesticides. Tissue-nonspecific alkaline phosphatase (TNALP) is a glycoprotein expressed by osteoblasts that promotes bone mineralization. TNALP catalyzes the hydrolysis of the mineralization inhibitor inorganic pyrophosphate and ATP to provide inorganic phosphate, thus controlling the inorganic pyrophosphate/inorganic phosphate ratio to enable the growth of hydroxyapatite crystals. N-linked glycosylation of TNALP is essential for protein stability and enzymatic activity and is responsible for the presence of different bone isoforms of TNALP associated with functional and clinical differences. The site-specific glycosylation profiles of TNALP are, however, elusive. TNALP has 5 potential N-glycosylation sites located at the asparagine (N) residues 140, 230, 271, 303, and 430. The objective of this study was to reveal the presence and structure of site-specific glycosylation in TNALP expressed in osteoblasts. Calvarial osteoblasts derived from Alpl +/- expressing SV40 Large T antigen were transfected with soluble epitope-tagged human TNALP. Purified TNALP was analyzed with a lectin microarray, matrix-assisted laser desorption/ionization-time of flight mass spectrometry, and liquid chromatography with tandem mass spectrometry. The results showed that all sites ( n  = 5) were fully occupied predominantly with complex-type N-glycans. High abundance of galactosylated biantennary N-glycans with various degrees of sialylation was observed on all sites, as well as glycans with no terminal galactose and sialic acid. Furthermore, all sites had core fucosylation except site N271. Modelling of TNALP, with the protein structure prediction software ColabFold, showed possible steric hindrance by the adjacent side chain of W270, which could explain the absence of core fucosylation at N271. These novel findings provide evidence for N-linked glycosylation on all 5 sites of TNALP, as well as core fucosylation on 4 out of 5 sites. Chitosan is a natural biopolymer with a proven ability to impart textile materials with antimicrobial properties when loaded onto them. The mechanism of its bacteriological activity depends on the contact between the positive and negative charges of the amino groups located on the surface of the microbes. Unfortunately, the type of microorganisms and pH influence this action-shortcomings that can be avoided by chitosan modification and by loading its film with substances possessing antimicrobial properties. In this study, chitosan was modified with benzaldehyde and crosslinked with glutaraldehyde to form a film on the surface of cotton fabric (CB). Also, another material was obtained by including zinc oxide particles (CBZ) synthesized in situ into the chitosan coating. The performed analyses (contact angle measurement, optical and scanning electron microscopy, FTIR, XRD, and thermal analysis) evidenced the modification of the cotton fabric and the alteration of the film properties after zinc oxide inclusion. A comparison of the antimicrobial properties of the new CB with materials prepared with chitosan without benzaldehyde from our previous study verified the influence of the hydrophobicity and surface roughness of the fabric surface on the enhancement of antimicrobial activity. The microbial growth inhibition increased in the following order: fungal strain Candida lipolytica >Gram-positive bacteria Bacillus cereus >Gram-negative bacteria Pseudomonas aeruginosa. The samples containing zinc oxide particles completely inhibited the growth of all three model strains. The virucidal activity of the CB was higher against human adenovirus serotype 5 (HAdV-5) than against human respiratory syncytial virus (HRSV-S2) after 60 min of exposure. The CBZ displayed higher virucidal activity with a Δlog of 0.9 against both viruses.

The difficult-to-heal wounds continue to be a problem for modern medicine. Chitosan and diosgenin possess anti-inflammatory and antioxidant effects making them relevant substances for wound treatment. That is why this work aimed to study the effect of the combined application of chitosan and diosgenin on a mouse skin wound model. For the purpose, wounds (6 mm diameter) were made on mice's backs and were treated for 9 days with one of the following: 50% ethanol (control), polyethylene glycol (PEG) in 50% ethanol, chitosan and PEG in 50% ethanol (Chs), diosgenin and PEG in 50% ethanol (Dg) and chitosan, diosgenin and PEG in 50% ethanol (ChsDg). Before the first treatment and on the 3rd, 6th and 9th days, the wounds were photographed and their area was determined. On the 9th day, animals were euthanized and wounds' tissues were excised for histological analysis. In addition, the lipid peroxidation (LPO), protein oxidation (POx) and total glutathione (tGSH) levels were measured. The results showed that ChsDg had the most pronounced overall effect on wound area reduction, followed by Chs and PEG. Moreover, the application of ChsDg maintained high levels of tGSH in wound tissues, compared to other substances. It was shown that all tested substances, except ethanol, reduced POx comparable to intact skin levels. Therefore, the combined application of chitosan and diosgenin is a very promising and effective medication for wound healing.

Background: Relatives of people with psychosis or bipolar disorder experience high levels of distress but are typically not offered the support they need. Online peer forums may offer a solution, but knowledge about who uses them, how, and why is limited. This study reported on online forum use during the Relatives Education and Coping Toolkit (REACT) trial., Objective: We aimed to report who used the forum and why; how sociodemographic factors are associated with participation; the relationship among frequency, type of use, and outcomes; and how the forum was used., Methods: The relationships between key sociodemographic characteristics, levels of forum use, and distress were statistically analyzed. We used thematic and semantic analyses to understand the reasons for relatives joining the forum and the key topics initiated by them. We also used the University Centre for Computer Corpus Research on Language Semantic Analysis System to compare how relatives and REACT supporters (moderators) used the forum., Results: A total of 348 participants with full forum use data from REACT were included in this study. The forum was accessed by 59.4% (207/348) of the relatives across the entire age range, with no significant associations between sociodemographic factors and forum participation, or between level or type of use and relatives' distress levels. Relatives joined the forum primarily to find people in similar circumstances, express concerns, and talk about stressful events. Relatives were most concerned about recent events, negative emotions linked to caring, experiences of conflict or threat, and concerns about suicide. These posts underscored both the challenges the relatives were facing and the fact that they felt safe sharing them in this context., Conclusions: Although only a proportion of REACT participants engaged actively with its forum, they were widely distributed across age and other sociodemographic groupings. Relatives used the forum for information, support, and guidance and to offer detailed information about their experiences. Multiple sclerosis (MS), an autoimmune-driven, inflammatory demyelinating disease of the central nervous system (CNS), causes irreversible accumulation of neurological deficits to a variable extent. Although there are potent disease-modifying agents for its initial relapsing-remitting phase, immunosuppressive therapies show limited efficacy in secondary progressive MS (SPMS). Although modulation of sphingosine-1 phosphate receptors has proven beneficial during SPMS, the underlying mechanisms are poorly understood. In this project, we followed the hypothesis that siponimod, a sphingosine-1 phosphate receptor modulator, exerts protective effects by direct modulation of glia cell function (i.e., either astrocytes, microglia, or oligodendrocytes). To this end, we used the toxin-mediated, nonautoimmune MS animal model of cuprizone (Cup) intoxication. On the histological level, siponimod ameliorated cuprizone-induced oligodendrocyte degeneration, demyelination, and axonal injury. Protective effects were evident as well using GE180 translocator protein 18-kDa (TSPO) imaging with positron emission tomography (PET)/computed tomography (CT) imaging or next generation sequencing (NGS). Siponimod also ameliorated the cuprizone-induced pathologies in Rag1 -deficient mice, demonstrating that the protection is independent of T and B cell modulation. Proinflammatory responses in primary mixed astrocytes/microglia cell cultures were not modulated by siponimod, suggesting that other cell types than microglia and astrocytes are targeted. Of note, siponimod completely lost its protective effects in S1pr5 -deficient mice, suggesting direct protection of degenerating oligodendrocytes. Our study demonstrates that siponimod exerts protective effects in the brain in a S1PR5-dependent manner. This finding is not just relevant in the context of MS but in other neuropathologies as well, characterized by a degeneration of the axon-myelin unit.

Surface-initiated photopolymerization has been run to synthesize a hydrogel with ZnO particles distributed uniformly along its structure, which has been loaded onto a polyamide fabric. Three samples have been obtained at different concentrations of zinc nitrate (10% (sample PA10); 20% (sample PA20) and 30% (sample PA30) of the weight of the fabric, respectively)) and subjected to gravimetric analysis, scanning electron microscopy and transmission electron microscopy. The effect of the adsorption parameters of the composite material on the removal Drimaren Rot K-7B dye from water has been studied. The Freundlich isotherm describes this process better than the Langmuir isotherm. As the results of the adsorption kinetics show, the process fits well with a pseudo-second-order equation and depends both on the boundary layer and on the structure of the adsorbent itself. The thermodynamic parameters have demonstrated that the process is endothermic and physical. When exposed to ultraviolet light, the discoloration of the dye solution accelerates due to the photocatalytic properties of the composite materials. The addition of H 2 O 2 also speeds up further the process, while the reuse of the materials slows it down, gradually changing the kinetic parameters. The reaction has been attributed to first-order kinetic model, when the active centers of the materials and the number of oxidative radicals formed are numerous and to the second-order kinetic model at a lower reaction activity. Moreover, 52% decolorization of the dye solution (50 mg L -1 ) in the dark was achieved from composite material PA 30 (13.3 g L -1 ) in 120 min and 89% under UV light irradiation. The H 2 O 2 addition (0.14 mmol L -1 ) enhanced it up to 98%. In the second and third use of the photocatalyst, the dye removal decreased to 80% and 60%. Composite material PA30 exhibits antibacterial activity against Gram-negative bacteria E. coli , being most effective at eliminating Gram-positive bacteria S. aureus.

Sclerostin, a glycoprotein encoded by the SOST gene, is mainly produced by mature osteocytes and is a critical regulator of bone formation through its inhibitory effect on Wnt signaling. Osteocytes are differentiated osteoblasts that form a vast and highly complex communication network and orchestrate osteogenesis in response to both mechanical and hormonal cues. The three most commonly described pathways of SOST gene regulation are mechanotransduction, Wnt/β-catenin, and steroid signaling. Downregulation of SOST and thereby upregulation of local Wnt signaling is required for the osteogenic response to mechanical loading. This review covers recent findings concerning the identification of SOST , in vitro regulation of SOST gene expression, structural and functional properties of sclerostin, pathophysiology, biological variability, and recent assay developments for measuring circulating sclerostin. The three-dimensional structure of human sclerostin was generated with the AlphaFold Protein Structure Database applying a novel deep learning algorithm based on the amino acid sequence. The functional properties of the 3-loop conformation within the tertiary structure of sclerostin and molecular interaction with low-density lipoprotein receptor-related protein 6 (LRP6) are also reviewed. Second-generation immunoassays for intact/biointact sclerostin have recently been developed, which might overcome some of the reported methodological obstacles. Sclerostin assay standardization would be a long-term objective to overcome some of the problems with assay discrepancies. Besides the use of age- and sex-specific reference intervals for sclerostin, it is also pivotal to use assay-specific reference intervals since available immunoassays vary widely in their methodological characteristics.

A new methodology for modifying textile materials with dendrimers containing nanoparticles was developed. This involved a combination of eosin Y and N -methyldiethanolamine (MDEA) for reducing the copper ions in the dendrimer complex by enabling a photochemical reaction under visible light and ambient conditions. The conversion of copper ions into nanoparticles was monitored using scanning electron microscopy (SEM) and by performing colorimetric, fluorescence, and electron paramagnetic resonance (EPR) studies. Regardless of the concentration of the photoinitiator eosin Y, it discolored completely upon illumination. Three types of cotton fabrics were compared as antimicrobial materials against Bacillus cereus . One of the fabrics was dyed with a first-generation PAMAM dendrimer which had been functionalized with eight 1,8-naphthalimide fluorophores. Another fabric was dyed with a dendrimer-copper complex, and the third was treated by conversion of the complex into copper nanoparticles encapsulated into the dendrimer. An enhancement in the antimicrobial activity of the textiles was achieved at higher dendrimer concentrations, under illumination with visible light. The fabric modified with the copper nanoparticles encapsulated inside the dendrimer exhibited the best antibacterial activity because it had two photosensitizers (PS), as both 1,8-naphthalimide fluorophores and copper nanoparticles were contained in the dendrimer molecules. The presence of oxygen and suitable illumination activated the photosensitizers to generate the reactive oxygen species (singlet oxygen ( 1 O 2 ) and other oxygenated products, e.g., anion radicals, hydroxyl radicals, and hydrogen peroxide) responsible for destroying the bacteria.

Background: In accordance with the meridian theory of Traditional Chinese Medicine (TCM), meridian phenomena are observed along the course of a meridian following acupuncture. Their visible manifestations include alterations in the color (reddening or whitening) of the skin as well as papule and vesicle formation., Objectives: The aim of the present work is to report a series of visible meridian phenomena manifested in human subjects and to correlate them to TCM concepts. A total of 1,200 patients, on whom classical acupuncture (with standard single-use needles without electrostimulation or moxibustion) was applied, were carefully observed and documented., Methods: Visible meridian phenomena were photographed using standard photographical equipment and compared to classical acupuncture channels used by TCM., Results: Ten patients (5 male, 5 female) exhibited visible meridian phenomena. Lines, concurring with the meridians, were observed: white lines in seven cases and red lines in three cases. The duration of the two kinds of phenomena was different. White lines remained visible for a shorter period (10-15 min), whereas red lines were seen for up to one hour after needle removal., Conclusion: These observations indicate that visible meridian phenomena following acupuncture are objective, albeit rare, findings that coincide with the acupuncture channels described in the classical works of TCM. The presence of such phenomena provides a new insight into the concept of meridians and explains the development of the idea in its historical context.

Clinically, temporal lobe epilepsy (TLE) is the most prevalent type of partial epilepsy and often accompanied by various comorbidities. The present study aimed to evaluate the effects of chronic treatment with the antiepileptic drug (AED) lacosamide (LCM) on spontaneous motor seizures (SMS), behavioral comorbidities, oxidative stress, neuroinflammation, and neuronal damage in a model of TLE. Vehicle/LCM treatment (30 mg/kg, p.o.) was administered 3 h after the pilocarpine-induced status epilepticus (SE) and continued for up to 12 weeks in Wistar rats. Our study showed that LCM attenuated the number of SMS and corrected comorbid to epilepsy impaired motor activity, anxiety, memory, and alleviated depressive-like responses measured in the elevated plus maze, object recognition test, radial arm maze test, and sucrose preference test, respectively. This AED suppressed oxidative stress through increased superoxide dismutase activity and glutathione levels, and alleviated catalase activity and lipid peroxidation in the hippocampus. Lacosamide treatment after SE mitigated the increased levels of IL-1β and TNF-α in the hippocampus and exerted strong neuroprotection both in the dorsal and ventral hippocampus, basolateral amygdala, and partially in the piriform cortex. Our results suggest that the antioxidant, anti-inflammatory, and neuroprotective activity of LCM is an important prerequisite for its anticonvulsant and beneficial effects on SE-induced behavioral comorbidities.

Textile materials, as a suitable matrix for different active substances facilitating their gradual release, can have an important role in skin topical or transdermal therapy. Characterized by compositional and structural variety, those materials readily meet the requirements for applications in specific therapies. Aromatherapy, antimicrobial substances and painkillers, hormone therapy, psoriasis treatment, atopic dermatitis, melanoma, etc., are some of the areas where textiles can be used as carriers. There are versatile optional methods for loading the biologically active substances onto textile materials. The oldest ones are by exhaustion, spraying, and a pad-dry-cure method. Another widespread method is the microencapsulation. The modification of textile materials with stimuli-responsive polymers is a perspective route to obtaining new textiles of improved multifunctional properties and intelligent response. In recent years, research has focused on new structures such as dendrimers, polymer micelles, liposomes, polymer nanoparticles, and hydrogels. Numerous functional groups and the ability to encapsulate different substances define dendrimer molecules as promising carriers for drug delivery. Hydrogels are also high molecular hydrophilic structures that can be used to modify textile material. They absorb a large amount of water or biological fluids and can support the delivery of medicines. These characteristics correspond to one of the current trends in the development of materials used in transdermal therapy, namely production of intelligent materials, i.e., such that allow controlled concentration and time delivery of the active substance and simultaneous visualization of the process, which can only be achieved with appropriate and purposeful modification of the textile material.

The alfalfa weevil ( Hypera postica ) is a well-known example of a worldwide-distributed pest with high genetic variation. Based on the mitochondrial genes, the alfalfa weevil clusters into two main mitochondrial lineages. However, there is no clear picture of the global diversity and distribution of these lineages; neither the drivers of its diversification are known. However, it appears likely that historic demographic events including founder effects played a role. In addition, Wolbachia , a widespread intracellular parasite/symbiont, likely played an important role in the evolution of the species. Wolbachia infection so far was only detected in the Western lineage of H. postica with no information on the infecting strain, its frequency, and its consequences on the genetic diversity of the host. We here used a combination of mitochondrial and nuclear sequences of the host and sequence information on Wolbachia to document the distribution of strains and the degree of infection. The Eastern lineage has a higher genetic diversity and is found in the Mediterranean, the Middle East, Eastern Europe, and eastern America, whereas the less diverse Western lineage is found in Central Europe and the western America. Both lineages are infected with the same common strain of Wolbachia belonging to Supergroup B. Based on neutrality tests, selection tests, and the current distribution and diversification of Wolbachia in H. postica , we suggested the Wolbachia infection did not shape genetic diversity of the host. Mast cells (MCs) are important object in experimental acupuncture due to their putative involvement in local reactions to needling. In the rat, they are shown to contain in their granules, among other tissue mediators, serotonin, also called 5-hydroxytryptamine (5-HT). The aim of this study is to examine the normal distribution of 5-HT-containing MCs in soft tissues of Zusanli (ST36) acupuncture point (acupoint) and their morphological changes caused by experimental acupuncture. We observed 5-HT-immunopositive MCs in the tissues and in the vicinity of the needle tract formed after acupuncture. As a result of acupuncture needling, the tissue integrity is disrupted and certain folds are formed in the direction of the needle tract. Connective tissue in the vicinity of the needle tract gets compressed and displaced, together with the 5-HT-immunoreactive MCs seen there. Some of those 5-HT-immunopositive MCs showed signs of degranulation with numerous discharged granules, some of them found at a considerable distance form the cell. Furthermore, 5-HT-immunopositive MCs are unevenly distributed in soft tissues of ST36 acupoint. Larger numbers of 5-HT-containing MCs were visualized in subcutis and dermis, compared to the observed in striated muscles. Placing the acupuncture needle into the rat skin caused a formation of an apparent needle tract, tissue displacement and degranulation of 5-HT-immunopositive MCs. The demonstrated serotonin release by means of MC degranulation might be involved in the local tissue response to acupuncture.

Objectives: To evaluate the prevalence of some virulence genes among 510 clinical Enterococcus spp. isolates and to assess the association of those genes with the species, infection site, and patient group (inpatients/outpatients)., Methods: Adhesins genes (aggregation substances agg and asa1 of Enterococcus faecalis and Enterococcus faecium, respectively), enterococcal surface protein (esp), endocarditis-specific antigen A (efaA), collagen-binding proteins (ace/acm)); invasins (hyaluronidase (hyl) and gelatinase (gelE)); cytotoxines (activation of cytolysin (cylA) in E. faecalis); and modulators of the host immunity and inflammation (enhanced expression pheromone (eep) in E. faecalis) were detected by polymerase chain reaction., Results: The overall prevalence was: esp - 44.3%, agg/asa1 - 38.4%, ace/acm - 64.3%, efaA - 85.9%, eep - 69.4%, gelE - 64.3%, hyl - 25.1%, and cylA - 47.1%. E. faecalis isolates had significantly higher frequency of adhesin genes (esp and agg/asa1) and gelatinase in comparison to E. faecium. Multiple virulence genes in E. faecalis were significantly more prevalent than in E. faecium isolates. Domination of E. faecium with or without only one gene compared to the isolates of E. faecalis were found. Enterococcus spp. isolates obtained from outpatients compared to inpatients isolates had significantly higher frequency of agg/asa1, eep, gelE and cylA. Some adhesins genes (esp, agg/asa1 and efaA) had higher prevalence among the non-invasive Enterococcus spp. isolates compared to those causing invasive bacteremia, while ace/acm revealed higher dissemination in isolates causing invasive infections compared to non-invasive isolates., Conclusion: Most E. faecalis attaches to abiotic surfaces in hospital environment, which correlates with higher prevalence of gene encoding for virulence factors involved in biofilm formation, such as enterococcal surface protein, aggregation substance, and gelatinase. The intestinal tract is an important reservoir for opportunistic enterococcal pathogens and allows them to access infectious sites through different virulence factors, demonstrated in outpatient isolates in this study., (Copyright © 2016 Elsevier Editora Ltda.
