Abstract

Abstract Background To date, only twenty-one cases diagnosed postnatally with mosaic trisomy 12 have been reported. The most frequent phenotypic manifestations are developmental delay, dysmorphic facial features, congenital heart defects, digital alterations, and pigmentary disorders. In the present report, detailed clinical and genetic profiles of three unrelated new patients with mosaic trisomy 12 are described and compared with previously reported cases. Case presentation In the present report, we include the clinical, cytogenetic, and molecular description of three Mexican patients diagnosed postnatally with mosaic trisomy 12. At phenotypic level, the three patients present with developmental delay, dysmorphic facial features, congenital heart defects and skin pigmentary anomalies. Particularly, patient 1 showed unique eye alterations as bilateral distichiasis, triple rows of upper lashes, and digital abnormalities. In patient 2 redundant skin, severe hearing loss, and hypotonia were observed, and patient 3 presented with hypertelorism and telecanthus. Hyperpigmentation with disseminated pigmentary anomalies is a common trait in all of them. The cytogenetic study was carried out under the strict criteria of analysis, screening 50–100 metaphases from three different tissues, showing trisomy 12 mosaicism in at least one of the three different tissues analyzed. With SNParray, the presence of low-level mosaic copy number variants not previously detected by cytogenetics, and uniparental disomy of chromosome 12, was excluded. STR markers allowed to confirm the absence of uniparental disomy as well as to know the parental origin of supernumerary chromosome 12. Conclusions The detailed clinical, cytogenetic, and molecular description of these three new patients, contributes with relevant information to delineate more accurately a group of patients that show a heterogeneous phenotype, although sharing the same chromosomal alteration. The possibility of detecting mosaic trisomy 12 is directly associated with the sensitivity of the methodology applied to reveal the low-level chromosomal mosaicism, as well as with the possibility to perform the analysis in a suitable tissue.

Abstract

Abstract Background Pigmentary mosaicism constitutes a heterogeneous group of skin pigmentation alterations associated with multisystem involvement. The aim of this study was to establish a complete cytogenetic and molecular characterization of PM patients, emphasizing on searching for possible low chromosomal mosaicism and on establishing an accurate genotype-phenotype correlation. Results A total of 73 patients were included (3 months to 18 years of age), 52% male and 48% female. Observed in 69 (95%) patients, the most frequent pattern of pigmentation was fine and whorled BL, which was associated with disseminated skin extent in 41 (59%) patients. Central nervous system (84%) alterations were the most frequent observed in the group of patients, followed by the musculoskeletal (53%) and ophthalmologic (27%) alterations. Considering the pattern of pigmentation, no significant differences in association with skin extent or extracutaneous manifestations were detected. Following a strict cytogenetic analysis strategy, screening metaphases from three different tissues (peripheral blood, hyperpigmented and hypopigmented skin) we found that 23/73 patients had chromosomal abnormalities classified as follows: 1) Mosaic with 2 or more different cell lines with structural alterations n = 19; 2) Polyploidy (mosaic) n = 1 and 3) Alterations in all cells in three different tissues n = 3. SNP array, array CGH and FISH were useful for the complete characterization of the chromosomal aberrations, for the detection of microdeletions in patients with normal karyotype but with strong clinical suspicious of chromosomal alteration, and for a better establishment of genotype-phenotype correlation. In 2 patients we found genes associated with some of the extracutaneous manifestations (SHH, MNX1, PPP2R2C). Conclusions This group of 73 patients finely described is the largest series of patients with pigmentary mosaicism reported worldwide. As we showed in this study, the followed analysis strategy allowed the detection of cytogenetic and molecular abnormalities, and made possible the establishment of genotype-phenotype associations in some patients. An important limitation of our study was the analysis of fibroblasts cultures instead of melanocytes and keratinocytes. In some cases the direct molecular DNA analysis of skin biopsy could be another choice.

Abstract

Abstract Background Malaria control in Panama is problematic due to the high diversity of morphologically similar Anopheles mosquito species, which makes identification of vectors of human Plasmodium challenging. Strategies by Panamanian health authorities to bring malaria under control targeting Anopheles vectors could be ineffective if they tackle a misidentified species. Methods A rapid mass spectrometry identification procedure was developed to accurately and timely sort out field-collected Neotropical Anopheles mosquitoes into vector and non-vector species. Matrix-assisted laser desorption/ionization (MALDI) mass spectra of highly-abundant proteins were generated from laboratory-reared mosquitoes using different extraction protocols, body parts, and sexes to minimize the amount of material from specimen vouchers needed and optimize the protocol for taxonomic identification. Subsequently, the mass spectra of field-collected Neotropical Anopheles mosquito species were classified using a combination of custom-made unsupervised (i.e., Principal component analysis—PCA) and supervised (i.e., Linear discriminant analysis—LDA) classification algorithms. Results Regardless of the protocol used or the mosquito species and sex, the legs contained the least intra-specific variability with enough well-preserved proteins to differentiate among distinct biological species, consistent with previous literature. After minimizing the amount of material needed from the voucher, one leg was enough to produce reliable spectra between specimens. Further, both PCA and LDA were able to classify up to 12 mosquito species, from different subgenera and seven geographically spread localities across Panama using mass spectra from one leg pair. LDA demonstrated high discriminatory power and consistency, with validation and cross-validation positive identification rates above 93% at the species level. Conclusion The selected sample processing procedure can be used to identify field-collected Anopheles species, including vectors of Plasmodium, in a short period of time, with a minimal amount of tissue and without the need of an expert mosquito taxonomist. This strategy to analyse protein spectra overcomes the drawbacks of working without a reference library to classify unknown samples. Finally, this MALDI approach can aid ongoing malaria eradication efforts in Panama and other countries with large number of mosquito’s species by improving vector surveillance in epidemic-prone sites such as indigenous Comarcas.

4. Transcriptome analysis of the binucleate ciliate Tetrahymena thermophila with asynchronous nuclear cell cycles

Abstract

As a prototypic ciliated protozoan, Tetrahymena thermophila harbors two functionally and physically distinct nuclei within a shared cytoplasm. During vegetative growth, the ‘cell cycles’ of the diploid germline micronucleus and polyploid somatic macronucleus are offset. Micronuclear S phase initiates just before cell division and is completed in daughter cells prior to the onset of macronuclear S phase. Whereas mitotic micronuclear division occurs mid-cell cycle, amitotic macronuclear division immediately precedes cytokinesis. Here we report the first RNA-seq analysis across the cell cycle of a binucleated organism. RNA was isolated at 30 min intervals across 1.5 vegetative cell cycles, starting with a macronuclear G1 population synchronized by centrifugal elutriation. Using MetaCycle, 3244 of the predicted 26,000+T. thermophila genes were shown to be cell cycle regulated. Proteins that are required in micro- and macronuclei exhibit a single mRNA peak that correlates with their macronuclear function, while the expression of nucleus-limited protein-coding genes, including nucleoporins and importins, peak prior to their respective nucleus-specific role. Cyclin D and cyclin A/B genes showed distinct expression patterns that predict nucleus-specific functions. Clustering of periodically expressed genes revealed seven gene expression patterns. Four clusters have known PANTHER GO biological processes that are overrepresented for G1/S and G2/M phase functions. We propose that these clusters encode known and novel factors that coordinate micro- and macronuclear-specific events such as mitosis, amitosis, DNA replication and cell division.

5. Coral-reefs off North and West Africa – Environmental settings

Abstract

As part of the EAF Nansen Program, three cold-water coral sites have been studied along the West African coast: in Mauritania-Morocco and Ghana/Côte d'Ivoire. The aim was to investigate their health status in contrasting environment to obtain knowledge on the physiological ranges and adaptive capacity to multiple stressors. The used data concerns the landscape, depth, water masses, temperature, dissolved oxygen concentration (DO), carbonate chemistry (pH, aragonite saturation WAr), macronutrients and particles (visual). The results are compared to the health status of other Lophelia pertusa reefs in such contrasting environments. The results show that large healthy reefs of L. pertusa reside off Ghana and Mauritania in the oxygen minimum zone (OMZ) with DO concentrations between 1.1 and 1.6 ml L− 1. They are located in corrosive waters with the lowest pH and WAr and high nutrient levels. The reefs off Morocco reside in waters similar to the Norwegian reefs in well oxygenated waters with oversaturated WAr but was showed to be less healthy (dead) compared to the reefs in the other two sites that resided in the OMZ. This study demonstrates that L. pertusa thrives in OMZ and corrosive aragonite waters, suggesting local adaptation to these conditions and supporting recommendations to continue research work on the same species in contrasting environments for potential effects of climate change.

Abstract

Phytoplankton blooms are sporadic events in time and isolated in space. This complex phenomenon is produced by a variety of both natural and anthropogenic causes. Early detection of this phenomenon, as well as the classification of a water body under conditions of bloom or non-bloom, remains an unresolved problem. This research proposes the use of Inherent Optical Properties (IOPs) in optically complex waters to detect the bloom or non-bloom state of the phytoplankton community. An IOP index is calculated from the absorption coefficients of the colored dissolved organic matter (CDOM), the phytoplankton (φ) and the detritus (d), using the wavelength (λ) 443 nm. The effectiveness of this index is tested in five bloom events in different places and with different characteristics from Mexican seas: (1) Dzilam (Caribbean Sea, Atlantic Ocean) a diatom bloom (Rhizosolenia hebetata); (2) Holbox (Caribbean Sea, Atlantic Ocean) a mixed bloom of dinoflagellates (Scrippsiella sp.) and diatoms (Chaetoceros sp.); (3) Campeche Bay in the Gulf of Mexico (Atlantic Ocean) a bloom of dinoflagellates (Karenia brevis); (4) Upper Gulf of California (UGC) (Pacific Ocean) a diatoms bloom (Planktoniella sol) and (5) Todos Santos Bay, Ensenada (Pacific Ocean) a dinoflagellates bloom (Lingulodinium polyedrum). The diversity of sites shows that the IOP index is a suitable method to determine the bloom conditions.

Abstract

The unicellular eukaryote Tetrahymena thermophila has seven mating types. Cells can mate only when they recognize cells of a different mating type as non-self. As a ciliate, Tetrahymena separates its germline and soma into two nuclei. During growth the somatic nucleus is responsible for all gene transcription while the germline nucleus remains silent. During mating, a new somatic nucleus is differentiated from a germline nucleus and mating type is decided by a stochastic process. We report here that the somatic mating type locus contains a pair of genes arranged head-to-head. Each gene encodes a mating type-specific segment and a transmembrane domain that is shared by all mating types. Somatic gene knockouts showed both genes are required for efficient non-self recognition and successful mating, as assessed by pair formation and progeny production. The germline mating type locus consists of a tandem array of incomplete gene pairs representing each potential mating type. During mating, a complete new gene pair is assembled at the somatic mating type locus; the incomplete genes of one gene pair are completed by joining to gene segments at each end of germline array. All other germline gene pairs are deleted in the process. These programmed DNA rearrangements make this a fascinating system of mating type determination.

Coconut Creek (Florida), ISSN 0749-0208.As an effort to mitigate the water scarcity and to achieve a water security status in remote coastal communities with few or no hydraulic systems, or in places with compromised services due to natural disasters, a new semi-portable wave driven desalination device is being developed and tested using a state of the art numerical model. In this early stage of development, the main challenge is to further optimize mechanisms that allow to adjust to tidal and wave variations and to efficiently resist the stresses exerted by cycle motions. The solution is partially handled through the use of a taut-line mooring mechanism which allows the device to work at large range of wave conditions. This prototype consists of two subsystems: a point absorber WEC conformed by a single buoy, and a standard reverse osmosis (RO) system. When interacting with the incoming waves, the mooring line is pulled and pressurizes seawater enough to drive it through several filters including the RO membranes. The FLOW-3D numerical model was used to test the efficiency of the WEC subsystem under several sea states associated with low energy wave conditions. The displacements and motions of the WEC as well as the forces in its anchor line were calculated. The results suggest that it provides the necessary force to pressurize the seawater for the desalination process. Further testing is needed to improve the reliability and survivability of the system which in turn will help to scale the prototype in order to obtain greater quantities of freshwater and thus be competitive with other technologies.

Abstract

In Mexico, as in many other Latin American countries, the lack of a development plan and/or prevention in aquaculture to environmental variability is evident. In the State of Colima, aquaculture presents lags in terms of technical and productive capacity especially for growing tilapia Oreochromis spp. Surveys were performed to farmers and experts, in addition to consulting scientific literature sources, and identifying different risk factors arising from the practice of aquaculture in order to assess the vulnerability of the sector. The main risk factor associated with hydrometeorological phenomena, for which the need to define high-risk areas, even more, given the uncertainty of the same due to global climate change is recognized. This study proposes to implement a comprehensive program of technological conversion and control measures for alien species that also consider aquaculture production schemes based on the efficient use of water, and to consider using this endemic species for cultivation.

Abstract

Background: Results from retrospective studies suggest that use of neuromuscular blocking agents during general anaesthesia might be linked to postoperative pulmonary complications. We therefore aimed to assess whether the use of neuromuscular blocking agents is associated with postoperative pulmonary complications. Methods: We did a multicentre, prospective observational cohort study. Patients were recruited from 211 hospitals in 28 European countries. We included patients (aged ≥18 years) who received general anaesthesia for any in-hospital procedure except cardiac surgery. Patient characteristics, surgical and anaesthetic details, and chart review at discharge were prospectively collected over 2 weeks. Additionally, each patient underwent postoperative physical examination within 3 days of surgery to check for adverse pulmonary events. The study outcome was the incidence of postoperative pulmonary complications from the end of surgery up to postoperative day 28. Logistic regression analyses were adjusted for surgical factors and patients' preoperative physical status, providing adjusted odds ratios (ORadj) and adjusted absolute risk reduction (ARRadj). This study is registered with ClinicalTrials.gov, number NCT01865513. Findings: Between June 16, 2014, and April 29, 2015, data from 22 803 patients were collected. The use of neuromuscular blocking agents was associated with an increased incidence of postoperative pulmonary complications in patients who had undergone general anaesthesia (1658 [7·6%] of 21 694); ORadj 1·86, 95% CI 1·53–2·26; ARRadj −4·4%, 95% CI −5·5 to −3·2). Only 2·3% of high-risk surgical patients and those with adverse respiratory profiles were anaesthetised without neuromuscular blocking agents. The use of neuromuscular monitoring (ORadj 1·31, 95% CI 1·15–1·49; ARRadj −2·6%, 95% CI −3·9 to −1·4) and the administration of reversal agents (1·23, 1·07–1·41; −1·9%, −3·2 to −0·7) were not associated with a decreased risk of postoperative pulmonary complications. Neither the choice of sugammadex instead of neostigmine for reversal (ORadj 1·03, 95% CI 0·85–1·25; ARRadj −0·3%, 95% CI −2·4 to 1·5) nor extubation at a train-of-four ratio of 0·9 or more (1·03, 0·82–1·31; −0·4%, −3·5 to 2·2) was associated with better pulmonary outcomes. Interpretation: We showed that the use of neuromuscular blocking drugs in general anaesthesia is associated with an increased risk of postoperative pulmonary complications. Anaesthetists must balance the potential benefits of neuromuscular blockade against the increased risk of postoperative pulmonary complications. Funding: European Society of Anaesthesiology.

Abstract

Background:COVID-19 outcomes in Mexican patients with rheumatic diseases (RDs) in comparison to general population patients are unknown.Objectives:To compare mortality and hospitalization of COVID-19 patients with RDs and those without.Methods:We included for this study all the Mexican patients with RDs and COVID-19 registered from April 17th to October 30th, 2020 in the COVID-19 Global Rheumatology Alliance registry. We compare clinical and demographic characteristics of patients with RDs and COVID-19 to patients with COVID-19 that were selected randomly from the Mexican Epidemiology database (1:3). A logistic regression analysis was performed to adjust for confusion variables.Results:We included 322 patients with COVID-19 and RDs and 969 controls without RDs. Table 1 shows the demographic characteristics and comorbidities of both groups. Bivariate analysis showed that patients with RDs had higher mortality, were older, and were more frequently hospitalized. Comorbidities, such as diabetes, hypertension, cardiovascular and renal diseases were also more frequent in patients with RDs. In the multivariate analysis, having a RD was no longer associated with mortality (Figure 1).Figure 1.Multivariate analysis of mortalityConclusion:Patients with RDs had higher comorbidities, hospitalizations, and mortality than the general population in the bivariate analysis. However, adjusted multivariate analysis showed that the odds for mortality were not increased because of having a RD. Abstract

Background:As of the 25th of January 2021, more than 150 thousand deaths as consequence of COVID-19 have been reported in Mexico [1]. Advanced age, male gender and comorbidities have been described as risk factors for severe disease and mortality in general population [2]. COVID-19 mortality in Mexican patients with rheumatic and musculoskeletal diseases (RMDs) is unknown.Objectives:To describe characteristics of Mexican patients with RMDs and COVID-19, and to analyse factors associated with mortality.Methods:The Global Rheumatology Alliance COVID-19 (GRA) physician reported registry, is an international effort to collect information on COVID19 in adult patients with RMDs. GRA is an observational registry. The first patient from Mexico was registered on April 17, 2020. All Mexican patients registered in GRA until October 30, 2020 were included in this analysis. The association of mortality with demographic and clinical variables was estimated using logistic regression analysis.Results:A total of 323 patients were registered, with a median age of 52 (IQR 41-61) years old, 166 (51.4%) patients lived in Mexico City. The most frequent RMDs were rheumatoid arthritis, 149 (46.1%) and systemic lupus erythematosus, 24 (19.8%). Over a third of patients with RMDs and COVID-19 (119 (36.8%)) were hospitalized, and 43 (13.3%) died. Table 1 shows clinical and demographic characteristics. In the univariable analysis, the absence of comorbidities was a protective factor, OR 0.3 (95% CI 0.1-0.6). Factors associated with mortality at COVID-19 diagnosis were age over 65 years old, having type 2 diabetes, chronic renal insufficiency, treatment at COVID-19 diagnosis with corticosteroids or with CD20 inhibitors. In the multivariable adjusted analysis, these factors remained independently associated with mortality. No associations with other treatments or comorbidities at COVID-19 diagnosis were found.Conclusion:Mexican patients with RMDs and COVID-19 in the GRA physician reported registry had a mortality of 13.3%. Abstract

[EN] The Yucatan Peninsula hosts worldwide-known tourism destinations that concentrate most of the Mexico tourism activity. In this region, tourism has exponentially increased over the last years, including wildlife oriented tourism. Rapid tourism development, involving the consequent construction of hotels and tourist commodities, is associated with domestic sewage discharges from septic tanks. In this karstic environment, submarine groundwater discharges are very important and highly vulnerable to anthropogenic pollution. Nutrient loadings are linked to harmful algal blooms, which are an issue of concern to local and federal authorities due to their recurrence and socioeconomic and human health costs. In this study, we used satellite products from MODIS (Moderate Resolution Imaging Spectroradiometer) to calculate and map the satellite Inherent Optical Properties (IOP) Index. We worked with different scenarios considering both holiday and hydrological seasons. Our results showed that the satellite IOP Index allows one to build baseline information in a sustainable mid-term or long-term basis which is key for ecosystem-based management., This research was funded by CONACYT with a doctorate scholarship to Jesús A. Abstract

Phytoplankton blooms are sporadic events in time and isolated in space. This complex phenomenon is produced by a variety of both natural and anthropogenic causes. Early detection of this phenomenon, as well as the classification of a water body under conditions of bloom or non-bloom, remains an unresolved problem. This research proposes the use of Inherent Optical Properties (IOPs) in optically complex waters to detect the bloom or non-bloom state of the phytoplankton community. An IOP index is calculated from the absorption coefficients of the colored dissolved organic matter (CDOM), the phytoplankton (φ) and the detritus (d), using the wavelength (λ) 443 nm. The effectiveness of this index is tested in five bloom events in different places and with different characteristics from Mexican seas: (1) Dzilam (Caribbean Sea, Atlantic Ocean) a diatom bloom (Rhizosolenia hebetata); (2) Holbox (Caribbean Sea, Atlantic Ocean) a mixed bloom of dinoflagellates (Scrippsiella sp.) and diatoms (Chaetoceros sp.); (3) Campeche Bay in the Gulf of Mexico (Atlantic Ocean) a bloom of dinoflagellates (Karenia brevis); (4) Upper Gulf of California (UGC) (Pacific Ocean) a diatoms bloom (Planktoniella sol) and (5) Todos Santos Bay, Ensenada (Pacific Ocean) a dinoflagellates bloom (Lingulodinium polyedrum). The diversity of sites shows that the IOP index is a suitable method to determine the bloom conditions.

Abstract

Summary CKS proteins are evolutionarily conserved cyclin-dependent kinase (CDK) subunits whose functions are incompletely understood. Mammals have two CKS proteins. CKS1 acts as a cofactor to the ubiquitin ligase complex SCFSKP2 to promote degradation of CDK inhibitors, such as p27. Little is known about the role of the closely related CKS2. Using a Cks2−/− knockout mouse model, we show that CKS2 counteracts CKS1 and stabilizes p27. Unopposed CKS1 activity in Cks2−/− cells leads to loss of p27. The resulting unrestricted cyclin A/CDK2 activity is accompanied by shortening of the cell cycle, increased replication fork velocity, and DNA damage. In vivo, Cks2−/− cortical progenitor cells are limited in their capacity to differentiate into mature neurons, a phenotype akin to animals lacking p27. We present a deformation of the Minkowski space as embedded into the conformal space (in the formalism of twistors) based in the quantum versions of the corresponding kinematic groups. We compute explicitly the star product, whose Poisson bracket is quadratic. We show that the star product although defined on the polynomials can be extended differentiably. Finally we compute the Eucliden and Minkowskian real forms of the deformation.

Abstract

Tetrahymena thermophila is a ciliate – a unicellular eukaryote. Remarkably, every cell maintains differentiated germline and somatic genomes: one silent, the other expressed. Moreover, the two genomes undergo diverse processes, some as extreme as life and death, simultaneously in the same cytoplasm . Conserved eukaryotic mechanisms have been modified in ciliates to selectively deal with the two genomes. We describe research in several areas of Tetrahymena biology, including meiosis, amitosis, genetic assortment, selective nuclear pore transport, somatic RNAi-guided heterochromatin formation, DNA excision and programmed nuclear death by autophagy, which has enriched and broadened knowledge of those mechanisms.

Abstract

The unicellular eukaryote Tetrahymena thermophila has seven mating types. Cells can mate only when they recognize cells of a different mating type as non-self. As a ciliate, Tetrahymena separates its germline and soma into two nuclei. During growth the somatic nucleus is responsible for all gene transcription while the germline nucleus remains silent. During mating, a new somatic nucleus is differentiated from a germline nucleus and mating type is decided by a stochastic process. We report here that the somatic mating type locus contains a pair of genes arranged head-to-head. Each gene encodes a mating type-specific segment and a transmembrane domain that is shared by all mating types. Somatic gene knockouts showed both genes are required for efficient non-self recognition and successful mating, as assessed by pair formation and progeny production. The germline mating type locus consists of a tandem array of incomplete gene pairs representing each potential mating type. During mating, a complete new gene pair is assembled at the somatic mating type locus; the incomplete genes of one gene pair are completed by joining to gene segments at each end of germline array. All other germline gene pairs are deleted in the process. These programmed DNA rearrangements make this a fascinating system of mating type determination.

Abstract

Ciliated protozoans present several features of chromosome segregation that are unique among eukaryotes, including their maintenance of two nuclei: a germline micronucleus, which undergoes conventional mitosis and meiosis, and a somatic macronucleus that divides by an amitotic process. To study ciliate chromosome segregation, we have identified the centromeric histone gene in the Tetrahymena thermophila genome (CNA1). CNA1p specifically localizes to peripheral centromeres in the micronucleus but is absent in the macronucleus during vegetative growth. During meiotic prophase of the micronucleus, when chromosomes are stretched to twice the length of the cell, CNA1p is found localized in punctate spots throughout the length of the chromosomes. As conjugation proceeds, CNA1p appears initially diffuse, but quickly reverts to discrete dots in those nuclei destined to become micronuclei, whereas it remains diffuse and is gradually lost in developing macronuclei. In progeny of germline CNA1 knockouts, we see no defects in macronuclear division or viability of the progeny cells immediately following the knockout. However, within a few divisions, progeny show abnormal mitotic segregation of their micronucleus, with most cells eventually losing their micronucleus entirely. This study reveals a strong dependence of the germline micronucleus on centromeric histones for proper chromosome segregation.

The unicellular eukaryote Tetrahymena thermophila has seven mating types. Cells can mate only when they recognize cells of a different mating type as non-self. As a ciliate, Tetrahymena separates its germline and soma into two nuclei. During growth the somatic nucleus is responsible for all gene transcription while the germline nucleus remains silent. During mating, a new somatic nucleus is differentiated from a germline nucleus and mating type is decided by a stochastic process. We report here that the somatic mating type locus contains a pair of genes arranged head-to-head. Each gene encodes a mating type-specific segment and a transmembrane domain that is shared by all mating types. Somatic gene knockouts showed both genes are required for efficient non-self recognition and successful mating, as assessed by pair formation and progeny production. The germline mating type locus consists of a tandem array of incomplete gene pairs representing each potential mating type. During mating, a complete new gene pair is assembled at the somatic mating type locus; the incomplete genes of one gene pair are completed by joining to gene segments at each end of germline array. All other germline gene pairs are deleted in the process. These programmed DNA rearrangements make this a fascinating system of mating type determination.

Abstract

The macronucleus of the binucleate ciliate Tetrahymena thermophila contains fragmented and amplified chromosomes that do not have centromeres, eliminating the possibility of mitotic nuclear division. Instead, the macronucleus divides by amitosis with random segregation of these chromosomes without detectable chromatin condensation. This amitotic division provides a special opportunity for studying the roles of mitotic proteins in segregating acentric chromatin. The Smc4 protein is a core component of the condensin complex that plays a role in chromatin condensation and has also been associated with nucleolar segregation, DNA repair, and maintenance of the chromatin scaffold. Mutants of Tetrahymena SMC4 have remarkable characteristics during amitosis. They do not form microtubules inside the macronucleus as normal cells do, and there is little or no bulk DNA segregation during cell division. Nevertheless, segregation of nucleoli to daughter cells still occurs, indicating the independence of this process and bulk DNA segregation in ciliate amitosis.

Abstract

Introduccion La enfermedad de Kawasaki es la causa mas frecuente de cardiopatia adquirida en el mundo desarrollado. La presentacion atipica es mas frecuente en edades extremas, en las cuales es mas elevado el riesgo de inicio como fallo cardiaco. Revisamos los casos de shock cardiogenico secundarios a enfermedad de Kawasaki que ingresaron en nuestra unidad en los ultimos 2 anos. Caso 1 Nina de 8 anos que ingresa en UCIP por mal estado general y signos de bajo gasto cardiaco en el contexto de un sindrome febril de 4 dias de evolucion. En la exploracion hiperemia faringea, exantema urticariforme en raiz de miembros, y eritema palmoplantar y edema de dorso de manos. Se realiza ecocardiografia donde se evidencia dilatacion del ventriculo izquierdo con fallo de contractilidad, con anatomia coronaria normal. En hemograma: leucocitosis con neutrofilia y PCR de 200 mg/l. Se inicia soporte inotropico. En el segundo y tercer dia de estancia en UCI aparecen adenopatias laterocerivales, inyeccion conjuntival, lengua aframbuesada y labio fisurado. Se inicia tratamiento con gammaglobulina a 2 g/kg y aspirina a dosis antiinflamatorias ante la sospecha clinica de enfermedad de Kawasaki. La evolucion es favorable. Caso 2 Nina de 12 anos, que ingresa por signos de bajo gasto cardiaco: obnubilacion, hipotension y oliguria, precedido de un sindrome febril de 6 dias de evolucion. Precisa soporte inotropico y resucitacion volumetrica agresiva para su estabilidad hemodinamica. Presenta leucocitosis con desviacion a la izquierda, PCR de 320 mg/l. En ecocardiografia se observa disminucion de la fraccion de eyeccion y de la contractilidad. Se instaura tratamiento antibiotico empirico, persistiendo la fiebre. Cultivos y serologia recogidas negativas. Tras 4 dias de ingreso en UCIP recupera la funcion miocardica y permite la disminucion de farmacos inovasodilatadores. Durante su estancia desarrollo en tiempos distintos exantema urticariforme cambiante, edemas de extremidades, labios fisurados y enrojecidos e inyeccion conjuntival no purulenta, diagnosticandose de enfermedad de Kawasaki. Tras la administracion de gammaglobulinas a 2 g/kg cede la fiebre y se normalizan los reactantes de fase aguda. La evolucion es favorable, con ecocardiografia posterior normal. Conclusiones 1 . La miocarditis en la enfermedad de Kawasaki es frecuente, aunque excepcionalmente cursa con shock cardiogenico. 2 . El diagnostico precoz permite la instauracion del tratamiento adecuado: gammaglobulinas y aspirina, asi como soporte inotropico, favoreciendo la recuperacion y previniendo las complicaciones coronarias.

Abstract

Introduccion El objetivo del trabajo es analizar los casos de pacientes con quemaduras graves, que precisaron ser ingresados en la unidad de cuidados intensivos pediatricos (UCIP) de nuestro hospital durante los ultimos 7 anos. Material y metodos Estudio retrospectivo y descriptivo. Se analizan los datos recogidos en las historias clinicas de los pacientes estudiados. Resultados Durante el periodo de estudio ingresaron en UCIP, 17 pacientes con quemaduras graves. De ellos, 10 tenian entre 1 y 3 anos. Al ser nuestro centro referencia en esta patologia, procedian de otras provincias andaluzas, 9 pacientes y de Canarias, 4. Fueron quemaduras por escaldadura, 8 casos; por incendio, 6 casos y por llama, 3 casos. El 60 % de los pacientes presentaron intoxicacion por monoxido de carbono. La extension de la quemadura fue entre el 10-30% en el 45 % de los pacientes y mayor del 30 % en el 33 %. En cuanto a la profundidad, todos presentaron quemaduras de segundo grado y 10 casos, quemaduras de tercer grado. Las localizaciones mas frecuentes fueron en cabeza y tronco superior. Precisaron intubacion y ventilacion mecanica convencional 13 casos. La alimentacion enteral se ha iniciado entre el segundo y tercer dia. En los 13 pacientes que necesitaron nutricion parenteral se inicio al segundo dia de estancia. El tratamiento quirurgico mas empleado fue la escarectomia con injerto. Este se inicio entre los dias 2.° y 51.° (mediana de 11 dias). Dos casos precisaron cirugia en las primeras 24 h. Entre las medidas terapeuticas mas usadas destacan: inotropicos (10), toxoide tetanico (9), hemoderivado (9), corticoides intravenosos (9), anticoagulacion (9), oxido nitrico (5) y surfactante pulmonar (5). Las complicaciones mas frecuentes fueron: distres respiratorio agudo (7), neumonia (7), derrame pleural (5), neumopatia intersticial (3), atelectasia (3), ulcera corneal (2) y edema de glotis (2). La estancia media en UCIP oscilo de 2 a 45 dias. Un caso reingreso desde la unidad de quemados. Fallecieron 2 pacientes. Conclusiones La edad de mayor riesgo es en los menores de 3 anos y la etiologia mas frecuente es la escaldadura. En nuestra experiencia el tiempo medio de escarectomia es de 11 dias y las complicaciones mas frecuentes son el distres respiratorio agudo y la neumonia. El paciente quemado grave presenta una elevada morbimortalidad, por tanto son fundamentales las medidas de prevencion. Una vez producida la quemadura grave es basico la resucitacion con liquidos, la nutricion precoz y evitar la infeccion.

Abstract

Introduccion La aparicion de un edema agudo de pulmon (EAP) secundario a la obstruccion de via aerea superior (VAS) esun hecho constatado en la bibliografia desde la primera publicacion por Oswalt en 1977. Los mecanismos implicados en su patogenia son multiples, aunque el factor principal es la generacion de una fuerte presion negativa intratoracica por maniobras de inspiracion forzada contra la obstruccion de la VAS. El laringoespasmo es la causa mas frecuentemente descrita de EAP despues de una anestesia general y su incidencia es mayor en la poblacion pediatrica, en el postoperatorio de intervenciones de la via aerea superior y en deportistas. Existen casos sin embargo de EAP en el postoperatorio inmediato de ninos previamente sanos y tras una intervencion banal, que no puede atribuirse directamente a esta circunstancia, y cuya etiologia permanece aun incierta (farmacos inductores de anestesia, origen neurogeno, sobrecarga de liquidos o cambios de presion intratoracica). El diagnostico diferencial se plantea principalmente con fallo ventricular izquierdo o con una broncoaspiracion. Es esencial para el manejo de estos pacientes pensar en esta posibilidad que responde adecuadamente a la administracion de oxigeno, diureticos y restriccion de liquidos, y habitualmente evoluciona hacia la curacion en unas horas. Casos clinicos Se presentan 2 casos de ninos previamente sanos de 10 y 7 anos, respectivamente, que ingresaron en nuestra unidad procedentes de quirofano por un cuadro de hipoxemia y dificultad respiratoria. En el primero tras una adenoidectomia y en el segundo tras una intervencion de fractura supracondilea. Ninguno de ellos presento incidencias durante la intervencion. En la radiografia de torax al ingreso se evidenciaron imagenes de infiltrado alveolo-intersticial difuso, con un indice cardiotoracico normal. En ninguno de los 2 casos existio laringoespasmo tras la extubacion y en ambos se descarto enfermedad cardiaca y sobrecarga de liquidos. Tras la administracion de furosemida y oxigeno, evolucionaron hacia la normalidad clinica y radiologica en unas horas. Conclusiones 1. El EAP por presion negativa puede ocurrir hasta en el 20 % de los casos de intervenciones de via aerea superior en la infancia. Los ninos sometidos a este tipo de intervenciones deben tener una vigilancia estrecha al despertar de la anestesia general especialmente si han presentado un episodio de laringoespasmo. 2. El pronostico del EAP postobstructivo es excelente si se detecta y se trata precozmente.

Abstract

We studied 13 patients with acute attacks of asthma to test the hypothesis that magnitude of dyspnea at rest correlates well with spirometry and with breath-holding time. Dyspnea ("falta de aire" in Spanish) was quantitated with a linear numerical scale from 0 to 10. We measured breath-holding time, breathing frequency (f), and FEV1 and FVC both expressed as percent of normal. Measurements were done on the patient's arrival and were repeated 3 to 6 times until dyspnea at rest disappeared or was minimal. Dyspnea magnitude, f, and breath-holding time correlated well with FEV1% and FVC% (r between 0.65 and 0.78), and better with changes in FEV1% and FVC% with respect to initial values (delta FEV1 y delta FVC; r between 0.80 and 0.89). Breath-holding time and f changed in proportion to magnitude of dyspnea (r = -0.85 and 0.87 respectively). Regression equations were: dyspnea = 6.34 -0.16 (delta FEV1) r = 0.80, and dyspnea = 7.82-0.105 (FEV1%) r = 0.62. Using multiple regression we improved prediction of FEV1% with easily obtained variables (R = 0.76). These results suggest that: 1) magnitude of dysnea, f, and breath-holding time correlate with severity of airflow obstruction in acute asthma attacks associated with dyspnea at rest; and 2) breath-holding time varies inversely with dyspnea magnitude when it is present at rest.

An anthropometric study was performed on 40 Caucasian children with familial short stature (FSS) and conpared with 40 age-, race-, and sex-matched children with normal height (C-NH), and 250 adolescent girls with normal height (AG-NH). Anthropometric measurements were also obtained on 30 short parents of the FSS (P-SS) and compared with 26 normal height parents of the FSS (P-NH) and 33 normal height adults (Ad-NH). Brachymetacarpy V, rhizomelia, and short upper and lower limbs were highly prevalent among the FSS patients as compared to the control groups. Abstract

Allergic asthma is characterized by increased type 2 inflammation, including eosinophils. Subjects with allergic asthma have recurrent symptoms due to their constant exposure to environmental allergens, such as house dust mite (HDM), which can be further exacerbated by respiratory infections like rhinovirus. The immunoproteasome (IP) is a proteolytic machinery that is induced by inflammatory mediators during virus infection, but the role of the IP in airway allergic inflammation during rhinovirus infection remains unknown. Wild-type (WT) and IP knockout (KO) mice were challenged with HDM. At 48 h after the last HDM challenge, mice were infected with rhinovirus 1B (RV-A1B) for 24 h. After HDM and RV-A1B treatment, IP KO (vs. WT) mice had significantly more lung eosinophils and neutrophils, as well as a significantly higher viral load, but less IFN-beta expression, compared to WT mice. A TLR3 agonist polyinosinic-polycytidylic acid (Poly I:C) treatment after RV-A1B infection in HDM-challenged IP KO mice significantly increased IFN-beta expression and reduced viral load, with a minimal effect on the number of inflammatory cells. Our data suggest that immunoproteasome is an important mechanism functioning to prevent excessive inflammation and viral infection in allergen-exposed mice, and that Poly I:C could be therapeutically effective in enhancing the antiviral response and lessening the viral burden in lungs with IP deficiency., Competing Interests: The authors declare no conflicts of interest.

Background: Influenza A virus (IAV) infection is a significant risk factor for respiratory diseases, but the host defense mechanisms against IAV remain to be defined. Immune regulators such as surfactant protein A (SP-A) and Toll-interacting protein (Tollip) have been shown to be involved in IAV infection, but whether SP-A and Tollip cooperate in more effective host defense against IAV infection has not been investigated., Methods: Wild-type (WT), Tollip knockout (KO), SP-A KO, and Tollip/SP-A double KO (dKO) mice were infected with IAV for four days. Lung macrophages were isolated for bulk RNA sequencing. Precision-cut lung slices (PCLS) from WT and dKO mice were pre-treated with SP-A and then infected with IAV for 48 h., Results: Viral load was significantly increased in bronchoalveolar lavage (BAL) fluid of dKO mice compared to all other strains of mice. dKO mice had significantly less recruitment of neutrophils into the lung compared to Tollip KO mice. SP-A treatment of PCLS enhanced expression of TNF and reduced viral load in dKO mouse lung tissue. Abstract

Gut microbiota exert functions of high importance in the intestine. Furthermore, there is increasing evidence for its role in immune regulation and maintenance of homeostasis in many physiological processes taking place in distant tissues. In particular, in this review, we explore the impact of metabolites produced by the gut microbiota on the development of atopic dermatitis (AD). Probiotics and prebiotics balance the microbiota and promote the generation of bacterial metabolites, such as short-chain fatty acids and tryptophan derivates, which promote the regulation of the exacerbated AD immune response through regulatory T cells and IL-10 and TGF-β cytokines. Metabolites also have a direct action on keratinocytes once they reach the bloodstream. Besides, probiotics decrease the levels of metabolites associated with AD onset, such as phenols. Understanding all these crosstalk processes between the gut and the skin reveals a number of possibilities, mainly through the manipulation of the gut microbiome, which may represent therapeutic strategies that can contribute to the standard treatments of AD patients to improve their quality of life.

Abstract

Vaping is an increasing health threat in the US and worldwide. The damaging impact of vaping on the human distal lung has been highlighted by the recent epidemic of electronic cigarette or vaping use-associated lung injury (EVALI). The pathogenesis of EVALI remains incompletely understood, due to a paucity of models that recapitulate the structural and functional complexity of the human distal lung and the still poorly defined culprit exposures to vaping products and respiratory viral infections. Our aim was to establish the feasibility of using single cell RNA-sequencing (scRNA-seq) technology in human precision-cut lung slices (PCLS) as a more physiologically relevant model to better understand how vaping regulates the antiviral and pro-inflammatory response to influenza A virus infection. Normal healthy donor PCLS were treated with vaping extract and influenza A viruses for scRNA-seq analysis. Vaping extract augmented host antiviral and pro-inflammatory responses in structural cells such as lung epithelial cells and fibroblasts, as well as in immune cells such as macrophages and monocytes. Our findings suggest that human distal lung slice model is useful to study the heterogeneous responses of immune and structural cells under EVALI conditions, such as vaping and respiratory viral infection.

46. Desert particulate matter from Afghanistan increases airway obstruction in human distal lungs exposed to type 2 cytokine IL-13.

Abstract

Introduction: Deployment related asthma-like symptoms including distal airway obstruction have been described in U.S. military personnel who served in Iraq and Afghanistan. The mechanisms responsible for the development of distal airway obstruction in deployers exposed to desert particulate matter (PM) is not well understood. We sought to determine if respiratory exposure to PM from Afghanistan (PMa) increases human distal airway hyperresponsiveness (AHR) with or without exposures to IL-13, a type 2 cytokine. We further tested whether mitochondrial dysfunction, such as ATP signaling and oxidative stress, may contribute to PMa- mediated AHR., Methods: Precision-cut lung slices from donors without a history of lung disease, tobacco smoking, or vaping were pre-treated with IL-13 for 24 h. This was followed by exposure to PMa or PM from California (PMc, control for PMa) for up to 72 h. The role of hydrogen peroxide and ATP in AHR was assessed using the antioxidant enzyme catalase or an ATP receptor P2Y13 antagonist MRS2211. AHR in response to methacholine challenges as well as cytokine IL-8 production were measured., Results: PMa alone, but not PMc alone, trended to increase AHR. Importantly, the combination of PMa and IL-13 significantly amplified AHR compared to control or PMc+IL-13. PMa alone and in combination with IL-13 increased IL-8 as compared to the control. PMa increased H2O2 and ATP. 47. Glycomacropeptide Protects against Inflammation and Oxidative Stress, and Promotes Wound Healing in an Atopic Dermatitis Model of Human Keratinocytes.

Abstract

Keratinocytes are actively implicated in the physiopathology of atopic dermatitis (AD), a skin allergy condition widely distributed worldwide. Glycomacropeptide (GMP) is a milk-derived bioactive peptide generated during cheese making processes or gastric digestion. It has antiallergic and skin barrier restoring properties when it is orally administered in experimental AD. This study aimed to evaluate the effect of GMP on the inflammatory, oxidative, proliferative, and migratory responses of HaCaT keratinocytes in an in vitro AD model. GMP protected keratinocytes from death and apoptosis in a dose dependent manner. GMP at 6.3 and 25 mg/mL, respectively, reduced nitric oxide by 50% and 83.2% as well as lipid hydroperoxides by 27.5% and 45.18% in activated HaCaT cells. The gene expression of TSLP , IL33 , TARC , MDC , and NGF was significantly downregulated comparably to control by GMP treatment in activated keratinocytes, while that of cGRP was enhanced. Finally, in an AD microenvironment, GMP at 25 mg/mL stimulated HaCaT cell proliferation, while concentrations of 0.01 and 0.1 mg/mL promoted the HaCaT cell migration. Therefore, we demonstrate that GMP has anti-inflammatory and antioxidative properties and stimulates wound closure on an AD model of keratinocytes, which could support its reported bioactivity in vivo.

48. Protective Effect of Glycomacropeptide on the Inflammatory Response of U937 Macrophages.

Abstract

Macrophages play crucial roles in inflammation and oxidative stress associated with noncommunicable diseases, such as cardiovascular diseases, diabetes, and cancer. Glycomacropeptide (GMP) is a bioactive peptide derived from milk κ-casein that contains abundant sialic acid and has shown anti-inflammatory, antioxidative, anti-obesity, and anti-diabetic properties when is orally administered. The aim of this study was to evaluate the effect of GMP on the regulation of the inflammatory response in human macrophages and the participation of sialic acid in this activity. GMP pretreatment decreased by 35%, 35%, and 49% the production of nitrites, interleukin (IL)-1β, and tumor necrosis factor (TNF)-α, respectively, in activated human macrophages U937. The same effect was obtained when cells were pretreated with asialo GMP, and no change on the gene expression of the lectins associated with the recognition of sialic acids, SIGLEC5 , 7, and 9, was induced by GMP on macrophages, which suggests that sialic acid might not be involved in this immunoregulatory effect. Interestingly, GMP increased 8.9- and 3.5-fold the gene expression of the canonical anti-inflammatory protein SOCS3 and the antioxidant enzyme HMOX1 , respectively, in U937 cells. Thus, GMP exerts anti-inflammatory and antioxidative activities on activated macrophages in a sialic acid-independent manner, which might be related to its in vivo reported bioactivity.

Abstract

Objectives: SSc is a devastating autoimmune disease characterized by fibrosis and obliterative vasculopathy affecting the skin and visceral organs. While the processes mediating excessive extracellular matrix deposition and fibroblast proliferation are clear, the exact link between autoimmunity and fibrosis remains elusive. Th17 cells have been proposed as critical drivers of profibrotic inflammation during SSc, but little is known about the immune components supporting their pathogenic role. Our aim was to determine cytokine responses of stimulated monocyte-derived dendritic cells (Mo-DCs) and to determine how they influence T-cell cytokine production in SSc., Material and Methods: Dendritic cells (DCs) activate and shape T cell differentiation by producing polarizing cytokines. Hence, we investigated the cytokine responses of monocyte-derived DCs (Mo-DCs) from patients with limited cutaneous SSc (lcSSc), diffuse cutaneous SSc (dcSSc) and healthy controls (HCs) after stimulation with toll-like receptor (TLR) agonists. Also, using co-culture assays, we analysed T cell subpopulations after contact with autologous TLR-activated Mo-DCs., Results: In general, we observed an increased production of Th17-related cytokines like IL-1β, IL-17F, IL-21 and IL-22 by SSc compared with HC Mo-DCs, with variations between lcSSc vs dcSSc and early- vs late-stage subgroups. Noticeably, we found a significant increment in IL-33 production by Mo-DCs in all SSc cases regardless of their clinical phenotype. Strikingly, T cells displayed Th2, Th17 and dual Th2-Th17 phenotypes after exposure to autologous TLR-stimulated Mo-DCs from SSc patients but not HCs. These changes were pronounced in individuals with early-stage dcSSc and less significant in the late-stage lcSSc subgroup., Conclusions: Our findings suggest that functional alterations of DCs promote immune mechanisms favouring the aberrant T cell polarization and profibrotic inflammation behind clinical SSc heterogeneity., (© The Author(s) 2022. Abstract

Background: Mild hypoxic-ischemic encephalopathy (HIE) is a condition that predisposes to negative outcomes such as neuroanatomical injury, mood disorders, and motor or cognitive disabilities. The neuroinflammation plays an important role in the neurological damage; therefore, reducing it could provide neuroprotection. The leuprolide acetate (LA) has shown to have neuroregenerative and immunomodulator properties in other nervous system injuries., Objective: The aim of this study was to evaluate the immunomodulatory effect of LA in the acute phase of mild HIE and its effects in motor activity and behavior in a subacute phase., Method: Forty-five Wistar rats on postnatal day 7 were divided into Sham, HIE treated with saline solution (HIE-SS), and HIE-LA. The HIE was performed cutting of the right carotid artery followed by 60 min of hypoxia. The expression of the inflammatory cytokines interleukin (IL)-1β, tumor necrosis factor (TNF)-α, interferon (IFN)-γ, and the chemokine CXCL-1 were evaluated 72 h after HIE by RT-qPCR and the motor activity and behavior were evaluated by open field test at postnatal day 33., Results: HIE-SS animals showed increased expression of IL-1β, TNF-α, IFN-γ, and CXCL-1 genes in injured tissue. However, the HIE-LA group exhibited similar expression levels of IL-1β and TNF-α to the Sham group, while IFN-γ and CXCL-1 mRNA expression were attenuated with LA treatment. LA treatment also prevented anxiety-like behavior in the open field test., Conclusion: Treatment with LA partially reverses HIE-induced neuroinflammation and prevents anxiety-like behavior in neonatal rats., (© 2023 The Author(s).
