Abstract

In this study, we investigated the effects of gamma irradiation on the antioxidant activity and metabolite profiles of Euphorbia maculata calli (PC3012). Gamma irradiation at various doses (0, 0.05, 0.5, and 10 kGy) significantly enhanced the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS+) radical scavenging activities of the callus extracts of PC3012 in a dose-dependent manner. High-performance liquid chromatography (HPLC) and ultra-performance liquid chromatography-quadrupole time-of-flight/mass spectrometry (UPLC-Q-TOF/MS) analyses revealed that irradiation increased the lysophospholipid content, although no new antioxidant compounds were formed. Furthermore, a PLS-DA analysis revealed evident metabolic differences between non-irradiated and irradiated samples, which were further verified by statistical validation. These findings suggest that gamma irradiation induces specific biochemical modifications that enhance the bioactive properties of PC3012 calli. This technology exhibits potential for utilization in the natural product and food sectors, particularly in the development of functional foods and nutraceuticals with improved health benefits. Abstract

Nialamide is a non-selective monoamine oxidase inhibitor that was widely used as an antidepressant. However, it has been prohibited for decades in the depressive medicine market due to the adverse hepatotoxic side effects. The re-use of drugs that have been withdrawn from the market represents a promising approach for the development of novel incrementally modified drugs and, in this context, ionizing radiation can serve as a powerful tool for producing new drug candidates. The present study exposed nialamide to γ radiation at 50 kGy to obtain the novel cyclized benzylamide, nialaminosin (compound 2), along with five known compounds, 3-amino-N-benzylpropanamide (compound 3), 3-methoxy-N-benzylpropanamide (compound 4), 3-hydroxy-N-benzylpropanamide (HBPA; compound 5), N-benzylpropanamide (compound 6) and isonicotinamide (compound 7). Among the isolated compounds, HBPA was established to inhibit the lipopolysaccharide-induced overproduction of pro-inflammatory mediators, including nitric oxide (NO) and prostaglandin E2 and cytokines including TNF-α, IL-6 and IL-10, without causing cytotoxicity to both RAW 264.7 and DH82 cells. Furthermore, HBPA was found to reduce the protein expression of inducible NO synthase and cyclooxygenase-2 in macrophages and compared with nialamide, it was established to have more potent radical scavenging activity. The present study therefore suggested the application of HBPA for the improvement of anti-inflammatory properties using ionizing radiation technology on the withdrawn drug nialamide. Abstract

Abstract Background Centipede grass (CG) originates from China and South America and is reported to contain several C-glycosyl flavones and phenolic constituents, including maysin and luteolin derivatives. This study aimed to investigate, for the first time, the antiobesity activity of CG and its potential molecular mechanism in 3T3-L1 cells. Methods To study the effect of CG on adipogenesis, differentiating 3T3-L1 cells were treated every day with CG at various concentrations (0–100 μg/ml) for six days. Oil-red O staining and triglyceride content assay were performed to determine the lipid accumulation in 3T3-L1 cells. The expression of mRNAs or proteins associated with adipogenesis was measured using RT-PCR and Western blotting analysis. We examined the effect of CG on level of phosphorylated Akt in 3T3-L1 cells treated with CG at various concentration s during adipocyte differentiation. Results Differentiation was investigated with an Oil-red O staining assay using CG-treated 3T3-L1 adipocytes. We found that CG suppressed lipid droplet formation and adipocyte differentiation in 3T3-L1 cells in a dose-dependent manner. Treatment of the 3T3-L1 adipocytes with CG resulted in an attenuation of the expression of adipogenesis-related factors and lipid metabolic genes. The expression of C/EBPα and PPARγ, the central transcriptional regulators of adipogenesis, was decreased by the treatment with CG. The expression of genes involved in lipid metabolism, aP2 were significantly inhibited following the CG treatment. Moreover, the CG treatment down-regulated the phosphorylation levels of Akt and GSK3β. Conclusions Taken collectively, these data indicated that CG exerts antiadipogenic activity by inhibiting the expression of C/EBPβ, C/EBPα, and PPARγ and the Akt signaling pathway in 3T3-L1 adipocytes.

Abstract

We investigated relations between physiological damages and gene expression and enzyme activities of superoxide dismutase(SOD) isoenzymes in leaves of rice (Oriza sativa L. cv. Ilpoombyeo) plants irradiated with a high-dose gamma-ray. Gamma-irradiation with 500 Gy caused significant decreases in the contents of protein, chlorophyll and carotenoid in the rice leaves by 24 h, especially reducing the chlorophyll contents up to 26% relative to the control. In contrast, gene expressions of SOD isoenzymes were kept higher in the irradiated leaves until 24 h after the irradiation than in the control and they started to noticeably decrease at 48 h, finally being lower in the irradiated leaves at 72 h than in the control. In the case of enzyme activities of SOD isoenzymes, some CuZn-SOD isoenzymes showed slightly increased activities until 48 h after the irradiation but at 72 h, all isoenzyme activities markedly decreased in the irradiated leaves below the control levels. In conclusion, 500 Gy gamma-irradiation used in the current study caused decreases in the contents of protein, chlorophyll and carotenoid as symptoms for physiological damages. Although such physiological damages were not directly related to the gene expressions and enzyme activities of SOD isoenzymes until 24 h after the irradiation, the damages at 72 h were reasonably attributable to their reduction.

Abstract

Summary: The plant‐specific transcription factor (TF) NAC103 was previously reported to modulate the unfolded protein response in Arabidopsis under endoplasmic reticulum (ER) stress. Alternatively, we report here that NAC103 is involved in downstream signaling of SOG1, a master regulator for expression of DNA damage response (DDR) genes induced by genotoxic stress. Arabidopsis NAC103 expression was strongly induced by genotoxic stress and nac103 mutants displayed substantial inhibition of DDR gene expression after gamma radiation or radiomimetic zeocin treatment. DDR phenotypes, such as true leaf inhibition, root cell death and root growth inhibition, were also suppressed significantly in the nac103 mutants, but to a lesser extent than in the sog1‐1 mutant. By contrast, overexpression of NAC103 increased DDR gene expression without genotoxic stress and substantially rescued the phenotypic changes in the sog1‐1 mutant after zeocin treatment. The putative promoters of some representative DDR genes, RAD51,PARP1,RPA1E,BRCA1 and At4g22960, were found to partly interact with NAC103. Together with the expected interaction of SOG1 with the promoter of NAC103, our study suggests that NAC103 is a putative SOG1‐dependent transcriptional regulator of plant DDR genes, which are responsible for DDR phenotypes under genotoxic stress. Significance Statement: While plants are inevitably subjected to environmental stress factors, genotoxic agents and endogenous metabolic processes that cause DNA damage, there is little information available about downstream signaling of SUPPRESSOR OF GAMMA RESPONSE1 (SOG1), a master regulator in the plant DNA damage response (DDR). Abstract

Using two japonica rice cultivars (Ilpumbyeo and Sanghaehyanghyella), which are distinguishable by the brown planthopper (BHP) resistance maker (R208), a relationship between the BPH resistance gene (Os-Bil) and salt-tolerance was investigated. To do this, changes in the expression level of Os-Bil by the salt stress were quantified by the real-time PCR in the two cultivars, and compared with those in other two indica rice cultivars (Pokkali and IR29). In Ilpumbyeo, the expression level of Os-Bil decreased by the treatments of 50 and 200 mM NaCl in a concentration-dependent manner, and in Sanghaehyanghyella it rather increased slightly at 50 mM but decreased drastically at 200 mM. Comparably, IR29, a salt-sensitive cultivar, showed a reduction of the Os-Bil gene expression after the treatment of 100 mM NaCl, but Pokkali, a salt-tolerance cultivar, rather increased about two times in the level of Os-Bil transcripts. These results suggest that the BPH resistance gene may involve in the difference in the salt-tolerance at least between the two indica rice cultivars.
