4. Study on impact resistance of Nano-Si[O.sub.2] reinforced STG/Kevlar composites

As a flexible protective material, shear thickening gel (STG)/Kevlar composite still has great potential to improve its impact resistance. In this paper, a novel STG/Si[O.sub.2]/Kevlar flexible protective composite was prepared by impregnating STG/Si[O.sub.2] into Kevlar. Due to the excellent Si[O.sub.2] enhancement effect, the STG/Si[O.sub.2]/Kevlar exhibited a higher safeguarding property than the STG/Kevlar. The maximum yam pull-out force of STG/Si[O.sub.2]/Kevlar reached 61.7 N, which was higher than the 41.9 N of STG/Kevlar. The uniform puncture results showed that the maximum bearing capacity of STG/Kevlar and STG/Si[O.sub.2]/Kevlar increased by 116.8% and 215.3%, respectively, compared with pure Kevlar. The impact test results indicated that 20 layers of STG/Si[O.sub.2]/Kevlar could not be penetrated by the knife stab and 24 layers of STG/Si[O.sub.2]/Kevlar could not be penetrated by 7.62 mm pistol bullets. Finally, the enhancement mechanism of impact resistance of STG/Si[O.sub.2]/Kevlar composites was studied. 5. Insights from Structure-Based Simulations into the Persulfidation of Uridine Diphosphate-Glycosyltransferase71c5 Facilitating the Reversible Inactivation of Abscisic Acid.

The action of abscisic acid (ABA) is closely related to its level in plant tissues. Uridine diphosphate-glycosyltransferase71c5 (UGT71C5) was characterized as a major UGT enzyme to catalyze the formation of the ABA-glucose ester (ABA-GE), a reversible inactive form of free ABA in Arabidopsis thaliana (thale cress). UGTs function in a mode where the catalytic base deprotonates an acceptor to allow a nucleophilic attack at the anomeric center of the donor, achieving the transfer of a glucose moiety. The proteomic data revealed that UGT71C5 can be persulfidated. Herein, an experimental method was employed to detect the persulfidation site of UGT71C5, and the computational methods were further used to identify the yet unknown molecular basis of ABA glycosylation as well as the regulatory role of persulfidation in this process. Our results suggest that the linker and the U-shaped loop are regulatory structural elements: the linker is associated with the binding of uridine diphosphate glucose (UPG) and the U-shaped loop is involved in binding both UPG and ABA.It was also found that it is through tuning the dynamics of the U-shaped loop that is accompanied by the movement of tyrosine (Y388) that the persulfidation of cysteine (C311) leads to the catalytic residue histidine (H16) being in place, preparing for the deprotonation of ABA, and then reorientates UPG and deprotonated ABA closer to the 'Michaelis' complex, facilitating the transfer of a glucose moiety. Ultimately, the persulfidation of UGT71C5 is in favor of ABA glycosylation. Our results provide insights into the molecular details of UGT71C5 recognizing substrates and insights concerning persulfidation as a possible mechanism for hydrogen sulfide (H

“English Curriculum Standards for Compulsory Education (2022 Edition)” puts forward new requirements for English teaching in compulsory education, which makes the training of English major normal students challenging. With the wide application of the online and offline blended teaching mode, its role in the teaching of the course “English Pedagogy” is prominent. In the process of constructing the “2+3+2” model (that is, two major sections of theoretical courses and practical courses + three links of pre-class, afterclass teaching + online and offline methods), the subject teaching team of Kashi University solved the problems existing in teaching the course “English Pedagogy”, such as, insufficient class hours, students’ insufficient language ability, insufficient integration of theory and practice, and untimely and unscientific teaching evaluation. And the attempt of constructing this model provides a reference for the application of the blended teaching mode to various courses.

19. Predicting chemosensitivity based on mini patient-derived xenografts in osteosarcoma patients: A retrospective study.

Context: The survival of patients diagnosed with osteosarcoma has not improved in the past three decades because of chemoresistance. Aim: This study aimed to improve the prognosis of patients with osteosarcoma. Settings and Design: From January 1, 2018, to June 30, 2019, a total of 14 patients with osteosarcoma were enrolled who underwent mini patient-derived xenograft (mini-PDX) assay in our hospital. Methods and Materials: We recruited 14 patients with osteosarcoma having acquirable lesions to establish PDX models and examine the sensitivity of nine drugs, including methotrexate (MTX), ifosfamide (IFO), epirubicin, and etoposide. Drug sensitivity was evaluated using the tumor relative proliferation rate (TRPR), and the patients' responses were assessed according to the RECIST 1.1 guidelines. Statistical Analysis Used: The difference in TRPR was analyzed using a paired t-test, while progression-free survival (PFS) was analyzed using the Kaplan–Meier method. Results: The mini-PDX results revealed that IFO had a lower tumor proliferation rate than MTX, indicating that IFO was more sensitive in patients with osteosarcoma (38.3% vs. 84.3%, P = 0.031). Thus, the regimen where IFO alternates with doxorubicin and cisplatin was recommended as adjuvant chemotherapy. MTX could replace IFO if the TRPR was better. Finally, 11 patients received adjuvant chemotherapy. A comparison of PFS revealed that sensitive patients with TRPR of <40% had a better prognosis (9.4 months vs. 3.7 months, P = 0.0324). Conclusions: Chemotherapy based on mini-PDX can improve the survival of patients with osteosarcoma whose TRPR was <40%, and that chemotherapy without MTX could be an alternative for osteosarcoma. Summary Limited by spatial resolution and visual contrast, bone scintigraphy interpretation is susceptible to subjective factors, which considerably affects the accuracy and repeatability of lesion detection and anatomical localization. In this work, we design and implement an end-to-end multi-task deep learning model to perform automatic lesion detection and anatomical localization in whole-body bone scintigraphy. A total of 617 whole-body bone scintigraphy cases including anterior and posterior views were retrospectively analyzed. The proposed semi-supervised model consists of two task flows. The first one, the lesion segmentation flow, received image patches and was trained in a supervised way. The other one, skeleton segmentation flow, was trained on as few as five labeled images in conjunction with the multi-atlas approach, in a semi-supervised way. The two flows joint in their encoder layers so each flow can capture more generalized distribution of the sample space and extract more abstract deep features. The experimental results show that the architecture achieved the highest precision in the finest bone segmentation task in both anterior and posterior images of whole-body scintigraphy. Such an end-to-end approach with very few manual annotation requirement would be suitable for algorithm deployment. Moreover, the proposed approach reliably balances unsupervised labels construction and supervised learning, providing useful insight for weakly labeled image analysis. OBJECTIVES The purpose of this study was to present our clinical experience of treating multifocal osteosarcoma (MFOS) in our center and gain more insight into the biology of this rare condition; in particular, to address with the help of precision genomic medicine the issue of whether the multiple osteosarcoma (OS) lesions in such patients are multi-centric or originate from one primary lesion and metastasize to other sites. Finally, we aimed to identify particular gene phenotypes and mutations that differentiate MFOS from OS with only one tumor. METHODS Clinical data of patients with MFOS treated at our center between June 2007 and October 2014 were collected and analyzed retrospectively. High throughput sequencing of the whole exome of normal tissue and multiple lesions had been performed on samples from two patients (HJF and JZ) diagnosed in 2014. To explore the particular gene phenotype and clinical significance of MFOS, these sequencing results were analyzed and compared with those from patients with osteosarcoma in a single site. Seven patients with MFOS (three male and four female; average age 19.71 ± 3.35 years were enrolled in this study. Two of these patients declined treatment and died after 4 and 6 months, respectively. The remaining patients received standard treatment comprising neoadjuvant chemotherapy, surgery and chemotherapy. The chemotherapy regimen was lobaplatin (45 mg/m(2) ), doxorubicin (60 mg/m(2) ) and ifosfamide (12 g/m(2) ). Patients were followed up every 3 months after completing treatment and evaluated by the Enneking and Response Evaluation Criteria in Solid Tumors scoring systems. RESULTS Up to the last follow-up on 1 December 2015, three patients were still alive. The event-free survival ranged from 4 to 144 weeks (median, 50.14 weeks), the mean (±SD) being 55.45 ± 45.47 weeks. Overall survival ranged from 16 to 388 weeks (median, 89 weeks; mean ± SD, 118.7 ± 147.7 weeks). The rates of mutation of the targeted drug-related genes were 133.5% ± 3.0% in the proximal tibia lesion and 113.1% ± 1.9% in the distal femur of patient HJF (P < 0.01) and 136.1% ± 10.8% in the proximal tibial lesion and 122.3% ± 5.5% in the proximal humerus of patient JZ (P = 0.0335). Furthermore, there were several anti-oncogenes in the somatic copy number variation lists analyzed from the two patients, especially TP53. However, no kataegis was found. CONCLUSIONS Early and radical surgery accompanied by appropriate chemotherapy is the optimal means of treating MFOS. These patients may benefit from precision genomic medicine.

23. Evaluation of bus depot’s environmental impact and recommendations for improvements by material optimisation and improved energy efficiency

The public transportation in Stockholm is expanding and in order to meet the new demand the amount of buses and depots will have to increase within the city. As a result, it is getting more important to evaluate and analyse the performance of bus depots in order to reduce its environmental impact. The aim of this work is to study the production and operational phase during a bus depot’s life cycle and introduce saving measures that can reduce the emission of carbon dioxide equivalents (CO2eq). This study is conducted in collaboration with Skanska and the depot chosen for this study is currently under construction and located in Charlottendal, Värmdö. A base model is created for the whole bus depot area and the environmental impact is evaluated regarding the activities and usage of materials during production and the energy usage during operation of the depot. The evaluation of the model is performed by using the calculation tools IDA ICE, Anavitor, SPIK and Excel, and the environmental impact is expressed in terms of emission of CO2eq during the lifetime of the depot, which is assumed to be 50 years. In order to investigate how bus depots can be built to be more climate neutral and energy efficient, several saving measures are evaluated in four cases. The first two cases are focusing on optimising the usage of materials in the building process, by reducing the material groups with the highest environmental impact and considering green construction solutions. The other two cases are aiming towards enhancing the energy performance of the depot, by reducing the usage of energy according to BBR and deliberating an indoor parking place for the buses. The total emission of CO2eq from the base model is determined to be approximately 16 000 tonnes during the lifetime of the depot. About 42 percent of the environmental impact is instigated during the production phase and the rest of the emission is caused by the use of electricity and heat during operation. By considering the implemented measures it can be concluded that the largest reduction in emission can be obtained by optimising the usage of materials on the site, which is achieved by reducing two of the largest materials groups consisting of concrete and asphalt. By reducing the usage of these materials the total emission from the production phase can be reduced by approximately 9 percent and the total emissions can be reduced by up to 4 percent. To verify the obtained results a sensitivity analysis is performed where three important parameters are investigated. The chosen parameters are; the assumption of the emission factors for the electricity and district heating mixes and the required heating demand for the buses. According to the sensitivity analysis the final results are highly related to the considered parameters. For instance, if the delivered district heating is assumed to be supplied by Fortum, which is the main distributor within Stockholm, it can be concluded that an indoor parking place for the buses is the most beneficial solution to reduce the total emissions. By building a new base hall the emissions instigated from the total heating demand can be reduced by 55 percent and the total emissions can be reduced by 25 percent. På grund av detta blir det mer och mer viktigt att utvärdera och analysera bussdepåernas prestanda för att kunna minska miljöpåverkan från dessa verksamheter. Syftet med detta arbete är att studera produktion- och driftfasen under en bussdepås livscykel samt presentera åtgärder som kan leda till en minskning i utsläppen av koldioxidekvivalenter (CO2ekv). Detta arbete utförs i sammarbete med Skanska och den studerade bussdepån, som ligger i Charlottendal på Värmdö, är för tillfället under konstruktion. För att kunna utvärdera den valda depån skapas en basmodell där klimatpåverkan utvärderas utifrån de aktiviteter och material som används i produktionsfasen och den energi som används under driften av depån. De beräkningsverktyg som har används i utvärderingen av depån består av IDA ICE, Anavitor, SPIK och Excel, och klimatpåverkan från depån uttrycks i ton CO2ekv under dess livslängd, vilken har antagits till 50 år. För att undersöka hur depåer kan konstrueras för att vara mer klimatneutrala och energieffektiva så utvärderas olika besparingsåtgärder i fyra fall. Fokus för de två första fallen ligger på att optimera materialanvändningen i byggprocessen, vilket inkluderar en minskning av de material som genererar i de största utsläppen samt en analys av mer miljövänliga konstruktionslösningar. De andra två fallen riktas istället mot att förbättra energiprestandan av depån genom att minska energianvändningen enligt BBR och överväga en inomhusparkering för bussarna. De totala utsläppen från basmodellen utgör cirka 16 000 ton, där 42 procent orsakas under produktionsfasen och de resterande utsläppen kommer från driften av depån. Utifrån analysen kan man dra slutsatsen att den största minskningen av utsläpp kan åstadkommas genom att dra ner på mängden betong och asfalt i produktionsfasen. Genom att minska på dessa material kan utsläppen från produktionsfasen minskas med ungefär 9 procent och de totala utsläppen kan minskas med 4 procent. I känslighetsanalysen undersöks tre huvudsakliga parametrar som har en stor inverkan på det slutliga resultatet av beräkningarna. Dessa parametrar består av de valda energimixerna för elektricitet och fjärrvärme samt värmebehovet för bussarna. Enligt resultatet i känslighetsanalysen så är en inomhusparkering den förbättringsåtgärd som leder till den största minskningen av utsläpp. Detta resultat fås då fjärrvärmen antas levereras av Fortum, som är den största leverantören i Stockholmsområdet. Genom att investera i en ny busshall kan det totala värmebehovet för bussdepån minskas med 55 procent och det totala utsläppet med 25 procent.

24. Estimation of Carbon Dioxide Emissions from the Residential Building Sector in the county of Stockholm

During the last decades the housing sector has increased continuously, and housings and services accounted for 40 % of the energy usage in Sweden during 2011. The expansion in number of buildings in society has resulted in an increase in both energy usage and emissions of greenhouse gases. It is crucial to enable a sustainable development of society and as a result, the demand to reduce the emissions of carbon dioxide is a current question. In this study, the carbon dioxide emitted from the housing sector in Stockholm is estimated. The housing sector can be divided into different types of houses such as apartment blocks, single-family houses and holiday houses. By collecting information of the energy purchased in respective households, a computational model is generated that calculates the total emission of carbon dioxide. The computational model is created in Excel and based on various assumptions and boundaries regarding the production of electricity and district heating. The environmental impact of the energy purchased is studied through a lifecycle perspective and the energy statistics is studied through an average perspective. The model is designed to calculate the emission factor from the electricity production through a Swedish, Nordic and Northern European perspective. The model also generates a value of the emission factor for the district heating system in Stockholm. By combining the emission factors and the energy statistics, a value of the total emission of carbon dioxide from the housing sector is obtained. A case study is implemented by examining the energy usage for the year of 2010. The emission of carbon dioxide during the year was approximately 1.3 million tones. By taking the import of electricity into account in the Swedish perspective, the result was obtained. When the Swedish electricity mix was replaced with the European mix, the amount of carbon dioxide increased approximately 3 times. As a result of the cold climate and low production due to recession during 2010, limited flow in water reservoirs and low production in nuclear power plants the emissions was approximately 30 000 tonnes higher compared to the year before. By assuming a Swedish electricity mix, the case study shows that the emission per square meter is the highest in apartment blocks, while the emissions from the single-family houses and holiday houses are relatively low in comparison. According to the sensitivity analysis the emission is essentially affected by the import of electricity. The efficiency in power plants also affects the result of the electricity mix in the Nordic and Northern European perspective. However, the selection of electricity mix in the production of district heating, biomass fuel in single-family houses and efficiency in boilers and burners are practically insignificant in the final result. Modellen konstrueras för beräkning av emissionsfaktorer för elmix ur ett svenskt, nordiskt och nordeuropeisk elsystem samt för fjärrvärmemixen i Stockholms län. Dessa emissionsfaktorer, inklusive inmatningsvärden för den använda energin, används för att beräkna det totala koldioxidutsläppet från bostadssektorn. En fallstudie genomförs för att beräkna koldioxidutsläppet från bostadssektorn år 2010 med hjälp av modellen. Resultatet visar att koldioxidutsläppet för detta år var ungefär 1,3 miljoner ton vid beräkning med svenskt medelel med hänsyn till nettoimport, och om beräkning utförs med europeisk elmix blir utsläppet tre gånger större. Koldioxidutsläppet år 2010 är nästan 30 tusen ton mer än föregående år och detta bland annat på grund av det kalla klimatet i kombination med dålig tillrinning i vattenmagasinen, låg kärnkraftproduktion och lågkonjunktur. Resultatet ur fallstudien visar även att koldioxidutsläppet per kvadratmeter är störst i flerbostadshus, medan utsläppet från småhus och fritidshus ligger på en förhållandevis låg nivå i jämförelse, då elen beräknas ur ett svensk elsystem. Känslighetsanalysen av fallstudien visar att importen av el har en stor inverkan på resultatet. Även verkningsgrad i kraftverken är en parameter som påverkar resultatet för nordisk och nordeuropeisk elmix. Däremot är valet av elmix i fjärrvärmeproduktionen, biobränslemix i småhusen samt verkningsgraden i pannor och brännare i de enskilda hushållen nästan obetydligt för slutresultatet.

Summary: Limited by spatial resolution and visual contrast, bone scintigraphy interpretation is susceptible to subjective factors, which considerably affects the accuracy and repeatability of lesion detection and anatomical localization. In this work, we design and implement an end-to-end multi-task deep learning model to perform automatic lesion detection and anatomical localization in whole-body bone scintigraphy. A total of 617 whole-body bone scintigraphy cases including anterior and posterior views were retrospectively analyzed. The proposed semi-supervised model consists of two task flows. The first one, the lesion segmentation flow, received image patches and was trained in a supervised way. The other one, skeleton segmentation flow, was trained on as few as five labeled images in conjunction with the multi-atlas approach, in a semi-supervised way. The two flows joint in their encoder layers so each flow can capture more generalized distribution of the sample space and extract more abstract deep features. The experimental results show that the architecture achieved the highest precision in the finest bone segmentation task in both anterior and posterior images of whole-body scintigraphy. Such an end-to-end approach with very few manual annotation requirement would be suitable for algorithm deployment. - Abstract
Background: Osteosarcoma (OS) is the most common primary malignant bone tumor in children and adolescents. OS is associated with locally aggressive growth and high metastatic potential. The mechanisms that underlie these processes are currently elusive. Reactive oxygen species (ROS) and Na
+ /H+ exchanger 1 (NHE1) have been suggested to regulate proliferation and migration of tumor cells. However, the relationship between NHE1 and ROS in OS proliferation and migration has not been investigated before., Aim: To investigate the role of NHE1 and ROS in the proliferation and migration of OS., Methods: ROS levels and NHE1 expression were studied in cultured human OS cells and human OS xenografts in nude mice. In vitro, OS cells were treated with different doses of tert-butyl hydroperoxide (tBHP), a ROS inducer, and cariporide, an NHE1 inhibitor, to study the effect on cell proliferation and migration. In vivo, nude mice bearing OS cells were administrated with NHE1 inhibitor or antioxidant and the tumor weights were measured., Results: This study reported for the first time that the expression of NHE1 and intracellular ROS level were both increased in OS tissues and cells. Exposure of OS cell to ROS derived from tBHP was able to accelerate cell proliferation and migration and also up-regulate NHE1 protein expression. Moreover, tBHP significantly increased intracellular pH (pHi), decreased extracellular pH (pHe) and induced upregulation of ERK, MMP2, and MMP9. Lowering of ROS levels with the anti-oxidant DMTU or inhibiting NHE1 activity via cariporide abolished the stimulatory effect of tBHP. However, there cariporide did not affect intracellular ROS levels. In vivo study we further confirmed that cariporide could inhibit tumor growth in the nude mouse xenografts of OS cells., Conclusions: The data demonstrate that up-regulation of NHE1 was induced by low concentrations of ROS contributes to the regulation of tumor proliferation and invasion of OS., Relevance for Patients: There is potential application for cariporide as an effective antitumor agent during the development of human osteosarcoma.
