Abstract

Developing catalysts with superior activity to hydrogen evolution reaction (HER) and oxygen evolution reaction (OER) is equally important to the overall photoelectrochemical water splitting to produce hydrogen. In this work, bismuth oxyiodide (BiOI), iron-modified bismuth iodide Fe/BiOI, and the sulfurized S-Fe/BiOI were prepared using the solvothermal method. The three materials all have good absorption ability for visible light. The photoelectrochemical catalytic activity of BiOI to oxygen evolution reaction (OER) is significantly enhanced after iron modification, while the sulfurized product S-Fe/BiOI exhibits better catalytic activity to hydrogen evolution reaction (HER). Hence, OER and HER can be simultaneously catalyzed by using Fe/BiOI and S-Fe/BiOI as anodic and cathodic catalysts to facilitate the overall photoelectrochemical water splitting process. 27. Recyclable green hydrogel adsorbents with excellent adsorption capacity for removal of methylene blue

Abstract

The recyclable green carboxymethyl chitosan-oxidized sodium alginate-gelatin/polyacrylamide double network hydrogel was simply and rapidly synthesized by one-pot method. This hydrogel with satisfactory mechanical properties was used to adsorb methylene blue, showing a high adsorption capacity, recyclability and environmental friendliness. The results showed that the adsorption of methylene blue solution (50 mL of 50 mg·L–1) with 15 mg of adsorbent resulted in a removal rate of over 93%, and the adsorption rate was still over 86% after 7 adsorption–desorption cycles. The adsorption of methylene blue by hydrogel adsorbents was consistent with the Langmuir adsorption isotherm model and pseudo-second-order kinetic adsorption process, mainly through electrostatic and hydrogen bonding interactions. In this work, the prepared hydrogels have a high adsorption capacity and stable cycle adsorption–desorption, which are beneficial to the application in the purification of dye wastewater.

Abstract

Abstract Background Stress, anxiety and depression can cause complex physiological and neuroendocrine changes, resulting in increased level of stress related hormone catecholamine, which may constitute a primary mechanism by which physiological factors impact gene expression in tumors. In the present study, we investigated the effects of catecholamine stimulation on MMP-7 expression in gastric cancer cells and elucidated the molecular mechanisms of the up-regulation of MMP-7 level by catecholamine through an adrenergic signaling pathway. Results Increased MMP-7 expression was identified at both mRNA and protein levels in the gastric cancer cells in response to isoproterenol stimulation. β2-AR antigonist effectively abrogated isoproterenol-induced MMP-7 expression. The activation of STAT3 and AP-1 was prominently induced by isoproterenol stimulation and AP-1 displayed a greater efficacy than STAT3 in isoproterenol-induced MMP-7 expression. Mutagenesis of three STAT3 binding sites in MMP-7 promoter failed to repress the transactivation of MMP-7 promoter and silencing STAT3 expression was not effective in preventing isoproterenol-induced MMP-7 expression. However, isoproterenol-induced MMP-7 promoter activities were completely disappeared when the AP-1 site was mutated. STAT3 and c-Jun could physically interact and bind to the AP-1 site, implicating that the interplay of both transcriptional factors on the AP-1 site is responsible for isoproterenol-stimulated MMP-7 expression in gastric cancer cells. The expression of MMP-7 in gastric cancer tissues was found to be at the site where β2-AR was overexpressed and the levels of MMP-7 and β2-AR were the highest in the metastatic locus of gastric cancer. Conclusions Up-regulation of MMP-7 expression through β2-AR-mediated signaling pathway is involved in invasion and metastasis of gastric cancer.

Abstract

Aberrant Ras/Raf/MAPK and PI3K/AKT/mTOR signaling pathways are found in hepatocellular carcinoma (HCC). This study reports how sorafenib (a multi-kinase inhibitor) and PI-103 (a dual PI3K/mTOR inhibitor) alone and in combination inhibit the proliferation of the HCC cell line, Huh7.Huh7 proliferation was assayed by 3H-thymidine incorporation and by MTT assay. Western blot was used to detect phosphorylation of the key enzymes in the Ras/Raf and PI3K pathways.Sorafenib and PI-103, as single agents inhibited Huh7 proliferation and epidermal growth factor (EGF)-stimulated Huh7 proliferation in a dose-dependent fashion; the combination of sorafenib and PI-103 produced synergistic effects. EGF increased phosphorylation of MEK and ERK, key Ras/Raf downstream signaling proteins; this activation was inhibited by sorafenib. However, sorafenib as a single agent increased AKT(Ser473) and mTOR phosphorylation. EGF-stimulated activation of PI3K/AKT/mTOR pathway components was inhibited by PI-103. PI-103 is a potent inhibitor of AKT(Ser473) phosphorylation; in contrast, rapamycin stimulated AKT(Ser473) phosphorylation. It was found that PI-103, as a single agent, stimulated MEK and ERK phosphorylation. However, the combination of sorafenib and PI-103 caused inhibition of all the tested kinases in the Ras/Raf and PI3K pathways.The combination of sorafenib and PI-103 can significantly inhibit EGF-stimulated Huh7 proliferation by blocking both Ras/Raf/MAPK and PI3K/AKT/mTOR pathways.
