Biogenic silver nanoparticles (AgNP) are among the fastest-growing nanomaterials due to the simplicity, efficiency, and sustainability of their biosynthesis using phytochemicals as reducing and coating agents. The agro-food industry generates large quantities of organic waste, a renewable source of biomolecules for AgNP biosynthesis. The main objective of this work was to prepare and characterize biogenic AgNP using a green tomato waste extract (TE) obtained by subcritical water extraction. To the best of our knowledge, this is the first report on the use of such an extract in the synthesis of AgNP. The effects of the TE and AgNO3 concentrations, reaction time, pH, and temperature on AgNP physico-chemical characteristics and on in vitro cytotoxicity against HaCaT and THP-1 cells were assessed. Antimicrobial activity was determined in vitro and ex vivo. The wound-healing capability of AgNP was evaluated in vivo in an incisional wound mouse model. The developed AgNP have a Surface Plasmon Resonance (SPR) band between 402 and 406 nm and a size of ±60 nm, and they are negatively charged (−42 mV) and spherical. In vitro and ex vivo studies prove that AgNP do not compromise skin cells and can decrease cutaneous irritation. The AgNP formulated in a gel revealed similar wound-healing properties to a commercial silver-containing topical ointment. Overall, the biogenic synthesis of AgNP employing an extract of agricultural waste obtained by an eco-friendly method is simple and cost-effective and presents the potential for application in skin disease management. Diabetes mellitus is a chronic disease, commonly associated with unhealthy habits and obesity, and it is becoming a serious health issue worldwide. As a result, new approaches to treat diabetes are required, and a movement towards more natural approaches is emerging. Consuming fruit and vegetables is advised to prevent diabetes since they contain several bioactive compounds. A simple and effective strategy to include them in the diets of diabetic and obese people is through beverages. This review aims to report the anti-diabetic potentials of different vegetable and fruit beverages. These functional beverages demonstrated in vitro potential to inhibit α-glucosidase and α-amylase enzymes and to improve glucose uptake. In vivo, beverage consumption showed a reduction of blood glucose, increase of insulin tolerance, improvement of lipid profile, control of obesity, and reduction of oxidative stress. This suggests the potential of vegetable- and fruit-based functional beverages to be used as a natural innovative therapy for the management of diabetes. [ABSTRACT FROM AUTHOR]

Triple-negative breast cancers (TNBCs) are more likely to occur in younger patients and have a poor prognosis. They are highly heterogeneous tumors consisting of different molecular subtypes. The only common characteristic among them is the absence of targets for endocrine therapy and human epidermal growth factor receptor 2 (HER2) blockade. In the past two decades, there has been an increased understanding of these tumors from a molecular perspective, leading to their stratification according to new therapeutic strategies. TNBC has ushered breast carcinomas into the era of immunotherapy. The higher frequency of germline BRCA mutations in these tumors enables targeting this repair defect by drugs like PARP inhibitors, resulting in synthetic lethality in neoplastic cells. Additionally, we have the identification of new molecules to which this generation of smart drugs, such as antibody-drug conjugates (ADCs), are directed. In this review, we will discuss the trajectory of this knowledge in a systematic manner, presenting the molecular bases, therapeutic possibilities, and biomarkers. [ABSTRACT FROM AUTHOR]

Atomic force microscopy (AFM) is a useful and powerful tool to study molecular interactions applied to nanomedicine. The aim of the present study was to implement a hands-on atomic AFM course for graduated biosciences and medical students. The course comprises two distinct practical sessions, where students get in touch with the use of an atomic force microscope by performing AFM scanning images of human blood cells and force spectroscopy measurements of the fibrinogen-platelet interaction. Since the beginning of this course, in 2008, the overall rating by the students was 4.7 (out of 5), meaning a good to excellent evaluation. Students were very enthusiastic and produced high-quality AFM images and force spectroscopy data. The implementation of the hands-on AFM course was a success, giving to the students the opportunity of contact with a technique that has a wide variety of applications on the nanomedicine field. In the near future, nanomedicine will have remarkable implications in medicine regarding the definition, diagnosis, and treatment of different diseases. AFM enables students to observe single molecule interactions, enabling the understanding of molecular mechanisms of different physiological and pathological processes at the nanoscale level. Therefore, the introduction of nanomedicine courses in bioscience and medical school curricula is essential.

OBJETIVO: avaliar a taxa de recorrência local, regional e a distância de nova técnica cirúrgica para tratamento conservador do câncer de mama de estádio inicial. A técnica é baseada na ressecção segmentar com dissecção axilar e preservação de pele por incisão periareolar única. MÉTODOS: cento e dezenove pacientes com câncer de mama de estádio I e II foram incluídas neste estudo. O grupo estudo foi constituído por 57 pacientes que se submeteram à cirurgia pela técnica proposta e 62 pacientes submetidas a quadrantectomia clássica constituíram o grupo controle. A radioterapia pós-operatória e o boost foram realizados em ambos os grupos. A média de seguimento foi de 50,1 meses para o grupo estudo e 51,2 para o grupo controle. As taxas de recidiva, sobrevida global e sobrevida livre de doença foram analisadas e comparadas entre os dois grupos. RESULTADOS: a taxa de recidiva local no período foi de 3,5% para o grupo estudo e 4,8% para o grupo controle. Não houve diferença significativa entre os grupos nas taxas de sobrevida livre de doença e sobrevida global. CONCLUSÕES: a técnica de ressecção segmentar com dissecção axilar por incisão única periareolar, preservando pele, não mostrou diferença, em relação à quadrantectomia clássica, nas taxas de recidiva, sobrevida global e sobrevida livre de doença.

Introduction: Nearly 85% of cervical cancer new cases are diagnosed in limited resources countries. Although several strategies have been proposed to reduce the disease burden, challenges remain to provide the best possible care. We report recommendations from an expert consensus meeting convened to address from prevention to management of cervical cancer in limited resources countries. Methods: The expert panel, composed by invited specialists from 38 developing countries in Africa, Asia, Eastern Europe, Latin America, and the Middle East, convened in Rio de Janeiro in September 2019, during the Global Meeting of the International Gynecological Cancer Society (IGCS). Panel members considered the published scientific evidence and their practical experience on the topics, as well as the perceived cost-effectiveness of, and access to, the available interventions. The focus of the recommendations was on geographic regions rather than entire countries because medical practice varies considerably in the countries represented. Resource limitation was qualified as limited access to qualified surgeons, contemporary imaging or radiation-oncology techniques, antineoplastic drugs, or overall funding for provision of state-of-the-art care. Consensus was defined as at least 75% of the voting members selecting a particular answer of the multiple-choice questionnaire, whereas the majority vote was considered as 50% to 74.9%. Results: Consensus was reached for 25 of the 121 (20.7%) questions, whereas for 54 (44.6%) questions there was one option garnering between 50% to 74.9% of votes (majority votes). For the remaining questions, considerable heterogeneity in responses was observed. Discussion: The implementation of international guidelines is challenging in countries with resource limitations or unique health-care landscapes. The development of guidelines by the health care providers in those regions is more reflective of the reality on the ground and may improve medical practice and patient care.
