While regular physical activity is a cornerstone of health, wellness, and vitality, the impact of endurance exercise training on molecular signaling within and across tissues remains to be delineated. The Molecular Transducers of Physical Activity Consortium (MoTrPAC) was established to characterize molecular networks underlying the adaptive response to exercise. Here, we describe the endurance exercise training studies undertaken by the Preclinical Animal Sites Studies component of MoTrPAC, in which we sought to develop and implement a standardized endurance exercise protocol in a large cohort of rats. To this end, Adult (6-mo) and Aged (18-mo) female (n = 151) and male (n = 143) Fischer 344 rats were subjected to progressive treadmill training (5 d/wk, ∼70%-75% VO2max) for 1, 2, 4, or 8 wk; sedentary rats were studied as the control group. A total of 18 solid tissues, as well as blood, plasma, and feces, were collected to establish a publicly accessible biorepository and for extensive omics-based analyses by MoTrPAC. Treadmill training was highly effective, with robust improvements in skeletal muscle citrate synthase activity in as little as 1-2 wk and improvements in maximum run speed and maximal oxygen uptake by 4-8 wk. For body mass and composition, notable age- and sex-dependent responses were observed. This work in mature, treadmill-trained rats represents the most comprehensive and publicly accessible tissue biorepository, to date, and provides an unprecedented resource for studying temporal-, sex-, and age-specific responses to endurance exercise training in a preclinical rat model.

Regular exercise promotes whole-body health and prevents disease, but the underlying molecular mechanisms are incompletely understood1-3. Here, the Molecular Transducers of Physical Activity Consortium4 profiled the temporal transcriptome, proteome, metabolome, lipidome, phosphoproteome, acetylproteome, ubiquitylproteome, epigenome and immunome in whole blood, plasma and 18 solid tissues in male and female Rattus norvegicus over eight weeks of endurance exercise training. The resulting data compendium encompasses 9,466 assays across 19 tissues, 25 molecular platforms and 4 training time points. Thousands of shared and tissue-specific molecular alterations were identified, with sex differences found in multiple tissues. Temporal multi-omic and multi-tissue analyses revealed expansive biological insights into the adaptive responses to endurance training, including widespread regulation of immune, metabolic, stress response and mitochondrial pathways. Many changes were relevant to human health, including non-alcoholic fatty liver disease, inflammatory bowel disease, cardiovascular health and tissue injury and recovery. The data and analyses presented in this study will serve as valuable resources for understanding and exploring the multi-tissue molecular effects of endurance training and are provided in a public repository ( https://motrpac-data.org/ ).

Wearable devices have revolutionized real-time health monitoring, yet challenges persist in enhancing their flexibility, weight, and accuracy. This paper presents the development of a wearable device employing a conductive polyacrylamide–lithium chloride–MXene (PLM) hydrogel sensor, an electronic circuit, and artificial intelligence (AI) for gait monitoring. The PLM sensor includes tribo-negative polydimethylsiloxane (PDMS) and tribo-positive polyurethane (PU) layers, exhibiting extraordinary stretchability (317% strain) and durability (1000 cycles) while consistently delivering stable electrical signals. The wearable device weighs just 23 g and is strategically affixed to a knee brace, harnessing mechanical energy generated during knee motion which is converted into electrical signals. These signals are digitized and then analyzed using a one-dimensional (1D) convolutional neural network (CNN), achieving an impressive accuracy of 100% for the classification of four distinct gait patterns: standing, walking, jogging, and running. The wearable device demonstrates the potential for lightweight and energy-efficient sensing combined with AI analysis for advanced biomechanical monitoring in sports and healthcare applications.

Photographic identification is increasingly being used as a cost-effective and minimally invasive method to monitor species, which is of particular importance for endangered populations that are vulnerable to intrusive research methods. The purpose of our study was to collect photographs of an endangered population of beluga whales (Delphinapterus leucas (Pallas, 1776)) in Cumberland Sound, Nunavut, Canada, for use in photographic identification. Rather than pursuing the whales with boats to collect photographs, drones were used to minimize disturbance. We analyzed drone photographs from 2017 to 2019 for distinctive markings on the whales, which were used to develop a photographic identification catalogue. In total, 93 individuals were identified, with 24 resightings of marked individuals over the survey period. Approximately 43.4% (standard error 3.3%) of the adult beluga population was uniquely marked. The beluga population has been harvested at a rate of 41 whales per year, not including struck and lost, since 2002. The markings were from unknown origins (61%), scars/wounds from gunshots (27%), anthropogenic or predatory given the size and severity (11%), or a satellite tag (1%). The continuation of the photographic identification program will allow for the estimation of important population demographics, such as abundance and calving interval, which are important parameters for population conservation and management.

ObjectiveThe United States faces an opioid crisis. Integrating prescription drug monitoring programs into electronic health records offers promise to improve opioid prescribing practices. This study aimed to evaluate 2 different user interface designs for prescription drug monitoring program and electronic health record integration.Materials and MethodsTwenty-four resident physicians participated in a randomized controlled experiment using 4 simulated patient cases. In the conventional condition, prescription opioid histories were presented in tabular format, and computerized clinical decision support (CDS) was provided via interruptive modal dialogs (ie, pop-ups). The alternative condition featured a graphical opioid history, a cue to visit that history, and noninterruptive CDS. Two attending pain specialists judged prescription appropriateness.ResultsParticipants in the alternative condition wrote more appropriate prescriptions. When asked after the experiment, most participants stated that they preferred the alternative design to the conventional design.ConclusionsHow patient information and CDS are presented appears to have a significant influence on opioid prescribing behavior.

In conducting Arctic field research, hiring local field guides has long been a necessity for providing field teams with local knowledge and fundamental needs of boat operation and navigation, general field logistics/safety, and traditional ecological knowledge (TEK) of local animal distribution and natural history. As new threats to Arctic wildlife emerge and as field research methods evolve, including local Inuit as long-standing members of research teams has provided additional collaborative benefits through expanded local knowledge, greater efficiency of data collection, and longer temporal sampling which provides the opportunity to study uncommon events. We describe the collaboration between southern-based scientists and local Inuit from the community of Pangnirtung, Nunavut, to conduct field research on marine mammals in Cumberland Sound from 1997 to 2021. Through a keen interest in marine mammal field research, Inuit partners in Pangnirtung have become highly proficient in all aspects of sample and data collection and have received advanced technical training to allow for an expanded role in achieving research objectives. This expanded role includes running field research operations independently, as well as the extensive use of drones to capture photographs of whales for the purposes of photographic-identification and to record behavior. Collaboration with local Inuit also provides benefits through employment opportunities, development of technical skills, and opportunities to actively participate in research that aims to conserve culturally important local wildlife populations.

Effective management of animal populations requires knowledge of life history parameters and estimates of population abundance. One method commonly used to estimate abundance is capture–recapture analyses of photographs. Small, relatively inexpensive, rotary-wing drones have become an effective platform for obtaining high-quality aerial photographs of whales. To conduct capture–recapture analyses the animal needs to be defined as marked or unmarked and the photographs must be of high quality. While a system for scoring quality and markedness has previously been developed for bowhead whales (Balaena mysticetus Linnaeus, 1758) (Rugh et al. 1998. Rep. int. Whal. Commn. 48: 501–512), a revised scoring system was needed to incorporate increased information in photographs taken by drones. We present a revised scoring system that enlarges two of the previously defined areas of the whale examined for markings and incorporates smaller markings into the definition of marked whales. We scored 30 whales using the previous criteria and the revised criteria developed in this paper. More whales were identified as marked (23%) and mark scores were higher for 30% of the zones scored using the new system. Increasing the number of marked whales during capture–recapture studies increases the precision of estimated parameters and permits us to make those estimates with smaller samples of photographs.

ObjectiveTo provide expert recommendations for the safe and effective application of repetitive transcranial magnetic stimulation (rTMS) in the treatment of major depressive disorder (MDD).ParticipantsParticipants included a group of 17 expert clinicians and researchers with expertise in the clinical application of rTMS, representing both the National Network of Depression Centers (NNDC) rTMS Task Group and the American Psychiatric Association Council on Research (APA CoR) Task Force on Novel Biomarkers and Treatments.EvidenceThe consensus statement is based on a review of extensive literature from 2 databases (OvidSP MEDLINE and PsycINFO) searched from 1990 through 2016. The search terms included variants of major depressive disorder and transcranial magnetic stimulation. The results were limited to articles written in English that focused on adult populations. Of the approximately 1,500 retrieved studies, a total of 118 publications were included in the consensus statement and were supplemented with expert opinion to achieve consensus recommendations on key issues surrounding the administration of rTMS for MDD in clinical practice settings.Consensus processIn cases in which the research evidence was equivocal or unclear, a consensus decision on how rTMS should be administered was reached by the authors of this article and is denoted in the article as "expert opinion."ConclusionsMultiple randomized controlled trials and published literature have supported the safety and efficacy of rTMS antidepressant therapy. These consensus recommendations, developed by the NNDC rTMS Task Group and APA CoR Task Force on Novel Biomarkers and Treatments, provide comprehensive information for the safe and effective clinical application of rTMS in the treatment of MDD.

The temporal evolution of intermediate-depth seismicity before and after the 2011 M9.0 Tohoku-oki earthquake reveals interactions between plate interface slip and deformation in the subducting slab. We investigate seismicity rate changes in the upper and lower planes of the double seismic zone beneath northeast Japan using both a β statistic approach and a temporal epidemic type aftershock sequence model. We do not observe an anomalous precursory increase in intermediate-depth earthquake activity preceding the mainshock; however, following the mainshock, we observe a rate increase for the intermediate-depth earthquakes in the upper plane. The average ratio of upper plane to lower plane activity and the mean deep aseismic slip rate both increased by factor of 2. An increase of downdip compression in the slab resulting from coseismic and postseismic deformation enhanced seismicity in the upper plane, which is dominated by events accommodating downdip shortening from plate unbending.

The properties of the first kilometers of the Martian atmospheric Planetary Boundary Layer have until now been measured by only a few instruments and probes. InSight offers an opportunity to investigate this region through seismoacoustics. On six occasions, its seismometers recorded short low‐frequency waveforms, with clear dispersion between 0.4 and 4 Hz. These signals are the air‐to‐ground coupling of impact‐generated infrasound, which propagated in an low‐altitude atmospheric waveguide. Their group velocity depends on the structure of effective sound speed in the boundary layer. Here, we conduct a Bayesian inversion of effective sound speed up to 2,000 m altitude using the group velocity measured for events S0981c, S0986c and S1034a. The inverted effective sound speed profiles are in good agreement with estimates provided by the Mars Climate Database. Differences between inverted and modeled profiles can be attributed to a local wind variation in the impact→station direction, of amplitude smaller than 2 m/s. Plain Language Summary: The Martian Planetary Boundary Layer corresponds to the first few kilometers of the atmosphere. The InSight lander offers the opportunity to investigate its properties via the coupling of seismic and acoustic waves. Impact‐generated infrasound waves were recorded for the first time on Mars by the seismometers of the InSight mission. These infrasound waves propagated in an atmospheric waveguide in the first kilometers above the Martian surface, and thus present a frequency‐dependent group velocity. This frequency‐dependence, also known as a dispersion relation, is influenced by the structure of the speed of sound in the waveguide. Here, we use group velocity measured for events S0981c, S0986c and S1034a to invert the variations of effective sound speed between 0 and 2,000 m altitude. For the three events, the inverted profiles are in good agreement with estimates provided by the Mars Climate Database using global climate modeling. The differences between inverted and modeled profiles can be attributed to a local variation in wind in the impact→station direction, with magnitude smaller than 2 m/s. Alkyl nitrites ("poppers") are a diverse class of volatile chemical compounds with a varied legal and medical history. Though once commonly prescribed to treat angina, popper use is now almost exclusively recreational. Currently, poppers are widely available and sold legally under labels like "solvent cleaner", despite marketing suggesting they are meant to be consumed. As a result, there is little incentive for producers to implement robust quality controls to protect users. In this study, nine common popper brands were analyzed using hydrogen-1 and carbon-13 nuclear magnetic resonance spectroscopy to assess the presence of impurities. Physical labels on all nine samples indicated the contents were "pure" isobutyl nitrite, despite contradictory online marketing in several cases. Spectral results showed isobutyl nitrite was present in all popper samples. However, there was evidence that various unlabeled compounds were also present in all samples. The identity and concentration of these contaminants were not clear, but the seemingly ubiquitous presence of impurities and lack of consistency in the tested samples are concerning and may represent a threat to users' health. We hope the results of this study draw attention to the potential dangers of recreational popper use and the need to reassess how these compounds are regulated. [ABSTRACT FROM AUTHOR]

In order to provide surface geodetic measurements with “landslide-wide” spatial coverage, we develop and validate a method for the characterization of 3-D surface deformation using the unique capabilities of the Uninhabited Aerial Vehicle Synthetic Aperture Radar (UAVSAR) airborne repeat-pass radar interferometry system. We apply our method at the well-studied Slumgullion Landslide, which is 3.9 km long and moves persistently at rates up to ∼2 cm/day. A comparison with concurrent GPS measurements validates this method and shows that it provides reliable and accurate 3-D surface deformation measurements. The UAVSAR-derived vector velocity field measurements accurately capture the sharp boundaries defining previously identified kinematic units and geomorphic domains within the landslide. We acquired data across the landslide during spring and summer and identify that the landslide moves more slowly during summer except at its head, presumably in response to spatiotemporal variations in snowmelt infiltration. In order to constrain the mechanics controlling landslide motion from surface velocity measurements, we present an inversion framework for the extraction of slide thickness and basal geometry from dense 3-D surface velocity fields. We find that the average depth of the Slumgullion Landslide is 7.5 m, several meters less than previous depth estimates. We show that by considering a viscoplastic rheology, we can derive tighter theoretical bounds on the rheological parameter relating mean horizontal flow rate to surface velocity. Using inclinometer data for slow-moving, clay-rich landslides across the globe, we find a consistent value for the rheological parameter of 0.85 ± 0.08.

Two instructors report our experience co-teaching an action research (AR) required as part of an e-learning master's degree. Adopting a practice-centered stance we focus on the course activities of participants (instructors and students), with particular attention to the careful crafting of course elements with the goal of achieving an excellent learning experience for students. The case narrative describes the course and ways in which we have modified the course based on a variety of considerations. We also outline problems and areas still in need of improvement. We reflect on the role of theory in our own pursuit of excellence, and the role of theory in our students' inquiry processes. We find that theory is just another tool or resource to apply to the work, with the core concerns being the needs of students and the learning environment.

Accounts of killer whale (Orcinus orca) predation on marine mammals in the Canadian Arctic are relatively uncommon. Although second-hand reports of killer whale predation events in the Arctic are more common in recent years, these observations are generally poorly documented and the outcome of attacks are often unknown. On 12 August 2016, a floating bowhead whale (Balaena mysticetus) carcass was found off-shore in Cumberland Sound, Nunavut — presumably predated by killer whales that were sighted in the area. Inspection of the carcass revealed injuries consistent with published accounts of killer whale predation on large whales and observations of killer whale predation on bowheads described in Inuit traditional knowledge. The bowhead was male, 6.1 m long in good nutritional condition and estimated between 14 and 16 months old. As a recently weaned yearling, this whale would have been highly vulnerable to killer whale predation. With decreasing summer sea ice making some areas of the Arctic more accessible, the incursion and presence of killer whales in the Arctic is expected to increase. A better understanding of Arctic killer whale predation pressure is needed to predict the potential impact they will have on the eastern Canada–west Greenland bowhead population as well as on other marine mammal prey.

The Canadian Arctic is warming at three times the rate of the rest of the planet and the effects of climate change on the Arctic marine carbon cycle remains unconstrained. Baffin Bay is a semi-enclosed, Arctic basin that connects the Arctic Ocean to the north to the Labrador Sea to the south. While the physical oceanography of surface Baffin Bay is well characterized, less is known about deep water formation mechanisms within the Basin. Only a few residence-time estimates for Baffin Bay Deep Water (BBDW) exist and range from 20 to 1450 years. Better residence time estimates are needed to understand the oceanographic significance of Baffin Bay. Here we report stable carbon (δ13C) and radiocarbon (Δ14C) values of dissolved inorganic carbon (DIC) collected aboard the CCGS Amundsen in 2019. DIC δ13C and Δ14C values between ranged between -0.7‰ to +1.9‰ and -90.0‰ to +29.8‰, respectively. Surface DIC δ13C values were between +0.7‰ to +1.9‰, while deep (>100m) values were 0.0 to -0.7‰. Surface DIC Δ14C values ranged between -5.4‰ to +22.9‰, while deep DIC (>1400m) DIC Δ14C averaged -82.2 ± 8.5‰ (n = 9). To constrain natural DIC Δ14C values, we quantified the amount of atmospheric “bomb” 14C in DIC (Δ14Cbomb; using the potential alkalinity method; Palk) and anthropogenic DIC (DICanth; using the ΔC* method). Both proxies indicate an absence of Δ14Cbomb and DICanth below 1000m. Using two previously proposed deep water formation mechanisms and our corrected DIC Δ14Cnatural values, we estimated a 14C-based residence time of 360-690 years for BBDW. Based on these residence times, we infer carbon is likely stored for centuries in deep Baffin Bay.

Learning communities can emerge spontaneously when people find common learning goals and pursue projects and tasks together in pursuit of those goals. "Bounded" learning communities (BLCs) are groups that form within a structured teaching or training setting, typically a course. Unlike spontaneous communities, BLCs develop in direct response to guidance provided by an instructor, supported by a cumulative resource base. This article presents strategies that help learning communities develop within bounded frameworks, particularly online environments. Seven distinguishing features of learning communities are presented. When developing supports for BLCs, teachers should consider their developmental arc, from initial acquaintance and trust-building, through project work and skill development, and concluding with wind-down and dissolution of the community. Teachers contribute to BLCs by establishing a sense of "teaching presence", including an atmosphere of trust and reciprocal concern. The article concludes with a discussion of assessment issues and the need for continuing research. (Contains 3 figures and 2 tables.)

A non-magnetic piston-cylinder pressure cell is presented for solution-state NMR spectroscopy at geochemical pressures. The probe has been calibrated up to 20 kbar using in situ ruby fluorescence and allows for the measurement of pressure dependencies of a wide variety of NMR-active nuclei with as little as 10 μL of sample in a microcoil. Initial (11)B NMR spectroscopy of the H3BO3-catechol equilibria reveals a large pressure-driven exchange rate and a negative pressure-dependent activation volume, reflecting increased solvation and electrostriction upon boron-catecholate formation. The inexpensive probe design doubles the current pressure range available for solution NMR spectroscopy and is particularly important to advance the field of aqueous geochemistry.

General Internal Medicine research evolves in response to the needs of the patients to whom we provide care. Currently, many studies exclude older adults who deeply affect the clinical care of this population. With the number of older adults increasing, creating research protocols that include older adults with multiple chronic comorbidities is imperative. Through funding from the Association of Specialty Physicians, a working group of aging-responsive researchers from the Society of General Internal Medicine was convened to tackle this issue. The goal of this article is threefold: 1) to shed light on the current exclusion of older adults in research; 2) to identify and propose research protocol solutions for overcoming barriers to including older adults in research; and 3) to provide suggestions for research funding. The extent to which these recommendations can create change depends greatly on our researcher colleagues. By embracing these challenges, we hope that the care provided to older adults with multiple chronic conditions will no longer be extrapolated, but become evidence-based.

Abstract Background Posterior reversible encephalopathy syndrome (PRES) is a rare neurological disorder usually associated with specific medical conditions that cause a disturbance of the CNS homeostasis. It has seldom been reported to be a consequence of an iatrogenic intervention causing intracranial hypotension. Case presentation We report the case of an individual 69-year-old male presenting with headache and blurred vision following cerebrospinal fluid (CSF) leak from resection of a sellar mass. The patient developed the condition following removal of the lumbar drain post-operatively. Magnetic Resonance Imaging showed bilateral occipital, parieto-occipital, and cerebellar T2 FLAIR hyper-intensities, suggesting a radiological diagnosis of posterior reversible encephalopathy syndrome (PRES). The patient’s symptoms started to improve shortly afterwards and had completely resolved at 3 months follow-up. Conclusions The absence of severe hypertension and presence of an intraoperative CSF leak requiring placement of the lumbar drain suggests that decreased CSF volume and associated reactive hyperemia could have a role in the pathophysiology of the disease.

ABSTRACT When conducting aerial surveys of wildlife populations, a number of animals go undetected by observers. The use of infrared imagery may offer a solution to improve detection rates and reduce visibility bias. Our objective was to compare the use of an infrared camera system with traditional methods using direct visual observations for estimating density of ringed seals (Pusa hispida) on ice. We conducted aerial surveys of ringed seals in the areas of Pond Inlet and western Hudson Bay in the Canadian Arctic during spring 2016 and 2017. Infrared videos were recorded within a 250‐m‐wide strip beneath the aircraft and potential seals were verified using corresponding visual images from a digital single‐lens reflex camera. Results from strip‐transect analysis of infrared observations were compared with results from strip‐transect analysis of direct visual observations and results from line‐transect analysis of a combined data set of infrared and direct visual observations. The different methods produced different density estimates with the infrared strip‐transect and line‐transect methods producing similar results that were, on average, approximately 2–3 times greater than strip‐transect analyses of observer data. Use of infrared imagery provides several advantages over traditional methods because it allows for more efficient and reliable detection of seals, eliminates the need for large teams of trained observers, and simplifies the process of data analysis and density estimation. © 2019 The Wildlife Society.

Resource development in Arctic waters is proceeding rapidly leading to increased interactions with Arctic wildlife. As sea ice extent decreases, the demand for shipping and ice-breaking operations will expand into winter and spring with greater impact on ice-dependent pinnipeds. However, knowledge of the distribution of these species, such as ringed seals (Pusa hispida (Schreber, 1775)), during spring within areas of resource development is lacking. Baffinland Iron Mines Corporation’s Mary River iron ore port in southern Milne Inlet, Nunavut opened in 2015 with proposed ice-breaking activities in spring — an important period in ringed seal seasonal life-history. We conducted infrared and photographic aerial surveys in June 2016 and 2017 to overlay the proposed ice-breaking route with ringed seal hotspots (i.e., areas of higher density). We identified four areas of overlap where proposed ice-breaking would traverse through ringed seal hotspots: eastern and western Eclipse Sound (a ringed seal pupping ground identified by local knowledge), the middle of Milne Inlet, and southern Milne Inlet. We identified potential negative implications of spring ice-breaking operations on ringed seals such as displacement, separation of mothers and pups, destruction of resting and birth lairs, and vessel–seal collisions. Results are relevant to policy decision-makers who can develop mitigation strategies in the rapidly melting and developing Arctic.

Abstract Visual and observer aerial surveys are important for monitoring wildlife populations but are subject to visibility biases where animals may go undetected. The use of infrared technology in aerial surveys has the potential to reduce visibility biases, both when recording data and in the retrospective processing of the footage, and thus complements visible wavelength photography. We used infrared video during marine mammal surveys in the high‐Arctic and indirectly detected narwhal (Monodon monoceros) via their thermal flukeprints (i.e., thermo‐stratified water mixing from fluke strokes). This novel indicator persisted for a longer duration than when the animal was at the water's surface, which likely improved the probability of an animal being observed by increasing the duration of its detectability. Using infrared to complement aerial photographic surveys may assist in monitoring whales, especially in remote areas. Abstract A climate‐driven decline of sea ice has increased international interest in understanding the ecology of seldom studied high‐Arctic regions projected to retain the last remnants of summer sea ice. Arctic marine mammals are vulnerable to future environmental change due to their high dependence on sea ice. We completed aerial surveys in August 2019 to document the occurrence and location of marine mammals within the recently designated Tuvaijuittuq Marine Protected Area which acts as a last refuge for ice‐associated species. We present photographic evidence of narwhals (Monodon monoceros), Atlantic walruses (Odobenus rosmarus), bearded seals (Erignathus barbatus), and ringed seals (Pusa hispida) in Archer Fjord, northern Ellesmere Island, Nunavut, Canada (81°14′–81°49′ N). We discuss these observations in the context of historical records and more recent opportunistic sightings from the region to support the hypothesis of an expansion north of the currently accepted range of narwhal. The diversity and number of observations of marine mammals in Archer Fjord is a striking contrast to the relative scarcity of the rest of the surveyed region, with very few observations of ringed seals throughout the more northerly study area. Our observations highlight opportunities to further explore high‐latitude areas that may host unique marine mammal biodiversity in the region, including narwhal.
