Nickel (Ni) is an essential micronutrient for plants although it is considered toxic when present in excess in the soil. This study investigated the transfer of Ni from the soil to rice in terraced paddy fields affected by serpentinite, which contains an anomalously higher Ni content compared with other geological materials. Soils, soil solutions, and rice plants were collected at several different growing stages from three adjacent terraced paddy fields subject to the same water and fertilizer management. Temporal changes in their elemental compositions revealed that a higher concentration of Ni was dissolved in the soil solution during flooded conditions, probably due to the co-solubilization with Mn oxides under low redox potential conditions. However, rice accumulated Ni at a higher rate during the drainage period than in the flooding period. Although the Ni concentration in the soil solution was lowest in the drainage period, the relative concentration to Fe (i.e., Ni/Fe ratio) was much higher than that in flooded conditions. These relationships suggest that a potential measure to counter the transfer of Ni from the soil to rice in serpentine-affected paddy fields is to increase Fe phytoavailability during the drainage period.

Active Al, Fe and Si (i.e., oxalate extractable fraction: Alo, Feo, Sio) strongly affect soil physical, chemical and biological properties. This study examined the pedogenic factors affecting Alo, Feo and Sio contents across a soil weathering sequence in the Cameroon volcanic line. We investigated the B horizon (∼50-cm depth) from 26 soils formed in basaltic materials at different elevations (110–2570 m) incorporating a wide range of temperature (14–27 °C) and precipitation (1520–3130 mm). The weathering sequence ranged from weakly weathered Andisols in the southwest region grading to strongly weathered Oxisols on the central highlands. We assumed pyrophosphate extractable Al/Fe (Alp/Fep) as organo-Al/Fe complexes, and Sio, (Alo − Alp) and (Feo − Fep) as short-range-order (SRO) minerals. Factor analysis of climatic (e.g., temperature and precipitation/leaching metrics) and soil geochemical properties (e.g., weathering indices) identified three independent factors representing temperature/dry season intensity, weathering degree and precipitation/leaching as the primary determinants of Alo, Feo and Sio concentrations. Organo-metal complexes (Alp and Fep) were negatively correlated with the temperature/dry season intensity factor, whereas the SRO mineral phases (Sio, Alo − Alp and Feo − Fep) were negatively correlated with weathering degree. The precipitation/leaching factor positively correlated with Alo, Feo and Sio. Our analysis infers that low temperature promotes the formation and preservation of organo-Al/Fe complexes, whereas weathering degree is more critical for SRO minerals. Further, increased weathering and a drier climate enhance the formation of crystalline clay minerals at the expense of SRO minerals. Allophanic materials (Sio) were evident (Sio: 9–43 g kg−1) only in weakly weathered soils. However, low allophanic contents were found in more highly weathered soils (Sio: 2–7 g kg−1) accompanied by high Alp and Fep, suggesting the importance of volcanic parent materials as a direct source of Al and Fe via weathering for the formation of organo-metal complexes. In sum, we clarified the discriminatory effects of climatic factors and degree of weathering in regulating the composition of the active Al, Fe and Si fractions along the Cameroon volcanic line.

BACKGROUND: Remote-access thyroidectomy and its cosmetic merit have been widely accepted, but remote-access parathyroidectomy has not become common. There are few reports about the risks and effectiveness of a remote-access endoscopic parathyroidectomy. Herein, we evaluated the risks and benefits of total endoscopic parathyroidectomy (TEP) for patients with primary hyperparathyroidism (PHPT). We retrospectively compared the surgical outcomes of TEP and open minimally invasive parathyroidectomy (MIP). METHODS: We analyzed the cases of 28 patients with PHPT who were scheduled to undergo a MIP at Mita Hospital (Tokyo) during the period from April 2015 to March 2019, all of whom were presumed preoperatively to have a single adenoma. RESULTS: Eleven of the patients underwent a TEP (10 females, one male; mean age 54.2 years). The other 17 patients underwent an open MIP (11 females, 6 males; mean age 63.5 years). The younger patients and the females tended to select endoscopic surgery as their treatment. International audience; The Fukushima nuclear accident in March 2011 generated a 3000 km2 plume of soils heavily contaminated with 137Cs. Decontamination was completed early in 2019. Typhoon Hagibis was the first extreme event that occurred in the region after decontamination. Its impact on sediment sources and sediment 137Cs contamination was investigated through the application of a sediment fingerprinting procedure using spectrocolorimetry and geochemical properties. Sediment deposits (n=24) were collected in the Mano and Niida River catchments after the 2019 typhoons, and their signature was compared to that of potential sources (e.g., cropland, forests, and subsurface; n=57). Results demonstrate the dominance of cropland as the main source of sediment (mean: 54%) followed by forests (41%) with much lower contributions of subsurface material (5%). Overall, 137Cs concentrations in sediment were on average 84%–93% lower than the levels recorded after the accident in 2011, which demonstrates the effectiveness of cropland decontamination.

International audience; Sediment that has deposited after the flood generated by the 2019 typhoon (referred to as Flood Sediment, FS) was collected along two rivers in eastern Fukushima Prefecture, Japan to determine the total and exchangeable radiocesium (137 Cs), and acid-extractable potassium (K) contents. Then, these FS parameters were compared with those of decontaminated soils (DS) in nearby agricultural fields to discuss the potential transfer risk of 137 Cs from rivers to nearby remediated soils. While FS had about four times higher total 137 Cs content, it showed a three-times lower exchangeable 137 Cs content than DS. Furthermore, acid-extractable K referred to as non-exchangeable K (Nex-K) content was high enough to restrict 137 Cs transfer from soil to crops for both FS and DS. From these comparisons, we concluded that deposition of FS onto DS may not increase the transfer of 137 Cs from soil to crops.

Large variations in the maximum earthquake magnitude ($$M_{{\text{max}}}$$ M max ) have been observed among the world’s subduction zones. There is still no universal relationship between $$M_{{\text{max}}}$$ M max and a given subduction-zone parameter, such as plate age, plate dip angle, or plate velocity, which suggests that multiple parameters control $$M_{{\text{max}}}$$ M max . Here, we conduct exhaustive variable selections that are based on three evaluation criteria; leave-one-out cross-validation errors (LOOCVE), Akaike information criterion (AIC), and Bayesian information criterion (BIC) to determine the combination of subduction-zone parameters that best explains $$M_{{\text{max}}}$$ M max . Multiple linear regression analyses are applied using 18 subduction-zone parameters as potential candidates for the explanatory variables of $$M_{{\text{max}}}$$ M max . The minimum BIC is obtained when five variables (trench sediment thickness, existence of an accretionary prism, upper-plate crustal thickness, bending radius of the subducting oceanic plate, and trench depth) are selected as explanatory variables; each variable contributes positively to $$M_{{\text{max}}}$$ M max . Minimum LOOCVE and AIC values are obtained when eight variables (the five parameters for BIC, plus the along-strike plate convergence rate, age of the subducting plate, and maximum depth of the subducting plate) are selected. Our selection of the trench sediment thickness and plate bending radius contributing to $$M_{{\text{max}}}$$ M max is consistent with previous studies. The results show that increasing upper-plate crustal thickness results in a large $$M_{{\text{max}}}$$ M max . In addition to smoothing the subducting-plate interface via subducted sediments, along-dip extension of the crustal area along the convergent plate boundary would be important for generating a large earthquake. Potassium (K) and radiocaesium (RCs) were chemically extracted from soils derived from granite (G soils) and sedimentary rock (S soils) in Fukushima, Japan. The extractants employed were 1 M HNO3, concentrated HNO3, and HF + HClO4. As S soils contain a lower amount of trioctahedral 2:1 phyllosilicates than G soils, the RCs/K ratio was higher in S soils than in G soils with 1 M HNO3 extraction, indicating that the potential risk of soil-to-plant transfer of RCs is higher in S soils than in G soils. In conclusion, information about surface geology is important in predicting the spatial pattern of soil characteristics related to transferability of RCs.

The Fukushima nuclear accident released large quantities of radionuclides into the environment in March 2011 and generated a 3000-km² plume of soils heavily contaminated with Cs-137. Soil erosion in the region mainly takes place during typhoons generally occurring between July and October (Laceby et al., 2016). During these events, rivers draining the main plume may transport large quantities of sediment and radiocesium. Typhoon Hagibis that occurred in October 2019 was the most intense rainfall event affecting the Fukushima region (rainfall range: 77–558 mm) since the nuclear accident in 2011. It led to extensive landsliding and river overflow.The impact of this event on sediment sources and Cs-137 contamination was quantified through the implementation of sediment fingerprinting using geochemistry and spectrocolorimetry as potential input properties. The signature of potential source material (including cropland prepared for recultivation after decontamination, forests and subsurface material originating from landslides and channel bank collapse; n=57) was compared with that of sediment deposits collected in the Mano and Niida River catchments late in October 2019. Results show that cropland supplied the main source of sediment (average: 54%) along with forests (41%). In contrast, the contribution of subsurface material (5%) was much lower, likely because landslides and channel bank erosion mainly took place after the flood peak (Evrard et al., 2020). However, this material that deposited at the foot of hillslopes after the typhoon may be mobilized and delivered to the river network by subsequent rainfall events.Overall, this flood did not modify the decreasing trend observed in terms of Cs-137 contamination in sediment transiting these rivers between 2011 and 2019. Concentrations in Cs-137 observed in sediment collected in 2019 were on average 84–93% lower than those measured after the accident in 2011. Radiocesium (RCs) is selectively adsorbed on interlayer sites of weathered micaceous minerals, which can reduce the mobility of RCs in soil. Therefore, soils developed from mica-deficient materials (e.g. serpentine soils) may have a higher risk of soil-to-plant transfer of RCs. Soils were collected from three serpentine soil profiles; Udepts in Oeyama, Japan, and Udepts and Udox in Kinabalu, Malaysia. Soil was sampled every 3 cm from 0 to 30 cm depth and sieved to isolate soil particles of ≤20 μm diameter for the assessment of radiocesium interception potential (RIP) after a series of pretreatments. Weathered micaceous minerals (micas) are able to release potassium ion (K+) and fix caesium-137 (137Cs), both of which reduce soil-to-plant transfer of 137Cs. Among micas, trioctahedral micas such as biotite is expected to have a stronger ability to supply nonexchangeable K+ and a higher amount of Cs fixation sites than dioctahedral micas such as illite. Although biotite is predominant in granitic soils (G soils), illite is mainly dominant in sedimentary rock soils (S soils). Therefore, we hypothesized that G soils have a lower 137Cs transfer risk than S soils because of this difference in mineralogy. The objective of the present study was to determine the transfer factor (TF) of 137Cs and stable Cs (SCs) and to elucidate the determinant factors of TFs for G and S soils in Fukushima, Japan. Pot experiments were carried out with rice (Oryza sativa L. cv. Hokuriku 193) in G and S soils to determine the TF of 137Cs (TF-137Cs) and stable Cs (TF-SCs) under K-deficient conditions. TF-137Cs and TF-SCs were highly correlated, and both were significantly lower for G soils than for S soils. Higher TF values were shown for soils with lower amounts of exchangeable and nonexchangeable K or with higher percentages of exchangeable 137Cs (ex137Cs). The percentage of ex137Cs was negatively correlated with the amount of Cs fixation sites, represented by the radiocaesium interception potential. Thus, we concluded that smaller TF values for G soils were caused by a stronger ability to supply nonexchangeable K+ and a higher amount of Cs fixation sites. These findings will contribute to the establishment of soil screening techniques based on 137Cs transfer risk in Fukushima prefecture.

Recent statistical analyses on the isotopic compositions of oceanic, arc, and continental basalts have revealed that the Earth's mantle is broadly divided into eastern and western hemispheres. The present study aimed to characterize the isotopically defined east–west geochemical hemispheres using trace-element concentrations. Basalt data with Rb, Sr, Nd, Sm, Pb, Th, and U in addition to the isotopic ratios 87Sr/86Sr, 143Nd/144Nd, 206Pb/204Pb, 207Pb/204Pb, and 208Pb/204Pb were selected mostly from the GEOROC and PetDB databases. A total of 4787 samples were used to investigate the global geochemical variations. The results show that the wide trace-element variations are broadly explained by the melting of melt-metasomatized and fluid-metasomatized mantle sources. The larger amount of the fluid component derived from subducted plates in the eastern hemisphere than that in the western hemisphere is inferred from the basalts. These characteristics support the hypothesis that focused subduction towards the supercontinent created the mantle geochemical hemispheres.

Artificial radionuclides including radiocesium (134Cs and 137Cs) and radiosilver (110mAg) were released into the environment following the Fukushima Dai-ichi nuclear power plant accident in March 2011. These particle-bound substances deposited on soils of north-eastern Japan, located predominantly within a ∼3000 km2 radioactive fallout plume and drained by several coastal rivers to the Pacific Ocean. The current dataset (Evrard et al., 2021), which can be accessed at https://doi.org/10.1594/PANGAEA.928594, compiles gamma-emitting artificial radionuclide activities measured in 782 sediment samples collected from 27 to 71 locations across catchments draining ∼6450 km2 during 16 fieldwork campaigns. These campaigns were conducted in Japan between November 2011 and November 2020 in river catchments draining the main radioactive plume. This database may be useful to evaluate and anticipate the post-accidental redistribution of radionuclides in the environment and for the spatial validation of models simulating the transfer of radiocesium across continental landscapes.

The effects of water on subduction dynamics, e.g., plate migration rate, slab geometry, stress field, and back-arc spreading, are investigated by using a 2-D self-consistent model for lithosphere subduction and whole mantle convection. We solve water transportation coupled with hydrous mineral phase changes. Mantle flows and water transportation are interactive through constitutive and state equations for hydrous rocks. Our model has successfully reproduced the water distribution in a mantle wedge and along the slab with sufficient resolution comparable to that of previous models that focus on the mantle wedge structure. As a result, low density owing to hydration reduces subduction rates, back-arc spreading, and slab stagnation on the phase boundary at 660-km depth, whereas low viscosity owing to hydration enhances rapid subduction, trench migration, and slab stagnation. We attribute these results to mechanisms that cause the hydrous buoyancy of subducting plates to reduce the slab pull force and the accompanying tensile stress on overlying lithosphere. In addition, hydrous weakening diminishes the mechanical coupling of the subducted slab with the wedge mantle and overriding lithosphere. Thus, water is capable of generating two opposite situations in the stress field of the overlying lithosphere and the subduction rate. Water is therefore expected to be an important mechanism for generating broad styles of the subduction structure and kinematics, as observed in actual subduction zones such as Tonga and Mariana. Such observed variation in the subduction mode can be caused by variation in buoyancy corresponding to the water content from relatively dry to several thousands of parts per million for the wedge mantle and slab surface, whereas the extremely buoyant case does not appear to occur in nature. Water in the mantle is thus key to better understand the whole-mantle-scale slab dynamics as well as island arc volcanic processes.

To evaluate the available silicon (Si) content in agricultural soils in Japan and to investigate the determining factors of this content, we collected 180 soil samples from the surface layer of paddies and upland fields in Japan and determined their available Si contents. A phosphate buffer (PB; 0.02 M, pH 6.9) or an acetate buffer (AB; 0.1 M, pH 4.0) was used to extract available Si from the soil samples, and the Si concentrations in the extracts were determined by inductively coupled plasma-atomic emissions spectroscopy (ICP-AES). The total Si content and selected physicochemical properties were also determined for the soil samples. The median values of the available Si contents by the PB and AB methods were 48.8 and 79.7 mg kg−1 and corresponded to 0.017% and 0.027% of the total Si content, respectively. The Fukushima Dai-ichi Nuclear Power Plant (FDNPP) accident in March 2011 resulted in the contamination of Japanese landscapes with radioactive fallout. Accordingly, the Japanese authorities decided to conduct extensive remediation activities in the impacted region to allow for the relatively rapid return of the local population. The objective of this review is to provide an overview of the decontamination strategies and their potential effectiveness in Japan, focussing on particle-bound radiocesium. In the Fukushima Prefecture, the decision was taken to decontaminate the fallout-impacted landscapes in November 2011 for the 11 municipalities evacuated after the accident (Special Decontamination Zones – SDZ, 1117 km2) and for the 40 non-evacuated municipalities affected by lower, although still significant, levels of radioactivity (Intensive Contamination Survey Areas, 7836 km2). Decontamination activities predominantly targeted agricultural landscapes and residential areas. No decontamination activities are currently planned for the majority of forested areas, which cover ~ 75 % of the main fallout-impacted region. Research investigating the effectiveness of decontamination activities underlined the need to undertake concerted actions at the catchment scale to avoid the renewed supply of contamination from the catchment headwaters after the completion of remediation activities. Although the impact of decontamination on the radioactive dose rates for the local population remains a subject of debate in the literature and in the local communities, outdoor workers in the SDZ represent a group of the local population that may exceed the long-term dosimetric target of 1 mSv yr−1. Decontamination activities generated ~ 20 million m3 of soil waste by early 2019. The volume of waste generated by decontamination may be decreased through incineration of combustible material and recycling of the less contaminated soil for civil engineering structures. However, most of this material will have to be stored for ~ 30 years at interim facilities opened in 2017 in the close vicinity of the FDNPP before being potentially transported to final disposal sites outside of the Fukushima Prefecture. Further research is required to investigate the perennial contribution of radiocesium from forest sources. In addition, the re-cultivation of farmland after decontamination raises additional questions associated with the fertility of remediated soils and the potential transfer of residual radiocesium to the plants. Overall, we believe it is important to synthesize the remediation lessons learnt following the FDNPP nuclear accident, which could be fundamental if a similar catastrophe occurs somewhere on Earth in the future.

Background We previously showed that glycated albumin (GA) is a useful glycemic control indicator in patients with neonatal diabetes mellitus (NDM), and that age-adjusted GA (Aa-GA) can reflect more accurately glycemic control status. Here, we investigated whether the age at diagnosis influences Aa-GA at diagnosis of NDM. Methods Eight patients with NDM whose GA was measured at diagnosis (age at diagnosis: 39 ± 18 days; GA: 31.3 ± 7.6%; Aa-GA: 47.1 ± 10.3%; plasma glucose: 525 ± 194 mg/dl) were included. Aa-GA was calculated as follows: Aa-GA = GA × 14.0/[1.77 × log-age (days) + 6.65]. Correlations of GA or Aa-GA at diagnosis with its logarithmically transformed age in days (log-age), plasma glucose, and their product were investigated. Results GA at diagnosis was not significantly correlated with log-age or plasma glucose. On the other hand, Aa-GA at diagnosis was significantly positively correlated with plasma glucose (R = 0.75, P = 0.031) and was more strongly positively correlated with the product of plasma glucose and log-age (R = 0.82, P = 0.012) although it was not correlated with log-age. Conclusion Aa-GA at diagnosis is influenced by both age in days and plasma glucose. This finding is likely to show the aspect that age in days is almost equal to diabetes duration because glycemic control indicators including GA reflect the weighted mean of plasma glucose.

In this paper we show three learning materials (examples) of the heuristic learning as one of the active leaning, which we did with an undergraduate student (second author). The first is some extension of the barycenter (or several centers) of triangles (originally H. Hamanaka). The second is also one of the trials to make the power of point with respect to quadratic curves instead of circles (inspired from H. Nakazato's work with respect to parabolas). The third is heuristic learning by using ICT, for example, rediscovering of Hervey’s point, Steiner's circle and Lambert's thoerem, and perhaps new theorems about circumcenters of triangles made by tangents lines of a parabola. Finally we discuss on the meaning or problems of heuristic learning for undergraduate students., 新しい学びのスタイルとして，アクティブ・ラーニングの必要性が最近いろいろなところで聞かれる．ここでは，アクティブ・ラーニングの一例として，教育学部の学部生(セカンドオーサー)と幾何の題材について，私が関わった発見的学習の事例を3つ紹介する．1つ目は，三角形の中心として少し変則的なものを探そうとする試みで，兵庫教育大学で濱中裕明氏が始めたものと聞いている．2つ目は，方べきの定理として円に関する結果として知られているものを2次曲線に関しても拡張してみようという試みで，中里仁謙氏の放物線の方べきの定理に刺激されてたものである．3つ目は，最近はやりのICTを用いての発見的学習として，Hervey点の再発見に関わるもので，発見に至った経緯や関連する知られていないであろう結果，放物線の接線によってできるいくつかの三角形の外心の配置について報告する．最後に大学生に対しての発見的学習の意味や問題点について考察する．

Here, we investigate a possible scenario of surface seawater evolution in the numerical simulations of surface plate motion driven by mantle dynamics, including thermo-chemical convection and water migration, from the early to present-day Earth to constrain the total amount of water in the planetary system. To assess the validity of two hypotheses of the total amount of water inferred from early planetary formation processes and mineral physics, we examine the model sensitivity to the total water in the planetary system (both surface and deep interior) up to 15 ocean masses. To explain the current size of the reservoir of surface seawater, the predictions based on the numerical simulations of hydrous mantle convection suggest that the total amount of water should range from 9 to 12 ocean masses. Incorporating the dense hydrous magnesium silicate (DHMS) with a recently discovered hydrous mineral at lower mantle pressures (phase H) indicates that the physical mechanism of the mantle water cycle would not be significantly influenced, but the water storage region would be expanded in addition to the mantle transition zone. The DHMS solubility field may have a limited impact on the partitioning of water in the Earth’s deep mantle.

To evaluate the content of nitrogen (N) fractions of agricultural soils in Java, Indonesia, in relation to soil type and land use, 46 surface soil samples, 23 from paddy and 23 from upland, were collected throughout Java to include various types of soils. Soil N was separated into four fractions according to form and availability: inorganic extractable nitrogen (Iex-N), fixed ammonium nitrogen (Ifix-N), organic mineralizable nitrogen (Omin-N) and organic stable nitrogen (Osta-N). The total-N content was determined by the dry combustion method. The Iex-N content was determined by extraction with a 2 mol L−1 potassium chloride (KCl) solution and the Ifix-N content by extraction with an hydrofluoric and hydrochloric acid (HF-HCl) solution after removal of organic-N. The Omin-N content was evaluated as the potentially mineralizable N based on a long-term incubation method. Pancreatic agenesis is a rare disease that causes neonatal diabetes mellitus and exocrine pancreatic insufficiency. We report the case of a very low birth weight infant with congenital diaphragmatic hernia, and congenital heart disease (ventricular septal defect and patent ductus arteriosus). Failure to gain weight, despite well-managed respiratory and heart failure, was improved by infusion of subcutaneous insulin, administration of pancreatic enzyme, and nutrition of medium-chain-triglyceride -enriched formula. This is the first case of pancreatic agenesis with both malformations where the patient is discharged from the hospital. Early diagnosis and adequate treatments to compensate pancreatic function may prevent mortality and improve growth.

Although it is well known that phytoavailability of radiocaesium is gradually lost after its deposition on the ground by fixation to soil minerals, the decreasing rates during early period after the deposition is not yet quantitatively evaluated. In this study, stable Cs was added to 5 types of soil, including Andosols, a sand-dune regosol and a smectic lowland soil, in a laboratory soil incubation experiment to assess the aging effect of radiocaesium. Aliquots of a soil sample were put into pots and incubated in an artificial climate chamber. Orchardgrass or red clover was cultivated for 28 d in soil pots containing one of the allophanic Andosol samples seven times during about 1200 d using new pots for each cultivation. The soil-to-plant transfer factors of Cs declined exponentially until about 100 d and were almost constant thereafter. The extractabilities of Cs by water and 1 M NH4OAc solution from this allophanic Andosol soil sample also decreased with time and their decreasing patterns were similar to that of the transfer factor. The temporal changes of extractabilities of Cs in other soil samples were also examined 6 times during about 600 d. Rate of decline for the extracted yield of the added Cs by 1 M NH4OAc varied widely among all the soil types. Two allophanic Andosol samples showed relatively higher extractabilities in comparison with the other soils throughout the incubation experiment, which may be attributable to the lower contribution of Cs specific sorption sites to total cation exchange capacity of the allophonic Andosol soil samples.

Cs accumulation by Pseudomonas fluorescens was examined under conditions of growth and resting to elucidate the interaction between Cs and bacteria. In the growth condition, P. fluorescens was cultured in citric acid media containing 50 ∝M CsCl and 0, 0.1, 1.0, and 10 mM K for 64 h at 30 ˚C. Adsorption and desorption behaviors of Cs were examined by the contact of the cells with 50 ∝M CsCl solution under the resting condition. Cs-accumulated P. fluorescens was exposed to a 1 M CH3COONH4 solution to examine the reversibility of Cs accumulation. In the growth condition, P. fluorescens did not accumulate Cs irrespective of the presence of K. In the resting condition, the cells quickly adsorbed approximately 5 ∝mol Cs/gcell dry-weight and subsequently released approximately 90% of the adsorbed Cs with 1 M CH3COONH4. The amount of Cs adsorption by cells of P. fluorescens varied with changing pH and ionic strength of the solution. These results indicate that Cs accumulation by P. fluorescens occurs mainly by reversible adsorption on the cell walls, but not by intracellular accumulation under nutrient conditions.

Background: Reticulated platelets (RPs) are newly synthesized platelets. Recently, an automatic method was established to detect RPs as a percentage of the immature platelet fraction (IPF%). Although, neonates often develop thrombocytopenia at some time during their hospitalization, the details of IPF% in neonates remain unclear. We, therefore, studied the relations between IPF% and other factors to gain a more detailed understanding of IPF% in neonates. Methods: The following clinical data were obtained from the medical records of 105 neonates who met our inclusion criteria: Gestational age, birth weight, IPF% and platelet count of neonatal peripheral blood at birth, and perinatal data. The subjects were divided into three groups: Group A, birth weight standard deviation score (SDS) ≥ −2 standard deviation (SD) and ≤ +2 SD; Group S, < −2 SD; and Group L, > +2 SD. Results: IPF% correlated negatively with platelet count at birth in the whole study population. IPF% was 2.8 ± 1.3% in term neonates, and IPF correlated negatively with gestational age and birth weight. Platelet count correlated positively with birth weight SDS in the whole study population and in Group S. IPF% correlated negatively with birth weight SDS in the whole study population and in Group S. In neonates with a platelet count below 25 × 104/μl, IPF% correlated negatively with platelet count. Among other neonates, however, IPF% remained almost constant. Conclusion: Monitoring of IPF% is useful for estimating the function of thrombocytopoiesis in neonates and preterm infants.

Trisomies 18 and 21 are genetic disorders in which cells possess an extra copy of each of the relevant chromosomes. Individuals with these disorders who survive birth generally have a shortened life expectancy. As telomeres are known to play an important role in the maintenance of genomic integrity by protecting the chromosomal ends, we conducted a study to determine whether there are differences in telomere length at birth between individuals with trisomy and diploidy, and between trisomic chromosomes and normal chromosomes. We examined samples of peripheral blood lymphocytes (PBLs) from 31 live neonates (diploidy: 10, trisomy 18: 10, trisomy 21: 11) and estimated the telomere length of each chromosome arm using Q-FISH. We observed that the telomeres of trisomic chromosomes were neither shorter nor longer than the mean telomere length of chromosomes as a whole among subjects with trisomies 18 and 21 (intra-cell comparison), and we were unable to conclude that there were differences in telomere length between 18 trisomy and diploid subjects, or between 21 trisomy and diploid subjects (inter-individual comparison). Although it has been reported that telomeres are shorter in older individuals with trisomy 21 and show accelerated telomere shortening with age, our data suggest that patients with trisomies 18 and 21 may have comparably sized telomeres. Therefore, it would be advisable for them to avoid lifestyle habits and characteristics such as obesity, cigarette smoking, chronic stress, and alcohol intake, which lead to marked telomere shortening.

It is difficult to use HbA(1c) as an indicator of glycaemic control in patients with neonatal diabetes mellitus (NDM) because of high levels of fetal haemoglobin (HbF) remaining in the blood. In this study, glycated albumin (GA), which is not affected by HbF, and HbA(1c) were compared to evaluate whether they reflect glycaemic control in patients with NDM.This study included five patients with NDM. Age at diagnosis was 38 ± 20 days. Insulin therapy was started in all patients, and levels of GA, HbA(1c) and HbF were measured monthly for 6 months. One-month average preprandial plasma glucose (aPPG) was calculated using self-monitoring of blood glucose.Plasma glucose and GA were elevated (29.7 ± 13.1 mmol/l [n = 5] and 33.3 ± 6.9% [n = 3], respectively) but HbA(1c) was within normal limits (5.4 ± 2.6% [35.5 ± 4.9 mmol/mol]; n = 4) at diagnosis. With diabetes treatment, aPPG (r = -0.565, p = 0.002), GA (r = -0.552, p = 0.003) and HbF (r = -0.855, p0.0001) decreased with age, whereas HbA(1c) increased (r = 0.449, p = 0.004). GA was strongly positively correlated with aPPG (r = 0.784, p0.0001), while HbA(1c) showed no correlation with aPPG (r = 0.221, p = 0.257) and was significantly inversely correlated with HbF (r = -0.539, p = 0.004).GA is a useful indicator of glycaemic control in patients with NDM, whereas HbA(1c) is influenced by age-related changes in HbF and does not accurately reflect glycaemic control.

We developed a system for bioconverting diverse compounds using P450s produced in Escherichia coli. Vectors for the expressing various P450 cDNAs quickly and easily in E. coli were developed by using several restriction enzyme sites. Three types of P450 (2C2, 2C29, and 2D22) were produced using these plasmids. Substrates were directly added to the incubation medium and metabolized. To obtain pure product from the medium, we first tried production of P450 in synthetic medium. The amount of another P450 2C43 produced in the synthetic medium was similar to the amount produced in Luria broth (LB) medium. Next, estradiol, a steroid, was added as a substrate, incubated, and the metabolite was extracted and analyzed by high-performance liquid chromatography. The metabolite extracted from synthetic medium was purer than that obtained from LB medium. Three P450s (2C29, 2C2, and 2A4) metabolized testosterone at different positions. P450 2C29 metabolized 7-ethoxycoumarin, androstendione, and dehydroepiandrosterone in this medium. P450s produced in the synthetic medium may be useful for producing various modified compounds for high-throughput screening.

A cDNA clone encoding methyl DNA binding domain-containing protein (bMBD2/3) was obtained by homology searches using a Bombyx mori fat body cDNA library. The cDNA encoded a polypeptide with 249 amino acids sharing 54% similarity with the methyl DNA binding protein from Drosophila melanogaster. To characterize the biochemical properties of bMBD2/3, the clone was expressed in Escherichia coli as His-tagged protein. The recombinant protein was purified to homogeneity using Ni-NTA superflow resin and heparin agarose. The protein showed specific methyl DNA binding activity and was phosphorylated by protein kinase in vitro. Immunoblotting using the purified antibody indicated that bMBD2/3 was expressed in almost all tissues. Using west-western blotting analysis, some proteins that interact with bMBD2/3 were identified in the brain. This is the first report that insect MBD is phosphorylated and is present in adult tissues. These results suggest that bMBD2/3 plays important roles in the DNA methylation-specific transcription of Bombyx mori. Abbreviation: EMSA Electro Phoretic Mobility Shift Assay
