Abstract This study focused on sustainable and novel development of concrete through treated and untreated plastic waste aggregates. In this study, the surface of Elec-waste coarse aggregates was treated with sulfuric acid, marble dust coating, silica fume coating, and alkaline bleach. Elec-waste aggregates were used in concrete by replacing natural coarse aggregates in amounts of 10%, 20%, and 30%. The 10%, 20%, and 30% Elec-waste replacement ratios resulted in the compressive strength reduction of 8.97%, 27.99%, and 42.01%, respectively. The same percentage of Elec-waste aggregates reduced the splitting tensile strength by 21.77%, 35.89%, and 47.89%, respectively. However, the slump was observed to increase by 41.4%, 52.1%, and 68.8%, respectively. All the considered treatments enhanced the performance of Elec-waste aggregate concrete (E-WAC). Overall, the best improvement in the performance of E-WAC was imparted by silica fume coating, irrespective of the quantity of Elec-waste aggregates. The present study proposed an expression to estimate the reduction of the compressive strength of E-WAC. It was demonstrated that the equation by JCI-08 predicted splitting tensile strength of E-WAC close to experimental results.

Recent research have highlighted the potential of hybrid confinement, combining high tensile strength fiber-reinforced polymers with large rupture strain confinement. This study presents experimental findings on 64 cylindrical and square-shaped specimens tested under axial compression, introducing a novel hybrid confinement method utilizing low-cost fiberglass chopped strand mat sheets and cotton ropes (COFS confinement). The experimental and analytical results yielded several key conclusions. Firstly, circular specimens exhibited significant peak strength increases in various subgroups, with enhancements ranging from 97.5 % to 285.5 %, and ultimate strain improvements ranging from 588.6 % to 1650.0 %. Similarly, square specimens under COFS confinement also demonstrated notable enhancements in ultimate strength and strain, with increases up to 244.7 % and 1083.0 %, respectively, particularly evident with higher levels of confinement. The influence of cross-sectional shape on compressive strength, strain, and energy dissipation was noted, with COFS confinement notably improving these factors for circular sections. Additionally, the study found that as the unconfined compressive strength increased, the enhancement in compressive strength, ultimate strain, and energy dissipation decreased. Moreover, the confinement ratio positively affected axial behavior improvement, with a proportional enhancement observed. However, the efficacy of the confinement ratio was influenced by cross-section type and plain concrete strength, emphasizing the need for considering these factors in COFS-based confinement design. Lastly, an analytical design-oriented model proposed for approximating stress vs. strain curves of COFS-confined concrete showed close agreement with experimental results, providing valuable insights for future design considerations.

The hybrid confinement system combines various fiber types within a single matrix, allowing for the adjustment of volumetric ratios to optimize confinement performance. Synthetic FRPs are more expensive and have a higher carbon footprint due to significant CO2 emissions during production. In response, this study presents an innovative hybrid confinement approach using two natural materials: cotton ropes and FSMS (CFS) to improve concrete strength and ductility. Specimens, standardized at 300 mm height and 150 mm diameter with longitudinal steel bars and stirrups, were divided into two groups based on CFS configurations. The stress-strain response of CFS-confined concrete displayed distinctive behavior: an initial parabolic phase leading to peak compressive stress (ultimate strength), followed by a linearly degrading phase. Across all subgroups, CFS confinement significantly enhanced ultimate strength and corresponding compressive strains, with Subgroup 2A achieving the highest improvements of 246 % in ultimate strength and 1477 % in strain. Moreover, the ductility gain was reported as high as 20 for CFS-confined concrete. A non-proportional enhancement in the compressive behavior was observed with the increase in confinement ratio. Predictive models were developed for the idealized two-branch response of CFS-confined concrete, encompassing expressions based on nonlinear regression for ultimate strength, corresponding strain, ultimate strain, and elastic modulus. Two existing models were modified to trach each branch of the response. Integrating these two adjusted models closely replicated the experimental compressive curves.

The precast industry offers slab panels of different geometries according to the field conditions. These slab panels are popular in temporary constructions and beneficial in sustainability but have some financial limitations and local constraints. For a long time, the construction industry has used the arch action method, which restricts the stresses to the compression zone in concrete members and develops the required load-carrying capacity. For the same motives, industrial buildings have preferred semi-circular precast roofs, but the morphology was not suitable. For the proposed slab in this research, firstly, the typical industrial precast slab panel was doubled in width to minimize the time and efforts required for its casting, curing, and placement. Secondly, that doubled-in-width slab was provided with the arch action to confine the stresses to compression and benefit from the section entirely. Lastly, the top of the slab was kept flat to take advantage of the roof space. All these changes aimed for structural stability, reduced material's weight, improved load-carrying capacity, appropriate mobilization, and financial viability. A numerical approach and practical testing were adopted using the finite element modeling software, ABAQUS, to analyze load–deflection responses of both slabs through the concrete damage plasticity model. The proposed slab exhibited better performance as its capacity enhanced by about 1.5 times that of a typical slab. Although the volume of the material in the proposed slab increased slightly from 0.040 m3 to 0.045 m3, the reductions in joint filler materials, reinforcements, and efforts required for mixing and lifting machinery compensated for this increase significantly. Hence, the slab can be recommended for the industry to save the costs while taking heavier loads efficiently.

Recent earthquakes have highlighted the need to strengthen existing structures with substandard designs. NFRPs provide a sustainable, cost-effective alternative for strengthening, but accurately predicting their performance remains a challenge. This study investigates the use of machine learning algorithms for predicting the compressive strength concrete specimens confined with various NFRPs. Four algorithms were employed: decision tree, random forest, neural network, and gradient boosting regressor. A diverse dataset encompassing various geometries, material properties, and confinement configurations was used to train and evaluate the models. Gradient boosting regressor (GBR) achieved the highest performance, with an average R-squared value of 0.94 and low mean absolute error (MAE) and root mean squared error (RMSE) during training and k-fold cross-validation. Neural network and random forest also demonstrated satisfactory performance, with average R-squared values of 0.88 and 0.86, respectively, during cross-validation. These results suggest that machine learning holds promise for predicting the compressive strength of concrete confined with NFRPs. GBR offers the most accurate predictions, making it a valuable tool for engineers seeking to optimize the design and performance of strengthened structures using sustainable materials.

This paper presents a comprehensive investigation into the behavior of concrete confined with hybrid Basalt and Chopped Strand Mat (B-CSM) fibers. The newly proposed B-CSM confinement technique substantially enhances the brittle compressive stress-strain behavior, leading to a noteworthy increase in peak strength (approximately 90%) and ultimate strain (approximately 461 %). The efficiency of B-CSM confinement is affected by the strength of plain concrete, with lower-strength specimens indicating a more pronounced enhancement. The performance of existing analytical models for FRP confinement in predicting ultimate strength and strain in B-CSM confined concrete is assessed, highlighting the need for tailored models. Regression-based equations are proposed for characteristic points along the stress-strain curve, enabling accurate prediction of material behavior. The predicted stress-strain curves exhibit a high level of agreement with experimental results. These findings provide valuable insights for the design and application of B-CSM confinement techniques in structural engineering, facilitating improved performance and ductility of concrete structures under compressive loading conditions.

The case report describes a patient with stage IV breast cancer which metastasized to the lungs. The patient's initial computed tomography (CT) scan revealed a malignant lesion in the upper outer quadrant of the left breast and multiple pulmonary nodules, suggesting pulmonary metastasis. After starting palliative chemotherapy with intravenous paclitaxel and subcutaneous injections of Herceptin, a follow-up CT scan 3 months after the initiation of treatment showed the disappearance of metastasis, and her cancer regressed to stage II breast cancer that could be surgically resected. This case report highlights the importance of timely and appropriate palliative treatment measures, which can lead to unexpected outcomes, such as the regression of metastatic lesions and the possibility of curative treatment in such advanced cancer.

Abstract This study presents an experimental and finite element analysis of reinforced concrete beams with solid, hollow, prismatic, or non-prismatic sections. In the first part, a total of six beams were tested under four-point monotonic bending. The test matrix was designed to provide a comparison of structural behavior between prismatic solid and hollow section beams, prismatic solid and non-prismatic solid section beams, and prismatic hollow and non-prismatic hollow section beams. The intensity of shear was maximum in the case of prismatic section beams. The inclusion of a tapered section lowered the demand for shear. In the second part, Nonlinear Finite Element Modeling was performed by using ATENA. The adopted modeling strategy resulted in close agreement with experimental crack patterns at ultimate failure. However, the ultimate failure loads predicted by nonlinear modeling were generally higher than their corresponding experimental results. Whereas in the last part, the developed models were further extended to investigate the effect of the strength of concrete and ratio of longitudinal steel bars on the ultimate load-carrying capacity and cracking behavior of the reinforced concrete beams with solid, hollow, prismatic, or non-prismatic sections. The ultimate loads for each beam predicted by the model were found to be in close agreement with experimental results. Nonlinear modeling was further extended to assess the effects of concrete strength and longitudinal reinforcement ratio on failure patterns and ultimate loads. The parametric study involved beams reinforced with glass fiber-reinforced polymer (GFRP) bars against shear and flexural failure. In terms of ultimate load capacities, diagonal cracking, and flexural cracking, beams strengthened with GFRP bars demonstrated comparable performance to the beams strengthened with steel bars.

This study presents an innovative approach to enhance the flexural capacity of cement-clay interlocking (CCI) brick walls using ferrocement jackets with an expanded wire mesh. The investigation involved 15 CCI brick walls subjected to flexural load testing, exploring various mesh sizes and anchor configurations on single and dual sides. The results revealed that while the primary failure mode involved dry joint opening and subsequent ferrocement debonding in the strengthened walls, effective anchorage mechanisms delayed dry joint opening until debonding occurred. The reinforced walls exhibited improved peak load capacity, notably in dual-sided reinforcement cases. With plastering on both sides and supplemented with anchors, an improvement of up to 221% in flexural capacity was obtained. The study emphasizes the influence of wire mesh size on anchorage effectiveness and its correlation with debonding susceptibility. Additionally, the moment-curvature analysis demonstrated resilient, ductile responses in load-bearing characteristics, with occasional abrupt declines due to ferrocement jacket debonding. Unlike control walls, the strengthened walls exhibited strains exceeding yield thresholds, indicating enhanced structural performance.

The utilization of recycled waste in the production of new concrete raises significant concerns regarding the quality of the resulting concrete. One prominent drawback of recycled aggregate concrete (RAC) is its inferior mechanical properties compared to natural aggregate concrete. In this study, we examine the impact of hemp fiber rope (HFR) confinement on improving the compressive stress-strain behavior of RAC, specifically using recycled brick aggregates from fired-clay solid bricks (RAC-FCSB) as a partial substitute for natural coarse aggregates. Furthermore, we explore and compare the effectiveness of HFR confinement in the form of strips versus fully wrapped confinement. To conduct the experiments, a comprehensive framework was developed, involving a total of 32 cylindrical specimens. The parameters of interest included the configuration of HFR confinement (strips or full wrapping), the number of HFR layers, and the inherent strength of the concrete. The results indicate that HFR strips can significantly enhance the compressive stress-strain response. However, their performance falls short when compared to the fully wrapped HFR confinement. Nonetheless, HFR strips were able to enhance the peak compressive stress and strain of RAC-FCSB up to 204% and 190%, respectively. The second part of the study investigated the performance of existing models of HFR confinement in predicting the peak compressive stress and strain of RAC-FCSB. The deficiencies in existing models were highlighted, and new models based on non-linear regression analysis were proposed to accurately predict the peak compressive stress and strain of HFR-confined RAC-FCSB.

Each year, an enormous amount of construction waste is produced worldwide. The reuse of construction waste in construction works is a sustainable solution. The present research work utilized recycled brick aggregates in the production of concrete. The resulting concrete exhibited substandard splitting tensile, flexural, and compressive properties. Steel fibers were used to improve these substandard properties of recycled brick aggregate concrete. The volume fractions of 1%, 2%, and 3% for steel fibers were mixed in concrete, whereas recycled brick aggregates were obtained from solid fired-clay bricks, hollow fired-clay bricks, and cement–clay interlocking bricks. The compressive strength was enhanced by up to 35.53% and 66.67% for natural and recycled brick aggregate concrete, respectively. Strengthened flexural specimens demonstrated up to 8765.69% increase in the energy dissipation. Specimens strengthened with steel fibers showed substantially improved splitting tensile, flexural, and compressive responses. Separate equations were proposed to predict the peak compressive strength, strain at peak compressive strength, elastic modulus, and post-peak modulus of recycled brick aggregate concrete. The proposed regression equations were utilized in combination with an existing compressive stress–strain model. A close agreement was observed between experimental and predicted compressive stress–strain curves of recycled brick aggregate concrete.

Flexible pavements are mostly affected by meteorological factors in addition to traffic loads, which results in premature pavement failures like rutting and moisture-induced damage. This study focuses on the impacts of adding various contents of nano-silica (NS), i.e., 2%, 4%, 6%, and 8% (percentage weight of asphalt), along with a constant value of 4.5% styrene-butadiene-styrene (SBS). To assess the effectiveness of modified and unmodified mixtures, the indirect tensile strength (ITS) test, resilient modulus (MR) test, and wheel tracking test were conducted. The MR test was performed at dual temperature values, i.e., 25 °C and 40 °C, and demonstrated different metrological conditions in this region. The tensile strength ratio was used to estimate the mitigation of water losses in hot mix asphalt (HMA) mixtures (specimens) utilizing ITS test results of the conditioned and unconditioned specimens. Moreover, a model was developed for the rutting potential of the modified specimens using multi expression programming (MEP), a sophisticated technique that employs experimental data and suggests an equation for different input variables. The results indicated that the addition of NS to SBS-modified bitumen enhanced different mechanical properties of the specimens, including the stiffness and moisture and rutting resistances. The temperature had adverse effects on the stiffness of the specimens, while the modifiers had a direct relationship with the stiffness. The two-way factorial method justified the effect of the temperature and modifiers on MR with 95% precision, while the MEP model for rutting showed an R2 value of >0.95, which revealed a good relationship between the experimental and predicted data. Furthermore, NS and SBS had a good impact on the mechanical properties of the HMA specimens.

This study investigated the influence of CFRP composite wrapping techniques on the load–deflection and strain relationships of non-prismatic RC beams. A total of twelve non-prismatic beams with and without openings were tested in the present study. The length of the non-prismatic section was also varied to assess the effect on the behavior and load capacity of non-prismatic beams. The strengthening of beams was performed by using carbon fiber-reinforced polymer (CFRP) composites in the form of individual strips or full wraps. The linear variable differential transducers and strain gauges were installed at the steel bars to observe the load–deflection and strain responses of non-prismatic RC beams, respectively. The cracking behavior of unstrengthened beams was accompanied by excessive flexural and shear cracks. The influence of CFRP strips and full wraps was primarily observed in solid section beams without shear cracks, resulting in enhanced performance. In contrast, hollow section strengthened beams exhibited minor shear cracks alongside the primary flexural cracks within the constant moment region. The absence of shear cracks was reflected in the load–deflection curves of strengthened beams, which demonstrated a ductile behavior. The strengthened beams demonstrated 40% to 70% higher peak loads than control beams, whereas the ultimate deflection was increased up to 524.87% compared to that of the control beams. The improvement in the peak load was more prominent as the length of the non-prismatic section increased. A better improvement in ductility was achieved for the case of CFRP strips in the case of short non-prismatic lengths, whereas the efficiency of CFRP strips was reduced as the length of the non-prismatic section increased. Moreover, the load–strain capacity of CFRP-strengthened non-prismatic RC beams was higher than the control beams.

Many masonry structures are constructed with cement clay interlocking brick (CCIB) due to its added benefits. Recent research has demonstrated the vulnerability of brick masonry walls against seismic loading. Various strengthening materials and techniques are extensively used to improve the structural behavior of brick walls. Carbon fiber-reinforced polymer (CFRP) composites are the most popular strengthening material due to their advantages of easy application, lightweight qualities, and superior tensile strength. The current research work aimed to explore the cost-effective solutions and feasibility of CFRP composite-based strengthening techniques to improve the load-bearing capacity of CCIB walls. Various configurations and combinations of strengthening materials were investigated to customize the cost of repair and strengthening. The experimental results indicated that CFRP composites in combination with cement-sand (CS) mortar are an efficient strengthening material to enhance the strength and ultimate deflection of CCIB walls. The ultimate load-bearing capacity and axial deformation of the strengthened CCIB wall (using two layers of CFRP strips and CS mortar of 10 mm thickness) remained 171% and 190% larger than the unstrengthened CCIB wall. The conclusions of this study are expected to enhance the seismic performance of masonry buildings in developing countries. It should be noted that due to the reduced number of tested specimens, the results to be assumed as general considerations need a wider experimental campaign and a large numbers of tests for each strengthening typology.

Worldwide, rheumatoid arthritis is a major cause of morbidity and mortality. In recent years, the domain of inorganic medicinal chemistry has shown more interest in metal-based drugs owing to their beneficial pharmacological activities. Since then, many useful anti-cancer metalbased drugs have been introduced. However, less attention has been paid to improving inflammatory drugs based on metal complexes. The objective of this review is to recapitulate previously published studies that concentrated on metal-based drugs used for the treatment of arthritis. It is anticipated that the current compilation of earlier studies will be helpful for the researcher to find the data and areas for future studies. The literature survey was done without any time limit using the help of various electronic databases, including ScienceDirect, SpringerLink, Web of Science, PubMed, and the Cochrane Library. Articles published in the English language are used for the current review. Only 20 studies were found to be pertinent to the present review. Metal complexes of non-steroidal anti-inflammatory drugs have been the subject of extensive research. To date, metal complexes of drugs such as aspirin, ibuprofen, anthranilic acid, methotrexate, indomethacin, and aceclofenac have been studied and evaluated for their possible antiarthritic activity. Gold metal complexes showed better efficacy and have been used in clinical practice as well. Due to their advantageous therapeutic properties, metals and their complexes are becoming more and more important today. Pakistani population is unique for the study of recessive genetic diseases due to a higher rate of consanguinity and Hearing Loss (HL) is one of them that affects more than 466 million people worldwide; will increase to 900 million by 2050. GJB2 mutation is one of the main causes of hearing loss in different populations, including Pakistan, that encodes a gap junction protein involved in the homeostasis of the inner ear by recycling potassium ion. This research aimed to find out mutations in the GJB2 gene and its protein structure. Both control and patient samples were collected from Gilgit-Baltistan for DNA isolation and PCR was done by using a specific primer while sequencing was done by Sanger sequencing. Mutations were detected by Mutation Surveyor and BLAST. Protein structures of both control and mutated samples were constructed by PHYRE2 and visualized by the software PyMOL. The detected mutations were 380G>A (R127H), 457G>A (V153I), 36T>C (G12G), 496C>T (L166L), 650 delA and 79G>A (V27I). This study revealed that the prevalence of GJB2 mutations in hearing-impaired patients of Gilgit-Baltistan is 11.66 %. The mutation rate is higher as compared to other populations of Pakistan. Objective: To look for the frequency of thyroid dysfunction in heart failure patients with reduced ejection fraction at Medicine/Cardiology Department. Study Design: Cross-sectional study. Place and Duration of Study: Combined Military Hospital Sialkot, from Nov 2019 to Apr 2020. Methodology: The sample population comprised of patients diagnosed with the heart failure and ejection fraction of 40% at Medicine/Cardiology unit. Patients underwent thyroid functions test from the laboratory of our hospital and were diagnosed as hypothyroid, hyperthyroid or euthyroid by consultant cardiologist and medical specialist. Age, gender, body mass index and New York heart association (NYHA) class were associated with thyroid dysfunction among the study participants. Results: Out of 110 patients with heart failure and low ejection fraction who underwent thyroid function testing during the study period, 92 (83.6%) patients were euthyroid, 14 (12.7%) had thyroid profile parameters showing hypothyroidism, while 04 (3.6%) had laboratory findings of hyperthyroidism. After chi-square, we found that patients with New York heart association class III and IV and high body mass index had a statistically significant relationship with thyroid dysfunction (pvalue 0.017 and <0.001 respectively) among the patients of heart failure with low ejection fraction. Conclusion: Thyroid dysfunction, especially hypothyroidism, emerged as a common finding among heart failure patients with low ejection fraction. Patients with New York heart association class III and IV and high body mass index should be considered high-risk and may be screened for thyroid problems if present with heart failure. Microscopic, phytochemical and pharmacological profiles are required for correct identification of a plant material to ensure consistent efficacy and safety. But such data are not available for the leaf of an important medicinal plant, Zizyphus oxyphylla Edgew. (Family: Rhamnaceae). Therefore, the current study aimed to investigate leaves of the plant for microscopic, phytochemical and antibacterial studies. Powdered material was subjected to microscopy, proximate analyses and estimation of total primary metabolites. Then, different types of extracts prepared using various solvents in order of increasing polarity were screened for antibacterial activity against seven standard strains. The most active extract was hydrolyzed and aglycone enriched fraction was extracted and screened for antibacterial activity. The powder microscopy indicated the presence of vascular bundles filled with cuboidal calcium oxalate crystals, anisocytic stomata and xylary vessels with reticulate and scalariform thickenings. Proximate features and primary metabolites provided characteristic identifying patterns. The most active extract (methanol) upon acidic hydrolysis exhibited higher activity against B. bronchiseptica (26.01±0.01 mm), S. aureus (26.00±0.00 mm), P. aeruginosa (24.03±0.02 mm) and M. luteus (24.02± 0.00 mm). The results of the current study provide identifying microscopic and phytochemical profiles that may be useful for correct identification of leaves of the plant. Aglycone enriched extract is having remarkable antibacterial activity hence may be used for activity-guided isolation. [ABSTRACT FROM AUTHOR]

The present study aimed to investigate the enzymetic, non-enzymetic toxicity and antioxidant potential of a drug candidate 5-Benzyl-1,3,4-Oxadiazole-2-Thiol(OXPA) using computational tools and in vivo models. The binding pattern of it, with different toxicity/oxidative enzymes was predicted using software pkCSM, Protox- II, LAZAR, Mcule 1-Click Docking 3D-Ligand binding Site and best score obtained used as an evaluating criterion. After acute oral toxicity, in vivo. antioxidant and hepato protective activity was investigated on male wistar rats, segregated into four groups as control (NS), toxic (INH-RIF), standard (Silymerin) and sample (OXPA, 100mg/Kg) for 21days. Level of antioxidant enzymes / histopathology and serum biochemical parameters in liver and blood of treated rats was assessed by using scientific tools. In silico study reveal no profound toxicity parameters however, LD50 found to be 560mg/Kg while in vivo study declared it safe till 1000mg/Kg, as having no toxicity symptoms. Molecular interaction score with GTH reductase, s-transferase and significant in vivo antioxidant effect on catalase, SOD, TBARS enzymes and histopathological assessment, declare OXPA a good antioxidant having significant (P< 0.05) hepato protective activity. Results of in silico, in vivo studies declare the propensity of 5-Benzyl-1, 3, 4-oxadiazole-2-thiol as potential antioxidant, for further investigations as a drug. [ABSTRACT FROM AUTHOR]

Objective: To compare the effect of SGLT-2 inhibitors with DPP-4 inhibitors as the add on therapy to Metformin on HBA1c level in type 2 diabetes patients in Pakistani population. Study Design: Quasi-experimental study. Place and Duration of Study: Combined Military Hospital Sialkot, Pakistan from Nov to Apr 2020. Methodology: The study population comprised of 300 type-II diabetes patients with poorly controlled diabetes on Metformin managed at Combined Military Hospital Sialkot, Pakistan. Patients were divided into two groups via lottery method. Group-A received the SGLT-2 inhibitors at standard dose while group-B received the DPP-4 inhibitors at the usual standard does as recommended by the consultant medical specialist. Values of glycosylated hemoglobin were compared three months after the start of study. Results: Mean age of the patients was 44.31 ± 5.841 years. Mean duration of type 2 diabetes mellitus in the study participants was 4.18 ± 6.369 years. HBA1c after three months of treatment in the SGLT-2 inhibitors-group was 7.01 ± 0.14 mmol/L while in the DPP-4 inhibitors-group was 7.89 ± 0.99 mmol/L. Difference between the two groups was statistically significant (pvalue <0.01). The result showed SGLT-2 inhibitors as the better option for add on therapy to Metformin as compared to DPP-4 inhibitors. Conclusion: SGLT-2 inhibitors emerged as better option to lower the HBA1c level as compared to DPP-4 inhibitors as the add on therapy to Metformin as there was statistically significant difference in HBA1c levels after three months of therapy in both the groups. The study aimed at simultaneous quantification of sumatriptan succinate (SUM) and prochlorperazine maleate (PCP) in an orodispersible film using two validated spectroscopic methods viz. simultaneous equation (Method I) and the Q-absorption ratio (Method II). The Method I involved measurement of absorbances at λmax of both drugs while in Method II, absorbances were measured at isosbestic wavelength and λmax of one of the two components. Method validation were accomplished as per the ICH guidelines. A 1:1 mixture of the drugs and an orodispersible film (ODF) containing these drugs were assayed by both methods. The absorbance data of SUM and PCP in both methods were linear at respective wavelengths with correlation coefficient values >0.995. Both methods were precise as % RSD in repeatability, interday and intraday precision was less than 2. The estimation of SUM and PCP from the film dosage form by method I was104.74% and 98.34% and by method II was 103.45% and 98.85%, respectively, with a standard deviation <2. The study concluded that both the methods were simple, reliable and robust and can be applied successfully for the simultaneous quantification of SUM and PCP in mixture and orodispersible film dosage form. This study aimed to fabricate and characterize polymeric microneedle patches for rapid and non-invasive administration of enoxaparin across skin layers. The patches comprising of PVA, sorbitol and enoxaparin sodium were prepared by employing micromolding technique. Formulated patches were characterized physicochemically by folding endurance, dimensional analysis and swelling study, morphologically by optical and scanning electron microscopy and thermally by thermogravimetric analysis. Moreover, performance efficiency of prepared polymeric device was analyzed by in-vitro drug release study and piercing ability. Prepared patches showed appropriate dimensions and folding endurance (i.e., ~1100) indicating satisfactory integrity of polymeric device. Patches exhibited appropriately distanced needles with pointed tips in optical and scanning electron microscopy analysis. Thermogravimetric analysis proved thermal stability of formulation ingredients and prepared patches. Swelling percentage was >110 % suggesting that prepared formulation would allow penetration of physiological fluids in its polymeric network. Maximum (~89%) drug was released within ~2 hours during in-vitro release study. In-vitro piercing ability experiments suggested that prepared patches successfully breached skin barrier stratum corneum. It is concluded that prepared microneedle device can serve as a potential alternative of currently employed invasive parenteral route for rapid and efficient administration of enoxaparin sodium in the systemic circulation. The modern trend of research is highly focused on finding new bioactive molecules from medicinal plants. As a functional bicyclic monoterpene, Bornyl acetate (BA) has displayed antioxidant and anti-inflammatory properties in different types of cells and tissues. The purpose of this research was to evaluate the probable hypotensive effect of BA, an underlying mechanism(s) backboned by in-silico studies. Mean arterial pressure and heart rate were recorded via invasive blood pressure measuring technique in normotensive Sprague-Dawley rats following the administration of BA (1-80mg/kg). Docking studies were carried out with various targets involved in the pathophysiology of hypertension.RO5 and ADMET properties were also evaluated. In the current study dose-dependent reduction in systolic, diastolic and mean arterial pressure was observed. Pretreatment with atropine and captopril significantly (p<0.001) reduced the hypotensive effect produced by BA. On the other hand docking studies showed pronounced interactions with M2 mAch receptor in an agonistic way and ACE protein in an antagonistic way. BA justified all cut-off limits of RO5 and had an acceptable predicted computational toxicity profile. Results postulate that dose-dependent hypotensive effect of BA is mediated through the muscarinic pathway and ACE inhibitory activity corresponding well with findings of in-silico studies. There is a long history of natural products for the treatment of infections and diseases. The objective of present study was to investigate the organoleptic, microscopic, physico-chemical, phytochemical, antidiarrheal and antidiabetic potential of leaf, flowering bud and stem bark of Moringa oleifera L. Macroscopic, microscopic, physicochemical parameters and phytochemical screening were carried out. Diarrhea was induced with castor oil (10ml/kg), verapamil (3, 10 and 30mg/kg) were used as standard antidiarrheal drug and extract of Moringa oleifera at (100, 300 and 1000mg/kg) was used for treatment. Alpha glucosidase inhibitory assay was carried out by using acarbose (0.5mM) and extracts (5.0 mg/Ml). Diabetes was induced by alloxan (150mg/kg), while glibenclamide (10mg/kg) was used as standard drug, and extracts (at the doses of 500mg/kg) were used to determine the antidiabetic activity. Results showed the presence of primary and secondary metabolites, treatment at the dose of 1.0g/kg of leaf, flowering bud and stem bark showed 94 ±2.527, 85.42±5.460 and 84.58±6.138% protection respectively whereas verapamil (10mg/kg) showed 94.84±3.27% protection. Alpha glucosidase inhibition of stem bark (0.5mg/ml) was 95.43±1.47 and flowering bud 94.78±1.25 whereas acarbose (5mM) inhibition was 92.23±0.14%. Stem bark and flowering bud extract (500mg/kg) decreases the blood glucose level from 388.5±35.83 to 226.3±47.10 and 322.5±48.35 to 173.8±29.5 respectively whereas glibenclamide (10 mg/kg) decreases the blood glucose level from 320.7±22.9 to 146.3±17.7 and increases the body weight of the experimental animal. This study aims at the development of a validated reverse phase-high performance liquid chromatography (RP-HPLC) method for the simultaneous quantification of levofloxacin (LVX) and clarithromycin (CAM). Eclipse X DB C18 column (5 µm, 4.6 × 250 mm) was used as a stationary phase, and monobasic potassium phosphate (KH2PO4) buffer (0.035 M): acetonitrile (75: 25) was used as an isocratic mobile phase. The pH of the mobile phase was adjusted to 4.5 with the help of diluted orthophosphoric acid. The diode array detector (DAD) was operated at 205 nm and 294 nm for the detection of CAM and LVX, respectively. Limit of detection (LOD) values for CAM and LVX were 1.22 µg/mL and 0.79 µg/mL, respectively. Limit of quantification (LOQ) values for CAM and LVX were 4.08 µg/mL and 2.64 µg/mL, respectively. The method was precise as % relative standard deviation (% RSD) values were less than 2% for all types of precision. Objective: To establish the correlation between glycated hemoglobin (HbA1c) and lipid profile in patients with type 2 diabetes mellitus (T2DM). Study Design: Correlation study. Place and Duration of Study: Pak Emirates Military Hospital Rawalpindi (PEMH), Medical Outpatient department (OPD), from Nov 2017 to May 2018. Methodology: A total of 160 patients with type 2 diabetes mellitus of both genders, 30-70 years of age presenting in OPD were included in study, while those with T1DM, CKD stage 3 and above, infection or diabetic foot, any endocrine disease and female patients with pregnancy were excluded from the study. The blood samples were taken for blood sugar random (BSR), blood sugar fasting (BSF), haemoglobin A1c (HbA1c), lipid profile and results were collected next day. The Pearson correlation test was used to analyze correlations. Results: Among 160 patients the mean age was 57.28 ± 10.13 years, 81 (50.6%) were males while 79 (49.4%) were females. The 34 (21.20%) patients had good, while 125 (78.80%) had poor glycaemic control using haemoglobin A1c of 7% as a cut off. A positive and statistically significant correlation was observed between haemoglobin A1c and total cholesterol (r=0.233, p<0.01), triglycerides (r=0.172, p<0.05), and a correlation which was positive but statistically not significant was observed between haemoglobin A1c and LDL-C (r=0.105, p=0.260) while a statistically insignificant negative correlation was observed between haemoglobin A1c and HDL-C (r=-0.041, p=0.652). Conclusion: A significant positive correlation was found between haemoglobin A1c and total cholesterol and triglycerides. Haemoglobin A1c can also be used as a biomarker to predict dyslipidemias in patients with type 2 diabetes mellitus. [ABSTRACT FROM AUTHOR]

Purpose: To evaluate 5-Benzyl-1,3,4-oxadiazole-2-thiol (OXPA) for antidiabetic and antioxidant properties. Methods: Antidiabetic activity was evaluated using three in vitro models, glucose uptake by yeast cells, alpha amylase inhibition assay and hemoglobin glycosylation inhibition assays. Antioxidant potential was determined by DPPH radical scavenging, reducing power and lipid peroxidation assays. Results: OXPA showed antidiabetic activity in all the three models. The activity of the compound was comparable with that of metronidazole in glucose uptake by yeast cells, but the alpha amylase inhibition activity of the compound was slightly lower than that of acarbose, whereas the hemoglobin glycosylation inhibition activity of the compound was higher than that of vitamin E. DPPH free radical and hydrogen peroxide scavenging activity of the compound was comparable with that of vitamin C. In reducing power assay, the activity of the compound was lower than that of vitamin C (p > 0.05). Conclusion: The results of antidiabetic and antioxidant activity indicate that OXPA may be a drugcandidate for treating both diabetes and its associated oxidative stress. [ABSTRACT FROM AUTHOR]
