Historical and ethnographic sources depict use of portable braced shaft weapons, or pikes, in megafauna hunting and defense during Late Holocene millennia in North and South America, Africa, Eurasia and Southeast Asia. Given the predominance of megafauna in Late Pleistocene North America during the centuries when Clovis points appeared and spread across much of the continent (13,050-12,650 cal BP), braced weapons may have been used in hunting of megaherbivores and defense against megacarnivores. Drawing from historical examples of pike use against lions, jaguars, boars, grizzlies, carabao and warhorses we consider the possibility of a fluted lithic pike. Associated osseous rods have been problematic as Clovis foreshafts due to the bevel angle and the apparent weakness of the splint haft when great strength is needed for deep penetration in megafauna hunting. However our review of Late Holocene pike use in megafauna encounters indicates the sharp tip becomes less important after hide or armor has been pierced because compression is sustained. Thus, foreshaft collapse after hide entry may not limit but rather increase the efficacy of the braced weapon. We conduct preliminary static experiments to model a fluted pike that adjusts during compression such that haft collapse and point detachment (when point jams on impact with bone) preserve the fluted biface, beveled rod and wooden mainshaft tip. In addition to Clovis point attributes and association with osseous rods, potential archaeological correlates of Clovis pike use include the high frequency of Clovis point isolates and concentrations of complete points with unbutchered mammoth remains at sites such as Naco in Arizona.

IntroductionEarly relapse in Crohn's disease (CD) is associated with a more severe disease course. The microbiome plays a crucial role, yet strategies targeting the microbiome are underrepresented in current guidelines. We hypothesise that early manipulation of the microbiome will improve clinical response to standard-of-care (SOC) induction therapy in patients with a relapse-associated microbiome profile. Many students, especially English language learners (ELLs), struggle when they write. This study examined the various writing strategies 3 Spanish-speaking bilingual student teachers used to help their elementary school ELL students write. This was a case study that looked at how these student teachers used their primary language, among other writing methods, to help their ELLs access writing strategies so these students could write effective English compositions. The authors used the interviews of student teachers, their lesson plans, and reflective journals to identify the instructional methods these student teachers used with their students. These methods included strategies such as helping their ELL students write their ideas in Spanish and English and using Spanish and English cognates to build students' word banks. The knowledge from this study is important because it showed how ELLs may benefit when their teachers use the students' primary language to help them write English compositions.

We present BVRI and unfiltered (Clear) light curves of 70 stripped-envelope supernovae (SESNe), observed between 2003 and 2020, from the Lick Observatory Supernova Search follow-up program. Our SESN sample consists of 19 spectroscopically normal SNe Ib, 2 peculiar SNe Ib, six SNe Ibn, 14 normal SNe Ic, 1 peculiar SN Ic, 10 SNe Ic-BL, 15 SNe IIb, 1 ambiguous SN IIb/Ib/c, and 2 superluminous SNe. Our follow-up photometry has (on a per-SN basis) a mean coverage of 81 photometric points (median of 58 points) and a mean cadence of 3.6 d (median of 1.2 d). From our full sample, a subset of 38 SNe have pre-maximum coverage in at least one passband, allowing for the peak brightness of each SN in this subset to be quantitatively determined. We describe our data collection and processing techniques, with emphasis toward our automated photometry pipeline, from which we derive publicly available data products to enable and encourage further study by the community. Using these data products, we derive host-galaxy extinction values through the empirical colour evolution relationship and, for the first time, produce accurate rise-time measurements for a large sample of SESNe in both optical and infrared passbands. By modelling multiband light curves, we find that SNe Ic tend to have lower ejecta masses and lower ejecta velocities than SNe Ib and IIb, but higher 56Ni masses.

Nitrate uptake by plants primarily relies on two gene families: Nitrate transporter 1/peptide transporter (NPF) and Nitrate transporter 2 (NRT2). Here, we extensively characterized the NPF and NRT2 families in the durum wheat genome, revealing 211 NPF and 20 NRT2 genes. The two families share many Cis Regulatory Elements (CREs) and Transcription Factor binding sites, highlighting a partially overlapping regulatory system and suggesting a coordinated response for nitrate transport and utilization. Analyzing RNA-seq data from 9 tissues and 20 cultivars, we explored expression profiles and co-expression relationships of both gene families. We observed a strong correlation between nucleotide variation and gene expression within the NRT2 gene family, implicating a shared selection mechanism operating on both coding and regulatory regions. Furthermore, NPF genes showed highly tissue-specific expression profiles, while NRT2s were mainly divided in two co-expression modules, one expressed in roots (NAR2/NRT3 dependent) and the other induced in anthers and/ovaries during maturation. Our evidences confirmed that the majority of these genes were retained after small-scale duplication events, suggesting a neo- or sub-functionalization of many NPFs and NRT2s. Altogether, these findings indicate that the expansion of these gene families in durum wheat could provide valuable genetic variability useful to identify NUE-related and candidate genes for future breeding programs in the context of low-impact and sustainable agriculture.

Abstract The salinity of soil is a relevant environmental problem around the world, with climate change raising its relevance, particularly in arid and semiarid areas. Arbuscular Mycorrhizal Fungi (AMF) positively affect plant growth and health by mitigating biotic and abiotic stresses, including salt stress. The mechanisms through which these benefits manifest are, however, still unclear. This work aimed to identify key genes involved in the response to salt stress induced by AMF using RNA-Seq analysis on durum wheat (Triticum turgidum L. subsp. durum Desf. Husn.). Five hundred sixty-three differentially expressed genes (DEGs), many of which involved in pathways related to plant stress responses, were identified. The expression of genes involved in trehalose metabolism, RNA processing, vesicle trafficking, cell wall organization, and signal transduction was significantly enhanced by the AMF symbiosis. A downregulation of genes involved in both enzymatic and non-enzymatic oxidative stress responses as well as amino acids, lipids, and carbohydrates metabolisms was also detected, suggesting a lower oxidative stress condition in the AMF inoculated plants. Interestingly, many transcription factor families, including WRKY, NAC, and MYB, already known for their key role in plant abiotic stress response, were found differentially expressed between treatments. This study provides valuable insights on AMF-induced gene expression modulation and the beneficial effects of plant-AMF interaction in durum wheat under salt stress.

Background PD-1 checkpoint blockade therapy (CBT) has greatly benefited patients with select solid tumors and lymphomas but has limited efficacy against diffuse large B-cell lymphoma (DLBCL). Because numerous inhibitory checkpoint receptors have been implicated in driving tumor-specific T cell dysfunction, we hypothesized that combinatorial CBT would enhance the activity of anti-PD-1-based therapy in DLBCL. T cell immunoglobulin and immunoreceptor tyrosine-based inhibitory motif domain (TIGIT) is a coinhibitory receptor expressed on dysfunctional tumor-infiltrating T cells, and TIGIT blockade has demonstrated encouraging activity in combination with PD-1 blockade in murine tumor models and in clinical studies. However, the degree to which TIGIT mediates T cell dysfunction in DLBCL has not been fully explored.Results Here, we demonstrate that TIGIT is broadly expressed on lymphoma-infiltrating T cells (LITs) across a variety of human lymphomas and is frequently coexpressed with PD-1. TIGIT expression is particularly common on LITs in DLBCL, where TIGIT+ LITs often form distinct cellular communities and exhibit significant contact with malignant B cells. TIGIT+/PD-1+ LITs from human DLBCL and murine lymphomas exhibit hypofunctional cytokine production on ex vivo restimulation. In mice with established, syngeneic A20 B-cell lymphomas, TIGIT or PD-1 mono-blockade leads to modest delays in tumor outgrowth, whereas PD-1 and TIGIT co-blockade results in complete rejection of A20 lymphomas in most mice and significantly prolongs survival compared with mice treated with monoblockade therapy.Conclusions These results provide rationale for clinical investigation of TIGIT and PD-1 blockade in lymphomas, including DLBCL.

The onion yellow dwarf virus (OYDV) represents a limiting biotic stress in onion (Allium cepa L.); little information is available regarding resistant varieties. In Italy, onion production is limited but represented by a wide diversity of ecotypes. A two-year trial was carried out to test the OYDV-susceptibility/tolerance of different Italian onion cultivars by rating symptom severity and plant growth parameters and assessing post-harvest secondary infections. The cultivar and ecotypes included in the study were characterized by simple sequence repeats (SSR) analysis, and the expression analysis of two genes (Eukaryotic translation initiation factors, EIFs) involved in potyvirus replication was also performed. Two susceptible and one tolerant cultivar were identified based on symptom expression and virus impact on plants. Although differences in growth parameters were limited to the first-year trial, the infection was correlated to a higher incidence of secondary infections in post-harvest, with altered water balance in infected bulbs. This correlation was also demonstrated during the long-term storage of bulbs. SSR analysis identified different clusters and only one gene isoform (EIF4eiso1) showed different expression levels in the OYDV/onion pathosystem. In conclusion, this study defines the genetic profile of Italian onion cultivars and provides evidence on susceptibility/tolerance features which will be useful in the future for the identification of viral resistance traits in onion.

This paper analyses the reception of Norbert Wiener’s cybernetics in France with particular attention to the perspective proposed by Gilbert Simondon. The debate that has emerged since the 1950s among the philosophers of the so-called French Theory opens up the possibility of speaking of a revised cybernetics on the basis of the need to go beyond the structural analogy between living being and machine. The present paper investigates the relationship between Simondon and cybernetics by considering his 1953 manuscript, his two doctoral theses and his speech at the Royaumont conference in 1962. It also focuses on the concept of ecceità that Simondon uses in order to speak from the qualitative side of information and to propose an ontological perspective in which it is that which goes beyond the limits of Aristotelian hylomorphism.

BackgroundUnderstanding the complex regulatory network underlying plant nitrogen (N) responses associated with high Nitrogen Use Efficiency (NUE) is one of the main challenges for sustainable cropping systems. Nitrate (NO3-), acting as both an N source and a signal molecule, provokes very fast transcriptome reprogramming, allowing plants to adapt to its availability. These changes are genotype- and tissue-specific; thus, the comparison between contrasting genotypes is crucial to uncovering high NUE mechanisms.MethodsHere, we compared, for the first time, the spatio-temporal transcriptome changes in both root and shoot of two NUE contrasting tomato genotypes, Regina Ostuni (high-NUE) and UC82 (low-NUE), in response to short-term (within 24 h) low (LN) and high (HN) NO3- resupply. ResultsUsing time-series transcriptome data (0, 8, and 24 h), we identified 395 and 482 N-responsive genes differentially expressed (DEGs) between RO and UC82 in shoot and root, respectively. Protein kinase signaling plant hormone signal transduction, and phenylpropanoid biosynthesis were the main enriched metabolic pathways in shoot and root, respectively, and were upregulated in RO compared to UC82. Interestingly, several N transporters belonging to NRT and NPF families, such as NRT2.3, NRT2.4, NPF1.2, and NPF8.3, were found differentially expressed between RO and UC82 genotypes, which might explain the contrasting NUE performances. Transcription factors (TFs) belonging to several families, such as ERF, LOB, GLK, NFYB, ARF, Zinc-finger, and MYB, were differentially expressed between genotypes in response to LN. A complementary Weighted Gene Co-expression Network Analysis (WGCNA) allowed the identification of LN-responsive co-expression modules in RO shoot and root. The regulatory network analysis revealed candidate genes that might have key functions in short-term LN regulation. In particular, an asparagine synthetase (ASNS), a CBL-interacting serine/threonine-protein kinase 1 (CIPK1), a cytokinin riboside 5’-monophosphate phosphoribohydrolase (LOG8), a glycosyltransferase (UGT73C4), and an ERF2 were identified in the shoot, while an LRR receptor-like serine/threonine-protein kinase (FEI1) and two TFs NF-YB5 and LOB37 were identified in the root. DiscussionOur results revealed potential candidate genes that independently and/or concurrently may regulate short-term low-N response, suggesting a key role played by cytokinin and ROS balancing in early LN regulation mechanisms adopted by the N-use efficient genotype RO.

Olive possesses excellent nutritional and economic values for its main healthy products. Among them, a high content of antioxidant compounds, balanced during the ripening process, are produced under genetic and environmental control, resulting in high variability among cultivars. The genes involved in these complex pathways are mainly known, but despite many studies which indicated the key role of light quality and quantity for the synthesis of many metabolites in plants, limited information on these topics is available in olive. We carried out a targeted gene expression profiling in three olive cultivars, Cellina di Nardò, Ruveia, and Salella, which were selected for their contrasting oleic acid and phenolic content. The –omics combined approach revealed a direct correlation between a higher expression of the main flavonoid genes and the high content of these metabolites in ‘Cellina di Nardò’. Furthermore, it confirmed the key role of FAD2-2 in the linoleic acid biosynthesis. More interestingly, in all the comparisons, a co-regulation of genes involved in photoperception and circadian clock machinery suggests a key role of light in orchestrating the regulation of these pathways in olive. Therefore, the identified genes in our analyses might represent a useful tool to support olive breeding, although further investigations are needed.

Recent advances in mission archaeology advocate for studies beyond the mission church and quadrangle in order to better understand their spatial organizations and how they were embedded within the landscapes of indigenous populations. This raises the question of how to implement such studies in areas impacted for years by urban development, which has made it difficult to detect archaeological remains using standard pedestrian-survey methods. This article advocates for the use of geophysical survey as part of the mix of field strategies. Archaeologists undertook fieldwork at Mission San Francisco Solano in Sonoma, California, to assess the potential of employing geophysical-survey methods in contexts characterized by extensive post-mission reuse. The results indicate that ground-penetrating radar and resistivity surveys are capable of detecting earlier mission architectural remains that can be differentiated from the remains of post-mission urban development from the late 19th and early 20th centuries.Avances recientes en la arqueología de misiones abogan por estudios más allá de la iglesia y del patio interior de la misión con el fin de comprender mejor sus organizaciones espaciales y cómo fueron incrustadas en los paisajes de las poblaciones indígenas. Esto plantea la cuestión de cómo implementar dichos estudios en áreas afectadas durante años por el desarrollo urbano, que ha dificultado detectar restos arqueológicos utilizando métodos estándar de estudios peatonales. El presente estudio aboga por el uso de estudios geofísicos como parte del mix de estrategias de campo. Los arqueólogos emprendieron trabajo de campo en la Misión San Francisco Solano en Sonoma (California) para evaluar el potencial de emplear métodos de estudio geofísicos en contextos caracterizados por una reutilización post-misión extensiva. Los resultados indican que el radar de penetración terrestre y los estudios de resistividad son capaces de detectar restos arquitectónicos de misiones anteriores que pueden ser diferenciados de los restos del desarrollo urbano post-misión de finales del siglo XIX y principios del siglo XX.Les progrès récents en archéologie de mission défendent les études en dehors de l’église de la mission et du quadrilatère afin de mieux comprennent leur organisation spatiale et comment elle était intégrée dans les paysages des populations autochtones. Cela soulève la question de l'application de ces études dans les zones touchées pendant des années par le développement urbain, qui a rendu difficile la détection des vestiges archéologiques à l’aide de méthodes normalisées de sondages dans la rue. Cet article préconise l’utilisation d'un levé géophysique dans le cadre de la combinaison des stratégies de terrain. Les archéologues ont entrepris des travaux sur le terrain à la Mission San Francisco Solano, à Sonoma (Californie) pour évaluer la possibilité d’utilisation des méthodes de levés géophysiques dans des contextes caractérisés par une vaste réutilisation après la mission. Les résultats indiquent que les enquêtes de géoradar et les levés de résistivité sont capables de détecter des vestiges architecturaux de mission antérieurs, qui peuvent être différenciés des vestiges du développement urbain postérieur à la mission de la fin du 19e siècle et au début du 20e siècle.

The remarkable potential of geophysical scanning—to assess the internal variability of sites in new ways, to highlight important phenomena in the field, to exercise co-creation of interpretation and commitment to minimal destruction of community partners’ resources, and to aid in the practice of due diligence in avoiding desecration of the sacred—continues to be underutilized in archaeology. While archaeological artifacts, features, and strata remain primary foci of archaeological geophysics, these phenomena are perceived quite differently in scans than in visual or tactile exposures. In turn, new registers of site exploration afforded by geophysical prospection may be constrained by the language of site excavation and visual observation, requiring adjustments in the ways of thinking about and describing what the instruments are measuring. The texture and form of site deposits as rendered in ground-penetrating radar scans can be examined in detail prior to making interpretations of cultural features or stratigraphy. Far more than simple “anomalies” demanding our attention for excavation, patterns in geophysical data can be the focus of extensive archaeological analysis prior to, in conjunction with, or independent from excavation.

Sex chromosomes evolved from autosomes many times across the eukaryote phylogeny. Several models have been proposed to explain this transition, some involving male and female sterility mutations linked in a region of suppressed recombination between X and Y (or Z/W, U/V) chromosomes. Comparative and experimental analysis of a reference genome assembly for a double haploid YY male garden asparagus (Asparagus officinalis L.) individual implicates separate but linked genes as responsible for sex determination. Dioecy has evolved recently within Asparagus and sex chromosomes are cytogenetically identical with the Y, harboring a megabase segment that is missing from the X. We show that deletion of this entire region results in a male-to-female conversion, whereas loss of a single suppressor of female development drives male-to-hermaphrodite conversion. A single copy anther-specific gene with a male sterile Arabidopsis knockout phenotype is also in the Y-specific region, supporting a two-gene model for sex chromosome evolution.

Abstract Molecular docking computationally predicts the conformation of a small molecule when binding to a receptor. Scoring functions are a vital piece of any molecular docking pipeline as they determine the fitness of sampled poses. Here we describe and evaluate the 1.0 release of the Gnina docking software, which utilizes an ensemble of convolutional neural networks (CNNs) as a scoring function. We also explore an array of parameter values for Gnina 1.0 to optimize docking performance and computational cost. Docking performance, as evaluated by the percentage of targets where the top pose is better than 2Å root mean square deviation (Top1), is compared to AutoDock Vina scoring when utilizing explicitly defined binding pockets or whole protein docking. Gnina, utilizing a CNN scoring function to rescore the output poses, outperforms AutoDock Vina scoring on redocking and cross-docking tasks when the binding pocket is defined (Top1 increases from 58% to 73% and from 27% to 37%, respectively) and when the whole protein defines the binding pocket (Top1 increases from 31% to 38% and from 12% to 16%, respectively). The derived ensemble of CNNs generalizes to unseen proteins and ligands and produces scores that correlate well with the root mean square deviation to the known binding pose. Understanding identity aspects of those labeled Genízaro during the late Spanish Colonial period of New Mexico benefits from finer-grained perspectives on what ranges and mixtures of practices persons bearing this casta designation may have performed while preparing cuisine. Materials from the northern frontier site of Casitas Viejas (LA 917) suggest that the closely related households of this fortified plaza may have departed from the less expansive culinary practices of colonial elites while drawing from their multiple social relationships at the various stages of production and consumption of foods. In other words, at different temporal and spatial scales, behaviors reflected in the material record refute historical notions about a creolized community that tried to diminish identity difference within the village. The goal of this work is to explore through the study of faunal remains some of the relationships between foodways and cultural identity in a manner that might assist in some disentangling of the sticky problems archaeologists face in interpreting traces of dynamic past situations of identity from a static material record recovered today.

Many students, especially English language learners (ELLs), struggle with writing expository texts. This study examined the impact of several writing strategies on ELLs' writing skills, including prewriting strategies and scaffolding strategies inherent in the Thinking Maps (TM) program. The purpose of the study was to see if ELLs were able to use these strategies to express their ideas more effectively in compositions in a more organized way. The participants were 8 students in grades 3 through 5 in the South Bay School District. The students were participating in an after-school writing class 2 days a week for 6 months. As a result, the overall average of students' writing scores in the areas of "Ideas" and "Organization" increased. While the overall averages were below the proficiency level (3.0), these writing strategies can be seen as having a positive impact on ELLs' writing skills.

In the Natufian lithic component at Hayonim, both in the cave and the terrace, numerous artifacts of pink/red color may be recognized. Cherts with similar appearance are not present in the geological environment surrounding the site in Northern Israel. Pink chert available in Jordan is shown to be of different nature. Thus this leaves us with the hypothesis of intentional heat treatment of locally available iron-rich yellow chert, of Cenomanian age. Based on experimental replication of chert firing and SEM analysis, we argue that a well-mastered and controlled use of fire was practiced by some skilled craftsmen at Hayonim throughtout the Late Epipalaeolithic.

After administration, patients underwent subsequent injections according to PRN criteria in case of edema relapse, but not earlier than 4 months after the previous treatment. Patients were evaluated at baseline, within 15 days of injection, and every month thereafter. During all visits, best-corrected visual acuity (BCVA) was recorded; central retinal thickness (CRT), type of edema, presence of SND, and presence and number of HRFs were evaluated using swept-source optical coherence tomography (SS-OCT) 3D. Treatment outcome was defined as changes in BCVA, CRT, SND and HRFs at 12 (T12) and 24 (T24) months compared with baseline (T0).Results: The study enrolled 24 eyes of 18 patients. The mean duration of follow-up was 18± 6.6 months; for all eyes, T12 data were available, while follow-up reached T24 for 12 eyes. BCVA improved significantly and CRT decreased significantly during treatment; the edema was no longer detectable in 13/24 eyes at T12 and 8/12 eyes at T24. No patient presented SND at T12 and T24, and the mean number of HRFs decreased significantly during treatment. Results with CRT and HRFs correlated with BCVA at 12 and 24 months. No significant adverse events were observed.Conclusion: In patients with DME, the intravitreal dexamethasone implant was effective and safe in improving both functional and tomographic parameters. Several Triticum species spread in cultivation in Sicily and neighboring regions over the centuries, which led to the establishment of a large genetic diversity. Many ancient varieties were widely cultivated until the beginning of the last century before being replaced by modern varieties. Recently, they have been reintroduced in cultivation in Sicily. Here, the genetic diversity of 115 and 11 accessions from Sicily and Calabria, respectively, belonging to Triticum species was evaluated using a high-density SNP array. Einkorn, emmer, and spelta wheat genotypes were used as outgroups for species and subspecies; five modern varieties of durum and bread wheat were used as references. A principal coordinates analysis (PCoA) and an unweighted pair group method with arithmetic mean (UPGMA) showed four distinct groups among Triticum species and T. turgidum subspecies. The population structure analysis distinguished five gene pools, among which three appeared private to the T. aestivum, T. turgidum subsp. Turgidum, and ‘Timilia’ group. The principal component analysis (PCA) displayed a bio-morphological trait relationship of a subset (110) of ancient wheat varieties and their wide variability within the T. turgidum subsp. durum subgroups. A discriminant analysis of principal components (DAPC) and phylogenetic analyses applied to the four durum wheat subgroups revealed that the improved varieties harbored a different gene pool compared to the most ancient varieties. The ‘Russello’ and ‘Russello Ibleo’ groups were distinguished; both displayed higher genetic variability compared to the ‘Timilia’ group accessions. This research represents a comprehensive approach to fingerprinting the old wheat Sicilian germplasm, which is useful in avoiding commercial fraud and sustaining the cultivation of landraces and ancient varieties.

Archaeological studies of movement between communities that had horses should use empirical evidence about horse travel over terrain types analogous to those traversed between historical archaeological sites. The experiences of equestrians are of interest to archaeologists because they reflect past processes of creating landscapes of warfare, communication, transportation, and trade. Late Spanish colonial New Mexico provides an example of how the potential of an equine perspective on landscape-scale choices might change archaeological interpretations of place and space. This article introduces an experimental approach and calls for modeling that accounts for different kinds of observed horse travel that can better articulate archaeological landscape studies with more realistic travel factors encountered by those who populated a dynamic and horse-connected frontier. Datasets generated by such a method will be well-positioned to aid in the interpretation of lived experiences on indigenous landscapes completely transformed by the colonial introduction of the horse.

Wild-game carcass processing behaviors, including marrow extraction and grease rendering, are traditionally inferred from the nutritional utility of recovered elements. The expected survivorship of bones processed for within-bone fats, as well as the inverse relationship between density and grease utility, diminishes the power to infer these behaviors by element counts alone. The palimpsests of transport and butchery decisions as well as non-human site formation processes are revealed most clearly by taphonomic and metric analysis for carcass handling and density-mediated attrition. This article presents a means of inferring ruminant marrow and grease extraction by synthesizing lines of evidence for density and bone survivorship, selectivity of marrow-rich and greasy elements, presence of percussive impact marks and fractures to fresh bone, small specimen sizes, and high fragmentation rates.

The domestication and spreading of grapevine as well as the gene flow history had been described in many studies. We used a high-quality 7k SNP dataset of 1,038 Eurasian grape varieties with unique profiles to assess the population genetic diversity, structure, and relatedness, and to infer the most likely migration events. Comparisons of putative scenarios of gene flow throughout Europe from Caucasus helped to fit the more reliable migration routes around the Mediterranean Basin. Approximate Bayesian computation (ABC) approach made possible to provide a response to several questions so far remaining unsolved. Firstly, the assessment of genetic diversity and population structure within a well-covered dataset of ancient Italian varieties suggested the different histories between the Northern and Southern Italian grapevines. Moreover, Italian genotypes were shown to be distinguishable from all the other Eurasian populations for the first time. The entire Eurasian panel confirmed the east-to-west gene flow, highlighting the Greek role as a “bridge” between the Western and Eastern Eurasia. Portuguese germplasm showed a greater proximity to French varieties than the Spanish ones, thus being the main route for gene flow from Iberian Peninsula to Central Europe. Our findings reconciled genetic and archaeological data for one of the most cultivated and fascinating crops in the world.

Human impacts have left and are leaving distinctive imprints in the geological record. Here we show that in North America, the human-caused changes evident in the mammalian fossil record since c. 14,000 years ago are as pronounced as earlier faunal changes that subdivide Cenozoic epochs into the North American Land Mammal Ages (NALMAs). Accordingly, we define two new North American Land Mammal Ages, the Santarosean and the Saintagustinean, which subdivide Holocene time and complete a biochronologic system that has proven extremely useful in dating terrestrial deposits and in revealing major features of faunal change through the past 66 million years. The new NALMAs highlight human-induced changes to the Earth system, and inform the debate on whether or not defining an Anthropocene epoch is justified, and if so, when it began.

The current project tested a diagnostic approach to selecting interventions to increase patron selection of reusable dinnerware in a cafeteria. An assessment survey, completed by a sample of 43 patrons, suggested that the primary causes of wasteful behavior were (a) environmental arrangement of dinnerware options and (b) competing motivational variables. A functional relation between environmental arrangement and reusable product selection was demonstrated in a reversal design. However, the largest effect occurred as function of a multicomponent intervention involving environmental arrangement, employee involvement, and personal spoken prompts with motivational signs. The results support the use of informant assessments when designing community interventions. (Contains 1 table and 1 figure.)

Eggplant (Solanum melongena L.), similar to many other crops, suffers from soil-borne diseases, including Fusarium oxysporum f. sp. melongenae (Fom), causing wilting and heavy yield loss. To date, the genetic factors underlying plant responses to Fom are not well known. We previously developed a Recombinant Inbred Lines (RILs) population using as a female parent the fully resistant line ‘305E40’ and as a male parent the partially resistant line ‘67/3’. The fully resistant trait to Fom was introgressed from the allied species S. aethiopicum. In this work, the RIL population was assessed for the responses to Fom and by using a genomic mapping approach, two major QTLs on chromosomes CH02 and CH11 were identified, associated with the full and partial resistance trait to Fom, respectively. A targeted BSAseq procedure in which Illumina reads bulks of RILs grouped according to their resistance score was aligned to the appropriate reference genomes highlighted differentially enriched regions between resistant/susceptible progeny in the genomic regions underlying both QTLs. The characterization of such regions allowed us to identify the most reliable candidate genes for the two resistance traits. With the aim of revealing exclusive species-specific contigs and scaffolds inherited from the allied species and thus associated with the full resistance trait, a draft de-novo assembly of available Illumina sequences of the ‘305E40’ parent was developed to better resolve the non-recombining genomic region on its CH02 carrying the introgressed Fom resistance locus from S. aethiopicum.

In this work, an electrochemical sensor for uric acid determination is shown with a preliminary study for its validation in real samples (milk and urine). Uric acid can be electrochemically oxidized in aqueous solutions and thus it is possible to obtain electrochemical sensors for this chemical by means of this electrooxidation reaction. Indium tin oxide coated on flexible polyethylene terephthalate substrate, modified with reduced graphene oxide and gold nanoparticles by co-electrodeposition, was used. Electrodeposition was performed at -0.8V vs SCE for 200 s. All samples were characterized by electron scan microscopy and electron diffraction spectroscopy. A careful investigation on the effect of pH was performed to understand its influence on uric acid oxidation. The detection of uric acid was using the linear sweep voltammetry. Results show that the peak current increases linearly with uric acid concentration from 10 to 1000 µM with a limit of detection of about 7.1 µM. The sensor shows high selectivity towards different interferents that can be found in the milk and urine matrix, such as chloride, calcium, sodium and ammonium ions. To prove the applicability of the proposed sensor, uric acid was quantified in real milk and urine samples with excellent results comparable to those of conventional techniques.
