【目的】 探讨雷帕霉素对肝癌肝移植术后复发影响?【方法】 应用MTT细胞增殖实验观察雷帕霉素对walker-256肿瘤细胞增殖的影响;应用流式细胞仪观察雷帕霉素对walker-256肿瘤细胞周期的影响;应用荧光定量RT-PCR观察雷帕霉素walker-256肿瘤细胞VEGFmRNA表达的影响?观察雷帕霉素对免疫抑制肝转移癌大鼠模型肿瘤生长情况?肿瘤组织PCNA指数?肿瘤复发率及生存情况的影响?【结果】 MTT细胞增殖实验示雷帕霉素对walker-256肿瘤细胞有抑制作用,IC50为9.60 ng/mL;细胞周期检测示雷帕霉素组G1/G0期比例为55.2% ± 0.1%较对照组(45.2%±0.4%)高(P < 0.05);雷帕霉素组VEGFmRNA表达为(4.50 ± 0.13)×10-2较对照组[(70.8 ± 1.3)×10-2]低(P < 0.05)?雷帕霉素组荷瘤大鼠荷瘤肝重为(12.5 ± 0.2)g,较对照组[(14.4 ± 0.3)g]轻,两组间差别有统计学意义(P < 0.05);雷帕霉素组荷瘤大鼠荷瘤肝质量比为(61.3 ± 0.6)×10-3,较对照组[(70.8 ± 1.3)×10-3]小,两组间差别有统计学意义(P < 0.05);雷帕霉素组大鼠肿瘤复发率为90%,对照组为95%,两组比较差异无统计学意义;雷帕霉素组生存时间为(26.5 ± 2.5)d,对照组为(14.8 ± 2.3)d,两组间差别有统计学意义(P < 0.05)?雷帕霉素组大鼠PCNA指数为64.1% ± 2.1%,较对照组(80.8% ± 1.0%)低,差别有统计学意义(P < 0.05)?【结论】 虽然雷帕霉素对降低免疫抑制下的肿瘤复发率作用不明显,但能明显抑制肿瘤生长速度,延长带瘤生存时间?

【目的】构建小鼠端粒酶蛋白亚单位（mouse telomerase reverse transcriptase，mTERT）基因重组腺病毒载体，为下一步研究其体外表达和动物实验研究提供基础。【方法】从肝癌细胞中提取总RNA。采用RT-PER技术扩增mTERT的基因编码区序列，将序列定向克隆至真核表达载体pAC，将此表达质粒与腺病毒重组质粒pJM17共同转染293细胞，经同源重组产生重组腺病毒载体Ad-mTERT，纯化后的重组腺病毒载体Ad-mTERT在293细胞大量扩增并通过氯化铯密度梯度离心法纯化，测定病毒滴度。【结果】PCR及酶切证实：mTERTDNA正确克隆到穿梭质粒pAC中，带mTERTDNA的表达盒成功重组到腺病毒载体基因组E1A缺失区，并在293细胞中成功包装出具有感染活性的重组腺病毒Ad-mTERT。【结论】成功构建了小鼠TERT基因重组腺病毒载体，为研究其用于肿瘤的基因治疗奠定基础。

目的: 探讨不同热、冷缺血时间对供肝胆道的影响。方法: 将经历不同热缺血时间的SD 大鼠肝脏用 4 ℃ UW 液( solution of University of Wisconsin)经门静脉灌注,胆管用 4 ℃ UW 液或 Ringer 液逆行灌注后置 4 ℃ UW 液中保存一定时限。光镜观察胆道粘膜。结果: UW 液灌注组: 热缺血时间在 3 min 以内胆道经历12 h 冷缺血后无明显改变; 热缺血时间达 5 min 的胆道在经历 12 h 冷缺血后出现严重的损 伤。用 R inger液进行胆道灌洗效果欠佳。结论:①UW 液对供肝胆道有保护作用。 ②关于供肝胆道的保存时限: 热缺血时间不应超过 5 min; 若热缺血时间在 3 min 以内,冷缺血时间可延长至 12 h; 热缺血时间达 5 min,则冷缺血时间不应超过 8 h。

Stroke is the leading cause of death and disability in Chinese residents, with ischemic stroke (IS) accounting for approximately 70% of stroke patients. In IS, cardioembolic stroke and embolic stroke of undetermined source account for 20%-30% and 30%-40%, respectively. Cardiac magnetic resonance (CMR) examination is an important adjunctive diagnostic tool, facilitating the identification of IS etiology while also revealing the novel pathogenesis. This article discussed the necessity of CMR examination in IS patients, the value of CMR examination techniques, and the indications for its application in IS patients. It also recommended the procedure of CMR examination for IS patients to provide better help for the diagnosis, treatment, and secondary prevention of IS. [ABSTRACT FROM AUTHOR]
