The basic physicochemical indicators, phenolic composition and aroma composition of fruits from five-year-old ‘Cabernet Sauvignon’ grapevines with four shoot densities of 15, 20, 25, and 30/m from the wine production area of the Qingtong Gorge at the eastern foothill of Helan Mountain in Ningxia were analyzed, and principal component analysis (PCA) was used to investigate monomeric anthocyanin contents and comprehensive fruit quality. The results showed that there were differences in the physical indicators of grape bunches and individual grapes and the chemical indicators such as sugar, acid, phenolics, and aroma substances among different shoot densities (P 20 > 30 > 25/m. In conclusion, for ‘Cabernet Sauvignon’ grapes in the wine production area of the Qingtong Gorge, the comprehensive fruit quality was best at a shoot density of 15–20/m.

The types and contents of organic acids in wine grapes determine wine quality. To explore the effects of different rootstocks on the acid metabolism of 'Cabernet Sauvignon' grapes, various perennial rootstock–scion combinations were used as experimental materials. High-performance liquid chromatography (HPLC) was used to determine citric acid, tartaric acid, and malic acid contents during fruit development. Succinic acid and oxalic acid contents and the activity of related enzymes were measured using spectrophotometry. The expression levels of related genes were measured using a real-time fluorescence quantitative method. The results showed that all four rootstock types significantly reduced oxalic acid and citric acid contents in the grapes, while increasing succinic acid content to varying degrees. Employing 110R, SO4, and Kangzhen3 rootstocks increased tartaric acid and malic acid contents. Enzyme activity analysis revealed that 110R, SO4, and Kangzhen3 rootstocks increased the NAD-MDH enzyme activity, which positively correlated with malic acid content. Simultaneously, these rootstocks reduced the NADP-ME enzyme activity level. NAD-MDH and PEPC gene expression levels were higher in 'Cabernet Sauvignon' grapes grafted with 110R, SO4, and Kangan3 rootstocks compared to control self-rooted seedlings. Field studies were conducted on Vitis vinifera cv. 'Cabernet Sauvignon' to evaluate the effects of the defoliation time (pre- and post-flowering and veraison) and severity on the 3-isobutyl-2-methoxypyrazine (IBMP) concentration and expression levels of the key related genes in grape berries. The IBMP concentration in the grapes decreased significantly at harvest after all the defoliation treatments. The earlier and heavier the defoliation treatment, the lower the IBMP concentration. The relative expression level of VvOMT3 in the berry skin was closely positively correlated with the IBMP accumulation in all the treatments. Early defoliation coupled with a 1-week delay in the harvest time may be an effective management strategy to control the IBMP concentration in grapes.

The aim of this work was to determine the effect of different drying treatments: freeze-drying, room-drying, and oven-drying on the grape phenolic composition of the 'Regent' and 'Cabernet Sauvignon' varieties. After drying, the samples were grounded, submerged to ultrasound-assisted extraction, and analyzed with high-performance liquid chromatography (HPLC). This study demonstrated that the use of different drying methods significantly affects the content of polyphenols in grape skins extracts. The greatest content of anthocyanins-diglucoside was preserved using freeze-drying, i.e. 15706.86 mg/kg for 'Regent', while the content of anthocyanins-monoglucoside was best preserved by room-drying, i.e. 216778.68 and 9220.30 mg/kg for 'Regent' and 'Cabernet Sauvignon', respectively. The highest content of flavonol glycosides (2583.04 and 1429.64 mg/kg for 'Regent' and 'Cabernet Sauvignon', respectively), hydroxycinnamic acids (1303.31 and 544.88 mg/kg for 'Regent' and 'Cabernet Sauvignon', respectively), and stilbene (2321.52 and 79.36 mg/kg for 'Regent' and 'Cabernet Sauvignon', respectively) was observed in the oven-dried samples. By applying freeze-drying the most optimal content of flavan-3-ol was preserved. Contents of polyphenolic compounds in oven-dried samples after 6 months of storage were almost identical to those in the samples analyzed immediately after drying. The greatest rate of degradation was observed in the room-dried samples while it was moderate in the freeze-dried ones. The results of this experiment demonstrate that it is necessary to dry samples in different ways to obtain the highest content of a certain polyphenolic group of compounds. The application of a drying method is determined by the goal of the final dried product in terms of content and composition of different polyphenolic compounds; thus, the obtained results could have an application in scientific research and for commercial purposes as well.

Abstract Mathematical models are indirect and non-destructive methods for estimating the leaf area (LA), an important parameter associated with canopy vigor in Vitis viniferavineyards. This work aims to adjust and test mathematical models for estimating LA using the midvein length of the Vitis vinifera cultivars conducted in horizontal and vertical training systems in the Serra Gaúcha region, Brazil. Leaves were collected in ‘Chardonnay’, ‘Cabernet Sauvignon’ vineyards conducted in trellis and lyre training systems, and in ‘Merlot’ vineyards conducted in trellis and espalier systems. Models were tested for establishing the correlations among the parameters LA and midvein length, considering the cultivar/training system and the general data set. For validation, linear regression between the observed and estimated LA was defined. The relationship between the midvein length and LA was defined by non-linear potential equations (R2aj 0.95-0.98). The performances of the specific and general models were similar regardless of the cultivar/training system. The adjusted models showed excellent results in the validation step (correlation 0.95-0.98; Wilmott index >0.97 and confidence index>0.92). It is possible to estimate the LA of Vitis vinifera Chardonnay, Cabernet Sauvignon, and Merlot cultivars using a simple non-destructive model that employs one single measurement of the leaf blade, to optimize the vegetative canopy quantification in vineyards.

Abstract: The objective of this work was to evaluate the yield and must composition of 'Cabernet Sauvignon' and 'Chardonnay' grapevines subjected to phosphorus applications to a soil from a high-altitude region of Southern Brazil, during three crop seasons. Experiments 1 and 2 were carried out in 'Cabernet Sauvignon' and 'Chardonnay' commercial vineyards, respectively, in the municipality of Água Doce, located in the Midwestern region of the state of Santa Catarina. The soil from the two vineyards was classified as a Typic Humicryept. The used source of P was triple superphosphate (45% P2O5). From 2011 to 2013, at flowering, a total of 0, 10, 20, 40, and 80 kg ha-1 P2O5 were applied in the crown projection area, to the soil surface, without incorporation, in both vineyards. Soil and leaf P concentration, yield parameters, and must quality were evaluated. The increase in P availability decreased total titratable acidity and tartaric acid in the must of the 'Cabernet Sauvignon' grapevine in the 2013/2014 crop season and increased pH and total soluble solids in the must of the 'Chardonnay' grapevine in the 2011/2012 crop season. Phosphorus application to the soil increases the levels of available phosphorus, but does not result in higher yields for 'Cabernet Sauvignon' and 'Chardonnay' grapevines.

Esca proper and Botryosphaeria dieback are among the most widespread Grapevine trunk diseases (GTDs), characterized by similar decline symptoms. In the present work, chronic, apoplexy and death symptoms were analysed separately in four vineyards and four different cultivars, on more than 1,000 vines per cultivar, taking into account ten-year annual surveys. The cumulative incidence of plants with chronic symptoms (CHR) reached high values on 'Sauvignon Blanc' (81.9 %), 'Cabernet Sauvignon' (79.4 %) and 'Cannonau' (66.5 %), but it was low on 'Merlot' (25.1 %). 'Sauvignon Blanc' showed the highest cumulative incidence of apoplectic events (23.1 %) and dead cordons (49.2 %), while 'Cannonau' had the greatest number of dead plants (28.8 %). In each symptom category, incidence among cultivars differed significantly according to χ² test at P ≤ 0.05. Annual incidence of foliar symptoms fluctuated over ten years (ranging from 0.9-9.5 % in 'Merlot' to 6.3-59.1 % in 'Cabernet Sauvignon'), mostly with regard to CHR. On average, every year only 33.9 % of plants showing CHR had expressed symptoms in the previous year, while 48.6 % did not show symptoms the following year. Conversely, most of the plants exhibiting apoplexy or death were symptomatic the previous year. According to Tuckey HSD test (P ≤ 0.05) 'Merlot' had the highest incidence of plants showing CHR symptoms for the first time (72.1 %) and of apparently recovered plants (76.3 %), while 'Cabernet Sauvignon' exhibited the highest incidence of plants showing CHR symptoms also the previous year (50.0 %). The 'Cabernet Sauvignon' attitude to show chronic symptoms with a certain continuity was also confirmed by the low incidence of plants with hidden symptoms (lack of symptoms in previously symptomatic vines). On the contrary, the incidence of acute symptoms (apoplectic events and dead plants) was quite low on 'Cabernet Sauvignon'. The present study confirms that GTD incidence is influenced by cultivar. All the cultivars assessed were susceptible, but with differences in intensity, type (chronic or acute) and fluctuation of symptoms. It cannot be excluded, however, that besides the genotype also external factors, as the vigour conferred by the type of soil or the combination with the rootstock, may have influenced the results. Potassium is the most abundant cation in grape berries. It has important roles in grapevine physiology and winemaking. This study investigates the feasibility of using rootstocks to lower berry potassium concentrations ([K]) in 'Cabernet Sauvignon' grapevines. The ultimate target is to achieve lower pH and higher titratable acidity (TA) in grape juice so as to bring down the cost of acid adjustment during winemaking. The specific objective here is to provide new insights into the potential of particular rootstocks to modify K uptake by 'Cabernet Sauvignon' grapevines and their partitioning and accumulation into grape berries. The vineyard soils of a replicated rootstock trial located in the Limestone Coast of South Australia were characterised. Petiole, berry and juice nutrient content were assessed at oenological maturity of 'Cabernet Sauvignon' grown on eight different rootstocks. Rootstock had an impact on cations of the vegetative tissue of 'Cabernet Sauvignon', with Merbein 5512 having the lowest petiole [K]. The concentrations of major cations in the berry were, however, not altered by rootstock. While no particular rootstock stood out in limiting 'Cabernet Sauvignon' berry K accumulation, berries grown on the 'Börner' rootstock tended to have slightly lower concentrations (< 10%) relative to vines on their own roots. Across the rootstocks, juice pH tended to increase with greater juice [K], while juice TA tended to decrease with greater juice [K]. It was found that juice TA was higher for the rootstocks 140 Ruggeri and 110 Richter, and juice pH tended to be lower for the rootstocks 110 Richter, 140 Ruggeri, Merbein 5512 and Merbein 5489. Grapevine leafroll disease (GLD) is present in all grape-growing regions of the world and is considered the most significant grapevine viral disease. Grapevine leafroll-associated virus 3 (GLRaV-3) is considered the primary cause of GLD and in South African vineyards five genetic variant groups (I, II, III, VI and VII) have been confirmed. Biological distinctions between GLRaV-3 variants have not been fully validated. By characterising virus concentration and stress-responsive microRNA expression in GLRaV-3 infected plants, this study aimed to glean a better understanding of the possible biological distinctions between GLRaV-3 variants. Quantitative reverse transcription PCR was utilised for virus concentration ratio (VCR) determination and miRNA quantitation in GLRaV-3 positive and negative grapevines grown under greenhouse and field conditions. This study found statistically significant differences in VCRs in plants singly infected with different GLRaV-3 variants. Interestingly, no difference in mean VCRs were observed between data sets, despite notable differences in plant age, duration of GLRaV-3 infection, scion/rootstock combination and growing conditions. Several miRNAs showed statistically significant expression modulation between infected and healthy samples. miRNA expression between data sets varied substantially and a greater overall miRNA response was observed in plants with more established GLRaV-3 infections. The lack of significant differences in mean VCRs between data sets, coupled with the consistent modulation of certain miRNAs in plants that have likely been infected for longer is a promising result. This finding could indicate that successful inhibition of further virus replication by plant defence mechanisms occurred, and that these miRNAs are implicated in this response. Adaptation of varieties to changing climatic conditions is a major breeding target, which includes the selection of late ripening varieties/clones, whose bunches may escape the warmer summer condition by postponing the ripening period. However, assessment of the genetic basis of phenology and quality related traits is a prerequisite to develop breeding programs for grapevine varieties adapted for the cultivation in specific viticultural areas and/or to identify the candidate genes for the new breeding technology approaches. The present work reports a two-season evaluation of traits segregation in populations derived from crosses of the red skinned cv. 'Corvina', the principal local variety of the Valpolicella wine area (Verona, Italy), with other two cultivars: the white skinned cv. 'Solaris', highly divergent from 'Corvina' for the phenology and fruit ripening traits, and the red skinned cv. 'Cabernet Sauvignon', whose bunches shows distinctive ripening and post-ripening traits from 'Corvina'. One hundred and fourty-two and one hundred and 29 seedlings were developed respectively for each population, propagated and grown under field conditions. During each season the main phenological stages from budbreak to berry maturation were determined for each genotype in the populations. At full ripening, several clusters from each plant were harvested for measuring main morphological and technological parameters. The data collected was then used to evaluate the distribution of each trait across the individuals in each population. Correlation analyses have been performed between traits collected in the two seasons or across traits. The comprehensive information obtained will be used for QTL mapping. The identification of genetic markers associated with the studied traits will help accelerating the selection of new cultivars more adapted to the changing climatic conditions. Altogether, the described approaches will finally allow to improve our current understanding of the genetic control of phenology and berry quality traits in grape, thus helping and assisting breeding. Effective abscission agents that decrease fruit detachment force (FDF) are sought by the California raisin industry to improve the continuous tray mechanical harvesting method. Such agents might also enable mechanical harvest of table and wine grapes (Vitis vinifera L.), but few agents are known to be effective for grape. Thus, methyl jasmonate (MeJA) and six other compounds known to stimulate abscission of other fruits were screened for their ability to reduce FDF of mature 'Thompson Seedless' grapes. Most compounds tested reduced FDF to some extent, but MeJA was particularly effective. Solutions containing between 45 and 4500 ppm MeJA reduced FDF by at least 50% to 85% compared with nontreated fruits. Application of 2250 and 4500 ppm MeJA to 'Thompson Seedless' vines caused 25% to 50% fruit drop, respectively, within 10 d after treatment (DAT). The efficacy of MeJA was verified in a second experiment in which solutions of 0, 1125, 2250, or 4500 ppm MeJA were applied to clusters of 'Crimson Seedless' grapes; at 14 DAT, FDF declined as a linear function of MeJA applied. The grapes did not abscise, but berries treated with 2250 to 4500 ppm MeJA had slightly lower soluble solids than nontreated fruits. Solutions of 0 or 4500 ppm MeJA applied to clusters of 'Cabernet Sauvignon' and 'Merlot' grapevines reduced FDF by 66% and 75%, respectively. Fruit drop was estimated to be less than 10%. Thus, a solution containing up to 4500 ppm MeJA may be an effective abscission agent to facilitate mechanical harvest of 'Cabernet Sauvignon' or 'Merlot'. [ABSTRACT FROM AUTHOR]

Sorti ‘Merlot’ in ‘Cabernet sauvignon’ sta najbolj razširjeni rdeči sorti vinske trte, saj jo gojijo na vseh celinah sveta. Sorti sta se po svetu širili istočasno, čeprav se razlikujeta tako v ampelografskih lastnostih kot agrotehničnih zahtevah. V Slovenijo oziroma Primorsko, kjer sta trenutno pomembnejši rdeči sorti, naj bi prišli proti koncu 19. stoletja. V Goriških brdih sta sorti posajeni po celem okolišu, čeprav razlike v kakovosti grozdja po lokacijah dokazujejo neprimernost določenih leg za gojenje rdečih sort vinske trte. Količinsko največ grozdja obeh sort pridelajo na lokacijah Biljana, Cerovo in Neblo. Na lokaciji Kozana pridelajo najkakovostnejše, medtem ko na lokaciji Kožbana najslabše grozdje obeh sort. Vzroke velike variabilnosti med lokacijami in leti v kakovosti grozdja pripisujemo interakcijskim vplivom vremenskih razmer, razgibanega reliefa ter tradicionalne agro-ampelotehnike, s katero ne dosegajo več nadpovprečne kakovosti grozdja.
