Local accumulation of the plant growth regulator auxin mediates pattern formation in Arabidopsis roots and influences outgrowth and development of lateral root- and shoot-derived primordia. However, it has remained unclear how auxin can simultaneously regulate patterning and organ outgrowth and how its distribution is stabilized in a primordium-specific manner. Here we show that five PIN genes collectively control auxin distribution to regulate cell division and cell expansion in the primary root. Furthermore, the joint action of these genes has an important role in pattern formation by focusing the auxin maximum and restricting the expression domain of PLETHORA (PLT) genes, major determinants for root stem cell specification. In turn, PLT genes are required for PIN gene transcription to stabilize the auxin maximum at the distal root tip. Our data reveal an interaction network of auxin transport facilitators and root fate determinants that control patterning and growth of the root primordium. [ABSTRACT FROM AUTHOR]

SUMMARY: The enzyme DWARF27 (D27) catalyzes the reversible isomerization of all‐trans‐ into 9‐cis‐β‐carotene, initiating strigolactone (SL) biosynthesis. Genomes of higher plants encode two D27‐homologs, D27‐like1 and ‐like2, with unknown functions. Here, we investigated the enzymatic activity and biological function of the Arabidopsis D27‐like1. In vitro enzymatic assays and expression in Synechocystis sp. PCC6803 revealed an unreported 13‐cis/15‐cis/9‐cis‐ and a 9‐cis/all‐trans‐β‐carotene isomerization. Although disruption of AtD27‐like1 did not cause SL deficiency phenotypes, overexpression of AtD27‐like1 in the d27 mutant restored the more‐branching phenotype, indicating a contribution of AtD27‐like1 to SL biosynthesis. Accordingly, generated d27 d27like1 double mutants showed a more pronounced branching phenotype compared to d27. The contribution of AtD27‐like1 to SL biosynthesis is likely a result of its formation of 9‐cis‐β‐carotene that was present at higher levels in AtD27‐like1 overexpressing lines. By contrast, AtD27‐like1 expression correlated negatively with the content of 9‐cis‐violaxanthin, a precursor of ABA, in shoots. Consistently, ABA levels were higher in shoots and also in dry seeds of the d27like1 and d27 d27like1 mutants. Transgenic lines expressing GUS driven by the AtD27LIKE1 promoter and transcript analysis of hormone‐treated Arabidopsis seedlings revealed that AtD27LIKE1 is expressed in different tissues and affects ABA and auxin. Taken together, our work reports a cis/cis‐β‐carotene isomerase that affects the content of both cis‐carotenoid‐derived plant hormones, ABA and SLs. A small organizing center, the quiescent center (QC), maintains stem cells in the Arabidopsis root and defines the stem cell niche. The phytohormone auxin influences the position of this niche by an unknown mechanism. Here, we identify the PLETHORA1 (PLT1) and PLT2 genes encoding AP2 class putative transcription factors, which are essential for QC specification and stem cell activity. The PLT genes are transcribed in response to auxin accumulation and are dependent on auxin response transcription factors. Distal PLT transcript accumulation creates an overlap with the radial expression domains of SHORT-ROOT and SCARECROW, providing positional information for the stem cell niche. Furthermore, the PLT genes are activated in the basal embryo region that gives rise to hypocotyl, root, and root stem cells and, when ectopically expressed, transform apical regions to these identities. Thus, the PLT genes are key effectors for establishment of the stem cell niche during embryonic pattern formation. [Copyright &y& Elsevier]

A retraction of the article "Generation of cell polarity in plants links endocytosis, auxin distribution and cell fate decisions," by Pankaj Dhonukshe and others, that was published in a 2008 issue is presented.

The article reports that the phytohormone auxin plays a major role in plant growth. To determine the direction of auxin flow researchers studied the directional auxin translocation during root gravitropism in Arabidopsis thaliana. They designed hemagglutinin epitope-tagged PIN1 and PIN2 genes under the transcriptional control of the PIN2 promoter. It was observed that in cortex cells, both proteins were localized at the lower basal side. Researchers tested the relationship between polarity and the direction of auxin flow by examining auxin translocation during gravity response. It was found that the localization of PIN proteins at the upper side of epidermal cells was related to their ability to facilitate upward auxin movement for root gravitropic response. Finally it was concluded that PIN polarity is a primary direction determining factor in auxin transport in meristematic tissues. It also provides information on how auxin flows can be redirected through rapid changes in PIN polarity in response to developmental and environmental signals.
