Optical lattices (OLs) with conventional spatial periodic λ/2, formed by interfering the counterpropagating laser beams with wavelength λ, are versatile tools to study the dynamical and static properties of ultracold atoms. OLs with subwavelength spatial structure have been realized in recent quantum-gas experiment, offering new possibility for nonlinear and quantum control of ultracold atoms at the nano scale. Herein, we study theoretically and numerically the formation, property, and dynamics of matter-wave localized gap modes of Bose–Einstein condensates loaded in a one-dimensional nanoscale dark-state OL consisted of an array of optical subwavelength barriers. The nonlinear localized modes, in the forms of on- and off-site fundamental gap solitons, and dipole ones, are demonstrated; and we uncover that, counterintuitively, these modes exhibit always a cusplike (side peaks) mode even for a deeply subwavelength adiabatic lattice, contrary to the previously reported results in conventional deep OLs where the localized gap modes are highly confined in a single lattice cell. The (in)stability features of all the predicted localized modes are verified through the linear-stability analysis and direct perturbed simulations. Our predicted results are attainable in current ultracold atoms experiments with the cutting-edge technique, pushing the nonlinear control of ultracold atoms with short-period OLs as an enabling technology into subwavelength structures.

【Background and objective】 Hainan province is a pepper base in China and its traditional production uses furrow fertigation, resulting in not only water and fertilizer waste but also nutrient leaching and contaminating the environment. The purpose of this paper is to explore suitable irrigation and fertilization methods to improve water and fertilizer use efficiency without compromising pepper yield. 【Method】 The experiment was conducted in field plots with four different irrigation - fertilization combinations: furrow irrigation without fertilization (CK), furrow irrigation with fertilization of 470.70 kg/hm2 of N + 511.22 kg/hm2 of P2O5 + 526.39 kg/hm2 of K2O (F1), furrow irrigation with fertilization of 376.55 kg/hm2 of N + 314.83 kg/hm2 of P2O5 + 526.39 kg/hm2 of K2O (F2), mulched micro-spray with fertilization of 376.55 kg/hm2 of N + 314.83 kg/hm2 of P2O5 + 526.39 kg/hm2 of K2O. In each treatment, we measured yield and fruit quality, photosynthesis, root growth, nutrient accumulation and nutrient distribution in soil, as well as water and fertilizer use efficiency. 【Result】 When N and P were initially high in the soil, F1 and F2 did not show significant difference in their yield, fruit quality, root growth, photosynthesis and accumulation of N, P and K in the plant. In contrast, F3 increased the yield, transpiration, root length, root surface area, root volume, accumulation of N, P and K by 8.03%, 16.91%, 60.69%, 43.00%, 40.37%, 19.30%, 14.19% and 14.61%, respectively, compared to F2; it also showed significant differences in fruit quality, net photosynthetic rate, stomatal conductance and intercellular CO2 concentration from F2. Compared with F1, F2 reduced the contents of total N, ammonium N, total P and available P in the 0~20 cm and 20~40 cm of soil after the harvest, reduced the apparent surplus N and P, and improved the partial productivity of N and P fertilizers. Compared with F2, F3 increased the contents of total N, nitrate N, ammonium N, total P and available P in the 0~20 cm of soil, reduced the apparent surplus N and P, and improved the production rate of the irrigation water. 【Conclusion】 For all treatments we compared, mulched micro-spray irrigation with reduced fertilization (F3) improved yield, water and fertilizer use efficiency of the pepper.

It is an increasingly mature application solution that triboelectric nanogenerator (TENG) supplies power to electronic devices through its power management system (PMS). However, the previous PMS is able to manage a limited voltage magnitude and the energy storage elements are limited to capacitors. This work proposes an ultrahigh voltage PMS (UV‐PMS) to realize the charging of commercial lithium cells (LCs) by TENG. The design of UV‐PMS enables energy management of TENGs with ultrahigh open‐circuit voltages up to 3500 V and boosts the peak charging current from 30.9 µA to 2.77 mA, an increase of 89.64 times. With the introduction of UV‐PMS, the effective charging capacity of LC charged by a TENG at a working frequency of 1.5 Hz for 1 h comes to 429.7 µAh, making a 75.3 times enhancement compared to charging by TENG directly. The maximum charging power comes to 1.56 mW. The energy storage efficiency is above 97% and the overall charge efficiency can be maintained at 81.2%. This work provides a reliable strategy for TENG to store energy in LC, and has promising applications in energy storage, LC's life, and self‐powered systems. [ABSTRACT FROM AUTHOR]

During continuous slab casting, strand electromagnetic stirring (S-EMS) has a significant effect on improving the slab quality. In the current work, a numerical model based on the practical slab continuous casting machine and coupled electromagnetic field, flow field, solidification, and solute transport was established to investigate and evaluate the effect of the S-EMS installation position with various current intensities on metallurgical behavior. The model was verified by magnetic field measurement, infrared camera, and nail shooting experiments. The results show that moving the S-EMS installation position to the solidification end reduces the stirring effect due to the skin effect and the increasing thickness of the slab shell. A higher installation position is beneficial for improving the equiaxed grain rate, while a lower one is beneficial for reducing carbon segregation. The maximum segregation index and range decrease from 1.26 to 1.2 and from 0.42 to 0.36 with the installation position being decreased from −3 m to −12.8 m, respectively. The industrial trials show that S-EMS installed at 3 m has a significant effect on expanding the equiaxed grain zone and a deteriorating effect on reducing carbon segregation. [ABSTRACT FROM AUTHOR]

Secondary cooling electromagnetic stirring (S-EMS) significantly impacts the internal quality of continuous casting slabs. In order to investigate the effects of S-EMS modes on segregation in slabs, a three-dimensional numerical model of the full-scale flow field, solidification, and mass transfer was established. A comparative analysis was conducted between continuous electromagnetic stirring and alternate stirring modes regarding their impacts on steel flow, solidification, and carbon segregation. The results indicated that adopting the alternate stirring mode was more advantageous for achieving uniform flow fields and reducing the disparity in solidification endpoints, thus mitigating carbon segregation. Specifically, the central carbon segregation index under continuous stirring at 320 A was 1.236, with an average of 1.247, while under alternate stirring, the central carbon segregation index decreased to 1.222 with an average of 1.227. In order to solve the problem that the apparent density measurement result of the coal body is inaccurate due to its small particle size, the feasibility of the coal seam apparent density measurement by simulating coal seam method is researched by theoretical analysis of the relationship between the coal sample particle size and the apparent density. Using the self-developed visual density measuring device, nine coal samples with different metamorphic degrees from different parts of the country were selected for experimental study. The results show that: in a very small range, the apparent density of coal samples increases with the decrease of the particle size of coal samples. When the particle size of coal samples is large enough, the apparent density of coal samples will tend to nearly constant value. With the increase of coal metamorphism, the apparent density of coal increases first and then decreases, which is same to the change rule of coal metamorphism degree and coal micropore volume, porosity and coal adsorption capacity. For each coal sample measured twice under the same moisture content, particle size, molding pressure and surrounding rock pressure, the analysis found that the apparent density measurement error is less than 0.5%; the error difference of the apparent density obtained is no more than 0.33%.

The heat transfer of a slab is significantly influenced by roll contact during the continuous casting process. The influence of roll contact calculation methods on the predicted heat transfer results has not been previously investigated. In this work, the non-uniform solidification of the wide-thick slab was studied with a 2D heat transfer model using real roll contact method (R. method) and equivalent roll contact method (E. method). The predicted slab surface temperature and shell thickness were verified with the measured results of the infrared camera and nail shooting experiments, respectively. Then, the predicted heat transfer results (including the slab surface temperature, mushy region length, and solidification end position) for the wide-thick slab with different thicknesses and different casting speeds were calculated using the E. method and R. method, and the influence of these two roll contact methods on the predicted heat transfer results was discussed for the first time. The results show that both these two roll contact methods could be applied to accurately predict the slab surface temperature without considering the transient temperature dips in the roll–slab contact regions. However, the deviation of the predicted mushy region length and solidification end position using the E. method are obvious. Compared with the R. method, the predicted mushy region length obtained using the E. method is larger and the solidification end obviously subsequently moves along the casting direction. In past years, optical lattices have been demonstrated as an excellent platform for making, understanding, and controlling quantum matters at nonlinear and fundamental quantum levels. Shrinking experimental observations include matter-wave gap solitons created in ultracold quantum degenerate gases, such as Bose–Einstein condensates with repulsive interaction. In this paper, we theoretically and numerically study the formation of one-dimensional gap soliton molecules and clusters in ultracold coherent atom ensembles under electromagnetically induced transparency conditions and trapped by an optical lattice. In numerics, both linear stability analysis and direct perturbed simulations are combined to identify the stability and instability of the localized gap modes, stressing the wide stability region within the first finite gap. The results predicted here may be confirmed in ultracold atom experiments, providing detailed insight into the higher-order localized gap modes of ultracold bosonic atoms under the quantum coherent effect called electromagnetically induced transparency. Background. Cervical cancer (CC) is one of the most frequent female malignancy. Cancer stem cells (CSCs) positively affect survival outcomes in cancer patients, but in cervical cancer, the mechanism of tumor stem cells is still uncertain. Methods. RNA-seq data and related clinical follow-up of patients suffering from CC were from TCGA. Consensus clustering screened prognostic mRNAsi-related genes and identified molecular subtypes for CC. Based on the overlapping differentially expressed genes (DEGs) in subtypes, we employed LASSO and multivariate Cox regression to screen prognostic-related genes and established the RiskScore system. The patients were grouped by RiskScore, the prognosis was analyzed by the Kaplan-Meier (K-M) curve among the various groups, and the precision of the RiskScore was assessed by the ROC curve. Finally, the potential worth of RiskScore in immunotherapy/chemotherapy response was assessed by evaluating TIDE scores and chemotherapy drug IC 50 values. Results. We noticed that patients with low mRNAsi had a shorter survival and then identified three molecular subtypes (C1-3), with the C1 having the worst prognosis and the lowest mRNAsi. Finally, we identified 7 prognostic-related genes (SPRY4, PPP1R14A, MT1A, DES, SEZ6L2, SLC22A3, and CXCL8) via LASSO and Cox regression analysis. We established a 7-gene model defined RiskScore to predict the prognosis of CC patients. K-M curve indicated that low RiskScore patients had improved prognosis, and ROC curves indicated that RiskScore could precisely direct the prognostic evaluation for those suffering from the cancer. This was also confirmed in the GSE44001 and GSE52903 external cohorts. Patients were more sensitive to immunotherapy if with low RiskScore, and RiskScore exhibited precise assessment ability in predicting response to immunological therapy in CC patients. Conclusion. CC stemness is associated with patient prognosis, and the RiskScore constructed based on stemness characteristics is an independent prognostic index, which is expected to be a guide for immunotherapy, providing a new idea for CC clinical practice. Highlights: A flexible tribotronic artificial synapse with bioinspired neurosensory behaviour was demonstrated, which establishes an active interaction mechanism with the environment. The device can well exhibit tuneable synaptic behaviours by changing the mechanical input modes, including excitatory postsynaptic current, paired-pulse facilitation, and the hierarchical memorial process. The device has excellent mechanical flexibility that can exhibit stable synaptic functions even under strain conditions with a bending radius of 20 mm after 1000 bending cycles. As key components of artificial afferent nervous systems, synaptic devices can mimic the physiological synaptic behaviors, which have attracted extensive attentions. Here, a flexible tribotronic artificial synapse (TAS) with bioinspired neurosensory behavior is developed. The triboelectric potential generated by the external contact electrification is used as the ion-gel-gate voltage of the organic thin film transistor, which can tune the carriers transport through the migration/accumulation of ions. The TAS successfully demonstrates a series of synaptic behaviors by external stimuli, such as excitatory postsynaptic current, paired-pulse facilitation, and the hierarchical memory process from sensory memory to short-term memory and long-term memory. Moreover, the synaptic behaviors remained stable under the strain condition with a bending radius of 20 mm, and the TAS still exhibits excellent durability after 1000 bending cycles. Finally, Pavlovian conditioning has been successfully mimicked by applying force and vibration as food and bell, respectively. This work demonstrates a bioinspired flexible artificial synapse that will help to facilitate the development of artificial afferent nervous systems, which is great significance to the practical application of artificial limbs, robotics, and bionics in future. In the era of the Internet of Things, various electronics play an important role in information interaction, in which the power supply is an urgent problem to be solved. Triboelectric nanogenerator (TENG) is an emerging mechanical energy harvesting technology that can serve as a power source for electronics, which is developing towards high performance, miniaturization and integration. Herein, the advanced micro-nano manufacturing technologies are systematically reviewed for TENGs. First, film preparation such as physical vapor deposition, chemical vapor deposition, electrochemical deposition, electrospinning and screen printing for triboelectric layers are introduced and discussed. Then, surface processing, such as soft lithography, laser ablation, inductively coupled plasma and nanoimprint for micro-nano structures on the surface of triboelectric layers are also introduced and discussed. In addition, micro-electromechanical system fabrication for TENG devices such as acoustic and vibration sensors, is introduced, and their current challenges are analyzed. Finally, the challenges of the advanced micro-nano manufacturing technologies for the TENGs are systematically summarized, and further development is prospected. [ABSTRACT FROM AUTHOR]

Owing to the insufficient converter heat, IF steel is produced via the BOF–LF–RH–CC process in Pangang Group Xichang Steel and Vanadium Co., Ltd. To clarify the evolution of inclusions and the control strategy to improve the cleanliness of molten steel in Ti-bearing IF steel produced via the long process, scanning electron microscopy with energy spectroscopy analysis and automatic scanning electron microscopy were employed to analyze the number, size, type and morphology of inclusions in IF steel from RH to tundish. The results show that the characteristics of inclusions are similar in two heats during RH treatment. In the tundish sample of Heat 2, the number density (ND) and area fraction (AF) of Al2O3 and Al2O3·TiOx inclusions increase significantly, and the size of Al2O3 inclusions decreases obviously, which is closely related to the serious reoxidation of molten steel caused by the slag with high oxidability during the holding process. Meanwhile, a new method of determining the number of cluster inclusions is used to evaluate the cleanliness of IF steel in this paper, and the obtained number of inclusion clusters is consistent with the trend of ND and AF of inclusions. The effects of reoxidation on the morphology, number and other indexes of Al2O3 and Al2O3·TiOx inclusions are discussed in detail, and there are two ways of forming Al2O3·TiOx inclusions in the case of serious reoxidation. To weaken the reoxidation process and enhance the cleanliness of IF steel produced via the long process, reducing the oxygen content in molten steel before Al deoxidation, minimizing the holding time and reducing the oxidability of slag after RH are helpful. The gas emission rate of boreholes is one of the most important indices for coal and gas outburst prediction. In this work, instantaneous gas emission velocity and environmental effects on borehole gas emission were studied. Through theoretical analysis, the mechanism of crack propagation in the coal borehole was clarified, and the effect of soft and hard coal on gas desorption and gas emission. The results of numerical simulations also indicated that the initial gas emission has a function relationship to the drilling distance and the physical characteristics of the coal seam. A novel dynamic testing technology was proposed to obtain gas emission velocity. Laboratory experiments under adsorption and desorption of CO2 and N2 were performed using coal samples from Xuehu, Fenghui, Weishe, and Wuzhong coal mines. The data of initial gas emission under different coal samples were recorded, and the fitting curves were obtained. The results show a positive correlation between initial gas emission and drilling depth. However, abnormal would occur when the drill pipe enters the soft stratification, and the maximum value of the initial gas emission of the abnormal part is 3.8 times the normal value, which indicates a high degree of sensibility to soft stratification. Myofibres (primary and secondary myofibre) are the basic structure of muscle and the determinant of muscle mass. To explore the skeletal muscle developmental processes from primary myofibres to secondary myofibres in pigs, we conducted an integrative three-dimensional structure of genome and transcriptomic characterization of longissimus dorsi muscle of pig from primary myofibre formation stage [embryonic Day 35 (E35)] to secondary myofibre formation stage (E80). In the hierarchical genomic structure, we found that 11.43% of genome switched compartment A/B status, 14.53% of topologically associating domains are changed intradomain interactions (D -scores) and 2,730 genes with differential promoter–enhancer interactions and (or) enhancer activity from E35 to E80. The alterations of genome architecture were found to correlate with expression of genes that play significant roles in neuromuscular junction, embryonic morphogenesis, skeletal muscle development or metabolism, typically, NEFL , MuSK , SLN , Mef2D and GCK. Significantly, Sox6 and MATN2 play important roles in the process of primary to secondary myofibres formation and increase the regulatory potential score and genes expression in it. In brief, we reveal the genomic reorganization from E35 to E80 and construct genome-wide high-resolution interaction maps that provide a resource for studying long-range control of gene expression from E35 to E80. Gas pressure is one of the major factors to characterize the outburst danger of coal seams. Generally, the process of gas pressure determination is complicate and cannot ensure the accuracy of results, which may cause a wrong identification of outburst danger. This paper discussed and revealed a positive correlation between initial gas emission and gas pressure, which provides a new approach to identify the outburst danger by initial gas emission. Through theoretical analysis and numerical simulation, the liner equation of gas pressure and initial gas emission was deduced. Laboratory experiments under adsorption and desorption of CO2 and N2 were performed using coal samples from Weishe coal mine. The data of initial gas emission under different gas pressure were recorded, and the fitting curves were obtained. The results show a positive correlation between initial gas emission and gas pressure, and the curve of N2 was below the CO2. Especially, while the adsorption gas pressure was under 0.663 and 0.675 MPa, the difference of corresponding initial gas emission was only 0.01 L/s, which indicates the high degree of correlation. The results validated the theoretical and practical feasibility to identify the coal and the gas outbursts through initial gas emission. [ABSTRACT FROM AUTHOR]

Porcine reproductive and respiratory syndrome virus (PRRSV) 1 and 2 differ in their recognition of CD163. Substitution of porcine CD163 SRCR5 domain with a human CD163-like SRCR8 confers resistance to PRRSV 1 but not PRRSV 2. The deletion of CD163 SRCR5 has been shown to confer resistance to PRRSV 1 in vivo and both PRRSV 1 and 2 in vitro. However, the anti-PRRSV 2 activity of modifying the CD163 SRCR5 domain has not yet been reported. Here, we describe the highly efficient generation of two pig breeds (Liang Guang Small Spotted and Large White pigs) lacking a short region of CD163 SRCR5, including the ligand-binding pocket. We generated a large number of gene-edited Large White pigs of the F0 generation for use in viral challenge studies. The results of this study show that these pigs are completely resistant to infection by species 2 PRRSV, JXA1, and MY strains. There were no clinical symptoms, pathological abnormalities, viremia, or anti-PRRSV antibodies in the CD163 SRCR5-edited pigs compared to wild-type controls after viral challenge. Porcine alveolar macrophages (PAMs) isolated from CD163 SRCR5-edited Large White pigs also displayed resistance to PRRSV in vitro. In addition, CD163 SRCR5-edited PAMs still exhibited a cytokine response to PRRSV infection, and no significant difference was observed in cytokine expression compared to wild-type PAMs. Taken together, these data suggest that CD163 SRCR5-edited pigs are resistant to PRRSV 2, providing a basis for the establishment of PRRSV-resistant pig lines for commercial application and further investigation of the essential region of SRCR5 involved in virus infection. Ubiquitin E3 Ligase MARCH7 is involved in T cell proliferation and neuronal development. In our previous study, we demonstrated MARCH7 promoted malignant behavior of ovarian cancer via the nuclear factor (NF)-κB and Wnt/β-catenin signaling pathway. However, the expression and function of MARCH7 in cervical cancer remains unknown. The present study aimed to unravel the expression and function of MARCH7 in cervical cancer to elucidate its potential role in the diagnosis and pathogenesis of cervical cancer. Results indicated that the expression of MARCH7 was abnormally high in cervical cancer tissues than normal cervical tissues. However, silencing the expression of MARCH7 in HeLa cells resulted in decreased cell proliferation and invasion. Mechanistic investigations revealed that MARCH7 interacted with VAV2. Silencing the expression of MARCH7 in HeLa cells inhibited the VAV2-RAC1-CDC42 signaling pathway. Overall, the results of the present study identified MARCH7 as a candidate oncogene in cervical cancer, and a potential target for cervical cancer therapy. [ABSTRACT FROM AUTHOR]

This paper describes the preparation of low-temperature sintered nanosilver paste with inverse microemulsion method with Span-80/Triton X-100 as the mixed-surfactant and analyzes the influence of different sintering parameters (temperature, pressure) on the shear properties of low-temperature sintering of nanosilver. Experimental results show that the shear strength of the low-temperature sintering of nanosilver increases as the temperature and pressure increase. But there are many pores and relative fewer cracks on the sintering layer after low-temperature sintered. The test thermal resistance of low-temperature sintered nanosilver paste is 0.795 K/W which is greater than SAC305 weld layer with a T3ster thermal analyzer. Based on the coupling effect of contact electrification and electrostatic induction, the triboelectric nanogenerator (TENG) as an emerging energy technology can effectively harvest mechanical energy from the ambient environment. However, due to its inherent property of large impedance, the TENG shows high voltage, low current and limited output power, which cannot satisfy the stable power supply requirements of conventional electronics. As the interface unit between the TENG and load devices, the power management circuit can perform significant functions of voltage and impedance conversion for efficient energy supply and storage. Here, a review of the recent progress of switching power management for TENGs is introduced. Firstly, the fundamentals of the TENG are briefly introduced. Secondly, according to the switch types, the existing power management methods are summarized and divided into four categories: travel switch, voltage trigger switch, transistor switch of discrete components and integrated circuit switch. The switch structure and power management principle of each type are reviewed in detail. Finally, the advantages and drawbacks of various switching power management circuits for TENGs are systematically summarized, and the challenges and development of further research are prospected. The wearable tactile sensors have attracted great attention in the fields of intelligent robots, healthcare monitors and human-machine interactions. To create active tactile sensors that can directly generate electrical signals in response to stimuli from the surrounding environment is of great significance. Triboelectric nanogenerators (TENGs) have the advantages of high sensitivity, fast response speed and low cost that can convert any type of mechanical motion in the surrounding environment into electrical signals, which provides an effective strategy to design the self-powered active tactile sensors. Here, an overview of the development in TENGs as tactile stimulators for multifunctional sensing and artificial synapses is systematically introduced. Firstly, the applications of TENGs as tactile stimulators in pressure, temperature, proximity sensing, and object recognition are introduced in detail. Then, the research progress of TENGs as tactile stimulators for artificial synapses is emphatically introduced, which is mainly reflected in the electrolyte-gate synaptic transistors, optoelectronic synaptic transistors, floating-gate synaptic transistors, reduced graphene oxides-based artificial synapse, and integrated circuit-based artificial synapse and nervous systems. Finally, the challenges of TENGs as tactile stimulators for multifunctional sensing and artificial synapses in practical applications are summarized, and the future development prospects are expected. Ultrasonic technology is often used to modify proteins. Here, we investigated the effects of ultrasound alone or in combination with other heating methods on emulsifying properties and structure of glycinin (11S globulin). Structural alterations were assessed with Sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS–PAGE), intrinsic fluorescence spectroscopy, ultraviolet (UV) absorption spectroscopy, and Fourier transform infrared (FTIR) spectroscopy. The size distribution and zeta-potential of 11S globulin were evaluated with a particle size analyzer. An SDS-PAGE analysis showed no remarkable changes in the primary structure of 11S globulin. Ultrasound treatment disrupted the 11S globulin aggregates into small particles with uniform size, narrowed their distribution and increased their surface charge density. Fluorescent spectroscopy and second-derivative UV spectroscopy revealed that ultrasound coupled with heating induced partial unfolding of 11S globulin, increasing its flexibility and hydrophobicity. FTIR further showed that the random coil and α-helix contents were higher while β-turn and β-sheet contents were lower in ultrasound combined with heating group compared to the control group. Consequently, the oil-water interface entirely distributed protein and reduced the surface tension. Moreover, ultrasound combined with heating at 60 °C increased the emulsifying activity index and emulsifying stability index of 11S globulins by 6.49-folds and 2.90-folds, respectively. As an essential synthetic material used in the continuous casting of steels, mold fluxes improve the surface quality of steel slabs. In this study, a CaO-SiO2-Na2O-based low-fluorine mold flux was solidified by an improved water-cooled copper probe with different temperatures of molten flux and different probe immersion times. The heat flux through solid films and the film structures were calculated and inspected, respectively. Internal cracks (formed in the glassy layer of films during solidification) were observed. The formation and evolution of those cracks contributed to the unstable heat flux density. The roughness of the surface in contact with the water-cooled copper probe formed as films were still glassy and the roughness had no causal relationship with crystallization or devitrification. The wyosine derivatives present at position 37 in transfer RNAs (tRNAs) are critical for reading frame maintenance. The methyltransferase Trm5a from Pyrococcus abyssi (PaTrm5a) plays a key role in this hypermodification process in generating m1G37 and imG2, two products of the wyosine biosynthetic pathway, through two methyl transfers to distinct substrates, but the mechanism is currently unknown. We report two cocrystal structures of PaTrm5a in complex with tRNAPhe and reveal the structural basis for substrate recognition, which was supported by in vitro activity assays. The crystal structures showed that the D1 domain of the enzyme undergoes large conformational changes upon the binding of tRNA. The deletion of this domain greatly reduces the affinity and activity of PaTrm5a toward its RNA substrate, indicating that the enzyme recognizes the overall shape of tRNA. Using the small-angle x-ray scattering technique and crystallographic analysis, we discovered that PaTrm5a adopts distinct open conformations before and after the binding of tRNA. Last, through structure comparison with its ortholog Methanococcus jannaschii Trm5b (MjTrm5b), we propose a reaction mechanism for the double methylation capability of this unique enzyme.
