No. of bitstreams: 0 Previous issue date: 2018-01-01 Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Successful disease treatment depends on molecular studies under indoor conditions with experimental infection protocols that facilitate understanding the disease and the drug`s efficacy. The internal transcribed spacer (ITS) region was sequenced from three isolates, which were identified as Saprolegnia aenigmatica. Subsequently, healthy fish were immunosuppressed with dexamethasone (1.2 mg kg-1) and descaled to the skin using a sharp scalpel. These individuals were isolated in individual aquariums maintained at 22°C. Individuals in one group were subcutaneously inoculated with 9,000 zoospores (DDZ treatment), a second group was exposed to oomycetes in water with three colonized baits (DDB), a third group was maintained in water without zoospores (DD), and a control group (C) consisted of healthy animals. After 48 and 96 hours, two animals from each group were euthanized for fungal re-isolation. The fish from groups DD and C did not show clinical signs, and no oomycetes were isolated. The animals from the DDZ and DDB groups showed cotton-wool-like masses on the skin, and S. aenigmatica was re-isolated. Thus, for infection using zoospores or baits parasitized by S. aenigmatica, an immunosuppressor (dexamethasone) and a sharp scalpel can be used effectively to establish an experimental infection in P. mesopotamicus. The knowledge of fish mineral requirements, such as selenium, is important to promote the growth and health status for fish in captivity, but excess or deficiency must be avoided. Therefore, this study evaluated the growth performance, hematology and the parasite load of Nile tilapia (Oreochromis niloticus) supplemented with organic selenium (0.0; 0.25; 0.5; 1.0 and 1.5 mg/Kg). The increased levels of selenium did not affect significantly (p<0.05) the growth performance of tilapia. However, the increases of thrombocytes and parasite load are proportional to the increases of selenium levels. Unlike expected, supplemented diets with selenium did not benefit growth performance and supplementation of 1.5 mg/kg caused deleterious effects on the health of tilapia. [ABSTRACT FROM AUTHOR]

No. of bitstreams: 0 Previous issue date: 2012-12-01 This trial was conducted in order to determine the 50% lethal concentration (LC (I) 50-96h) of the aqueous extract of dried leaves of Terminalia catappa, a plant with antifungal, antibacterial and antiparasitic activity. Due to the increasing use of herbal medicines in aquaculture, its use is an option for controlling diseases in fish. Toxicity tests are important before recommending any treatment, since some products have therapeutic concentration close to lethal. To conduct the study used 135 fish species Guarus (Phalloceros caudimaculatus), exposed to increasing concentrations of: 0.0, 50.0, 100.0, 150.0, 200.0, 250.0, 300.0 mL of stock solution per liter of water. The results were calculated by the method Trimmed Spearman Karbo, demonstrating that the 50% lethal concentration (LC (I) 50-96h) estimated was 208.52 mL / L, with lower limit of 187.79 mL / L and higher 231.54 mL / L. Observed changes in behavior of the test organisms at concentrations above 250 mL / L decrease in the levels of dissolved oxygen and pH. No. of bitstreams: 0 Previous issue date: 2011-08-22 The oxytetracyline (OTC) and florfenicol (FLO) are the most used antibiotics for bacterial disease control in fish. The aims are: estimating the lethal concentration 50% initial (LC(I)50;48h) of OTC (Terramicina ®) and FLO (Aquaflor ®) for pacu (Piaractus mesopotamicus); classifying OTC and FLO according to the acute toxicity and the risk of environmental intoxication; and assessing OTC and FLO dilution effect in water quality variables. Fish were acclimatized for 10 days in a bioassay room at 27 ± 2 °C. Then nine animals (three each concentration) were exposed to 7.0; 7.5; 8.0 or 8.5 mg L -1 of OTC or to 600.0; 700.0; 800.0; 900.0 and 1000.0 mg L -1 of FLO and a control treatment. The water quality variables recorded daily were: pH, temperature, electric conductivity, and dissolved oxygen. OTC and FLO were classified according to acute toxicity and environmental risk. The estimated OTC LC(I)50;48h for pacu is 7.6 mg L -1 and the estimated FLO is > 1000 mg L -1. OTC reduced the dissolved oxygen concentration, and it was considered moderately toxic, causing risk of environmental intoxication to pacu. FLO is not toxic; it does not cause risk of environmental intoxication to pacu and does not change water quality variables. Some studies aimed to demonstrate the effectiveness of herbicides to control these macrophytes; however, few studies report the possible ecotoxicological effects. The objective of this study was to estimate the acute toxicity (LC (I)50;96h) and assess water quality variables for glyphosate in the Rodeo® formulation, Aterbane® BR surfactant and mixtures of glyphosate + 0.5% and 1.0% of surfactant, for the guaru fish (Phallocerus caudimaculatus). The guaru was exposed to increasing concentrations of glyphosate and a mixture of glyphosate + 0.5 and 1.0% of surfactant. The mixture of glyphosate and glyphosate + 0.5 and 1.0% of surfactant showed (LC (I)50;96h) > 975.0 mg L -1. For the surfactant, the rate was 5.81 mg L -1. The glyphosate and mixtures of glyphosate + 0.5% and 1.0% of surfactant caused a decrease in pH and dissolved oxygen and increased the electrical conductivity of water. Glyphosate in the Rodeo® formulation and the mixtures with surfactant Aterbane® BR can be classified as practically nontoxic, whereas surfactant Aterbane® BR can be considered as moderately toxic to guaru. A RT de codorna é predominantemente do tipo intra-albugínica e constituída por canais epiteliais tortuosos estendendo-se pela túnica albugínea do testículo e a seguir penetram na estrutura microscópica da região epididimária (RE), pelo seu tecido matricial. Os canais da RT são contínuos com os ductos eferentes proximais da RE. A ultraestrutura do epitélio de revestimento da RT mostra diferenças marcantes na primavera - a fase proliferativa do ciclo reprodutivo, e em meados do outono - a fase quiescente do ciclo reprodutivo nesta ave. As características ultraestruturais marcantes dos epiteliócitos da RT, nestas duas fases, foram a baixa eletrodensidade citoplasmática e ocorrência de poucas vesículas e lisossomos no outono, que aparecem com maior freqüência na RT primaveril. Aspectos degenerativos de organelas, e presença de extrusões citoplasmáticas apicais, apareceram em número expressivo no outono, não se observando aspectos ultraestruturais marcantes nas estações de inverno e verão, relativamente às observações de primavera.

The RT of domestic quail from Italian variety showed mainly an albuginic pattern being represented by tortuous channels lined predominately by a simple cubic epithelium. RT channels extended along the testicular albuginea and penetrates into the epididymal region (ER) through its myoconnective matrix. Passageways were continuous to proximal efferent ducts of the ER. Epithelium lining ultrastructure of RT passageways showed some differences between the spring and the inactive phase at middle fall, concerning the quail testicular reproductive cycle. The features observed in RT epitheliocytes in fall were the low cytoplasmic electrodensity, paucity of supranuclear vesicles, which were abundant and variable in form and shape in spring, and some degenerative aspects of cell organelles mainly in ER lamellae. Moreover, presence of apical cytoplasmic extrusions were verified in the fall. No marked features were seen in the RT ultrastructure during the winter and summer comparatively to the active phase of spring. The surface epithelium of the vas deferens of Agouti paca, a wild and large South American rodent, was basically formed by principal and basal cells being only the principal cells related to endocytosis processes and also secretion taking base on their cytoplasmic ultrastructural features. Principal cell of vas deferens epithelium were characterized mainly by presence of vesicles with several shapes, sizes and internalized content at their apical cytoplasm occurring smaller pits and pale small vesicles seen next to the apical brush border of microvillus. Moreover, coated vesicles, smooth surface vesicles and great vesicles; multivesicular bodies, endosomes and lysosomes were seen. Presence of an apocrine secretory apparatus was also viewed, showing apical cytoplasmic expansions protruding into the vas deferens luminal compartment. The basal flattened cells, without luminal surface contact, occurred next to the basement membrane of the ductus, and did no exhibit special ultrastructural features. El epitelio que recubre la luz del conducto deferente de la laca (Agouti paca), un roedor silvestre de Sudamérica, está formado por células principales (P) y básales (B), en donde las células principales están asociadas a los procesos de endocitosis y secreción, teniendo una base en su característica ultraestructural citoplasmática. Las células principales de los vasos deferentes del epitelio se caracteriza principalmente por la presencia de vesículas con variadas formas, tamaños y contenido interior en su citoplasma apical produciendo pequeñas invaginaciones y pequeñas vesículas pálidas ubicadas continuas al borde en cepillo de las microveilosidades apicales. Además, fueron observados, vesículas de superficie lisa y vesículas de gran tamaño; órganos multivesiculares, endosomas y lisosomas. También se observó la presencia de un aparato secretor apocrino, con expansiones citoplasmáticas apicales que se protruyen en el compartimiento luminar del conducto deferente. Células básales aplanadas, sin contacto con la superficie luminal, se encuentran junto a la membrana basal del conducto, las que no presentan características ultraestructurales especiales.

The objective of this work was to estimate the acute toxicity of copper sulfate pentahydrate (CuSO4.5H2O) and the aqueous extract of dried neem leaves on snails (P. canaliculata) under laboratory conditions. In order to estimate the lethal concentration 50% (LC (I)50;96h), snails were exposed to six increasing copper sulfate concentrations (0.0; 0.01; 0.03; 0.05; 0.07 and 0.1 mg L-1) and six increasing concentrations of aqueous extract of dried neem leaves 0.0; 100; 125; 150; 175 and 200 mL aqueous extract of dried neem leaves L-1 water, equivalent to (0.0; 1.18; 1.47; 1.77; 2.06 and 2.36 mg azadirachtin L -1), in triplicate and one control treatment in an entirely random delineation. Estimated LC (I)50;96h, of copper sulfate was 0.02 mg copper sulfate L-1, with a 0.01 mg L-1 lower limit and a 0.03 mg L-1 upper limit. Estimated lethal concentration 50% of the aqueous extract of dried neem leaves was 142.75 mL L-1, equivalent to 1.68 mg L-1 of azadirachtine, with a 130.89 mL L-1 (1.54 mg L-1) low limit and 155.69 mL L-1 (1.83 mg L -1) as the upper limit. The lethal concentration of 50% (LC (I) 50-96h) and the histopathologic effects of diquat herbicide on Nile tilapia (Oreochromis niloticus) fish were evaluated in three experiments. The fishes were exposed to concentrations of 0, 25, 30, 35, 40, 45, 50, 55 and 60 mg diquat L-1, and gill and liver histology were evaluated in the surviving fishes. The estimated LC (I) (50-96h) of diquat was 37.28 mg L-1, with lower limits of 33.12 mg L-1 and upper limits of 41.44 mg L-1. In the treatment with 30, 35 and 40 mg L-1, signs of apical fusion of the secondary lamellae were observed; with 45 and 50 mg L-1, congestion of the primary lamellae was observed; in the treatment with 55 mg L-1, congestion of blood vessels on secondary lamellae took place. The livers of fishes in treatments with 0, 25, 30 and 35 mg L-1 showed cordonal organization of hepatocytes. In the treatments with 40 and 45 mg L-1, hypertrophy of hepatocytes took place; with 50 and 55 mg L-1, cell fusion and the presence of vacuoles inside hepatocytes were observed. Diquat presented low risk of toxicity for Nile tilapia, as the more severe histopathologic alterations occurred only in higher concentrations A concentração letal 50% e os efeitos histopatológicos do herbicida diquat para a tilápia nilótica (Oreochromis niloticus) foram avaliados em três experimentos. Os peixes foram expostos às concentrações de 0; 25; 30; 35; 40; 45; 50; 55; e 60 mg de diquat L-1 e a histologia da brânquia e do fígado foi avaliada nos peixes sobreviventes. A CL (I) 50-96h do diquat estimada foi de 37,28 mg L-1, com limite inferior de 33,12 mg L-1 e superior de 41,44 mg L-1. No tratamento com 30, 35 e 40 mg L-1, ocorreram início de fusão apical das lamelas secundárias; com 45 e 50 mg L-1 ocorreram congestão nas lamelas primárias e no tratamento com 55 mg L-1, ocorreu congestão sangüínea nas lamelas secundárias. O fígado dos peixes dos tratamentos controle, 30 e 35 mg L-1 estavam com organização cordonal dos hepatócitos. Histologic studies on the seminiferous epithelium were performed to determine the reproductive annual cycle of domestic duck in mesothermic tropical region and to correlate it with metabolic parameters such as blood glucose, hepatic and muscular glycogen and lipids, which are basic metabolic substrates related to the reproductive process. The annual reproductive cycle of the duck could be divided in four successive phases along the year, which are named reproductive, regressive, quiescent and recrudescent phases. The reproductive phase began in winter (July) reaching its maximum peak in October. The regressive phase occurred in the end of spring and in the beginning of summer (December). The quiescent phase started in the summer between January and February. The recrudescent phase occurred in autumn and winter (June) when the spermatogenetic process resumed. Concerning the energetic metabolism only the blood glucose was related to the reproductive cycle, giving energetic support to this reproductive process Estudos histológicos do epitélio seminífero foram realizados para determinar o ciclo reprodutivo anual do pato doméstico na região mesotérmica tropical e correlacioná-lo aos parâmetros de glicose sangüínea, de glicogênio e de lipídeos hepáticos e musculares, substratos metabólicos básicos ligados ao processo reprodutivo. O ciclo reprodutivo anual do pato pôde ser dividido em quatro fases sucessivas ao longo do ano, denominadas de reprodução, de regressão, de quiescência e de recrudescência testicular. A fase reprodutiva iniciou-se na estação de inverno (julho), atingindo seu pico máximo em outubro. A fase de regressão ocorreu no final da primavera (novembro) e no início do verão (dezembro). A fase de quiescência testicular iniciou-se no verão (janeiro e fevereiro). A fase de recrudescência ocorreu do outono (março) até o inverno (junho), com a retomada do processo espermatogenético. Quanto ao metabolismo energético, somente a glicose sangüínea esteve correlacionada ao ciclo reprodutivo do pato, fornecendo suporte energético para o processo reprodutivo

Twenty adult partridges Rhynchotus rufescens were used to study the morphology of oesophagus and crop. Materials to the morphologic study were collected and lengths of the oesophagus and of the crop were measured. For histological study, fragments of the oesophagus and of the crop were stained routinely with Masson’s trichrome stain. Total oesophagus was larger in females. In the entrance of the thorax, its ventral wall is enlarged broadly, forming the crop, which is larger in males. Oesophagus mucosa is constituted by stratified squamous epithelium, with mucous glands and lymphatic tissues. There are three layers of smooth muscle, involved in serosa. The histological structure of the crop is similar to the oesophagus. The mucous glands are reduced in size and the lymphatic tissues are dispersed in the connective tissue. There is a thin stratum of smooth muscle and other two wider layers of longitudinal and circular musculature. Vinte exemplares de Rhynchotus rufescens foram utilizadas para estudar a morfologia do esôfago e do papo. O material para o estudo morfológico foi coletado e o comprimento dos órgãos foi avaliado. Para o estudo histológico, as secções histológicas foram coradas com tricromo de Masson, posteriormente analisadas e descritas. O comprimento médio total do esôfago foi maior em fêmeas. Na entrada do tórax, a parede ventral do esôfago se dilata, formando o papo, maior em machos. A mucosa do esôfago é formada por epitélio escamoso estratificado, com glândulas mucosas e nódulos linfáticos. Há três camadas de músculo liso, as quais são envolvidas por serosa. A característica histológica do papo é semelhante à do esôfago. No papo, as glândulas mucosas são menores e os nódulos linfáticos estão dispersos no tecido conjuntivo. Há uma camada delgada de músculo liso e outras duas camadas mais largas de musculaturas longitudinal e circular.

No. of bitstreams: 0 Previous issue date: 2008-01-01 The objective of this work was to evaluate the skin, liver and kidney histology of young pacus supplemented in diet with trivalent chromium and to evaluate the residue of chromium in water effluent. Randomised delineation by using factorial scheme 4x2, with four levels of chromium (0, 6, 12, 18 mg/L), two stocking densities (4 and 20 kg/m(3)), and four repetitions were used. Pacus with 100 g were used, for 90 days. An increase of the number of mucus cells and club cells in the epidermis of the pacus supplemented with 18 chromium mg/kg was observed in any stocking density. The same effect was observed in the liver, and in the highest density (20 kg/m(3)) the alterations already occurred at the level of 6 mg/kg with the inclusion of chromium, initially with congestion of sinusoids and getting worse at the level of 18 mg/kg with necrosis. The kidney didn't present visible morphologic alterations when supplemented and submitted to the stocking densities. The concentration of chromium in the effluent of pacus was 3.27 mu g/L in the higher level of supplementation (18 mg/L). In conclusion the level of 18 supplement mg/kg was not shown beneficial to the growth of pacus and the stocking density, due to the possible stress, accelerated the alteration histology process of tissues. No presente estudo investigou-se a estrutura histológica do nervo radial do gato doméstico. Foram utilizados cinco gatos, três fêmeas e dois machos, sem raça definida, provenientes do CCZ de Jaboticabal, SP. Dividiu-se o nervo em três regiões e foram coletados três segmentos: proximal, médio e distal. Após procedimentos de rotina histológica convencional, observou-se três tecidos de sustentação: o epineuro, o perineuro e o endoneuro. O epineuro é formado por tecido conjuntivo denso modelado e não modelado, e também por conjuntivo frouxo. Do epineuro partem septos, constituídos por conjuntivo denso não modelado e frouxo, rico em células adiposas. O perineuro, que reveste grupo de feixes nervosos, formando os fascículos, possui de duas a quinze camadas de células justapostas concêntricas com núcleo fusiforme, citoplasma delgado e alongado, diferenciando duas camadas, uma externa e outra interna. O espaço interlaminar da camada externa é preenchido por fibras colágenas e o da camada interna por fibras colágenas e reticulares. O endoneuro é constituído por tecido conjuntivo frouxo que circunda axônios predominantemente mielinizados, apresentando fibras colágenas e reticulares. Analisou-se, também, no presente estudo, o arranjo dos fascículos nervosos, notando-se no segmento proximal média de 11,50 ï‚±ï€ 5,40 pequenos fascículos de diâmetros variados. O segmento médio apresentou média de 10,20 ï‚±ï€ 2,94 fascículos com diâmetros maiores que os do segmento proximal. O segmento distal apresentou média de 15,30 ï‚±ï€ 3,89 fascículos exibindo várias dimensões e menores que os observados no segmento médio. - Abstract
A gramínea B. decumbens é a mais cultivada em formação de pastagens no Brasil, em áreas são utilizadas para outros cultivos torna-se uma das plantas daninhas mais indesejadas, devido o seu difícil controle e rusticidade. Assim, o objetivo deste estudo foi determinar a emergência das plântulas de B. decumbens em profundidades de semeadura. Os ensaios foram realizados em casa de vegetação com as seguintes profundidades de semeadura (0,0; 0,5; 1,0; 1,5; 2,0; 2,5; 3,0; 4,0; 5,0; 6,0; 7,0; 8,0; 9,0 e 10,0 cm). A partir de 7,0 cm de profundidade ocorreu redução significativa no índice de velocidade de emergência (IVE) e na porcentagem de emergência das plântulas (%E) de B. decumbens. A emergência de plântulas de B. decumbens é afetada pela profundidade das sementes no perfil do solo.

- Abstract
O objetivo deste trabalho foi avaliar a histologia e a histoquímica do intestino anterior de tilápia do Nilo alimentadas com dietas contendo farinha de peixe ou silagem de peixe como fonte de proteína de origem animal. A espessura da vilosidade intestinal dos peixes alimentados com silagem fermentada de resíduo de tilápia foi influenciada pelos teores protéicos, independente das proporções de proteína de origem animal das dietas. Observou-se que a variação da intensidade de secreção de glicoproteínas pelas células caliciformes está diretamente ligada com o tipo de dieta fornecida aos animais. O intestino médio de O. niloticus apresentou diferenças no padrão de secreção de muco glicoproteíco neutro, glicoproteínas ácidas e glicoconjugados, dependendo da origem da proteína e da porcentagem utilizada na dieta, demonstrando que esta espécie pode adaptar seu sistema de secreção para a proteção do aparelho digestório durante a absorção diferentes fontes protéicas.

Uncaria tomentosa is a medicinal plant used in folk medicine by Amazon tribes. In this study the constituents of aqueous extract of U. tomentosa bark were quantified by chromatographic technique and its lethal concentration 50 (48h) in Hyphessobrycon eques was determined. The chromatography showed high levels of oxindole alkaloids, quinovic acid glycosides, and low molecular weight polyphenols. The CL50 48h was 1816 mg/L. Fish showed behavior changes at concentrations above 2000 mg/L, accompanied by a significant decrease of dissolved oxygen. At the highest concentration 100% mortality was observed attributed to oxygen reduction by the amount of oxindole alkaloids, polyphenols accumulation of the extract in the gills, and the interaction of these compounds with dopamine. In conclusion, the aqueous extract of U. tomentosa did not alter the chemical components and it was shown that U. tomentosa has low toxicity to H. eques; therefore, it can be used safely in this species. - Abstract
The features of paca epididymis, based on its appearance in light microscope, is described in this paper. The cellular population of the epithelial lining comprises principal cells, basal cells, apical cells, narrows cells, and hallo cells. The epididymis is divided in five distinct and continuous regions, Zone I, or initial segment, and zone II, are both localized into the head. Zone III comprises the distal head and all the body. Zones IV and V are restricted to the tail, in the proximal and distal cauda epididymis respectively. Each zone can be readily distinguished on the basis of morphological characteristics. The height of epididymal epithelium is greater in zone I. There is a progressive increase in the diameter of the tubular lumen through the different areas, with the maximum in the zone V. The presence of a high epithelium, and the virtual absence of sperm in zone I suggest fast transit of spermatozoa in this region. Zone V comprises the distal tail, has smaller epithelial lining, greater luminal diameter, shorter stereocilia than the other zones, and contains spermatozoa packed inside the lumen, that characterizes this zone as a place of sperm storage. The findings are compared with other reports in rodents and other domestic animals, to contribute to the understanding of epididymal morphophysiology.
