Objective: The purpose of this study was to explore the potential mechanism of Helicobacter pylori (Hp) eradication by probiotic therapy through 16S rRNA gene sequencing technology and untargeted metabolomics. Methods: Twenty four Hp-infected children were recruited from the Shanxi Bethune Hospital, and 24 healthy children were recruited as a blank control group. Group A: fecal samples from 24 healthy children. Group B: fecal samples of 24 children with Hp infection. Group B1 (n = 15): fecal samples of group B treated with probiotic therapy for 2 weeks. Group B2 (n = 19): fecal samples of group B treated with probiotic therapy for 4 weeks. The above fecal samples were analyzed by 16S rRNA gene sequencing technology and untargeted metabolomics. Results: There was no significant difference in alpha diversity and beta diversity among the four groups, but many bacteria with statistical difference were found in each group at the bacterial genus level and phylum level. LEfSe results showed that in group B, Porphyromonadaceae , Shigella and other microorganisms related to intestinal microecological dysbiosis were enriched. And in group B2, abundant characteristic microorganisms were found, namely Bacillales and Prevotella. KEGG metabolic pathway enrichment analysis showed that groups B1 and B2 were involved in 10 metabolic pathways potentially related to probiotic treatment: purine metabolism, nitrogen metabolism, arginine biosynthesis, alanine, aspartic acid and glutamate metabolism, glyoxylic acid and dicarboxylic acid metabolism, unsaturated fatty acid biosynthesis, fatty acid extension, fatty acid degradation, pyrimidine metabolism, fatty acid biosynthesis. Conclusion: Probiotic therapy can inhibit Hp to some extent and can relieve gastrointestinal symptoms, making it a preferred therapy for children with Hp infection and functional abdominal pain. Hp infection can reduce the diversity of intestinal microbes, resulting in the disturbance of intestinal microbiota and changes in the relative abundance of microbiota in children, while probiotic therapy can restore the diversity of intestinal microbes and intestinal microecological balance. In order to verify the advantage of CaO·6Al2O3 (CA6)‐based refractories on the inclusions of aluminum deoxidized steel, the five refractories, CA6, alumina, spinel, and CA6‐alumina and CA6‐spinel composition refractories were prepared into crucibles, and then the laboratory smelting experiments were conducted. After experiment, the slag resistance of the crucible and the variation on inclusions in steel were characterized and discussed. A dense CaO·2Al2O3 (CA2) layer, which was produced by CA6 reacting with the slag, was distributed between the original bricklayer and the slag layer, improving the slag resistance of refractories. Meanwhile, the 12CaO·7Al2O3 (C12A7), generated by the reaction between CA2 and refining slag, would release much Ca into the molten steel. The Ca would react with inclusions to produce low melting point substance to float up and remove, contributing to the reduction of the proportion of large size inclusions. In addition, typical inclusions in steel smelted with CA6 crucible were small‐sized MgO·Al2O3 inclusions, whereas those of other crucibles are MnS–MgO·Al2O3 composite inclusions with MgO·Al2O3 as the core, implying CA6 may absorb sulfur during the smelting process. Abstract Background Tumor necrosis factor alpha (TNF-α) plays a key role in the containment of tuberculosis. The relationship between the TNF -238G/A polymorphism and tuberculosis susceptibility remains inconclusive. A comprehensive meta-analysis was made to provide a more precise estimate of the relationship between them. Methods Multiple search strategies were used. A fixed effect model was takentook to estimate pooled OR with 95% confidence interval (CI) for the association between the TNF -238G/A polymorphism and tuberculosis susceptibility. The Chi-squared-based Q-test and I-squaredI2 statistic were calculated to examine heterogeneity. Begg’s funnel plot and Egger’s test were used to assess publication bias. Results 9 case-control studies were included in this meta-analysis. No significant heterogeneity was demonstrated, and no obvious publication bias was detected among the included studies. The meta-analysis indicated that there was no significant association between the TNF -238G/A polymorphism and tuberculosis susceptibility (GA+AA versus GG model: OR=1.005, 95% CI: 0.765-1.319; A versus G model: OR=1.000, 95% CI: 0.769-1.300). In the subgroup analyses by ethnicity, types of TB and human immunodeficiency virus (HIV) status, no significant association were identified. Conclusions The meta-analysis involving 2723 subjects did not detect any association between the TNF -238G/A polymorphism and tuberculosis susceptibility.

Background/aims: A large number of studies have shown that polymorphisms in the tumor necrosis factor-α (TNF-α, TNFA) gene are implicated in susceptibility to tuberculosis (TB). However, the results are inconsistent. We performed this meta-analysis to estimate the association between polymorphisms in the TNFA gene and TB susceptibility., Methods: Relevant studies published before March 2012 were identified by searching PubMed, ISI web of knowledge, EBSCO and CNKI. The strength of relationship between the TNFA gene and TB susceptibility was assessed using odds ratios (ORs)., Results: A total number of twenty-three case-control studies including 3630 cases and 4055 controls were identified referring to three previously chosen single-nucleotide polymorphisms (SNPs): -308G>A, -863C>A and -857C>T. No association was found between -308G>A, -863C>A and TB susceptibility: -308G>A (GG+GA vs. AA): OR 0.85, 95%CI: 0.55-1.30, P=0.44; -863C>A (CC+CA vs. AA): OR 0.93, 95%CI: 0.84-1.81, P=0.83. Increased risk of TB was associated with -857C>T in the dominant genetic model (CC+CT vs. TT: OR 2.13, 95%CI: 1.25-3.63, P=0.01), the heterozygote comparison (CT vs. TT: OR 2.69, 95%CI: 1.44-5.02, P=0.00) and the homozygote comparison (CC vs. TT: OR 2.08, 95%CI: 1.22-3.53, P=0.01) in Asian subjects., Conclusion: There is an increased association between TNFA -857C>T polymorphism and TB risk among Asian subjects. No association was found between -308G>A and -863C>A with TB risk.
