This study aims to identify key regulators of paraptosis in gastric cancer (GC) and explore their potential in guiding therapeutic strategies, especially in stomach adenocarcinoma (STAD). Genes associated with paraptosis were identified from the references and subjected to Cox regression analysis in the TCGA-STAD cohort. Using machine learning models, LPAR1 consistently ranked highest in feature importance. Multiple sequencing data showed that LPAR1 was significantly overexpressed in cancer-associated fibroblasts (CAFs). LPAR1 expression was significantly higher in normal tissues, and ROC analysis demonstrated its discriminative ability. Copy number alterations and microsatellite instability were significantly associated with LPAR1 expression. High LPAR1 expression correlated with advanced tumor grades and specific cancer immune subtypes, and multivariate analysis confirmed LPAR1 as an independent predictor of poor prognosis. LPAR1 expression was associated with different immune response metrics, including immune effector activation and upregulated chemokine secretion. High LPAR1 expression also correlated with increased sensitivity to compounds, such as BET bromodomain inhibitors I-BET151 and RITA, suggesting LPAR1 as a biomarker for predicting drug activity. FOXP2 showed a strong positive correlation with LPAR1 transcriptional regulation, while increased methylation of LPAR1 promoter regions was negatively correlated with gene expression. Knockdown of LPAR1 affected cell growth in most tumor cell lines, and in vitro experiments demonstrated that LPAR1 influenced extracellular matrix (ECM) contraction and cell viability in the paraptosis of CAFs. These findings suggest that LPAR1 is a critical regulator of paraptosis in GC and a potential biomarker for drug sensitivity and immunotherapy response. This underscores the role of CAFs in mediating tumorigenic effects and suggests that targeting LPAR1 could be a promising strategy for precision medicine in GC.

In order to study the change rules of mineral composition, micro-structure evolution characteristics and their relationship with dynamic properties of granite after high temperature treatment, the high temperature tests on Beishan granite in Gansu Province, China, were carried out at 20−1 000 ℃. Microscopic images of granite after high temperature treatment were obtained by slicing technique and polarizing microscope. The evolution of mineral composition, mineral content and micro-structure characteristic parameters of granite with temperature were studied through mineral composition analysis, micro-cracks characteristic identification and parameters calculation. Subsequently, dynamic impact compression tests of high temperature treated granite were carried out by split Hopkinson pressure bar (SHPB) system, to analyze the effects of temperature, impact rate and micro-structure characteristics on dynamic peak stress. On this basis, the quantitative relationship between micro-structure characteristics and macroscopic dynamic properties was established. The results show that high temperature has a significant effect on the mineral composition, micro-structure and impact compressive strength of granite, and 600 ℃ is taken as the threshold temperature. The loading rate also significantly affects the impact compressive strength, and the larger the loading rate is, the less the influence of temperature on the macroscopic dynamic properties is. The evolution process of the micro-structure of granite after high temperature treatment is the internal mechanism for the change of its macroscopic dynamic properties with temperature. The microcrack characteristics have a significant correlation with the dynamic peak stress, and the average crack width is the main factor affecting the dynamic peak stress of granite.

Introduction: Aconiti Lateralis Radix Praeparata (lateral roots of Aconitum carmichaelii Debeaux, Fuzi), is commonly used to treat various cardiovascular diseases, particularly heart failure. However, its strong cardiotoxicity limits its clinical applicability. Glycyrrhizae radix et rhizoma, (the root of Glycyrrhiza uralensis Fisch., Gancao), is known to synergistically increase the cardiotonic effects of Fuzi and alleviate the myocardial injury caused by Fuzi to some extent. However, the detailed mechanism via which the combination of Fuzi and Gancao reduces toxicity and increases or preserves the efficacy of Fuzi requires further investigation. Methods: Oxidative stress injury models in H9C2 cells and mice with acute heart failure were established to evaluate the optimal synergistic protective concentration of Fuziline and Glycyrrhetinic acid (GA). A GA probe was then synthesized and used for target fishing using chemical and biological methods. Finally, the target and its function were verified using fluorescence co-localization, Western blotting, protein interaction analysis, molecular docking, and calcium ion imaging. Results: The best pharmacodynamic potential was achieved with a 1:1 or 2:1 ratio of Fuziline and GA concentrations. At these ratios, they regulated the protein levels of the downstream players of the Ca2+ signaling pathway via MDH2 and CALR, thereby balancing Ca2+ homeostasis in the myocardial tissue and mitigating the effects of heart failure. Conclusion: This study aimed to investigate the compatibility of Fuziline and GA, the active metabolites of a traditional Chinese medicine (TCM) pair, in exerting their cardiac effects, identify the direct biological targets and verify the mechanism of compatibility. Revise ISO 22768∶2017 based on China′s industrial standards, and expand the scope of application to latex. The effects of sample preparation method, gas flow rate, heating rate and sample amount on the determination results of glass transition temperature were investigated, and the precision of the method was established. The results showed that, when the drying method was selected, the gas flow rate was 20 mL/min, the sample amount was 5-20 mg, and the heating rate was 20 ℃/min, the method had good precision, the standard deviation of repeatability was less than 0.30 ℃ and the standard deviation of reproducibility was less than 1.01 ℃.

The contents of soap and organic acid in emulsion polymerized styrene-butadiene rubber determined by indicator titration and potentiometric titration in ISO 7781 were compared. The differences between the determination principles and results of the two methods were compared, and the accuracy of the results was investigated by standard addition experiment. The results showed that the results of potentiometric titration and indicator method were consistent. When testing the soap content in emulsion polymerized styrene-butadiene rubber, the indicator method was greatly affected by amine antioxidant and filling oil. But the potentiometric titration method was less affected by amine antioxidant and was not affected by filling oil, so the determination result was more accurate.

The curing characteristics and tensile properties of sulfur and mercaptan modified chloroprene rubber(CR) were evaluated using carbon black free formulation were studied .The results showed that the specific surface area of magnesium oxide(MgO) affected the curing rate of CR, with the increase of the specific surface area of MgO, the curing efficiency improved. The specific surface area of MgO also effected the tensile properties of vulcanizated CR. The carbon black free formulation A and formulation B were applicable to evaluate curing characteristics and tensile properties of mercaptan modified CR, and the evaluation results may not be compareable. The carbon black free formulation was suitable for evaluating the curing characteristics and tensile properties of sulfur modified and mercaptan modified CR, and the precision of the test results could meet the requirements of ASTM D 3190—2006.

Soil thermal conductivity in the near-phase-transition zone is a key parameter affecting the thermal stability of permafrost engineering and its catastrophic thermal processes. Therefore, accurately determining the soil thermal conductivity in this specific temperature zone has important theoretical and engineering significance. In the present work, a method for testing the thermal conductivity of fine sandy soil in the near-phase-transition zone was proposed by measuring thermal conductivity with the transient plane heat source method and determining the volumetric specific heat capacity by weighing unfrozen water contents. The unfrozen water content of sand specimens in the near-phase-transition zone was tested, and a corresponding empirical fitting formula was established. Finally, based on the testing results, temperature variation trends and parameter influence laws of thermal conductivity in the near-phase-transition zone were analyzed, and thermal conductivity prediction models based on multiple regression (MR) and a radial basis function neural network (RBFNN) were also established. The results show the following: (1) The average error of the proposed test method in this work and the reference steady-state heat flow method is only 7.25%, which validates the reliability of the proposed test method. (2) The variation in unfrozen water contents in fine sandy soil in the range of 0~−3 °C accounts for over 80% of the variation in the entire negative temperature range. The unfrozen water content and thermal conductivity curves exhibit a similar trend, and the near-phase-transition zone can be divided into a drastic phase transition zone and a stable phase transition zone. (3) Increases in the thermal conductivity of fine sandy soil mainly occur the drastic phase transition zone, where these increases account for about 60% of the total increase in thermal conductivity in the entire negative temperature region. With the increase in density and total water content, the rate of increase in thermal conductivity in the drastic phase transition zone gradually decreases. (4) The R2, MAE, and RSME of the RBFNN model in the drastic phase transition zone are 0.991, 0.011, and 0.021, respectively, which are better than those of the MR prediction model. The 3D geological model serves as a cornerstone, establishing the essential foundation and prerequisite for numerical simulations, analytical assessments, and evaluative procedures that are integral to the disciplines of geotechnical and geological engineering. The task of integrating vast quantities of multi-source, heterogeneous modeling data, the absence of stratigraphic division standards to meet varying precision requirements, and addressing the application demands of 3D geological models beyond the geological domain, pose significant challenges to the endeavor of 3D geological modeling. This is especially true for the construction of large-scale, high-precision 3D geological models at the urban level. Our research introduces a methodology for 3D geological modelling, with a particular focus on the dual-weighted interpolation technique for the integration of land-sea data and the establishment of a three-tiered coding system for bedrock layers. Focusing on Qingdao, a city characterized by its bedrock geology and situated along the eastern coastal region of China, our proposed method has successfully catalyzed the creation of models across four distinct scales: a comprehensive city-wide model covering an area of 11,282 square kilometers, five general models, two demonstration models, and five refined models. Further application cases have demonstrated that these models are not only capable of meeting the requirements of geological and geotechnical engineering across various scenarios, but also exhibit excellent interoperability and significant application potential when integrated with Building Information Modeling (BIM), Geographic Information Systems (GIS), and Internet of Things (IoT) technologies. The outcomes of this study are significant for the theoretical development and technological advancement of 3D geological modeling, and can also bring inspiration and reference to the work of 3D geological modeling in other areas. As a result of global warming, the thawing settlement disasters of permafrost in the Qinghai–Tibet Engineering Corridor (QTEC) have intensified, which has serious effects on the safe operation of permafrost highway engineering. In this work, a prediction model for the thawing depth of permafrost subgrade in the QTEC under the climate warming scenario was established. Based on the survey results of permafrost ice content along the QTEC and the classification of thawing settlement risks, the zoning characteristics of thawing settlement of permafrost subgrade in the QTEC were obtained and analyzed. The results showed that the thawing depth of permafrost underlying the 26 m width subgrade in the QTEC will mainly remain below 9 m, and the area with a thawing depth of 6~9 m will have the widest spread within the next 20 years. The thawing settlement will be between 0.02 m and 5.45 m, with an average value of about 0.93 m after 20 years. Furthermore, after 50 years, the thawing depth of permafrost underlying the 26 m width subgrade will almost always be greater than 9 m, and the average thawing settlement will be about 1.12 m. Within the next 20 to 50 years, the risk of permafrost subgrade thawing settlement in the QTEC will be the most significant risk type, and this effect will mainly be distributed in the Kunlun Mountains, Chumar River Plain, Kekexili Mountains, Beiluhe Basin, Tanggula Mountains and intermountain Basins.
