17. Identification of Key Genes and Competitive Endogenous RNA Network Construction in Osteonecrosis of the Femoral Head by Integrated Bioinformatics Analysis

Abstract

Background: Despite cumulative evidence shows osteonecrosis of the femoral head (ONFH) could result in the progressive collapse of the femoral head. The pathogenesis of ONFH remains unclear. Early ONFH is difficult to diagnose due to the lack of effective biomarkers. Method: In Gene Expression Omnibus (GEO) database, we searched the Microarray datasets for serum (GSE123568) in ONFH and normal controls to identify differentially expressed genes (DEGs) by R software. The enrichment analyses were performed to enrich pathways of DEGs. Protein–protein interaction (PPI), miRNA-mRNA co-expression, ceRNA networks were constructed using Cytoscape to identity top15 hub genes, target miRNAs of hub genes and potential regulatory pathways. Furthermore, hub genes validated in GSE74089 with high diagnostic value for ONFH were selected as key genes. The Human Protein Atlas (HPA) and Bgee Database were used to find out the subcellular and tissue distribution of key genes.Results: A total of 568 DEGs were identified between 30 ONFH samples and 10 normal controls. Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology (GO) enrichment analysis showed that DEGs are mostly enriched in innate immune responses, thrombosis and signal transduction. Fifteen hub genes were identified by PPI network using Cytoscape. The 15 hub genes were almost all positively correlated with each other. The expression of PLEK (P＜0.001), TLR2 (P＜0.05), and TREM1 (P＜0.001) were validated in dataset GSE74089 and they had high diagnostic value (AUC＞0.8) for ONFH. MALAT1-miR-146b-5p-TLR2, MALAT1-miR-664b-3p-PLEK, NORAD-miR-106b-5p-TLR2, and MSMO1-miR-106b-5p-TLR2 might be potential RNA regulatory pathways in the disease progression of ONFH. PLEK mainly expressed in nucleus, TREM1 in dictyosome, and TLR2 in nucleoplasm and mitochondria.Conclusions: In this study, we found that PLEK, TLR2, and TREM1 might be potential biomarkers in diagnostic and play a vital role in the progression of ONFH.

27. Seawater Acidification Reduced the Resistance of <italic>Crassostrea gigas</italic> to <italic>Vibrio splendidus</italic> Challenge: An Energy Metabolism Perspective.

Abstract

Negative physiological impacts induced by exposure to acidified seawater might sensitize marine organisms to future environmental stressors, such as disease outbreak. The goal of this study was to evaluate if ocean acidification (OA) could reduce the resistance capability of the Pacific oyster (Crassostrea gigas) to Vibrio splendidus challenge from an energy metabolism perspective. In this study, the Pacific oyster was exposed to OA (pH 7.6) for 28 days and then challenged by V. splendidus for another 72 h. Antioxidative responses, lipid peroxidation, metabolic (energy sensors, aerobic metabolism, and anaerobic metabolism) gene expression, glycolytic enzyme activity, and the content of energy reserves (glycogen and protein) were investigated to evaluate the environmental risk of pathogen infection under the condition of OA. Our results demonstrated that following the exposure to seawater acidification, oysters exhibited an energy modulation with slight inhibition of aerobic energy metabolism, stimulation of anaerobic metabolism, and increased glycolytic enzyme activity. However, the energy modulation ability and antioxidative regulation of oysters exposed to seawater acidification may be overwhelmed by a subsequent pathogen challenge, resulting in increased oxidative damage, decreased aerobic metabolism, stimulated anaerobic metabolism, and decreased energy reserves. Overall, although anaerobic metabolism was initiated to partially compensate for inhibited aerobic energy metabolism, increased oxidative damage combined with depleted energy reserves suggested that oysters were in an unsustainable bioenergetic state and were thereby incapable of supporting long-term population viability under conditions of seawater acidification and a pathogen challenge from V. splendidus. 28. Effects of dietary supplementation of Clostridium butyricumH129 on growth performance and intestinal health of turbot, Scophthalmus maximus

Abstract

In the present study, the effect of Clostridium butyricumH129 on the growth performance and intestinal health of Scophthalmus maximuswas investigated in an indoor circulating aquaculture system during an 8-week trial period. Three hundred sixty healthy turbots were randomly divided into four groups fed with C. butyricumH129 at different concentrations (0, 106, 107, and 108CFU/g), respectively. In terms of terminal body weight, weight gain rate and specific growth rate of turbot, the CB2 (107CFU/g) group performed best. The amylase enzyme activity of the intestine was significantly increased in CB2 groups (p < 0.05). Moreover, it was demonstrated that C. butyricumincreased the thickness, width and height of the intestine epithelium of turbot, parallel with the significantly transcriptional up-regulation of tight junction proteins. Furthermore, the transcripts of proinflammatory cytokine were down-regulated and intestinal inflammation was inhibited by regulating the immune system in C. butyricum-administered groups, especially in CB2 group. As a result, C. butyricumH129 might enhance intestinal physical barrier function and anti-infection capacity in turbot. Moreover, dietary supplementation with C. butyricumH129 significantly improved the diversity of the intestinal microflora as well as increased the relative abundance of potential probiotic-related genera including Clostridialesand Bacillalesin turbot. In conclusion, dietary supplementation with C. butyricumH129 can enhance the mucosal barrier function, and actively regulate the intestinal microflora, resulting in improving the growth performance of S. maximus.
