Bacillus mojavensis KRS009 was identified as an antagonistic strain with a strong inhibitory effect on various phytopathogenic fungi. To provide further insight into its biocontrol mechanisms and ability to improve plant salt tolerance, the high quality complete genome of KRS009 was sequenced by PacBio platform and assembled using PacBio-HiFiReads. The KRS009 genome consists of one circular chromosome of 4,089,687 bp with 43.5% GC content and 4,062 open reading frames, of which 3,920 protein-coding genes, 86 transfer RNA, 10 ribosomal RNA, and 26 small RNA were identified in this genome. Among them, genes related to compatible solutes, including treC, galU, proX, and proW, are involved in the synthesis of trehalose, proline, and betaine metabolism and play an important role in relieving osmotic stress. In addition, the KRS009 genome contains genes associated with high salinity tolerance, including those responsible for Na+/H+ antiporters, K+ transporters, TrkH thiamin phosphate synthase, K+-sensing histidine kinase, aryl-phospho-beta-D-glucosidase, flavoprotein CzcO associated with the cation diffusion facilitator CzcD, and chaperonin GroEL. Together, the high-quality genome resource of strain KRS009 would provide a molecular basis for further research on its biocontrol and plant salt tolerance mechanisms. Abstract Background Verticillium wilt, caused by the fungus Verticillium dahliae, is a soil-borne vascular fungal disease, which has caused great losses to cotton yield and quality worldwide. The strain KRS010 was isolated from the seed of Verticillium wilt-resistant Gossypium hirsutum cultivar “Zhongzhimian No. 2.” Results The strain KRS010 has a broad-spectrum antifungal activity to various pathogenic fungi as Verticillium dahliae, Botrytis cinerea, Fusarium spp., Colletotrichum spp., and Magnaporthe oryzae, of which the inhibition rate of V. dahliae mycelial growth was 73.97% and 84.39% respectively through confrontation test and volatile organic compounds (VOCs) treatments. The strain was identified as Bacillus altitudinis by phylogenetic analysis based on complete genome sequences, and the strain physio-biochemical characteristics were detected, including growth-promoting ability and active enzymes. Moreover, the control efficiency of KRS010 against Verticillium wilt of cotton was 93.59%. After treatment with KRS010 culture, the biomass of V. dahliae was reduced. The biomass of V. dahliae in the control group (Vd991 alone) was 30.76-folds higher than that in the treatment group (KRS010+Vd991). From a molecular biological aspect, KRS010 could trigger plant immunity by inducing systemic resistance (ISR) activated by salicylic acid (SA) and jasmonic acid (JA) signaling pathways. Its extracellular metabolites and VOCs inhibited the melanin biosynthesis of V. dahliae. In addition, KRS010 had been characterized as the ability to promote plant growth. Conclusions This study indicated that B. altitudinis KRS010 is a beneficial microbe with a potential for controlling Verticillium wilt of cotton, as well as promoting plant growth.

Bacillus altitudinis strain KRS010 was isolated from the seed of Verticillium wilt-resistant Gossypium hirsutum cultivar Zhongzhimian No. 2. An in vitro assay revealed that the isolate KRS010 exhibited significant antagonistic activity against Verticillium dahliae. Strain KRS010 could be used as a potential biocontrol agent against Verticillium wilt. The KRS010 genome consisted of a single circular chromosome 3,753,708 bp in length, with an average GC content of 41.85%. Genome annotation predicted 3,519 protein-coding genes and 128 noncoding RNA. Improving soil quality while achieving higher productivity is the major challenge in the agricultural industry. Wheat (Triticum aestivum L.)–maize (Zea mays L.) (W–M) rotation is the dominant planting pattern in the Huang-Huai-Hai Plain and is important for food security in China. However, the soil quality is deteriorating due to the W–M rotation's long-term, intensive, and continuous cultivation. Introducing legumes into the W–M rotation system may be an effective way to improve soil quality. In this study, we aimed to verify this hypothesis by exploring efficient planting systems (wheat–peanut (Arachis hypogaea L.) (W–P) rotation and wheat rotated with maize and peanut intercropping (W–M/P)) to achieve higher agricultural production in the Huang-Huai-Hai Plain. Using traditional W–M rotation as the control, we evaluated crop productivity, net returns, soil microorganisms (SMs), and soil organic carbon (SOC) fractions for three consecutive years. The results indicated that wheat yields were significantly increased under W–P and W–M/P (382.5–579.0 and 179.8–513.1 kg ha−1, respectively) compared with W–M. W–P and W–M/P provided significantly higher net returns (58.2 and 70.4%, respectively) than W–M. W–M/P and W–M retained the SOC stock more efficiently than W–P, increasing by 25.46–31.03 and 14.47–27.64%, respectively, in the 0–20 cm soil layer. Compared with W–M, W–M/P improved labile carbon fractions; the sensitivity index of potentially mineralizable carbon, microbial biomass carbon (MBC), and dissolved organic carbon was 31.5, 96.5–157.2, and 17.8% in 20–40, 10–40, and 10–20 cm soil layers, respectively. The bacterial community composition and bacteria function were altered as per the soil depth and planting pattern. W–M/P and W–M exhibited similar bacterial community composition and function in 0–20 and 20–40 cm soil layers. Compared with W–P, a higher abundance of functional genes, namely, contains mobile elements and stress-tolerant, and a lower abundance of genes, namely, potentially pathogenic, were observed in the 10–20 cm soil layer of W–M and the 0–20 cm soil layer of W–M/P. SOC and MBC were the main factors affecting soil bacterial communities, positively correlated with Sphingomonadales and Gemmatimonadales and negatively correlated with Blastocatellales. Organic input was the main factor affecting SOC and SMs, which exhibited feedback effects on crop productivity. In summary, W–M/P improved productivity, net returns, and SOC pool compared with traditional W–M rotation systems, and it is recommended that plant–soil–microbial interactions be considered while designing high-yield cropping systems.

Abstract Diabetic nephropathy (DN) represents a prevalent chronic microvascular complication of diabetes mellitus (DM) and is a major cause of end‐stage renal disease. The anfractuous surrounding of DN pathogenesis and the intricate nature of this metabolic disorder often pose challenges in both the diagnosis and treatment of DN compared to other kidney diseases. Hyperglycaemia in DM predispose vulnerable renal cells into microenvironmental disequilibrium and thereby results in innate immunocytes infiltration including neutrophils, macrophages, myeloid‐derived suppressor cells, dendritic cells, and so forth. These immune cells play dual roles in kidney injury and closely correlated with the degree of proteinuria in DN patients. Additionally, innate immune signaling cascades, initiated by altered metabolic and hemodynamic in diabetic context, are crucial in instigating and perpetuating renal inflammation, which detrimentally contribute to DN pathogenesis. As such, anti‐inflammatory therapies, particularly those targeting innate immunity, hold renoprotective promise in DN. In this article, we reviewed the origin and feature of the above four prominent kidney innate immune cells, analyze their pathogenic role in DN, and discuss potential targeted‐therapeutic strategies, aiming to enhance the current understanding of renal innate immunity and hence help to discover promising therapeutic approaches for DN.

Functional nano-coatings have been a research hotspot in recent years. The nano-size composition units provide good volume effect and surface effect for functional nano-coatings, and greatly optimize the mechanical properties, corrosion resistance, oxidation resistance and wear resistance of the coatings. The preparation technology and properties of functional nano-coatings determine the application range of nano-coatings. However, most reviews focus on certain properties, preparation methods or applications of functional nano-coatings, and lack of reviews on the integrity of functional nano-coatings. Firstly, this work introduced the concept and classification of functional nano-coatings briefly. Secondly, the preparation techniques of functional nano-coatings were described in detail, including traditional chemical vapor deposition, physical vapor deposition and sol-gel method, the latest plasma spraying technology and other preparation techniques. Moreover, the principles, advantages and disadvantages of various nano-coatings preparation technologies and the application of nano-coatings in materials were analyzed and summarized. Finally, the latest and popular applications of functional nano-coatings were reviewed in detail, and the application prospect and potential of functional nano-coatings were prospected.

Objective To investigate the prevalence of depression and anxiety in patients undergoing maintenance hemodialysis (MHD) in Hohhot, a large city on the northern border of China, and to identify independent risk factors for depression and anxiety in these patients.Methods Patients receiving MHD for >3 months were enrolled in the four largest hemodialysis centers between September 2020 and December 2020. Depression and anxiety were assessed using the Zung self-rated depression scale (SDS) and Zung self-rated anxiety scale (SAS), respectively, with demographic and other data collected for logistic regression analyses.Results Among 305 MHD patients included in this study, the prevalence of depression was 55.1%, including 27.5%, 21.0%, and 6.6% with mild, moderate and severe cases, respectively. The prevalence of anxiety was 25.9%, with 20.0%, 4.6%, and 1.3% having mild, moderate, and severe cases, respectively. To establish a safe, efficient, and simple biocontrol measure for gray mold disease caused by Botrytis cinerea, the basic characteristics and antifungal activity of KRS005 were studied from multiple aspects including morphological observation, multilocus sequence analysis and typing (MLSA–MLST), physical-biochemical assays, broad-spectrum inhibitory activities, control efficiency of gray mold, and determination of plant immunity. The strain KRS005, identified as Bacillus amyloliquefaciens, demonstrated broad-spectrum inhibitory activities against various pathogenic fungi by dual confrontation culture assays, of which the inhibition rate of B. cinerea was up to 90.3%. Notably, through the evaluation of control efficiency, it was found that KRS005 fermentation broth could effectively control the occurrence of tobacco leaves gray mold by determining the lesion diameter and biomass of B. cinerea on tobacco leaves still had a high control effect after dilution of 100 folds. Meanwhile, KRS005 fermentation broth had no impact on the mesophyll tissue of tobacco leaves. Further studies showed that plant defense-related genes involved in reactive oxygen species (ROS), salicylic acid (SA), and jasmonic acid (JA)-related signal pathways were significantly upregulated when tobacco leaves were sprayed with KRS005 cell-free supernatant. In addition, KRS005 could inhibit cell membrane damage and increase the permeability of B. cinerea. Overall, KRS005, as a promising biocontrol agent, would likely serve as an alternative to chemical fungicides to control gray mold.

Abstract Background Both extracellular vesicles from mesenchymal stromal cell-derived human umbilical cords (hUC-EVs) and arsenic trioxides (ATOs) have been demonstrated to treat acute graft-versus-host disease (aGVHD) via immunomodulation. Apart from immunomodulation, hUC-EVs have a unique function of drug delivery, which has been proposed to enhance their efficacy. In this study, we first prepared ATO-loaded hUC-EVs (hUC-EVs-ATO) to investigate the therapeutic effect and potential mechanisms of hUC-EVs-ATO in a mouse model of aGVHD after allogeneic hematopoietic stem cell transplantation (HSCT). Methods An aGVHD model was established to observe the therapeutic effects of hUC-EVs-ATO on aGVHD. Target organs were harvested for histopathological analysis on day 14 after transplantation. The effects of hUC-EVs-ATO on alloreactive CD4+ were evaluated by flow cytometry in vivo and in vitro. Flow cytometry, RT-PCR, immunofluorescence colocalization analysis and Western blot (Wb) analysis were performed to examine macrophage polarization after hUC-EV-ATO treatment. The cytokines in serum were measured by a cytometric bead array (CBA). TEM, confocal microscopy and Wb were performed to observe the level of autophagy in macrophages. A graft-versus-lymphoma (GVL) mouse model was established to observe the role of hUC-EVs-ATO in the GVL effect. Results The clinical manifestations and histological scores of aGVHD in the hUC-EVs-ATO group were significantly reduced compared with those in the ATO and hUC-EVs groups. The mice receiving hUC-EVs-ATO lived longer than the control mice. Notably, hUC-EVs-ATO interfering with alloreactive CD4+ T cells differentiation were observed in aGVHD mice but not in an in vitro culture system. Additional studies showed that depletion of macrophages blocked the therapeutic effects of hUC-EVs-ATO on aGVHD. Mechanistically, hUC-EVs-ATO induced autophagic flux by inhibiting mammalian target of rapamycin (mTOR) activity to repolarize M1 to M2 macrophages. Additionally, using a murine model of GVL effects, hUC-EVs-ATO were found not only to reduce the severity of aGVHD but also to preserve the GVL effects. Taken together, hUC-EVs-ATO may be promising candidates for aGVHD treatment. Conclusions hUC-EVs-ATO enhanced the alleviation of aGVHD severity in mice compared with ATO and hUC-EVs without weakening GVL activity. hUC-EVs-ATO promoted M1 to M2 polarization via the mTOR-autophagy pathway. hUC-EVs-ATO could be a potential therapeutic approach in aGVHD after allo-HSCT.

Abstract Active crosstalk between the nervous system and breast cancer cells has been experimentally demonstrated in vitro and in animal models. However, low frequencies of peripheral nerve presence in human breast cancers reported in previous studies (~30% of cases) potentially negate a major role of the nervous system in breast cancer development and progression. This study aimed to clarify the incidence of nerves within human breast cancers and to delineate associations with clinicopathological features. Immunohistochemical staining was conducted in formalin‐fixed paraffin‐embedded breast cancer tissue sections using antibodies against the pan‐neuronal markers protein gene product 9.5 and growth‐associated protein 43, and the sympathetic nerve‐specific marker tyrosine hydroxylase. Nerve trunks and isolated nerve fibers were quantitated. The chi‐squared test was used to determine the associations between nerve counts and clinicopathological parameters. The log‐rank test was used to compare differences in patient progression‐free survival (PFS) and overall survival (OS). The overall frequency of peripheral nerves in breast cancers was 85%, a markedly higher proportion than reported previously. Of note, most nerves present in breast cancers were of the sympathetic origin. While high density of nerve trunks or isolated nerve fibers was associated with poor PFS and OS of patients, high nerve trunk density appeared also to predict poor patient PFS independently of lymph node metastasis. Innervation of breast cancers is a common event correlated with poor patient outcomes. These findings support the notion that the nervous system plays an active role in breast cancer pathogenesis.

Currently available deep learning methods for sound source localization encounter the problems of poor accuracy, high complexity, and susceptibility to environmental interference. To solve these problems, this paper proposes an icosahedral feature-based method of sound source localization that uses hybrid dilated convolutions. The network uses icosahedral convolutional features of the phase-transformed steering response power as input features. A downsampling module consisting of the icosahedral convolution, dilated convolution, and a normalization layer is stacked to form the main structure of the network. In addition, the hybrid dilated convolution is used to enhance the receptive field and acquire multi-scale spatial information for accurate sound source localization. The proposed method was compared with several state-of-the-art models of sound source localization on multiple tasks from the LOCATA database. When tested on audio signals without and with silent segments, the direction of arrival (DOA) obtained by the proposed model had root mean-squared angular errors of 6.34° and 7.15°, respectively, at spherical distances. Both these results were superior to those of advanced models from the literature, and this shows that the proposed method is more accurate and robust than currently used techniques. We propose a scheme for realizing broadband and tunable transmission non-reciprocity by utilizing two-photon near-resonant transitions in thermal atoms as single-photon far-detuned transitions can be eliminated. Our basic idea is to largely reduce the Doppler broadenings on a pair of two-photon, probe and coupling, transitions and meanwhile make the only four-photon transition Doppler-free (velocity-dependent) for a forward (backward) probe field. One main advantage of this scheme lies in that the transmission non-reciprocity can be realized and manipulated in a frequency range typically exceeding 200 MHz with isolation ratio above 20 dB and insertion loss below 1.0 dB by modulating an assistant field in frequency and amplitude. The intersecting angle between four applied fields also serves as an effective control knob to optimize the nonreciprocal transmission of a forward or backward probe field, e.g. in a much wider frequency range approaching 1.4 GHz.

Congenital nasolacrimal duct obstruction(CNLDO)has an incidence rate of 5.7% and there are many clinical treatments for the disease. But these treatments have been taken very randomly. Therefore, it is still a controversial topic on how to choose the best treatment. This paper will discuss the treatments of CNLDO so as to know the characteristics of each treatment for better clinical services.

A two-dimensional multi-element phase grating has been designed in terms of the offset refractive index to exhibit the spatially odd symmetry ( antisymmetry ) along one transparent diagonal line with an even number of rectangular elements while leaving other elements in a unit cell opaque. This grating can be engineered to attain a few intriguing phenomena of asymmetric diffraction, including the elimination of equally spaced oblique diffraction lines, the elimination of alternately crossed oblique diffraction lines, and the selection of equally spaced oblique diffraction lines. These phenomena of engineered asymmetric diffraction are well explained via destructive interference between transmitted field amplitudes from paired, dual-paired, and successive elements along the transparent diagonal line.

Due to the complexity and non-stationarity of the voice generation system, the nonlinearity of speech signals cannot be accurately quantified. Recently, the recurrence quantification analysis method has been used for voice disorder detection. In this paper, multiscale recurrence quantification measures (MRQMs) are proposed. The signals are reconstructed in the high-dimensional phase space at the equivalent rectangular bandwidth scale. Recurrence plots (RPs) combining the characteristics of human auditory perception are drawn with an appropriate recurrence threshold. Based on the above, the nonlinear dynamic recurrence features of the speech signal are quantized from the recurrence plot of each frequency channel. Furthermore, this paper explores the recurrence quantification thresholds that are most suitable for pathological voices. Our results show that the proposed MRQMs with support vector machine (SVM), random forest (RF), Bayesian network (BN) and Local Weighted Learning (LWL) achieve an average accuracy of 99.45%, outperforming traditional features and other complex measurements. In addition, MRQMs also have the potential for multi-classification of voice disorder, achieving an accuracy of 89.05%. This study demonstrates that MRQMs can characterize the recurrence characteristic of pathological voices and effectively detect voice disorders.

Phase gratings, each period of which consists of even numbers of equal-width and equal-thickness elements, can be devised to attain asymmetric diffraction patterns. We show that engineering of offset refractive indices in different elements exhibiting a spatially odd symmetry leads to the elimination of a single diffraction order (directional elimination), while further manipulations on offset refractive indices may lead to the elimination of all odd or even orders (grouped elimination) or all orders but one or two selected (directional selection). These intriguing effects arise from destructive interference between diffracted amplitudes contributed by paired or successive elements, and violation of Friedel's law in such transparent gratings is an effect of higher order multiple scattering.

We report here the purification of a novel metal-binding protein from Oratosquilla oratoria (O. oratoria MT-1) by gel and ion-exchange chromatography. SDS-PAGE and MALDI-TOF analyses demonstrated that isolated O. oratoria MT-1 was of high purity with a molecular weight of 12.4 kDa. The fluorescence response to SBD-F derivatives revealed that O. oratoria MT-1 contained a large number of sulfhydryl groups, which is a general property of metallothioneins. Zn and Cu metal stoichiometries for O. oratoria MT-1 were 3.97:1 and 0.55:1, respectively. The proportion of cysteine (Cys) residues in the amino acid composition was 32.69%, and aromatic amino acids were absent. The peptide sequence coverage with Macrobrachium rosenbergii calmodulin (accession AOA3S8FSK5) was 60%. Infrared spectroscopy of O. oratoria MT-1 revealed two obvious peaks at absorption frequencies for the amide I band and the amide II band. CD spectra revealed that the secondary structure was mainly composed of random coil (57.6%) and β-sheet (39.9%). An evaluation of in vitro antioxidant activity revealed that isolated O. oratoria MT-1 has strong reducing activities, exhibiting scavenging rates for DPPH and OH of 77.8% and 75.8%, respectively (IC50 values 0.57 mg/mL and 1.1 mg/mL). O. oratoria MT-1 may be used as a functional additive in cosmetics, health foods, and medical products, as well as a reference material for quantitative analysis of metallothionein in such products.

Phytoene synthase (PSY) plays a key role in the biosynthesis and accumulation of carotenoids as the first rate-limiting enzyme in the plant carotenoid pathway. In the present study, the ‘Granny Smith’ apple PSY gene, MdPSY, was cloned using in silico cloning. The length of the MdPSY was 1500 bp, which included a complete open reading frame of 1194 bp, encoding a predicted protein of 397 amino acids. Sequence analysis revealed that the ‘Granny Smith’ apple PSY protein shared high homology with MdPSY2 (99%), MdPSY1 (93.9%), FaPSY (71.8%), CuPSY (70.7%) and NtPSY (69.7%). Phylogenetic analysis showed that the MdPSY protein was closely related to the PSY protein of Rosaceae. An expression profile analysis of different tissues indicated that MdPSY expression in the roots was higher than that in other tissues. MdPSY expression was tested throughout the fruit development period. The results showed that MdPSY plays a role in carotenoid biosynthesis in ‘Granny Smith’ apple. In addition, bagging treatment inhibits PSY gene expression, which in turn affects the synthesis and accumulation of carotenoids.

Abstract We theoretically study the optically tunable gratings based on a L-type atomic medium using coherent population oscillations from the angle of reflection and transmission of the probe field. Adopting a standing-wave driving field, the refractive index of the medium as well as the absorption are periodically modified. Consequently, the Bragg scattering causes the effective reflection. We show that different intensities of the control field lead to three types of reflection profile which actually correspond to different absorption/amplification features of the medium. We present a detailed analyses about the influence of amplification on the reflection profile as well. The coherent population oscillation is robust to the dephasing effect, and such induced gratings could have promising applications in nonlinear optics and all-optical information processing.

We investigate the four-wave mixing (FWM) nonlinearity in an ensemble of cold Rydberg atoms with each of them regarded as a double-ladder system. The interaction is studied from the view of generating a signal field in virtue of three applied lasers. Using an approach beyond mean-field theory, we solved the equations for the one-body and two-body correlators under perturbation, and show that the system possesses not only a local FWM nonlinearity, but also a much larger nonlocal nonlinearity due to the Rydberg–Rydberg interaction which can be further strengthened by increasing the atomic density. The results obtained may have promising applications in the quantum information processes involving the FWM nonlinearity, such as the generation of squeezed or entangled states.

As an unconventional resource, oil shale possesses abundant reserves and significant potential for industrial applications. The rational and efficient development of oil shale resources holds immense importance in reducing national energy demand. In-situ catalytic technology, characterized by its high efficiency, low pollution, and minimal energy consumption, represents a key direction for future oil shale development. This paper provides a comprehensive review of research progress in in-situ oil shale mining technology, oil shale pyrolysis catalysts, the pyrolysis mechanism of kerogen, and the compatibility of different heating processes and catalysts. Furthermore, the paper proposes future research directions and prospects for oil shale in-situ catalytic technology, including reservoir modification, highefficiency catalyst synthesis, injection processes, and high-efficiency heating technology. Abstract Background Xiao’er Qixingcha (EXQ) has been extensively applied to relieve dyspepsia and constipation in children for hundreds of years in China. However, the therapeutic mechanism underlying its efficacy remained to be defined. The present study aimed to clarify the possible laxative and immune-regulating effects of EXQ on two models of experimental constipation in mice, which mimicked the pediatric constipation caused by high-heat and high-protein diet (HHPD). Methods The two models of constipated mice were induced by HHPD or HHPD + atropine respectively. To investigate the laxative and immune-regulating activities of EXQ, animals were treated with three doses of EXQ (0.75, 1.5 and 3 g/kg) for 7 consecutive days. The fecal output parameters (number and weight), weight of intestinal content and, the thymus and spleen indexes were measured. The levels of sIgA, IL-10, TNF-α and LPS in colon and serum were determined by ELISA. Furthermore, the pathological changes of colon tissue were examined after routine H&E staining. Results Both HHPD and HHPD + atropine treatments obviously inhibited the fecal output and reduced the colonic sIgA, prominently increased the levels of IL-10 and TNF-α in colonic tissue and elevated the contents of LPS in serum and colonic tissues. In contrast, oral administration of EXQ significantly improved the feces characters and dose-dependently decreased the intestinal changes in both models. In HHPD model test, EXQ efficaciously boosted the sIgA level in a dose-dependent manner, significantly elicited decreases in TNF-α and IL-10 levels, and evidently decreased the spleen and thymus indexes. In HHPD + atropine model test, EXQ treatment reversed the pathological changes by not only dramatically decreasing the spleen index and the levels of LPS and IL-10, but also markedly elevating the thymus index. Furthermore, microscopic observation revealed that EXQ treatment maintained the integrity of colonic mucosa, and protected the colonic tissues from inflammation in the both models. Conclusions EXQ exhibited prominent laxative activity and effectively protected the colonic mucosal barrier in two models of constipated mice, of which the mechanism might be closely associated with its propulsive and immune-regulating properties. The current results not only validated the rationale for the clinical application of EXQ in pediatric constipation related symptoms, but also threw new light on the immune-inflammatory responses accompanied with chronic constipation pathology.

In this paper, we propose a method of extracting features from ECG signals based on the sparse representation of double-threshold Stagewise orthogonal matching Pursuit. First, the ECG signal was de-noised and then the K singular value decomposition algorithm (KSVD) was used to iterate the ECG data set to obtain a supercomplete dictionary of ECG signals. Then, through the double-threshold Stagewise orthogonal matching Pursuit, a sparse atomic matrix with the best reconstruction effect is finally selected to obtain the characteristics of ECG signals. Normal, atrial fibrillation, and ventricular fibrillation ECG signals from MIT ’ s ECG Signal Database (MIT-BIH) were used to evaluate the proposed method. Experimental results show that in the binary classification experiment, the comprehensive recognition rate of the proposed algorithm for normal ECG signals, ventricular fibrillation and atrial fibrillation is 93.96%, 1.26% higher than that of CNN, and 3.99% higher than that of wavelet transform. Meanwhile, the five parameters of the proposed algorithm, such as comprehensive Kappa and comprehensive root relative square error, are the best. In addition, this paper carries out three-classification recognition of normal, atrial fibrillation, and ventricular fibrillation signals. The average recognition rate of the proposed algorithm among the three classifiers is 89.17%. The average recognition rate is 3.2% and 6.67% higher than that of convolutional neural networks and wavelet transform, respectively. Objective To investigate the distribution of abnormal neurological activity of patients with recurrent optic neuritis (ON) by resting-state functional magnetic resonance imaging (fMRI), and to explore the relationship between baseline neurological function and clinic. Methods Resting-state fMRI were collected from 35 patients with recurrent ON and 50 normal controls who were matched in sex, age and education with patients. The amplitude of low-frequency fluctuation (ALFF) difference was investigated and compared between 2 groups. The correlation of brain regions with significantly different mALFF with duration, Paced Auditory Serial Addition Test (PASAT) and vision was further explored. Results Compared to normal controls, patients with recurrent ON showed significantly decreased mALFF in left cuneus/precuneus, left superior temporal gyrus, bilateral inferior temporal gyri, bilateral lingual gyri, right middle occipital gyrus (P < 0.01), and significantly increased mALFF in bilateral inferior frontal gyri and left medial frontal gyrus (P < 0.01). The positive correlation between duration and mALFF in left superior temporal gyrus (r = 0.403, P < 0.05) and right lingual gyrus (r = 0.472, P < 0.05) was observed in recurrent ON patients. Conclusions Resting-state fMRI can detect decreased neurological function in the temporal and occipital lobes which are involved in visual processing and increased neurological function in the frontal lobe which is involved in default mode network (DMN) in recurrent ON. Ulcerative colitis (UC) is a chronic relapsing disease without satisfactory treatments, in which intestinal inflammation and disrupted intestinal epithelial barrier are two main pathogeneses triggering UC. Berberrubine (BB) is deemed as one of the major active metabolite of berberine (BBR), a naturally-occurring isoquinoline alkaloid with appreciable anti-UC effect. This study aimed to comparatively investigate the therapeutic effects of BB and BBR on dextran sodium sulfate (DSS)-induced mouse colitis model, and explore the potential underlying mechanism. Results revealed that BB (20 mg/kg) produced a comparable therapeutic effect as BBR (50 mg/kg) and positive control sulfasalazine (200 mg/kg) by significantly reducing the disease activity index (DAI) with prolonged colon length and increased bodyweight as compared with the DSS group. BB treatment was shown to significantly ameliorate the DSS-induced colonic pathological alternations and decreased histological scores. In addition, BB markedly attenuated colonic inflammation by alleviating inflammatory cell infiltration and inhibiting myeloperoxidase (MPO) and cytokines (TNF-α, IFN-γ, IL-1β, IL-6, IL-4 and IL-10) productions in DSS mice. Furthermore, BB treatment substantially upregulated the expression of tight junction (TJ) proteins (zonula occludens-1, zonula occludens-2, claudin-1, occludin) and mRNA expression of mucins (mucin-1 and mucin-2), and decreased the Bax/Bcl-2 ratio. In summary, BB exerted similar effect to its analogue BBR and positive control in attenuating DSS-induced UC with much lower dosage and similar mechanism. The protective effect observed may be intimately associated with maintaining the integrity of the intestinal mucosal barrier and mitigating intestinal inflammation, which were mediated at least partially, via favorable modulation of TJ proteins and mucins and inhibition of inflammatory mediators productions in the colonic tissue. This is the first report to demonstrate that BB possesses pronounced anti-UC effect similar to BBR and sulfasalazine with much smaller dosage. BB might have the potential to be further developed into a promising therapeutic option in the treatment of UC.

Neuromyelitis optica spectrum disorders (NMOSDs) include classic neuromyelitis optica (NMO), opticospinal multiple sclerosis (OSMS), limited form of NMO and isolated optic neuritis or myelitis accompanied by either systemic autoimmune diseases or typical MRI findings of NMO. The common neuro-ophthalmic features of NMOSDs include simultaneous or consecutive bilateral optic neuritis, more commonly seen optic disk edema and surrounding exudate, poor visual recovery, steroid responsiveness and dependency. Combined with serum aquaporin 4 (AQP4) antibody and brain MRI examination, these clinical features can be helpful to the early differential diagnosis between NMOSDs and MS. Some types of eye movement abnormalities have been reported in patients with NMOSDs, but further investigation needs to be done before the specificity of these features are confirmed. doi: 10.3969/j.issn.1672-6731.2014.10.003

AIM:To investigate triple channel double ring guided treatment of lacrimal obstruction and its clinical therapeutic effect. METHODS: Fifty cases(54 eyes)of lacrimal obstruction were treated by laser or micro-drill under triple channel lacrimal endoscope with double ring guided intubation created by our department. The silicone tube was extracted at 3～6mo of postoperation. All cases were followed up 3～6mo. RESULTS:Of total 54 eyes, 47 eyes were cured, and lacrimal passage was clearly flushed without epiphora; 5 eyes were improved, but lacrimal passage was clearly flushed with slight epiphora; 2 eyes were recurred, and lacrimal passage was not clearly flushed with epiphora. Total effective rate was 96%. CONCLUSION:Triple channel double ring guided treatment of lacrimal obstruction has greatly improved traditional operation. It has dramatically increased success rate of lacrimal obstruction operation with little invasion, avoiding formation of false lacrimal passage, quick recovery and less complications.

Propagation measurements of wireless channels performed in the tunnel environments at 6 GHz are presented in this paper. Propagation characteristics are simulated and analyzed based on the method of shooting and bouncing ray tracing/image (SBR/IM). A good agreement is achieved between the measured results and simulated results, so the correctness of SBR/IM method has been validated. The measured results and simulated results are analyzed in terms of path loss models, received power, root mean square (RMS) delay spread, Ricean K-factor, and angle of arrival (AOA). The omnidirectional path loss models are characterized based on close-in (CI) free-space reference distance model and the alpha-beta-gamma (ABG) model. Path loss exponents (PLEs) are 1.50–1.74 in line-of-sight (LOS) scenarios and 2.18–2.20 in non-line-of-sight (NLOS) scenarios. Results show that CI model with the reference distance of 1 m provides more accuracy and stability in tunnel scenarios. The RMS delay spread values vary between 2.77 ns and 18.76 ns. Specially, the Poisson distribution best fits the measured data of RMS delay spreads for LOS scenarios and the Gaussian distribution best fits the measured data of RMS delay spreads for NLOS scenarios. Moreover, the normal distribution provides good fits to the Ricean K-factor. The analysis of the abovementioned results from channel measurements and simulations may be utilized for the design of wireless communications of future 5G radio systems at 6 GHz.

Severe burn-induced liver damage and dysfunction is associated with endoplasmic reticulum (ER) stress. ER stress has been shown to regulate global protein synthesis. In the current study, we induced ER stress in vitro and estimated the effect of ER stress on hepatic protein synthesis. The aim was two-fold: (1) to establish an in vitro model to isotopically measure hepatic protein synthesis and (2) to evaluate protein fractional synthetic rate (FSR) in response to ER stress. Human hepatocellular carcinoma cells (HepG2) were cultured in medium supplemented with stable isotopes 1,2-13C2-glycine and L-[ring-13C6]phenylalanine. ER stress was induced by exposing the cells to 100 nM of thapsigargin (TG). Cell content was collected from day 0 to 14. Alterations in cytosolic calcium were measured by calcium imaging and ER stress markers were confirmed by Western blotting. The precursor and product enrichments were detected by GC-MS analysis for FSR calculation. We found that the hepatic protein FSR were 0.97±0.02 and 0.99±0.05%/hr calculated from 1,2-13C2-glycine and L-[ring-13C6]phenylalanine, respectively. Our goal was to assess the validity of the enrichments of plasma free palmitate and intramuscular (IM) fatty acid metabolites as precursors for calculating the IM triglyceride fractional synthetic rate. We infused U-13C16-palmitate in anesthetized rabbits for 3 h and sampled adductor muscle of legs using both freeze-cut and cut-freeze approaches. We found that IM free palmitate enrichment (0.70 ± 0.07%) was lower (P < 0.0001) than IM palmitoyl-CoA enrichment (2.13 ± 0.17%) in samples taken by the freeze-cut approach. The latter was close (P = 0.33) to IM palmitoyl-carnitine enrichment (2.42 ± 0.16%). The same results were obtained from the muscle samples taken by the cut-freeze approach, except the enrichment of palmitoyl-CoA (2.21 ± 0.08%) was lower (P = 0.02) than that of palmitoyl-carnitine (2.77 ± 0.17%). Plasma free palmitate enrichment was ∼2-fold that of IM palmitoyl-CoA enrichment and palmitoyl-carnitine enrichment (P < 0.001). These findings indicate that plasma free palmitate overestimated IM precursor enrichment owing to in vivo IM lipid breakdown, whereas IM free palmitate enrichment underestimated the precursor enrichment because of lipid breakdown during muscle sampling and processing. IM palmitoyl-carnitine enrichment was an acceptable surrogate of the precursor enrichment because it was less affected by in vitro lipid breakdown after sampling.

We have developed a new method for the simultaneous measurements of stable isotopic tracer enrichments and concentrations of individual long-chain fatty acyl-carnitines in muscle tissue using ion-pairing high-performance liquid chromatography-electrospray ionization quadrupole mass spectrometry in the selected ion monitoring (SIM) mode. Long-chain fatty acyl-carnitines were extracted from frozen muscle tissue samples by acetonitrile/methanol. Baseline separation was achieved by reverse-phase HPLC in the presence of the volatile ion-pairing reagent heptafluorobutyric acid. The SIM capability of a single quadrupole mass analyzer allows further separation of the ions of interest from the sample matrixes, providing very clean total and selected ion chromatograms that can be used to calculate the stable isotopic tracer enrichment and concentration of long-chain fatty acyl-carnitines in a single analysis. The combination of these two separation techniques greatly simplifies the sample preparation procedure and increases the detection sensitivity. Applying this protocol to biological muscle samples proves it to be a very sensitive, accurate, and precise analytical tool.

The supercritical-carbon dioxide fluid extract of Chrysanthemum indicum Linné. (CFE) has been demonstrated to be effective in suppressing inflammation. The aim of this study is to investigate the preventive action and underlying mechanisms of CFE on acute lung injury (ALI) induced by lipopolysaccharide (LPS) in mice. ALI was induced by intratracheal instillation of LPS into lung, and dexamethasone was used as a positive control. Results revealed that pretreatment with CFE abated LPS-induced lung histopathologic changes, reduced the wet/dry ratio and proinflammatory cytokines productions (TNF-α, IL-1β, and IL-6), inhibited inflammatory cells migrations and protein leakages, suppressed the levels of MPO and MDA, and upregulated the abilities of antioxidative enzymes (SOD, CAT, and GPx). Furthermore, the pretreatment with CFE downregulated the activations of NF-κB and the expressions of TLR4/MyD88. These results suggested that CFE exerted potential protective effects against LPS-induced ALI in mice and was a potential therapeutic drug for ALI. Its mechanisms were at least partially associated with the modulations of TLR4 signaling pathways.

The aim of this study was to investigate the anti-inflammatory property of the ethanol extract of the root and rhizome of Pogostemon cablin (ERP). The anti-inflammatory effect was evaluated using four animal models including xylene-induced mouse ear edema, acetic acid-induced mouse vascular permeability, carrageenan-induced mouse pleurisy, and carrageenan-induced mouse hind paw edema. Results indicated that oral administration of ERP (120, 240, and 480 mg/kg) significantly attenuated xylene-induced ear edema, decreased acetic acid-induced capillary permeability, inhibited carrageenan-induced neutrophils recruitment, and reduced carrageenan-induced paw edema, in a dose-dependent manner. Histopathologically, ERP (480 mg/kg) abated inflammatory response of the edema paw. Preliminary mechanism studies demonstrated that ERP decreased the level of MPO and MDA, increased the activities of anti-oxidant enzymes (SOD, GPx, and GRd), attenuated the productions of TNF-α, IL-1β, IL-6, PGE2 and NO, and suppressed the activities of COX-2 and iNOS. This work demonstrates that ERP has considerable anti-inflammatory potential, which provided experimental evidences for the traditional application of the root and rhizome of Pogostemon cablin in inflammatory diseases.
