Abstract The aim of this study was to investigate the vertical transfer of microbiota from dams to the offspring. We studied a pair of 20 dams and its offspring. Maternal sources (colostrum, feces and vaginal secretion) and newborn fecal samples were analyzed using 16S rDNA amplicon sequencing on days 1, 3, 7, 14 and 28. Overall, newborns were maintained healthy and did not receive antimicrobial therapy. The Source Tracker analysis indicated that the newborn fecal microbiota was similar to colostrum and vaginal secretion from day 1 up to 7. However, an unknown source (probably from the environment) showed a gradual increase in its similarity with fecal samples from calves measured from day 3 to 28. The most abundant bacteria groups on meconium (day 1) and calf fecal samples on day 3 were Escherichia-Shigella and Clostridium, respectively. On day 7, the predominant genus were Bifidobacterium and Lactobacillus, while Fusobacterium was the most abundant genus on day 14, coinciding with the diarrhea peak. Faecalibacterium showed a gradual increase throughout the neonatal period. Maternal sources contribute to the neonatal microbiota, however other unknown sources (probably environment) had a strong influence on development of the gut microbiota later in the neonate period.

ABSTRACT: Bovine viral diarrhea virus (BVDV) is a productive and reproductive virus with a high global economic impact on dairy production systems. We investigated the prevalence of BVDV at the herd and individual levels in most dairy-producing regions of Brazil. The frequency of BVDV at the herd level was investigated using quantitative reverse transcription polymerase chain reaction (RT-qPCR) in bulk milk tanks monitored at 289 dairy farms between August 2020 and January 2022. Among these farms, 68 production systems were selected to investigate the prevalence of persistently infected (PI) animals using two antigen-enzyme-linked immunosorbent assays (Ag-ELISAs), at 21-d intervals, from ear-notch samples. In total, 2,902 RT-qPCR and 23,466 Ag-ELISAs were performed. At the herd level, 23.87% (69/289) of dairy farms were considered infected, presenting at least one qPCR test positive. At the individual level, 41.2% (28/68) of the subgroup of selected farms had at least one animal positive in the Ag-ELISA test. Association between tests allowed the classification of farms into the following four categories: level 0, negative for both tests (41.2%, 28/68); level 1, RT-qPCR positive and Ag-ELISA negative (17.6%, 12/68); level 2, RT-qPCR negative and Ag-ELISA positive (13.2%, 9/68); and level 3, positive for both tests (27.9%, 19/68). Multiple correspondence analysis (MCA) suggested a possible association between BVDV positivity and large farms, average daily milk production (herd), breed, and somatic cell counts. The confinement and intensification of animals from different categories, and use of artificial bedding are associated with BVDV infection. Using waste or bulk tank milk to fed calves was also a risk factor for BVDV positivity in RT-qPCR and Ag-ELISA. Despite the use of reproductive vaccines by most producers, their use seems to be associated with BVDV-positive farms. This study presented the epidemiological frequencies of BVDV at the individual and herd levels in the Campos Gerais Paranaense region. The region ranks among the top milk-producing areas in Brazil. Additionally, the association between BVDV tests and farm characteristics indicated the farm risk for BVDV and guides specific control programs.

ABSTRACT: Local adverse reactions following vaccination, often tied to the adjuvant type, can lead to pain, fever, reduced intake, and weight loss. Infrared thermography (IRT), a non-invasive human and veterinary medicine technique, can assess local inflammation. This study aimed to evaluate such reactions induced by reproductive vaccines with different adjuvants, using IRT alongside cardinal signs, rectal temperature, and haptoglobin concentration. Thirty-five Holstein heifers were grouped by vaccine type: Ah (aluminum hydroxide), Ow (oil-in-water), QAD (amphigen and quil A cholesterol and dimethyl-dioctadecyl ammonium bromide adjuvant), and a Control (saline solution). Assessments were made at 0, 6, 24-, 48-, 72-, and 168 hours post-vaccination for both doses, with an interval of 21 days. The local reactions were evaluated using the inflammatory cardinal signs and surface temperature measurement using IRT. The systemic reactions were identified by rectal temperature and the concentration of haptoglobin. A larger proportion of animals exhibiting local reactions based on scores assigned to the cardinal signs was found in the Vaccine QAD group, as well as the rectal temperature and the frequency of heifers with inflammation (Hp ≥2mg/dL). Nevertheless, Vaccine Ow demonstrated higher temperature at the site after the first vaccination dose for the IRT. Therefore, this approach is a valuable tool in classifying responses and local inflammation following vaccination in heifers with reproductive vaccines. Concurrently evaluating systemic manifestations, facilitates the surveillance of adverse reactions, thereby improving the discernment of the extent of systemic and local effects.

ABSTRACT: This study determined the association between biosecurity practices and the status of bovine viral diarrhea virus (BVDV) in dairy production systems. Approximately 280 herds were screened for BVDV virus detection. Following the screening, 68 herds were selected to identify individual BVDV PI animals using an ear notch biopsy and ELISA-antigen. All offspring of the last generation were tested, and the maternal lineage of positive cases was examined. A questionnaire on BVDV biological risk assessment was completed. A multiple correspondence analysis (MCA) was conducted to determine the association between herds with or without BVDV circulation and biosecurity practices. The MCA revealed that farms with virus circulation lacked knowledge about the disease and wrongly perceived their herds as protected, while farms without virus circulation were aware of the disease but considered their herds unprotected. Vaccination practices differed between positive and negative herds, with positive herds using vaccines only for reproductive diseases and negative herds vaccinating for respiratory and reproductive issues. Biosecurity practices such as frequent visitation, contact between animals of different ages, and annual introduction of new animals were linked to viral circulation, while virus-free herds implemented measures like controlled visitation, no contact between different age groups, and quarantine. Lastly, herds with virus circulation acquired pregnant females without prior testing. This study emphasized the crucial role of biosecurity practices in controlling BVDV in dairy herds. It highlighted the sharper risk perception and better application of biosecurity practices in negative herds compared to BVDV-positive herds.

ABSTRACT: This study aimed to evaluate the effect of bovine leukemia virus (BLV) on the systemic profile of naturally infected dairy heifers during the transition period. Pregnant Holstein and Jersey heifers (n=24) were distributed in pairs into two experimental groups: (BLV+) and (BLV-). Animals in the BLV+ group were divided into two subgroups based on the median BLV proviral load (high and low). The animals were then assessed at weeks -3, -2, -1, calving time (0), +1, +2, and +3. Blood samples were obtained for hematological and biochemical analyses, as well as haptoglobin measurements. Farm BLV screening revealed a herd BLV prevalence of 57.25% and heifer BLV prevalence of 38.7%. Mean corpuscular hemoglobin concentration was the only hematological variable for which group interaction was observed, with BLV+ cattle having higher values (33.29 ± 3.39%) than BLV- cattle (31.08 ± 2.31%). Aspartate aminotransferase activity was higher in the BLV+ heifers. The BLV+ group had greater incidence of inflammation (haptoglobin ≥ 2.0 mg/dL). Fibrinogen concentrations were also higher at weeks 0 and +1 in BLV+ heifers than in BLV- heifers. A high proviral load affected total leukocyte and lymphocyte count; however, this profile was not observed in the low proviral load and paired BLV- heifers. To our knowledge, this is the first study to report the impact of BLV infection on the health of dairy heifers during the transition period, demonstrating the effect of proviral load on white blood cell changes and early inflammation in infected animals.

Maternal status during the transition period can significantly impact the health and performance of Holstein dairy calves, with lasting effects on various variables. However, the relationship between maternal late gestation metabolic status, seasonality, and their impact on offspring remains unclear. This study aimed to assess the influence of maternal variables at calving on the performance, metabolism, and immunity of 28 dairy calves during their first month of life. Blood samples were collected from 28 Holstein cows at calving. Median results for maternal variables including non-esterified fatty acids (NEFA), β-hydroxybutyrate (BHB), glucose, total protein (TP), albumin, triglycerides (TG), total cholesterol (TC), haptoglobin (Hp), body weight (BW), and body condition score (BCS) were determined. These median values served as a basis for categorizing the offspring into two groups based on their dams’ high or low degree of each maternal variable. Additionally, calves were categorized by the season of birth (Spring vs. Winter), with 14 in each. Blood samples were collected from the calves at birth and on days 1, 7, 14, and 28 to assess IgG, biochemical parameters, and haptoglobin concentration. Reactive oxygen species (ROS) production by polymorphonuclear cells stimulated by various agents was also evaluated. Clinical assessments were conducted for diarrhea and bovine respiratory disease frequencies. Despite the overall health of the cows, differences were observed in the calves between maternal groups. Heavier cows with high maternal BCS tended to have larger offspring, while high maternal BCS was associated with increased diarrhea prevalence. Low maternal BCS resulted in a stronger innate immune response, indicated by higher ROS production. Calves from cows experiencing metabolic changes during calving displayed elevated Hp concentrations. Spring-born calves were larger but had lower serum IgG concentration and reduced innate immune response compared to winter-born calves. Additionally, spring-born calves exhibited higher Hp and increased diarrhea prevalence on day 28. These findings underscore the importance of the prenatal period in determining neonatal health and suggest further research to elucidate the long-term clinical implications of maternal effects on offspring health and growth. Investigating offspring constituents later in life can provide insight into the persistence of maternal effects over time.

Biosecurity refers to a set of practices that prevents and/or controls the introduction, spread, and elimination of harmful biological agents in a production system. In this study, we aimed to survey the biosecurity practices and determine their correlation with the size of production systems. A biosecurity assessment form was provided to 69 farms in the Campos Gerais region of Paraná, Brazil. The questionnaire was divided into two sections: general and bovine viral diarrhea virus- and bovine herpesvirus type-1-specific sections. The general section covered topics on traffic control, quarantine and animal isolation, hygiene practices, carcass disposal, and disease monitoring/control. The specific section consisted of questions on the reproductive and respiratory factors, use of antimicrobials, and vaccination schedule. The 69 farms were also classified into small (≤ 61), medium (62–201), and large (≥ 202) size farms based on the number of lactating cows. Moreover, multiple correspondence analysis (MCA) was performed between the biosecurity measures and farm size. The main risk factors and variability were related to the traffic control of people, animals, and vehicles/equipment, animal quarantine/isolation, and hygiene practices. MCA revealed that the small farms exhibited a lack of biosecurity measures, including those related to traffic control, animal quarantine, and hygiene. In medium-size farms, contact between bovine animals of different ages and difficulty in animal isolation in the quarantine system were among the main risk factors. In contrast, isolation of sick animals was easy, but the need to frequently purchase cattle was an important risk factor in large farms. These results highlight the relationship between biosecurity measures and farm size, providing valuable insights for the development of better biosecurity plans for production systems.

Bovine alphaherpesvirus 1 (BoAHV-1) infection affects the production and reproductive performance of dairy and beef livestock, resulting in considerable economic losses. In addition to biosecurity measures, vaccination programs are effective strategies for controlling and preventing BoAHV-1 infection and transmission. We evaluated the serological immune response against BoAHV-1 induced by eight different formulations of commercial vaccines: three modified live vaccines and five killed vaccines containing BoAHV type 1 or types 1 and 5. In the first experiment, 50 BoAHV-1-seronegative guinea pigs were assigned to eight groups; each individual in the treatment groups received two doses (one-fifth of the bovine dose). The second experiment was conducted using 29 crossbred Holstein × Gir heifers in four groups of six to nine animals each. The serological immune response against BoAHV-1 was measured using virus neutralization and enzyme-linked immunosorbent assays to measure the total IgG against BoAHV. We evaluated the effects of the vaccine, time, and interaction of the vaccine and time on neutralizing antibodies against BoAHV-1. Killed vaccines produced low levels of antibodies against BoAHV-1, whereas modified live vaccines produced high levels of antibodies capable of providing neutralizing titers in the vaccinated animals, with the thermosensitive modified live vaccine showing the highest levels of antibodies.

ABSTRACT: This study evaluated the vaccine-induced serological response after administering four commercial formulations containing Bovine Viral Diarrhea Virus (BVDV) type-1, BVDV-2, and Bovine Herpesvirus type 1 (BoHV-1) to young heifers with circulating maternal antibody titers. The study also determined the anamnestic response to vaccinations after the complete metabolization of maternal antibodies when the calves reached six months. Eighty-seven Holstein heifers were selected and randomly distributed into four experimental groups based on the vaccine administered. The four vaccine-based experimental groups were as follows: vaccine A (n = 22), vaccine B (n = 22), vaccine C (n = 24), and vaccine D (n = 19), given on the 60th, 90th, and 180th day of life, respectively. Virus-neutralization (VN) tests were performed at baseline (day 60) and 30 days after administering the second (day 120) and third doses (day 210). We analyzed the effect of vaccine group (P ≤ 0.001), day of vaccination (P ≤ 0.001), and group versus vaccine day interaction (P ≤ 0.001) for antibody titers produced against BVDV-1, BVDV-2, and BoHV-1 using the PROC MIXED method (Statistical Analysis System -SAS 9.4). Antibody titers against BVDV-1, BVDV-2, and BoHV-1 were similar at baseline and on day 60 of life. The mean antibody titers were constant and persisted against BVDV-1 in heifers immunized with vaccines A and C. Heifers immunized with vaccine A alone had a similar effect against BVDV-2. Regarding BoHV-1, the antibody titers decreased between days 60 and 210 in groups B, C, and D. The antibody titer for heifers in group A also decreased between days 60 and day 120, and an intense increase in titers was observed on day 210. After being immunized with formulations B, C, and D, the frequency of animals with titers above protective levels for BVDV-1, BVDV-2, and BoHV-1 was very low or null. Indicators of anamnestic response were observed in heifers vaccinated with formulation A only. Therefore, it can be concluded that passive immunity negatively interferes with antibody production after vaccination.

ABSTRACT: Bovine respiratory disease (BRD) is a multifactorial disease which causes short- and long-term negative effects. Early detection is crucial for a prompt response to therapy, as well as to decrease mortality risk. Clinical scoring systems have been developed mostly in North America for screening calves at risk or suspected of having BRD, and these tools have also been applied in subtropical and tropical countries. However, it has been unknown whether these scoring systems had the same accuracy in tropical environmental conditions. Therefore, this study evaluated the accuracy of 4 different field techniques, as well as serum haptoglobin (HAP), to diagnose BRD in Holstein dairy calves in subtropical conditions. The tests used to diagnose BRD were thoracic ultrasound (TUS; positive if consolidation depth ≥1 cm), thoracic auscultation (AUSC; positive if crackles, wheezes, or silent areas were present), Wisconsin score (WISC; ≥2 categories with scores of ≥2), and California score (CALIF; positive if total score ≥5). Also, HAP was measured and classified as positive if ≥15 mg/dL. Heifers between 30 d of age and weaning (n = 482), residing on 17 commercial dairies in São Paulo state, were enrolled in this study. Bayesian latent class models were used with informative priors to evaluate the accuracy of TUS, AUSC, and HAP, and noninformative priors for the accuracy of WISC and CALIF. The percentage of calves positive for each test on each farm ranged from 0 to 56% for WISC, 11–51% for CALIF, 0–72% for TUS, 0–32% for AUSC, and 0–100% for HAP. The sensitivity (Se; 95% credible interval) and specificity (Sp) for WISC were 77.9% (64.8–90.2) and 81.9% (76.3–88.2). For CALIF, the Se was 67.1% (53.6–80.1) and Sp 79.1% (73.9–84.6). For TUS Se was 59.8% (46.5–73.1) and Sp was 84.8% (80.0–89.5), and for AUSC, Se was 58.8% (41.3–79.8) and Sp was 98.6% (95.7–99.9). The Se and Sp of HAP was 67.6% (55.3–78.8) and 46.7% (41.4–52.2), respectively. The performance of the scoring systems was similar to, or better than, the performance found in North American studies, despite the fact that calves were in a tropical environment.

Abstract Background Neonatal calf diarrhea (NCD) is the leading cause of calf morbidity and mortality in beef cattle. Cow’s vaccination in last stage of pregnancy is one of the most important measures to mitigate the risk of NCD outbreaks. The aim of this study was to evaluate the efficacy of prepartum single dose vaccination against NCD, especially Bovine Rotavirus type A (BoRVA) and Bovine Coronavirus (BCoV), in Nelore dams and offspring. A total of 117 pregnant cows (n = 81) and heifers (n = 36) were distributed in two groups, vaccinated (VAC: cows = 40; heifers = 19) and non-vaccinated (NVAC: cows = 41; heifers = 17). Vaccination occurred between 60 to 50 days before the expected calving date with a single dose of a water-in-oil (W/O) vaccine, and NVAC group received a dose of saline solution 0.9%. Blood samples were collected before vaccination and 30 days after to evaluate the antibody (Ab) response. Specific IgG1 Abs against BoRVA and BCoV were measured by using an Enzyme Linked Immuno Sorbent Assay (ELISA). Calves’ births were monitored, and the transference of passive immunity was evaluated. Diarrhea was monitored in the first 30 days of age, and fecal samples were collected for identification of the etiological agent. This study aimed to assess the impact of prepartum maternal diphenyl diselenide (PhSe)2 supplementation on the development, biochemical, immune, and antioxidant parameters of calves. Eighteen Holstein breed calves were used, born to females who were or were not subjected to supplementation, at 42, 28, and 14 days prior to calving. The (PhSe)2 group (DDG) was administered 3 μmol/kg of (PhSe)2 in 4 mL of dimethyl sulfoxide (DMSO), while the DMSO and NaCl groups were administered 4 mL of DMSO and 0.9% NaCl, subcutaneously. The calves were evaluated based on their weight, withers height, body condition score 24 h post-birth (0), as well on days 14, 28, 42, 56, 70. Blood samples were also taken to determine serum variables. Calves on the DDG showed higher average levels of total protein, albumin, and globulins on day 0, and the immunoglobulin G level was significantly higher than the other groups on days 0, 14, 56, 70. Maternal supplementation showed immunomodulatory effect on calves, evidenced by the exceptional rates of passive immunity transfer, as well as the enhancement of humoral immunity. Our research offers fresh insights into the immunomodulatory potential of (PhSe)2, making it a viable alternative in facing this challenging phase, rearing dairy calves.

ABSTRACT: The early use of antimicrobial therapy has been introduced in many farms to prevent diarrhea and respiratory disease in young calves; however, there is controversy about whether this practice has a beneficial effect on the health of these animals. This study evaluated the influence of the early use of antimicrobials on the health and performance of neonatal Holstein calves. Twenty-six Holstein calves were screened and divided into two groups, according to the administration (ATB+), or not (ATB-) of tulathromycin (2.5mg/kg, subcutaneously) within the first 12 hours of life. Calves were evaluated by general clinical examination, fecal score, respiratory score, and external palpation of the umbilical region, besides fecal output of dry matter. Anemia was determined by using an automatic system and, also, using a commercial kit for iron dosage. Diarrhea was diagnosed by a centrifuge-flotation technique using a sugar solution (Cryptosporidium) and multiplex semi-nested RT-PCR (rotavirus/coronavirus). The performance of the calves was estimated by Daily Weight Gain (DWG). The young dairy calves were evaluated within 12 hours of birth (≤12h) and at 3-5th (D3-5), 7-9th (D7-9), 13-15th (D13-15), 20-23rd (D20-23), and 27-30th (D27-30) days of life. No difference was noted between the ATB+ and ATB- groups concerning heart rate, respiratory frequency, and rectal temperature. Erythrogram showed a higher frequency of anemia in ATB- group (P=0.016) at the D3-5 check-up; lower values of serum iron were also observed simultaneously (P=0.051). Thirteen cases of respiratory disease were detected during this study; however, no significant difference was observed between the groups in this regard. The frequency of diarrhea (fecal score 2-3) was high in both groups, peaking at D13-D15. No differences were noted between the groups regarding the frequency of diarrhea when considering the dry fecal matter. The predominant etiological agent for diarrhea was Cryptosporidium spp.. The DWG was similar between groups, with maximum weight reduction on D13-15. The administration of tulathromycin in prophylactic dose (2.5mg/kg) at birth decreased the frequency of anemia but did not influence weight gain or the prevalence of diarrhea.

ABSTRACT: Vaccination is a strategy to the prevention and control of reproductive diseases caused by bovine viral diarrhea virus (BVDV) and bovine herpesvirus type 1 (BoHV-1), however the various compositions of commercial vaccines should be evaluated for their ability to induce protection mediated by antibodies. The objective of this research was to evaluate the production of specific neutralizing Abs against BVDV-1 and 2, and BoHV-1 induced by commercial vaccines composed by different adjuvants. Holstein heifers were vaccinated and distributed in three experimental groups: Group I (G1) was vaccinated with a commercial vaccine containing inactivated BVDV-1, BVDV-2 and BoHV-1 diluted in alum hydroxide as adjuvant (n=9); Group II (G2) was vaccinated with an product containing inactivated strains of BVDV-1, BVDV-2, BoHV-1 and BoHV-5 diluted in oil emulsion as adjuvant (n=10); Group III (G3) was vaccinated with a commercial vaccine containing inactivated BVDV-1 and BVDV-2, besides live modified thermosensitive BoHV-1, diluted in Quil A, amphigen and cholesterol (n=10); A control, non-vaccinated group (n=6) was mock vaccinated with saline. Heifers received two subcutaneous doses of 5mL of each commercial vaccine on the right side of the neck, with 21 days interval. Humoral immune response was assessed by the virus neutralization test (VN) against BVDV-1 (NADL and Singer strains), BVDV-2 (SV253 strain) and BoHV-1 (Los Angeles strain) in serum samples collected on vaccination days zero (D0), 21 (D21) and 42 (D42; 21 days after boosting). Neutralizing Abs against BVDV-1 NADL was detected only in D42, regardless of the vaccine used. Similar geometric mean titers (GMT) for BVDV-1 NADL were observed between G1 (log2=5.1) and G3 (log2=5.1). The seroconversion rate (%) was higher in G1 (78%) when compared to G2 (10%) and G3 (40%). For BVDV-1 Singer, it was also possible to detect Abs production in G1 (log2=5.8, 100% seroconversion rate) and G3 (log2=3.5, seroconversion rate = 60%), only after the booster dose (D42). Neutralizing Abs to BVDV-2 (SV253) were detected only in G3, observing 90% seroconversion associated with high titers of Abs (log2=6.7) after the 2nd dose of vaccine (D42). Heifers from G1 and G3 responded to BoHV-1 after the first dose (D21): G1 (log2=2.5, seroconversion rate = 67%) and G3 (log2=0.7, seroconversion rate = 80%). In D42, a higher magnitude response was observed in the heifers from G3 (log2=6.1, 100%) compared with G1 (log2=4.3, 100%) and G2 (log2=2.7, 60%). Based on the data obtained, it can be concluded that the commercial vaccine contained aluminum hydroxide (G1) was most effective in the induction of antibodies against BVDV-1. On the other hand, this vaccine did not induce the production of neutralizing Abs against BVDV-2. Only the heifers from G3 (Quil A, amphigen and cholesterol) generated neutralizing Abs against BVDV-2. The animals that received commercial vaccine containing oil emulsion as adjuvant (G2) had a weak/undetectable response against BVDV-1 and BVDV-2. The best protective response against BoHV-1 was observed in heifers vaccinated with the live modified thermosensitive virus.

During the period called “transition”, from the ceasing of milk production to the reestablishment of full milk production, it is postulated that the microbiota of cows undergo changes in composition driven by the fluxes in systemic energetics and that these changes appear to impact the health of cows. The primary objective of this study was to document the make-up of the microbiota in the mammary gland compared with those in the vagina and in feces in an attempt to determine any correlations between the composition of the microbiota, the impact of blood indicators of energetic metabolites and the health of the mammary gland at the time of calving. Samples were collected from 20 Holstein dairy cows immediately following calving to assess their general health and measure the microbiomes associated with each cow using 16S rRNA sequencing. The results indicated that the microbiomes found within each maternal niche were different. A set of significant negative associations between the blood energetic biomarkers (NEFAs, BHB, triglycerides and cholesterol) and the taxa Pseudomonas, Christensenellaceae and Methanobrevibacter were observed in this study. In contrast, Escherichia and Romboutsia were positively correlated with the same energetic metabolites. Therefore, it was concluded that there appears to be a set of relationships between the microorganisms that colonize several niches of cows and the sufficiency of systemic energy metabolism. Furthermore, both the microbiome and energy dynamics impact the health of the mammary gland of the host.

A fase neonatal é representada pela incapacidade adaptativa dos recém-nascidos ao ambiente extrauterino e execução de funções vitais até então exercidas pela mãe, como a termorregulação, equilíbrio acidobásico, funções cardiorrespirató- rias, nutrição e desenvolvimento do sistema imunológico. Objetivou-se com este estudo, avaliar as adaptações fisiológicas de bezerros Holandeses durante a fase de aleitamento (60 dias). Foram selecionados 22 bezerros recém-nascidos machos da raça Holandesa, nascidos entre os meses de setembro à março (n=18) e Abril à Agosto (n=4). Valores de frequência cardíaca (FC), frequência respiratória (FR) foram registrados diariamente durante 60 dias pela técnica semiológica da auscultação, bem como a temperatura retal (TR) mensurada com a utilização de termômetro clínico digital. Os resultados obtidos diariamente durante os 60 dias do experimento foram agrupados em períodos compostos por seis dias cada (P1 à P10). A primeira semana de vida pós-natal foi marcada por modificações fisiológicas inerentes a adaptação das funções cardiorrespiratórias dos recém-nascidos, observando-se estabilização da FC e FR nos períodos subsequentes, com algumas variações provavelmente atribuídas à oscilação climática (10 a 37ºC) do ambiente de criação dos bezerros. Estes dados demonstraram impacto direto do período de adaptação extrauterina nos parâmetros fisiológicos dos neonatos bovinos.

The detection of Bovine Viral Diarrhea virus (BVDV) infection, especially among persistently infected calves (PI), should be performed earlier in order to eliminate the source of the infection and to prevent the spread of the disease in the herd. However, colostrum intake can influence the results of some of the tests used to diagnose the BVDV infection. Therefore, this study evaluated the efficacy of serum neutralization (SN) test in conjunction with reverse transcription-polymerase chain reaction (RT-PCR) for the diagnosis of BVDV infection before colostrum intake. The deliveries of the animals were assisted to select 52 newborn Holstein calves for inclusion in the study. Initially, the whole blood and serum samples were collected from the calves before (T0) and after (T1) the colostrum intake. The calves that were RT-PCR positive at any of the time-points were retested on the 30th day post birth (T2). The presence of specific antibodies for BVDV was evaluated by SN, and that of viral RNA by the RT-PCR. The BVDV-specific antibodies were observed in the serum of 13.46% (7/52) of the calves at T0 because of fetal infection. At T1, seroconversion was observed in 100% (52/52) of the calves. The geometric mean titers (GMT) of the antibodies for BVDV increased significantly from T0 (14.52) to T1 (2490) (P = 0.0001). Of the four calves that were RT-PCR positive before colostrum intake (T0), two were seronegative and two, seropositive. Of the forty-eight RT-PCR negative calves, five were seropositive. After 30 days post birth, all of the animals tested negative by RT-PCR, thus excluding the possibility of persistent infection. The association observed between the results of the SN and RT-PCR assays at T0 (P = 0.025) could not be observed at T1 (P > 0.05). The SN test before the colostrum intake allowed the detection of fetal infection in the herd; however, this test was ineffective as a diagnostic method after the transfer of passive immunity. The confirmation of the results of the SN assay by those of the RT-PCR was essential for the identification of the infected calves before colostrum intake.

The aim of this study was to evaluate secondary clinical disease, milk production efficiency and reproductive performance of heifers and cows persistently infected (PI) with bovine viral diarrhea virus type 2 (BVDV type 2). PI animals (n = 25) were identified using an antigen capture ELISA of ear notch samples. They were distributed into three age groups: ≤ 12 (n = 8), 13 to 24 (n = 6) and 25 to 34 (n = 11) months old. A control group of BVDV antigen ELISA negative female cattle that were age matched to the PI animals was utilized from the same herd. The PI group had a 1.29 higher odds ratio for diarrhea than controls (p = 0.001, IC95% = 1.032–1.623) and 1.615 greater chance of developing bovine respiratory disease (BRD) (p = 0.012, IC95% = 1.155–2.259). The age at first insemination (p = 0.012) and number of insemination attempts required to establish the first pregnancy (p = 0.016) were both higher for PI than controls. Milk production was higher for control cows than PI cows during most of the sampling periods. Somatic cell counts (SCC) were higher in PI cows than the controls at all sampling points across lactation (p ≤ 0.042). PI cattle had a higher incidence of disease, produced less milk, a higher SCC, and poorer reproductive performance than control cattle in this study.

RESUMO: Este trabalho avaliou a influência da infecção bacteriana da glândula mamária (GM) sobre a transferência de imunidade passiva (TIP) em bezerros recém-nascidos. Vacas holandesas (n=13) foram observadas no momento da parição e ordenhadas para a obtenção de forma asséptica das amostras de colostro (n=52) para os testes microbiológicos. Os recém-nascidos receberam 6 litros de colostro de uma ordenha nas primeiras 12 horas de vida, proveniente de suas respectivas mães. Amostras de sangue foram colhidas antes (D0) e após (D2) o manejo do colostro. A TIP foi avaliada por meio de testes bioquímicos, eletroforese e leucograma. Os bezerros foram distribuídos conforme a ausência (IB-) ou presença (IB+) de infecção mamária em pelo menos uma GM de suas respectivas mães. Todas as amostras de colostro (n=52) foram negativas ao cultivo fúngico. Das 13 fêmeas, 8 (61%) apresentaram crescimento bacteriano em ≥1 quartos mamários. Considerando-se os quartos mamários, foi obtido isolamento bacteriano em 21,15% (11/52), observando-se predomínio de espécies bacterianas do grupo Staphylococcus coagulase negativa. Não foram encontradas diferenças entre os parâmetros de acordo com os grupos experimentais. Em relação aos momentos, foi possível verificar aumento nos valores de proteína total, globulinas, atividade sérica da gama glutamiltransferase e frações eletroforéticas beta e gamaglobulina após a ingestão do colostro materno. A mastite subclínica não influencia a transferência de imunidade passiva em bezerros recém-nascidos da raça holandesa, avaliados por teste bioquímicos, eletroforese e leucograma.

RESUMO: O periparto caracteriza-se por mudanças imunes e fisiológicas na glândula mamária das fêmeas bovinas, tornando-as mais susceptiveis às mastites. Este trabalho teve o objetivo de avaliar a fagocitose da bactéria Escherichia coli pelos fagócitos do sangue e do colostro de vacas hígidas da raça Holandesa. Amostras provenientes de 22 fêmeas, entre novilhas e vacas, foram obtidas durante a primeira ordenha pós-parto, para avaliação da fagocitose, em lamínula de vidro, após estímulo com E. coli. A proporção de fagócitos espraiados do sangue foi maior quando as células foram estimuladas com bactérias (p=0,003), no entanto a taxa de fagocitose foi baixa e semelhante à observada quando as células não foram estimuladas (p=0,299). No colostro, foram constatados maiores índices de fagocitose (p=0,001) e espraiamento (p=0,000) quando os fagócitos foram estimulados com E. coli. Comparando-se os resultados obtidos para o sangue e o colostro, verificou-se, no sangue, maior proporção de fagócitos não espraiados e que não apresentavam bactérias no seu interior, estimulados (p=0,000) ou não (p=0,002) com bactéria. Houve maior taxa de espraiamento nos fagócitos do colostro quando estimulados com E. coli (p=0,002). Não se viu diferença nos percentuais de fagocitose entre as amostras de sangue e colostro incubadas (p=0,478) ou não (p=0,071) com bactérias. Os resultados permitiram concluir que E. coli influenciou no espraiamento dos fagócitos do sangue e do colostro de fêmeas bovinas da raça Holandesa no período do pós-parto imediato e auxiliarão na compreensão dos mecanismos imunológicos ocorridos nesse período, considerado o de maior susceptibilidade das fêmeas bovinas às infecções.

Para a avaliação da transferência de citocinas para o sangue de bezerros neonatos via ingestão de colostro de fêmeas bovinas holandesas, foram utilizados 15 bezerros nascidos de parto eutócico, distribuídos igualmente por três grupos experimentais (n=5): G1- receberam dois litros de colostro fresco provenientes de suas próprias mães; G2- receberam dois litros de colostro provenientes de "pool" de colostro congelado e o G3- foram alimentados apenas com leite. Nestes grupos foram coletadas amostras de sangue em cinco tempos durante os primeiros quinze dias de vida e mensuradas as concentrações das citocinas Interleucina-1 β (IL-1b), Interleucina-6 (IL-6), Fator de necrose tumoral- α (TNF-a) e Interferon-γ (IFN-γ). Também se mensurou tais citocinas (IL-1 β, IL-6 e TNF-α) nos sobrenadantes do colostro de do "pool" de colostro fornecidos aos bezerros dos grupos G1 e G2 respectivamente. Verificou-se a transferência das citocinas IL-1b, IL-6, TNF-a e IFN-γ pela presença no soro dos bezerros do grupo G1, enquanto que nos demais grupos (G2 e G3) não foram detectadas.

The aim of this study was to identify etiologic agents and risk factors for mastitis in dairy goats. For this, two aliquots were collected from 257 milk samples, between April and September, 2012, from 136 Saanen goats from six intensive management farms, with manual and mechanical milking system located in the state of São Paulo. The first aliquot of milk samples was obtained in vials containing preservative bronopol for electronic somatic cell count (SCC), and the second in sterile bottles for bacteriological examination. For risk factors evaluation, epidemiologic questionnaires were performed by the research team by visual observation and survey with handler and owners. Analyzes univariate and multivariate were performed using multiple logistic regression. Was observed prevalence of 28.79% of mammary infection, and predominance of coagulase-negative Staphylococcus (CNS). The SCC median was 4,7x105 cells per mL (cells mL-1) of milk. In univariate analysis, was observed risk for mammary infection in places where, the goat was not the main activity of the property (p=0.03); when the diagnosis of clinical mastitis by proof of dark background was not performed (p=0.07); when chlorine was not used in the cleaning of the teats (p=0.29); improper cleaning of the milking parlor (p= 0.06); inadequate hygiene in the milking process (p=0.03) and inadequate hygiene of the milkers (p=03). The main group of pathogen isolated in this study was CNS. The biggest risk associated with mammary infection was the hygiene of ceilings, room, milker and also the process of milking resulting from these microorganisms. Thus, despite indications of hygienic measures in the programs of milk quality and mastitis control, the applicability and awareness of employees involved in the production chain of milk goats are not sensitized. Therefore, the personnel training is the principal measure recommended for the control of mastitis in dairy goats.

After vaccination, vaccine components must activate the immune response, but the ideal vaccine should not result in undesirable effects in cattle. The aim of this study was to evaluate the inflammatory and humoral responses and adverse reactions induced by three adjuvanted commercial vaccines against bovine viral diarrhea virus (BVDV) and bovine herpesvirus 1 (BHV-1). Holstein heifers (n = 35) were divided into four groups by adjuvant compounds: Vaccine A (Alum; n = 9), Vaccine B (Oil-in-water; n = 10), Vaccine C (Amphigen/Quil A cholesterol and dimethyl-dioctadecyl ammonium (DDA) bromide (QAD; n = 10), and Control (n = 6). Heifers were assessed at 0 h, 6, 24, 48, 72 and 168 h post-vaccination; serology was evaluated at first dose (D0), booster (D21) and D42. Heifers vaccinated with Vaccine B (p = 0.0001) and C (p = 0.0001) had a more intense local reaction, while there was a higher rectal temperature detected in heifers vaccinated with Vaccine C (p = 0.020). There was greater systemic reaction observed for heifers vaccinated with Vaccines B and C at 48 h (p = 0.002) after a second dose. Clinical pathology parameters [white blood count (WBC) (p = 0.001), neutrophils (p = 0.0001) and haptoglobin concentrations (p = 0.0001)] were higher in animals vaccinated with Vaccine C. Neutralizing Abs against BVDV type 1 strains, NADL and Singer, were detected in animals vaccinated with Vaccines A or C at D42, while BVDV-2 antibodies were detected only in animals vaccinated with Vaccine C. A BHV-1 antibody was detected in all three vaccine groups (Vaccines A, B or C) at day 42 (21 days post booster vaccination). The findings of this research were based on three different commercial laboratory formulations and also according to the conditions which the study was conducted. In this context, vaccine containing mineral oil or Amphigen/QAD presented greater local reactivity and induced a significant systemic inflammatory response. Vaccinated heifers with Alum and Amphigen/QAD commercial vaccines enhanced humoral immune response against BVDV and BHV-1.

The purpose of the present trial was to investigate the influence of lactation stage on milk cellularity and physicochemical characteristics. Thus, 143 milk samples from 33 Santa Ines ewes negative in the bacteriological examination and without any alteration in the strip cup test were used to determine the automatic and microscopic somatic cell count (SCC), and the fat, lactose, protein and total solids content. The results of the present study showed an augment in the SCC and in the fat and protein content, besides a reduction in lactose content at the last lactation stage. Otherwise, no alteration was observed in the total solids content during the lactation period. This study pointed out to a significant correlation between the automatic SCC and the microscopic SCC (r = 0.80), percentage of polymorphonuclear leukocytes (r = 0.45), percentage of mononuclear leukocytes (r = -0.45), lactose (r = - 0.44), protein (r = 0.37) and fat (r = 0.30) content. Nevertheless, significant correlation between the automatic SCC and the total solids content was not observed.

In order to establish hematological and biochemical Dorper sheep values and evaluate the influence of sex and age in these determinations, 72 animals were assigned to three experimental groups according to age: Group 1 (G1) consisting of animals up to 12 months of age (n=28), Group 2 (G2) composed of animals from 13 to 36 months (n=37) and Group 3 (G3) for animals from 37 to 60 months (n=7). The animals were still divided in another two groups according to gender: Group 4 (G4) – females (n=57) and Group 5 (G5) – males (n=15). Evaluating the results obtained for hematological and biochemical examinations it was found that the mean values of RBC (13.4 ± 2.5x106/mL), hemoglobin (12.6 ± 1.3 g/dL), packed cell volume (37.5 ± 3.7%), total leukocytes (7116.0 ± 1967.0/mL) and segmented neutrophils (4168.0 ± 1501.0/mL) were higher in the animals of G1, as well as urea, GGT and ALP (52.0 ± 17.0 mg/dL, 93.0 ± 19.0 IU/L, 257.0 ± 125.0 IU/L, respectively). Comparing the gender, it was found that the values of total leukocytes (7576.0 ± 2082.0/mL), segmented neutrophils (4296.0 ± 1510.0/mL) and lymphocytes (2428.0 ± 1084.0/mL) were greater in males (G5). The values of creatinine (1.3 ± 0.4 mg/dL), serum total protein (6.3 ± 0.9 g/dL) and globulins (3.7 ± 1.0 g/dL) were higher in females (G4). Statistical analysis of the results allowed to determine reference values for hematological and biochemical constituents of Dorper sheep breed, and concluded that there were variations in these parameters influenced by gender and different age groups. A fim de estabelecer os valores hematológicos e bioquímicos de ovinos da raça Dorper e avaliar a influência do sexo e idade nestas determinações, 72 animais foram distribuídos em três grupos experimentais de acordo com a faixa etária, sendo o Grupo 1 (G1) composto por animais com até 12 meses de idade (n=28), o Grupo 2 (G2) composto por animais de 13 a 36 meses de idade (n=37) e o Grupo 3 (G3) por animais de 37 a 60 meses de idade (n=7). Os animais foram distribuídos ainda em outros dois grupos exprimentais, de acordo com o sexo, sendo o Grupo 4 (G4) composto por fêmeas (n=57) e o grupo 5 (G5) por machos (n=15). Avaliando-se os resultados obtidos para os exames hematológicos e bioquímicos verificou-se que os valores médios de hemácias (13,4 ± 2,5x106/mL), hemoglobina (12,6 ± 1,3 g/dL), volume globular (37,5 ± 3,7%), leucócitos totais (7.116,0 ± 1.967,0/mL) e neutrófilos segmentados (4.168,0 ± 1.501,0/mL) foram maiores nos animais do G1, assim como a ureia, GGT e FA (52,0 ± 17,0 mg/dL, 93,0 ± 19,0 UI / L e 257,0 ± 125,0 UI / L, respectivamente). Comparando-se os sexos, verificou-se que os valores de leucócitos totais (7.576,0 ± 2.082,0/mL), neutrófilos segmentados (4.296,0 ± 1.510,0/mL) e linfócitos (2.428,0 ± 1.084,0/mL) foram maiores nos machos (G5). Os valores de creatinina (1,3 ± 0,4 mg/dL), proteínas totais séricas (6,3 ± 0.9 g/dL) e globulinas (3,7 ± 1.0 g/dL) foram maiores nas fêmeas (G4). A análise estatística dos resultados encontrados permitiu determinar os valores de referência para os constituintes hematológicos e bioquímicos de ovinos da raça Dorper, além de concluir que ocorreram variações nos referidos parâmetros, influenciados pelo sexo e por diferentes faixas etárias.

O presente estudo objetivou avaliar a correlação entre a contagem de células somáticas (CCS) automática e a CCS microscópica direta por diferentes colorações no leite de ovelhas da raça Santa Inês. Avaliou-se a CCS automática por citometria de fluxo, e a CCS microscópica foi realizada por esfregaços lácteos corados com as colorações de Broadhurst-Paley (BP), Hematoxilina-eosina (HE) e Rosenfeld. O presente trabalho objetivou realizar um estudo retrospectivo sobre os prolapsos vaginal e uterino em ovelhas atendidas no Serviço de Clínica de Bovinos e Pequenos Ruminantes (CBPR) da FMVZ/USP no período compreendido entre 2000 a 2010, no qual, foram atendidas 56 ovinos com problemas inerentes ao sistema reprodutivo, dessas, 25 apresentaram prolapso vaginal ou uterino (44,6%). O prolapso vaginal total foi o de maior frequência (72%). As ovelhas acometidas, em sua maioria, possuíam idade superior a quatro anos (64%), eram sem raça definida (44%) ou da raça Ile de France (40%). As manifestações clínicas observadas durante a maioria dos atendimentos foram: taquipnéia, taquicardia, mucosas oculares avermelhadas indicando estado de toxemia, decúbito esternal ou lateral, apatia e anorexia. O tratamento instituído para todos os casos foi a limpeza, desinfecção e reintrodução do órgão prolapsado. A sutura de Bühner foi feita em 84% dos casos e a histeropexia em um caso (4%). A evolução foi satisfatória em 80% dos casos atendidos, nos demais casos (20%) observou-se óbito da fêmea acometida. Do total de óbitos, os prolapsos vaginais foram responsáveis por 60% (3/5) e os prolapsos uterinos por 40% (2/5). A etiologia dos prolapsos não foi definida nos casos atendidos, sendo esses associados com o período pós-parto em sua maioria (56%), provavelmente associados com quadros de hipocalcemia, altas concentrações séricas de estrógeno e hipertonia uterina. Além disso, a predisposição genética não pode ser descartada.

The somatic cell count (SCC) is a screening test for the evaluation of intramammary infection; however, changes in mammary secretion during colostrogenesis can promote a physiological increase in the SCC, potentially reducing its reliability in the diagnosis of mastitis. Therefore, the aim of this study was to evaluate bovine colostrum SCC as an indicative parameter of breast infection in Holstein periparturient cows. A total of 80 samples were harvested from the first milking colostrum of 20 cows and were subjected to manual SCC and bacteriological examination. Bacterial growth was present in 36.62% of the crops; coagulase-negative Staphylococcus (CNS) was the predominant microorganism (76.92%). The median SCC in infected cows (1.8 × 106 cells/mL) was significantly higher than in uninfected cows (0.9 × 106 cells/mL) (p = 0.0451). The sensitivity (100–15%), specificity (100–2.2%), and false positive (100–2.2%) of the SCC decreased gradually when thresholds of 0.2–10.0 × 106 cells/mL were adopted. In contrast, the proportion of false negatives (0–84.6%) revealed an opposite trend. The threshold of greatest concordance between SCC and bacteriological examination was 10.0 × 106 cells/mL; however, the sensitivity rates (15.4%), specificity (2.2%), and false positive (2.2%) were very low. Based on these results, we conclude that SCCs increased prior to the infectious processes of the mammary gland, particularly in the CNS group. However, physiological changes caused by colostrogenesis resulted in poor concordance between the SCC and bacteriological examination of the colostrum.

To establish reference values and to assess the influence of age on the leukograms of healthy Holstein calves, blood samples were obtained from 300 animals. These samples were distributed equally (n=20) among 15 experimental groups according to age: birth to 8 hours, 9 to 16 hours, 17 to 24 hours, 2 days, 3 days, 4 days, 5 days, 6 to 7 days, 8 to 9 days, 10 to 11 days, 12 to 13 days, 14 to 15 days, 16 to 20 days, 21 to 25 days and 26 to 30 days of age. The maximum numbers of leukocytes (9,305.0/mL), segmented neutrophils (6,551.2/mL) and total neutrophils (6,678.3/mL) were noted within the first 8 hours of life, while band neutrophils peaked in number (133.3/mL) between 9 and 16 hours after birth. Meanwhile, the maximum total lymphocyte (4,992.1/µL) and typical lymphocyte (4,686.1/µL) counts occurred between 21 and 25 days, whereas atypical lymphocytes (388.5/µL) reached their maximum number between 26 and 30 days, demonstrating an inversion of the neutrophil:lymphocyte ratio over time. Thus, the influence of age on the leukocyte count of the evaluated calves was verified. The release of endogenous corticosteroids during labor or at birth may contribute to this variation in leukograms with age.

This study analyzed the influence of the number of milkings, number of births, and udder quarter in immunoglobulin (Ig) concentration in the colostrum of healthy Holstein cows. It was collected two samples of colostrum by manual milking, getting the first jets to completion of bacteriological examination and immunoglobulin levels by radial immunodiffusion test in agar gel. Positive samples for bacteriological examination were excluded from this investigation. Medians of immunoglobulin's G, A and M in the colostrum collected before the first and second milking were respectively 9,200 and 6,400mg/dL (p=0.0029); 400 and 200mg/dL (p=0.0018); 800 and 400mg/dL (p=0.0001). Median immunoglobulin concentration in animals that calved once, twice or three times or in cows that calved 4 to 6 times were 6,400; 6,400; 3,200 and 11,200mg/dL IgG; 100, 200, 100 and 800mg/dL IgA ; and 400, 400, 100 and 800mg/dL IgM, respectively. The measurement of pH in milk is one of the most important and frequent procedures performed in biochemistry practices. This variable is applied to the diagnosis of mammary gland disease, assessment of industrial yields, and identification of metabolic diseases. The objective of this study was to evaluate de effect of freezing the milk from healthy and infected mammary glands of Santa Ines ewes on the pH. The bacteriological culture of 183 milk samples was analyzed resulting in 146 negative and 37 positive samples. From these, the hydrogen ion concentration was determined on sampling day and after freezing at -20°C for 30 months. A reduction on pH was observed after freezing, and the bacteriological positive samples showed the highest pH values. A higher decrease of the pH after freezing was seen in positive samples. Thus, the kind of conservation of milk samples should be considered, as well as the results of parameters that could be influenced by such variation.

A particularidade da secreção láctea caprina, do tipo apócrina, diferente da secreção merócrina da vaca, leva a erros de interpretação durante a realização de técnicas de avaliação da celularidade do leite de fêmeas desta espécie. Portanto, o presente trabalho teve o objetivo de determinar a contagem de células somáticas pelo método indireto California Mastitis Test (CMT), e por métodos diretos, incluindo a contagem por citometria de fluxo e a contagem microscópica direta, através da coloração de verde de metil e pironina-Y, além de comparar os métodos de contagem celular. Foram analisadas 102 amostras de 51 fêmeas caprinas, das raças Saanen, Parda Alpina e Toggenburg, criadas no Estado de São Paulo. Os animais foram categorizados segundo a fase da lactação, exame físico da glândula mamária e exame do leite. As amostras foram colhidas, após a realização do exame Califórnia Mastitis Test, em duas alíquotas, uma destinada à contagem celular automática e a outra, a contagem microscópica direta, utilizando-se o corante verde de metil e pironina- Y. De acordo com os diferentes escores do CMT, observou- se 74,5% de amostras negativas, 8,8% de amostras com escore traços, 8,8% de amostras ligeiramente positivas (+), 6,8% de amostras fracamente positivas (++) e 0,9% de amostras fortemente positivas (+++). Os valores medianos das contagens de células somáticas presentes no leite de cabras, avaliadas através de contador automático e microscopia direta, e analisadas de acordo com os diferentes escores do CMT, foram, respectivamente, 181.000, 578.000, 628.000, 1.421.500 e 5.542.000 células/mL de leite e 74.991, 271.396, 71.420, 640.995 e 5.049.394 células/ mL de leite, nos escores negativo, traços, +, ++ e +++. Os valores medianos obtidos através da contagem de células somáticas pelo método automático e microscópico direto, de acordo com as fases de lactação foram de 159.500, 508.000 e 277.500 células/mL de leite, e 62.493, 89.275 e 146.411. A correlação obtida entre a contagem celular automática e microscópica direta foi de 88%. A partir dos resultados observados pode-se concluir que existe diferença na contagem celular determinada através do método automático e microscópico sendo este último o mais adequado para a determinação da celularidade no leite de cabras.The particular apocrine secretion of goat milk different from the merocrine one observed in cows, may lead to errors in interpreting cellularity evaluations in the milk of this species. Thus, the objective of the present trial was to determine Somatic Cell Counts by means of one indirect methods, the California Mastitis Test (CMT), and direct methods, flow cytometry and direct microscopic count using methyl green-pyronine-Y stain, beyond comparing the methods of cellular counting. A total of 102 samples from 51 Saanen, Brown Alpine and Toggenburg female goats, bred in the state of São Paulo, were analyzed. Goats were separated in groups according to the phase of lactation and to physical examination of the mammary gland, and milk examination. Samples were divided into two aliquots, and were collected after California Mastitis Test evaluation. One aliquot was used in automatic cell counts, and the other, in direct microscopic count using methyl green-pyronine-Y stain. CMT results were as follows: 74.5% of the samples were negative, 8.8% yielded traces, 8.8% were weak positive (1), 6.8% were distinct positive (2) and 0.9% were strong positive (3). Medians of somatic cell counts in goat milk as evaluated by means automatic cell counter and direct microscopy, and grouped according to the different CMT scores, were as follows: 181,000, 578,000, 628,000, 1,421,500, and 5,542,000 cells/mL of milk and 74,991, 271,396, 71,420, 640,995, and 5,049,394 cells/mL of milk in scores negative, traces, 1, 2 and 3, respectively. Medians obtained in automatic cell counts and direct microscopic counts, grouped according to the phase of lactation were 159,500; 508,000; and 277,500 cells/mL of milk and 62,493; 89,275; and 146,411 cells/ml of milk, respectively. The correlation between the automatic and microscopic methods for somatic cell counts was 88%. Based on the results obtained, it could be concluded that there were differences between the automatic and microscopic methods for somatic cell counts, being this most adequate for the determination of the celularidade in the cellularity of goat milk.

O eritrograma é uma avaliação que representa um auxílio ao clínico no diagnóstico de afecções que acometem os animais domésticos. No entanto, estudos hematológicos em bubalinos são escassos, havendo poucas informações na literatura a respeito de valores de referência dos constituintes sanguíneos desta espécie. Assim, este trabalho teve como objetivo a análise do eritrograma de búfalos de diferentes faixas etárias da raça Murrah, machos e fêmeas. Os animais em estudo foram distribuídos em quatro grupos experimentais, de acordo com as idades: Grupo 1: animais com idade entre o nascimento e 3 meses (n=15); Grupo 2: animais com idade entre 4 e 6 meses (n=50); Grupo 3: animais com idade entre 7 e 12 meses (n=50); e Grupo 4: animais com idade superior a 12 meses (n=50). Como resultados das análises foram encontrados os seguintes valores médios, no Grupo 1: 7,9x106 hemácias/mL de sangue (He); 13,0 g/dL de hemoglobina (Hb); hematócrito (Ht) de 38,9%; Volume Corpuscular Médio (VCM) de 49,0 fl; Concentração Hemoglobínica Corpuscular Média (CHCM) de 33,6 %, e Hemoglobina Corpuscular Média (HCM) de 16,4 pg; no Grupo 2: He: 7,1 x10(6)/mL; Hb: 12,5 g/dL; Ht: 36,8%; VCM: 52,4 fl; CHCM: 33,9%, e HCM: 17,8 pg; no Grupo 3: He: 7,9 x10(6)/mL; Hb: 12,0 g/dL; Ht: 33,8%; VCM: 43,1 fl; CHCM: 35,4%, e HCM: 15,34 pg; e no Grupo 4: He: 6,7 x10(6)/mL; Hb: 11,7 g/dL; Ht: 34,4%; VCM: 53,4 fl; CHCM: 34,4%. e HCM: 17,4 pg. A análise estatística dos resultados encontrados para as diferentes faixas etárias permitiu concluir que ocorreram variações determinadas pela evolução da idade, caracterizadas por diminuição do número médio de hemácias (He), da concentração de hemoglobina (Hb) e do valor do hematócrito (Ht). However, hematological studies involving buffaloes are scarce, with little information in the literature on reference values for blood components in this species. Therefore, the objective of this study was analyze erythrograms of Murrah breed buffaloes of different ages, both males and females. Buffaloes in the study were distributed into four experimental groups, according to their ages: Group 1, animals from birth to 3 months of age (n=15); Group 2, animals from 4 to 6 months of age (n=50); Group 3, animals from 7 to 12 months of age (n=50); and Group 4, animals from 13 months to five years of age (n=50). The following values were obtained, for Group 1: 7.9x10(6) erythrocytes/mL (He), 13.0g/dL hemoglobin (Hb), hematocrit (Ht) 38.9%, Mean Corpuscular Volume (MCV) 49.0 fl, Mean Hemoglobin Corpuscular Concentration (MCHC) 33.6%, and Mean Corpuscular Hemoglobin (MCH) 16.4 pg; for Group 2: He7.1x10(6)/mL, Hb12.5g/dL, Ht 36.8%, MCV 52.4 fl; MCHC 33.9%; and MCH 17.8 pg; for Group 3: He 7.9x10(6)/mL, Hb12.0g/dL, Ht 33.8%, MCV 43.1 fl, MCHC 35.4%, and MCH 15.34pg; for Group 4: He 6.7x10(6)/mL, Hb 11.7g/dL, Ht 34.4%, MCV 53.4 fl, MCHC 34.4%, and MCH 17.4 pg. Statistical analysis of the results for the different age ranges permitted the conclusion that when the animals got older there was a decrease in the mean red cell counts (He), hemoglobin concentration (Hb) and hematocrit values (Ht). Although hematometric indexes showed significant variations in G3.

The objectives of the present study were the analysis of cell counts, the comparison between different methods for counting cells and the quantification of different types of cells observed in the milk of Lacaune sheep, using automatic somatic cell counts (SCC) and direct microscopic SCC. Milk smears were stained with methyl green-pyronin-Y and Rosenfeld stain. Mean of automatic cell counts was 0.145 x106 cells/mL of milk, and mean of direct microscopic counts using methyl green-pyronin-Y and Rosenfeld were respectively 0.157 and 0.193 x106 cells /mL of milk. Samples used in direct microscopic counts were evaluated according to the different types of cells and the following results were recorded: 0.094 and 0.14 x106 mononuclear cells /mL, 0.063 and 0.051 x106 polymorphonuclear cells /mL of milk using methyl green-pyronin-Y and Rosenfeld stain, respectively. It may be concluded that there were differences in the methods used for counting cells. The number of cells recorded was lower when methyl green-pyronin-Y and automatic counts were used, compared with direct microscopy using Rosenfeld stain.KEY WORDS: somatic cells and cytology, lacaune, milk, sheep. O objetivo deste trabalho foi avaliar o perfil microbiológico e celular do leite no período lactante e de involução ativa de ovelhas da raça Santa Inês. Foram avaliadas amostras lácteas de 12 ovelhas durante estes distintos períodos. Realizou-se o exame físico da mama, sendo as amostras lácteas submetidas à contagem de células somáticas (CCS), ao California Mastitis Test (CMT), ao exame microbiológico e aos testes de sensibilidade in vitro dos patógenos encontrados. Foram observados maiores escores do exame físico, CCS, CMT durante o período de involução ativa, além de uma alta persistência da infecção durante estes períodos. O período de involução ativa não se mostrou como um momento de alta susceptilidade. Os estafilococos coagulase negativa representaram o único gênero isolado das glândulas infectadas. Uma alta sensibilidade dos agentes etiológicos envolvidos frente aos diferentes antimicrobianos in vitro foi também observada.The aim was to evaluate and compare the microbiological and cellular profile of the milk of Santa Ines ewes during the lactation period and the active involution. Milk samples were analyzed from 12 ewes during these distinct periods. Clinical examination of the mammary gland, somatic cell count (SCC), California Mastitis Test (CMT), bacteriologic screening and sensibility of the pathogens in vitro were performed. Most alterations were observed in the active involution period. SCC and CMT were higher in this same period. Besides this, a high persistency of infection occurred. The active involution period did not show high susceptibility. Coagulase-negative staphylococci were the only isolated bacteria. A high antimicrobial sensibility of these pathogens was also encountered.

The aim of this study was to evaluate the hematological profile of Holstein calves during the first month of life. Blood samples were harvested (n = 208) from 26 calves, from birth until 30 days of life. Hematologic values were determined by an automatic system associated with differential leukocyte count by manual methods. Variations in the erythrogram components were detected from birth up to the 30th day of life, except for hemoglobin (Hb) concentration and Mean Corpuscular Hemoglobin Concentration (MCHC). At birth, higher values were observed for hematocrit, Mean Corpuscular Volume (MCV) and Mean Corpuscular Hemoglobin (MCH) that decreased in subsequent moments. During the first days of life, leukocytosis was found due to neutrophilia and eosinopenia. A gradual increase of lymphocytes with the increase of age was also observed. Finally, the present research showed that the first month of life is a hematological adaptation period. Based on the results, it detected that blood component variations, characterized by hemoconcentration and leukocyte prolife compatible with glucocorticoids response up to the 4th day of life, were responsible for neutrophil lymphocyte ratio > 1.0 at birth.

Foi realizado o estudo retrospectivo das afecções do sistema locomotor de ruminantes atendidos no Serviço de Clínica de Bovinos e Pequenos Ruminantes da Faculdade de Medicina Veterinária e Zootecnia da Universidade de São Paulo (FMVZ-USP), no período de 2000 a 2012. Nesse período, foram atendidos 209 casos de animais com problemas locomotores, dos quais 62,7% localizados na região distal dos membros e 37,3% na região proximal. Na espécie bovina, com 121 (57,9%) casos atendidos, o comprometimento da região distal dos membros foi observado em 86 (71,07%) e da região proximal em 35 (28,93%) dos casos, respectivamente. A afecção mais frequente observada em bovinos foi a hiperplasia interdigital com 26,74% (23) dos casos atendidos. Entre as afecções na região proximal, houve maior ocorrência de fraturas, com 48,6% (17) dos casos. Foram atendidos 88 (42,1%) pequenos ruminantes, apresentando lesões na região distal em 51,1% (45) dos casos e 48,9% (43) com lesões na região proximal. Nessas espécies, as lesões de maior ocorrência nas regiões distais e proximais foram, respectivamente, o foot-rot (60%) e as fraturas (77,4%). As afecções do sistema locomotor dos ruminantes foram pouco frequentes entre os animais atendidos no período estudado. Quanto ao prognóstico, foi bom nos animais acometidos com doenças podais, diferentemente das afecções proximais dos bovinos, principalmente fraturas, que apresentaram prognóstico mau.

A influência do estágio de lactação na composição do leite de cabras foi estudada em 27 animais lactantes da raça Saanen durante oito meses, colhendo-se o total de 304 amostras. Foram selecionados e mantidos, no decorrer do trabalho, animais saudáveis, sem qualquer alteração no exame físico da glândula mamária e cujo leite manteve-se negativo ao exame microbiológico. Avaliaram-se os teores de gordura, proteína, lactose e sólidos totais. A concentração de sólidos totais, gordura e lactose declinaram durante a lactação, porém os teores de proteína foram praticamente estáveis durante o período estudado. Os resultados obtidos permitem concluir que a constituição do leite de cabras sofreu influência do estágio de lactação.

ABSTRACT: The objective of this study was to evaluate the influence of viable cells from colostrum on immune development in dairy heifer calves during the first 28 days of life. The animals were distributed between 2 groups: COL+ (n=9) receiving fresh whole colostrum from their own damns; and COL- (n=10) receiving pooled frozen colostrum, containing no viable cells, from a pool of donor cows. These calves were assessed before colostrum intake (D0), 48 hours of age (D2), and weekly from D7 to D28. The development of immunity was evaluated by assessment of the phenotype of blood leukocyte subsets, and induced cytokine production after 72 hours of stimulation in culture with concanavalin A (ConA), killed Staphylococcus aureus (S.aureus) and killed Escherichia coli (E. coli) by peripheral blood mononuclear cell (PBMC). The clinical history of these calves was marked by a high frequency of diarrhea in both groups. However, COL- had greater diarrhea intensity scores (fecal score~3 of 4), and rectal temperature on D7 than COL+ calves. Moreover, bronchopneumonia (n=1) and navel inflammation were observed only in COL- calves. COL- had a lower concentration of serum iron, and a higher absolute number of lymphocytes on D7 than COL+. COL- also had a higher percentage of anemic calves than the COL+ calves on D21 and D28. In general, the percent of cells within each subset of leukocytes was similar between the groups over the experiment, except on week 1 when COL- calves had a higher percentage of lymphocytes expressing CD45RO+ (P=0.07). A steady increase in CD45RO+ and concomitant decline in CD45RO- leukocytes was observed over the course of the study, indicating the development of immune memory. The proportion of CD14MHCII+ leukocytes increased with age (P≤0.05). The median background cytokine production by PBMC that were not stimulated was below the level of detection of the assays used for both groups. The PBMC from COL+ calves stimulated with ConA secreted a larger quantity of IL-17 week 2 (COL+=2060.0pg/mL and COL-=0.0pg/mL, P=0.00). PBMC from COL+ calves stimulated with killed S. aureus whole cell antigen (P=0.05) and killed E. coli whole cell antigen (P=0.05) also secreted higher levels of IL17 than COL- calves at week 4. Clear production of IL17 was observed in PBML from COL+ calves at week 2, but the difference was not statistical different between groups. In conclusion, calves fed fresh and frozen colostrum showed no difference in cells subset profile overall. The increased percentage of leukocytes expressing the memory CD45RO+ or CD14MHCII+ over the course of the experiment indicated a maturation of the adaptive immune response after natural exposure to pathogens in the environment of the calf. The enhanced IL17 secretion by COL+ calves indicated that viable maternal cells modulated T-cell Th17 production that was primed by bacterial antigens. This mechanism could be responsible for quick and efficient activation of neutrophils for bacterial clearance. The differences in cytokine production observed between groups may help to explain the different clinical pictures observed for calves COL+ and COL- calves.
