Currently, submerged cultivation of the Bacillus anthracis STI-1 strain for live anthrax vaccine production requires liquid nutrient media, which have disadvantages of a short shelf life (no more than one month) and a narrow range of storage temperatures (2–8 °С). Dry media, in contrast, have a number of indisputable advantages: such media are transportable and easy to use, have a standard capability to retain properties, and can be stored without preservatives at 2–30 °С for 2–5 years. The aim of this work was to develop a technology for the preparation of a dry nutrient medium for anthrax vaccine production. Materials and methods: The study used the Bacillus anthracis STI-1 vaccine strain and a nutrient medium for its cultivation, containing a 70:30 mixture of an enzymatic digest of casein and a pre-processed corn extract solution. Drying of the nutrient medium was carried out on a spray-drying unit. The authors evaluated physicochemical parameters of experimental medium batches. The shelf life was determined by an accelerated stability study. The dry nutrient medium was used to produce a live anthrax vaccine. Quality attributes of the vaccine were assessed for compliance with regulatory requirements. Results: The authors developed the dry media production technology. According to it, the liquid nutrient medium is fed to the drying unit at a rate of 20–25 dm3/h. The spray air pressure is 0.02 MPa. Temperatures at the drying chamber inlet and outlet are 118–122 °С and 85–90 °С, respectively. The technology was used to obtain 3 experimental batches of the dry medium. The study results demonstrate that the technology is reproducible, and the tested quality attributes of experimental medium batches are consistent with the requirements. According to the accelerated stability study, the shelf life of the dry nutrient medium at 2–30 °С is at least 3 years. Experiments demonstrated the possibility of using the dry nutrient medium for live anthrax vaccine production. Critical quality attributes of the vaccine obtained with the medium met regulatory requirements. Conclusions: The developed technology allows for the production of a standard dry nutrient medium with a prolonged shelf life, which is convenient for live anthrax vaccine production.

The aim of the study was to improve the procedure for the Yersinia pestis EV strain cell concentration using the system for tangential flow microfiltration with the ASF-020 filter support unit.Materials and methods. The study used the vaccine Y. pestis EV strain derived from NIIEG cell line. Submerged cultivation of the native culture was performed using BIOR-0.25 reactor with automated control system. Microbial suspension concentrate was produced through microfiltration applying (Adaptive filtration system) AFS-009 and AFS-020 installations. The content of live microbial cells was determined by cytorefractometry. Assessment of the resistance of Y. pestis EV strain cells to technological factors was performed by photometric registration of changes in the optical density of bacterial suspension during the lytic response of cells to sodium dodecyl sulfate. Physical-chemical and immunobiological properties of the dry live plague vaccine were determined in accordance with the pharmacopoeial item.3.3.1.0021.15 of the State Pharmacopoeia of the Russian Federation, 14th edition.Results and discussion. The design features of the equipment introduced made it possible to carry out membrane filtration of microbial suspension, using BIOR-0.25 reactor as an intermediate storage unit, thereby excluding three technological stages. The total concentration of microbes in the suspension obtained by routine and improved methods was more than 150 billion microbial cells per ml. A comparative study of the effect of various hydrodynamic regimes in the working cavities of AFS-009 and AFS-020 filter units did not significantly affect the morphometric properties and resistance of microbial cultures to extreme (technological) factors. Based on the experimental data, the mass balance of the membrane filtration process has been determined. The optimized technology gave 0.13 liter yield of concentrate from 1 liter of native culture, and the process duration was reduced to 5 hours, the yield of the finished product in one production cycle was increased by 3 times. Thus, the process of concentrating Y. pestis EV strain cells during the production of the tablet form of live plague vaccine has been enhanced. A comparative study of the morphometric properties and resistance of plague microbe cultures to technological factors in the process of their concentration using optimized technology did not reveal any significant differences as compared to the routine one. Technological stage of concentrating has been reduced to 5 h term with a three-fold increase in the yield of finished product.

Bladder cancer relapses are associated with diffuse pattern of neoplastic changes in the mucous layer of the bladder; treatment efficacy depends on the degree of structural alterations in the bladder wall.Materials and methods: for evaluation of microcirculation characteristics and structural alterations of mucous layer of the bladder we have used laser analyzer of capillary blood circulation («ЛАКК-01» produced by НПП «ЛАЗМА», Russia) and compact mobile optic tomograph (ИПФ РАН, Russia).Results: conducted researches have shown alterations affecting both epithelial and submucous layers with their microcirculatory bed in perifocal to bladder tumor tissues.Conclusions: complimentary application of laser Doppler flowmeter and compact mobile optic tomograph allows defining the type and extent of pathologic process in the bladder wall in superficial tumors and choosing optimal method of correction.

Introduction. The effectiveness of methods for treating chronic prostatitis, which are aimed at improving the hemodynamics of the pelvic organs, has been proven by numerous studies conducted over more than thirty years. At the same time, pelvic myofascial syndrome can lead to impaired pelvis' hemodynamics. Purpose of research. This study aimed to determine the effect of myofascial syndromes on the blood circulation in the prostate in patients with chronic bacterial prostatitis, as well as an assessment of the dynamics of changes in blood flow in the prostate on the treatment of concomitant myofascial syndrome. Materials and methods. 59 men 34-52 years old with various clinical manifestations of myofascial syndrome (patients of the neurological clinic) were examined to assess the dynamics of blood circulation in the prostate. In addition, 127 men aged 28-52 years (mean age 38 years) with typical clinical manifestations of chronic prostatitis, who repeatedly and unsuccessfully received treatment for this disease, evaluated the effect of treatment of concomitant myofascial syndrome on prostate hemodynamics Results. The results suggest violations of microcirculation in the prostate in patients with chronic pelvic pain with myofascial syndrome. At the same time, hemodynamic impairment in the prostate is directly associated with impaired pelvic hemodynamics in general. The state of microcirculation in the prostate depends on the severity of pain symptoms in nonspecific chronic pelvic pain, and the treatment of nonspecific chronic pelvic pain leads to a reduction in the main symptom of the disease (pain) and to the normalization of microcirculation in the prostate. Treatment of concomitant myofascial syndrome in patients with chronic bacterial prostatitis allowed to achieve the disappearance of clinical manifestations of the disease and normalization of hemodynamics in the prostate, as well as a decrease in laboratory signs of inflammation. Conclusions. Concomitant myofascial syndrome in patients with chronic pelvic pain is a cause of pelvic hemodynamic disorders and because of the development of the inflammatory process. The severity of hemodynamic disturbances directly depends on the intensity of painful manifestations of the myofascial syndrome. Treatment of myofascial syndrome in patients with chronic bacterial prostatitis is accompanied by the disappearance of disease's clinical symptoms and the restoration of prostate's blood circulation, as well as the reduction or disappearance of inflammation's signs.

To date, the technology of live plague vaccine production uses a combined method of concentrating microbial cells which consists of three operations with a total duration of 18 hours. The procedure of obtaining concentrate, which is used in the current vaccine production technology, has a number of disadvantages, namely: multiple operations, high energy consumption, long duration, and, as a consequence, low yield of concentrated suspension (0.04 l from 1 l of native culture). The aim of the study was to optimise the procedure of Yersinia pestis ЕV microbial cell concentration using the system for tangential flow microfiltration with the ASF-020 filter support unit. Materials and methods: the vaccine strain used in the study was Yersinia pestis ЕV derived from NIIEG cell line (the strain of the Research Institute of Epidemiology and Hygiene). Submerged cultivation of the native culture was performed using the BIOR-0.25 reactor. The content of live microbial cells was determined by cytorefractometry. Oxidative metabolism was assessed using the chronoamperometric method. Physico-chemical and immunobiological properties of the dry live plague vaccine were assessed according to the monograph FS.3.3.1.0022.15 of the State Pharmacopoeia of the Russian Federation, 14 edition. Results: the equipment’s design features made it possible to carry out membrane filtration of the microbial suspension using the BIOR-0.25 reactor as an intermediate storage unit, thereby excluding three technological stages. The total concentration of microbes in the suspension obtained by the routine and the optimised methods was not less than 120 billion microbial cells/ml. A comparative study of the effect of various hydrodynamic regimes in the working cavities of ASF-009 and ASF020 filter units did not significantly affect the morphometric and physiological properties of microbial cultures. Experimental data helped to determine the process mass balance of membrane filtration. The optimised technology gave 0.17 l yield of the concentrate from 1 l of native culture, and the process duration was reduced to 4 hours. Conclusions: the process of concentrating Y. pestis EV microbial cells during production of plague vaccines was optimised. A comparative study of morphometric and physiological properties of plague microbe cultures that was carried out during their concentration using the optimised technology did not reveal any significant differences as compared to the routine one.

Metabolic syndrome can lead to several challenging complications including degeneration of the pancreas and hypogonadism. Recently, we have shown that Bisamide Derivative of Dicarboxylic Acid (BDDA) can contribute to pancreatic restoration in mice with metabolic disorders via its positive effects on lipid and glucose metabolism, and by increasing the numbers of pancreatic stem cells. In the present study, we hypothesized that BDDA might also be effective in restoring hypogonadism caused by metabolic syndrome. Experiments were performed on male C57BL/6 mice with hypogonadism, where metabolic disorders have been introduced by a combination of streptozotocin treatment and high fat diet. Using a combination of histological and biochemical methods along with a flow cytometric analysis of stem and progenitor cell markers, we evaluated the biological effects of BDDA on testicular tissue, germ cells, spermatogonial stem cells in vitro and in vivo, as well as on fertility. We demonstrate that in mice with metabolic disorders, BDDA has positive effects on spermatogenesis and restores fertility. We also show that BDDA exerts its therapeutic effects by reducing inflammation and by modulating spermatogonial stem cells. Thus, our results suggest that BDDA could represent a promising lead compound for the development of novel therapeutics able to stimulate regeneration of the testicular tissue and to restore fertility in hypogonadism resulting from complications of metabolic syndrome.

Idiopathic pulmonary fibrosis (IPF) is a chronic progressive disease characterized by interstitial fibrosis and progressive respiratory failure. Pirfenidone and nintedanib slow down but do not stop the progression of IPF. Thus, new compounds with high antifibrotic activity and simultaneously regenerative activity are an unmet clinical need. Recently, we showed that Treamid can help restoring the pancreas and testicular tissue in mice with metabolic disorders. We hypothesized that Treamid may be effective in antifibrotic therapy and regeneration of damaged lung tissue in pulmonary fibrosis. In this study, experiments were performed on male C57BL/6 mice with bleomycin-induced pulmonary fibrosis. We applied histological and immunohistochemical methods, ELISA, and assessed the expression of markers of endothelial and epithelial cells in primary cultures of CD31+ and CD326+ lung cells. Finally, we evaluated esterase activity and apoptosis of lung cells in vitro. Our data indicate that Treamid exhibits antifibrotic activity in mice with pulmonary fibrosis and has a positive effect on capillaries of the lungs. Treamid also increases the number of endothelial progenitor cells in the lungs of animals with pulmonary fibrosis. Lastly, Treamid increases esterase activity and decreases apoptosis of CD31+ lung cells in vitro. Based on these findings, we suggest that Treamid may represent a promising compound for the development of new antifibrotic agents, which are capable of stimulating regeneration of lung endothelium in IPF patients.

In clinical practice, the metabolic syndrome can lead to multiple complications, including diabetes. It remains unclear which component of the metabolic syndrome (obesity, inflammation, hyperglycemia, or insulin resistance) has the strongest inhibitory effect on stem cells involved in beta cell regeneration. This makes it challenging to develop effective treatment options for complications such as diabetes. In our study, experiments were performed on male C57BL/6 mice where metabolic disorders have been introduced experimentally by a combination of streptozotocin-treatment and a high-fat diet. We evaluated the biological effects of Bisamide Derivative of Dicarboxylic Acid (BDDA) and its impact on pancreatic stem cells in vivo. To assess the impact of BDDA, we applied a combination of histological and biochemical methods along with a cytometric analysis of stem cell and progenitor cell markers. We show that in mice with metabolic disorders, BDDA has a positive effect on lipid and glucose metabolism. The pancreatic restoration was associated with a decrease of the inhibitory effects of inflammation and obesity factors on pancreatic stem cells. Our data shows that BDDA increases the number of pancreatic stem cells. Thus, BDDA could be used as a new compound for treating complication of the metabolic syndrome such as diabetes.

In clinical practice, the metabolic syndrome (MetS) is often associated with chronic obstructive pulmonary disease (COPD). Although gender differences in MetS are well documented, little is known about sex-specific differences in the pathogenesis of COPD, especially when combined with MetS. Consequently, it is not clear whether the same treatment regime has comparable efficacy in men and women diagnosed with MetS and COPD. In the present study, using sodium glutamate, lipopolysaccharide, and cigarette smoke extract, we simulated lipid metabolism disorders, obesity, hyperglycemia, and pulmonary emphysema (comorbidity) in male and female C57BL/6 mice. We assessed the gender-specific impact of lipid metabolism disorders and pulmonary emphysema on angiogenic precursor cells (endothelial progenitor cells (EPC), pericytes, vascular smooth muscle cells, cells of the lumen of the nascent vessel), as well as the biological effects of pegylated glucagon-like peptide 1 (pegGLP-1) in this experimental paradigm. Simulation of MetS/COPD comorbidity caused an accumulation of EPC (CD45&minus, CD31+CD34+), pericytes, and vascular smooth muscle cells in the lungs of female mice. In contrast, the number of cells involved in the angiogenesis decreased in the lungs of male animals. PegGLP-1 had a positive effect on lipids and area under the curve (AUC), obesity, and prevented the development of pulmonary emphysema. The severity of these effects was stronger in males than in females. Furthermore, PegGLP-1 stimulated regeneration of pulmonary endothelium. At the same time, PegGLP-1 administration caused a mobilization of EPC (CD45&minus, CD31+CD34+) into the bloodstream in females and migration of precursors of angiogenesis and vascular smooth muscle cells to the lungs in male animals. Gender differences in stimulatory action of pegGLP-1 on CD31+ endothelial lung cells in vitro were not observed. Based on these findings, we postulated that the cellular mechanism of in vivo regeneration of lung epithelium was at least partly gender-specific. Thus, we concluded that a pegGLP-1-based treatment regime for metabolic disorder and COPD should be further developed primarily for male patients.

The investigations of tungsten transport in T-10 plasma are presented in the current paper. The process of tungsten accumulation in OH discharges and its dependence on parameters ne, Ip, and Zeff are discussed. Peaked tungsten density profiles nW(r) are observed in plasmas with Zeff ≈ 1–1.5. In plasmas with Zeff ≈ 3–4, W accumulation is mainly integral with no profile peaking with regard to electron density ne(r). Lower densities of W are observed in discharges with a lithized T-10 vacuum vessel in comparison with plasma contaminated with light impurities. The disposal of W accumulation is produced by auxiliary ECRH input. During the central ECRH, the tungsten density profile flattens with the central density drop off by ~10 times together with W source increase up to 4 times.

Dirofilariasis is the only human filariasis, found in the territory of the Russian Federation. Mosquitoes are carriers of the disease. Changing climate conditions in the direction of warming, the availability of international travel, non-compliance with preventive measures lead to an increase in the number of cases of infection by helminths, including rare species.

The role ofimmunomodulatory therapy to stimulate bactericidal properties humoral defensecells of neutrophilsin patients suffering fromrecurrent bladder infection. Found that the inclusion ofthe immunomodulatorin the treatment regimen of exacerbations of chronic cystitis contributes to increasing the activation of phagocytic index of neutrophils and reserve leads to a significant decrease in the frequency of exacerbations of the disease after basic andpreventive courses of treatment.

The article represents the study of quality of life in patients with muscle-invasive bladder cancer stages T2b and T3a after surgery, depending on the operation type. The study was conducted using questionnaires EORTC QLQ-30 and FACT-BL. The best levels of quality of life were observed in patients after organ-saving treatment of muscle-invasive bladder cancer.

The authors determined the detection rates of prostatic intraepithelial neoplasia (PIN) in 2317 patients with benign prostatic hyperplasia (BPH) and prostate cancer (PC); and those of chronic prostatitis and fibrosis in patients with PIN, BPH, or PC. There was no difference in median age between the groups. PC was found to be more concurrent with PIN 2 than with BPH. The severer inflammation or fibrosis is, more likely there is a concomitance with PIN 2 or PC. There is evidence for the theory of inflammation is a factor of carcinogenesis. Prostatic fibrosis may also initiate carcinogenesis.

The investigation examined the feasibility of cross-polarization optical cohe-rence tomography (CP OCT) to detect early urinary bladder cancer (UBC). Studies were performed in 376 patients; 5290 images were obtained using an OCT 133-U optical coherence tomograph. To acquire and compare intrared-light scattering images in baseline and orthogonal polarizations is the basis of CP OCT; their analysis makes it possible to judge from the state of the epithelium/connective tissue system and to obtain information on changes in tissue depolarizing components, collagen in particular. The authors elaborated criteria as determinants of the nature of CP OCT changes in direct and orthogonal polarizations in health, inflammatory changes, and UBC at its early stage - urothelial dysplasia and carcinoma in situ in flat suspected areas.

Whether cross-polarization (CP) optical coherence tomography (OCT) could be used to detect early bladder cancer was ascertained; it was compared with traditional OCT within the framework of blind (closed) clinical statistical studies. One hundred and sixteen patients with local nonexophytic (flat) pathological processes of the bladder were examined; 360 CP OCT images were obtained and analyzed. The study used an OCT 1300-U CP optical coherence tomographer. CP OCT showed a high (94%) sensitivity and a high (84%) specificity in the identification of suspected nonexophytic areas in the urinary bladder.

In clinical practice, there are patients with a combination of metabolic syndrome (MS) and chronic obstructive pulmonary disease (COPD). The pathological mechanisms linking MS and COPD are largely unknown. It remains unclear whether the effect of MS (possible obesity) has a major impact on the progression of COPD. This complicates the development of effective approaches for the treatment of patients with a diagnosis of MS and COPD. Experiments were performed on female C57BL/6 mice. Introduction of monosodium glutamate and extract of cigarette smoke was modeled to simulate the combined pathology of lipid disorders and emphysema. Biological effects of glucagon-like peptide 1 (GLP-1) and GLP-1 on endothelial progenitor cells (EPC) in vitro and in vivo were evaluated. Histological, immunohistochemical methods, biochemical methods, cytometric analysis of markers identifying EPC were used in the study. The CD31+ endothelial cells in vitro evaluation was produced by Flow Cytometry and Image Processing of each well with a Cytation&trade, 3. GLP-1 reduces the area of emphysema and increases the number of CD31+ endothelial cells in the lungs of mice in conditions of dyslipidemia and damage to alveolar tissue of cigarette smoke extract. The regenerative effects of GLP-1 are caused by a decrease in inflammation, a positive effect on lipid metabolism and glucose metabolism. EPC are proposed as pathogenetic and diagnostic markers of endothelial disorders in combination of MS with COPD. Based on GLP-1, it is proposed to create a drug to stimulate the regeneration of endothelium damaged in MS and COPD.

Carried out have been two experimental studies on hybridization of mouse myeloma cells and lymphocytes of BALB/c mice immunized with inactivated microbe Francisella tularensis cultures. As a result obtained have been hybridomas-producers of monoclonal antibodies (MAb) specific to the antigens of tularemia agent. Evaluated have been the prospects of its application for the detection of the agent under discussion using enzyme-linked immunoassay. Established is the fact that monoclonal antibodies produced by 31G1F10, 32E5D3, 35B11C8, 36C2F11 hybridomas make it possible to identify microbe cells of various tularemia agent strains when concentrated up to 0.5·106 mc/sm3, and do not interact with cultures of heterologous microorganisms when concentrated to 1.0·108 mc/sm3, which testifies to their specificity. These MAb are planned to be used for the construction of immune-enzyme and immune-chromatographic test-systems designed for tularemia agent detection.

The article represents the study of survival rate (common, oncology-specific, disease-free) and frequency and nature of the recurrence after organ-saving surgery of patients with muscleinvasive bladder cancer stages T2b and T3aN0M0G1-G3 with full diagnosis of bladder mucosa, the adjuvant intravesical chemotherapy on pre-operative stage. Oncology-speсific and diseasefree survival 5-year rates with full diagnosis of bladder mucosa, the adjuvant intravesical chemotherapy on pre-operative stage were higher. Recurrence of bladder cancer were on the fields, other than the zones of operations, the stages of disease were lower than that of the original tumor. These factors indicated the presence of an unrecognized cancer in situ on the walls of the urine bladder.

Cрывы разряда в крупных токамаках и будущих реакторах являются серьёзной проблемой, ограничивающей срок их службы. Представлены результаты исследований влияния напуска благородных газов на динамику развития срыва разряда в токамаке Т-10. Срывы разряда вызывались разными способами: наращиванием плотности плазмы до предельной и/или инжекцией дейтериевой или примесной (углеродной) макрочастицы. Инжекция благородных газов (гелия, аргона или ксенона) проводилась как в стационарной стадии для инициации срыва разряда, так и во время спада тока. Обнаружено, что спад тока при срыве разряда в Т-10 происходит в две фазы (медленную и быструю), отличающиеся существенно разным характерным временем. Инжекция дейтериевых макрочастиц при параметрах имеющегося инжектора не влияет на динамику спада тока, а массивный напуск благородных газов при выcоком давлении приводит к переходу спада тока из медленной фазы в быструю и инициации вторичных МГД-возмущений во время не слишком быстрого спада тока, а также к подавлению пучка ускоренных электронов.

The article represents the study of frequency and nature of the recurrence and survival rate (common, oncology-specific, disease-free) after organ-saving surgery of patients with muscle-invasive bladder cancer stages T2b and T3a. Oncology-speсific and disease-free survival rates were much higher if full diagnosis of bladder mucosa, the adjuvant intravesical chemotherapy had been on pre-operative and intra-operative stages than in the absence of these diagnosis and therapy. Recurrentes of bladder cancer which appeared in the absence of diagnosis and combination therapy, statistically reliably occured at another location other than the zone of operation, stage of recurrentes and degree of differentiation of recurrents were less than the original tumor. This information confirms the existence of foci of cancer in situ which have not been identified on the diagnostic stage.

A diagnostic neutral beam injector DINA-KI60 has been developed and produced for upgraded T-15 tokamak (Kurchatov Institute, Moscow). The system will be primarily used for measurement of the ion temperature by charge-exchange recombination spectroscopy (CXRS), for magnetic field measurements via motional Stark effect (MSE) and investigation of plasma turbulence by Beam emission spectroscopy (BES). The ion source provides 60 keV, 6 A hydrogen beam with low angular divergence less than 10 mrad. Ions are extracted from a plasma created by an arc-discharge source with cold cathode and, after accelerating and focusing, are neutralized in a gas target. The extracted beam has about 86% of full energy specie. The grids of ion optical system are spherically curved providing geometric focusing of the beam at a distance 4 m. Diagnostic neutral beam has been successfully tested at Budker Institute of Nuclear Physics at 2 seconds pulse duration with modulation of the beam (duty ratio from 1:1 to 1:3).

Tungsten limiters were tested in the T-10 tokamak. The limiters were made from the ITER-grade WMP "POLEMA" tungsten. The influence of the edge tokamak plasma on tungsten limiters leads to significant cracking of tungsten. The heat load of up to 2 MW m−2 leads to the micro-crack development at the grain boundaries accompanied by the loss of grains. The heat loads that exceed 5 MW m−2 lead to the macro crack development. Under the present T-10 tokamak conditions, the heat and particle fluxes in the edge plasma lead to the significant destruction of tungsten limiters during the experimental campaign. During the disruption and runaway electron formation, extreme heat loads of more than 1 GW/m2 cause strong melting of tungsten on the inner and outer part of the ring limiter.

Bleomycin-induced lung inflammation and fibrosis were assessed in a model of idiopathic pulmonary fibrosis in C57BL/6 mice and effect of spiperone on histopathological lung indicators, hematopoietic (HSC) and mesenchymal (MSC) stem cells and progenitor cells was characterized. Spiperone is a selective antagonist of D2 dopamine receptors which disturbs dopamine mediation. Hematoxylin and eosin staining showed spiperone decreased alveolar epithelial edema, exudation and infiltration of the alveoli walls and lumen by inflammatory cells (neutrophils, macrophages, plasma cells) after bleomycin instillation. Picrofuchsin staining by Van Gieson revealed spiperone decreases the area of connective tissue in the lung fibrotic phase of pulmonary fibrosis. ELISA assay determined the decrease levels of collagen type I, hydroxyproline and total collagen in lung homogenate after spiperone treatment. Number of “long-term” HSCs (Lin- Sca-1+c-Kit+CD34-), «short-term” HSCs (Lin-Sca-1+c-Kit+CD34+), hematopoietic progenitor cells and MSC-like cells in lung with pneumofibrosis decreased after spiperone treatment. This spiperone effect we connect to the violation of immature bone marrow cells migration. Additionally, spiperone inhibited clonal activity of hematopoietic (CFU-GEMM, CFU-G) and mesenchymal (CFU-F) progenitor of bone marrow, blood and lungs. An additional feature of the spiperone action was the ability to decrease the capacity of self-renewal and MSCs differentiation activity into adipocytes, osteoblasts, chondrocytes and fibroblast cells. Thus, a selective antagonist D2 of dopamine receptors spiperone can act as a potential antifibrotic agent for the treatment of toxic pulmonary fibrosis. The overall conclusion was that the neurotropic agent spiperone is able to influence the stem and progenitor cells in lung pathology effectively.

The parameters of molecular plasma in closed magnetic trap is studied in this paper. Using the system of molecular diagnostics, which was designed by the authors on the «Tokamak T-10» facility, the radiation of hydrogen isotopes at the plasma edge is investigated. The scheme of optical radiation registration within visible spectrum is described. For visualization, identification and processing of registered molecular spectra a new software is developed using MatLab environment. The software also includes electronic atlas of electronic-vibrational-rotational transitions for molecules of protium and deuterium. To register radiation from limiter cross-section a network control system is designed using the means of the Internet/Intranet. Remote control system diagram and methods are given. The examples of web-interfaces for working out equipment control scenarios and viewing of results are provided. After test run in Intranet, the remote diagnostic system will be accessible through Internet.

In order to obtain breakdown and suitable plasma parameters for low-voltage OH start-up, high level of EC-power was injected into T-10 tokamak. Input HF-power was varied in the range of 0.15-1.0 MW. Two HF-launcher systems with different output beams allowed to inject EC-waves with maximum power density 0.25 MW/cm 2 and 0.01 MW/cm 2 . Dependence of breakdown time delay on HF-power was obtained. It was shown, that optimal plasma parameters were achieved in presence of plasma equilibrium currents I=3 kA (input HF-power=1.0 MW). Electron temperature Te=100÷150 eV and electron density ne=5•10 12 cm -3 was measured in these discharges. These parameters remained constant during full HF-pulse-length.

Hyaluronidases are groups of enzymes that degrade hyaluronic acid (HA). To stop enzymatic hydrolysis we modified testicular hyaluronidase (HYAL) by activated polyethylene oxide with the help of electron-beam synthesis. As a result we received pegylated hyaluronidase (pegHYAL). Spiperone is a selective D2 dopamine receptor antagonist. It was demonstrated on the model of a single bleomycin damage of alveolar epithelium that during the inflammatory phase monotherapy by pegHYAL or spiperone reduced the populations of hematopoietic stem /progenitor cells in the lung parenchyma. PegHYAL also reduced the levels of transforming growth factor (TGF)-β, interleukin (IL)-1β, tumor necrosis factor (TNF)-α in the serum and lungs, while spiperone reduced the level of the serum IL-1β. Polytherapy by spiperone and pegHYAL caused the increase of the quantity of hematopoietic stem/ progenitor cells in the lungs. Such an influx of blood cell precursors was observed on the background of considerable fall level of TGF-β and the increase level of TNF-α in the serum and lungs. These results show pegHYAL reduced the bleomycin-induced fibrosis reaction (production and accumulation of collagen) in the lung parenchyma. This effect was observed at a single and repetitive bleomycin damage of alveolar epithelium, the antifibrotic activity of pegHYAL surpassing the activity of testicular HYAL. The antifibrotic effect of pegHYAL is enhanced by an additional instillation of spiperone. Therapy by pegHYAL causes the flow of CD31‒ CD34‒ CD45‒ CD44+ CD73+ CD90+ CD106+-cells into the fibrous lungs. These cells are incapable of differentiating into fibroblast cells. Spiperone instillation separately or together with pegHYAL reduced the MSC-like cells considerably. These data enable us to assume, that pegHYAL is a new and promising instrument both for preventive and therapy of toxic pneumofibrosis. The blockage of D2 dopamine receptors with the following change of hyaluronan matrix can be considered as a new strategy in treatment of pneumofibrosis.

The results of the evaluation of the immunogenic properties of B-antigen, earlier identified in the culture fluid of Yersinia pseudotuberculosis submerged culture, with respect to experimental plague and pseudotuberculosis are presented. B-antigen has been shown to produce protective effect in guinea pigs and, probably, hamadryas baboons, but not in white mice infected with the causative agent of plague. Immunizaton with B-antigen protects guinea pigs from primary pneumonic plague caused by both capsule-forming and noncapsular Y. pestis virulent strains. Passive immunization with antibodies to B-antigen induces limitedly pronounced protective effect in guinea pigs and is not effective for white mice with respect to experimental plague. No active or passive protection of white mice or guinea pigs, infected with Y. pseudotuberculosis cultures, has been achieved by the injection of B-antigen or antibodies to it.

The investigation data of testicles bioptates of 153 subfertile men are given. The examined patients are divided into 2 groups: the first group includes subfertile men with epidemic parotidits in anamnesis (91), the second one includes subfertile men, never being ill with epidemic parotiditis (62). The peculiarity lies in the fact that the morphologic variations of testicles of subfertile men with epidemic parotiditis in anamnesis are similar to the variations of autoimmune nature.

In this study was found that inflammation in interstitial lung tissue of mice C57Bl/6 after intratracheal injection of the bleomycin is accompanied by the increase of the bone marrow and circulating blood cells with the phenotype (CD3, CD45R (B220), Ly6C, Ly6G (Gr1), CD11b (Mac1), TER-119), Sca-1 + , c-Kit + , CD34 - (hematopoietic stem cells (HSCs)) and hematopoietic progenitor cells (granulocyte-erythroid-macrophage-megakaryocytic (CFU-GEMM) and granulocyte precursors (CFU-G)). These results demonstrate that the intraperitoneal reserpine injection reduces alveolar interstitium and alveolar passages infiltration by inflammatory cells and prevents the connective tissue growth in the lung parenchyma. Supposedly, the reserpine anti-inflammatory effect is caused by the reduced activity of the bone marrow HSCs differentiation in CFU-G, the decrease of circulating HSCs and progenitor hematopoietic cells and the violation of their migration in bleomycin-treated lungs. These data suggest the decrease caused by the reserpine in deposition of collagen fibers in the lung's parenchyma associated with the decrease in the inflow of multipotent mesenchymal stromal cells (MSCs) and fibroblast progenitor bone marrow-derived cells to lungs. Thus, reserpine violates the MSCs differentiation into fibroblast-like cells.

Problems of spectroscopic diagnostics of ITER plasma are under consideration. Three types of diagnostics are presented: 1) Balmer lines spectroscopy in the edge and divertor plasmas; 2) Thomson scattering, 3) charge exchange recombination spectroscopy. The Zeeman-Stark structure of line shapes is discussed. The overlapping of isotopes H-D-T spectral line shapes are presented for the SOL and divertor conditions. The polarization measurements of H-alpha spectral lines for H-D mixture on T-10 tokamak are shown in order to separate Zeeman splitting in more details. The problem of plasma background radiation emission for Thomson scattering in ITER is discussed in details. The line shape of P-7 hydrogen spectral line having a wave length close to laser one is presented together with continuum radiation. The charge exchange recombination spectroscopy (CXRS) is discussed in details. The data on Dα, HeII and CVI measurements in CXRS experiments on T-10 tokamak are presented.

The effect of aflatoxin in the doses 0.25, 0.5, 1 and 2 mg per 1 kg of mixed feed and residual aflatoxin in the tissue have been studied. Residual aflatoxin B1 in the liver and muscles of broilers appeared to surpass those reported in the literature. When 49-day-old broilers were deprived of the toxin, its levels in the liver, muscles decreased for 10 days up to the levels 1.1-4.3 micrograms/kg. the inclusion in the chicken feed of methionine (0.3%) or meat-bone flour (5%) weakens the toxin effect and reduces toxin residues content in the tissues 2-3-fold.

The aim of our study was to determine the pathogen supporting the inflammatory process in the genitals of infertile patients, its sensitivity to antibiotics and the effectiveness of treatment measures, carried out taking into account the data obtained by ejaculate culture. Eighty patients aged 21 to 33 years old with secretory toxic form of male infertility were examined. Clinical examination revealed signs of chronic inflammation in the prostate in 32 men, in the testicular appendages in 12 men, and chronic urethritis in 2 men. In 34 patients the inflammatory process had no clinical manifestations, but the ejaculate showed a large number of leukocytes.

Infertile marriage is one of the urgent problems of modern medicine. It is established that 10-15% of couples remain childless, and in 35-60% of cases the cause of infertility is male infertility. The etiology and pathogenesis of the disease have not yet been sufficiently explored, and the results of its treatment are not encouraging. However, there is evidence that male infertility is completely incurable in only 10-12.5% of cases, so the development of new methods to improve the diagnosis and treatment of this pathology is of great importance.
