Upon a variety of environmental stresses, eukaryotic cells usually recruit translational stalled mRNAs and RNA-binding proteins to form cytoplasmic condensates known as stress granules (SGs), which minimize stress-induced damage and promote stress adaptation and cell survival. SGs are hijacked by cancer cells to promote cell survival and are consequently involved in the development of anticancer drug resistance. However, the design and application of chemical compounds targeting SGs to improve anticancer drug efficacy have rarely been studied. Here, we developed two types of SG inhibitory peptides (SIPs) derived from SG core proteins Caprin1 and USP10 and fused with cell-penetrating peptides to generate TAT-SIP-C1/2 and SIP-U1-Antp, respectively. We obtained 11 SG-inducing anticancer compounds from cell-based screens and explored the potential application of SIPs in overcoming resistance to the SG-inducing anticancer drug sorafenib. We found that SIPs increased the sensitivity of HeLa cells to sorafenib via the disruption of SGs. Therefore, anticancer drugs which are competent to induce SGs could be combined with SIPs to sensitize cancer cells, which might provide a novel therapeutic strategy to alleviate anticancer drug resistance.

Understanding the ice formation process in cement-based materials is necessary to evaluate the freeze-thaw durability of concrete structures serviced in cold regions. However, the influencing mechanism of deicing salt on the ice formation process in cement-based material remains unclear. In this paper, the influence of NaCl on the ice formation process in the mortar was investigated by tracking the state and movement of pore water during the ice formation process using the low-field nuclear magnetic resonance (LF-NMR) technique. The obtained results show that during the ice formation process, the movement of pore water after being saturated with NaCl solution from the gel pore to the capillary pore is more distinct, regardless of ordinary and air-entrained mortar. Moreover, the increase of the signal amplitude of LF-NMR illustrates that NaCl solution promotes the conversion of chemically bound water to physically absorbed water. This effect leads to an increase in the free water content, and the saturation of the capillary pores is vulnerable to reaching critical saturation.

IntroductionMajor Depressive Disorder (MDD) is a leading cause of worldwide disability, and standard clinical treatments have limitations due to the absence of neurological evidence. Electroencephalography (EEG) monitoring is an effective method for recording neural activities and can provide electroneurophysiological evidence of MDD.MethodsIn this work, we proposed a probabilistic graphical model for neural dynamics decoding on MDD patients and healthy controls (HC), utilizing the Hidden Markov Model with Multivariate Autoregressive observation (HMM-MAR). We testified the model on the MODMA dataset, which contains resting-state and task-state EEG data from 53 participants, including 24 individuals with MDD and 29 HC.ResultsThe experimental results suggest that the state time courses generated by the proposed model could regress the Patient Health Questionnaire-9 (PHQ-9) score of the participants and reveal differences between the MDD and HC groups. Meanwhile, the Markov property was observed in the neuronal dynamics of participants presented with sad face stimuli. Coherence analysis and power spectrum estimation demonstrate consistent results with the previous studies on MDD.DiscussionIn conclusion, the proposed HMM-MAR model has revealed its potential capability to capture the neuronal dynamics from EEG signals and interpret brain disease pathogenesis from the perspective of state transition. Compared with the previous machine-learning or deep-learning-based studies, which regarded the decoding model as a black box, this work has its superiority in the spatiotemporal pattern interpretability by utilizing the Hidden Markov Model.

Abstract Detecting changes is an important issue for ophthalmology to compare longitudinal fundus images at different stages and obtain change regions. Illumination variations bring distractions on the change regions by the pixel-by-pixel comparison. In this paper, a new unsupervised change detection method based on sparse representation classification (SRC) is proposed for the fundus image pair. First, the local neighborhood patches are extracted from the reference image to build a dictionary of the local background. Then the current image patch is represented sparsely and its background is reconstructed by the obtained dictionary. Finally, change regions are given through background subtracting. The SRC method can correct automatically illumination variations through the representation coefficients and filter local contrast and global intensity effectively. In experiments of this paper, the AUC and mAP values of SRC method are 0.9858 and 0.8647 respectively for the image pair with small lesions; the AUC and mAP values of the fusion method of IRHSF and SRC are 0.9892 and 0.9692 separately for the image pair with the big change region. Experiments show that the proposed method in this paper is more robust than RPCA for the illumination variations and can detect change regions more effectively than pixel-wised image differencing.

Abstract To conquer the worldwide outbreak of COVID-19 virus, a large number of studies have been carried out on COVID-19 infection, transmission and treatment. However, few studies have been conducted from the perspectives of circRNA and lncRNA, which are known to be involved in regulating many life activities, such as immune tolerance and immune escapes, and hence may provide invaluable information in the emerging COVID-19 infection and recurrence. Moreover, exosomes has been reported to play an important role in COVID-19 recurrence, and thus may interact with the expression of circRNA and lncRNA. In this work, we sequenced circRNA, lncRNA and mRNA from recurrent COVID-19 patients and healthy people, and compared the differences. GO and KEGG enrichment analysis show that differentially expressed circRNA and lncRNA are mainly involved in the regulation of host cell cycle, apoptosis, immune inflammation, signaling pathway and other processes. The comparison to exosomes related databases shows that there are 114 differentially expressed circRNA, and 10 differentially expressed lncRNA related to exosomes. These studies provide reference for exploring circRNA and lncRNA to study the infection mechanism of COVID-19, their diagnostic and therapeutic values, as well as the possibility to employ them as biomarkers.

The hedgehog (HH) signaling pathway is one of the key pathways that is indispensable for many developmental processes and postnatal tissue homeostasis. Dysregulated HH signaling could lead to developmental disorders and tumorigenesis in a variety of tissues via inherited or sporadic mutation, gene overexpression, and crosstalk with other signaling pathways. Recently, accumulating evidence has shown that HH signaling is targeted by viruses to facilitate viral transcription, immune evasion, and uncontrolled growth, leading to effective viral replication and pathogenesis. In this study, we will summarize recent advances in functional interaction between HH signaling and different types of viruses, particularly focusing on the pathological role of HH signaling in viral infections and related diseases.

Compared with the traditional fixed relay assisted communication network, the mobile relay assisted wireless communication network has greater communication coverage and better flexibility. It enables to extend the lifetime of a communication system and improve the throughput of uses. Due to the time-varying nature of the actual communication environment and the existence of various interferences, there are uncertain factors in the communication channel that affect the quality of service of users. In this paper, we study the energy efficiency maximization problem based on power control, mobile relay selection, and subcarrier allocation under uncertain channel. The proposed mobile relay selection and subcarrier allocation algorithm improves the energy efficiency of the system while ensuring the normal communication of mobile users. Probabilistic constraints are introduced into the optimization problem to describe the uncertain channel, and after the mathematical transformation, it becomes a solvable form. The subcarrier allocation algorithm and distributed robust power control algorithm are also presented. Simulation results show that the improved system energy efficiency has been achieved and the proposed algorithm behaves better scalability and robustness under dynamic communication environment.

To improve the endurance and charging flexibility of electric vehicle battery packs, this paper proposes a multi-battery block module (MBM) topology for four-phase switched reluctance motors (SRMs), which not only allows flexible electric vehicle operation, but also achieves fast demagnetization and excitation. By integrating the multi-battery block module and photovoltaic (PV) panel into an asymmetrical half-bridge (AHB) converter, the MBM topology is designed to supply a multilevel bus voltage for the SRM drive. To improve the endurance of battery packs, a PV panel is also added to the topology to charge battery packs when the system is stationary. According to the different operation requirements, multiple power supply modes and charging modes can be realized by controlling the power devices in the proposed MBM topology. The simulation results based on the MATLAB/Simulink platform and the experimental results on a four-phase 8 / 6 switched reluctance motor verify the effectiveness of the proposed design.

3C proteases (3Cpros) of picornaviruses and 3C-like proteases (3CLpros) of coronaviruses and caliciviruses represent a group of structurally and functionally related viral proteases that play pleiotropic roles in supporting the viral life cycle and subverting host antiviral responses. The design and screening for 3C/3CLpro inhibitors may contribute to the development broad-spectrum antiviral therapeutics against viral diseases related to these three families. However, current screening strategies cannot simultaneously assess a compound’s cytotoxicity and its impact on enzymatic activity and protease-mediated physiological processes. The viral induction of stress granules (SGs) in host cells acts as an important antiviral stress response by blocking viral translation and stimulating the host immune response. Most of these viruses have evolved 3C/3CLpro-mediated cleavage of SG core protein G3BP1 to counteract SG formation and disrupt the host defense. Yet, there are no SG-based strategies screening for 3C/3CLpro inhibitors. Here, we developed a fluorescence resonance energy transfer (FRET) and SG dual-based system to screen for 3C/3CLpro inhibitors in living cells. We took advantage of FRET to evaluate the protease activity of poliovirus (PV) 3Cpro and live-monitor cellular SG dynamics to cross-verify its effect on the host antiviral response. Our drug screen uncovered a novel role of Telaprevir and Trifluridine as inhibitors of PV 3Cpro. Moreover, Telaprevir and Trifluridine also modulated 3Cpro-mediated physiological processes, including the cleavage of host proteins, inhibition of the innate immune response, and consequent facilitation of viral replication. Taken together, the FRET and SG dual-based system exhibits a promising potential in the screening for inhibitors of viral proteases that cleave G3BP1.

Water repellent treatment is one of the effective means to improve the durability of concrete. This paper aims to investigate the water repellency of cement-based materials treated with silane-based water repellent agent under different exposure environments. Five different exposure conditions were applied in this experiment, namely, standard moist curing room, in-door laboratory atmosphere, pre-oven-dried condition, and out-door natural environment with or without shelter. Three different types of mortars with water-to-cement ratios of 0.4, 0.5 and 0.6 were prepared. And three different dosages (200, 400, 600 g/m2) of silane gel were utilized on the surface of specimens. The impregnation depth and water absorption after hydrophobic treatment have been measured. Results indicate that the initial moisture condition of mortar has a significant influence on the efficiency of surface silane impregnation. The depth of silane impregnation gradually decreases with the increase of initial moisture condition. The water repellency of surface impregnation of cement-based materials was well exhibited under the condition of about 50% relative humidity. It illustrated that the moisture within the cement-based materials affects the hydrolysis reaction process and the formation of hydrophobic membrane. Thus, the water repellency of cement-based materials greatly depends on the initial moisture content and the thickness of hydrophobic layer.

Floral initiation is an important transition point from vegetative growth to reproductive growth in tomatoes and is known to be affected by light intensity, temperature, and nutrients. However, the regulation between flower formation and environmental factors, including nutrient conditions, due to source–sink dynamics (supply and demand of photoassimilates) is seldom documented. To evaluate the effects of light intensity and nutrition conditions on prefloral formation and development, dynamic floral characteristics during development were fitted with sigmoidal logistic curves under four light treatments with shading nets in two nutrient conditions. Source activity and sink strength were altered, which caused differences in the floral positions, length of floral shoots, floral initiation dates, and leaf numbers under the different treatments. Accumulated light acts upstream of nutrition supply during the formation of buds and leads to the accumulation of carbohydrates in source organs. Leaf area reached ≈500 cm2, and dry matter weights reached ≈3 g in each treatment until the flowering day, revealing that some level of photoassimilates are necessary for floral initiation. Both days to flowering and bud number were highly correlated with daily light integral (DLI) from 6 to 12 days before anthesis, which means this period is important for anthesis in tomato. Our results highlight regulation of the transition from vegetative growth to reproductive growth by tomato seedlings due to environmental factors and nutrients. A better understanding of communication between source organs and sink organs during floral initiation response to different environments is expected to provide management strategies for greenhouse tomato production.

The applied load has significantly impacted on the permeability behavior of concrete. Although many investigations have reported the impact of loading level on the permeability, the influence of loading direction on the permeability received little consideration in previous publications. Therefore, this work gives a comprehensive study on the influence of loading level and its direction on water and chloride permeability. Applied loading experiment, water absorption experiment and chloride diffusion test were conducted in this work. The findings showed that the applied load increased the specific crack area and pore size distribution, and that the permeability increased as the applied loading level and duration increased. The water and chloride penetration whose direction is vertical to the applied loading direction is greater than that whose direction parallels the loading direction; in addition, the applied load direction shows larger effects on water ingress than on chloride ingress. Capillary absorption coefficient and chloride diffusion coefficient grow linearly as total porosity increases, and the chloride diffusion coefficient is in direct proportion to the capillary absorption coefficient. When the loading direction is vertical to water penetration, the capillary absorption coefficient of concrete after 75 % fc and 85 %fc increases by 25.76 and 28.27 (g/m2h1/2), compared with the case that the loading direction is parallel with water penetration. This paper further gives a relationship between concrete permeability and the induced loading damage.

Ethylene is not only well known as a unique gaseous hormone, which has various impacts on plant growth, but also considered to be closely related to tomato productivity and occurrence of fruit physiological disorders. On the other hand, the different tomato cultivars were also considered as key factors influencing tomato productivity and quality. In this study, we examined the differences between ethylene emission characteristics of different tomato cultivars. Our results confirmed that the Dutch tomato cultivar ‘Endeavour’ emitted less ethylene compared to other Japanese tomato cultivars, whereas ethylene emission in ‘Rinnka409’ was significantly higher than that in other cultivars. We observed that the wounding treatment resulted in rapid increase in ethylene emission in tomato, and the increase in ethylene emission was much higher in the ‘Momotarou Youku’ cultivar than that in the ‘Endeavour’ cultivar. The results will be useful to maximize tomato productivity and reduce the occurrence of fruit physiological disorders in tomato production at plant factories.

Long non-coding RNAs (lncRNAs) are emerging as crucial regulators of gene expression and physiological processes. LncRNAs are a class of ncRNAs of 200 nucleotides in length. HOX transcript antisense RNA (HOTAIR), a trans-acting lncRNA with regulatory function on transcription, can repress gene expression by recruiting chromatin modifiers. HOTAIR is an oncogenic lncRNA, and numerous studies have determined that HOTAIR is highly upregulated in a wide variety of human cancers. In this review, we briefly summarize the impact of lncRNA HOTAIR expression and functions on different human solid tumors, and emphasize the potential of HOTAIR on tumor prognosis and therapy. Here, we review the recent studies that highlight the prognostic potential of HOTAIR in drug resistance and survival, and the progress of therapies developed to target HOTAIR to date. Furthermore, targeting HOTAIR results in the suppression of HOTAIR expression or function. Thus, HOTAIR knockdown exhibits great therapeutic potential in various cancers, indicating that targeting lncRNA HOTAIR may serve as a promising strategy for cancer therapy. We also propose that preclinical studies involving HOTAIR are required to provide a better understanding of the exact molecular mechanisms underlying the dysregulation of its expression and function in different human cancers and to explore effective methods of targeting HOTAIR and engineering efficient and targeted drug delivery methods in vivo.

Photoplethysmography is a widely used technique to noninvasively assess heart rate, blood pressure, and oxygen saturation. This technique has considerable potential for further applications—for example, in the field of physiological and mental health monitoring. However, advanced applications of photoplethysmography have been hampered by the lack of accurate and reliable methods to analyze the characteristics of the complex nonlinear dynamics of photoplethysmograms. Methods of nonlinear time series analysis may be used to estimate the dynamical characteristics of the photoplethysmogram, but they are highly influenced by the length of the time series, which is often limited in practical photoplethysmography applications. The aim of this study was to evaluate the error in the estimation of the dynamical characteristics of the photoplethysmogram associated with the limited length of the time series. The dynamical properties were evaluated using recurrence quantification analysis, and the estimation error was computed as a function of the length of the time series. Results demonstrated that properties such as determinism and entropy can be estimated with an error lower than 1% even for short photoplethysmogram recordings. Additionally, the lower limit for the time series length to estimate the average prediction time was computed.

Abstract Background Adult T-cell leukemia (ATL) is an aggressive neoplasm caused by human T-cell leukemia virus type 1 (HTLV-1). ATL carries a poor prognosis due to chemotherapy resistance. Thus, it is urgent to develop new treatment strategies. Hypericin (HY) is a new-type of photosensitizer in the context of photodynamic therapy (PDT) due to its excellent photosensitizing properties and anti-tumor activities. Results In the present study, we investigated the efficacy of hypericin in ATL cells. Clinically achievable concentrations of hypericin in association with PDT induced the inhibition of cell proliferation in ATL cell lines with minimal effect on peripheral blood CD4+ T lymphocytes. Moreover, hypericin-PDT treatment caused apoptosis and G2/M phase cell cycle arrest in leukemic cells. Western blot analyses revealed that hypericin-PDT treatment resulted in downregulation of Bcl-2 and enhanced the expression of Bad, cytochrome C, and AIF. Cleavage of caspases-3/-7/-9/-8, Bid, and PARP was increased in hypericin-PDT-treated ATL cells. In a luciferase assay, hypericin-PDT treatment was able to activate the promoter activity of Bax and p53, resulting in enhanced expression of Bax and p53 proteins. Finally, hypericin-PDT treatment suppressed the expression of viral protein HBZ and Tax by blocking the promoter activity via HTLV-1 5′LTR and 3′LTR. Conclusions Our results revealed that hypericin-PDT is highly effective against ATL cells by induction of apoptosis and suppression of viral transcription. These studies highlight the promising use of hypericin-PDT as a targeted therapy for ATL.

This study presents a finite element model to investigate the bidirectional seismic behavior of masonry infill walls. The test data are utilized to verify the numerical model. The comparison between the analytical and the experimental results indicates that the finite element model can successfully predict the failure mode, stiffness, and strength of the masonry infill wall. Based on the model, the effects of aspect ratio (height to length), slenderness ratio (height to thickness), and masonry strength on the out-of-plane (OOP) response of infill wall with in-plane (IP) damage are explored. Considering the aspect ratio, slenderness ratio, and masonry strength of infill wall, the OOP behavior of infill wall with and without IP damage is studied. Finally the reduction of the stiffness and strength in the OOP direction, due to the IP damage, is discussed.

The Hippo signaling pathway is a novel tumor suppressor pathway, initially found in Drosophila. Recent studies have discovered that the Hippo signaling pathway plays a critical role in a wide range of biological processes, including organ size control, cell proliferation, cancer development, and virus-induced diseases. In this review, we summarize the current understanding of the biological feature and pathological role of the Hippo pathway, focusing particularly on current findings in the function of the Hippo pathway in virus infection and pathogenesis.

Machine translation, which will be used widely in human-computer interaction services to Internet of Things (IoT), is a key technology in artificial intelligence field. This paper presents a minimum Bayes-risk (MBR) phrase table pruning method for pivot-based statistical machine translation (SMT). The SMT system requires a great amount of bilingual data to build a high-performance translation model. For some language pairs, such as Chinese-English, massive bilingual data are available on the web. However, for most language pairs, large-scale bilingual data are hard to obtain. Pivot-based SMT is proposed to solve the data scarcity problem: it introduces a pivot language to bridge the source language and the target language. Therefore, a source-target translation model based on well-trained source-pivot and pivot-target translation models can be derived with the pivot-based approach. However, due to the ambiguities of the pivot language, source and target phrases with different meanings may be wrongly matched. Consequently, the derived source-target phrase table may contain incorrect phrase pairs. To alleviate this problem, we apply the MBR method to prune the phrase table. The MBR pruning method removes the phrase pairs with the lowest risk from the phrase table. Experimental results on Europarl data show that the proposed method can both reduce the size of phrase tables and improve the performance of translations. This study also gives a useful reference to many IoT research field and smart web services.

A variety of 7 Tesla RF coil systems have been proposed to produce spin excitation (B1+ field) and MR image acquisition. Different groups have attempted to mitigate the challenges at high and ultra-high field MRI by proposing novel hardware and software solutions to obtain uniformly high spin excitation at acceptable RF absorption levels. In this study, we extensively compare the designs of two distributed-circuit based RF coils: the Tic-Tac-Toe (TTT) head coil and TEM head coil on multiple anatomically detailed head models and in-vivo. Bench measurements of s-parameters and experimental B1+ field distribution were obtained in volunteers and compared with numerical simulations. RF absorption, quantified by both average and peak SAR, and B1+ field intensity and homogeneity, calculated/measured in terms of maximum over minimum and coefficient of variation (CV) in the region of interest (ROI), are presented for both coils. A study of the RF consistency of both coils across multiple head models for different RF excitation strategies is also presented.

Although studies on water-repellent properties of silicon resin (SR) on cement-based materials have received tremendous attention around the world in recent years, they mainly focus on individual properties of water-repellent concrete treated with SR such as strength and water permeability resistance. However, to date, a systematic and comprehensive investigation is missing. The current study therefore investigates the influence of SR on the durability properties of cementitious materials by adding SR emulsion into integral concrete mixture and coating concrete surface with SR. The microscopic and mechanical properties and the durability performance were tested on the integral SR water‐repellent concrete (C-ISR) and SR surface water-repellent concrete (C-SSR). The obtained results indicate that SR application has no impact on hydration reaction and corresponding composition of cement-based materials, while SR addition leads to a reduction in strength and early shrinkage of cement mortar because the added SR delays the hydration reaction. Water-repellent treatment via SR can significantly reduce the water permeability of concrete, which is more apparent for surface water-repellent treatment (SSR). Furthermore, the durability properties were investigated, and the results indicate that integral SR water-repellent treatment (ISR) possesses better properties than SSR, which suggests water-repellent treatment via surface coating as an effective means to improve concrete durability.

Magnetic resonance fingerprinting (MRF) requires a uniform B1+ radiofrequency field. Here the authors present plug-n-play MRF, a technique that enables multiparametric imaging with heterogeneous B1+fields, and demonstrate its utility in the presence of RF distortion caused by a metallic orthopaedic implant.

Irrigation management continues to be an important issue for tomato cultivation, especially in plant factories. Accurate and timely assessment of tomato leaf water status is a key factor in enabling appropriate irrigation, which can save nutrition solution and labor. In recent decades, hyperspectral imaging has been widely used as a nondestructive measurement method in agriculture to obtain plant biological information. The objective of this research was to establish an approach to obtain the tomato leaf water status—specifically, the relative water content (WC) and equivalent water thickness (MC)—for five different tomato cultivars in real time by using hyperspectral imaging. The normalized difference vegetation index (NDVI) and two-band vegetation index (TBI) analyses were performed on the tomato leaf raw relative reflection (RAW), the inversion-logarithm relative reflection (LOG), and the first derivative of relative reflection (DIFF) from wavelengths of 900 nm to 1700 nm. The best regression model for WC assessment was obtained by TBI regression using DIFF at wavelengths of 1410 nm and 1520 nm, and the best regression model for MC assessment was obtained by NDVI regression using RAW at wavelengths of 1300 nm and 1310 nm. Higher model performance was obtained with MC assessment than with WC assessment. The results will help improve our understanding of the relationship between hyperspectral reflectance and leaf water status.

Traditional cement-based repair materials are brittle and prone to cracking. The failure of more than half of repaired concrete structure is due to the re-cracking of the repair material itself or delamination and peeling from the concrete matrix. Thus, a second repair is required in a short period, increasing the maintenance cost. To reduce cracking, Strain Hardening Cement-based Composite (SHCC), with strain hardening and multiple cracking property, is prepared to study the influence of interface roughness and repair layer thickness on the shrinkage, cracking and delamination modes of SHCC-repaired concrete beams. The results show that under the shrinkage stress, multiple fine cracks instead of local fractures occur in the SHCC repair layer, and the interfacial delamination is effectively controlled. Interfacial bonding property is the main factor that affects the shrinkage and deformation coordination of SHCC-repaired beams. When the interface roughness is different, the crack width of the SHCC repair layer is similar. However, it has a greater influence on the interfacial delamination length and maximum delamination height of the repaired beam. With the increase of interface roughness, the delamination length and height of the repaired beam are greatly reduced. Therefore, before using SHCC to repair the existing structures or components, the bonding surface should be roughened to improve the bond strength between SHCC and the old concrete. With the increase of the repair layer thickness, the cracking and delamination of the repair layer tend to be alleviated. Although the crack width of the repair layer can be effectively controlled after cracking, the overlarge shrinkage (985.35 × 10−6, about twice the shrinkage value of ordinary concrete) of the SHCC prepared in this research results in the cracking of the repair layer and the delamination of the repair interface under the restraint of concrete; thus, SHCC fails to repair the concrete efficiently. In terms of shrinkage deformation control, materials with high toughness and low shrinkage are required to repair the existing concrete structures. The implication of this research may provide a theoretical basis for the preparation and application of SHCC with high toughness and low shrinkage.

An Ag/AgCl electrode used as a corrosion sensor in a reinforced concrete structure is considered as having good application prospect. However, its performance under complex conditions, such as dry-wet cycle condition, is not affirmed. In the current study, the performance of Ag/AgCl as chloride selective electrode in mortar exposed to dry-wet cycle condition was investigated. A simple Ag/AgCl electrode was prepared and fabricated by electrochemical anodization. These Ag/AgCl electrodes were embedded into a mortar specimen with temperature sensors, humidity sensors and anode ladder monitoring system (ALS). After 28 d curing time, the upper surface of mortar specimen was wetted (with 5% NaCl solution) and dried regularly. The obtained results indicate that Ag/AgCl electrode responds to the ingress of chloride ion, sensitively. The chloride ion concentration variation can be reflected by the potential trend. Furthermore, the balance potential of Ag/AgCl electrodes is influenced by dry-wet cycles. Compared with ALS, it demonstrates that Ag/AgCl electrodes are more sensitive to chloride. The research provides the key element for the specific application of Ag/AgCl electrode for corrosion monitoring in the future.

Iridium oxide pH electrodes employing the carbonate melt oxidation method were fabricated with oxidation temperatures of 750 °C, 800 °C and 850 °C, respectively. Scanning electron microscope (SEM) and atomic force microscope (AFM) images showed that the oxide film regularized with the increase in oxidation temperatures. The pH response, response time and long-term stability of the electrodes indicated that the electrodes made at 850 °C had the best performance. X-ray photoelectron spectra (XPS) surveys investigated the change in the electrodes’ chemical composition and element oxidation states at 850 °C, and the results showed that the relative content of Ir3+ had increased by 23.9%, and the Ir4+ and Ir6+ had decreased by 10.9% and 13%, respectively, in the surface oxide layer after one month of aging. However, the relative contents of Ir3+, Ir4+ and Ir6+ were almost constant for the inner oxide layer. Meanwhile, the XPS result also indicated that the outer oxide layer of the electrode had a higher hydration degree than the inner oxide layer.

In this study, we examined the influence of moisture content on the electromagnetic response of concrete. A novel homemade electromagnetic monitoring apparatus was developed and used to evaluate the Hall effect voltage at both ends of concrete based on our previous study of the Hall effect. We used four different concrete mix water/binder ratios: 0.30, 0.28, 0.26, and 0.24, and three conditions (relative humidity, carbonation, and water absorption) were examined in this experiment. The results show that the moisture content inside concrete influences the relative permeability of concrete. The variation in the Hall effect voltage is more influenced by carbonation than changes in relative humidity; water absorption increases the Hall effect voltage the least amongst the other examined factors. According to the experiment, a calibration system was established, and the relevant correction factors are provided.

Radio-frequency (RF) field inhomogeneities and higher levels of specific absorption rate (SAR) still present great challenges in ultrahigh-field (UHF) MRI. In this study, an in-depth analysis of the eigenmodes of a 20-channel transmit Tic-Tac-Toe (TTT) RF array for 7T neuro MRI is presented. The eigenmodes were calculated for five different Z levels (along the static magnetic field direction) of the coil. Four eigenmodes were obtained for each Z level (composed of 4 excitation ports), and they were named based on the characteristics of their field distributions: quadrature, opposite-phase, anti-quadrature, and zero-phase. Corresponding finite-difference time-domain (FDTD) simulations were performed and experimental B1+ field maps were acquired using a homogeneous spherical phantom and human head (in-vivo). The quadrature mode is the most efficient and it excites the central brain regions; the opposite-phase mode excites the brain peripheral regions; anti-quadrature mode excites the head periphery; and the zero-phase mode excites cerebellum and temporal lobes. Using this RF array, up to five eigenmodes (from five different Z levels) can be simultaneously excited. The superposition of these modes has the potential to produce homogeneous excitation with full brain coverage and low levels of SAR at 7T MRI.

ObjectiveThe purpose of this study was to explore the efficacy of Ultra-high field [UHF] 7 Tesla [T] MRI as compared to 3T MRI in non-contrast enhanced [nCE] imaging of structural anatomy in the elbow, forearm, and hand [upper extremity].Materials and methodA wide range of sequences including T1 weighted [T1] volumetric interpolate breath-hold exam [VIBE], T2 weighted [T2] double-echo steady state [DESS], susceptibility weighted imaging [SWI], time-of-flight [TOF], diffusion tensor imaging [DTI], and diffusion spectrum imaging [DSI] were optimized and incorporated with a radiofrequency [RF] coil system composed of a transverse electromagnetic [TEM] transmit coil combined with an 8-channel receive-only array for 7T upper extremity [UE] imaging. In addition, Siemens optimized protocol/sequences were used on a 3T scanner and the resulting images from T1 VIBE and T2 DESS were compared to that obtained at 7T qualitatively and quantitatively [SWI was only qualitatively compared]. DSI studio was utilized to identify nerves based on analysis of diffusion weighted derived fractional anisotropy images. Images of forearm vasculature were extracted using a paint grow manual segmentation method based on MIPAV [Medical Image Processing, Analysis, and Visualization].ResultsHigh resolution and high quality signal-to-noise ratio [SNR] and contrast-to-noise ratio [CNR]-images of the hand, forearm, and elbow were acquired with nearly homogeneous 7T excitation. Measured [performed on the T1 VIBE and T2 DESS sequences] SNR and CNR values were almost doubled at 7T vs. 3T. Cartilage, synovial fluid and tendon structures could be seen with higher clarity in the 7T T1 and T2 weighted images. SWI allowed high resolution and better quality imaging of large and medium sized arteries and veins, capillary networks and arteriovenous anastomoses at 7T when compared to 3T. 7T diffusion weighted sequence [not performed at 3T] demonstrates that the forearm nerves are clearly delineated by fiber tractography. The proper digital palmar arteries and superficial palmar arch could also be clearly visualized using TOF nCE 7T MRI.ConclusionUltra-high resolution neurovascular imaging in upper extremities is possible at 7T without use of renal toxic intravenous contrast. 7T MRI can provide superior peripheral nerve [based on fiber anisotropy and diffusion coefficient parameters derived from diffusion tensor/spectrum imaging] and vascular [nCE MRA and vessel segmentation] imaging.

Water repellent treatment has been considered an effective preventive method against water and aggressive ions penetration into concrete and consequently can improve the durability of concrete structures. In reality, many concrete structures are exposed to conditions with high risk of carbonation. In this contribution, one type of ordinary concrete had been prepared and surface impregnated by 400 g/m2 silane cream and 100 g/m2 and 400 g/m2 silane gel. In addition, integral water repellent concrete was produced by adding 2% silane emulsion. Then, the specimens were exposed to accelerated carbonation for 7, 28, and 72 days. The effect of water repellent treatment on carbonation of concrete has been investigated. The results indicate that surface impregnation reduced carbonation depth of concrete under RH 70%, but integral water repellent concrete increased carbonation. Carbonation reaction started behind the hydrophobic layer in the surface-impregnated concrete. The coefficient of carbonation can be described better by a hyperbolic function of time. Treatment by 400 g/m2 silane gel and silane cream showed better efficiency on reducing carbonation than usage of 100 g/m2. Coefficient of water capillary suction was decreased significantly by both surface impregnation and integral water repellent treatment. It is an effective method to protect concrete from water penetration into the material.

In many cases, service life of reinforced concrete structures is severely limited by chloride penetration until the steel reinforcement or by carbonation of the covercrete. Water repellent treatment on the surfaces of cement-based materials has often been considered to protect concrete from these deteriorations. In this paper, three types of water repellent agents have been applied on the surface of concrete specimens. Penetration profiles of silicon resin in treated concrete have been determined by FT-IR spectroscopy. Water capillary suction, chloride penetration, carbonation, and reinforcement corrosion in both surface impregnated and untreated specimens have been measured. Results indicate that surface impregnation reduced the coefficient of capillary suction of concrete substantially. An efficient chloride barrier can be established by deep impregnation. Water repellent surface impregnation by silanes also can make the process of carbonation action slow. In addition, it also has been concluded that surface impregnation can provide effective corrosion protection to reinforcing steel in concrete with migrating chloride. The improvement of durability and extension of service life for reinforced concrete structures, therefore, can be expected through the applications of appropriate water repellent surface impregnation.

Reinforced bar corrosion induced by chloride ingression is one of the most significant threats to the durability of concrete structures in marine environments. The concrete cover thickness, compressive strength, chloride diffusion coefficient, and surface defects of reinforced concrete in the Jiaozhou Bay sea-crossing railway bridge were measured. The temperature and relative humidity in the concrete and the loading applied onto the reinforced concrete were monitored. Based on the DuraCrete model, a revised model for the service life prediction of concrete structures was established, considering the effects of temperature and loading on the chloride diffusion coefficient. Further, the reliability indexes of the reinforced concrete box girder, pier, and platform, located in the marine and land sections, in relation to service lives lasting various numbers of years, were calculated. The measured and calculated results show that the mean cover thicknesses of concrete piers in the marine and land sections are 52 mm and 36 mm, respectively, and the corresponding standard deviations are 5.21 mm and 3.18 mm, respectively. The mean compressive strengths of concrete in the marine and land sections are 56 MPa and 46 MPa, respectively. The corresponding standard deviations are 2.45 MPa and 2.67 MPa, respectively. The reliability indexes of the reinforced concrete box girder and platform in the marine section, under the condition of a service life of 100 years, are 1.81 and 1.76, respectively. When the corrosion-resistant reinforced bar was used in the pier structure in the marine section, its reliability index increased to 2.01. Furthermore, the reliability index of the reinforced concrete damaged by salt fog in the land section was 1.71.

This paper mainly studies the behavior of steel corrosion in various reinforced concrete under freeze-thaw environment. The influence of thickness of concrete cover is also discussed. Additionally, the bond-slip behavior of the reinforced concrete after suffering the freeze-thaw damage and steel corrosion has also be presented. The results show that the freeze-thaw damage aggravates the steel corrosion in concrete, and the results become more obvious in the concrete after suffering serious freeze-thaw damage. Compared with the ordinary concrete, both air entrained concrete and waterproofing concrete possess better resistance to steel corrosion under the same freeze-thaw environment. Moreover, increasing the thicknesses of concrete cover is also an effective method of improving the resistance to steel corrosion. The bond-slip behavior of reinforced concrete with corroded steel decreases with the increase of freeze-thaw damage, especially for the concrete that suffered high freeze-thaw cycles. Moreover, there exists a good correlation between the parameters of bond-slip and freeze-thaw cycles. The steel corrosion and bond-slip behavior of reinforced concrete should be considered serious under freeze-thaw cycles environment, which significantly impact the durability and safety of concrete structure.

Background and objective The brachytherapy with iodine seeds is characterized by radioactive 125I very close to or in contact with the target organ. This study is to evaluate the clinical value of implanted radioactive 125I-seed in the treatment of lung cancer patients. Methods Sixty-three cases of lung cancer were enrolled in this study, and in every case, 125I-seeds were planted into tumor mass guided by CT. Serum CEA, CA125, NSE and CYFRA21-1 weredetected by radioimmunoassay in lung cancer patients before and after planting. The therapeutic effect was evaluated by WHO standards. The relation between the therapeutic effect and the tumor markers levels was observed. Results Just one month after planting, the levels of serum CEA, CA125, NSE and CYFRA21-1 were induced significantly in all cases. There was no significant difference among 1 month, 3 month and 6 month in the levels of tumor markers after therapy. Conclusion 125I-seed could effectively decrease the level of tumor markers in the treatment of lung cancer patients.

Background and Objective. Exosomes secreted from mesenchymal stem cells (MSC) have demonstrated cardioprotective effects. This study examined the role of exosomes derived from MSC overexpressing CXCR4 for recovery of cardiac functions after myocardial infarction (MI). Methods. In vitro, exosomes from MSC transduced with lentiviral CXCR4 (ExoCR4) encoding a silencing sequence or null vector were isolated and characterized by transmission electron microscopy and dynamic light scattering. Gene expression was then analyzed by qPCR and Western blotting. Cytoprotective effects on cardiomyocytes were evaluated and effects of exosomes on angiogenesis analyzed. In vivo, an exosome-pretreated MSC-sheet was implanted into a region of scarred myocardium in a rat MI model. Angiogenesis, infarct size, and cardiac functions were then analyzed. Results. In vitro, ExoCR4 significantly upregulated IGF-1α and pAkt levels and downregulated active caspase 3 level in cardiomyocytes. ExoCR4 also enhanced VEGF expression and vessel formation. However, effects of ExoCR4 were abolished by an Akt inhibitor or CXCR4 knockdown. In vivo, ExoCR4 treated MSC-sheet implantation promoted cardiac functional restoration by increasing angiogenesis, reducing infarct size, and improving cardiac remodeling. Conclusions. This study reveals a novel role of exosomes derived from MSCCR4 and highlights a new mechanism of intercellular mediation of stem cells for MI treatment.

GbaSM-4 cells, vascular smooth muscle cells (VSMCs) derived from brain basilar arteries, were shown to migrate toward d-nicotine by augmenting the actin cytoskeleton in their cell bodies and lamellipodia, and expression of nicotinic acetylcholine receptor (α7-nAChR) was detected in GbaSM-4 cells. Their chemotaxis was antagonized by an α7-nAChR antagonist of methyllycaconitine. It was also antagonized by inhibiting myosin light chain (MLC) kinase and by down-regulating MLC kinase. However, the changes in MLC phosphorylation were not associated with the nicotine treatment, suggesting the involvement of non-kinase activity of MLC kinase as reviewed by Gao et al. (IUBMB Life. 2001;51:337). This plot may work to induce arteriosclerosis during cigarette smoking. Triple-negative breast cancers are associated with poor clinical outcomes and new therapeutic strategies are clearly needed. Gallotannin (Gltn) has been previously demonstrated to have potent anti-tumor properties against cholangiocarcinoma in mice, but little is known regarding its capacity to suppress tumor outgrowth in breast cancer models. We tested Gltn for potential growth inhibitory properties against a variety of breast cancer cell lines in vitro. In particular, triple-negative breast cancer cells display higher levels of sensitivity to Gltn. The loss of proliferative capacity in Gltn exposed cells is associated with slowed cell cycle progression and S phase arrest, dependent on Chk2 phosphorylation and further characterized by changes to proliferation related genes, such as cyclin D1 (CcnD1) as determined by Nanostring technology. Importantly, Gltn administered orally or via intraperitoneal (IP) injections greatly reduced tumor outgrowth of triple-negative breast cells from mammary fat pads without signs of toxicity. In conclusion, these data strongly suggest that Gltn represents a novel approach to treat triple-negative breast carcinomas.

Human T-cell leukemia virus type 1 (HTLV-1) causes adult T-cell leukemia (ATL) and chronic inflammatory diseases. HTLV-1 bZIP factor (HBZ) is transcribed as an antisense transcript of the HTLV-1 provirus. Among the HTLV-1-encoded viral genes, HBZ is the only gene that is constitutively expressed in all ATL cases. Recent studies have demonstrated that HBZ plays an essential role in oncogenesis by regulating viral transcription and modulating multiple host factors, as well as cellular signaling pathways, that contribute to the development and continued growth of cancer. In this article, I summarize the current knowledge of the oncogenic function of HBZ in cell proliferation, apoptosis, T-cell differentiation, immune escape, and HTLV-1 pathogenesis.

BACKGROUND:The prognostic significance of survivin for survival of patients with esophageal squamous cell carcinoma (ESCC) remains controversial. Thus, meta-analysis of the literatures was performed in order to demonstrate its expression impact on ESCC clinicopathological features and prognosis. METHODOLOGY:Relevant literatures were searched using PubMed, EMBASE and Medline Databases. Revman5.0 software was used to pool eligible studies and summary hazard ratio (HR). Correlation between survivin expression and clinicopathological features of ESCC was analyzed. PRINCIPAL FINDINGS:Final analysis of 523 patients from 7 eligible studies was performed. Human T-cell leukemia virus type 1 (HTLV-1) is the causal agent of a neoplastic disease of CD4+ T cells, adult T-cell leukemia (ATL), and inflammatory diseases including HTLV-1 associated myelopathy/tropical spastic paraparesis, dermatitis, and inflammatory lung diseases. ATL cells, which constitutively express CD25, resemble CD25+CD4+ regulatory T cells (T(reg)). Approximately 60% of ATL cases indeed harbor leukemic cells that express FoxP3, a key transcription factor for T(reg) cells. HTLV-1 encodes an antisense transcript, HTLV-1 bZIP factor (HBZ), which is expressed in all ATL cases. In this study, we show that transgenic expression of HBZ in CD4+ T cells induced T-cell lymphomas and systemic inflammation in mice, resembling diseases observed in HTLV-1 infected individuals. In HBZ-transgenic mice, CD4+Foxp3+ T(reg) cells and effector/memory CD4+ T cells increased in vivo. As a mechanism of increased T(reg) cells, HBZ expression directly induced Foxp3 gene transcription in T cells. The increased CD4+Foxp3+ T(reg) cells in HBZ transgenic mice were functionally impaired while their proliferation was enhanced. HBZ could physically interact with Foxp3 and NFAT, thereby impairing the suppressive function of T(reg) cells. Thus, the expression of HBZ in CD4+ T cells is a key mechanism of HTLV-1-induced neoplastic and inflammatory diseases.

The primary role of the innate immune response is to limit the spread of infectious pathogens, with activation of Toll-like receptor (TLR) and RIG-like receptor (RLR) pathways resulting in a pro-inflammatory response required to combat infection. Limiting the activation of these signaling pathways is likewise essential to prevent tissue injury in the host. Triad3A is an E3 ubiquitin ligase that interacts with several components of TLR signaling and modulates TLR activity. In the present study, we demonstrate that Triad3A negatively regulates the RIG-I RNA sensing pathway through Lys48-linked, ubiquitin-mediated degradation of the tumor necrosis factor receptor-associated factor 3 (TRAF3) adapter. Triad3A was induced following dsRNA exposure or virus infection and decreased TRAF3 levels in a dose-dependent manner; moreover, Triad3A expression blocked IRF-3 activation by Ser-396 phosphorylation and inhibited the expression of type 1 interferon and antiviral genes. Lys48-linked ubiquitination of TRAF3 by Triad3A increased TRAF3 turnover, whereas reduction of Triad3A expression by stable shRNA expression correlated with an increase in TRAF3 protein expression and enhancement of the antiviral response following VSV or Sendai virus infection. Triad3A and TRAF3 physically interacted together, and TRAF3 residues Y440 and Q442--previously shown to be important for association with the MAVS adapter--were also critical for Triad3A. Point mutation of the TRAF-Interacting-Motif (TIM) of Triad3A abrogated its ability to interact with TRAF3 and modulate RIG-I signaling. TRAF3 appears to undergo sequential ubiquitin "immuno-editing" following virus infection that is crucial for regulation of RIG-I-dependent signaling to the antiviral response. Thus, Triad3A represents a versatile E3 ubiquitin ligase that negatively regulates RIG-like receptor signaling by targeting TRAF3 for degradation following RNA virus infection.

BACKGROUND: Interferon regulatory factor (IRF) family members have been implicated as critical transcription factors that function in immune response, hematopoietic differentiation and cell growth regulation. Activation of IRF-5 results in the production of pro-inflammatory cytokines such as TNFalpha, IL6 and IL12p40, as well as type I interferons. METHODOLOGY/PRINCIPAL FINDINGS: In this study, we identify a G202C (position relative to translation start codon) missense-mutation transcript of IRF-5 in transformed B and T cell lines, which were either infected or non-infected by viruses, and peripheral blood from ATL or CLL patients. The mutated transcript encodes a novel protein in which the sixty-eighth amino acid, Alanine, is substituted by Proline (IRF-5P68) in the DNA binding domain of IRF-5. IRF-5P68 phenotype results in a complete loss of its DNA-binding activity and functions as a dominant negative molecule through interacting with wild type IRF-5. Co-expression of IRF-5P68 inhibits MyD88-mediated IRF-5 transactivation. Moreover, Toll-like receptor (TLR)-dependent IL6 and IL12P40 production induced by lipopolysaccharide (LPS), R837 or CpG ODN 1826 was reduced in IRF-5 (P68) expressing cells as compared to the control cells. CONCLUSION: IRF-5P68 acts as a dominant negative regulator that interferes with IRF-5-mediated production of pro-inflammatory cytokines. The functional characterization of the novel IRF-5 mutant in transformed B and T cell lines and in ATL and CLL patients may lead to a better understanding of the role of these transcriptional regulators in hematopoietic malignancies.
