Abstract

The concept of space agriculture is developed to support manned exploration on a distant planet Mars. Three core items of our innovative life support system are hyper-thermophilic aerobic composting bacterial ecology, cultivation of trees for producing excess oxygen, and entomophagy. This proposal is well matured based on our cultural and historical background. Candidate insect species for supplying animal proteins are discussed together with tree species for those insects. 第25回宇宙利用シンポジウム(2009年1月14日-15日, 宇宙航空研究開発機構宇宙科学研究本部相模原キャンパス), The Twenty-fifth Space Utilization Symposium (January 14-15, 2009: ISAS/JAXA Sagamihara, Japan), Engineering target of space agriculture is to create bio-regenerative life support system that enables to send human to distant Mars for astrobiological exploration. Even manned space activities are driven by national prestige, manned Mars exploration might be realized under multinational cooperation and coordination. In such project, commonly shared objective should be defined clearly. Participant member should be complimentary each others in their contribution. They should be equal partner, and necessary at providing their excellence to compose the whole system. Space agriculture is a good subject of space engineering with multi-partner contribution. We are living in the world of diverse culture of agriculture and foods. Implementing good components from diverse background might enforce the robustness of space agriculture and make it resistive against catastrophic disturbance. We have raised component items for space agriculture concept, which is based on historical and cultural background of Japan and east Asia. Even countries or area where their own space activities is not yet flourished, they might have excellent items to contribute to the space exploration. Abstract

Purpose: This study was conducted to investigate the effect of flavonoids, a major family of antioxidants contained in foods, on retinal ganglion cell (RGC) death induced by hypoxia, excessive glutamate levels, and oxidative stress. Moreover, to assess the structure-activity relationships of flavonoids, three types of flavonoids with different numbers of hydroxyl groups and varieties of sugar chains were studied., Methods: Three kinds of flavonoids-nicotiflorin, rutin, and quercitrin-were used. The death of neonatal rat purified RGCs was induced by hypoxic conditions (5% O(2), 5% CO(2), 37 °C) for 12 h, 25 µM glutamate over three days, or oxidative stress by depleting antioxidants from the medium for 24 h. RGC survival rates were calculated under each condition and compared with vehicle cultures. Modification of cell death signaling after stress-induced apoptosis and necrosis by flavonoids was assessed using caspase-3 and calpain immunoreactivity assays., Results: Under hypoxic and glutamate stress, both nicotiflorin and rutin significantly increased the RGC survival rate at 1 nM or higher, while quercitrin increased it at 100 nM or higher. Under oxidative stress, nicotiflorin, rutin, and quercitrin also significantly increased the RGC survival rate at 1 nM, 0.1 nM, and 100 nM or higher, respectively. Rutin significantly inhibited the induction of caspase-3 under both hypoxia and excessive glutamate stress, as well as blocking the induction of calpain during oxidative stress., Conclusions: Nicotiflorin and rutin showed neuroprotective effects on hypoxia-, glutamate- or oxidative stress-induced RGC death at concentrations of 1 nM or higher. The presence of a specific sugar side chain (rutinoside) may enhance neuroprotective activity.
