4. The contribution of the Fc RIIB immunoreceptor tyrosine-based inhibitory motif (ITIM) to the modulation of antibody immunotherapy

Abstract

Monoclonal antibodies (mAb) are an increasingly important therapeutic tool in the treatment of cancers and autoimmunity. Many mAbs elicit their efficacy through engagement of activatory Fc gamma receptors (Fc R), resulting in target cell depletion by immune effector cells. These activatory Fc Rs are negatively influenced by the single inhibitory Fc R, Fc RIIB. Fc RIIB has been shown to reduce the efficacy of mAbs through impairment of activatory Fc R function and upregulation of Fc RIIB has been identified as a resistance mechanism of mAb therapeutics in cancer. Fc RIIB contains an intracellular immunoreceptor-tyrosine-based inhibition motif (ITIM), which delivers inhibitory signalling. Whether the ITIM is required for all Fc RIIB-mediated inhibitory activities of therapeutic mAbs is not yet clear. To address this issue, we developed and characterised a novel transgenic mouse model (NoTIM), in which the endogenous inhibitory mouse (m)Fc RII is replaced with a non-signalling ITIM mutant of human (h)Fc RIIB. Cells from mice expressing the NoTIM receptor were no longer able to elicit phosphorylation of the Fc RIIB ITIM, mediate efficient internalisation of immune complexes or prevent anti-IgM-mediated calcium flux, confirming the expected phenotype. To understand how Fc RIIB inhibitory signalling impacted mAb-mediated depletion of target cells, NoTIM mice were treated with anti-mouse CD20 mAbs. The extent of depletion was compared to mice either lacking mFc RII or where mFc RII was replaced with a signalling competent hFc RIIB (hFc RIIB Tg). B cell depletion was reduced in both the NoTIM and hFc RIIB Tg mice compared to those lacking mFc RII. Subsequent experiments revealed this was not due to accelerated mAb internalisation nor enhanced clearance from the serum. Using a series of adoptive transfer models we determined that the NoTIM hFc RIIB was mediating inhibition, by competing with activatory Fc Rs on the surface of myeloid cells for the Fc of the direct targeting mAb. These findings were then assessed with respect to T regulatory cell and malignant murine B cell depletion. It was found that in both cases, depletion was negatively impacted by the expression of Fc RIIB at themyeloid surface rather than by its ability to signal. These data indicate that signalling through the ITIM is not critical for the ability of hFc RIIB to prevent mAb-mediated depletion of target cells, questioning current paradigms for its mechanism of action.

10. Evaluating the multiple sulfur isotope signature of Eoarchean rocks from the Isua Supracrustal Belt (Southwest-Greenland) by MC-ICP-MS:Volcanic nutrient sources for early life

Abstract

On the anoxic Archean Earth, prior to the onset of oxidative weathering, electron acceptors were relatively scarce, perhaps limiting microbial productivity. An important metabolite may have been sulfate produced during the photolysis of volcanogenic SO2 gas. Multiple sulfur isotope data can be used to track this sulfur source, and indeed this record indicates SO2 photolysis dating back to at least 3.7 Ga, that is, as far back as proposed evidence of life on Earth. However, measurements of multiple sulfur isotopes in some key strata from that time can be challenging due to low sulfur concentrations. Some studies have overcome this challenge with NanoSIMS or optimized gas-source mass spectrometry techniques, but those instruments are not readily accessible. Here, we applied an aqua regia leaching protocol to extract small amounts of sulfur from whole rocks for analyses of multiple sulfur isotopes by multi-collector inductively coupled plasma mass spectrometry (MC-ICP-MS). Measurements of standards and replicates demonstrate good precision and accuracy. We applied this technique to meta-sedimentary rocks with putative biosignatures from the Eoarchean Isua Supracrustal Belt (ISB, >3.7 Ga) and found positive ∆33S (1.40–1.80‰) in four meta-turbidites and negative ∆33S (−0.80‰ and −0.66‰) in two meta-carbonates. Two meta-basalts do not display significant mass-independent fractionation (MIF, −0.01‰ and 0.16‰). In situ Re–Os dating on a molybdenite vein hosted in the meta-turbidites identifies an early ca. 3.7 Ga hydrothermal phase, and in situ Rb–Sr dating of micas in the meta-carbonates suggests metamorphism affected the rocks at ca. 2.2 and 1.7 Ga. We discuss alteration mechanisms and conclude that there is most likely a primary MIF-bearing phase in these meta-sediments. Abstract

This research project is a profile of inverted social deviance that speaks to base issues of morality and justice in the organisation of economic life. Focusing on the City of London, the aim of the research project is to create a sociological snapshot of the organising interests and ideologies functioning within spaces and institutions of global finance. Engaging in the everyday routines, expectations assumptions of economic actors embedded within the financial services industry, this study contributes towards an enhanced understanding of how the cultural sensibilities that underpin a market based reality perpetuate gross social and economic inequalities through the shifting of negative market costs on vulnerable sections of society. Leading to a critical evaluation of the organising ideologies and taken-for-granted assumptions that function within the City of London, the principal findings detail the relational struggles, strategies and vested interests that dominate the field of finance life.

Abstract

This thesis describes novel developments in the fabrication and understanding of type IA fibre Bragg gratings, the uses of said gratings as optical sensors and the interrogation of optical sensors using tilted fibre Bragg gratings. This thesis presents the most detailed study of type IA gratings performed to date and provides the basis of a dual grating optical sensor capable of independently measuring strain and temperature. Until this work it was not known how to reliably fabricate type IA gratings or how they would react to high ambient temperatures, nor was it known what effect external parameters such as fibre type, dopant levels, inscription laser intensity, or hydrogenation levels would have on the physical properties of the grating. This comprehensive study has yielded answers to all of these unknowns and produced several unexpected uses for type IA gratings, such as the use of the previously unreported strong loss band at 1400nm to locally heat fibres by optical absorption and thereby fabricate optically tuneable gratings which do not affect directly adjacent standard gratings. Blazed fibre Bragg gratings have been studied in detail and used to produce several high quality prototype sensor interrogation systems yielding stability an accuracy values unsurpassed by similar devices reported in literature. An accurate distribution map of light radiated by blazed gratings is shown for the first time and has been studied in respect of polarisation state showing that for certain easily achievable conditions a blazed grating spectrometer may be deemed to be polarisation insensitive. In a novel implementation of the system, it is shown that the dynamic wavelength range of a blazed grating spectrometer may be at least doubled by superimposing blazed gratings.

23. A Comprehensive Framework to Advance Equity, Diversity, and Inclusion in a Forensic Service.

Abstract

Minority and Indigenous populations have disproportionate representation within forensic mental health services. Social determinants of health and systemic discrimination have contributed to the difficulties these populations have in accessing care, as well as significant differences in care trajectories. In addition, staffing and structural equity, diversity, and inclusion (EDI) challenges permeate forensic systems as in other health care settings. There is little literature to guide forensic mental health services in how best to provide equitable, diverse, and inclusive practices for patients, families, and staff. The forensic service at a major urban center in the Canadian province of Ontario has adapted an EDI framework to describe the processes employed to organize and integrate EDI principles and initiatives within a culture of learning and continuous improvement. This Forensic EDI Framework is composed of six domains: Organizational Commitment, Staff/Workforce Competencies, Service Access and Delivery, Promoting Responsiveness, Community Outreach, and Data Collection. Initiatives within each of these domains form the foundation of a sustainable platform for forensic service EDI practices that will promote lasting change. [ABSTRACT FROM AUTHOR]

30. A qualitative exploration of the mental health challenges and therapeutic needs of Canadian correctional workers

Abstract

PurposeCorrectional work is described as a high-stress environment associated with increased prevalence of mental health disorders in employees. Identifying appropriate healthcare services necessitates investigating the mental health challenges and needs of correctional workers (CWs).MethodsIndividual interviews (n = 9; 5 M and 4 W) and a mixed gender focus group (n = 6; 3 M and 3 W) were conducted to gather a general sense of the mental health landscape. Data were analyzed to develop a targeted and comprehensive question guide for gender-specific focus groups (n = 14 unique participants; 6 M and 8 W).ResultsEight themes emerged from the gender-specific focus groups. Themes focusing on work culture described the negative repercussions of job stress and the inability to discuss challenges openly due to confidentiality concerns and feelings of seclusion associated with the CW profession. Men were more likely to be subjected to physical violence and women to emotional and sexual harassment from staff and inmates. Themes related to mental health care described the benefits and limitations of the current services and digital mental healthcare. Stigma and accessibility were notable treatment barriers. Lastly, sector-specific therapy was seen as an important component in enhancing engagement and therapist interaction.ConclusionThe study demonstrates the interconnection between work culture and CW mental health that needs to be acknowledged when addressing mental health care.

Abstract

Objective: Little is known about the health care costs of individuals with chronic psychotic disorders who experience incarceration. This study sought to address this knowledge gap. Methods: The authors analyzed linked 2007?2010 correctional and administrative health care data on sex- and age-matched individuals with chronic psychotic disorders with and without known incarceration in prison for up to 2 years in the Ontario correctional system. Mean 1-year health care costs (overall and by sex) in the year before incarceration (when release occurred in 2010) were estimated from third-party payer data and compared between the two groups. Costs were calculated in 2018 Canadian dollars. Results: Individuals who experienced incarceration (N=3,197) had mean 1-year costs of $15,728 in the year before incarceration, whereas those who did not (N=6,393) had 1-year costs of $11,588. This difference was mostly due to costs arising from psychiatric hospitalizations, emergency department visits, and physician services. Abstract

U-Pb dating of calcite veins allows direct dating of brittle deformation events. Here, we apply this method to hydrothermal calcite veins in a fold-and-thrust belt and a large scale strike-slip fault zone in central and western Thailand, in an attempt to shed new light on the regional upper crustal deformation history. Calcite U-Pb dates for the Khao Khwang Fold and Thrust Belt (KKFTB) of 221 ± 7 Ma and 216 ± 3 Ma demonstrate that calcite precipitated during tectonic activity associated with stage II of the Indosinian Orogeny (Late Triassic – Early Jurassic). One additional sample from the KKFTB suggests that the Indosinian calcite has locally been overprinted by a Cenozoic fluid event with a different chemistry. For the Three Pagodas Fault Zone (TPFZ), our calcite U-Pb results suggest a complex, protracted history of Cenozoic brittle deformation. Petrographic information combined with contrasting redox-sensitive trace elemental signatures suggest that the vein arrays in the TPFZ precipitated during two distinct events of brittle deformation at ~48 and ~23 Ma. These dates are interpreted in the context of far-field brittle deformation related to the India-Eurasia collision. The presented calcite U-Pb dates are in excellent agreement with published age constraints on the deformation history of Thailand, demonstrating the utility of the method to decipher complex brittle deformation histories. The paper further illustrates some of the complexities in relation to calcite U-Pb dating and provides suggestions for untangling complex datasets that could be applied to future studies on the deformation history of Thailand and other regions.

Abstract

The Nevada Seismological Laboratory has posted a public database of Refraction Microtremor (ReMi) survey results of shear-wave velocity (Vs) versus depth in the Reno-area basin, and additional locations. Most of the Vs profiles were published previously in peer-reviewed journal articles. The database collects values of Vs30, Z1.0, and Z2.5 measured at 170 sites throughout the basin. Vs30 is the time-averaged Vs between the surface and 30 m depth. Z1.0 is the depth to the first occurrence of Vs = 1.0 km/s or greater, and Z2.5 is the depth to Vs = 2.5 km/s. The database contains many more Reno-area sites with Vs30 measurements that did not also have a Z1.0 measurement; this report does not examine sites without Z1.0 values. All but a few sites have Vs30 between 260 m/s and 760 m/s, with a majority in NEHRP hazard class C. Sites that are geologically on bedrock have unexpectedly low Vs30, 0.1 km, and some basin sites have Z1.0 Data File Format Assembled measurements of Vs30, Z1.0, and Z2.5 in Reno are given in Table 1. Predicted Z1.0 and Z2.5 results are contained within three plain text data files in the .zip file accompanying this report. Within each text file, Z1.0 and Z2.5 have been estimated for each location listed in the corresponding gravity data set of Zg values: Abbott and Louie (2000) for Reno; Langenheim et al. (1998, 2001) for Las Vegas; and Saltus and Jachens (1995) for the entire Basin and Range. Each location’s estimate is one row in the text file. The columns of the text files, from left to right, are: latitude, degrees WGS84; longitude, degrees WGS84; Zg, km; Z1.0min, km; Z1.0, km; Z1.0max, km; Z2.5min, km; Z2.5, km; Z2.5max, km; Z1.0error, percent; and Z2.5error, percent. The latitude and longitude are converted directly from the published gravity databases, and the Zg values are taken directly from their published results. The Z values are determined as described above from the best-fit coefficients in Equations 1 and 2, estimated from the Zg values. Z values are not forced to any nearby measured Z values that appear in Table 1. The minimum Z values result from the application of the 95% confidence minimum coefficients in Equations 1 and 2; the maximum Z values result from the application of the 95% confidence maximum coefficients in Equations 1 and 2. The Z errors are the percentages resulting from subtracting the minimum Z from the best-fit Z, and then dividing by the best-fit Z. Users should bear in mind that the Z1.0 estimates are not thought to be valid outside the Reno-area basin. All Z1.0 predictions in the Langenheim and Saltus files, outside the Reno area, should be ignored. Geologically, Z2.5 is thought to coincide with the depth to the floor of Tertiary and Quaternary basins filled with either clastic or volcanic sediments having slower velocity and lesser density, atop denser and faster Mesozoic basement. Figures 12 through 15 present maps of Z1.0 and Z2.5 for Reno, and of Z2.5 for Las Vegas and all of Nevada and Eastern California, respectively. The accompanying .zip file has the map images, as well as a KML file allowing Google Earth to show the Z maps.
