Fast and accurate identification of mine water source is the key condition to prevent and control mine water damage. This study takes Mindong No.1 Mine in grassland mining area of east China as the research area, collects a variety of mine water source samples, and analyzes the hydro-chemical characteristics of different types of water samples by drawing Piper three-line diagram and Gibbs diagram and ionic proportional coefficients diagram, and qualitatively identifies several types of mine water source. In addition, the proportion of mine water source is quantitatively analyzed by the calculation of hydrogen and oxygen isotopes. The hydro-chemical type of mine indirect water sources (atmospheric precipitation, surface water, quaternary diving and aquifer water I) is HCO3-Ca, and the hydro-chemical type of aquifer water II is HCO3-Ca mixed with HCO3-Na. The hydro-chemical types of aquifer III water and mine gushing water are both HCO3-Na type, and aquifer III water is the main recharge source of mine gushing water. The formation of Ca2+, Na+ and HCO3− in groundwater and mine water is controlled by rock weathering, and mainly comes from the dissolution of silicate minerals such as potassium feldspar, albite and calcium feldspar. Combined with the calculation of isotope tritium content, the relative average age of water in the Quaternary, I aquifer, II aquifer and III aquifer is 18.48, 57.88, 69.24, 69.24 years, respectively. The average age of mine water gushing is 69.24 years, and the average values of δD and δ18O of mine water are closest to aquifer II and aquifer III water. Mine water gushing is mainly a mixture of aquifer II and III water, accounting for 11.1% and 89.9% of the total mine water gushing, respectively. The mixing ratio is about 1∶9.

Summary: Background: In ORIENT-15 study, sintilimab plus chemotherapy demonstrated significant improvement on overall survival (OS) versus placebo plus chemotherapy in first-line treatment of advanced esophageal squamous cell carcinoma (ESCC). Here, we report effect of sintilimab plus chemotherapy on health-related quality of life (HRQoL) in patients with advanced ESCC. Methods: From December 14, 2018 to August 28, 2022, HRQoL was evaluated in all randomized patients using European Organization for Research and Treatment of Cancer (EORTC) Quality of Life Questionnaire Core 30 items (QLQ-C30), EORTC Quality of Life Questionnaire Oesophageal Cancer Module 18 items (QLQ-OES18), and visual analogue scale (VAS) of the EuroQol five-dimensional five-level questionnaire (EQ-5D-5L). Mean scores of each scale were described by treatment group through week 60. Least-squares mean (LSM) score change from baseline through week 24 were analyzed using the mixed-model repeated-measures method. Time to the first onset of deterioration (TTD) and OS for each scale were estimated. Clinical Trials Registration: NCT03748134. Findings: As of August 28, 2022, 689 of 690 enrolled patients were assessed for HRQoL analysis (sintilimab group: 340, placebo group: 349). Median follow-up was 32.2 months. Differences in LSM favored sintilimab over placebo for QLQ-C30 social functioning (LSM difference: 3.06, 95% CI: 0.55 to 5.57; P = 0.0170), pain (−2.24, 95% CI: −4.30 to −0.17; P = 0.0337), fatigue (−2.24, 95% CI: −4.46 to −0.02; P = 0.0479), constipation (−3.27, 95% CI −5.49 to −1.05; P = 0.0039), QLQ-OES18 pain (−1.77, 95% CI −3.11 to −0.43; P = 0.0097), trouble swallowing saliva (−2.09, 95% CI: −3.77 to −0.42; P = 0.0146), and choked when swallowing (−3.23, 95% CI: −5.60 to −0.86; P = 0.0076). Variations in vegetation composition and structure are significant signals of the succession of mudflat ecosystems and have important indicative roles in mudflat ecosystem degradation. Due to poor accessibility of vast even mudflat areas, variation in vegetation composition and structure across mudflat areas remains unclear in the Yellow River Delta (YRD), China. We provided an UAV multispectral orthomosaic with 10 cm ground sample distance to classify and compare the vegetation composition and structure across mudflat areas in the YRD. The vegetation classification overall accuracy achieved 95.0%. We found that although a significant difference (p < 0.05) was checked out in terms of the Shannon–Wiener diversity index (from 1.33 to 0.92) and evenness index (from 0.96 to 0.66) among the eight subareas from land to sea, all four dominant vegetation communities (S. salsa, L. bicolor, T. chinensis, and P. australis) were discovered at all eight subareas. Our findings support the idea that the regional environment and local microtopography are the predominant forces for variation in vegetation composition and structure across mudflat areas. From the perspective of vegetation restoration and conservation, changing the local microtopography will be an interesting way to enhance the vegetation diversity of the mudflat ecosystems in the YRD.

In order to solve the problem of difficult solid-liquid separation in the treatment of mine water containing suspended solids, based on the theory of nuclear crystal coagulation induced granulation, the process of “micro flocculation multi stage filtration” of mine water is studied through laboratory experiments. First of all, through the adjustment of reagent dosage and water conservancy conditions, the suspended particles and flocculants are induced to form a copolymer to form a floc settling time curve, and the floc granulation and densification are completed through the selection of optimal parameters. Secondly, four filter media of quartz sand, artificial zeolite, active carbon and aeolian sand are selected to investigate the effects of different media types and particle sizes on the removal efficiency of pollutants, and the physical and chemical reactions generated at the solid-liquid interface are analyzed. Finally, field tests are carried out in typical mines to verify the removal effect of the process on different pollution components. The results show that (1) the sedimentation time can be greatly reduced by adding polyaluminum chloride of 200 mg/L and diatomite of 200 mg/L in the micro flocculation stage. (2) The turbidity removal rate of 1−2 mm quartz sand in the multi-stage filtration stage can reach 97.9%, while the artificial zeolite in the four filter media is the best for the removal of inorganic ions. The removal rates of \begin{document}${\rm{SO}}_4^{2 -} $\end{document}, \begin{document}${\rm{HCO}}_3^- $\end{document} and Na+ in the filtered water can reach 25.5%, 44.1% and 69.9%, respectively. The micro flocculation multi-stage filtration process formed through the research results can greatly reduce the residence time in the process of mine water treatment and improve the solid-liquid separation efficiency. On this basis, the selection of natural diatomite as nucleating agent instead of polyacrylamide can effectively inhibit the generation of acrylamide monomer and reduce environmental risks. In addition, the process has a certain removal effect on inorganic ions. It is good application value for the treatment of mine water containing suspended solids.

Abstract UBR5, a HECT-domain E3 ubiquitin ligase, is an attractive therapeutic target for aggressive breast cancers. Defining the substrates of UBR5 is crucial for scientific understanding and clinical intervention. Here, we demonstrate that CDC73, a component of the RNA polymerase II-associated factor 1 complex, is a key substrate that impedes UBR5’s profound tumorigenic and metastatic activities in triple-negative breast cancer (TNBC) via mechanisms of regulating the expression of β-catenin and E-cadherin, tumor cell apoptosis and CD8+ T cell infiltration. Expression of CDC73 is also negatively associated with the progression of breast cancer patients. Moreover, we show that UBR5 destabilizes CDC73 by polyubiquitination at Lys243, Lys247, and Lys257 in a non-canonical manner that is dependent on the non-phosphorylation state of CDC73 at Ser465. CDC73 could serve as a molecular switch to modulate UBR5’s pro-tumor activities and may provide a potential approach to developing breast cancer therapeutic interventions.

Persimmon peel pectin (PPP) was extracted by subcritical water via the response surface methodology. The optimal crude PPP extraction yield of 7.62 ± 0.7 % was found at 138 °C, 2.84 min, and liquid–solid ratio of 1:10.02. After treatment of deproteinization and decolorization with papain and hydrogen peroxide, 83.19 % of protein and 78.56 % of the colour in crude PPP were removed, respectively. PPP owned the Mw of 21.79 kDa and its uronic acids content was 64.03 %. PPP was further affirmed by fourier transform infrared, X-ray diffractometer and 1H NMR analysis. Moreover, the degradation temperature (228.05 ℃) of PPP was verified via differential scanning calorimetry. Then, the IC50 of PPP to ABTS·+ was 9.8 times that of commercial citrus pectin. Moreover, PPP could change microbial communities and selectively enrich Bacteroides, Cetobacterium, Erysipelatoclostridium, Parabacteroides and Phocaeicola sartorii. This study demonstrated that subcritical water was practicable for extraction of persimmon peel pectin.

Mariculture is an important part of aquaculture, and it is important to address global food security and nutrition issues. However, seawater environmental conditions are complex and variable, which causes large uncertainties in the remote sensing spectral features. At the same time, mariculture types are distinct because of the different types of aquaculture (cage aquaculture and raft aquaculture). These factors bring great challenges for mariculture extraction and mapping using remote sensing. In order to solve these problems, an optical remote sensing aquaculture index named the marine aquaculture index (MAI) is proposed. Based on this spectral index, using time series Sentinel-1 and Sentinel-2 satellite data, a random forest classification scheme is proposed for mapping mariculture by combining spectral, textural, geometric, and synthetic aperture radar (SAR) backscattering. The results revealed that (1) MAI can emphasize the difference between mariculture and seawater; (2) the overall accuracy of mariculture in the Bohai Rim is 94.10%, and the kappa coefficient is 0.91; and (3) the area of cage aquaculture and raft aquaculture in the Bohai Rim is 16.89 km2 and 1206.71 km2, respectively. This study details an effective method for carrying out mariculture monitoring and ensuring the sustainable development of aquaculture.

Multiterritory perforator flap is an important plastic surgery technique, yet its efficacy can be limited by partial necrosis at the choke Ⅱ zone. Butylphthalide (NBP) has been used for many diseases but has not been studied in the multiterritory perforator flap. With the effect of NBP, we observed increasing in capillary density, inhibition of autophagy and oxidative stress, and a reduction in apoptosis of cells, all consistent with increased flap survival. However, the protective effect of NBP on multiterritory perforator flap was lost following administration of the autophagy agonist rapamycin (Rap). Through the above results, we assumed that NBP promotes flap survival by inhibiting autophagy. Thus, this study has found a new pharmacological effect of NBP on the multiterritory perforator by inhibiting autophagy to prevent distal postoperative necrosis and exert effects on angiogenesis, oxidative stress, and apoptosis within the flap.

Estrogen deficiency is associated with increased incidence of cardiovascular diseases. But merely estrogen supplementary treatment can induce many severe complications such as breast cancer. The present study was designed to elucidate molecular mechanisms underlying increased susceptibility of arrhythmogenesis during myocardial infarction with estrogen deprivation, which provides us a new target to cure cardiac disease accompanied with estrogen deprivation. We successfully established a rat model of myocardial ischemia (MI) accompanied with estrogen deprivation by coronary artery ligation and ovariectomy (OVX). Vulnerability and mortality of ventricular arrhythmias increased in estrogen deficiency rats compared to non estrogen deficiency rats when suffered MI, which was associated with down-regulation of microRNA-151-5p (miR-151-5p). Luciferase Reporter Assay demonstrated that miR-151-5p can bind to the 3'-UTR of FXYD1 (coding gene of phospholemman, PLM) and inhibit its expression. We found that the expression of PLM was increased in (OVX+MI) group compared with MI group. More changes such as down-regulation of Kir2.1/IK1, calcium overload had emerged in (OVX+MI) group compared to MI group merely. Transfection of miR-151-5p into primary cultured myocytes decreased PLM levels and [Ca(2+)]i, however, increased Kir2.1 levels. These effects were abolished by the antisense oligonucleotides against miR-151-5p. Co-immunoprecipitation and immunofluorescent experiments confirmed the co-localization between Kir2.1 and PLM in rat ventricular tissue. We conclude that the increased ventricular arrhythmias vulnerability in response to acute myocardial ischemia in rat is critically dependent upon down-regulation of miR-151-5p. These findings support the proposal that miR-151-5p could be a potential therapeutic target for the prevention of ischemic arrhythmias in the subjects with estrogen deficiency.

为进一步提高辣木籽蛋白资源的开发利用，采用盐提法提取辣木籽蛋白，再采用超声波辅助酶法制备辣木籽ACE抑制肽。以水解度和ACE抑制率为评价指标，通过单因素实验探究超声波功率、超声酶解时间、超声酶解温度及料液比对制备ACE抑制肽的影响,采用响应面法对制备工艺条件进行优化。结果表明：超声波辅助酶法制备辣木籽ACE抑制肽的最佳酶解工艺条件为碱性蛋白酶添加量5.5 mg/mL、pH 9、超声波功率500 W、超声酶解时间1.7 h、超声酶解温度55 ℃、料液比1∶ 45，在此条件下制备的酶解物ACE抑制率达到78.32%，水解度为778%。以辣木籽为原料制备ACE抑制肽作为功能性蛋白肽产品，可有效提高辣木籽蛋白资源的开发利用。 To further improve the exploitation of Moringa oleifera seed protein resources, the protein in Moringa oleifera seeds was extracted by salt extraction method, and then Moringa oleifera seeds ACE inhibitory peptide was prepared by ultrasound-assisted enzymatic hydrolysis. With the degree of hydrolysis and ACE inhibition rate as the evaluation indexes, the effects of the ultrasonic power, ultrasonic enzymolysis time, ultrasonic enzymolysis temperature and ratio of material to liquid on the preparation of ACE inhibitory peptide were studied through single factor experiment. The preparation process conditions of ACE inhibitory peptide were optimized by response surface methodology. The results showed that the optimal ultrasound-assisted enzymatic hydrolysis conditions were obtained as follows: Alkaline protease dosage 5.5 mg/mL,pH 9,ultrasonic power 500 W, ultrasonic enzymolysis time 1.7 h, ultrasonic enzymolysis temperature 55 ℃ and ratio of material to liquid 1∶ 45. Under these conditions, the ACE inhibition rate of the prepared product reached 78.32% and the degree of hydrolysis was 7.78%. Using ACE inhibitory peptide prepared from Moringa oleifera seeds as a functional protein peptide product can effectively improve the exploitation of Moringa oleifera seed protein resources.
