Arbuscular mycorrhizal fungi (AMF) and the antioxidant germanium (Ge) are promising tools for boosting bioactive compound synthesis and producing healthier foods. However, their combined effect remains unexplored. This study demonstrates the synergistic impact of AMF and Ge on the growth, metabolite accumulation, biological activities, and nutritional qualities of Spinacia oleracea L. (spinach), a globally significant leafy vegetable. Individually, Ge and AMF increased biomass by 68.1% and 22.7%, respectively, while their combined effect led to an 86.3% increase. AMF and Ge also improved proximate composition, with AMF–Ge interaction enhancing crude fiber and mineral content (p < 0.05). Interestingly, AMF enhanced photosynthesis-related parameters (e.g., total chlorophyll) in Ge treated plants, which in turn increased carbohydrate accumulation. This accumulation could provide a route for the biosynthesis of amino acids, organic acids, and fatty acids, as evidenced by increased essential amino acid and organic acid levels. Consistently, the activity of key enzymes involved in amino acids biosynthesis (e.g., glutamine synthase (GS), methionine biosynthase (MS), lysine biosynthase (LS)) showed significant increments. Furthermore, AMF improved fatty acid levels, particularly unsaturated fatty acids in Ge-treated plants compared to the control. In addition, increased phenylalanine provided a precursor for the production of antioxidants (e.g., phenols and flavonoids), through the action of the enzyme phenylalanine ammonia-lyase (PAL), resulting in improved antioxidant activity gains as indicated by FRAP, ABTS, and DPPH assays. This study is the first to show that Ge enhances the beneficial effect of AMF on spinach, improving growth and nutritional quality, with promising implications for agricultural practices. Fungal infection in agricultural grains is a global problem, particularly if it is accompanied by mycotoxin production. In this study, the degradation of mycotoxins by laccase and manganese peroxidase was investigated. Aspergillus flavus, Aspergillus fumigatus, and Fusarium graminearum were recorded in infected maize grains. Aflatoxin B1 (AF B1) was detected (from 3.38 to 2.60 ppm) on the infected samples by fungi compared to other detected aflatoxins. Trichothecene (T-2) toxin and deoxynivalenol (DON) were recorded with concentrations ranging from 0.464 to 0.184 ppm and 0.370 to 0.214 ppm, respectively. The addition of laccase and manganese peroxidase to the inoculated medium with A. flavus and F. graminearum individually degraded the produced AF B1, B2, G1, G2, T-2 toxin, and DON from 5.0, 1.33, 0.76, 0.61, 0.63, and 0.38 ppm to 2.77, 0.66, 0.37, 0.15, 0.45, and 0.38 ppm using laccase, to 3.08, 1.25, 0.61, 0.39, 0.55, and 0.36 ppm using manganese peroxidase. The computational technique (docking) demonstrated the laccase and manganese peroxidase activities on aflatoxin and DON degradation. Consequently, the results suggested that laccase (PDB ID: 1HFU) and manganese peroxidase (PDB ID: 1MNP) promise innovative activity toward aflatoxin degradation, while 1HFU has more effect than 1MNP on DON degradation. Cinnamon is a plant with significant medicinal value that is used extensively as a spice, flavoring, and fragrance ingredient. Phytochemical characterization via high-performance liquid chromatography, antimicrobial activity against various microorganisms, anti-diabetic properties by α-glucosidase and α-amylase assessments, antioxidant activity via 2,2-diphenyl-1-picrylhydrazyl-hydrate, and total antioxidant capacities of cinnamon bark were evaluated. Moreover, antitumor potential against human breast cancer (MCF-7) and human fetal lung fibroblast (WI-38) cells of the bark extract were detected. A promising antimicrobial action towards Enterococcus faecalis, Staphylococcus aureus, Klebsiella pneumoniae, Salmonella typhi, Candida albicans, and Mucor circinelloides where the inhibition areas were 25, 23, 18, 19, 29, and 16 mm, respectively, was recorded. Cinnamon bark has IC50 of 5.01 and 2.58 µg/mL compared to standard acrobose with their IC50 values 4.32 and 1.99 µg/mL, respectively for α-amylase and α-glucosidase inhibitions. Cinnamon bark extract has IC50 of 77.39 ± 0.84 and 162.67 ± 0.28 µg/mL toward cell lines of MCF-7 and WI-38, respectively. The protective effect of cinnamon in accelerating the apoptosis of MCF-7 cells has been verified by flow cytometric evaluation employing Annexin-V and cell cycle kits, as well as the increasing levels of malondialdehyde, hydrogen peroxide, and nitric oxide with decreasing glutathione and catalase levels. Nanotechnology is an exciting area with great potential for use in biotechnology due to the far-reaching effects of nanoscale materials and their size-dependent characteristics. Silver and other metal nanoparticles have attracted a lot of attention lately because of the exceptional optical, electrical, and antimicrobial characteristics they possess. Silver nanoparticles (AgNPs) stand out due to their cost-effectiveness and abundant presence in the earth's crust, making them a compelling subject for further exploration. The vital efficacy of silver nanoparticles in addressing environmental concerns is emphasized in this thorough overview that dives into their significance in environmental remediation. Leveraging the distinctive properties of AgNPs, such as their antibacterial and catalytic characteristics, innovative solutions for efficient treatment of pollutants are being developed. The review critically examines the transformative potential of silver nanoparticles, exploring their various applications and promising achievements in enhancing environmental remediation techniques. As environmental defenders, this study advocates for intensified investigation and application of silver nanoparticles. Furthermore, this review aims to assist future investigators in developing more cost-effective and efficient innovations involving AgNPs carrying nanoprobes. The functional and structural features of microbial load in soil are influenced by the presence of insecticides. This study examined the impact of two organophosphorus insecticides, dimethoate and parathion on the microbial load of soil. The colony count of fungi, actinomycetes, bacteria, and nitrogen fixing bacteria was reduced after insecticides application at 7 and 14 days, but at 28 days the colony count began to increase. The growth of two white rot fungi including Pleurotus sajor-caju and Phanerochaete chrysosporium was affected by parathion, which was reflected by a decrease in colony radius to 1.85±0.05 and 0.75±0.06 cm, respectively, and by dimethoate, reflected by a decrease in colony radius to 3.33±0.12 and 1.85±0.05 cm, respectively at 40 mg/L compared to colony radius at control 7.90±0.12 and 7.50±0.06 cm, respectively. The applied low concentration (10 mg/L) encouraged P. sajor-caju and P. chrysosporium to remove up to 87.7% and 81.8% of dimethoate, respectively, and 69.20 and 68.30% of parathion, respectively compared with the decomposition at high dose (40 mg/L) at 28 days. Water pollution has become a worldwide issue, particularly in developing countries. This study aimed to cleanse water contaminated with reactive red (RR-198) and reactive blue (RB-19) dyes via adsorption onto nonliving white rot fungal biomass Phanerochaete chrysosporium under different operational conditions. It was found that P. chrysosporium removed more of RB-19 than RB-198. The conditions of pH 3, temperature 50 °C, adsorbent dosage 0.6 g, particle sizes of adsorbent (0.25 mm), and contact time (30 min) were optimum for removal of the dyes. RR-198 and RB-19 were removed with levels of 78.4% and 86.4%, respectively at pH 3; 79.6% and 90.0%, respectively using 0.6 g of adsorbent dosage; 82.0% and 87.0%, respectively at 30 min; and 82.0% and 87.2%, respectively at 50 °C. The production of lignocellulytic enzymes by microwave-radiated Pleurotus sajor-caju was assayed. Wheat straw was employed as substrate to P. sajor-caju for production of laccase, manganese peroxidase (MnPase), filter-paperase (FPase), carboxmethyl cellulase (CMCase), and cellulase (as evaluated using microcrystalline cellulose). P. sajor-caju exposed to 10 s of microwave radiation (MR) showed maximum growth with colony radius of 7.17 ± 0.45 cm, while with increasing the exposure time up to 50 s the growth decreased up to 2.67 ± 0.22 cm. Moreover, it failed to grow at 80 s of exposure time. Cellulase, MnPase, FPase, CMCase, and laccase activities were induced to 37 ± .0.54, 49 ± 2.36, 189 ± 2.12, 0.37 ± 0.06, and 1.58 ± 0.03 U/mL compared to that at control 31 ± 0.25, 46 ± 1.25, 177 ± 1.65, 0.28 ± 0.03, and 1.37 ± 0.12 U/mL, respectively as a result of P. sajor-caju exposure to 10 s of MR. As the exposure time increased, these enzymes activity decreased. Different levels of moisture with surfactant (polysorbate 80) were applied to optimize the enzymes activities at 10 s of exposure time. The optimum activities 3.15 ± 0.23, 0.62 ± 0.06, 269 ± 5.36, 65 ± 1.63, and 48 ± 0.98 U/mL were recorded for cellulase, MnPase, FPase, CMCase, and laccase, respectively at 70% of moisture and 0.15 mL/L of polysorbate 80. The extraction methods used to obtain natural products face some problems, such as solvent toxicity, high extraction time, and low yields. Supercritical carbon dioxide fluid extraction (SFE-CO2) is an encouraging extraction system for obtaining high-yield of natural extracts. In this work, Schinus terebinthifolia fruits were extracted via SFE-CO2 using two conditions: A (static extraction) (SE) for 15 min, followed by dynamic extraction (DE) for 45 min, and B (without SE but with DE for 60 min). The extract yield was 0.205 g and 0.236 g via condition A and B, respectively. High-performance liquid chromatography assessment revealed the occurrence of several constituents with high quantities in the extract at condition B. The well diffusion test showed inhibition of 26 ± 0.1, 25 ± 0.2, 29 ± 0.1, 33 ± 0.2, 27 ± 0.1, and 8.0 ± 0.1 mm zones using the extract at condition B, while at condition A there were low inhibition zones towards Staphylococcus aureus, Pseudomonas areginosa, Bacillus subtilis, Escherichia coli, Candida albicans, and Aspergillus niger, correspondingly. Lipase (obesity stimulant) and butyrylcholinesterase (Alzheimer stimulant) were inhibited by the extract at condition B with IC50 quantities of 27.03 and 4.83 µg/mL, while it was 37.45 and 17.57 µg/mL, respectively at condition A. Mangiferin is a bioactive substance present in high concentration in mangoes and also in some other fruits. Owing to its potential as a chemopreventive and chemotherapeutic agent against several types of cancer, this unique, significant, and well‐researched polyphenol has received a lot of attention recently. It possesses the ability to treat cancers, including rectal cancer, prostate cancer, ovarian cancer, leukemia, gastric cancer, liver cancer, chronic pancreatitis, and lung cancer. It can control/regulate multiple key signaling pathways, such as signal transducer and activator of transcription 3 (STAT3), second mitochondria‐derived activator of caspases/direct inhibitor of apoptosis (IAP)‐binding protein with low propidium iodide (pl) (Smac/DIABLO) nuclear factor kappa B (NF‐κB), phosphatidylinositol 3 kinase/protein 3 kinase (PI3K/Akt), transforming growth factor beta/suppressor of mothers against decapentaplegic (TGF‐β/SMAD), c‐jun N‐terminal kinase/p38 mitogen‐activated protein kinase (JNK/p38‐MAPK), and phosphor‐I kappa B kinase (p‐IκB), which are crucial to the development of cancers. By triggering apoptotic signals and halting the advancement of the cell cycle, it can also prevent some cancer cell types from proliferating and developing. It has been revealed that mangiferin targets a variety of adhesion molecules, cytokines, pro‐inflammatory transcription factors, kinases, chemokines, growth factors, and cell‐cycle proteins. By means of preventing the onset, advancement, and metastasis of cancer, these targets may mediate the chemopreventive and therapeutic effects of mangiferin. Mangiferin has confirmed potential benefits in lung, cervical, breast, brain, and prostate cancers as well as leukemia whether administered alone or in combination with recognized anticancer compounds. It is thought to be risk-free, environmentally benign, and safe for biological processes to produce zinc oxide nanoparticles from renewable resources. This study examined Cassia javanica's ability to create ZnONPs. The generated ZnONPs were analyzed using a variety of techniques, such as TEM, FTIR spectroscopy, UV-Vis spectroscopy, and XRD analysis. The antibacterial potential of ZnONPs has been investigated using both Agar well diffusion and microtitreplate (MTP) methods. One method used to evaluate ZnONPs' capacity to scavenge free radicals at different concentrations was the DPPH method. The permanent zinc oxide (ZnO) shape and the naturally occurring crystal structure of ZnONPs were validated by the XRD data. ZnONPs showed antibacterial activity with MICs of 31.7 μg/mL toward Bacillus subtilis, 62.5 μg/mL for Salmonella typhimurium, Escherichia coli while Clostridium sporogenes and Bacillus pumilus was 125μg/mL. Furthermore, ZnONPs demonstrated a range of antibiofilm activities toward Staphylococcus aureus (MRSA). ZnONPs showed an intriguing antioxidant capacity, achieving IC50 of 109.3 μg/ml μg/mL. Additionally, ZnONPs demonstrated low toxic effect on Vero cell with IC50 154.01 μg/mL as well as possible anticancer action when applied to the carcinoma cell lines HepG2 with IC50 of 47.48 μg/mL. Efficacies of plant metabolites are known to be dependent on their extraction methods. Yields and compositions of phytoconstituents in the extract were evaluated following supercritical fluid extraction (SFE) of Vitex agnus-castus leaves, static extraction times (SET) for 30 min, subsequently dynamic extraction time (DET) for 30 min (condition A) and SET for 0 min followed by DET for 60 min (condition B). The extract exposed to condition B gave an extraction yield of 0.169 g compared to 0.115 g for condition A. High-performance liquid chromatography analysis revealed compounds including cinnamic acid, kaempferol, ferulic acid, rutin, and caffeic acid, in high concentrations in the extract exposed to condition B. Staphylococcus aureus, Pseudomonas aeruginosa, Klebsiella pneumoniae, Enterococcus faecalis, and Candida albicans were more affected by the condition B with 32 ± 0.1, 20 ± 0.2, 32 ± 0.2, 35 ± 0.2, and 40 ± 0.1 mm inhibition zones, respectively. Less MIC and MBC were noticed of the exposed extract to condition B than to condition A against C. albicans and bacteria. The IC50 of the extract exposed to condition B was high against ovarian tumor cells. Presently the efficacy of the exposed extract to condition B for wound healing process was documented. Plants represent a great source of medicines, and for their components to be discovered, extraction processes must be developed, especially methods based on green technology. Supercritical fluid extraction (SFE) was employed as a green method for Moringa oleifera extraction in the present investigation. The maximum yield of extraction was obtained at 25 MPa. Moreover, the extraction at 25 MPa induced the release of various phenols and flavonoids, as analyzed via high-performance liquid chromatography. The investigation revealed the concentrations of chlorogenic, gallic, rosmarinic, and coumaric acids to be 150.59, 89.90, 44.75, and 29.41 μg/mL, respectively at 25 MPa. However, their concentrations were 0.73, 1.53, 0.24, and 0.04 μg/mL, respectively at 15 MPa; vs. 4.73, 2.62, 1.06, and 0.50 at 35 MPa, respectively. Totals of saponin, flavonoid, phenolic, tannins, and alkaloid were recorded in maximum yield at 25 MPa. Moringa oleifera extracted at 35 MPa reflected highest inhibition zones of 27 ± 0.1, 30 ± 0.2, and 30 ± 0.1 mm against C. glabrata, C. tropicalis, and C. albicans, correspondingly. α-Amylase and α-glucosidase activities were greatly suppressed by the M. oleifera extract at 25 MPa with less IC50 (12.97 μg/mL and 6.0 µg/mL), than the IC50 (53.46 and 22.02 μg/mL) at 15 MPa, compared with acarbose IC50 (5.52 and 2.64 μg/mL), correspondingly. Green routes for the bio-designing of bicomponent nanocomposites and their utilizations have attracted many investigators. Bio-designing of CuO@ZnO nanocomposites was performed using spent mushroom substrate (SMS). Ultraviolet-spectrophotometry, transmission electron microscopy, Fourier transform infrared (FT-IR), and energy dispersive Xray (EDX), besides X-ray diffraction (XRD) were exploited to characterize the synthesized CuO@ZnO. The dimensions of CuO@ZnO nanocomposites ranged from 31.4 and 95.9 nm. Both FT-IR and EDX analyses displayed the presence of some organic constituents from the SMS that joined to the surface of the fabricated CuO@ZnO nanocomposite. CuO@ZnO nanocomposite succeeded in inhibiting Candida albicans with an inhibition zone of 33.5 ± 2 mm. C. albicans biofilm was affected by CuO@ZnO nanocomposite with biofilm inhibition of 25.08, 68.70, and 88.56% at 25, 50, and 75% of minimum inhibitory concentration, respectively. Molecular docking studies showed substantial binding affinities, as well as common hydrogen bonds. Optimum binding sites for CuO and ZnO nanoparticles were found to have binding affinities of interactions with 4YDE, 3DRA, and 1EAG proteins of C. albicans, resulting in, respectively, -2.7942, -3.30097, and -2.52129 kcal/mol, and - 3.78244, -4.6029, and -4.1352 kcal/mol values. In the current investigation, the antibacterial activity of zinc oxide nanoparticles (ZnO-NPs) formed by an aqueous extract of Psidium guajava leaves against foodborne pathogenic bacterial strains was investigated. To achieve this goal, 33 bacterial isolates were obtained from spoiled fruits. Among these isolates, 79% showed cellulase activity, 82% showed amylase activity, 81% exhibited xylanase potential, and 65% exhibited lipase activity. Moreover, 12 isolates showed complete hemolysis (β-hemolysis). The identification of these isolates was done using sequencing and amplification of 16s rRNA as Staphylococcus aureus (two strains), Pseudomonas syringae (one strain), E. coli (two strains), Salmonella typhimurium (two strains), Listeria monocytogenes (one isolate), Bacillus cereus (two isolates), and Bacillus subtilis (two isolates). The formed ZnO-NPs by aqueous Psidium guajava leaf extract were characterized using UV, FT-IR, TEM, EDX, XRD, DLS, and Zeta potential. The data revealed the successful formation of a spherical shape, crystallographic structure, and well-arranged ZnO-NPs. FT-IR showed the effect of different functional groups in the plant extract in the formation of ZnO-NPs through reducing, capping, and stabilizing of end products. Moreover, EDX analysis showed that the Zn ion occupied the main component of the produced NPs. Interestingly, the obtained bacterial strains showed varied sensitivity toward green-synthesized ZnO-NPs. The growth inhibition of foodborne pathogenic strains by ZnO-NPs was concentration dependent, forming a zone of inhibition in the range of 20–23 mm at a concentration of 200 µg mL−1, which decreased to 15–18 mm at 100 µg mL−1. Moreover, the values of MIC were 25 and 50 µg mL−1 based on the bacterial strain. Overall, the green-synthesized ZnO-NPs can be a useful approach for inhibiting the growth of spoilage bacterial strains that destroy fruits and hence reduce the harmful effects of traditional treatment methods on the environment and human health. The biomass filtrate of the endophytic bacterial strain Bacillus amyloliquefaciens Fa.2 was utilized for the eco-friendly production of silver nanoparticles (Ag-NPs). The yellowish-brown color's optical properties showed a maximum surface plasmon resonance at 415 nm. The morphological and elemental composition analysis reveals the formation of spherical shapes with sizes of 5–40 nm, and the Ag ion comprises the major component of the produced Ag-NPs. X-ray diffraction confirmed the crystalline structure, whereas dynamic light scattering reveals the high stability of synthesized Ag-NPs with a polydispersity index of 0.413 and a negative zeta potential value. The photocatalytic experiment showed the efficacy of Ag-NPs to degrade methylene blue with maximum percentages of 73.9 ± 0.5 and 87.4 ± 0.9% under sunshine and UV irradiation, respectively, compared with 39.8% under dark conditions after 210 min. Additionally, the reusability of Ag-NPs was still more active for the fifth run, with a percentage decrease of 11.6% compared with the first run. Interestingly, the biogenic Ag-NPs showed superior antimicrobial activity against different pathogenic Gram-negative bacteria (MIC = 6.25 µg mL−1), Gram-positive bacteria (MIC = 12.5 µg mL−1), and uni- and multicellular fungi (MIC = 12.5 µg mL−1). Moreover, the biosynthesized Ag-NPs could target cancer cells (Pc3 and Mcf7) at low concentrations compared with normal cell (Vero) lines. The IC50 of normal cells is 383.7 ± 4.1 µg mL−1 compared with IC50 Pc3 (2.5 ± 3.5 µg mL−1) and McF7 (156.1 ± 6.8 µg mL−1). Salinity has become a major environmental concern of agricultural lands, impairing crop production. The current study aimed to examine the role of zinc oxide nanoparticles (ZnO NPs) in reducing the oxidative stress induced by salinity and the overall improvement in phytochemical properties in barley. A total of nine different barley genotypes were first subjected to salt (NaCl) stress in hydroponic conditions to determine the tolerance among the genotypes. The genotype Annora was found as most sensitive, and the most tolerant genotype was Awaran 02 under salinity stress. In another study, the most sensitive (Annora) and tolerant (Awaran 02) barley genotypes were grown in pots under salinity stress (100 mM). At the same time, half of the pots were provided with the soil application of ZnO NPs (100 mg kg−1 ), and the other half pots were foliar sprayed with ZnO NPs (100 mg L−1 ). Salinity stress reduced barley growth in both genotypes compared to control plants. However, greater reduction in barley growth was found in Annora (sensitive genotype) than in Awaran 02 (tolerant genotype). The exogenous application of ZnO NPs ameliorated salt stress and improved barley biomass, photosynthesis, and antioxidant enzyme activities by reducing oxidative damage caused by salt stress. However, this positive effect by ZnO NPs was observed more in Awaran 02 than in Annora genotype. Furthermore, the foliar application of ZnO NPs was more effective than the soil application of ZnO NPs. A new series of 2-aminobenzothiazole hybrids linked to thiazolidine-2,4-dione 4a–e, 1,3,4-thiadiazole aryl urea 6a–d, and cyanothiouracil moieties 8a–d was synthesised. The in vitro antitumor effect of the new hybrids was assessed against three cancer cell lines, namely, HCT-116, HEPG-2, and MCF-7 using Sorafenib (SOR) as a standard drug. Among the tested compounds, 4a was the most potent showing IC50 of 5.61, 7.92, and 3.84 µM, respectively. Furthermore, compounds 4e and 8a proved to have strong impact on breast cancer cell line with IC50 of 6.11 and 10.86 µM, respectively. The three compounds showed a good safety profile towards normal WI-38 cells. Flow cytometric analysis of the three compounds in MCF-7 cells revealed that compounds 4a and 4c inhibited cell population in the S phase, whereas 8a inhibited the population in the G1/S phase. The most promising compounds were subjected to a VEGFR-2 inhibitory assay where 4a emerged as the best active inhibitor of VEGFR-2 with IC50 91 nM, compared to 53 nM for SOR. A novel series of 12 antipyrine derivatives containing 1,3,4-oxadiazoles (4a-d), 1,3,4-thiadiazoles (6a-d), and pyrimidines (8a-d), was preparedand assessed for its potential in vitro COX-2 inhibitors. Compared to Celecoxib, compounds 4b-d and 8d were the most potent derivatives c with a half-maximal inhibitory concentration range of 53–69 nM. Considering COX-2 selectivity index, compounds 4 b and 4c were chosen among these most potent derivatives for further investigation. The in vivo ability of compounds 4 b and 4c to counteract carrageenan-induced paw edoema has been assessed and their potential underlying mechanisms have been elucidated and the results have been further validated using molecular docking simulations.
