How does one determine if a collection of bars joined by freely rotating hinges cannot be deformed without changing the length of any of the bars? In other words, how does one determine if a bar-joint graph is rigid? This question has been definitively answered using combinatorial rigidity theory in two dimensions via the Geiringer-Laman Theorem. However, it has not yet been answered using combinatorial rigidity theory in higher dimensions, given known counterexamples to the trivial dimensional extension of the Geiringer-Laman Theorem. To work towards a combinatorial approach in dimensions beyond two, we present a theorem for gluing two rigid bar-joint graphs together that remain rigid. When there are no overlapping vertices between the two graphs, the theorem reduces to Tay's theorem used to identify rigidity in body-bar graphs. When there are overlapping vertices, we rely on the notion of pinned rigid graphs to identify and constrain rigid motions. This theorem provides a basis for an algorithm for recursively constructing rigid clusters that can be readily adapted for computational purposes. By leveraging Henneberg-type operations to grow a rigid (or minimally rigid) graph and treating simplices-where every vertex connects to every other vertex-as fundamental units, our approach offers a scalable solution with computational complexity comparable to traditional methods. Tumor spheroids are in vitro three-dimensional, cellular collectives consisting of cancerous cells. Embedding these spheroids in an in vitro fibrous environment, such as a collagen network, to mimic the extracellular matrix (ECM) provides an essential platform to quantitatively investigate the biophysical mechanisms leading to tumor invasion of the ECM. To understand the mechanical interplay between tumor spheroids and the ECM, we computationally construct and study a three-dimensional vertex model for a tumor spheroid that is mechanically coupled to a cross-linked network of fibers. In such a vertex model, cells are represented as deformable polyhedrons that share faces. Some fraction of the boundary faces of the tumor spheroid contain linker springs connecting the center of the boundary face to the nearest node in the fiber network. As these linker springs actively contract, the fiber network remodels. By toggling between fluid-like and solid-like spheroids via changing the dimensionless cell shape index, we find that the spheroid rheology affects the remodeling of the fiber network. More precisely, fluid-like spheroids displace the fiber network more on average near the vicinity of the spheroid than solid-like spheroids. We also find more densification of the fiber network near the spheroid for the fluid-like spheroids. These spheroid rheology-dependent effects are the result of cellular motility due to active cellular rearrangements that emerge over time in the fluid-like spheroids to generate spheroid shape fluctuations. Our results uncover intricate morphological-mechanical interplay between an embedded spheroid and its surrounding fiber network with both spheroid contractile strength and spheroid shape fluctuations playing important roles in the pre-invasion stages of tumor invasion., Comment: 19 pages, 12 figures, 7 SI figures

Cross-sections of cell shapes in a tissue monolayer typically resemble a tiling of convex polygons. Yet, examples exist where the polygons are not convex with curved cell-cell interfaces, as seen in the adaxial epidermis. To date, two-dimensional vertex models predicting the structure and mechanics of cell monolayers have been mostly limited to convex polygons. To overcome this limitation, we introduce a framework to study curvy cell-cell interfaces at the subcellular scale within vertex models by using a parameterized curve between vertices that is expanded in a Fourier series and whose coefficients represent additional degrees of freedom. This extension to non-convex polygons allows for cells with same shape index, or dimensionless perimeter, to be, for example, either elongated or globular with lobes. In the presence of applied, anisotropic stresses, we find that local, subcellular curvature, or buckling, can be energetically more favorable than larger scale deformations involving groups of cells. Inspired by recent experiments, we also find that local, subcellular curvature at cell-cell interfaces emerges in a group of cells in response to the swelling of additional cells surrounding the group. We explore rectification phenomena in a system where two-dimensional random walkers interact with a funnel-shaped ratchet under two distinct classes of reflection rules. The two classes include the angle of reflection exceeding the angle of incidence ($\theta_{reflect} > \theta_{incident}$), or vice versa ($\theta_{reflect} < \theta_{incident}$). These generalized boundary reflection rules are indicative of non-equilibrium conditions due to the introduction of energy flows at the boundary. Our findings reveal that the nature of such particle-wall interactions dictates the system's behavior: the funnel either acts as a pump, directing flow, or as a collector, demonstrating a ratchet reversal. Importantly, we provide a geometric proof elucidating the underlying mechanism of rectification, thereby offering insights into why certain interactions lead to directed motion, while others do not., Comment: 5 pages, 6 figures

In the realm of biological flow networks, the ability to dynamically adjust to varying demands is paramount. Drawing inspiration from the remarkable adaptability of Physarum polycephalum, we present a novel physical mechanism tailored to optimize flow networks. Central to our approach is the principle that each network component -- specifically, the tubes -- harnesses locally available information to collectively minimize a global cost function. Our findings underscore the scalability of this mechanism, making it feasible for larger, more complex networks. We construct a comprehensive phase diagram, pinpointing the specific network parameters under which successful adaptation, or tuning, is realized. By training linear physical networks to learn linear transformations, we discern how their physical properties evolve due to weight update rules. Our findings highlight a striking similarity between the learning behaviors of such networks and the processes of aging and memory formation in disordered and glassy systems. We show that the learning dynamics resembles an aging process, where the system relaxes in response to repeated application of the feedback boundary forces in presence of an input force, thus encoding a memory of the input-output relationship. With this relaxation comes an increase in the correlation length, which is indicated by the two-point correlation function for the components of the network. We also observe that the square root of the mean-squared error as a function of epoch takes on a non-exponential form, which is a typical feature of glassy systems. This physical interpretation suggests that by encoding more detailed information into input and feedback boundary forces, the process of emergent learning can be rather ubiquitous and, thus, serve as a very early physical mechanism, from an evolutionary standpoint, for learning in biological systems., Comment: 11 pages, 7 figures

The mechanics of a foam typically depends on the bubble geometry, topology, and the material at hand, be it metallic or polymeric, for example. While the foam energy functional for each bubble is typically minimization of surface area for a given volume, biology provides us with a wealth of additional energy functionals, should one consider biological cells as a foam-like material. Here, we focus on a mean field approach to obtain the elastic moduli, within linear response, for an ordered, three-dimensional vertex model using the space-filling shape of a truncated octahedron and whose energy functional is characterized by a restoring surface area spring and a restoring volume spring. The tuning of the three-dimensional shape index exhibits a rigidity transition via a compatible-incompatible transition. Specifically, for smaller shape indices, both the target surface area and volume cannot be achieved, while beyond some critical value of the three-dimensional shape index, they can be, resulting in a zero-energy state. As the elastic moduli depend on curvatures of the energy when the system, we obtain these as well. In addition to analytically determining the location of the transition in mean field, we find that the rigidity transition and the elastic moduli depend on the parameterization of the cell shape with this effect being more pronounced in three dimensions given the array of shapes that a polyhedron can take on (as compared to a polygon). We also uncover nontrivial dependence on the deformation protocol in which some deformations result in affine motion of the vertices, while others result in nonaffine motion. Such dependencies on the shape parameterization and deformation protocol give rise to a nontrivial shape landscape and, therefore, nontrivial mechanical response even in the absence of topology changes., Comment: 18 pages, 24 figures

How genes affect tissue scale organization remains a longstanding biological puzzle. As experimental efforts are underway to solve this puzzle via quantification of gene expression and sub-cellular, cellular and tissue structure, computational efforts remain far behind. To potentially help accelerate the computational efforts, we review two recent publications, the first on a cellular-based model for tissues and the second on a cell nucleus model consisting of chromatin and a lamina shell. We then give a perspective on how the two models can be combined to test multiscale hypotheses linking the chromatin scale and the tissue scale. To be concrete, we turn to an in vitro system for the brain known as a brain organoid. We provide a multiscale hypothesis to distinguish structural differences between brain organoids built from induced-pluripotent human stem cells and from induced-pluripotent gorilla and chimpanzee stem cells. Recent experiments discover that a cell fate transition from neuroepithelial cells to radial glial cells includes a new intermediate state that is delayed in human-derived brain organoids as compared to their genetically-close relatives, which significantly narrows and lengthens the cells on the apical side [1]. Additional experiments revealed that the protein ZEB2 plays a major role in the emergence of this new intermediate state with ZEB2 mRNA levels peaking at the onset of the emergence [1]. We postulate that the enhancement of ZEB2 expression driving this intermediate state is potentially due to chromatin reorganization. More precisely, there exists critical strain triggering the reorganization that is higher for human-derived stem cells, thereby resulting in a delay. Such a hypothesis can readily be tested experimentally within individual cells and within brain organoids as well as computationally to work towards solving the gene-to-tissue organization puzzle., Comment: 13 pages, 8 figures

Nanoparticles, such as viruses, can enter cells via endocytosis. During endocytosis, the cell surface wraps around the nanoparticle to effectively eat it. Prior focus has been on how nanoparticle size and shape impacts endocytosis. However, inspired by the noted presence of extracellular vimentin affecting viral and bacteria uptake, as well as the structure of coronaviruses, we construct a computational model in which both the cell-like construct and the virus-like construct contain filamentous protein structures protruding from their surfaces. We then study the impact of these additional degrees of freedom on viral wrapping. We find that cells with an optimal density of filamentous extracellular components (ECCs) are more likely to be infected as they uptake the virus faster and use relatively less cell surface area per individual virus. At the optimal density, the cell surface folds around the virus, and folds are faster and more efficient at wrapping the virus than crumple-like wrapping. We also find that cell surface bending rigidity helps generate folds, as bending rigidity enhances force transmission across the surface. However, changing other mechanical parameters, such as the stretching stiffness of filamentous ECCs or virus spikes, can drive crumple-like formation of the cell surface. We conclude with the implications of our study on the evolutionary pressures of virus-like particles, with a particular focus on the cellular microenvironment that may include filamentous ECCs., Comment: 15 pages, 8 figures

We introduce frequency propagation, a learning algorithm for nonlinear physical networks. In a resistive electrical circuit with variable resistors, an activation current is applied at a set of input nodes at one frequency, and an error current is applied at a set of output nodes at another frequency. The voltage response of the circuit to these boundary currents is the superposition of an `activation signal' and an `error signal' whose coefficients can be read in different frequencies of the frequency domain. Each conductance is updated proportionally to the product of the two coefficients. The learning rule is local and proved to perform gradient descent on a loss function. We argue that frequency propagation is an instance of a multi-mechanism learning strategy for physical networks, be it resistive, elastic, or flow networks. Multi-mechanism learning strategies incorporate at least two physical quantities, potentially governed by independent physical mechanisms, to act as activation and error signals in the training process. Locally available information about these two signals is then used to update the trainable parameters to perform gradient descent. We demonstrate how earlier work implementing learning via chemical signaling in flow networks also falls under the rubric of multi-mechanism learning., Comment: 9 pages, 0 figures

The ASTRI Mini-Array (MA) is an INAF project to build and operate a facility to study astronomical sources emitting at very high-energy in the TeV spectral band. The ASTRI MA consists of a group of nine innovative Imaging Atmospheric Cherenkov telescopes. The telescopes will be installed at the Teide Astronomical Observatory of the Instituto de Astrofisica de Canarias (IAC) in Tenerife (Canary Islands, Spain) on the basis of a host agreement with INAF. Thanks to its expected overall performance, better than those of current Cherenkov telescopes' arrays for energies above \sim 5 TeV and up to 100 TeV and beyond, the ASTRI MA will represent an important instrument to perform deep observations of the Galactic and extra-Galactic sky at these energies., Comment: 19 pages, 22 figures

Employing the elemental sensitivity of x-ray absorption spectroscopy (XAS) and x-ray magnetic circular dichroism (XMCD), we study the valence and magnetic order in the heavy fermion superconductor CeCoIn$_5$. We probe spin population of the f-electrons in Ce and d-electrons in Co as a function of temperature (down to 0.1 K) and magnetic field (up to 6 T). From the XAS we find a pronounced contribution of Ce$^{4+}$ component at low temperature and a clear temperature dependence of the Ce valence below 5 K, suggesting enhanced valence fluctuations, an indication for the presence of a nearby quantum critical point (QCP). We observe no significant corresponding change with magnetic field. The XMCD displays a weak signal for Ce becoming clear only at 6 T. This splitting of the Kramers doublet ground state of Ce$^{3+}$ is significantly smaller than expected for independent but screened ions, indicating strong antiferromagnetic pair interactions. The unconventional character of superconductivity in CeCoIn$_5$ is evident in the extremely large specific heat step at the superconducting transition., Comment: 5 pages, 5 figures. We study geometrical clues of a rigidity transition due to the emergence of a system-spanning state of self stress in under-constrained systems of individual polygons and spring networks constructed from such polygons. When a polygon with harmonic bond edges and an area spring constraint is subject to an expansive strain, we observe that convexity of the polygon is a necessary condition for such a self stress. We prove that the cyclic configuration of the polygon is a sufficient condition for the self stress. This correspondence of geometry and rigidity is akin to the straightening of a one dimensional chain of springs to rigidify it. We predict the onset of the rigidity transition using a purely geometrical method. We also estimate the transition strain for a given initial configuration by approximating irregular polygons as regular polygons. These findings help determine the rigidity of an area-preserving polygon just by looking at it. Since two-dimensional spring networks can be considered as a network of polygons, we look for similar geometric features in under-constrained spring networks under isotropic expansive strain. In particular, we observe that all polygons attain convexity at the rigidity transition such that the fraction of convex, but not cyclic, polygons predicts the onset of the rigidity transition. Organoids are in vitro cellular collectives from which brain-like, or gut-like, or kidney-like structures emerge. To make quantitative predictions regarding the morphology and rheology of a cellular collective in its initial stages of development, we construct and study a three-dimensional vertex model. In such a model, the cells are represented as deformable polyhedrons with cells sharing faces such that there are no gaps between them, otherwise known as confluent. In a bulk model with periodic boundary conditions, we find a rigidity transition as a function of the target cell shape index $s_0$ with a critical value $s_0^*=5.39\pm0.01$. For a confluent cellular collective with a finite boundary, and in the presence of lateral extensile and in-plane, radial extensile deformations, we find a significant boundary-bulk effect that is one-cell layer thick. More specifically, for lateral extensile deformations, the cells in the bulk are much less aligned with the direction of the lateral deformation than the cells at the boundary. For in-plane, radial deformations, the cells in the bulk exhibit much less reorientation perpendicular to the radial direction than the cells at the boundary. In other words, for both deformations, the bulk, interior cells are topologically-protected from the deformations, at least over time scales much slower than the timescale for cellular rearrangements and up to reasonable amounts of strain. Our results provide an underlying mechanism for some observed cell shape patterning in organoids. Finally, we discuss the use of a cellular-based approach to designing organoids with new types of morphologies to study the intricate relationship between structure and function at the multi-cellular scale for example.

Both non-neural and neural biological systems can learn. So rather than focusing on purely brain-like learning, efforts are underway to study learning in physical systems. Such efforts include equilibrium propagation (EP) and coupled learning (CL), which require storage of two different states-the free state and the perturbed state-during the learning process to retain information about gradients. Inspired by slime mold, we propose a new learning algorithm rooted in chemical signaling that does not require storage of two different states. Rather, the output error information is encoded in a chemical signal that diffuses into the network in a similar way as the activation/feedforward signal. The steady state feedback chemical concentration, along with the activation signal, stores the required gradient information locally. We apply our algorithm using a physical, linear flow network and test it using the Iris data set with 93% accuracy. We also prove that our algorithm performs gradient descent. Finally, in addition to comparing our algorithm directly with EP and CL, we address the biological plausibility of the algorithm., Comment: 9 pages, 2 figures

A bar-joint mechanism is a deformable assembly of freely rotating joints connected by stiff bars. Here we develop a formalism to study the equilibration of common bar-joint mechanisms with a thermal bath. When the constraints in a mechanism cease to be linearly independent, singularities can appear in its shape space, which is the part of its configuration space after discarding rigid motions. We show that the free-energy landscape of a mechanism at low temperatures is dominated by the neighborhoods of points that correspond to these singularities. We consider two example mechanisms with shape-space singularities and find that they are more likely to be found in configurations near the singularities than others. The interface separating a liquid from its vapor phase is diffuse: the composition varies continuously from one phase to the other over a finite length. Recent experiments on dynamic jamming fronts in two dimensions [Waitukaitis et al., Europhysics Letters 102, 44001 (2013)] identified a diffuse interface between jammed and unjammed discs. In both cases, the thickness of the interface diverges as a critical transition is approached. We investigate the generality of this behavior using a third system: a model of cyclically-sheared non-Brownian suspensions. As we sediment the particles towards a boundary, we observe a diffuse traveling front that marks the interface between irreversible and reversible phases. We argue that the front width is linked to a diverging correlation lengthscale in the bulk, which we probe by studying avalanches near criticality. Disordered spring networks can exhibit rigidity transitions, due to either the removal of materials in over-constrained networks or the application of strain in under-constrained ones. While an effective medium theory (EMT) exists for the former, there is none for the latter. We, therefore, formulate an EMT for random regular networks, under-constrained spring networks with purely geometrical disorder, to predict their stiffness via the distribution of tensions. We find a linear dependence of stiffness on strain in the rigid phase and a nontrivial dependence on both the mean and standard deviation of the tension distribution. While EMT does not yield highly accurate predictions of shear modulus due to spatial heterogeneities, the noninvasiveness of this EMT makes it an ideal starting point for experimentalists quantifying the mechanics of such networks., Comment: 10 pages, 8 figures

Brain organoids recapitulate a number of brain properties, including neuronal diversity. However, do they recapitulate brain structure? Using a hydrodynamic description for cell nuclei as particles interacting initially via an effective, attractive force as mediated by the respective, surrounding cytoskeletons, we quantify structure development in brain organoids to determine what physical mechanism regulates the number of cortex-core structures. Regions of cell nuclei overdensity in the linear regime drive the initial seeding for cortex-core structures, which ultimately develop in the non-linear regime, as inferred by the emergent form of an effective interaction between cell nuclei and with the extracellular environment, as mediated by a dynamic cytoskeleton. Individual cortex-core structures then provide a basis upon which we build an extended version of the buckling without bending morphogenesis (BWBM) model, with its proliferating cortex and constraining core, to predict foliations/folds of the cortex in the presence of a nonlinearity due to cortical cells actively regulating strain. In doing so, we obtain asymmetric foliations/folds with respect to the trough (sulci) and the crest (gyri). In addition to laying new groundwork for the design of more familiar and less familiar brain structures, the hydrodynamic description for cell nuclei during the initial stages of brain organoid development provides an intriguing quantitative connection with large-scale structure formation in the universe., Comment: 13 pages, 9 figures. The ability of cells to move through small spaces depends on the mechanical properties of the cellular cytoskeleton and on nuclear deformability. In mammalian cells, the cytoskeleton is comprised of three interacting, semi-flexible polymer networks: actin, microtubules, and intermediate filaments (IF). Recent experiments of mouse embryonic fibroblasts with and without vimentin have shown that the IF vimentin plays a role in confined cell motility. We, therefore, develop a minimal model of cells moving through confined geometries that effectively includes all three types of cytoskeletal filaments with a cell consisting of an actomyosin cortex and a deformable cell nucleus and mechanical connections between the two cortices the outer actomyosin one and the inner nuclear one. By decreasing the amount of vimentin, we find that the cell speed is typically faster for vimentin-null cells as compared to cells with vimentin. Vimentin-null cells also contain more deformed nuclei in confinement. Finally, vimentin affects nucleus positioning within the cell. By positing that as the nucleus position deviates further from the center of mass of the cell, microtubules become more oriented in a particular direction to enhance cell persistence or polarity, we show that vimentin-nulls are more persistent than vimentin-full cells. The enhanced persistence indicates that the vimentin-null cells are more subjugated by the confinement since their internal polarization mechanism that depends on cross-talk of the centrosome with the nucleus and other cytoskeletal connections is diminished. In other words, the vimentin-null cells rely more heavily on external cues. Our modeling results present a quantitative interpretation for recent experiments and have implications for understanding the role of vimentin in the epithelial-mesenchymal transition., Comment: 16 pages, 13 figures

The KArlsruhe TRItium Neutrino (KATRIN) experiment, which aims to make a direct and model-independent determination of the absolute neutrino mass scale, is a complex experiment with many components. More than 15 years ago, we published a technical design report (TDR) [https://publikationen.bibliothek.kit.edu/270060419] to describe the hardware design and requirements to achieve our sensitivity goal of 0.2 eV at 90% C.L. on the neutrino mass. Since then there has been considerable progress, culminating in the publication of first neutrino mass results with the entire beamline operating [arXiv:1909.06048]. Modeling living systems at the collective scale can be very challenging because the individual constituents can themselves be complex and the respective interactions between the constituents are not fully understood. With the advent of high throughput experiments and in the age of big data, data-driven methods are on the rise to overcome these challenges. To directly uncover the underlying physical principles, we present a data-driven method for obtaining the phase-space density such that the solution to the stochastic dynamic equation for active matter readily emerges, from which time and space dependence of physical order parameters can be readily extracted. If the system is near a steady state, we illuminate how to construct a field theory to subsequently make physical predictions about the system. The method is first developed analytically and subsequently calibrated using simulated data. The method is then applied to an experimental system of particles actively driven by a {\it Serratia marcescens} bacterial swarm and in the presence of spatially localized UV light. The analysis demonstrates that the particles are in the steady-state before and sometime after the UV light and obey a Gaussian field theory with a spatially-varying "mass" in those regimes. This novel, yet simple, finding is surprising given the complex dynamics of the bacterial swarm. In response to the UV light, we demonstrate that there is a net flow of the particles away from the UV light and that the entropy of the particles increases away from the light. We conclude with a discussion of additional potential applications of our data-driven method such as when the internal structure of the individual constituents dynamically changes to result in a modified stochastic dynamic equation governing the system., Comment: 15 pages, 9 figures, comments are welcome

During morphogenesis, a featureless convex cerebellum develops folds. As it does so, the cortex thickness is thinnest at the crest (gyri) and thickest at the trough (sulci) of the folds. This observation cannot be simply explained by elastic theories of buckling. A recent minimal model explained this phenomenon by modeling the developing cortex as a growing fluid under the constraints of radially spanning elastic fibers, a plia membrane and a nongrowing sub-cortex (Engstrom, et. al., PRX 2019). In this minimal buckling without bending morphogenesis (BWBM) model, the elastic fibers were assumed to act linearly with strain. Here, we explore how nonlinear elasticity influences shape development within BWBM. The nonlinear elasticity generates a quadratic nonlinearity in the differential equation governing the system's shape and leads to sharper troughs and wider crests, which is an identifying characteristic of cerebellar folds at later stages in development. As developing organs are typically not in isolation, we also explore the effects of steric confinement, and observe flattening of the crests. Finally, as a paradigmatic example, we propose a hierarchical version of BWBM from which a novel mechanism of branching morphogenesis naturally emerges to qualitatively predict later stages of the morphology of the developing cerebellum., Comment: 12 pages, 7 figures

In dense biological tissues, cell types performing different roles remain segregated by maintaining sharp interfaces. To better understand the mechanisms for such sharp compartmentalization, we study the effect of an imposed heterotypic tension at the interface between two distinct cell types in a fully 3D model for confluent tissues. We find that cells rapidly sort and self-organize to generate a tissue-scale interface between cell types, and cells adjacent to this interface exhibit signature geometric features including nematic-like ordering, bimodal facet areas, and registration, or alignment, of cell centers on either side of the two-tissue interface. The magnitude of these features scales directly with the magnitude of imposed tension, suggesting that biologists can estimate the magnitude of tissue surface tension between two tissue types simply by segmenting a 3D tissue. To uncover the underlying physical mechanisms driving these geometric features, we develop two minimal, ordered models using two different underlying lattices that identify an energetic competition between bulk cell shapes and tissue interface area. The cell nucleus houses the chromosomes, which are linked to a soft shell of lamin filaments. Experiments indicate that correlated chromosome dynamics and nuclear shape fluctuations arise from motor activity. To identify the physical mechanisms, we develop a model of an active, crosslinked Rouse chain bound to a polymeric shell. System-sized correlated motions occur but require both motor activity {\it and} crosslinks. Contractile motors, in particular, enhance chromosome dynamics by driving anomalous density fluctuations. Nuclear shape fluctuations depend on motor strength, crosslinking, and chromosome-lamina binding. Therefore, complex chromatin dynamics and nuclear shape emerge from a minimal, active chromosome-lamina system., Comment: 18 pages, 21 figures

Cellularized tissue and polymer networks can both transition from floppy to rigid as a function of their control parameters, and, yet, the two systems often mechanically interact, which may affect their respective rigidities. To study this interaction, we consider a vertex model with interfacial tension (a spheroid) embedded in a spring network in two dimensions. We identify two regimes with different global spheroid shapes, governed by the pressure resulting from competition between interfacial tension and tension in the network. In the first regime, the tissue remains compact, while in the second, a cavitation-like instability leads to the emergence of gaps at the tissue-network interface. Intriguingly, compression of the tissue promotes fluidization, while tension promotes cellular alignment and rigidification, with the mechanisms driving rigidification differing on either side of the instability., Comment: Main text: 5 pages, 3 figures. We show how rigidity emerges in experiments of sheared frictional granular materials by using generalizations of two methods for identifying rigid structures. Both approaches, the force-based dynamical matrix and the topology-based rigidity percolation, agree with each other and identify similar rigid structures. As the system becomes jammed, at a contact number of $z=2.4\pm 0.1$, a rigid backbone interspersed with floppy, particle-filled holes of a broad range of sizes emerges, creating a sponge-like morphology. We also find that the pressure within rigid structures always exceeds the pressure outside the rigid structures, i.e. that the backbone is load-bearing. These findings shows that it is necessary to go beyond mean-field theory to capture the physics of frictional jamming and also suggests that mechanical stability arises through arch structures and hinges at the mesoscale., Comment: 19 pages, 19 figures

We report on the first detection of very high-energy (VHE) gamma-ray emission from the Crab Nebula by a Cherenkov telescope in dual-mirror Schwarzschild-Couder (SC) configuration. The result has been achieved by means of the 4 m size ASTRI-Horn telescope, operated on Mt. Etna (Italy) and developed in the context of the Cherenkov Telescope Array Observatory preparatory phase. The dual-mirror SC design is aplanatic and characterized by a small plate scale, allowing us to implement large field of view cameras with small-size pixel sensors and a high compactness. The curved focal plane of the ASTRI camera is covered by silicon photo-multipliers (SiPMs), managed by an unconventional front-end electronics based on a customized peak-sensing detector mode. The system includes internal and external calibration systems, hardware and software for control and acquisition, and the complete data archiving and processing chain. The observations of the Crab Nebula were carried out in December 2018, during the telescope verification phase, for a total observation time (after data selection) of 24.4 h, equally divided into on- and off-axis source exposure. The camera system was still under commissioning and its functionality was not yet completely exploited. Furthermore, due to recent eruptions of the Etna Volcano, the mirror reflection efficiency was reduced. Nevertheless, the observations led to the detection of the source with a statistical significance of 5.4 sigma above an energy threshold of ~3 TeV. This result provides an important step towards the use of dual-mirror systems in Cherenkov gamma-ray astronomy. We report on the neutrino mass measurement result from the first four-week science run of the Karlsruhe Tritium Neutrino experiment KATRIN in spring 2019. Beta-decay electrons from a high-purity gaseous molecular tritium source are energy analyzed by a high-resolution MAC-E filter. A fit of the integrated electron spectrum over a narrow interval around the kinematic endpoint at 18.57 keV gives an effective neutrino mass square value of $(-1.0^{+0.9}_{-1.1})$ eV$^2$. From this we derive an upper limit of 1.1 eV (90$\%$ confidence level) on the absolute mass scale of neutrinos. This value coincides with the KATRIN sensitivity. It improves upon previous mass limits from kinematic measurements by almost a factor of two and provides model-independent input to cosmological studies of structure formation.

Both animal and plant tissue exhibit a nonlinear rheological phenomenon known as compression stiffening, or an increase in moduli with increasing uniaxial compressive strain. Does such a phenomenon exist in single cells, which are the building blocks of tissues? One expects an individual cell to compression soften since the semiflexible biopolymer-based cytoskeletal network maintains the mechanical integrity of the cell and in vitro semiflexible biopolymer networks typically compression soften. To the contrary, we find that mouse embryonic fibroblasts (mEFs) compression stiffen under uniaxial compression via atomic force microscopy (AFM) studies. To understand this finding, we uncover several potential mechanisms for compression stiffening. First, we study a single semiflexible polymer loop modeling the actomyosin cortex enclosing a viscous medium modeled as an incompressible fluid. Second, we study a two-dimensional semiflexible polymer/fiber network interspersed with area-conserving loops, which are a proxy for vesicles and fluid-based organelles. Third, we study two-dimensional fiber networks with angular-constraining crosslinks, i.e. semiflexible loops on the mesh scale. In the latter two cases, the loops act as geometric constraints on the fiber network to help stiffen it via increased angular interactions. We find that the single semiflexible polymer loop model agrees well with our AFM experiments until approximately 35% compressive strain. We also find for the fiber network with area-conserving loops model that the stress-strain curves are sensitive to the packing fraction and size distribution of the area-conserving loops, thereby creating a mechanical fingerprint across different cell types. Finally, we make comparisons between this model and experiments on fibrin networks interlaced with beads as well as discuss the tissue-scale implications of cellular compression stiffening., Comment: 19 pages, 17 figures

Surface tension governed by differential adhesion can drive fluid particle mixtures to sort into separate regions, i.e., demix. Does the same phenomenon occur in confluent biological tissues? We begin to answer this question for epithelial monolayers with a combination of theory via a vertex model and experiments on keratinocyte monolayers. Vertex models are distinct from particle models in that the interactions between the cells are shape-based, as opposed to distance-dependent. We investigate whether a disparity in cell shape or size alone is sufficient to drive demixing in bidisperse vertex model fluid mixtures. Surprisingly, we observe that both types of bidisperse systems robustly mix on large lengthscales. On the other hand, shape disparity generates slight demixing over a few cell diameters, a phenomenon we term micro-demixing. This result can be understood by examining the differential energy barriers for neighbor exchanges (T1 transitions). Experiments with mixtures of wild-type and E-cadherin-deficient keratinocytes on a substrate are consistent with the predicted phenomenon of micro-demixing, which biology may exploit to create subtle patterning. The robustness of mixing at large scales, however, suggests that despite some differences in cell shape and size, progenitor cells can readily mix throughout a developing tissue until acquiring means of recognizing cells of different types., Comment: 32 pages, 17 figures

We introduce two new concepts, frictional rigidity percolation and minimal rigidity proliferation, to help identify the nature of the frictional jamming transition as well as significantly broaden the scope of rigidity percolation. For frictional rigidity percolation, we construct rigid clusters in two different lattice models using a $(3,3)$ pebble game, while taking into account contacts below and at the Coulomb threshold. The first lattice is a honeycomb lattice with next-nearest neighbors, the second, a hierarchical lattice. For both, we generally find a continuous rigidity transition. Our numerical results suggest that, for the honeycomb lattice, the exponents associated with the transition found with the frictional $(3,3)$ pebble game are distinct from those of a central-force $(2,3)$ pebble game. We propose that localized motifs, such as hinges, connecting rigid clusters that are allowed only with friction could give rise to this new frictional universality class. However, the closeness of the order parameter exponent between the two cases hints at potential superuniversality. To explore this possibility, we construct a bespoke cluster generating algorithm invoking generalized Henneberg moves, dubbed minimal rigidity proliferation. The minimally rigid clusters the algorithm generates appear to be in the same universality class as connectivity percolation, suggesting superuniversality between all three types of transitions. Finally, the hierarchical lattice is analytically tractable and we find that the exponents depend both on the type of force and on the fraction of contacts at the Coulomb threshold. These combined results allow us to compare two universality classes on the same lattice via rigid clusters for the first time to highlight unifying and distinguishing concepts within the set of all possible rigidity transitions in disordered systems., Comment: 18 pages, 16 figures

A curious feature of organ and organoid morphogenesis is that in certain cases, spatial oscillations in the thickness of the growing "film" are out-of-phase with the deformation of the slower-growing "substrate," while in other cases, the oscillations are in-phase. The former cannot be explained by elastic bilayer instability, and contradict the notion that there is a universal mechanism by which brains, intestines, teeth, and other organs develop surface wrinkles and folds. Inspired by the microstructure of the embryonic cerebellum, we develop a new model of 2d morphogenesis in which system-spanning elastic fibers endow the organ with a preferred radius, while a separate fiber network resides in the otherwise fluid-like film at the outer edge of the organ and resists thickness gradients thereof. The tendency of the film to uniformly thicken or thin is described via a "growth potential". Several features of cerebellum, +blebbistatin organoid, and retinal fovea morphogenesis, including out-of-phase behavior and a film thickness amplitude that is comparable to the radius amplitude, are readily explained by our simple analytical model, as may be an observed scale-invariance in the number of folds in the cerebellum. We also study a nonlinear variant of the model, propose further biological and bio-inspired applications, and address how our model is and is not unique to the developing nervous system., Comment: version accepted by Physical Review X

We define a class of quantum linear Galois algebras which include the universal enveloping algebra Uq(gln), the quantum Heisenberg Lie algebra and other quantum orthogonal Gelfand-Zetlin algebras of type A, the subalgebras of G-invariants of the quantum affine space, quantum torus for G = G(m, p, n), and of the quantum Weyl algebra for G = Sn. We show that all quantum linear Galois algebras satisfy the quantum Gelfand-Kirillov conjecture. Moreover, it is shown that the the subalgebras of invariants of the quantum affine space and of quantum torus for the reflection groups and of the quantum Weyl algebra for symmetric groups are, in fact, Galois orders over an adequate commutative subalgebras and free as right (left) modules over these subalgebras. In the rank 1 cases the results hold for an arbitrary finite group of automorphisms when the field is C.

Compression stiffening, or an increase in shear modulus with increasing compressive strain, has been observed in recent rheometry experiments on brain, liver, and fat tissues. Here, we extend the known types of biomaterials exhibiting this phenomenon to include agarose gel and fruit flesh. Further, we show that two different results from classic elasticity theory can account for the phenomenon of linear compression stiffening. One result is due to Barron and Klein, extended here to the relevant geometry and pre-stresses; the other is due to Birch. For incompressible materials, there are no adjustable parameters in either theory. Which one applies to a given situation is a matter of reference state, suggesting that the reference state is determined by the tendency of the material to develop, or not develop, axial stress (in excess of the applied pre-stress) when subjected to torsion at constant axial strain. Our experiments and analysis also strengthen the notion that seemingly distinct animal and plant tissues can have mechanically similar behavior under certain conditions., Comment: 7 pages, 1 figure

Particle suspensions, present in many natural and industrial settings, typically contain aggregates or other microstructures that can complicate macroscopic flow behaviors and damage processing equipment. Recent work found that applying uniform periodic shear near a critical transition can reduce fluctuations in the particle concentration across all length scales, leading to a hyperuniform state. However, this strategy for homogenization requires fine tuning of the strain amplitude. Here we show that in a model of sedimenting particles under periodic shear, there is a well-defined regime at low sedimentation speed where hyperuniform scaling automatically occurs. Our simulations and theoretical arguments show that the homogenization extends up to a finite lengthscale that diverges as the sedimentation speed approaches zero., Comment: 11 pages, 6 figures

How can dense biological tissue maintain sharp boundaries between coexisting cell populations? We explore this question within a simple vertex model for cells, focusing on the role of topology and tissue surface tension. We show that the ability of cells to independently regulate adhesivity and tension, together with neighbor-based interaction rules, lets them support strikingly unusual interfaces. In particular, we show that mechanical- and fluctuation-based measurements of the effective surface tension of a cellular aggregate yield different results, leading to mechanically soft interfaces that are nevertheless extremely sharp.

Harnessing a model from composite materials science, we show how point-like cusped surface features arise as quasi-particle excitations, termed "ghost fibers", on the surface of a homogeneous soft elastic material. These deformations appear above a critical compressive strain at which ghost fiber dipoles unbind, analogous to vortices in the Kosterlitz-Thouless transition. Finite-length creases can be described in the same framework. Our predictions for crease surface profiles and onset strain agree with previous experiments and simulations, and further experimental tests are proposed., Comment: 5 pages, 3 figures

Cell migration in morphogenesis and cancer metastasis typically involves interplay between different cell types. We construct and study a minimal, one-dimensional model comprised of two different motile cells with each cell represented as an active elastic dimer. The interaction between the two cells via cadherins is modeled as a spring that can rupture beyond a threshold force as it undergoes dynamic loading via the attached motile cells. We obtain a phase diagram consisting of chase-and-run dynamics and clumping dynamics as a function of the stiffness of the interaction spring and the threshold force. We also find that while feedback between cadherins and cell-substrate interaction via integrins accentuates the chase-run behavior, feedback is not necessary for it.

The KATRIN experiment will probe the neutrino mass by measuring the beta-electron energy spectrum near the endpoint of tritium beta-decay. An integral energy analysis will be performed by an electro-static spectrometer (Main Spectrometer), an ultra-high vacuum vessel with a length of 23.2 m, a volume of 1240 m^3, and a complex inner electrode system with about 120000 individual parts. The strong magnetic field that guides the beta-electrons is provided by super-conducting solenoids at both ends of the spectrometer. Its influence on turbo-molecular pumps and vacuum gauges had to be considered. A system consisting of 6 turbo-molecular pumps and 3 km of non-evaporable getter strips has been deployed and was tested during the commissioning of the spectrometer. In this paper the configuration, the commissioning with bake-out at 300{\deg}C, and the performance of this system are presented in detail. The vacuum system has to maintain a pressure in the 10^{-11} mbar range. It is demonstrated that the performance of the system is already close to these stringent functional requirements for the KATRIN experiment, which will start at the end of 2016., Comment: submitted for publication in JINST, 39 pages, 15 figures

Hyperbolic lattices interpolate between finite-dimensional lattices and Bethe lattices and are interesting in their own right with ordinary percolation exhibiting not one, but two, phase transitions. We study four constraint percolation models---$k$-core percolation (for $k=1,2,3$) and force-balance percolation---on several tessellations of the hyperbolic plane. By comparing these four different models, our numerical data suggests that all of the $k$-core models, even for $k=3$, exhibit behavior similar to ordinary percolation, while the force-balance percolation transition is discontinuous. We also provide a proof, for some hyperbolic lattices, of the existence of a critical probability that is less than unity for the force-balance model, so that we can place our interpretation of the numerical data for this model on a more rigorous footing. Finally, we discuss improved numerical methods for determining the two critical probabilities on the hyperbolic lattice for the $k$-core percolation models., Comment: 10 pages, 15 figures

The Cherenkov Telescope Array (CTA) is the next-generation atmospheric Cherenkov gamma-ray observatory. CTA will consist of two installations, one in the northern, and the other in the southern hemisphere, containing tens of telescopes of different sizes. The CTA performance requirements and the inherent complexity associated with the operation, control and monitoring of such a large distributed multi-telescope array leads to new challenges in the field of the gamma-ray astronomy. The ACTL (array control and data acquisition) system will consist of the hardware and software that is necessary to control and monitor the CTA arrays, as well as to time-stamp, read-out, filter and store -at aggregated rates of few GB/s- the scientific data. The ACTL system must be flexible enough to permit the simultaneous automatic operation of multiple sub-arrays of telescopes with a minimum personnel effort on site. One of the challenges of the system is to provide a reliable integration of the control of a large and heterogeneous set of devices. Moreover, the system is required to be ready to adapt the observation schedule, on timescales of a few tens of seconds, to account for changing environmental conditions or to prioritize incoming scientific alerts from time-critical transient phenomena such as gamma ray bursts.
