Abstract We calculate several diagonal and non-diagonal fluctuations of conserved charges in a system of 2+1+1 quark flavors with physical masses, on a lattice with size 483 × 12. Higher order fluctuations at μ B = 0 are obtained as derivatives of the lower order ones, simulated at imaginary chemical potential. From these correlations and fluctuations we construct ratios of net-baryon number cumulants as functions of temperature and chemical potential, which satisfy the experimental conditions of strangeness neutrality and proton/baryon ratio. Our results qualitatively explain the behavior of the measured cumulant ratios by the STAR collaboration.

BackgroundGlucose-6-phosphate isomerase and collagen type II antibody-induced arthritis models (K/BxN and CAIA, respectively) have an inflammatory and a post-inflammatory phase. Both phases display robust tactile allodynia. In previous work, inflammatory phase allodynia was reversed by gabapentin and ketorolac, whereas in late phase only gabapentin was effective. Here, we sought to determine if the effects of these two drugs during the early and late phases of the two arthritis models were observed in the conditioned place preference (CPP) paradigm, indicating a differential drug effect on the aversive state.MethodsMale C57BL/6 mice received K/BxN serum intraperitoneally, while male BALB/c mice received collagen type II antibody cocktail intravenously. After onset of inflammation and allodynia, we assessed effects of i.p. gabapentin (100 mg/kg) or ketorolac (15 mg/kg) using a CPP paradigm: 2 days adaptation, 2 days conditioning (vehicle in morning and drug in afternoon), preference testing on day 5.ResultsConsistent with the effects upon allodynia, both gabapentin and ketorolac produced a preference for the drug-paired compartment in the early phase of the K/BxN model, while gabapentin, but not ketorolac, resulted in a place preference during late phase. In the CAIA model, consistent with differential effects upon allodynia, gabapentin produced a preference in the early phase and a trend in the late phase, whereas ketorolac was ineffective at either time.ConclusionsCPP validated the aversive state in the inflammatory and post-inflammatory phases of the K/BxN and CAIA arthritis models and correspondence between the anti-hyperpathic pharmacology as defined by thresholds and CPP.

Here we explore the role of spinal TNF and JNK in carrageenan-induced hypersensitivity. As links between TNF and JNK have been demonstrated in vitro, we investigated if TNF regulates spinal JNK activity in vivo.MethodsTNF levels in lumbar cerebrospinal fluid (CSF) were measured by enzyme-linked immunosorbent assay, spinal TNF gene expression by real-time polymerase chain reaction and TNF protein expression, JNK and c-Jun phosphorylation by western blotting. The role of spinal TNF and JNK in inflammation-induced mechanical and thermal hypersensitivity was assessed by injecting the TNF inhibitor etanercept and the JNK inhibitors SP600125 and JIP-1 intrathecally (i.t.). TNF-mediated regulation of JNK activity was examined by assessing the effect of i.t. etanercept on inflammation-induced spinal JNK activity.ResultsTNF levels were increased in CSF and spinal cord following carrageenan-induced inflammation. While JNK phosphorylation followed the same temporal pattern as TNF, c-jun was only activated at later time points. Intrathecal injection of TNF and JNK inhibitors attenuated carrageenan-induced mechanical and thermal hypersensitivity. TNF stimulation induced JNK phosphorylation in cultured spinal astrocytes and blocking the spinal actions of TNF in vivo by i.t. injection of etanercept reduced inflammation-induced spinal JNK activity.ConclusionsHere we show that spinal JNK activity is dependent on TNF and that both TNF and the JNK signalling pathways modulate pain-like behaviour induced by peripheral inflammation.

Abstract Biofilms are mucilaginous-organic layers produced by microbial activity including viruses. Growing biofilms form microbial mats which enhance sediment stability by binding particles with extracellular polymeric substances and promoting growth through nutrient cycling and organic matter accumulation. They preferentially develop at the sediment-water interface of both marine and non-marine environments, and upon the growing surfaces of modern tufa and travertine. In this context, however, little is known about the factors, environmental or anthropogenic, which affect viral communities in freshwater spring settings. To explore this issue, geochemical and metagenomic data were subjected to multidimensional analyses (Principal Component Analysis, Classical Multidimensional Scaling, Partial Least Squares analysis and cluster analysis based on beta-diversity), and these show that viral composition is specific and dependent on environment. Indeed, waters precipitating tufa and travertine do vary in their geochemistry with their viruses showing distinct variability between sites. These differences between virus groups allow the formulation of a viral proxy, based on the Caudoviricetes/Megaviricetes ratio established on the most abundant groups of viruses. This ratio may be potentially used in analysing ancient DNA preserved in carbonate formations as an additional source of information on the microbiological community during sedimentation.

Sustainable crop cultivation is a crucial goal in modern agriculture, aiming to attain high productivity while conserving natural resources. This requires the implementation of rational cultivation techniques, with proper irrigation and fertilization practices playing a crucial role in ensuring plant well-being and providing natural protection against biotic and abiotic stresses. The health-promoting properties of crops are also significantly influenced by irrigation and fertilization. This study investigated the productivity, nutritional composition, bioactive compounds, and antioxidant capacity of Vineta early potato cultivar tubers under drip irrigation conditions, combined with nitrogen fertilization through fertigation or broadcasting. Two-factor trials included drip irrigation (control or drip) and nitrogen application (broadcast or fertigation). Precise treatments, such as drip irrigation and N-fertigation were found to enhance all productivity traits. Both practices positively impacted tuber nutrient content. The highest levels of total polyphenols and chlorogenic acid were observed in non-irrigated and broadcasted tubers. Drip-irrigated and N-fertigated tubers exhibited high levels of vitamin C and antioxidant activity. In summary, the combination of drip irrigation with appropriate fertilization methods positively influenced potato crops, as evidenced by an increase in productivity and the quality of tubers.

Material extrusion (MEX) is one of the most widely used additive manufacturing techniques these days. This study investigates how the properties of MEX 3D-printed objects depend on the relative humidity (RH) conditions in which filaments are stored before and during the manufacturing process. Poly(lactic acid) (PLA) filament was drawn directly from a humidity-controlled chamber into the MEX 3D printer's nozzle. For each set of samples, the filaments were conditioned under different RH conditions, ranging from 10 % to 90 %. The macrostructure of the fabricated products was characterized using computed tomography, revealing increased porosity at higher RH values (from 0.84 % to 4.42 %). The increased porosity at higher storage RH is attributed to under-extrusion and volatile entrapment due to excess moisture. With growing storage RH, the melt flow rate of PLA also gradually increased, indicating a plasticizing effect of humidity on the biopolymer. Gel permeation chromatography and differential scanning calorimetry analyses were conducted to determine whether hydrolytic chain scission took place when PLA was processed in the presence of excessive moisture. Neither measurement indicated any considerable alteration in molecular integrity and crystalline structure as a function of storage RH. Mechanical tests, however, revealed a reduced load-bearing capacity of the manufactured PLA specimens. Flexural strength decreased from 103.0 to 99.6 MPa, and the impact strength dropped from 18.2 to 16.2 kJ/m2, which is ascribed to the increasing size of pores inside the specimens with increasing storage RH. These findings should be taken into account when designing and processing PLA products by MEX-based additive manufacturing.

Habitat loss and forest fragmentation are often linked to increased pathogen transmission, but the extent to which habitat isolation and landscape connectivity affect disease dynamics through movement of disease vectors and reservoir hosts has not been well examined. Tick-borne diseases are the most prevalent vector-borne diseases in the United States and on the West Coast, Ixodes pacificus is one of the most epidemiologically important vectors. We investigated the impacts of habitat fragmentation on pathogens transmitted by I. pacificus and sought to disentangle the effects of wildlife communities and landscape metrics predictive of pathogen diversity, prevalence and distribution. We collected pathogen data for four co-occurring bacteria transmitted by I. pacificus and measured wildlife parameters. We also used spatial data and cost-distance analysis integrating expert opinions to assess landscape metrics of habitat fragmentation. We found that landscape metrics were significant predictors of tick density and pathogen prevalence. However, wildlife variables were essential when predicting the prevalence and distribution of pathogens reliant on wildlife reservoir hosts for maintenance. We found that landscape structure was an informative predictor of tick-borne pathogen richness in an urban matrix. Our work highlights the implications of large-scale land management on human disease risk.

Abstract Soybean (Glycine max L.) is an important leguminous plant, in which pests trigger significant damage every year. Important members of this community are insects with piercing‐sucking mouthpart, especially the southern green stinkbug, Nezara viridula L.. This insect with its extraoral digestion causes visible alterations (morphological and color changes) in the seeds. We aimed to obtain precise information about the extent and nature of damage in soybeans caused by N. viridula using nondestructive imaging methods. Two infestation conditions were applied: one with controlled numbers of pests (six insects/15 pods) and another with naturally occurring pests (samples collected from the apical part of the plant and samples from whole plants). An intact control group was also included, resulting in four treatment groups. Seed samples were analyzed by computed tomography (CT) and image color analysis under laboratory conditions. According to our CT findings, the damage caused by N. viridula changed the radiodensity, volume, and shape (Solidity) of the soybean seeds during the pod‐filling and maturing period. Radiodensity was significantly reduced in all three damaged categories compared to the intact sample; the mean radiodensity reduction range was 49–412 HU. The seed volume also decreased significantly (25%–80% decrease), with a threefold reduction for samples exposed to regulated damage compared to natural ones. The samples exposed to natural damage showed significant but minor reduction in solidity, while samples exposed to regulated damage showed a prominent decrease (~12%). Image color analysis showed that the damaged samples were well distinguishable, and the differences were statistically verifiable. The achieved data derived from our external and internal imaging approaches contribute to a better understanding of the internal chemical processes, and CT analysis helps to understand the alteration trends of the hidden structure of seeds caused by a pest. Our results can contribute to the development of a practically applicable system based on image analysis, which can identify lots damaged by insects.

Freshwater crayfish are amongst the largest macroinvertebrates and play a keystone role in the ecosystems they occupy. Understanding the global distribution of these animals is often hindered due to a paucity of distributional data. Additionally, non-native crayfish introductions are becoming more frequent, which can cause severe environmental and economic impacts. Management decisions related to crayfish and their habitats require accurate, up-to-date distribution data and mapping tools. Such data are currently patchily distributed with limited accessibility and are rarely up-to-date. To address these challenges, we developed a versatile e-portal to host distributional data of freshwater crayfish and their pathogens (using Aphanomyces astaci, the causative agent of the crayfish plague, as the most prominent example). Populated with expert data and operating in near real-time, World of Crayfish™ is a living, publicly available database providing worldwide distributional data sourced by experts in the field. The database offers open access to the data through specialized standard geospatial services (Web Map Service, Web Feature Service) enabling users to view, embed, and download customizable outputs for various applications. The platform is designed to support technical enhancements in the future, with the potential to eventually incorporate various additional features. This tool serves as a step forward towards a modern era of conservation planning and management of freshwater biodiversity.

ABSTRACT Survival factor 1 (Svf1) protein has been described in some ascomycetous fungi where it was found to be contributing to several essential physiological processes, such as response to osmotic, oxidative and cold stresses, sphingolipid biosynthesis, morphogenesis, sporulation, antifungal resistance, and pathogenicity. It was also suggested that it can be a novel central regulator affecting the expression of various genes. In the present study, function of this protein and the encoding genes is described for the first time in a fungus (i.e., in Mucor lusitanicus) belonging to the order Mucorales. M. lusitanicus has two putative svf1 genes named svf1a and svf1b. Expression of both genes was proven. Although the expression of svf1a was affected by several environmental stresses and knocking out the gene affected adaptation to low temperatures and the sporulation ability, the main survival factor functions, such as participation in the maintenance of the viability, the response to oxidative and cold stresses, and the sphingolipid biosynthesis, could be associated with Svf1b, suggesting a central regulatory role to this protein. Interestingly, knockout of both genes affected the pathogenicity of the fungus in a Drosophila model.IMPORTANCEMucor lusitanicus is a widely used model organism to study various biological processes in the basal fungal group Mucorales. Several members of this group can be agents of mucormycosis, an opportunistic fungal infection, which is associated with high mortality, rapid progression, and wide resistance to the commonly used antifungal agents. Svf1 proteins have so far only been identified in fungi, where they have been involved in pathogenicity and resistance to antifungal agents in many cases. Only a limited number of factors affecting the stress response, antifungal resistance, and virulence of Mucorales fungi have been revealed. Elucidating the function of a fungus-specific protein that may regulate these processes may bring us closer to understanding the pathogenesis of these fungi.

European species of the Cochylimorpha perfusana (Guenée, 1845) species group are revised. Four species are recognized, all closely related to C. perfusana (Guenée, 1845) with which they have previously been confused. Based on morphological differences and DNA barcode data, C. callosana (Herrich-Schäffer, 1856), sp. rev. and C. dorsimaculana (Preissecker, 1908), sp. rev., are re-instated to their original species rank. Inconsistencies in their taxonomy leading to their synonymy are shown and corrected. Cochylimorpha bucegiana Z. Kovács, S. Kovács & P. Buchner, sp. nov. is described from the subalpine meadows of the Bucegi and Făgăraș Mountains (Southern Carpathians, Romania), and the other three species are redescribed. Adults and genitalia of both sexes of all four, and the habitat of three taxa are figured. The geographic distribution of each species is re-evaluated based on the examined material and a critical analysis of the literature. Literature data for which no material could be found for examination are treated as doubtful and in need of confirmation. Cochylimorpha perfusana is widely distributed in mountain areas of Europe (Alps, Carpathians and Pirin Mountains), C. callosana is widespread only in the north-western Balkan Peninsula and north-eastern Italy, and local in eastern France, Corsica and Hungary, the other two seem to be endemics, C. dorsimaculana to Wachau and Retz in Lower Austria and C. bucegiana sp. nov. to the Southern Carpathians in Romania. Cochylimorpha perfusana is recorded for the first time from Bulgaria; the first confirmed records of C. callosana from Slovenia and Hungary are given; and in the north-eastern Italian fauna C. callosana replaces C. perfusana. Records of C. perfusana from Ukraine, the Czech Republic, Slovenia, and north-western Italy require confirmation.

Abstract Objective Examining the distribution of breast cancer (BC) stage and molecular subtype among women aged below ( 65 years) the recommended screening age range helps to understand the screening program's characteristics and contributes to enhancing the effectiveness of BC screening programs. Methods In this retrospective study, female patients with newly diagnosed BC from 2010 to 2020 were identified. The distribution of cases in terms of TNM stages, severity classes, and subtypes was analysed according to age groups. Results A total of 3282 women diagnosed with BC were included in the analysis. Among these cases 51.4% were detected outside the screening age group, and these were characterized by a higher TNM stage compared to those diagnosed within the screening age band. We observed significantly higher relative frequency of advanced BC in the older age group compared to both the screening age population and women younger than 45 years (14.9% vs. 8.7% and 7.7%, P

Understanding the recombination lifetime of charge carriers (τc) is essential for the diverse applications of photovoltaic materials, such as perovskites. The study on the inorganic perovskite, CsPbBr3, reveals recombination dynamics exceeding 1 ms below 200 K and τc approaching 100 μs at room temperature. Utilizing time‐resolved microwave‐detected photoconductivity decay in conjunction with injection dependence, it is found that τc is dominated by impurity charge trapping. The observed injection dependence is well corroborated by modeling of the trap mechanism. The ultralong decay time is also consistent with photoconductivity measurements with a continuous‐wave excitation at powers corresponding to around 1 Sun irradiation. While charge‐carrier trapping may, in theory, impose limitations on the photovoltaic efficiency of single‐cell devices, it can also contribute to increased efficiency in tandem cells and find applications in photodetection, photocatalysis, and quantum information storage.

The purpose of this research was to develop “green” materials by combining poly(lactic acid) (PLA) with two agricultural by-products, namely flax seed meal (FSM) and rapeseed straw (RSS). The natural fillers (0–20 wt.%) were mixed with PLA through extrusion and then injection molded into specimens. The samples were analyzed for their thermal, morphological, mechanical, and physical features and biodegradability. Thermal properties and crystallinity were analyzed using Differential Scanning Calorimetry (DSC), while the morphology was investigated by Scanning Electron Microscopy (SEM). Mechanical properties were characterized through tensile, flexural, and impact measurements, while surface hardness was evaluated by Shore D tests. Water absorption and biodegradability of the samples were also examined. DSC measurements revealed a nucleating effect of both bio-fillers. Based on the tensile tests, major improvement in stiffness was found with the biocomposites having up to ∼16 % higher Young's modulus than neat PLA (2.5 GPa). It came, however, at the cost of tensile strength, which decreased from 56 to 51 MPa even in the presence of the lowest amount (2.5 wt.%) of FSM. Loss in strength was due to the limited adhesion between the components, as also supported by SEM images. The hardness slightly (1–2 %) improved in the presence of even 2.5 wt.% bio-filler and it remained at that level at higher filler loading as well. Laboratory-scale composting revealed that both fillers facilitated biodegradation with FSM being superior. In the presence of 10–20 wt.% FSM, the rate of decomposition was found to be twice as fast compared to neat PLA.

The two naphthol isomers were investigated in different organic solvents by taking cyclic voltammograms, and fouling took place on a platinum electrode surface, except for dimethyl sulfoxide and dimethyl formamide. Studies in allyl alcohol rarely used in electrochemical investigations pointed to the importance of the carbon–carbon double bond as electrode deactivation was remarkably faster compared with its saturated analog solvent. Similarly, the use of the other unsaturated solvent mesityl oxide in the electropolymerization of naphthols resulted in different findings compared with methyl isobutyl ketone. As dimethyl formamide was the best choice concerning the solubility of products, it was successfully tested in electrode renewal after deactivation in an aqueous solution. The increase in dimethyl formamide content led to more and more improved reproducibility of the currents of the outlined aromatic compounds. Naphthol isomers were assessed in the suppression of layer growth originating from the electrooxidation of another monomer phloroglucinol. Its simultaneous electrooxidation with naphthol monomers had a dramatic effect on layer morphology and it was found that instead of a coherent organic layer originating from the homopolymerization of phloroglucinol, the copolymerization with naphthols led to the development of more porous and rougher deposits. The suppressed electropolymerization thus increased sensitivity towards a chosen redox active compound, 4-methoxyphenol.

Abstract Antimicrobial resistance (AMR) is a major public health threat, reducing treatment options for infected patients. AMR is promoted by a lack of access to rapid antibiotic susceptibility tests (ASTs). Accelerated ASTs can identify effective antibiotics for treatment in a timely and informed manner. We describe a rapid growth-independent phenotypic AST that uses a nanomotion technology platform to measure bacterial vibrations. Machine learning techniques are applied to analyze a large dataset encompassing 2762 individual nanomotion recordings from 1180 spiked positive blood culture samples covering 364 Escherichia coli and Klebsiella pneumoniae isolates exposed to cephalosporins and fluoroquinolones. The training performances of the different classification models achieve between 90.5 and 100% accuracy. Independent testing of the AST on 223 strains, including in clinical setting, correctly predict susceptibility and resistance with accuracies between 89.5% and 98.9%. The study shows the potential of this nanomotion platform for future bacterial phenotype delineation.

The authors, as practising law enforcement experts, examine the historical antecedents of the current system of Courts of Honour in Hungary, which are not bound by previous legislation and norms. They look into the past and present of the commissions, which were not composed of professional judges in the past, but which are still deciding on the service relationships of professional members of law enforcement bodies, which were once established to investigate cases concerning the honour of a body or community with the aim of protecting the honour and reputation of the body. Travelling through time, from the 13th century penitential regulation of the Order of St John through the Visegrád curia militaris of Charles I, established for the internal investigation of knightly affairs and dissensions, to the “officer’s honour” of the military bodies of the Kingdom of Hungary, and then analysing the functioning of the socialist courts of honour after the Second World War, the article arrives at the legal institution of the courts of honour of today, regulated by the law of the service. In the words of Gusztáv Fabinyi, we point out that, although its institutions have changed from era to era, its purpose has always remained the same: to serve material justice in the truest sense of the word, to cultivate, strengthen, and establish the military spirit, chivalry, and nobility of thought in the officer corps – to reprimand in minor cases and to remove permanently from the corps in more serious cases those individuals whose conduct is contrary to the feelings of the educated and noble-minded individual.

Abstract Subjective cognitive complaints (SCC) is a self-reported experience of persistently impaired cognitive functions which could be the earliest red flag of neurocognitive disorders. The COVID-19 pandemic and related restriction measures changed the lifestyle and behaviour of older adults. The aim of this study was to assess the relation of these changes and SCC status in Hungary. This cross-sectional study analysed the data of 359 elderly Hungarians who filled out the WW-FINGERS-SARS-CoV2 survey. A quarter of the respondents (n:88) reported SCC in connection with the pandemic. We compared sociodemographic features, health status, lifestyle, and social life parameters between subjects with reported SCC and without. To eliminate the potential interrelation across group differences, stepwise logistic regression was applied. Participants with SCC showed the following characteristics, compared to individuals without: (1) they were older; (2) they were more likely to be women; (3) they had a higher number of chronic disorders; (4) showed more prominent impairment in physical mobility; (5) had worse sleep quality; (6) spent less time with family; and (7) used internet more frequently during the pandemic (all p’s

Abstract Background The measurement of root dielectric response is a useful non-destructive method to evaluate root growth and function. Previous studies tracked root development throughout the plant growing cycle by single-time electrical measurements taken repeatedly. However, it is known that root conductivity and uptake activity can change rapidly, coupled with the day/night cycles of photosynthetic and transpiration rate. Therefore, the low-frequency dielectric monitoring of intact root–substrate systems at minute-scale temporal resolution was tested using a customized impedance measurement system in a laboratory environment. Electrical capacitance (CR) and conductance (GR) and the dissipation factor (DR) were detected for 144 h in potted maize, cucumber and pea grown under various light/dark and temperature conditions, or subjected to progressive leaf excision or decapitation. Photosynthetic parameters and stomatal conductance were also measured to evaluate the stress response. Results The CR and GR data series showed significant 24-h seasonality associated with the light/dark and temperature cycles applied. This was attributed to the diurnal patterns in whole-plant transpiration (detected via stomatal conductance), which is strongly linked to the root water uptake rate. CR and GR decreased during the 6-day dark treatment, and dropped proportionally with increasing defoliation levels, likely due to the loss of canopy transpiration caused by dark-induced senescence or removal of leaves. DR showed a decreasing trend for plants exposed to 6-day darkness, whereas it was increased markedly by decapitation, indicating altered root membrane structure and permeability, and a modified ratio of apoplastic to cell-to-cell water and current pathways. Conclusions Dynamic, in situ impedance measurement of the intact root system was an efficient way of following integrated root water uptake, including diurnal cycles, and stress-induced changes. It was also demonstrated that the dielectric response mainly originated from root tissue polarization and current conduction, and was influenced by the actual physiological activity of the root system. Dielectric measurement on fine timescale, as a diagnostic tool for monitoring root physiological status and environmental response, deserves future attention.

We developed and applied a new calibration method to make more accurate measurements with our multi-color ellipsometric mapping tool made from cheap parts. Ellipsometry is an optical technique that measures the relative change in the polarization state of the measurement beam induced by reflection from or transmission through a sample. During conventional ellipsometric measurement, the data collection is relatively slow and measures one spot at a time, so mapping needs a long time compared with our new optical mapping equipment made by an ordinary color LED monitor and a polarization-sensitive camera. The angle of incidence and the incident polarization state is varied point by point, so a special optical calibration method is needed. Three SiO2 samples with different thicknesses were used for the point-by-point determination of the angle of incidence and rho (ρ) corrections. After the calibration, another SiO2 sample was measured and analyzed using the calibrated corrections; further, this sample was independently measured using a conventional spectroscopic ellipsometer. The difference between the two measured thickness maps is less than 1 nm. Our optical mapping tool made from cheap parts is faster and covers wider area samples relative to conventional ellipsometers, and these correction enhancements further demonstrate its performance.

The measurement and description of the charge-carrier lifetime (τc) is crucial for the wide-ranging applications of lead-halide perovskites. We present time-resolved microwave-detected photoconductivity decay (TRMCD) measurements and a detailed analysis of the possible recombination mechanisms including trap-assisted, radiative, and Auger recombination. We prove that performing injection-dependent measurement is crucial in identifying the recombination mechanism. We present temperature and injection level dependent measurements in CsPbBr3, which is the most common inorganic lead-halide perovskite. In this material, we observe the dominance of charge-carrier trapping, which results in ultra-long charge-carrier lifetimes. Although charge trapping can limit the effectiveness of materials in photovoltaic applications, it also offers significant advantages for various alternative uses, including delayed and persistent photodetection, charge-trap memory, afterglow light-emitting diodes, quantum information storage, and photocatalytic activity.

IntroductionThe field of Medicines Development faces a continuous need for educational evolution to match the interdisciplinary and global nature of the pharmaceutical industry. This paper discusses the outcomes of a 7-year collaboration between King’s College London and the Global Medicines Development Professionals (GMDP) Academy, which aimed to address this need through a blended e-learning program.MethodsThe collaboration developed a comprehensive curriculum based on the PharmaTrain syllabus, delivered through a combination of asynchronous and synchronous e-learning methods. The program targeted a diverse range of professionals serving in areas related to Medical Affairs.ResultsOver seven annual cohorts, 682 participants from eighty-six countries were enrolled in the program. The program’s effectiveness was assessed using Kirkpatrick’s model, showing elevated levels of satisfaction (over 4.0 on a five-point scale), suggesting significant gains in competence at the cognitive level and leveraged performance. Notably, 70% of responding alumni reported significant improvement in their functions, corroborated by 30% of their supervisors. The further long-term impact of the program on their respective organization has not been established.DiscussionThe GMDP Academy’s program has significantly contributed to life-long learning in Medicines Development, addressing educational gaps and fostering interdisciplinary collaboration. Its success highlights the importance of continuous education in keeping pace with the industry’s evolving demands and underscores the potential of blended learning in achieving educational objectives in pharmaceutical medicine.

With the emergence of modern food supply chains, there has been a noticeable decline in consumer trust and an increase in information asymmetry. Short food supply chains, including farmers' markets, offer potential solutions to these issues. Currently, farmers' markets are primarily found in the United States and the European Union, and their impact on sustainability has gained significant attention. However, the relevance of this traditional approach within modern supply chains remains largely unexplored. Thus, this study aims to examine the existing literature on farmers' markets using bibliometric techniques applied to 1765 documents sourced from the Scopus and Web of Science databases spanning from 1955 to 2022. The paper tracks the research trends associated with farmers' markets by identifying the stages of evolution of key topics, articles, journals, author citations, and co-citation networks. The findings demonstrate an increasing trend in publication of papers on this subject, highlight five interconnected areas of market research, and provide a foundation for future research and policy making by outlining the main and specific research avenues to explore.

A well-known insect phenomenon is colour polymorphism, from which the species hopes to gain some ecological advantage. Herbivores adapt to the colours of their environment to reduce their chances of becoming prey, while predators use camouflage colours to succeed in hiding from prey. The larvae of the cosmopolitan, highly invasive cotton bollworm (Helicoverpa armigera Hbn., Lep.: Noctuidae) also benefit from this morphological polymorphism. Their fully-developed larvae can take on various colours when feeding on foliage. Our study aimed to determine the host plant-related colour adaptation of H. armigera larvae collected from different plant organs using different spectral analysis methods. Our studies, based on colour analysis of photographic images, showed that the colour of fully-developed larvae of the species is highly correlated with the colour of the damaged nutrient plant. The dominant colours of the larvae show a high similarity with the colours of the consumed host plant parts. The RGB-based analysis confirmed that larval colours are clustered according to the host plant’s organs and are mainly located in the yellow and green regions of the visible light spectrum. All these results confirm the extraordinary adaptive capabilities of this invasive species, which, among other things, verify its worldwide distribution.

Abstract Isomerization Metathesis (ISOMET) reaction is an emerging tool for “open loop” chemical recycling of polyethylene to propylene. Novel, latent N‐Alkyl substituted Cyclic(Alkyl)(Amino)Carbene (CAAC)–ruthenium catalysts (5a‐Ru, 3b‐Ru – 6c‐Ru) are developed rendering “inverted” chemical structure while showing enhanced ISOMET activity in combination with (RuHCl)(CO)(PPh3)3 (RuH) double bond isomerization co‐catalyst. Systematic investigations reveal that the steric hindrance of the substituents on nitrogen and carbon atom adjacent to carbene moiety in the CAAC ligand have significantly improved the catalytic activity and robustness. In contrast to the NHC‐Ru and CAAC‐Ru catalyst systems known so far, these systems show higher isomerization metathesis (ISOMET) activity (TON: 7400) on the model compound 1‐octadecene at as low as 3.0 bar optimized pressure, using technical grade (3.0) ethylene. The propylene content formed in the gas phase can reach up to 20% by volume.

Introduction: The 21-gene analysis (OncotypeDX) is validated test for pT1-3, pN0-1 with hormone receptor (HR) positive and normal expression of human epidermal growth factor receptor-2 (HER2) breast cancer (BC) to determine the aggressiveness of the disease based on the calculation of Recurrence Score (RS).Methods: In this retrospective study the authors correlated pathological characteristics and Recurrence Score (RS) by traditional statistical methods and Observed Oriented Modeling (OOM) in a realistic cohort of BC patients.Results: OncotypeDX tests were performed in 94 tumour specimens of 90 BC patients. >83% of node-negative (pN0) and >72% of node-positive (pN1) cases could avoid chemotherapy. For pN0 cases, non-parametric correlation and tests demonstrated significant association in eight types of characteristics [progesterone receptor (PR) expression, Ki-67 value, Ki-67 group, PR group, grade, estrogen receptor (ER) expression, Nottingham Prognostic Index (NPI) and Clinical Risk]. For pN1 cases, parametric correlation and tests showed significant association in six characteristic types (number of positive nodes, ER and PR expression, PR group, Ki-67 group and NPI). Based on OOM for pN0 cases, significant associations were established in three characteristics (Ki-67 group, grade and NPI group). For pN1 cases OOM found significant associations in seven characteristics (PR group, PNI, LVI, Ki-67 group, grade, NPI group and number of positive nodes).Conclusion: First in oncology, OOM was applied, which found some other significant characteristics associated with RS than traditional statistical methods. There were few patients, where no clinical associations were found between characteristics and RS contrary to statistically significant differences. Therefore, the results of these statistical analyses can be neither applied for individual cases nor able to provide the bases for screening patients, i.e., whether they need for OncotypeDX testing or not. OncotypeDX still provides a personalised approach in BC.

After decades of development, neonicotinoids were widely used in Hungarian sugar beet production and seeds included thiamethoxam treatment by 2019 increasing protection against early spring pests. In Hungary, neonicotides’ influence on bee health led to their removal of emergency authorization. In the absence of thiamethoxam in 2020, experience showed that multiple treatments were needed to protect beet from pests early spring pests. Repeated applications of mostly pyrethroids have proved suitable; however, in sugar beet, this has resulted in a multiple increase in environmental stress and honey bee mortality, as well as increased input values. The aim of this work is to explore the current state of production and collect new alternative options for applying effectively in Hungarian sugar beet production technology, against early spring pests.

The World Medical Association’s Declaration of Helsinki is in the process of being revised. The following amendments are recommended to be incorporated in pursuit of the common goal of promoting health for all. 1. Data-driven research that facilitates broad informed consent and dynamic consent, assuring participant’s rights, and the sharing of individual participant data (IPD) and research results to promote open science and generate social value. 2. Risk minimisation in a placebo-controlled study and post-trial access to the best-proven interventions for all who need them. 3. A future-oriented research framework for co-creation with all the relevant stakeholders.

The paper discusses Radu Muntean 2021 film, Întregalde focusing on the representation of rural landscapes and the encounter between different social classes. The film marks multiple displacements within the director’s oeuvre, epitomized as “the chronicler” of the middle class in contemporary Romanian cinema. The spatial displacement from middle class urban spaces towards mountain and rural areas and the shift from the distant picturesqueness of landscape to the experience of landscape as dwelling, as inhabited and sensed environment mediated through textural images enables unsettling embodied, affective encounters both with the natural environment and between different social classes. In this way, the film addresses the question of class differences and lays bare the socio-economic inequalities between the urban middle class and the countryside without reducing rural characters to clichéd figurants in a picturesque or sombre countryside decorum. The film’s critical reflection on a form of occasional humanitarian aid and middle class philanthropy does not relativize the concept of charity, altruism and help but rather points to the growing gap of social inequalities (and the crisis of care) in the context of contemporary capitalism.1

Abstract Background Data harmonisation is essential in real-world data (RWD) research projects based on hospital information systems databases, as coding systems differ between countries. The Hungarian hospital information systems and the national claims database use internationally known diagnosis codes, but data on medical procedures are recorded using national codes. There is no simple or standard solution for mapping the national codes to a standard coding system. Our aim was to map the Hungarian procedure codes (OENO) to SNOMED CT as part of the European Health Data Evidence Network (EHDEN) project. Methods We recruited 25 professionals from different specialties to manually map the procedure codes used between 2011 and 2021. A mapping protocol and training material were developed, results were regularly revised, and the challenges of mapping were recorded. Approximately 7% of the codes were mapped by more people in different specialties for validation purposes. Results We mapped 4661 OENO codes to standard vocabularies, mostly SNOMED CT. We categorized the challenges into three main areas: semantic, matching, and methodological. Semantic refers to the occasionally unclear meaning of the OENO codes, matching to the different granularity and purpose of the OENO and SNOMED CT vocabularies. Lastly, methodological challenges were used to describe issues related to the design of the above-mentioned two vocabularies. Conclusions The challenges and solutions presented here may help other researchers to design their process to map their national codes to standard vocabularies in order to achieve greater consistency in mapping results. Moreover, we believe that our work will allow for better use of RWD collected in Hungary in international research collaborations.

Abstract Molecular data storage is becoming a viable alternative to traditional information storage systems. Here, we propose a method where the presence or absence of a given molecule in a mixture of compounds represents a bit of information. As a novel approach, direct analysis in real time (DART) ionization mass spectrometry is used to recover and decode the information stored at the molecular level. Nicotinic acid derivatives were synthesized and used as the ‘bit compounds’. Their volatility and ease of ionization make these molecules especially suitable for DART-MS detection. The application of DART-MS as a method with an ambient ionization technique, enables the re-reading of digital chemical codes embedded in the material of ordinary objects. Our method is designed to store and read back short pieces of digital information, up to several hundred bits. These codes can have the function of barcodes or QR codes, as shown in our proof-of-principle applications. First, modelling a QR code as a link to our university's website, three solutions were prepared, each representing 22 bits. Proceeding further, the bit compounds were incorporated into a polymer matrix that is suitable for 3D printing, and a toy ship was created with a hidden barcode. In addition, decoding software was developed to process the DART-MS spectra. The nicotinic acid components representing the bits dominated the DART-MS spectra and error-free decoding was achieved.

The Varroa destructor is the most significant bee parasite and the greatest threat to bee health all around the world. Due to its hidden lifestyle, detection within the brood cell is only possible through invasive techniques. Enhancing detection methods is essential for advancing research on population dynamics, spread, selection efforts, and control methodologies against the mite. In our study, we employed infrared imaging to measure the thermal differences in parasite and intact Apis mellifera worker broods. Experiments were conducted over two years at the MATE Kaposvár Campus in Hungary involving five beehives in 2022 and five beehives in 2023. A FLIR E5-XT WIFI handheld infrared camera was used to create a heat map of capped brood frames. Our results indicate that the resolution of these cameras is sufficient to provide detailed IR images of a bee colony, making them suitable to detect temperature differences in intact and Varroa parasitized capped brood cells. Mite parasitism causes a time-dependent and sustained temperature increase in developing bee pupae, observable regardless of mite number. Our work demonstrates two different heating patterns: hotspot heating and heating cells that are responsible for the elevated temperature of the Varroa-infested cells as a social fever response by the worker bees. Based on our results, future research combined with AI-based image evaluation software could offer beekeepers and researchers practical and valuable tools for high-throughput, non-invasive Varroa detection in the field.

In the first part of this study, the electrochemical polymerization of two compounds, 3,5-dihydroxybenzoic acid and 2′,6′-dihydroxyacetophenone, was compared in dimethyl sulfoxide solvent on platinum and glassy carbon electrodes. The voltammograms obtained showed remarkable differences between the two monomers and between the two electrode materials. The acetophenone derivative formed electropolymer remnants at the electrodes, while in the case of the benzoic acid derivative, practically no passivation occurred, and the scanning electron microscopic results reinforced this. A few stackings adsorbed only after electropolymerization from a highly concentrated solution of dihydroxybenzoic acid. As a modifying layer on the platinum and glassy carbon electrodes, the prepared films from 2′,6′-dihydroxyacetophenone were tested for tributylamine in acetonitrile and in an aqueous solution of a redox-active compound, hydroquinone, during the stirring of the solution. More stable amperometric current signals could be reached with modified platinum than with glassy carbon, and the significant influence of the organic washing liquid after deposition was established via the study of noise level. In this respect, acetone was the best choice. The amperometric signals with the modified platinum obtained upon the addition of aliquots of the stock solution resulted in a 3.29 μM detection limit.

The climate-change-coupled fungal burden in crop management and the need to reduce chemical pesticide usage highlight the importance of finding sustainable ways to control Aspergillus flavus. This study examines the effectiveness of 50 Pseudomonas isolates obtained from corn rhizospheres against A. flavus in both solid and liquid co-cultures. The presence and quantity of aflatoxin B1 (AFB1) and AFB1-related compounds were determined using high-performance liquid chromatography–high resolution mass spectrometry analysis. Various enzymatic- or non-enzymatic mechanisms are proposed to interpret the decrease in AFB1 production, accompanied by the accumulation of biosynthetic intermediates (11-hydroxy-O-methylsterigmatocystin, aspertoxin, 11-hydroxyaspertoxin) or degradation products (the compounds C16H10O6, C16H14O5, C18H16O7, and C19H16O8). Our finding implies the upregulation or enhanced activity of fungal oxidoreductases and laccases in response to bacterial bioactive compound(s). Furthermore, non-enzymatic reactions resulted in the formation of additional degradation products due to acid accumulation in the fermented broth. Three isolates completely inhibited AFB1 or any AFB1-related compounds without significantly affecting fungal growth. These bacterial isolates supposedly block the entire pathway for AFB1 production in the fungus during interaction. Apart from identifying effective Pseudomonas isolates as potential biocontrol agents, this work lays the foundation for exploring new bacterial bioactive compounds.

Candida albicans is an emerging pathogen that poses a significant challenge due to its multidrug-resistant nature. There are two types of antifungal agents, fungicidal and fungistatic, with distinct mechanisms of action against fungal pathogens. Fungicidal agents kill fungal pathogens, whereas fungistatic agents inhibit their growth. The growth can be restored once the agent is removed and favorable conditions are established. Recognizing this difference is crucial as it influences treatment selection and infection prognosis. We present a technique based on optical nanomotion detection (ONMD) (i.e., observing the movement of the cells using an optical microscope) to discriminate rapidly between fungicidal (caspofungin) and fungistatic (fluconazole) drugs. The technique is based on the change in a yeast cell’s nanomotion as a function of time during a two-hour treatment with the antifungal of interest followed by a one-hour growth period. The cells are entrapped in microwells in a microfluidic chip, which allows a quick exchange of growth medium and antifungal agent, enabling ONMD measurements on the same individual cells before and after treatment. This procedure permits to discriminate between fungicidal and fungistatic antifungals in less than 3 h, with single-cell resolution by observing if the nanomotion recovers after removing the treatment and reintroducing growth medium (YPD), or continues to drop. The simplicity of the approach holds promise for further development into a user-friendly device for rapid antifungal susceptibility testing (AFST), potentially being implemented in hospitals and medical centers worldwide in developed and developing countries.

Abstract Background Several meta-analyses demonstrated the efficacy of unilateral High-Frequency Left-sided (HFL) repetitive Transcranial Magnetic Stimulation (rTMS) for individuals with Major Depressive Disorder (MDD); however, results are contradictory due to heterogeneity of the included studies. Methods A systematic literature review (SLR) of English language articles published since 2000 was performed in March 2022 on PubMed and Scopus databases. Empirical evidence on the relative efficacy of rTMS treatment compared with standard pharmacotherapy in Treatment-Resistant Depression (TRD) were extracted. Random effects models were used to assess the effects of rTMS on response and remission rates. Results 19 randomized double-blinded sham-controlled studies were included for quantitative analysis for response (n = 854 patients) and 9 studies for remission (n = 551 patients). The risk ratio (RR) for response and remission are 2.25 and 2.78, respectively for patients after two treatment failures using rTMS as add-on treatment compared to standard pharmacotherapy. Cochrane’s Q test showed no significant heterogeneity. No publication bias was detected. Conclusions rTMS is significantly more effective than sham rTMS in TRD in response and remission outcomes and may be beneficial as an adjunctive treatment in patients with MDD after two treatment failures. This finding is consistent with previous meta-analyses; however, the effect size was smaller than in the formerly published literature.

Recent freshwater tufa precipitation and its parent water were investigated at Szalajka valley, Sebesvíz and Dobrica Spring (Bükk Mts., Hungary). The aim of the study is to analyse the carbon isotope dynamics of freshwater tufa precipitated in karstic streams between the spring water and the first significant tufa barrage using field measurements, water chemistry, and carbon isotope analysis. A further aim was to examine the fossil tufa precipitations in recently active areas and their neighbourhood to determine their age using the 14C method. Abstract Travertines, which precipitate from high temperature water saturated with calcium carbonate, are generally considered to be dominated by physico-chemical and microbial precipitates. Here, as an additional influence on organomineral formation, metagenomic data and microscopic analyses clearly demonstrate that highly diverse viral, bacterial and archaeal communities occur in the biofilms associated with several modern classic travertine sites in Europe and Asia, along with virus-like particles. Metagenomic analysis reveals that bacteriophages (bacterial viruses) containing icosahedral capsids and belonging to the Siphoviridae, Myoviridae and Podoviridae families are the most abundant of all viral strains, although the bacteriophage distribution does vary across the sampling sites. Icosahedral shapes of capsids are also the most frequently observed under the microscope, occurring as non-mineralized through to mineralized viruses and virus-like particles. Viruses are initially mineralized by Ca-Si amorphous precipitates with subordinate Mg and Al contents; these then alter to nanospheroids composed of Ca carbonate with minor silicate 80–300 nm in diameter. Understanding the roles of bacteriophages in modern carbonate-saturated settings and related organomineralization processes is critical for their broader inclusion in the geological record and ecosystem models.

Abstract Several Eurydema species (Hemiptera: Pentatomidae) are considered as pests, however, reports on their chemical ecology are scarce. In the current study we focused on Eurydema ornata (Linnaeus) a pentatomid pest of several brassicaceous crops. Since the species is known to feed preferably on generative parts of plants, a series of floral and green leaf volatiles were tested by electroantennography and compounds eliciting remarkable responses were also tested in the field. Three compounds elicited the most outstanding responses from antennae of E. ornata: allyl isothiocyanate, phenylacetaldehyde and ± linalool. Field experiments were conducted in Hungary between 2017 and 2021 to test the potential attractive effects of the compounds. Three Eurydema species were caught in the experiments E. ornata, E. oleracea (Linnaeus) and E. ventralis Kolenati. In the experiments combinations containing allyl isothiocyanate attracted both males and females of E. ornata. The compound was also attractive on its own, in a positive, dose-dependent manner. When presented alone, neither phenylacetaldehyde nor ± linalool was attractive to the species, furthermore, addition of these compounds to allyl isothiocyanate did not affect attraction considerably. To our knowledge this is the first demonstration of field attration of an Eurydema species to a semiochemical and one of the few reports on trapping of a pentatomid species with a synthetic plant volatile in the field. Perspectives regarding research and potential practical applications are discussed in the paper.

Understanding the structure-property relationship is of paramount importance for tailoring copolymers for specific applications. Poly(N-acryloylmorpholine)-block-poly(N-isopropylacrylamide) (PNAM-b-PNIPAM) diblock copolymers were synthesized by reversible addition–fragmentation chain transfer (RAFT) polymerization with varying Mn and composition, providing the basis for deducing structure-property relationships. The chemical structure of the copolymers was analyzed by mass spectrometry (MS). A novel and efficient mass spectrum processing methodology was developed for the detailed analysis of polymers/copolymers that greatly expands the upper mass limit of the time-of-flight (TOF) analyzers in the linear mode up to 20,000 Da. Our method “makes visible” the mass peaks of the individual copolymer species and their isotopologues providing effective and fast automatized analysis. The self-assembly property of the thermoresponsive PNAM-b-PNIPAM diblocks in aqueous solutions was investigated by dynamic light scattering (DLS) experiments, and quantified by determining the incipient temperature of the phase transition. For rapid evaluation, an artificial neural network (ANN) was created to explore the hidden relationships between the structural information obtained by our novel mass analysis method and the properties as well as to predict the self-assembly behavior of the copolymers.

The World Health Organization highlights the urgent need to address the global threat posed by antibiotic-resistant bacteria. Efficient and rapid detection of bacterial response to antibiotics and their virulence state is crucial for the effective treatment of bacterial infections. However, current methods for investigating bacterial antibiotic response and metabolic state are time-consuming and lack accuracy. To address these limitations, we propose a novel method for classifying bacterial virulence based on statistical analysis of nanomotion recordings. We demonstrated the method by classifying living Bordetella pertussis bacteria in the virulent or avirulence phase, and dead bacteria, based on their cellular nanomotion signal. Our method offers significant advantages over current approaches, as it is faster and more accurate. Additionally, its versatility allows for the analysis of cellular nanomotion in various applications beyond bacterial virulence classification.

We present a novel optical nanomotion-based rapid antibiotic and antifungal susceptibility test. The technique consisted of studying the effects of antibiotics or antifungals on the nanometric scale displacements of bacteria or yeasts to assess their sensitivity or resistance to drugs. The technique relies on a traditional optical microscope, a video camera, and custom-made image analysis software. It provides reliable results in a time frame of 2–4 h and can be applied to motile, non-motile, fast, and slowly growing microorganisms. Due to its extreme simplicity and low cost, the technique can be easily implemented in laboratories and medical centers in developing countries.
