Nonlinear damping with respect to vibration amplitude is particularly important in mechanical dynamics. The addition of short fibers to damping materials is considered to result in strong nonlinear damping due to interfacial peeling at the edges of the fibers. However, little has been reported on the occurrence of nonlinear damping in short-fiber reinforced rubber due to compounding difficulties. In this study, we investigated the relationship between the damping characteristics and deformation behavior of microdeformed short-fiber reinforced rubber by X-ray computed tomography (CT). We prepared a damping material with a natural rubber (NR) matrix and micrometer-sized polyethylene terephthalate (PET) fiber filler. The loss factor was identified by dynamic mechanical analysis, and three-dimensional strain maps were obtained using marker tracking in the CT data. The addition of 5 wt% PET fibers to NR resulted in an increase in the loss factor. Experimentally, we found that the nonlinear damping of the composite rubber is affected by the peeling of the filler/matrix interface and the strain inside the material.

Toxoplasma is a common parasite worldwide that mainly affects the brain, lungs and eyes. Although toxoplasmic encephalitis is a lethal disease without treatment, past case reports show most patients with systemic lupus erythematosus who developed toxoplasmic encephalitis were misdiagnosed and treated as neuropsychiatric systemic lupus erythematosus, which led to unfavorable outcomes. We herein describe a case of disseminated toxoplasmosis affecting all the above organs with atypical symptoms, which developed with exacerbation of systemic lupus erythematosus. She had initially manifested with retinochoroiditis without vitritis, mild cognitive impairment and an isolated lung mass. These are completely different from the classic symptoms of toxoplasmosis that have been reported in patients with HIV infection and/or those after hematopoietic transplantation. Our case, together with previously reported cases, suggests the manifestation of toxoplasmosis that develops in systemic lupus erythematosus patients can be different from that seen in conventional cases and varies between individual patients. Our case highlights both the difficulty in and the importance of diagnosing toxoplasmosis in patients with systemic lupus erythematosus and provides helpful information to identify this rare, devastating, yet treatable disease.

Summary The waste biomass residue from satsuma mandarin juice production was extracted by wet-fine grinding and separated by centrifugation into insoluble and soluble fractions. The insoluble material was wet-screened into four fractions and analysed for crude fibre and total hydrolysable sugars. Crude fibre was almost exclusively in fractions containing particles over 50 μm, comprising about 65% of the insoluble material; this could provide a raw material high in crude fibre. The remaining insoluble fraction was almost devoid of crude fibre. The soluble extract contained up to 63% of the total pectic substances with very similar methoxyl contents and degree of esterification, but double the viscometric molecular weight of a standard citrus pectin. The method provides an alternative to that of hot acid extraction.

We have routinely applied an extra-strong graduated compression stocking to cesarean section patients to reduce the incidence of spinal anesthesia hypotension. Because bupivacaine has recently become available in Japan, we compared the incidence of spinal hypotension using either 2.0 ml of hyperbaric 0.3% dibucaine or 0.5% bupivacaine. There were 98 full-term parturients wearing the stocking who received 2.0 ml injection of dibucaine or bupivacaine for elective cesarean section. When systolic blood pressure decreased to 90-100 mm Hg or to less than 70% of the pre-anesthesia value, ephedrine was injected intravenously. There was no significant difference in systolic blood pressure or heart rate during spinal anesthesia between the dibucaine and bupivacaine groups. Although the demographic data and various data related to anesthesia or surgery were similar in the groups, the fluid volume and the dose and incidence of ephedrine injection during anesthesia showed significant differences: the mean dose was 3.6 and 1.5 mg and the incidence was 41% and 19% in the dibucaine and bupivacaine groups, respectively. Spinal anesthesia using bupivacaine results in a lower incidence of spinal hypotension compared with dibucaine and, in combination with fitting the extra-strong stockings onto both legs, is clinically useful for cesarean sections.

Twelve elderly patients undergoing total knee arthroplasty received lumbar epidural anaesthesia and propofol infusion at 5 mg.kg(-1).h(-1) following a 1.5-2.0 mg.kg(-1) bolus dose with preservation of spontaneous respiration via a laryngeal mask airway. Circulatory, respiratory and metabolic variables were measured before and 1, 3, 5, 15 and 30 min after release of a pneumatic thigh tourniquet. The blood pressure was decreased at all time-points and the respiratory rate increased at 1 min. The P(a)CO(2) was increased only at 1 min. Arterial blood pH and base excess were decreased at 1 and 3 min and 1, 3 and 5 min, respectively. Arterial blood lactate levels were increased at all times. These characteristics were considered to be identical to those under regional anaesthesia with conscious spontaneous respiration, showing that spontaneous respiration under this anaesthetic regimen has a similar respiratory capacity to that of conscious spontaneous respiration.

Because a standard type of graduated compression stocking is not effective for the prevention of spinal anesthesia hypothension during cesarean section, we have used an extra-strong type of graduated compression stocking. This study examined whether the extra-strong stocking reduces usage of vasopressor agents. Forty-eight and 47 full term parturients were fitted with the standard stocking and the extra-strong stocking, respectively. Spinal anesthesia was performed by injecting 2.0 ml 0.3% dibucaine hyperbaric solution. When systolic blood pressure decreased to 90-100 mmHg or to less than 70% of the pre-anesthesia value, ephedrine was injected. There was no significant difference in systolic blood pressure or heart rate during spinal anesthesia between the groups. The mean dose of ephedrine injected during anesthesia was 12.2 and 4.3 mg, and the incidence of ephedrine injected was 85 and 49%, in the standard and extra-strong stocking groups, respectively. The extra-strong stocking group showed significantly lower values. Fitting the extra-strong stocking on both legs for cesarean sections undergoing spinal anesthesia reduced usage of vasopressor agents, suggesting a promising use of this stocking for non-invasive prophylaxis.

The Lithium Battery Energy Storage Technology Research Association (LIBES) conducts research and development on ‘Dispersed Battery Energy Storage Technology’ of the New Sun-Shine Program of the Agency of Industrial Science and Technology (AIST), Ministry of International Trade and Industry (MITI). This national project is being carried out based on the contract with New Energy and Industrial Technology Development Organization (NEDO). This R&D project for ten years with the budget of approximately Y14 billion has been started in the fiscal year 1992, and includes: (i) R&D of 20–30 kWh class high-performance ambient-temperature lithium secondary batteries, and (ii) study on the total system of lithium-battery application technology. This paper describes the project and current research activities.

An experimental study on the removal of NH/sub 3/, NO, and NO/sub x/ in concentrations of 10-40 ppm in air has been carried out using plasma chemical reaction in a streamer corona discharge. The results of the performance of dry-type and semi-wet-type reactors are compared. The effect of different types of applied voltages such as rectangular pulse, 60 Hz sinusoidal, and 18 kHz alternating voltages is investigated. During NO removal, O/sub 3/ and NO/sub 2/ are produced. NO/sub 2/ can, partially, be removed with higher power input into the discharge. Another undesirable pollutant, namely N/sub 2/O, is also produced, especially in the case of dry reactors having long residence time ( approximately 2.4 s). N/sub 2/O production decreases, essentially, to zero at 0.6 s residence time when using a semiwet reactor. In general, higher removal efficiency has been obtained with pulse voltage in a wet reactor. NH/sub 3/ in air appears to produce ozone and ammonium nitrate in a discharge. The performance of semiwet reactors with regard to the removal of submicron dust particles has also been investigated and very high removal efficiency ( approximately 93% at 0.6 s residence time) has been obtained. >

In this paper, the authors report the results of laboratory experiments on NO/sub x/ removal (DeNOx) using a nonthermal plasma process. Studies were conducted on a dry-type plasma reactor and several wet-type reactors to evaluate DeNOx efficiency. The reactor geometry was coaxial with an inner discharge electrode and an outer ground electrode wrapped on an insulating glass tube. Simulated flue gas was used in the experiment. The dry-type reactor performed better with the addition of ammonia and water vapor to the simulated gas. In the wet-type reactors, water and NaOH solution were used as absorbents and DeNOx performance was found to be the same in each case. Also, with water as the absorbent and with its pH value dropping below 3, the performance of the wet-type reactors remained constant. Studies on wet-type reactors, further, suggested that about half of NO removed by the plasma was dissociated into N/sub 2/ and O/sub 2/ and the rest was absorbed by water. The studies indicate that wet-type reactors performed better than the dry-type reactor in the removal of NO/sub x/.

Weekly survival of 39 wild (4 native and 35 translocated) versus 32 pen-reared birds were compared at 2 sites (~2,000 ha each) during the fall 1996 agricultural harvests. Pheasant survival after 1 week was wild 74% and 79% versus pen-reared 61% and 57% at the Meridian and Nicolaus sites, respectively. Thereafter, pooled sites survival was ~linear with ~1 wild bird dying every 2.8 weeks for 7 weeks and ~1 pen-reared bird death occurred every 4.7 days for 3 weeks. Several relocated and pen-reared pheasants joined wild flocks, and their survival improved with one of each harvested during the 1997 hunting season. Both survived g400 days. All affected pheasants changed their habitat utilization and movement ecology following the harvest of their primary cover and forage crop(s). Chi square analysis of habitat use by 30 wild and 19 pen-reared pheasants demonstrated habitat preferences were greater than its availability (P l 0.01) for milo (planted only in Meridian), weeds, and corn. Their preferences for alfalfa, beets, and safflower were equal to their availability. Rice was preferred when the fields were dry, but overall it was not preferred (P l 0.01). Also, orchards (cleared of ground vegetation for nut harvests) and fallow habitats were not preferred. Movement ecology data were separated by study site because of significant habitat differences. Home ranges (95% utilization areas) using the minimum convex polygon method to compare wild versus pen-reared pheasants averaged 74 and 67 ha at Meridian and 73 and 140 ha at Nicolaus. Daily rooster and hen movements averaged 295 m and 276 m for wild birds and 335 m and 382 m for pen-reared birds at the Meridian and Nicolaus sites, respectively. Results from the first fall pheasant study in California crops demonstrated they preferred the dynamic juxtaposition of grains and weeds for cover, shelter and forage with water. Twenty-nine pheasants (58%) demonstrated habitat preferences for grains– milo, rice, and corn. Pheasant survival was related to post-harvest habitat changes. Their home range, survival, and movements were very similar at both sites although the crop mosaic and habitat relationships were very different. These new results should be included in both public and private pheasant management practices.

In the surface acoustic wave quantum computer, the spin state of an electron trapped in a moving quantum dot comprises the physical qubit of the scheme. Via detailed analytic and numerical modeling of the qubit dynamics, we discuss the effect of excitations into higher-energy orbital states of the quantum dot that occur when the qubits pass through magnetic fields. We describe how single-qubit quantum operations, such as single-qubit rotations and single-qubit measurements, can be performed using only localized static magnetic fields. The models provide useful parameter regimes to be explored experimentally when the requirements on semiconductor gate fabrication and the nanomagnetics technology are met in the future., 13 pages, 10 figures, submitted to Phys. Incoherence in the controlled Hamiltonian is an important limitation on the precision of coherent control in quantum information processing. Incoherence can typically be modelled as a distribution of unitary processes arising from slowly varying experimental parameters. We show how it introduces artifacts in quantum process tomography and we explain how the resulting estimate of the superoperator may not be completely positive. We measured the temperature dependence of electrical resistivity of non-fluorine- substituted, oxygen-deficient LnFeAsO1-y (Ln = La, Ce, Nd, Tb) under hydrostatic high pressure up to '8 GPa in order to observe pressure dependence of superconducting transition temperature Tc. In LaFeAsO0.65, onset Tc initially enhances below 4 GPa, then decreases by applying further pressure. However, the zero-resistivity temperature does not show enhancement by applying pressure. In the case of NdFeAsO1-y and TbFeAsO1-y, Tc shows monotonic decrease as increasing pressure. Tc is much more likely to go down faster if Ln with small ionic size was taken. Therefore, the bulk superconductivity is suppressed finally at ~7 GPa in TbFeAsO0.7.

We have purified and identified a 32-kDa protein interacting with the Dbl oncogene homology domain of mSos1(Sos-DH) from rat brains by glutathione S-transferase-Sos-DH affinity chromatography. Peptide sequencing revealed that the protein is identical to a positive regulatory E subunit (V-ATPase E) of a vacuolar H(+)-ATPase, which is responsible for acidification of endosome and alkalinization of intracellular pH. The interaction between V-ATPase E and Sos-DH was confirmed by yeast two-hybrid assay. A coimmunoprecipitation assay demonstrated that a V-ATPase E protein physiologically bound to mSos1, and the protein was colocalized with mSos1 in the cytoplasm, as determined by immunohistochemistry. mSos1 was found in the early endosome fraction together with V-ATPase E and Rac1, suggesting the functional involvement of mSos1/V-ATPase E complexes in the Rac1 activity at endosomes. Overexpression of V-ATPase E in COS cells enhanced the ability of mSos1 to promote the guanine nucleotide exchange activity for Rac1 and stimulated the kinase activity of Jun kinase, a downstream target of Rac1. Thus, the data indicate that V-ATPase E may participate in the regulation of the mSos1-dependent Rac1 signaling pathway involved in growth factor receptor-mediated cell growth control.

A 46-year-old male presented with a huge facial schwannoma extending into both the middle cranial fossa and the cerebellopontine angle but without manifesting facial nerve palsy. Neurological examination on admission revealed no deficits except for speech disturbance. Computed tomography showed a multicystic tumor extending into the middle cranial fossa and the cerebellopontine angle, with destruction of the petrous bone. The tumor was totally grossly removed. Histological examination identified schwannoma. Total facial nerve palsy appeared postoperatively, but hearing acuity was preserved at a useful level. Facial nerve palsy is one of the most typical symptoms in patients with facial schwannoma, but is not always manifested even if the tumor extends into both the middle cranial fossa and the cerebellopontine angle.

Summary: A 12-year-old female patient presented with unilateral sensorineural hearing loss. Distortion-product otoacoustic emission testing failed to reveal any measurable emissions in the affected side. MR imaging did not reveal labyrinthine malformation. Three-dimensional Fourier transformation–constructive interference in steady-state MR images showed a thin cochlear branch. We speculated that mumps infection or developmental malformation caused the unilateral sensorineural hearing loss.

BACKGROUND AND PURPOSE: In childhood-onset moyamoya disease, the angiographic disease process of stenoocclusive lesions is progressive, and cerebral infarctions often develop as a result of ischemia. Our purpose was to determine how the severity of stenoocclusive lesions in the anterior and posterior circulations affects the distribution of cerebral infarction in patients with childhood-onset moyamoya disease. METHODS: In 69 patients with childhood-onset moyamoya disease, angiograms were reviewed for stenoocclusive lesions, and CT scans, MR images, or both were reviewed for the sites and extent of cerebral infarction. The relationship between the angiographic and CT/MR findings was examined. RESULTS: The prevalence and degree of stenoocclusive lesions of the posterior cerebral artery (PCA) significantly correlated with the extent of lesions around the terminal portion of the internal carotid artery (ICA). The prevalence of infarction significantly correlated with the degree of stenoocclusive changes of both the ICA and PCA. Infarctions tended to be distributed in the anterior borderzone in less-advanced cases, while in more advanced cases lesions were additionally found posteriorly in the territory of the middle cerebral artery, the posterior borderzone, and the PCA territory. CONCLUSION: Our results indicate that progressive changes of the anterior and posterior circulations are associated with the distribution of cerebral infarction, culminating in a patchily disseminated or honeycomb pattern of infarction on CT and MR studies in late stages of the disease.

To assess the clinical, angiographic and early follow-up findings of young patients suffering an acute myocardial infarction, in comparison with older patients with infarction, in the thrombolytic era.A retrospective analysis of the medical records of 46 patients40 years-old (group I) at the time of an acute myocardial infarction was compared with that of 46 older patients, randomly selected, presenting with this syndrome between february, 1991 and february, 1996 (group II). In both groups a comparison was conducted regarding the proportions of gender, risk factors, type of infarction (Q vs non-Q), left ventricular function, coronary anatomy and early mortality (1 month). The medical treatment was comparable for both groups, including the utilization of thrombolytics.The groups were discriminated only by: higher prevalence of smoking, of angiographically normal coronary arteries, and of non-critical (75% reduction of luminal diameter) coronary stenosis in group I; in the older group a higher proportion of patients had multivessel disease. Although not reaching statistical significance, a trend was observed to a more benign early course of the infarction in the patients less than40 years-old.The present findings are similar to those described in the pre-thrombolytic era, for young patients suffering an acute myocardial infarction.

From March 1986 to September 1991, the right gastroepiploic artery has been used for coronary artery bypass grafting in 200 patients (171 male and 29 female patients, mean age 58 years, range 6 to 80 years. They were followed up from 6 to 70 months with a mean of 27 months. There were 16 reoperations and 176 patients had triple vessel or left main disease. The gastroepiploic artery (182 in situ and 18 free grafts) was anastomosed to 11 anterior descending, 3 diagonal, 26 circumflex, and 160 right coronary arteries. The internal thoracic artery was concomitantly used in 192 patients. The mean number of distal anastomoses was 2.3 with arterial grafts and 3.3 including additional saphenous vein grafts. Postoperative angiography was performed in 152 patients within 6 months after the operation (mean 2 months) and after the operation second angiograms were done sequentially 1 to 5 years (mean 2 years) after the operation in 40 patients. There were 6 early and 4 late deaths. A new Q wave was noted in 4 patients. Duration of the operation and postoperative complications did not increase with the use of the gastroepiploic artery. Relief of angina was noted in 186 patients. Gastroepiploic artery graft patency was 95% (144/152) in the early postoperative period and 95% (38/40) in the late postoperative period. Percutaneous transluminal coronary angioplasty was done successfully through the in situ gastroepiploic artery graft for anastomotic stenosis in four cases. In stress myocardial scintiscans, performed sequentially preoperatively and in the immediate, 1-year, and 2-year postoperative periods in 11 patients, washout rate of the gastroepiploic artery-grafted area improved from 35% +/- 10% to 45% +/- 15% (p0.05) and was maintained to 43% +/- 6% and 48% +/- 9% at respective periods. In conclusion, the gastroepiploic artery is a suitable conduit for coronary artery bypass grafting in terms of low surgical risk, high patency rate, and excellent patient outcome.

The linewidth enhancement factor (α-factor) of a commercially available 6.1 μm InGaAs/InAlAs distributed-feedback quantum cascade laser capable of continuous-wave operation with a Peltier cooler and its current dependence were investigated. α tended to increase slowly with an increase in current. α, probably affected by detuning, was still smaller than that of conventional 1.55 μm semiconductor laser diodes.

A 70-year-old woman was admitted for evaluation of hepatosplenomegaly, fever and elevated serum LDH levels. A biopsy specimen of the liver revealed histiocytic proliferation at the portal triad, and a mild degree of hepatitis. A bone marrow biopsy specimen showed proliferation of histiocytes with minimal immaturity and atypism, and haemophagocytosis by the proliferated histiocytes. Fever, hepatosplenomegaly and elevation of LDH levels all disappeared spontaneously, and presumptive diagnosis of benign reticulosis with haemophagocytosis was made. One year later, fever, hepatosplenomegaly and elevation of LDH levels redeveloped, and the liver and bone marrow biopsy specimen showed proliferation of unequivocally malignant histiocytes. The patient died as a result of disseminated intravascular coagulation with shock 20 d later. We concluded that, in this case, malignant histiocytosis first presented as benign haemophagocytic reticulosis and, 1 year later, there was a typical malignant presentation.

Infants often take an asymmetrical posture and muscle tone, that is, one sided flattening of the skull, postural scoliosis, limitation of abduction of the flexed hip, pelvic obliquity and so on.In this study, we described the significance of the these oblique posture in infancy and the techniques of their handlings.In order to prevent the occurrance of deformity of the skull, new-born babies should be put into the side position for the first four months. After that they should be kept in the prone position.When an oblique posture was noticed in infants, we advised their mothers to do some handlings which were based on the righting and balance reactions: for example, oblique suspension.

長野県松本市近郊A村の住民を対象とし,昭和57年より3年間継続して大腸集検を実施した.集検方式は問診と便潜血による一次スクリーニング後,二次スクリーニングとしてsigmoidoscopyを行う方法を採用した.参加者はのべ総数4,388人,3年間の平均が対象者(40歳以上)の67%1,463人で,このうち問診陽性者,便潜血陽性者,要内視鏡検査者はそれぞれ5.0%,8.1%,11.6%であった.癌7人,0.16%(早期癌3人,0.07%,進行癌4人,0.09%),ポリープ54人,123%が発見された.2年目,3年目に発見された進行癌の3症例は,いずれも前年度の便潜血は陰性であった.ポリープも3年間を通じ高率に発見された。なお内視鏡検査の脱落者のなかに他医により早期癌と診断された例もある.以上より問診便潜血によるスクリーニングでは,false negativeの欠点を補充するため,逐年検診が必須と考えられる,また精検過程での脱落者への対応も必要と思われる.

A case with persistent atrial standstill is presented which developed from atrial fibrillation associated with rheumatic combined valvular heart disease. In addition to clinical and electrocardiographic findings, electrophysiological and histological studies by using microelectrode technique and electron microscopy, respectively, were carried out on specimens from the right atrial appendage resected at mitral valve surgery. Scattered but severe loss of myocardial cells in the atria and deterioration of the action potential of these poorly sustained myocardial cells were confirmed. The suggestive evidence is also shown that the deteriorated action potential observed by micro-electrode technique in this case may be composed of calcium current.

To investigate the existence of heterogeneity of beta-type myosin isozymes (HC beta) in human hearts, immunohistochemical studies using monoclonal antibodies (MoAbs) raised against human ventricular myosin heavy chains were performed. Two types of MoAbs recognized some muscle fibers in the atrium, whereas both reacted with all ventricular muscle fibers. Since atrial muscle fibers reactive with each MoAb were found to be clearly different, the existence of two immunologically distinct HC beta (beta 1, and beta 2) was suggested in the atrium. By using affinity chromatography, two molecular variants of HC beta were isolated from the bovine atrium, and differences in the primary structure of beta 1 and beta 2 were confirmed by analysis of peptides produced by chymotryptic digestion. In pressure-overloaded human atria, myofibers containing beta 1 and/or beta 2 increased in accordance with decrement of myofibers containing alpha-type myosin isozyme (P less than 0.01). But they differed in expression during the developmental stage, since beta 2 did not exist in the early embryonic bovine heart, but beta 1 did. Thus, there are two distinct HC beta whose expression is regulated by at least two factors: pressure overload and developmental stage.

For the studies on the mechanism of induction of peroxisomal beta-oxidation enzymes and biogenesis of the organelle, we have isolated cDNA clones for rat peroxisomal enoyl-CoA: hydratase-3-hydroxyacyl-CoA dehydrogenase bifunctional enzyme. On blotting experiments with liver RNA, the cDNAs hybridized to a 3.0-kilobase RNA which was increased 5-7-fold by the administration of di-(2-ethylhexyl)phthalate to rats. Nucleotide sequencing was carried out for four cloned cDNAs and one obtained by a primer extension method. By overlapping these sequences with each other, we identified 20 nucleotides of 5'-noncoding, 2,166 nucleotides of coding, and 910 nucleotides of 3'-noncoding regions. The deduced amino acid sequence of the enzyme is composed of 722 residues, and the composition agrees with that of the protein data. The sequence was confirmed by the amino acid compositions and sequence analyses of some of the tryptic peptides. The molecular weight of the mature enzyme is calculated to be 78,511 from the predicted amino acid sequence. The enzyme has no terminal peptide extension as a signal for translocation into peroxisomes.

An immunohistochemical study using monoclonal antibodies specific for the heavy chains of either human atrial (HC alpha) or ventricular (HC beta) myosin was performed to clarify the distribution of each isozyme in normal as well as pressure-overloaded human hearts. In normal human ventricles, all muscle fibers were stained by a monoclonal antibody (HMC14) specific for HC beta, whereas a small number of fibers reacted with a monoclonal antibody (CMA19) specific for HC alpha. In contrast, in normal human atria, almost all muscle fibers were stained by CMA19, and a relatively larger number of muscle fibers also reacted with HMC14. Furthermore, in pressure-overloaded atria, muscle fibers reactive with HMC14 were strikingly increased while those reactive with CMA19 showed a corresponding decrease. The extent of this isozymic redistribution was in good correlation with atrial pressure. These results not only confirmed the existence of isoforms of myosin heavy chain in human hearts, but also demonstrated that redistribution of iso-myosins could occur as an adaptation to pressure overload.

Two familial cases of dilated cardiomyopathy were evaluated by HL-A typing. In the case of the first family, the mode of inheritance is likely to be an autosomal dominant trait. Only the affected individuals carried the identical HL-A haplotype (A2, Bw54, Cw1, DR4, DQw3), while the unaffected members do not share this pattern. In the second family case, the disease is probably inherited by autosomal recessive traits. All of the family members examined shared the identical HL-A haplotype (A24, Bw52, DR2, DQw1), but only the affected individuals were homozygous for this haplotype.

cDNA clones for rat acyl-CoA oxidase were isolated. The 3.8-kilobase mRNA sequence of the enzyme was completely covered by two overlapping clones. The composite cDNA sequence consisted of 3741 bases and contained a 1983-base open reading frame which encodes a polypeptide of 661 amino acid residues. Two species of acyl-CoA oxidase cDNA were identified. They differed in their coding nucleotide sequences, only within a small region. They contained the same number of nucleotides and can be translated in a common reading frame. They are 55% and 50% homologous in the above region at the nucleotide and the amino acid levels, respectively. Both types of cDNA were isolated from a library constructed from mRNA of a single rat, thereby suggesting the occurrence of two species of acyl-CoA oxidase in each rat. The amino terminus of the enzyme was determined to be N-acetylmethionine, which corresponds to the initiator methionine, thus confirming the absence of a terminal presequence. We reported previously that a purified preparation of the enzyme contained three polypeptide components, A, B, and C, and suggested that components B and C are produced by a proteolytic cleavage of component A (Osumi, T., Hashimoto, T., and Ui, N. (1980) J. Biochem. (Tokyo) 87, 1735-1746). We located components B and C on the amino- and the carboxyl-terminal sides of component A. Possible functional significances of several stretches of amino acids of the enzyme are discussed, based on the sequence comparison data between rat and yeast acyl-CoA oxidases.

Recent data from our laboratory have shown that patients with the indeterminate form of Chagas' disease can have impairment of left ventricular contractility, as evaluated by the slope of the left ventricle end-systolic pressure-dimension relationship. We also showed that Chagas' disease patients with minimal baseline wall motion abnormalities detected by two-dimensional echocardiography have more intense contractility impairment when compared to patients with the indeterminate form of the disease without this abnormality. The prognostic implications of these findings have not been established. We evaluated 59 patients (37-76 years, mean = 55 years) with different clinical forms of Chagas' disease, who had normal left ventricular global systolic function at baseline (57.6 ± 6.9%) and who had at least one additional echo during clinical follow-up (0.4-17.6; mean 4.6 years). Group 1 consisted of 14 patients with minor baseline left ventricle wall motion abnormalities and group 2 consisted of 45 patients without these abnormalities. During follow-up, global left ventricle systolic function deterioration was observed in 10 group 1 patients (71.4%) and in only 10 group 2 patients (22.2%; P < 0.005). Age and duration of follow-up were not independent determinants of left ventricular function deterioration in these patients. The present data indicate that mild segmental left ventricular wall motion abnormalities are associated with worsening of systolic function in Chagas' disease patients who have normal baseline global systolic performance.
