As the demand for biobased lubricating oils continues to rise, there is a growing focus on exploring diverse oil types. Particularly noteworthy is the surge in demand for high oleic oils, which offer enhanced stability, and a richer oleic acid content compared to their regular oil counterparts. However, the performance of high oleic soybean oil (HOSO) with additives compared to regular soybean oil (RSO), remains unclear. This study is focused on revealing the compatibility of both regular soybean oil (RSO) and high oleic soybean oil (HOSO) with select antiwear and antioxidant additives, specifically zinc dialkyl dithiophosphate (ZDDP), and zinc dialkyl dithiocarbamate (ZDDC) combined with antimony dialkyldithiocarbamate (ADDC), along with a comparative performance analysis of these additives. Reciprocating friction, wear, and electrical contact resistance-based analyses were conducted to evaluate additive compatibility and wear mechanisms at room temperature lubrication conditions. Interestingly, it was observed that for the select additives, the compatibility with regular soybean oil (RSO) was better than that of high oleic soybean oil (HOSO). RSO with additives showed around 28% reduction of wear volume whereas, it was only 8% for HOSO with additives. Additional physiochemical property analyses were conducted on the lubricants to correlate the observed tribological behavior. Fragile X syndrome (FXS) is the most common cause of heritable intellectual disability and autism and affects ~1 in 4000 males and 1 in 8000 females. The discovery of effective treatments for FXS has been hampered by the lack of effective animal models and phenotypic readouts for drug screening. FXS ensues from the epigenetic silencing or loss-of-function mutation of the fragile X mental retardation 1 (FMR1) gene, which encodes an RNA binding protein that associates with and represses the translation of target mRNAs. We previously found that the activation of LIM kinase 1 (LIMK1) downstream of augmented synthesis of bone morphogenetic protein (BMP) type 2 receptor (BMPR2) promotes aberrant synaptic development in mouse and Drosophila models of FXS and that these molecular and cellular markers were correlated in patients with FXS. We report that larval locomotion is augmented in a Drosophila FXS model. Genetic or pharmacological intervention on the BMPR2-LIMK pathway ameliorated the synaptic abnormality and locomotion phenotypes of FXS larvae, as well as hyperactivity in an FXS mouse model. Our study demonstrates that (i) the BMPR2-LIMK pathway is a promising therapeutic target for FXS and (ii) the locomotion phenotype of FXS larvae is a quantitative functional readout for the neuromorphological phenotype associated with FXS and is amenable to the screening novel FXS therapeutics.

Epigenetic silencing of fragile X mental retardation 1 (FMR1) causes fragile X syndrome (FXS), a common inherited form of intellectual disability and autism. FXS correlates with abnormal synapse and dendritic spine development, but the molecular link between the absence of the FMR1 product FMRP, an RNA binding protein, and the neuropathology is unclear. We found that the messenger RNA encoding bone morphogenetic protein type II receptor (BMPR2) is a target of FMRP. Depletion of FMRP increased BMPR2 abundance, especially that of the full-length isoform that bound and activated LIM domain kinase 1 (LIMK1), a component of the noncanonical BMP signal transduction pathway that stimulates actin reorganization to promote neurite outgrowth and synapse formation. Heterozygosity for BMPR2 rescued the morphological abnormalities in neurons both in Drosophila and in mouse models of FXS, as did the postnatal pharmacological inhibition of LIMK1 activity. Compared with postmortem prefrontal cortex tissue from healthy subjects, the amount of full-length BMPR2 and of a marker of LIMK1 activity was increased in this brain region from FXS patients. These findings suggest that increased BMPR2 signal transduction is linked to FXS and that the BMPR2-LIMK1 pathway is a putative therapeutic target in patients with FXS and possibly other forms of autism.

Recent findings in several organ systems show that cytoneme-mediated signaling transports signaling proteins along cellular extensions and targets cell-to-cell exchanges to synaptic contacts. This mechanism of paracrine signaling may be a general one that is used by many (or all) cell types in many (or all) organs. We briefly review these findings in this perspective. We also describe the properties of several signaling systems that have previously been interpreted to support a passive diffusion mechanism of signaling protein dispersion, but can now be understood in the context of the cytoneme mechanism. Long-distance cell-cell communication is essential for organ development and function. Whereas neurons communicate at long distances by transferring signals at sites of direct contact (i.e., at synapses), it has been presumed that the only way other cell types signal is by dispersing signals through extracellular fluid--indirectly. Recent evidence from Drosophila suggests that non-neuronal cells also exchange signaling proteins at sites of direct contact, even when long distances separate the cells. We review here contact-mediated signaling in neurons and discuss how this signaling mechanism is shared by other cell types.

Decapentaplegic (Dpp), a Drosophila morphogen signaling protein, transfers directly at synapses made at sites of contact between cells that produce Dpp and cytonemes that extend from recipient cells. The Dpp that cytonemes receive moves together with activated receptors toward the recipient cell body in motile puncta. Genetic loss-of-function conditions for diaphanous, shibire, neuroglian, and capricious perturbed cytonemes by reducing their number or only the synapses they make with cells they target, and reduced cytoneme-mediated transport of Dpp and Dpp signaling. These experiments provide direct evidence that cells use cytonemes to exchange signaling proteins, that cytoneme-based exchange is essential for signaling and normal development, and that morphogen distribution and signaling can be contact-dependent, requiring cytoneme synapses.

Objectives: From the treatment of damaged teeth to replacing missing teeth, dental biomaterials cover the scientific interest of many fields. Dental biomaterials are one of the implants whose effective life depends vastly on their material and manufacturing techniques. The purpose of this review is to summarize the important aspects for metallic dental implants from biomedical, mechanical and materials science perspectives. The review article will focus on five major aspects as mentioned below. Tooth anatomy: Maximizing the implant performance depends on proper understanding of human tooth anatomy and the failure behavior of the implants. Major parts from tooth anatomy including saliva characteristics are explored in this section. Wear mechanisms: The prominent wear mechanisms having a high impact on dental wear are abrasive, adhesive, fatigue and corrosion wear. To imitate the physiological working condition of dental implants, reports on the broad range of mastication force and various composition of artificial saliva have been included in this section, which can affect the tribo-corrosion behavior of dental implants. Dental implants classifications: The review paper includes a dedicated discussion on major dental implants types and their details for better understanding their applicability and characteristics. Implant materials: As of today, the most established dental implant materials are SS316L, cobalt chrome alloy and titanium. Detailed discussion on their material properties, microstructures, phase transformations and chemical compositions have been discussed here. Manufacturing techniques: In terms of different production methods, the lost wax casting method as traditional manufacturing is considered. Selective Laser Melting (SLM) and Directed Energy Deposition (DED) as additive manufacturing techniques (AM) have been discussed. For AM, the relationships between process–property–performance details have been explored briefly. The effectiveness of different manufacturing techniques was compared based on porosity distribution, mechanical and biomechanical properties. Summary: Despite having substantial research available on dental implants, there is a lack of systematic reviews to present a holistic viewpoint combining state-of-the-art from biomedical, mechanical, materials science and manufacturing perspectives. This review article attempts to combine a wide variety of analyzing approaches from those interdisciplinary fields to deliver deeper insights to researchers both in academia and industry to develop next-generation dental implants. Multiple promoters drive the expression of the principal cell division gene, ftsZ, in bacterial systems. Primer extension analysis of total RNA from Mycobacterium tuberculosis and a Mycobacterium smegmatis transformant containing 1.117 kb of the upstream region of M. tuberculosis ftsZ and promoter fusion studies identified six ftsZ transcripts and their promoters in the ftsQ open reading frame and ftsQ-ftsZ intergenic region. The presence of multiple promoters reflects the requirement to maintain a high basal level of, or to differentially regulate, FtsZ expression during different growth conditions of the pathogen in vivo.

Background and Aims: Since December 2019, an increasing number of pneumonia cases of unknown reasons emerged in Wuhan, China. Deep-sequencing analysis from nasopharyngeal swabs, sputum, lower respiratory tract samples, and blood indicated a novel coronavirus, known as 2019-nCoV. Our aim was to determine the association of haematological parameters such as haemoglobin (Hb%), total leukocyte count (TLC), platelet count (Plt) with severity of coronavirus disease 19 (COVID 19) infection among patients admitted in the COVID intensive care unit (ICU) and COVID ward, and our objectives were to estimate haemoglobin (Hb), TLC, differential leukocyte count (DLC), and platelet count of patients admitted in the COVID ward and ICU; to derive neutrophil lymphocyte ratio (NLR) from the DLC of the patients admitted in the COVID ward and ICU; and to determine the correlation between Hb, platelet count, TLC, and NLR with severity of disease among COVID patients. Methods: After receiving approval from institutional ethics committee, data were collected according to our study parameter (age, weight, height, Hb%, TLC, platelet, NLR) from the medical record section for ICU-admitted (Group A) and ward-admitted (Group B) patients and compared. Here, SPSS version 22 was used for analysis, and a p value <0.05 was considered significant. Results: A decreasing trend in Hb%, TLC, and platelet count was seen in both the groups but more in group A than group B, which is statistically significant; NLR got an increasing trend in these two groups, and the increase in group A is more and statistically significant compared to group B. Conclusion: Our study tries to explore the feasibility of such parameters in this regard and found that a combination of NLR, Hb%, TLC, and platelet count can be considered together as an effective prognostic indicator in severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Study of activity of cloned promoters in slow-growing Mycobacterium tuberculosis during long-term growth conditions in vitro or inside macrophages, requires a genome-integration proficient promoter probe vector, which can be stably maintained even without antibiotics, carrying a substrate-independent, easily scorable and highly sensitive reporter gene. In order to meet this requirement, we constructed pAKMN2, which contains mycobacterial codon-optimized gfp(m) (2+) gene, coding for GFP(m) (2+) of highest fluorescence reported till date, mycobacteriophage L5 attP-int sequence for genome integration, and a multiple cloning site. pAKMN2 showed stable integration and expression of GFP(m) (2+) from M. tuberculosis and M. smegmatis genome. Expression of GFP(m) (2+), driven by the cloned minimal promoters of M. tuberculosis cell division gene, ftsZ (MtftsZ), could be detected in the M. tuberculosis/pAKMN2-promoter integrants, growing at exponential phase in defined medium in vitro and inside macrophages. Stable expression from genome-integrated format even without antibiotic, and high sensitivity of detection by flow cytometry and fluorescence imaging, in spite of single copy integration, make pAKMN2 useful for the study of cloned promoters of any mycobacterial species under long-term in vitro growth or stress conditions, or inside macrophages., (© 2011 The Authors. Echinococcus granulosus causes a zoonotic infection called Cystic Echinococcosis (CE). Surgeons meet with hydatid cysts of the liver and lungs with reasonable frequency. However hydatid cyst may also appear in other parts of the body. A 30 years old lady presented with a smooth slow-growing subcutaneous nodule on the anteromedial side of the right thigh with no detectable primary site in the liver or lung. The case was subsequently diagnosed as hydatid cyst of muscle and radical surgery was done under coverage of anihelminthic drug. The common practice in this type of case is to do fine needle aspiration cytology (FNAC) taking the lesion to be a soft tissue neoplasm. The aim of this case presentation is to be aware of that in case of a diffuse non-tender swelling with history of gradual increase in size, hydatid cyst should be considered in the differential diagnosis.
