A major breakthrough in the field of rare earth element (REE) geochemistry has been the recent discovery of their utility to microbial life, as essential metalloenzymes catalyzing the oxidation of methanol to formaldehyde. Lanthanide-dependent bacteria are thought to be ubiquitous in marine and terrestrial environments, but direct field evidence of preferential microbial utilization of REE in natural systems is still lacking. In this study, we report on the REE and trace element composition of the tube of a siboglinid worm collected at a methane seep in the Gulf of Guinea; a tube-dwelling annelid that thrives in deep-sea chemosynthetic ecosystems. High-resolution trace element profiles along the chitin tube indicate marked enrichments of lanthanum (La) and cerium (Ce) in its oxic part, resulting in REE distribution patterns that depart significantly from the ambient seawater signature. Combined with various geochemical and microbiological evidence, this observation provides direct support for an active consumption of light-REE at cold seeps, associated with the aerobic microbial oxidation of methane. To further evaluate this hypothesis, we also re-examine the available set of REE data for modern seep carbonates worldwide. While most carbonate concretions at cold seeps generally display REE distribution patterns very similar to those for reduced pore waters in marine sediments, we find that seafloor carbonate pavements composed of aragonite commonly exhibit pronounced light-REE enrichments, as inferred from high shale-normalized La/Gd ratio (>~0.8), interpreted here as possibly reflecting the signature of lanthanide-dependent methanotrophic activity. This finding opens new perspectives for revisiting REE systematics in ancient seep carbonates and other microbialites throughout the Earth's history. In particular, the geochemical imprint of aerobic methane oxidation could be possibly traced using REE in Archaean stromatolites and other archives of Precambrian seawater chemistry, potentially providing new insights into the oxygenation of early Earth's oceans and associated microbiogeochemical processes.

The Regab pockmark is a large cold seep area located 10 km north of the Congo deep sea channel at about 3160 m water depth. The associated ecosystem hosts abundant fauna, dominated by chemosynthetic species such as the mussel Bathymodiolus aff. boomerang, vestimentiferan tubeworm Escarpia southwardae, and vesicomyid clams Laubiericoncha chuni and Christineconcha regab. The pockmark was visited during the West African Cold Seeps (WACS) cruise with RV Pourquoi Pas? in February 2011, and a 14,000-m 2 high-resolution videomosaic was constructed to map the most populated area and to describe the distribution of the dominant megafauna (mussels, tubeworms and clams). The results are compared with previous published works, which also included a videomosaic in the same area of the pockmark, based on images of the BIOZAIRE cruise in 2001. The 10-year variation of the faunal distribution is described and reveals that the visible abundance and distribution of the dominant megafaunal populations at Regab have not changed significantly, suggesting that the overall methane and sulfide fluxes that reach the faunal communities have been stable. Nevertheless, small and localized distribution changes in the clam community indicate that it is exposed to more transient fluxes than the other communities. Observations suggest that the main megafaunal aggregations at Regab are distributed around focused zones of high flux of methane-enriched fluids likely related to distinct smaller pockmark structures that compose the larger Regab pockmark. Although most results are consistent with the existing successional models for seep communities, some observations in the distribution of the Regab mussel population do not entirely fit into these models. This is likely due to the high heterogeneity of this site formed by the coalescence of several pockmarks. We hypothesize that the mussel distribution at Regab could also be controlled by the occurrence of zones of both intense methane fluxes and reduced efficiency of the anaerobic oxidation of methane possibly limiting tubeworm colonization.

This work provides new insights on different aspects of seabed fluid flow processes based on seafloor observations. The methods used entirely rely on ROV-based high-resolution imaging and mapping techniques. Optical data are used to produce visual maps of the seafloor, in the form of geo-referenced video- and photo-mosaics, whereas acoustic techniques allow mapping the micro-bathymetry of the seabed, as well as the signal reflectivity of the sediment surface and of the water column. This work presents three case studies, about two sites of seabed fluid flow: the Menez Gwen hydrothermal vent on the MAR and the REGAB pockmark in the Lower Congo Basin. On the technical side, some of the high-resolution techniques used in this thesis are not commonly used by the marine scientific community. This is particularly the case for large-area photo-mosaics. Although the interest in mosaicking is growing, there are still no tools freely and readily available to scientists to routinely construct large-area photo-mosaics. Therefore, this work presents a MATLAB toolbox for large-area photo-mosaicking (LAPM toolbox), which was developed as part of this thesis.

The microbial diversity in sediments from four different assemblages of chemosynthetic organisms, Mytilidae, Vesicomyidae, Siboglinidae and a bacterial mat, was investigated using comparative 16S rRNA gene sequence analysis. Aggregates of anaerobic methanotrophic archaea (ANME-2) and bacteria of the Desulfosarcina/Desulfococcus cluster were found in all four chemosynthetic habitats. Fluorescence in situ hybridization targeting the ANME-2/Desulfosarcina/Desulfococcus aggregates showed their presence few centimeters (3-5 cm) below the surface of sediment. 16S rRNA gene sequences from all known marine ANME groups were detected in the pockmark sediments, as well as from both known bacterial partners. The archaeal diversity was limited to the ANME cluster for all investigated samples. The bacterial diversity included members of the Proteobacteria, Bacilliales, Cytophaga/Flavobacteria, Verrucomicrobia, JS1 and Actinobacteria clusters. Bacterial 16S rRNA gene sequences related to those of known sulphide-oxidizing symbionts were recovered from tissues of several invertebrates including vesicomyid clams and siboglinid tubeworms of REGAB.

International audience; During the ZaïRov2 cruise of the ZaïAngo project (1998-2000) on the passive Congo-Angola margin, several gravity cores were analyzed for helium isotopic composition of sedimentary pore waters in two cold fluid seepage zones: the Astrid slide area and the Regab giant pockmark. Gas concentration and isotopic composition are presented along with thermal data in terms of the origin and circulation of fluids. Helium isotope data lie on a mixing line between bottom seawater and an almost pure radiogenic. Helium and temperature vertical profiles are well described by the classic diffusion-advection equation. On the basis of He profiles, we estimate the advection rate on the rim of the pockmark between 1.2 and 2.3 mm/a. The He flux derived for a pure diffusive regime (2.4 Â 10 À8 mol/m 2 /a) can favorably be compared to literature data and contrasts with the flux computed close to the pockmark center (1.9 Â 10 À7 mol/m 2 /a). Helium depth profiles turned to be more sensitive to advection rate than temperature profiles are.
