Background and aimsThe precise roles of screen media activity (SMA) and sleep problems in relation to child/adolescent psychopathology remain ambiguous. We investigated temporal relationships among sleep problems, SMA, and psychopathology and potential involvement of thalamus-prefrontal-cortex (PFC)-brainstem structural covariation.MethodsThis study utilized data from the Adolescent Brain Cognitive Development study (n = 4,641 ages 9-12) at baseline, Year1, and Year2 follow-up. Cross-Lagged Panel Models (CLPMs) investigated reciprocal predictive relationships between sleep duration/problems, SMA, and psychopathology symptoms. A potential mediating role of baseline Thalamus-PFC-brainstem covariation on SMA-externalizing relationships was examined.ResultsParticipants were divided into discovery (n = 2,359, 1,054 girls) and replication (n = 2,282, 997 girls) sets. CLPMs showed 1) bidirectional associations between sleep duration and SMA in late childhood, with higher frequency SMA predicting shorter sleep duration (β = -0.10 [95%CI: -0.16, -0.03], p = 0.004) and vice versa (β = -0.11 [95%CI: -0.18, -0.05], p < 0.001); 2) externalizing symptoms at age 10-11 predicting sleep problems (β = 0.11 [95%CI: 0.04, 0.19], p = 0.002), SMA (β = 0.07 [95%CI: 0.01, 0.13], p = 0.014), and internalizing symptoms (β = 0.09 [95%CI: 0.05, 0.13], p < 0.001) at age 11-12; and 3) externalizing behavior at age 10-11 partially mediating the relationship between baseline thalamus-PFC-brainstem covariation and SMA at age 11-12 (indirect effect = 0.032 [95%CI: 0.003, 0.067], p-value = 0.030). Findings were replicable.ConclusionWe found bi-directional SMA-sleep-duration associations in late childhood. Externalizing symptoms preceded future SMA and sleep disturbances and partially mediated relationships between structural brain covariation and SMA. The findings emphasize the need for understanding individual differences and developing and implementing integrated strategies addressing both sleep concerns and screen time to mitigate potential impacts on psychopathology.

Abstract Objectives To analyse clinical outcomes of a non-medical switch from originator adalimumab (ADA) to its ABP501 biosimilar (ABP) over 6 months in patients with inflammatory rheumatic musculoskeletal diseases (RMD) in relation to comorbidity as a risk factor for therapy discontinuation. Methods RMD patients switching from originator ADA to ABP were identified from a large routine database from October 2018 onwards. Documented clinical data at the time of non-medical switching (baseline), and at 3 and 6 months were collected. Comorbidities were represented by the Charlson Comorbidity Index (CCI) at baseline and patients were categorized based on CCI > 0. Differences in the ABP retention rate over 6 months between patients with CCI = 0 and patients with CCI > 0 were analysed using Bayesian exponential regression. Results A total of 111 patients with axial spondyloarthritis (n = 68), rheumatoid arthritis (n = 23) and psoriatic arthritis (n = 15), were identified, 74.8% of whom had continued treatment with ABP after 6 months, while a smaller proportion had either switched to another ADA biosimilar (10.8%), switched back to originator ADA (7.2%), switched to a different biologic (3.6%), or dropped out (3.6%). At baseline, a CCI > 0 was found in 38% of patients. Cardiovascular comorbidities (40%) were most prevalent followed by diseases of the skin (33%), the gastrointestinal tract (20%) and the eye (20%). ABP treatment was continued after 6 months in 74% of patients with CCI = 0 and in 76% with CCI > 0. Bayesian analysis showed only a small difference (months) in the APB continuation rate between groups (estimate 0.0012, 95% credible interval (CrI) -0.0337 to 0.0361). Adjusting for age, sex, and disease subtype revealed somewhat shorter retention rates for patients with CCI > 0, but the distribution of the difference included 0 (estimate -0.0689, 95% CrI -0.2246 to 0.0234). Conclusion In a non-medical switch scenario of RMD patients, there was no evidence for a considerable difference in ABP retention rates over 6 months between comorbidity groups.

Abstract Recent advances in DNA nanotechnology allow for the assembly of nanocomponents with nanoscale precision, leading to the emergence of DNA-based material fabrication approaches. Yet, transferring these nano- and micron-scale structural arrangements to the macroscale morphologies remains a challenge, which limits the development of materials and devices based on DNA nanotechnology. Here, we demonstrate a materials fabrication approach that combines DNA-programmable assembly with actively driven processes controlled by acoustic fields. This combination provides a prescribed nanoscale order, as dictated by equilibrium assembly through DNA-encoded interactions, and field-shaped macroscale morphology, as regulated by out-of-equilibrium materials formation through specific acoustic stimulation. Using optical and electron microscopy imaging and x-ray scattering, we further revealed the nucleation processes, domain fusion, and crystal growth under different acoustically stimulated conditions. The developed approach provides a pathway for the fabrication of complexly shaped macroscale morphologies for DNA-programmable nanomaterials by controlling spatiotemporal characteristics of the acoustic fields.

Despite growing descriptions of wild-type Huntingtin (wt-HTT) roles in both adult brain function and, more recently, development, several clinical trials are exploring HTT-lowering approaches that target both wt-HTT and the mutant isoform (mut-HTT) responsible for Huntington's disease (HD). This non-selective targeting is based on the autosomal dominant inheritance of HD, supporting the idea that mut-HTT exerts its harmful effects through a toxic gain-of-function or a dominant-negative mechanism. However, the precise amount of wt-HTT needed for healthy neurons in adults and during development remains unclear. In this study, we address this question by examining how wt-HTT loss affects human neuronal network formation, synaptic maturation, and homeostasis in vitro. Our findings establish a role of wt-HTT in the maturation of dendritic arborization and the acquisition of network-wide synchronized activity by human cortical neuronal networks modeled in vitro. Interestingly, the network synchronization defects only became apparent when more than two-thirds of the wt-HTT protein was depleted. Our study underscores the critical need to precisely understand wt-HTT role in neuronal health. It also emphasizes the potential risks of excessive wt-HTT loss associated with non-selective therapeutic approaches targeting both wt- and mut-HTT isoforms in HD patients.

Abstract Background WNT signaling is pivotal in embryogenesis and tissue homeostasis. Aberrant WNT signaling, due to mutations in components of this pathway, contributes to the development and progression of human cancers, including colorectal cancer. AXIN2, encoded by the AXIN2 gene, is a key negative regulator and target of the canonical WNT signaling pathway. Germline mutations in AXIN2 are associated with absence of permanent teeth (hypo- and oligodontia) and predisposition to gastrointestinal polyps and cancer. The limited number of patients makes an accurate genotype–phenotype analysis currently challenging. Case presentation We present the case of a 55-year-old male with colorectal polyposis and hypodontia. Genetic testing confirmed a novel frameshift germline mutation in exon 8 of the AXIN2 gene. In addition, we provide an updated overview of germline AXIN2 mutations reported in literature. Conclusions Although the number of missing teeth is less severe in our patient than in some previously reported cases, our findings provide additional evidence that missing teeth and gastrointestinal neoplasia are associated with rare pathogenic AXIN2 germline mutations.

The Net Zero Industry Act (NZIA) was touted as the European Union’s big response to the US Inflation Reduction Act. A year ago, the Biden administration’s new green subsidy program spooked the EU into a flurry of industrial policy announcements. Now, the political dust has settled, and the EU’s main green industry initiatives will finally hit the legislative books. So, what has become of the EU’s new green industry agenda, and what can we learn from it about Europe’s role in the new global age of industrial policy?

In order to achieve the Paris climate targets, the German government has developed phased plans and anchored these in an amendment to the Renewable Energy Sources Act (EEG). This not only defines expansion targets for renewable energy producers, but also the additional amounts of energy that must be available in the electricity grid in future if it is to supply the mobility and building heating sectors, which are currently still powered by fossil fuels. While the lack of grid transport capacities is recognized and discussed in the public debate, another known and critical hurdle is rarely presented here. This is the temporal distribution of power from renewable energies and its effects on the processes involved in controlling the grid balance. By analyzing a target scenario specified in the EEG for 2030 and another for 2040+, which a German economic research institute recommends to policymakers, the aim is to investigate whether their targets can be achieved under the conditions of the natural fluctuations occurring in Germany and the quantities of renewable energy that can be harvested with a reasonable use of resources. The focus is on simulating future generation curves using historical data in such a way that they show the typical weather-driven behaviour to be expected in Germany. Unavoidable Surplus generation is used as efficiently as possible with electricity and hydrogen storage systems. Both scenarios fall considerably short of their targets. CO2-free coverage of the demand specified for the long-term future is only possible with a quantity of generators that is at the limit of what is feasible. The reason for this is that the current plans are obviously based on an unrealistic picture of the temporal distribution of renewable energy output. Achieving the transformation goals in Germany could be much more complex than politicians currently believe and may require a change in strategy sooner or later.

Glacial groundwater can mobilize deep-seated methane from beneath glaciers and permafrost in the Arctic, leading to atmospheric emissions of this greenhouse gas. We present a temporal, hydro-chemical dataset of methane-rich groundwater collected during two melt seasons from a high Arctic glacial forefield to explore the seasonal dynamics of methane emissions. We use methane and ion concentrations and the isotopic composition of water and methane to investigate the sources of groundwater and the origin of the methane that the groundwater transports to the surface. Our results suggest two sources of groundwater, one shallow and one deep, which mix, and moderate methane dynamics. During summer, deep methane-rich groundwater is diluted by shallow oxygenated groundwater, leading to some microbial methane oxidation prior to its emergence at the surface. Characterization of the microbial compositions in the groundwater shows that microbial activity is an important seasonal methane sink along this flow-path. In the groundwater pool studied, we found that potential methane emissions were reduced by an average of 29% (±14%) throughout the summer due to microbial oxidation. During winter, deep groundwater remains active while many shallow systems shut down due to freezing, reducing subsurface methane oxidation, and potentially permitting larger methane emissions. Our results suggest that ratios of the different groundwater sources will change in the future as aquifer capacities and recharge volumes increase in a warming climate.

The rare decay K_{L}→π^{0}νν[over ¯] was studied with the dataset taken at the J-PARC KOTO experiment in 2016, 2017, and 2018. With a single event sensitivity of (7.20±0.05_{stat}±0.66_{syst})×10^{-10}, three candidate events were observed in the signal region. After unveiling them, contaminations from K^{±} and scattered K_{L} decays were studied, and the total number of background events was estimated to be 1.22±0.26. We conclude that the number of observed events is statistically consistent with the background expectation. For this dataset, we set an upper limit of 4.9×10^{-9} on the branching fraction of K_{L}→π^{0}νν[over ¯] at the 90% confidence level.

Astronauts undertaking long-duration space missions may be vulnerable to unique stressors that can impact human aging. Nevertheless, few studies have examined the relationship of mission duration with DNA-methylation-based biomarkers of aging in astronauts. Using data from the six participants of the Mars-500 mission, a high-fidelity 520-day ground simulation experiment, we tested relationships of mission duration with five longitudinally measured blood DNA-methylation-based metrics: DNAmGrimAge, DNAmPhenoAge, DNA-methylation-based estimator of telomere length (DNAmTL), mitotic divisions (epigenetic mitotic clock [epiTOC2]), and pace of aging (PoA). We provide evidence that, relative to baseline, mission duration was associated with significant decreases in epigenetic aging. However, only decreases in DNAmPhenoAge remained significant 7 days post-mission. We also observed significant changes in estimated proportions of plasmablasts, CD4T, CD8 naive, and natural killer (NK) cells. Only decreases in NK cells remained significant post-mission. If confirmed more broadly, these findings contribute insights to improve the understanding of the biological aging implications for individuals experiencing long-duration space travel.

Objective The objective of this study is to build a structural model visualising and quantifying the interrelationships of different disease outcomes with the Assessment of SpondyloArthritis International Society Health Index (ASAS HI) in patients with axial spondyloarthritis (axSpA).Methods Cross-sectional data collected at month 72 of the Devenir des Spondylarthropathies Indifferénciées Récentes cohort was analysed. Combining prior knowledge and observed data, probabilistic Bayesian network modelling was used to study how the interplay of different disease outcomes affects the ASAS HI, which measures disease-specific overall functioning and health. Disease outcomes comprised, among others, the Ankylosing Spondylitis (AS) Disease Activity Score (ASDAS) and the Bath AS Functional Index (BASFI).Results Data of 384 patients were analysed. The obtained structure suggests that ASAS HI is determined by both patient-reported physical function (BASFI) and disease activity (ASDAS). The parameters of the structural model show that an increase of ASDAS or BASFI by 1 unit corresponds to an increase of ASAS HI by 0.70 or 1.25 units, respectively. Moreover, the model suggests that disease activity has an indirect impact on ASAS HI via BASFI. No relationship between spinal mobility or structural damage and ASAS HI was found.Conclusions This is the first structural model developed to better understand the construct and the interplay between clinically relevant outcomes related to ASAS HI in axSpA patients. It shows that disease activity and physical function have a strong impact on ASAS HI, confirming it to be a valid construct of overall functioning and health in axSpA patients.

Scientific innovations for sustainable development often make huge promises about overcoming climate change, or promote technological innovations that have potential development implications. However, the social outcomes of such interventions are often considered superficially; thus potentially reinforcing injustices or producing unforeseen, undesirable outcomes. Increasingly, projects claim to have an interdisciplinary dimension, but processes of interdisciplinary dialogue can be difficult to maintain. This means that researchers have to be prepared to engage in potentially extensive, transformative dialogue and be willing to learn from the disciplinary knowledge of the other. This can be methodologically challenging both for natural and social scientists. This paper reports meta-research exploring the methodological processes and learning experiences of academics involved in research on the implementation of second-generation biofuel technology. Through interdisciplinary meetings, workshops, reflections and interviews we built up sustainable relationships and bridged epistemological divides. Our approach to interdisciplinary dialogue offers insights into the methodological challenges of interdisciplinary work. We used the framework of transformative learning theory to identify key aspects of the interdisciplinary process and reflected on the need for sustained and open opportunities for dialogue in order to find genuine ways to communicate across disciplines. We explicitly revealed and considered our taken-for-granted assumptions to identify what we understood by key terms and processes, including ‘sustainable’, ‘development’, ‘methodology’, ‘truth’, ‘marginal land’ and ‘outputs’. We found these encounters created opportunities to influence the trajectory of each other’s research and thinking, with the ideal of social and environmental justice prominent in all our discussions. It was through ongoing learning and dialogue that we found that the multifaceted challenges of sustainable development research can become more open, more critical and more able to reveal appropriate solutions and promote relevant ongoing scientific research. This interplay between disciplines is an innovative way to influence decision-making directly.

Background: Previous experiences with non-medical switching of adalimumab (ADA) in patients with chronic inflammatory rheumatic diseases (CIRD) come mainly from phase III extension of randomised clinical trials and little from routine care. Objectives: To analyse treatment trajectories over 2 years in patients with CIRD conducting a non-medical switch from originator to biosimilar ADA. Design: A retrospective observational cohort study was conducted with data from a third-level rheumatology centre in Germany. CIRD patients on originator ADA who switched to ADA biosimilar from October 2018 onwards were identified and followed until September 2020. Methods: Patients’ characteristics were compared between the four a priori defined treatment trajectories ‘continued biosimilar ADA therapy’, ‘back-switch to originator ADA therapy’, ‘switch to another biological disease-modifying anti-rheumatic drug (bDMARD) therapy’ and ‘stopped bDMARD therapy/death/drop out’. Factors associated with continuing biosimilar ADA therapy were analysed using Cox proportional hazards regression analyses. Results: A total of 121 CIRD patients were included. Most patients (66.9%) continued therapy with biosimilar ADA over 2 years, with a treatment retention rate of 73.1%. Whereas 21 patients (17.4%) switched back to originator ADA, mainly due to adverse events, and 8 patients (6.6%) switched to a different bDMARD, mainly due to lack of effect. The estimated risk of withdrawal was lower for longer prior duration on originator ADA [hazard ratio (HR): 0.82; 95% CI: 0.69–0.97] and higher for higher C-reactive protein levels at baseline (HR: 1.18; 95% CI: 1.00–1.39). Male patients, older patients and those for whom originator ADA was their first bDMARD tended to have a lower risk of withdrawal. Conclusion: Our results indicated that three of four patients continue biosimilar ADA over 2 years with lower risks of withdrawal for male sex, older age, longer prior duration on originator ADA and originator ADA as first bDMARD.

BackgroundChronic diseases, such as opioid use disorder (OUD) require a multifaceted scientific approach to address their evolving complexity. The Council for the Advancement of Nursing Science's (Council) four nursing science priority areas (precision health; global health, determinants of health, and big data/data analytics) were established to provide a framework to address current complex health problems.PurposeTo examine OUD research through the nursing science priority areas and evaluate the appropriateness of the priority areas as a framework for research on complex health conditions.MethodOUD was used as an exemplar to explore the relevance of the nursing science priorities for future research.FindingsResearch in the four priority areas is advancing knowledge in OUD identification, prevention, and treatment. Intersection of OUD research population focus and methodological approach was identified among the priority areas.DiscussionThe Council priorities provide a relevant framework for nurse scientists to address complex health problems like OUD.

Abstract Semiarid rangelands are identified as at high risk of degradation due to anthropogenic pressure and climate change. Through tracking timelines of degradation we aimed to identify whether degradation results from a loss of resistance to environmental shocks, or loss of recovery, both of which are important prerequisites for restoration. Here we combined extensive field surveys with remote sensing data to explore whether long-term changes in grazing potential demonstrate loss of resistance (ability to maintain function despite pressure) or loss of recovery (ability to recover following shocks). To monitor degradation, we created a bare ground index: a measure of grazeable vegetation cover visible in satellite imagery, allowing for machine learning based image classification. We found that locations that ended up the most degraded tended to decline in condition more during years of widespread degradation but maintained their recovery potential. These results suggest that resilience in rangelands is lost through declines in resistance, rather than loss of recovery potential. We show that the long-term rate of degradation correlates negatively with rainfall and positively with human population and livestock density, and conclude that sensitive land and grazing management could enable restoration of degraded landscapes, given their retained ability to recover.

A fluorinated thiol was systematically investigated against a diverse set of –ene- and –yne-bearing imidazole and imidazolium substrates in the pursuit of new cation-fluorinated, omniphobic (hydrophobic and oleophobic) ionic liquids for aerospace applications. Compounds were prepared using UV-initiated click chemistry at quantitative conversion and purified with relative ease. Seven ionic liquids were synthesized and have isolated yields ranging from 74% to >99%. Five of the new cation-fluorinated ionic liquids are hydrophobic and two are liquids at room temperature. In addition to hydrophobicity, the seven ionic liquids’ solubilities were qualitatively examined across a wide spectrum of conventional organic solvents. Of note, the majority of the prepared ionic liquids are insoluble in water and hexanes yet soluble in methanol, acetone, acetonitrile, and dichloromethane. In addition to intriguing solubility properties, the ionic liquids in this study have high densities without the incorporation of heavy atoms, ranging from 1.596 to 1.924 g cm−3. The fluorinated salts also show good short-term thermal stability in air, with decomposition temperatures ranging from 215 °C to 320 °C. Other relevant characterization methods include spectroscopy, thermal analysis, and physical property measurements where appropriate. The X-ray crystal structure of a divalent, cation-fluorinated ionic liquid from this study was also obtained and shows unique fluorous domains in the extended structure, providing valuable insight into the materials’ physical properties. This work highlights a family of compounds moving towards omniphobic ionic liquids yielding potential possibilities for future applications.
