The human macula is a highly specialized retinal region with pit‐like morphology and rich in cones. How Müller cells, the principal glial cell type in the retina, are adapted to this environment is still poorly understood. We compared proteomic data from cone‐ and rod‐rich retinae from human and mice and identified different expression profiles of cone‐ and rod‐associated Müller cells that converged on pathways representing extracellular matrix and cell adhesion. In particular, epiplakin (EPPK1), which is thought to play a role in intermediate filament organization, was highly expressed in macular Müller cells. Furthermore, EPPK1 knockout in a human Müller cell‐derived cell line led to a decrease in traction forces as well as to changes in cell size, shape, and filopodia characteristics. We here identified EPPK1 as a central molecular player in the region‐specific architecture of the human retina, which likely enables specific functions under the immense mechanical loads in vivo.

Keratins exert important structural but also cytoprotective functions. They have to be adaptable to support cellular homeostasis. Epiplakin (EPPK1) has been shown to decorate keratin filaments in epithelial cells and to play a protective role under stress, but the mechanism is still unclear. Using live-cell imaging of epithelial cells expressing fluorescently tagged EPPK1 and keratin, we report here an unexpected dynamic behavior of EPPK1 upon stress. EPPK1 was diffusely distributed throughout the cytoplasm and not associated with keratin filaments in living cells under standard culture conditions. However, ER-, oxidative and UV-stress, as well as cell fixation, induced a rapid association of EPPK1 with keratin filaments. Photopolymerizations, in which the initiation of a chemical-physical reaction occurs by the exposure of photosensitive monomers to a high-intensity light source, have become a well-accepted technology for manufacturing polymers. Providing significant advantages over thermal-initiated polymerizations, including fast and controllable reaction rates, as well as spatial and temporal control over the formation of material, this technology has found a large variety of industrial applications. The reaction mechanisms and kinetics are quite complex as the system moves quickly from a liquid monomer mixture to a solid polymer. Therefore, the study of curing kinetics is of utmost importance for industrial applications, providing both the understanding of the process development and the improvement of the quality of parts manufactured via photopolymerization. Consequently, this review aims at presenting the materials and curing chemistry of such ultrafast crosslinking polymerization reactions as well as the research efforts on theoretical models to reproduce cure kinetics and mechanisms for free-radical and cationic photopolymerizations including diffusion-controlled phenomena and oxygen inhibition reactions in free-radical systems.

Background: Currently, the clinical outcome of patients with very severe head injuries and subsequent chronic disorders of consciousness is seen as generally very poor. Here, we specify individual outcome chances for disorders of consciousness (DOC) patients on the basis of clinical and EEG data and identify different subgroups of DOC patients, who vary substantially regarding their outcome chances. Methods: We employed data from 102 patients with DOC and used standard clinical protocol data (age, etiology, diagnosis, gender) and early sensory (N100, Mismatch Negativity) and late cognitive (P300, N400) EEG event related potentials (ERPs) to predict patients recovery rates. Results: Two significant prediction models emerged: In both, subgroups of patients with good (51%, tree 1) to very good recovery chances (97%, tree 2) could be identified. The first model was obtained from standard clinical data. The second model included late cognitive ERPs (N400 and P300) and resulted in considerably better patient classification. Moreover, when taking cognitive event related potentials into account, the standard protocol data did not add further significant information, neither did early ERPs (N100, MMN). This highlights that cognitive ERPs provide decisive information about an individual patient’s prognosis. Conclusion: The presented information about outcome chances of individual disorders of consciousness patients will be vital for these patients and critical for clinical professionals who have to direct specialized treatments and council relatives. Legal guardians and families, in turn, need to know what to expect in the future in order to prepare for the challenges ahead. Compositional grading has been widely exploited in highly efficient Cu(In,Ga)Se2, CdTe, GaAs, quantum dot solar cells, and this strategy has the potential to improve the performance of emerging perovskite solar cells. However, realizing and maintaining compositionally graded perovskite absorber from solution processing is challenging. Moreover, the operational stability of graded perovskite solar cells under long‐term heat/light soaking has not been demonstrated. In this study, a facile partial ion‐exchange approach is reported to achieve compositionally graded perovskite absorber layers. Incorporating compositional grading improves charge collection and suppresses interface recombination, enabling to fabricate near‐infrared‐transparent perovskite solar cells with power conversion efficiency of 16.8% in substrate configuration, and demonstrate 22.7% tandem efficiency with 3.3% absolute gain when mechanically stacked on a Cu(In,Ga)Se2 bottom cell. Non‐encapsulated graded perovskite device retains over 93% of its initial efficiency after 1000 h operation at maximum power point at 60 °C under equivalent 1 sun illumination. The results open an avenue in exploring partial ion‐exchange to design graded perovskite solar cells with improved efficiency and stability.

One of the most significant limitations for a wider utilisation of the renewable and CO2-storing resource wood is its low ultraviolet (UV) light stability. The protection of the wood surface without altering its aesthetic appeal requires an optically transparent but UV protective coating which should be strongly attached to the rough and inhomogeneous substrate. For this purpose, ZnO nanostructures were deposited onto the wood surface via a chemical bath deposition process. The morphology of crystalline ZnO was controlled by aluminium nitrate or ammonium citrate in the growth step resulting in nanorod arrays or platelet structures, respectively. Detailed structural, chemical and mechanical characterisations as well as accelerated weathering exposure revealed the effective performance of the platelet structure, which formed a dense and thin ZnO coating on spruce. The total colour change (ΔE in the CIE system) was calculated to be 20.5 for unmodified wood, while it was about three for the modified samples after 4 weeks accelerated weathering test. Inverted PTB7/PC71BM polymer solar cells are prepared on solution-processed Al:ZnO transparent contacts on PET substrates. Al:ZnO is deposited by a low temperature chemical bath deposition route (T< 100°C at any step) to comply with the temperature sensitive substrate. A maximum conversion efficiency of 6.4% and 6.9% is achieved for the indium-free solar cells on PET and glass substrates, respectively. The devices are relatively stable in air whereby an initial efficiency loss in the order of 15% after storage for 15 days can be fully recovered by light soaking.

The electrical properties of polycrystalline ZnO thin films with grain boundary limited charge transport depend significantly on the degree of chemisorbed adsorbate (CxOyHz) coverage at grain boundaries and on the surface. Here we report the evolution of carbon adsorbates in chemical bath deposited (CBD) aluminum doped ZnO (AZO) thin films during post-deposition UV light exposure and degradation in humid atmosphere. As CBD AZO intrinsically contains carbon impurities stemming from the addition of citrate during the deposition, sputtered AZO is examined in parallel as a nominally carbon-free reference. UV illumination decomposes citrate impurities in CBD AZO, but the residual carbon compounds are not effectively removed from the bulk part of the layer. At the same time, charge-trapping oxygen species at grain boundaries are desorbed, increasing the carrier density and mobility. Exposure to humid atmosphere decreases the electrical conductivity of CBD AZO, which is caused by the chemisorption of environmental oxygen species and the formation of zinc carbonate.

Subdivision comparisons of mass measurement are usually used in the mass dissemination for class $E_{1}$ mass standards from 1 kg to 1 mg or from 1 to 50 kg. Many equations of mass measurement are included in the subdivision comparisons. Therefore, matrix calculations for mass measurement are very convenient to get the true or conventional mass value of every weight compared. In conventional matrix equations, the important air buoyancy corrections are completely neglected, and uncertainties are normally calculated using the variance–covariance matrix. In this paper, we propose a new matrix calculation for both the system of weighing equations as well as the corresponding uncertainty budget using directly partial derivations of the weighting equations. The method is applied to a set of weights from 500 to $50~\mu $ g. An alternative method for evaluating uncertainties of matrix equations is by Monte Carlo simulation, introduced in this paper, to validate the calculation of uncertainty budget by means of the matrix method. Evaluating and improving the performance of mix-based anonymity systems in a real-world setting is critical to foster their adoption. However, current research in this field mostly employs unrealistic models for evaluation purposes. Moreover, previously documented results are often difficult to reproduce. We propose two complementary models tailored to the evaluation of mix-based anonymity services. The models enable realistic experiments and are easy to use as they allow to automatically extract workloads from trace files recorded in real networks and replay them in simulations. We also describe our ready-to-use open source evaluation suite that implements the models. Given the suite, researchers can easily create and re-use well-defined workload sets for evaluation purposes. The workloads can be replayed both in discrete-event simulations and distributed experiments. With this initiative we want to foster open research in our discipline.

Solution processing of inorganic thin films has become an important thrust in material research community because it offers low-cost and high-throughput deposition of various functional coatings and devices. Especially inorganic thin film solar cells – macroelectronic devices that rely on consecutive deposition of layers on large-area rigid and flexible substrates – could benefit from solution approaches in order to realize their low-cost nature. This article critically reviews existing deposition approaches of functional layers for chalcogenide solar cells with an extension to other thin film technologies. Only true solutions of readily available metal salts in appropriate solvents are considered without the need of pre-fabricated nanoparticles. By combining three promising approaches, an air-stable Cu(In,Ga)Se2 thin film solar cell with efficiency of 13.8% is demonstrated where all constituent layers (except the metal back contact) are processed from solutions. Notably, water is employed as the solvent in all steps, highlighting the potential for safe manufacturing with high utilization rates.

Large-grained CuInSe2 absorber layers are synthesized using a non-vacuum process based on nanoparticle ink precursors and selenization by rapid thermal processing (RTP). The use of hydroxide-based particles in organic solvents allows for the conversion with elemental selenium without the need to employ explosive and/or toxic H2 or H2Se gasses. Lateral grain sizes up to 4 µm are obtained through a novel RTP route, overcoming the inherently high layer porosity for previous nanoparticle processes. Morphological and elemental characterization at interrupted selenization steps suggests that liquid selenium can play a beneficial role in promoting layer densification and grain growth. Long carrier collection lengths in CuInSe2 enable notable conversion efficiencies, despite the low minority carrier lifetimes of below 1 ns. Record efficiencies up to 8.73% highlight the potential of low-cost, non-vacuum deposition of chalcopyrite absorber layers with safe and simple precursors and processing routes. Dysfunction of plectin, a 500-kD cytolinker protein, leads to skin blistering and muscular dystrophy. Using conditional gene targeting in mice, we show that plectin deficiency results in progressive degenerative alterations in striated muscle, including aggregation and partial loss of intermediate filament (IF) networks, detachment of the contractile apparatus from the sarcolemma, profound changes in myofiber costameric cytoarchitecture, and decreased mitochondrial number and function. Analysis of newly generated plectin isoform–specific knockout mouse models revealed that IF aggregates accumulate in distinct cytoplasmic compartments, depending on which isoform is missing. Our data show that two major plectin isoforms expressed in muscle, plectin 1d and 1f, integrate fibers by specifically targeting and linking desmin IFs to Z-disks and costameres, whereas plectin 1b establishes a linkage to mitochondria. Furthermore, disruption of Z-disk and costamere linkages leads to the pathological condition of epidermolysis bullosa with muscular dystrophy. Our findings establish plectin as the major organizer of desmin IFs in myofibers and provide new insights into plectin- and desmin-related muscular dystrophies.

Epiplakin is a large (>725 kDa) cytoskeletal protein exclusively expressed in epithelial tissues. It has a unique structure, consisting entirely of plakin repeat domains (PRDs), one of the hallmarks of spectraplakin protein family members. Previous studies, including the phenotypic analyses of knockout mice, failed to reveal the biological function of epiplakin. Using in vitro binding assays, we show here that all but one of the 16 PRDs of mouse epiplakin bind to keratins of basal keratinocytes. Nevertheless, in primary keratinocyte cell cultures, epiplakin only partially colocalized with keratin intermediate filament networks. However, upon application of cellular stress in the form of keratin hyperphosphorylation, osmotic shock or UV irradiation, the entire cytoplasmic epiplakin pool became associated with keratin. In response to such types of stress, epiplakin initially translocated to the still-intact keratin filament network and remained associated with keratin after its disruption and transformation into granular aggregates. Time-course experiments revealed that serine/threonine (okadaic acid) and tyrosine (orthovanadate) phosphatase inhibitor-induced filament disruption in differentiated keratinocytes proceeded faster in epiplakin-deficient cells compared with wild-type cells. Our data suggest that epiplakin plays a role in keratin filament reorganization in response to stress, probably by protecting keratin filaments against disruption in a chaperone-like fashion.

Plectin, a widespread intermediate filament-based cytolinker protein capable of interacting with a variety of cytoskeletal structures and plasma membrane-bound junctional complexes, serves essential functions in maintenance of cell and tissue cytoarchitecture. We have generated a mouse line bearing floxed plectin alleles and conditionally deleted plectin in stratified epithelia. This strategy enabled us to study the consequences of plectin deficiency in this particular type of tissues in the context of the whole organism without plectin loss affecting other tissues. Conditional knockout mice died early after birth, showing signs of starvation and growth retardation. Blistering was observed on their extremities and on the oral epithelium after initial nursing, impairing food uptake. Knockout epidermis was very fragile and showed focal epidermal barrier defects caused by the presence of small skin lesions. Stratification, proliferation and differentiation of knockout skin seemed unaffected by epidermis-restricted plectin deficiency. In an additionally generated mouse model, tamoxifen-induced Cre-ERT-mediated recombination led to mice with a mosaic plectin deletion pattern in adult epidermis, combined with microblister formation and epidermal barrier defects. Our study explains the early lethality of plectin-deficient mice and provides a model to ablate plectin in adult animals which could be used for developing gene or pharmacological therapies.

Epiplakin, a giant epithelial protein of >700 kDa, belongs to the plakin family of cytolinker proteins. It represents an atypical family member, however, as it consists entirely of plakin repeat domains but lacks any of the other domains commonly shared by plakins. Hence, its putative function as a cytolinker protein remains to be shown. To investigate epiplakin's biological role, we generated epiplakin-deficient mice by gene targeting in embryonic stem cells. Epiplakin-deficient mice were viable and fertile, without developing any discernible phenotype. Ultrastructurally, their epidermis revealed no differences compared to wild-type littermates, and cornified envelopes isolated from skin showed no alterations in shape or stability. Furthermore, neither embryonal formation nor later function of the epithelial barrier was affected. In primary cultures of epiplakin-deficient keratinocytes, the organization of actin filaments, microtubules, and keratin networks was found to be normal. Similarly, no alterations in keratin network organization were observed in simple epithelia of small intestine and liver or in primary hepatocytes. We conclude that, despite epiplakin's abundant and highly specific expression in stratified and simple epithelia, its absence in mice does not lead to severe skin dysfunctions, nor has it detectable consequences for keratin filament organization and cytoarchitecture of cells.

Plectin, a typical cytolinker protein, is essential for skin and skeletal muscle integrity. It stabilizes cells mechanically, regulates cytoskeleton dynamics, and serves as a scaffolding platform for signaling molecules. A variety of isoforms expressed in different tissues and cell types account for this versatility. To uncover the role of plectin 1, the major isoform expressed in tissues of mesenchymal origin, against the background of all other variants, we raised plectin isoform 1-specific antibodies and generated isoform-deficient mice. In contrast to plectin-null mice (lacking all plectin isoforms), which die shortly after birth because of severe skin blistering, plectin isoform 1-deficient mice were viable at birth, had a normal lifespan, and did not display the skin blistering phenotype. However, dermal fibroblasts isolated from plectin 1-deficient mice exhibited abnormalities in their actin cytoskeleton and impaired migration potential. Similarly, plectin 1-deficient T cells isolated from nymph nodes showed diminished chemotactic migration in vitro . Most strikingly, in vivo we found that leukocyte infiltration during wound healing was reduced in the mutant mice. These data show a specific role of a cytolinker protein in immune cell motility. Single isoform-deficient mice thus represent a powerful tool to unravel highly specific functions of plectin variants.

The widely expressed cytolinker protein plectin shows extensive isoform diversity both at the N-terminus and in the central part of the molecule. Judged on mRNA data, plectin variants lacking the central rod domain are expressed at a approximately 20-fold lower level than full-length proteins and their detection on the protein level can be difficult. Here we present data on the expression of plectin rodless isoforms in mouse brain and in rat glioma C6 cells on RNA and protein levels. Our data indicate that among the rodless variants expressed in neuronal tissues, those starting with exon 1c (plectin 1c) seem to be the most prominent ones. In addition, we show that similar to other monoclonal antibodies reported in the literature, the widely used mAb 7A8 recognizes an epitope within plectin's rod domain and therefore is unsuited to detect rodless variants of plectin.

We have recently identified PARP10 as a novel functional poly(ADP-ribose) polymerase . The gene encoding PARP10 is conserved in vertebrates but no orthologs were found in lower organisms. In addition to the poly(ADP-ribose) polymerase domain, PARP10 possesses several additional sequence motifs , including an RNA recognition motif and two ubiquitin interaction motifs. We characterized the murine genomic locus of the Parp10 gene. We noticed that 3′ Parp10 sequences overlapped with the plectin 1 gene in a head-to-tail arrangement. Detailed analyses revealed that the two most 3′ Parp10 exons (exons 10 and 11) are also used for plectin 1 . While these two exons code for part of the poly(ADP-ribose) polymerase domain in Parp10, they are noncoding for plectin 1 due to the lack of appropriate start codons . Furthermore our findings suggest that at least one of the plectin 1 promoters is located within intron 9 of the Parp10 gene.

Plectin is a large cytoskeletal linker protein expressed as several different isoforms from a highly complex gene. This transcript diversity is mainly caused by short 5'-sequences contained in alternative first exons. To elucidate the influence of these sequence differences and to determine potential differential functionality of the resulting protein forms, we conducted a systematic investigation of plectin isoforms on transcript and protein levels. Isoform expression was highly dependent on the different 5' ends, largely due to effects of the 5'-untranslated regions. Initiation of translation downstream of the expected start site led to loss of actin- and integrin beta4-binding in some isoforms. The small alternative N-terminal sequences (5-180 residues) profoundly affected the subcelluar localization of this >500 kDa protein. Specifically, plectin 1f was concentrated at focal adhesion contacts and plectin 1b was exclusively targeted to mitochondria, providing a connection of these organelles to intermediate filaments. Thus, with plectin as a model, we demonstrate a role for 5'-untranslated regions and alternative 5'-splicing as an important regulatory mechanism of protein expression and protein function.

The various plectin isoforms are among the major crosslinking elements of the cytoskeleton. The importance of plectin in epithelia is convincingly supported by the severe skin blistering observed in plectin-deficient humans and mice. Here, we identified plectin 1a (> 500 kDa), a full length plectin variant containing the sequence encoded by the alternative first exon 1a, as the isoform most prominently expressed in human and mouse keratinocytes. In skin sections and cultured keratinocytes, plectin 1a was shown to colocalize with hemidesmosomal structures. In contrast, a second isoform expressed in epithelia, plectin 1c, differing from 1a merely by a short N-terminal sequence, colocalized with microtubules. Expression of plectin 1a, but not of its N-terminal fragment alone, or of a third alternative full length isoform (plectin 1), restored the reduced number of hemidesmosome-like stable anchoring contacts in cultured plectin-null keratinocytes. Our results show for the first time that different isoforms of a cytolinker protein expressed in one cell type perform distinct functions. Moreover, the identification of plectin 1a as the isoform defects in which cause skin blistering in plectin-related genetic diseases, such as epidermolysis bullosa simplex MD and epidermolysis bullosa simplex Ogna, could have implications for the future development of clinical therapies for patients.

In this paper we present Laribus, a peer-to-peer network designed to detect local man-in-the-middle attacks against secure socket layer/transport layer security (SSL/TLS). With Laribus, clients can validate the authenticity of a certificate presented to them by retrieving it from different vantage points on the network. Unlike previous solutions, clients do not have to trust a central notary service nor do they have to rely on the cooperation of website owners. The Laribus network is based on a social network graph, which allows users to form notary groups that improve both privacy and availability. It integrates several well-known techniques, such as secret sharing, ring signatures, layered encryption, range queries, and a distributed hash table (DHT), to achieve privacy-aware queries, scalability, and decentralization. We present the design and core components of Laribus, discuss its security properties, and also provide results from a simulation-based feasibility study.

The chitinase gene chiA was identified on the Clostridium thermocellum genome downstream of the endoglucanase gene celA . It contains a catalytic module of glycosyl hydrolase family 18 and a cellulosomal dockerin module. Chi18A hydrolyzes aryl-acetyl-chito-oligosaccharides preferentially. In denaturing electrophoresis of purified cellulosomes, a single chitinase activity band was identified in zymograms and Western blots, indicating that Chi18A is the only chitinase in the cellulosome.

Epstein-Barr virus nuclear antigen 2 (EBNA2) and the Notch protein both function within the nucleus as transcriptional adaptor proteins. EBNA2 plays a key role during the immortalization of primary B-cells by Epstein-Barr virus (EBV). Notch proteins are involved in lymphomagenesis as well as in multiple cell fate decisions during tissue differentiation and development. Both, EBNA2 and Notch interact with the DNA binding protein RBP-J and thereby gain access to the promoter of their target genes. In order to identify regions within the J recombination signal sequence binding protein (RBP-J), that are relevant for either the Notch or the EBNA2 interaction, we have performed a mutational analysis of RBP-J. A library of RBP-J mutants was screened by a reverse two-hybrid system for alleles that fail to bind to either EBNA2 or Notch. The sequence analysis of these alleles reveals that a limited and particularly distinct number of amino-acid positions are relevant for either interaction only. Given the important role of RBP-J in B-cell immortalization, the EBNA2/RBP-J protein-protein interaction could be a candidate target for therapeutic intervention in EBV related diseases.

Gab1 is a substrate of the receptor tyrosine kinase c-Met and involved in c-Met–specific branching morphogenesis. It associates directly with c-Met via the c-Met–binding domain, which is not related to known phosphotyrosine-binding domains. In addition, Gab1 is engaged in a constitutive complex with the adaptor protein Grb2. We have now mapped the c-Met and Grb2 interaction sites using reverse yeast two-hybrid technology. The c-Met–binding site is localized to a 13–amino acid region unique to Gab1. Insertion of this site into the Gab1-related protein p97/Gab2 was sufficient to confer c-Met–binding activity. Association with Grb2 was mapped to two sites: a classical SH3-binding site (PXXP) and a novel Grb2 SH3 consensus-binding motif (PX(V/I)(D/N)RXXKP). To detect phosphorylation-dependent interactions of Gab1 with downstream substrates, we developed a modified yeast two-hybrid assay and identified PI(3)K, Shc, Shp2, and CRKL as interaction partners of Gab1. In a trk-met-Gab1–specific branching morphogenesis assay, association of Gab1 with Shp2, but not PI(3)K, CRKL, or Shc was essential to induce a biological response in MDCK cells. Overexpression of a Gab1 mutant deficient in Shp2 interaction could also block HGF/SF-induced activation of the MAPK pathway, suggesting that Shp2 is critical for c-Met/Gab1-specific signaling.

Plectin, the most versatile cytolinker identified to date, has essential functions in maintaining the mechanical integrity of skin, skeletal muscle and heart, as indicated by analyses of plectin-deficient mice and humans. Expression of plectin in a vast variety of tissues and cell types, combined with a large number of different binding partners identified at the molecular level, calls for complex mechanisms regulating gene transcription and expression of the protein. To investigate these mechanisms, we analyzed the transcript diversity and genomic organization of the murine plectin gene and found a remarkable complexity of its 5'-end structure. An unusually high number of 14 alternatively spliced exons, 11 of them directly splicing into plectin exon 2, were identified. Analysis of their tissue distribution revealed that expression of a few of them is restricted to tissues such as brain, or skeletal muscle and heart. In addition, we found two short exons tissue-specifically spliced into a highly conserved set of exons encoding the N-terminal actin binding domain (ABD), common to plectin and the superfamily of spectrin/dystrophin-type actin binding proteins. Using recombinant proteins we show that a novel ABD version contained in the muscle-specific isoform of plectin exhibits significantly higher actin binding activity than other splice forms. This fine tuning mechanism based on alternative splicing is likely to optimize the proposed biological role of plectin as a cytolinker opposing intense mechanical forces in tissues like striated muscle.

Evaluating and improving the performance of anonymity systems in a real-world setting is critical to foster their adoption. However, current research in this field mostly employs unrealistic models for evaluation purposes. Moreover, previously documented results are often difficult to reproduce. We propose two complementary workload models that operate on network traces in order to improve the evaluation of anonymity systems. In comparison to other approaches our workload models are more realistic, as they derive characteristics from trace files recorded in real networks and preserve dependencies of the flows of individual hosts. We also describe our ready-to-use open source evaluation suite that implements our models. Given our tools, researchers can easily create and re-use well-defined workload sets for evaluation purposes. Finally, we demonstrate the importance of realistic workload models by evaluating a well-known dummy traffic scheme with our tools.

The development of a novel chemistry for the chemical bath deposition of Zn(O,S) buffer layers for Cu(In,Ga)Se2 (CIGS) solar cells is desired for a higher growth rate, hence reduced deposition time, while reducing simultaneously the required concentration of reactants. State-of-the-art recipes are based on thiourea as sulphide precursor requiring a high molarity of reactants and relatively long deposition times due to the slow decomposition rate of thiourea. In this contribution thioamide based sulphide precursors were investigated for their decomposition and growth behaviour. A co-solvent approach in an ethanolic/aqueous ammonia medium was evaluated omitting the need for additional complexants. By replacing thiourea with the investigated thioamides, homogeneous dense layers of around 30 nm were grown with a greatly decreased deposition time of 8 min compared to 25 min for thiourea. Likewise, the concentration of the sulphide precursor was 40-fold reduced. The photovoltaic performance as characterized by external quantum efficiency and current–voltage measurements, showed conversion efficiencies of 15% comparable to the thiourea based process.

The problem of coordination in humanitarian emergencies arising from conflicts is not a new subject. So many seminars, round tables, declarations and publications have tackled it. Most came to the logical conclusion that stronger coordination between the humanitarian agencies was necessary, that the money had to be spent in the most effective way, and that new coordination bodies had to be created to make sure that there was no waste of operational energy.

Le problème de la coordination des opérations humanitaires lors des situations d'urgence provoquées par les conflits n'est pas nouveau. Le sujet a déjà été maintes fois abordé dans de nombreux séminaires, tables rondes, déclarations et publications. La plupart sont parvenus à la conclusion logique qu'une coordination plus étroite s'imposait entre les acteurs de l'humanitaire, que l'argent devait être dépensé le plus efficacement possible et que de nouveaux organes de coordination devaient être créés pour éviter toute perte d'énergie lors des opérations.

We report the fabrication of single mode quantum cascade lasers using a shallow-etched distributed Bragg reflector as frequency selective element. Quasi-continuous single mode tuning over 15 cm-1 at room temperature and 25 cm-1 via temperature tuning at Peltier temperatures is demonstrated. The behavior of both electro-optic and spectral characteristics under variation of the segment currents is analyzed, showing a maximum peak output power at room temperature of 600 mW. Thermal crosstalk between the laser segments is investigated. The spectral resolution of a gas absorption experiment is determined to be better than 0.0078 cm-1.

Clostridium thermocellum recombinant β-(1,3–1,4)-glucanase LicB was expressed inEscherichia coli K-12 and purified to electrophoretic homogeneity. Specific activity of the final preparation was 12,000 U/mg. Antibodies against recombinant LicB protein reacted with a specific band of purifiedC. thermocellum F7 cellulosomes, suggesting localization in the extracellular multienzyme complex. This indicates that the cellulosome takes part in the hydrolysis of other polysaccharides besides cellulose.
