Abstract

Resveratrol is a stilbene-type bioactive molecule with a broad spectrum of reported biological effects. In this sense, the current work provides a comprehensive literature analysis on resveratrol, representing a highly-researched commercially available dietary ingredient. Bibliometric data were identified by means of the search string TOPIC=("resveratrol*") and analyzed with the VOSviewer software, which yielded 17,561 publications extracted from the Web of Science Core Collection electronic database. 'I'he ratio of original articles to reviews was 9.5:1. More than half of the overall manuscripts have been published since 2013. Major contributing countries were USA, China, Italy, South Korea, and Spain. Most of the publications appeared in journals specialized in biochemistry and molecular biology, pharmacology and pharmacy, food science technology, cell biology, or oncology. The phytochemicals or phytochemical classes that were frequently mentioned in the keywords of analyzed publications included, in descending order: resveratrol, trans-resveratrol, polyphenols, flavonoids, quercetin, stilbenes, curcumin, piceatannol, cis-resveratrol, and anthocyanins.

6. Cytotoxicity assessment of aroylhydrazone derivatives

Abstract

Aroylhydrazones have been intensively investigated in recent years due to their versatile properties and applications. Many compounds of this type are biologically active as anticancer, antibacterial and antimicrobial agents. In addition, hydrazones as chelating agents were investigated as potential drugs for treatment of iron-overload associated diseases. In the present work, a group of 17 derivatives was studied for cytotoxic activity, as a part of our investigations on aroylhydrazones. The cytotoxic effect of the compounds was tested on non-tumor cells 3T3 and MCF10 and tumor cell lines HepG2, MCF7 and MDA-MB-231. Compound 11 exhibited the highest growth inhibitory effects on cell lines and significantly higher in tumor cell lines than in non-tumor cell lines. Compounds 5, C and E also show significantly higher cytotoxic activity in tumor cell lines than non-tumor cell lines in the experiment. The study makes it possible to assess the interaction between the structure and activity of the compounds and to understand the further development of hydrazones as more potent and selective antineoplastic agents.

Abstract

Aroyl hydrazones have been intensively investigated over the last decades due to their various biological properties such as anticancer, antibacterial and antimicrobial effects. Herein, we examine the anti-proliferative effect of two new aroyl hydrazones used in a combination with AVPI- and RGD -- peptides. The anti-proliferative activity is checked by MTT assay after 72h of treatment on MDA-MB-231 breast cancer cells. Though the aroyl hydrazone derivatives used as single agents shows a high activity, the combination is found not beneficial. [ABSTRACT FROM AUTHOR]

Abstract

AVPI-(Ala-Val-Pro-Ile) and RGD-(Arg-Gly-Asp) peptides have recently attracted a growing interest in the anticancer drug design and development. Following the concept of combining two fragments with different functionalities in order to achieve increased effects, we present herein the synthesis of two AVPI-RGD peptides. The impact of the performed chemical modifications on the anti-proliferative activity of these peptides was evaluated at MDA-MB-231 cancer cells. It was found that the conjugated peptide with modifications at proline and arginine residues showed a higher anti-proliferative effect than the parent AVPI-RGD molecule. Furthermore, it was observed that the effect of AVHypI-AgbGD vs. that of its co-administrated structural subunits was higher. The results obtained in this work suggested that the introduced substitutions and the conjugation of the two functional motifs benefit the anti-proliferative activity of the parent peptides. Abstract

Peptides have been developed during the last decade as an alternative for delivery and targeting of drugs, and for the visualization of tumors. In contrast to the antibodies, the peptides are smaller and can be delivered to the cytoplasm by various mechanisms. RGD peptides have been developed as potent integrin ligands and antagonists that inhibit the adhesion and formation of tumor metastases or directly induce apoptosis by activating caspases. A series of short RGD mimetics and their conjugates are synthesized and their cytotoxic activity against four cancer cell lines is examined. RGD peptides modified by shorter homologues of with arginine (α-amino-4-guanidinobutyric acid (Agb) and α-amino-3-guanidino-propionic acid (Agp)) show an increase of the cytotoxic effect compared to that of the parent RGD peptide. Abstract

The natural tridecapeptide neurotensin has been emerged as a promising therapeutic scaffold for the treatment of neurological diseases and cancer. In this work, we aimed to identify the top 100 most cited original research papers as well as recent key studies related to neurotensins. The Web of Science Core Collection database was searched and the retrieved research articles were analyzed by using the VOSviewer software. The most cited original articles were published between 1973 and 2013. The top-cited article was by Carraway and Leeman reporting the discovery of neurotensin in 1973. The highly cited terms were associated with hypotension and angiotensin-converting-enzyme.
