Abstract The black widow spider (BWS) is a venomous spider whose bite can cause various clinical conditions that range from local damage to serious systemic complications, including death. Cases of myocarditis following a BWS bite are rare but they can be fatal on occasion. However, the prognostic significance of the bite and presentation of myocarditis is unknown. Our case involved a 50-year-old man who presented with myocarditis after being bitten by a BWS and subsequently admitted to the intensive care unit for cardiac monitoring. During the hospital stay, he showed worsening signs on both the electrocardiographic and echocardiographic evaluations despite therapeutic success. Subsequent cardiac magnetic resonance and coronary angiography investigations showed no significant alterations; blood and instrumental test results slowly improved, and the patient was discharged home after 12 days of hospitalization without complications. This case illustrates that acute myocarditis, although an infrequent complication of BWS bite, has the potential to be lethal. The correct diagnosis, which is not always easy to formulate, is important to identify those patients who can benefit from careful monitoring and specific therapies aimed at reducing the risk of life.

Three files have been created in SEG-Y format after conversion from the raster images of selected parts of the ViDEPI open project: seismic reflection profiles D-445, D-451 and D-452, crossing the Mattinata–Gondola fault system. This dataset was obtained using the freely downloadable MATLAB© program IMAGE2SEGY. The program requires input TIFF and TXT files containing the image and specific parameters for the seismic reflection profiles to convert the raster images to the SEG-Y file format. The TIFF file of each profile was made using Adobe Photoshop®; the TXT format file was created with comma-separated number values and has a three-row and six-column structure, according to the program requirements. Once the raster images were converted to the SEG-Y file format, a light processing using DELPH Seismic® software improved the obtained seismic images. Creating this freely available dataset overcomes the problem of poor-quality representation of the original raster seismic reflection profiles. Hence, this dataset allows geologists to better interpret these profiles, especially by using dedicated software that can modify the colours of the reflectors as desired. Thus, these data are useful for researchers who want to investigate the deeper structures associated with the seismogenic Mattinata–Gondola fault system.

Abstract The knowledge of geomorphodynamic aspects is crucial for understanding marine and coastal processes/dynamics as well as for characterizing coastal environments heavily affected by anthropogenic activities. To provide a framework of analysis that can be applied in a consistent way for the geo-environmental characterization of highly contaminated coastal sites, in this paper a set of operational guidelines is proposed. Special attention is given to the role of geomorphological-based surveys and analyses in defining (i) the site-specific geological model of the investigated site, (ii) the anthropogenic impacts on marine and coastal sediments, (iii) the expected morphodynamic variations induced by climate change and anthropogenic interventions, (iv) tailored dissemination activities and community engagement plans. Then, an evaluation of the state of the art of activities already performed for the characterization of the coastal contaminated sites located in the Apulia region (southern Italy) is provided. The outcomes of this research are also provided in the form of infographics to favor their dissemination among communities and stakeholders.

Abstract Extreme atmospheric-marine events, known as medicanes (short for “Mediterranean hurricanes”), have affected the Mediterranean basin in recent years, resulting in extensive coastal flooding and storm surges, and have occasionally been responsible for several casualties. Considering that the development mechanism of these events is similar to tropical cyclones, it is plausible that these phenomena are strongly affected by sea surface temperatures (SSTs) during their development period (winter and autumn seasons). In this study, we compared satellite data and the numerical reanalysis of SSTs from 1969 to 2023 with in situ data from dataloggers installed at different depths off the coast of southeastern Sicily as well as from data available on Argo floats on the Mediterranean basin. A spectral analysis was performed using a continuous wavelet transform (CWT) for each SST time series to highlight the changes in SSTs prior to the occurrence of Mediterranean Hurricanes as well as the energy content of the various frequencies of the SST signal. The results revealed that decreases in SST occurred prior to the formation of each Mediterranean hurricane, and that this thermal drop phenomenon was not observed in intense extra-tropical systems. The spectral analyses revealed that high CWT coefficients representing high SST energy contents were observed before the occurrence of a Mediterranean hurricane. This information may provide a useful fingerprint for distinguishing Mediterranean hurricanes from common seasonal storms at the onset of these events.

Abstract Background Antimicrobial resistance has been identified as a major threat to global health. The pig food chain is considered an important source of antimicrobial resistance genes (ARGs). However, there is still a lack of knowledge on the dispersion of ARGs in pig production system, including the external environment. Results In the present study, we longitudinally followed one swine farm located in Italy from the weaning phase to the slaughterhouse to comprehensively assess the diversity of ARGs, their diffusion, and the bacteria associated with them. We obtained shotgun metagenomic sequences from 294 samples, including pig feces, farm environment, soil around the farm, wastewater, and slaughterhouse environment. We identified a total of 530 species-level genome bins (SGBs), which allowed us to assess the dispersion of microorganisms and their associated ARGs in the farm system. We identified 309 SGBs being shared between the animals gut microbiome, the internal and external farm environments. Specifically, these SGBs were characterized by a diverse and complex resistome, with ARGs active against 18 different classes of antibiotic compounds, well matching antibiotic use in the pig food chain in Europe. Conclusions Collectively, our results highlight the urgency to implement more effective countermeasures to limit the dispersion of ARGs in the pig food systems and the relevance of metagenomics-based approaches to monitor the spread of ARGs for the safety of the farm working environment and the surrounding ecosystems.

Summary: Aiming to shed light on the biology of wild ruminants, we investigated the gut microbiome seasonal dynamics of the Alpine ibex (Capra ibex) from the Central Italian Alps. Feces were collected in spring, summer, and autumn during non-invasive sampling campaigns. Samples were analyzed by 16S rRNA amplicon sequencing, shotgun metagenomics, as well as targeted and untargeted metabolomics. Our findings revealed season-specific compositional and functional profiles of the ibex gut microbiome that may allow the host to adapt to seasonal changes in available forage, by fine-tuning the holobiont catabolic layout to fully exploit the available food. Besides confirming the importance of the host-associated microbiome in providing the phenotypic plasticity needed to buffer dietary changes, we obtained species-level genome bins and identified minimal gut microbiome community modules of 11–14 interacting strains as a possible microbiome-based solution for the bioconversion of lignocellulose to high-value compounds, such as volatile fatty acids.

In this work, we study a Mediterranean cyclone, Helios, which took place during 9–11 February 2023 in the southeastern part of Sicily and Malta, by a multiparametric approach combining microseism results with sea state and meteorological data provided by wavemeter buoy, HF radar, hindcast maps and satellite SEVIRI images. The sub-tropical system Helios caused heavy rainfall, strong wind gusts and violent storm surges with significant wave heights greater than 5 m. We deal with the relationships between such a system and the features of microseism (the most continuous and ubiquitous seismic signal on Earth) in terms of spectral content, space–time variation of the amplitude and source locations tracked by means of two methods (amplitude-based grid search and array techniques). By comparing the location of the microseism sources and the area affected by significant storm surges derived from sea state data, we note that the microseism location results are in agreement with the real position of the storm surges. In addition, we are able to obtain the seismic signature of Helios using a method that exploits the coherence of continuous seismic noise. Hence, we show how an innovative monitoring system of the Mediterranean cyclones can be designed by integrating microseism information with other techniques routinely used to study meteorological phenomena.

Abstract Background The clam Chamelea gallina is an ecologically and economically important marine species in the Northwestern Adriatic Sea, which currently suffers from occasional, and still unexplained, widespread mortality events. In order to provide some glimpses in this direction, this study explores the connections between microbiome variations at the clam-sediment interface and the nutritional status of clams collected at four Italian production sites along the Emilia Romagna coast, with different mortality incidence, higher in the Northern sites and lower in the Southern sites. Results According to our findings, each production site showed a peculiar microbiome arrangement at the clam-sediment interface, with features that clearly differentiate the Northern and Southern sites, with the latter also being associated with a better nutritional status of the animal. Interestingly, the C. gallina digestive gland microbiome from the Southern sites was enriched in some health-promoting microbiome components, capable of supplying the host with essential nutrients and defensive molecules. Furthermore, in experiments conducted under controlled conditions in aquaria, we provided preliminary evidence of the prebiotic action of sediments from the Southern sites, allowing to boost the acquisition of previously identified health-promoting components of the digestive gland microbiome by clams from the Northern sites. Conclusions Taken together, our findings may help define innovative microbiome-based management strategies for the preservation of the productivity of C. gallina clams in the Adriatic Sea, through the identification and maintenance of a probiotic niche at the animal-sediment interface.

Abstract To understand the microbiome composition and interplay among bacterial communities in different compartments of a coupled freshwater aquaponics system growing flathead grey mullet (Mugil cephalus) and lettuces (Lactuca sativa), 16S rRNA gene amplicon sequencing of the V3–V4 region was analysed from each compartment (fish intestine, water from the sedimentation tank, bioballs from the biological filter, water and biofilm from the hydroponic unit, and lettuce roots). The bacterial communities of each sample group showed a stable diversity during all the trial, except for the fish gut microbiota, which displayed lower alpha diversity values. Regarding beta diversity, the structure of bacterial communities belonging to the biofilm adhering to the hydroponic tank walls, bioballs, and lettuce roots resembled each other (weighted and unweighted UniFrac distances), while bacteria from water samples also clustered together. However, both of the above-mentioned bacterial communities did not resemble those of fish gut. We found a low or almost null number of shared Amplicon Sequence Variants (ASVs) among sampled groups which indicated that each compartment worked as an independent microbiome. Regarding fish health and food safety, the microbiome profile did not reveal neither fish pathogens nor bacterial species potentially pathogenic for food health, highlighting the safety of this sustainable food production system.

Summary: The human gut microbiome is losing biodiversity, due to the “microbiome modernization process” that occurs with urbanization. To keep track of it, here we applied shotgun metagenomics to the gut microbiome of the Baka, a group of forager-horticulturalists from Cameroon, who combine hunting and gathering with growing a few crops and working for neighboring Bantu-speaking farmers. We analyzed the gut microbiome of individuals with different access to and use of wild plant and processed foods, to explore the variation of their gut microbiome along the cline from hunter-gatherer to agricultural subsistence patterns. We found that 26 species-level genome bins from our cohort were pivotal for the degradation of the wild plant food substrates. These microbes include Old Friend species and are encoded for genes that are no longer present in industrialized gut microbiome. Our results highlight the potential relevance of these genes to human biology and health, in relation to lifestyle.

IntroductionIn our paper, we explored the impact of different anthropogenic stressors, namely, mussel farming, methane extraction platforms, and summer mass tourism, on the epipelagic microbiomes of the North-Western Adriatic Sea.MethodsBy 16S rRNA gene sequencing, we provided the epipelagic microbiome layout from selected sites corresponding to each of the considered impacts. As an unimpacted reference, we used already published 16S rRNA sequencing data.ResultsAccording to our findings, each one of the anthropogenic stressors resulted in a peculiar increase of specific epipelagic microbial components, including copiotrophic R-strategists and host-restricted bacteria, as well as some pathobiome components, the latter being detected exclusively in impacted sites. Particularly, potentially harmful pathogenic species such as Legionella impletisoli and Staphylococcus epidermidis have been detected in proximity to the mussel farms, and Escherichia coli and Campylobacter ureolyticus were present close to the methane extraction platform and at the summer mass tourism site, respectively. Particularly, C. ureolyticus is an emerging human gastrointestinal pathogen, capable of destroying intestinal microvilli.DiscussionIn addition to providing evidence supporting the existence of recognizable and impact-driven fingerprints on the epipelagic marine microbiome peculiar to the different anthropogenic stressors, our findings also raise concern about the ecological relevance of the observed changes, in terms of possible loss of ecosystem services and also for the potential release of pathogenic microorganisms in the environment.

Mediterranean hurricanes, also known as medicanes, can cause significant damage to coastal communities. Consequently, they have been the subject of intense study in recent decades. Geographic Information Systems (GISs) and Web-GIS applications are essential for processing and analyzing geographic data from various sources, particularly in the field of spatial planning. Due to their ability to integrate heterogeneous information, these tools can provide experts with a comprehensive overview of relevant geospatial data. Here, we present ARCHIMEDE, a relational geodatabase connected to an open-source Web-GIS platform focused on Mediterranean hurricanes that contains data from recent research on these extreme weather phenomena. These datasets comprise climatic and oceanographic data obtained from remote sensing techniques as well as seismic and geomorphological data obtained from field observations. Consequently, this Web-GIS platform can enhance our understanding of Mediterranean cyclones by relating the physical properties of these natural phenomena with their impacts on coastal regions. ARCHIMEDE is an innovative tool for the dissemination of geographic information to stakeholders, researchers, and decision-makers, offering valuable support for the development of robust coastal management strategies aimed at mitigating the challenges posed by Mediterranean hurricanes.

Lipid disorders represent one of the most worrisome cardiovascular risk factors. The focus on the impact of lipids on cardiac and vascular health usually concerns low-density lipoprotein cholesterol, while the role of triglycerides (TGs) is given poor attention. The literature provides data on the impact of higher plasma concentrations in TGs on the cardiovascular system and, therefore, on the outcomes and comorbidities of patients. The risk for coronary heart diseases varies from 57 to 76% in patients with hypertriglyceridemia. Specifically, the higher the plasma concentrations in TGs, the higher the incidence and prevalence of death, myocardial infarction, and stroke. Nevertheless, the metabolism of TGs and the exact physiopathologic mechanisms which try to explain the relationship between TGs and cardiovascular outcomes are not completely understood. The aims of this narrative review were as follows: to provide a comprehensive evaluation of the metabolism of triglycerides and a possible suggestion for understanding the targets for counteracting hypertriglyceridemia; to describe the inner physiopathological background for the relationship between vascular and cardiac damages derived from higher plasma concentrations in TGs; and to outline the need for promoting further insights in therapies for reducing TGs plasma levels.

Skeletal muscle regeneration entails a multifaceted process marked by distinct phases, encompassing inflammation, regeneration, and remodeling. The coordination of these phases hinges upon precise intercellular communication orchestrated by diverse cell types and signaling molecules. Recent focus has turned towards extracellular vesicles (EVs), particularly small EVs, as pivotal mediators facilitating intercellular communication throughout muscle regeneration. Notably, injured muscle provokes the release of EVs originating from myofibers and various cell types, including mesenchymal stem cells, satellite cells, and immune cells such as M2 macrophages, which exhibit anti-inflammatory and promyogenic properties. EVs harbor a specific cargo comprising functional proteins, lipids, and nucleic acids, including microRNAs (miRNAs), which intricately regulate gene expression in target cells and activate downstream pathways crucial for skeletal muscle homeostasis and repair. Furthermore, EVs foster angiogenesis, muscle reinnervation, and extracellular matrix remodeling, thereby modulating the tissue microenvironment and promoting effective tissue regeneration. This review consolidates the current understanding on EVs released by cells and damaged tissues throughout various phases of muscle regeneration with a focus on EV cargo, providing new insights on potential therapeutic interventions to mitigate muscle-related pathologies.

The latest progress in deep learning approaches has garnered significant attention across a variety of research fields. These techniques have revolutionized the way marine parameters are measured, enabling automated and remote data collection. This work centers on employing a deep learning model for the automated evaluation of tide and surge, aiming to deliver accurate results through the analysis of surveillance camera images. A mode of deep learning based on the Inception v3 structure was applied to predict tide and storm surges from surveillance cameras located in two different coastal areas of Italy. This approach is particularly advantageous in situations where traditional tide sensors are inaccessible or distant from the measurement point, especially during extreme events that require accurate surge measurements. The conducted experiments illustrate that the algorithm efficiently measures tide and surge remotely, achieving an accuracy surpassing 90% and maintaining a loss value below 1, evaluated through Categorical Cross-Entropy Loss functions. The findings highlight its potential to bridge the gap in data collection in challenging coastal environments, providing valuable insights for coastal management and hazard assessments. This research contributes to the emerging field of remote sensing and machine learning applications in environmental monitoring, paving the way for enhanced understanding and decision-making in coastal regions.

ABSTRACTFabry disease (FD) is an X-linked metabolic disease caused by a deficiency in α-galactosidase A (α-Gal A) activity. This causes accumulation of glycosphingolipids, especially globotriaosylceramide (Gb3), in different cells and organs. Neuropathic pain and gastrointestinal (GI) symptoms, such as abdominal pain, nausea, diarrhea, constipation, and early satiety, are the most frequent symptoms reported by FD patients and severely affect their quality of life. It is generally accepted that Gb3 and lyso-Gb3 are involved in the symptoms; nevertheless, the origin of these symptoms is complex and multifactorial, and the exact mechanisms of pathogenesis are still poorly understood. Here, we used a murine model of FD, the male α-Gal A (-/0) mouse, to characterize functionality, behavior, and microbiota in an attempt to elucidate the microbiota-gut-brain axis at three different ages. We provided evidence of a diarrhea-like phenotype and visceral hypersensitivity in our FD model together with reduced locomotor activity and anxiety-like behavior. We also showed for the first time that symptomology was associated with early compositional and functional dysbiosis of the gut microbiota, paralleled by alterations in fecal short-chain fatty acid levels, which partly persisted with advancing age. Interestingly, most of the dysbiotic features suggested a disruption of gut homeostasis, possibly contributing to accelerated intestinal transit, visceral hypersensitivity, and impaired communication along the gut-brain axis.

Introduction: Detachment from the extracellular matrix (ECM) is the first step of the metastatic cascade. It is a regulated process involving interaction between tumor cells and tumor microenvironment (TME). Iron is a key micronutrient within the TME. Here, we explored the role of iron in the ability of ovarian cancer cells to successfully detach from the ECM.Methods: HEY and PEO1 ovarian cancer cells were grown in 3D conditions. To mimic an iron rich TME, culture media were supplemented with 100 μM Fe3+. Cell mortality was evaluated by cytofluorimetric assay. The invasive potential of tumor spheroids was performed in Matrigel and documented with images and time-lapses. Iron metabolism was assessed by analyzing the expression of CD71 and FtH1, and by quantifying the intracellular labile iron pool (LIP) through Calcein-AM cytofluorimetric assay. Ferroptosis was assessed by quantifying mitochondrial reactive oxygen species (ROS) and lipid peroxidation through MitoSOX and BODIPY-C11 cytofluorimetric assays, respectively. Ferroptosis markers GPX4 and VDAC2 were measured by Western blot. FtH1 knockdown was performed by using siRNA.Results: To generate spheroids, HEY and PEO1 cells prevent LIP accumulation by upregulating FtH1. 3D HEY moderately increases FtH1, and LIP is only slightly reduced. 3D PEO1upregulate FtH1 and LIP results significantly diminished. HEY tumor spheroids prevent iron import downregulating CD71, while PEO1 cells strongly enhance it. Intracellular ROS drop down during the 2D to 3D transition in both cell lines, but more significantly in PEO1 cells. Upon iron supplementation, PEO1 cells continue to enhance CD71 and FtH1 without accumulating the LIP and ROS and do not undergo ferroptosis. HEY, instead, accumulate LIP, undergo ferroptosis and attenuate their sphere-forming ability and invasiveness. FtH1 knockdown significantly reduces the generation of PEO1 tumor spheroids, although without sensitizing them to ferroptosis.Discussion: Iron metabolism reprogramming is a key event in the tumor spheroid generation of ovarian cancer cells. An iron-rich environment impairs the sphere-forming ability and causes cell death only in ferroptosis sensitive cells. A better understanding of ferroptosis sensitivity could be useful to develop effective treatments to kill ECM-detached ovarian cancer cells.

Abstract Acid sphingomyelinase deficiency (ASMD)—also known as Niemann–Pick (NP) disease—is a rare, autosomal recessive disorder which is characterized by deficiency of the lysosomal enzyme acid sphingomyelinase (ASM), resulting in excessive storage of lipids in organs (i.e., spleen, liver, lung, bone marrow, lymph nodes, and vascular system). Only a few cases of moderate-to-severe valvular heart disease due to ASMD are described in the literature, mostly in adulthood. We report here the case of a patient with NP disease subtype B that was diagnosed during adulthood. NP disease in this patient was found to be associated with situs inversus. Specifically, a severe, symptomatic aortic stenosis was identified, and the need for surgical or percutaneous intervention was discussed. The heart team chose transcatheter aortic valvular implantation (TAVI), which was successfully performed with no complications on follow-up.

In response to the accidental exhumation of three ancient trees by farmers, we conducted a multidisciplinary study based on the stratigraphic analysis of boreholes, carbon-14 dating, aerial photo interpretation, and analysis of palaeobotanical and archaeological evidences. We reconstructed the formation and evolution during Roman times of a first “continuous” and then “discontinuous” deltaic strandplain at the mouth of the Carapelle Stream in the Tavoliere di Puglia Plain—the second-largest plain in Italy. Ttwo main phases can be recognised in the evolution of the Carapelle deltaic strandplain: 1) a first phase, lasted until ca. the birth of Christ, was characterised by a regular and continuous construction of sand ridges one leaning against the other; 2) a second phase, lasted more or less from the birth of Christ to the termination of the construction of the deltaic strandplain, was characterised by the discontinuous construction of sand ridges/coastal barriers with the isolation of lagoons/ponds, and by evidences of alluvial events. The most probable climatic–environmental scenario to have formed the Roman-period deltaic strandplain implied that: 1) the first phase was triggered by a higher total amount of precipitations, but with less extreme alluvial events; 2) the second phase was triggered by a total amount of precipitation lower than the previous period, but with higher occurrence of extreme alluvial events and/or by extreme alluvial events separated by longer period of low precipitations. This second phase was enhanced by the opening of vegetation. The passage between the first and second phase of the Carapelle deltaic strandplain coincides with the passage from overall negative NAO index to an overall positive NAO index.

Abstract Because of their recognized global importance, there is now the urgent need to map diversity and distribution patterns of marine microbial communities. Even if available studies provided some advances in the understanding the biogeographical patterns of marine microbiomes at the global scale, their degree of plasticity at the local scale it is still underexplored, and functional implications still need to be dissected. In this scenario here we provide a synoptical study on the microbiomes of the water column and surface sediments from 19 sites in a 130 km2 area located 13.5 km afar from the coast in the North-Western Adriatic Sea (Italy), providing the finest-scale mapping of marine microbiomes in the Mediterranean Sea. Pelagic and benthic microbiomes in the study area showed sector specific-patterns and distinct assemblage structures, corresponding to specific variations in the microbiome network structure. While maintaining a balanced structure in terms of potential ecosystem services (e.g., hydrocarbon degradation and nutrient cycling), sector-specific patterns of over-abundant modules—and taxa—were defined, with the South sector (the closest to the coast) characterized by microbial groups of terrestrial origins, both in the pelagic and the benthic realms. By the granular assessment of the marine microbiome changes at the local scale, we have been able to describe, to our knowledge at the first time, the integration of terrestrial microorganisms in the marine microbiome networks, as a possible natural process characterizing eutrophic coastal area. This raises the question about the biological threshold for terrestrial microorganisms to be admitted in the marine microbiome networks, without altering the ecological balance.

Body lesions in pigs are a common welfare concern, particularly during the weaning period. These lesions can lead to pain, infection, and impaired mobility, resulting in reduced growth performance and increased mortality. Moreover, weaning stress can affect gut microbiota, immune response and increase the oxidative stress of piglets during this transition period. It has been hypothesised that social stress and body lesions could contribute to affect the gut microbiota, physiological and immune response of piglets. The study aims to evaluate the impact of the body lesions due to social stress on microbial profile, immune response, and oxidative status of weaned piglets. Lesion score (LS) on skin, tail, ear, neck, middle trunk, and hind quarters was measured 1 week (28 days of age, T1) and 7 weeks postweaning (T2) on 45 tail-docked pigs according to the method suggested from the Walfer Quality® (2009) on a scale from 0 to 2. Based on the LS, at T1, piglets were classified as High LS (n = 16), when LS was >1 in at least two of the areas considered, or Low LS (n = 29). Abstract The disastrous earthquake of 1693 AD caused over 60,000 causalities and the total destruction of several villages and towns in south-eastern Sicily. Immediately after the earthquake, a tsunami struck the Ionian coasts of Sicily and the Messina Strait and was probably recorded even in the Aeolian Islands and Malta. Over the last few decades, the event has been much debated regarding the location of the seismogenic source and the possible cause of the associated tsunami. The marine event has been related to both a submarine landslide and a coseismic displacement at the seafloor. To better define the most reliable sources and dynamics of the tsunami, we couple high-resolution marine seismic survey data with hydrodynamic modelling to simulate various scenarios of tsunami generation and propagation. Results from the simulations are compared with geomorphological evidence of past tsunami impacts, described in previous work along the coast of south-eastern Sicily, and within historical chronicles and reports. The most reliable scenario considers the 1693 event composed by two different tsunami waves: a first wave generated by the coseismic fault displacement at the seafloor and a second wave generated by a submarine landslide, triggered by the earthquake shaking. Tsunami modelling shows that a simultaneous movement between fault displacement and submarine mass movement could determine a destructive interference on the tsunami waves, resulting in a reduction in wave height. For this reason, the second tsunami wave probably occurred with a maximum delay of few minutes after the one generated by the earthquake and induced a greater flooding. The double-source model could explain the observation because in the course of other destructive earthquakes in south-eastern Sicily, such as that of 1169 AD, the associated tsunami caused less damages. This implies the need to better map, define and assess the hazard responsible for this type of tsunami events.

The assessment of past sea-level positions requires a multidisciplinary approach that involves both scientific and historical humanistic fields. The use of a multidisciplinary approach allows us to obtain reliable information on the relative sea-level position, the determination of which requires the evaluation of the eustatic and steric components as well as an assessment of the vertical ground displacements, such as the isostatic adjustments and tectonic movements. In this context, coastal geoarchaeological markers play a fundamental role since their architectural height (generally defined as functional height) was relative to the sea level at the time of their construction. Thus, a comparison between the current elevation of geoarchaeological structures (or depth in the case they are currently submerged) with their estimated functional height allows us to obtain the relative sea-level variation. In this study, we applied a methodological procedure for the evaluation of the functional height of architectural elements using modern technologies (Terrestrial Laser Scanner and GPS-Real Time Kinematic) and detailed sea-level analysis. The proposed methodology was applied to coastal quarries located along the coast of Bari (Apulia region, southern Italy). The results allowed us to confirm the functional height of the detachment surface reported in the literature and to assess the sea-level position in the fifth and fourth centuries before Christ.

IntroductionWines produced from the same grape cultivars but in different locations possess distinctive qualities leading to different consumer’s appreciation, preferences, and thus purchase choices. Here, we explore the possible importance of microbiomes at the soil–plant interface as a determinant of the terroir properties in grapevine production, which confer specific growth performances and wine chemo-sensory properties at the local scale.MethodsIn particular, we investigated the variation in microbial communities associated with the roots of Vitis vinifera cultivar Lambrusco, as well as with surrounding bulk soils, in different vineyards across the “Consorzio Tutela Lambrusco DOC” protected designation of origin area (PDO, Emilia Romagna, Italy), considering viticultural sites located both inside and outside the consortium in two different seasons (June and November 2021).ResultsAccording to our findings, rhizospheric and soil microbiomes show significant structural differences in relation to the sampling site, regardless of seasonality, while endophytic microbiomes seem to be completely unaffected by such variables. Furthermore, a deeper insight into the microbial terroir of PDO areas highlighted the presence of some rhizospheric microorganisms enriched inside the consortium and characterizing the PDO regardless of both sampling season and farming strategy. These include Bacillus, Paenibacillus, and Azospirillum, which are all well-known plant growth-promoting bacteria.DiscussionTaken together, our results suggest a connection between soil and root microbiomes of V. vinifera cultivar Lambrusco and the local designation of origin, emphasizing the potential role of PDO-enriched plant growth-promoting bacteria in vine growing and final quality of the Lambrusco DOC wine.

Cancer-associated thrombosis (CAT) is a devastating complication of cancer that can significantly impact a patient’s health and life. The incidence of CAT is approximately 20%, and 1 in 5 cancer patients will develop CAT annually. Indeed, CAT can promote pulmonary embolism and deep vein thrombosis, leading to increased morbidity and mortality that dramatically impact survival. CAT can also provoke delay or discontinuation of anticancer treatment, which may result in a lack of treatment efficacy and high costs for patients, institutions, and society. Current guidelines advocate direct oral anticoagulants (DOACs) as the first-line anticoagulant option in CAT. Compared to low-molecular-weight-heparins (LMWHs), DOACs are advantageous in that they typically have an oral route of administration, do not require laboratory monitoring, and have a more predictable anticoagulant effect. However, in patients with thrombocytopenia, renal failure, or those receiving anticancer regimens with potential for drug-drug interactions, LMWH is still the mainstay of care. The main limitation of current anticoagulant agents is related to bleeding risk (BR), both for DOACs and LMWHs. Specifically, DOACs have been associated with high BR in gastrointestinal and genitourinary cancers. In this challenging scenario, abelacimab, an anti-factor XI agent, could represent a viable option in the management of CAT due to its “hemostasis sparing” effect. The safe profile of abelacimab could be useful in patients with active malignancy and CAT, as long-term anticoagulant therapy is often required. Two ongoing international phase III trials (Aster and Magnolia) compare abelacimab with the standard of care (i.e., apixaban in patients with CAT and dalteparin in those with CAT and high BR, respectively). Abelacimab is a new and attractive anticoagulant for the management of CAT, especially in the insidious and critical scenario of active cancer patients with venous thromboembolism and high BR. The aim of this narrative review is to discuss the updated evidence on the performance of DOACs and LMWHs in the treatment of CAT and to focus on the potential role of abelacimab in CAT and its promising associated clinical trials.

Contaminated marine and coastal sediments represent the main source of secondary pollution for the aquatic environment and marine fauna, affecting, directly and indirectly, ecosystems and human health. The assessment of the distribution of chemical pollutants in marine sediments can therefore be considered a preliminary step for understanding the possible circulation of pollutants in the marine environment and planning any targeted and efficient reclamation activity. This study provides new insights on the environmental status of Bay I of Mar Piccolo basin (Southern Italy) by proposing an integrated investigation approach to define the distribution of trace metals and evaluate the thickness of the sediments potentially affected by pollution. To this aim, the concentrations of As, Cd, Cr, Cu, Hg, Ni, Pb, Sn, and Zn are estimated for sediment samples collected from 19 cores, and specific environmental indices are calculated. Due to its remarkable environmental and economic relevance, the area of Taranto has been selected as a case study to evaluate the effectiveness of the proposed method in supporting the identification of hotspot areas for which priority remediation activities are needed.

The human zinc finger protein 521 (ZNF521) is a co-transcriptional factor with multiple recognized regulatory functions in a range of normal, cancer and stem cell compartments. ZNF521 regulates proliferation, progression and CSC (cancer stem cell) compartments in human ovarian cancer (hOC), which is a very aggressive and late-diagnosed female tumor. Two other important regulators of hOC are the NRF2 and NOTCH signaling pathways. In the present paper, the mRNA and protein levels of ZNF521 were correlated with those of the NRF2-NOTCH signaling components in two different hOC cell lines and in a public dataset of 381 hOC patients. The data show that high levels of ZNF521 significantly increase NRF2-NOTCH signaling expression; conversely, the silencing of ZNF521 impairs NRF2-NOTCH signaling. This experimental work shows that, in hOC, different levels of ZNF521 modulate the NRF2-NOTCH signaling pathway and also influences hOC CSC properties.

Coastal erosion occurs due to different processes involving physical and ecological systems. One of these factors is the degree of water stress experienced by dune vegetation. While healthy dune vegetation can help to stabilize the dune systems, water-stressed vegetation can instead enhance dune erosion. In this study, remote sensing techniques were used to monitor the water stress affecting the dune vegetation in dune systems along the alluvial plain of the Chiatona coast (Apulia, Southern Italy) located on the Ionian Arc. Multispectral satellite data from Landsat 8/9 and Sentinel-2 were used to assess the water stress at different spatial scales over a 4-year monitoring period from 2019 to 2023. The normalized difference vegetation index (NDVI) and the normalized difference moisture index (NDMI) were used to identify dune surfaces that were experiencing water stress. Furthermore, a terrestrial laser scanner and LiDAR data were acquired at different temporal ranges in areas affected by water stress to highlight coastal changes in areas associated with unhealthy dune vegetation. A large drop in NDVI values was observed in May 2020 due to the occurrence of coastal fires in some parts of the Chiatona coast. Geoelectrical surveys were conducted to investigate if coastal fires were capable of saline groundwater contamination, potentially enhancing dune erosion in these areas. The joint analysis of remote sensing, topographical, and geoelectric data showed that water stress reduced the amount of healthy dune vegetation, triggering dune deflation processes that resulted in increased coastal erosion rates, while also leading to the saline contamination of groundwater.

BackgroundEven in the early stages, global crises such as the COVID-19 pandemic lead to serious dislocations of social life, secondary adjustment reactions to external restrictions and individual concerns. Coping mechanisms may also include dysfunctional strategies like an increase of drug use. Considering the wide-spread use of cannabis, the aim of this study was to elucidate the interplay of social restrictions, psychopathology, concerns related to the pandemic in addition to the users’ experiences, motivations and consumption quantities during the early COVID-19 pandemic. It was presumed that cannabis intake would increase during the early phase of the crisis and that consumption quantities would be related to corona-related restrictions, concerns as well as subjective substance effects and psychopathology.Materials and methodsAs part of an international, cross-sectional, internet-based survey (N = 5,049) available in five languages, consumption quantities and patterns of cannabis use in the early phase of the pandemic from April to August 2020 were examined. Participants retrospectively rated restrictions and concerns related to the pandemic, motives of cannabis use prior to and during 1 month the pandemic, and subjective consumption effects.ResultsCannabis use behavior showed no significant differences when consumption quantities prior and during 1 month after the COVID-19 outbreak were compared. Higher quantities of cannabis intake prior and during 1 month of the pandemic as well as more corona-related concern were associated with an increased perception of positive effects of cannabis during the pandemic. Predictors of its use during 1 month of pandemic were higher pre-pandemic consumption quantity, older age, quarantinization, a lesser degree of being affected by negative effects of the pandemic and a stronger subjective experience of corona-related positive effects of cannabis. Comparisons of the motives for cannabis intake in the pre-pandemic versus the pandemic period showed that all rationales for consumption were reported less frequently, except boredom.ConclusionFrequencies of cannabis intake remained relatively stable in the early pandemic phase. Risk factors for increased use seem related to habitual consumption patterns that become more prominent under quarantinization. The use of cannabis as a dysfunctional coping strategy might not be amenable via self-report and should therefore receive special attention in clinical contexts.

Background: Uterine fibroids are the most common benign gynaecological tumours in women. When symptomatic, heavy menstrual bleeding is the principal manifestation of uterine fibroids. Nowadays, several conservative procedures have been utilized to discourage hysterectomy or myomectomy and great interest has been focused on laser energy technology associated with mini-invasive approaches. Diode lasers demonstrated many advantages in laparoscopic and hysteroscopic surgery. Methods: To evaluate the Dual Wavelength Diode Laser System (DWLS) ablation techniques for treating fibroids, we devised an experimental system for characterizing the myolysis fibre ablation performance with a particular fibre designed to create a thermal ablation in the centre of the myoma. All the myomas were used for the experiment after a total hysterectomy and their excision from the fresh uteri. The experiment setup was composed of two beaker containers filled with isotonic saline water, temperature monitoring myoma surface and water bath, a diode laser fibre and a diode laser energy deposition system. Results: Good macroscopic results have been found for fibroids of about 60 cm3 ablated with a setting of 10 W of 980 nm + 10 W of 1470 nm and for samples of about 15 cm3 ablated with a setting of 5 W of 980 nm + 5 W of 1470 nm, where the volume of the ablated fibroids were 46.6% and 41.6%, respectively. Conclusion: This is the first ex vivo study with a new DWLS, which made it possible to understand which one of its settings could be recommended for the safest and best macroscopic results in myolysis.

c-MET/hepatocyte growth factor (HGF) system deregulation is a well-known feature of malignancy in several solid tumors, and for this reason this system and its pathway have been considered as potential targets for therapeutic purposes. In previous manuscripts we reported c-MET/HGF expression and the role in testicular germ cell tumors (TGCTs) derived cell lines. We demonstrated the key role of c-Src and phosphatidylinositol 3-kinase (PI3K)/AKT adaptors in the HGF-dependent malignant behavior of the embryonal carcinoma cell line NT2D1, finding that the inhibition of these onco-adaptor proteins abrogates HGF triggered responses such as proliferation, migration, and invasion. Expanding on these previous studies, herein we investigated the role of mitogen-activated protein kinase (MAPK)/extracellular signal regulated kinase (ERK) pathways in the HGF-dependent and HGF-independent NT2D1 cells biological responses. To inhibit MAPK/ERK pathways we chose a pharmacological approach, by using U0126 inhibitor, and we analyzed cell proliferation, collective migration, and chemotaxis. The administration of U0126 together with HGF reverts the HGF-dependent activation of cell proliferation but, surprisingly, does not exert the same effect on NT2D1 cell migration. In addition, we found that the use of U0126 alone significantly promotes the acquisition of NT2D1 «migrating phenotype», while collective migration of NT2D1 cells was stimulated. Notably, the inhibition of ERK activation in the absence of HGF stimulation resulted in the activation of the AKT-mediated pathway, and this let us speculate that the paradoxical effects obtained by using U0126, which are the increase of collective migration and the acquisition of partial epithelium–mesenchyme transition (pEMT), are the result of compensatory pathways activation. These data highlight how the specific response to pathway inhibitors, should be investigated in depth before setting up therapy.

Beach rocks are located along many coasts of the Mediterranean basin. The early diagenesis environment and the mean sea level along the shoreline make these landforms useful in the reconstruction of relative sea-level changes and, in particular, as SLIPs (sea-level index points). The beach rocks surveyed along the Ionian coast of Apulia were found to be well preserved at three specific depth ranges: 6–9 m, 3–4 m, and from the foreshore to about 1.20 m. Morpho-bathymetric and dive surveys were performed to assess both the geometries and the extension of the submerged beach rocks. Samples were collected at these different depths in the localities of Lido Torretta, Campomarino di Maruggio, San Pietro in Bevagna, and Porto Cesareo. Bivalve shells were identified and isolated from the beach rock samples collected at a depth of 7 m; AMS dating provided a calibrated age of about 7.8 ka BP. Their morphology and petrological features, along with the time constraints, enabled us to (i) reconstruct the local sea-level curve during the Holocene, (ii) corroborate acquired knowledge of the relative sea-level history, and (iii) identify possible local vertical land movement (VLM).

Space Archaeology (SA), also known as Satellite Archaeology, Satellite Remote Sensing for Archaeology, or Archaeology from Space, is part of the wider interdisciplinary field of Remote Sensing for Archaeology. The application of satellite data in archaeological investigations has proven useful for landscape observation and analysis, the detection of archaeological traces, the reconstruction and monitoring of natural and anthropic processes, and the management and promotion of archaeological heritage. During the last few decades, the increasing number of SA studies has demonstrated innovative growth in archaeological disciplines due to the significant enhancement of spatial technologies, the advancement of visual inspection and image processing techniques, the development of data fusion methodologies, and the improvement of multi-temporal analysis methods. Therefore, a broad overview of the current situation in the concepts and approaches of SA is necessary to gain greater awareness of the current potentialities and limitations of this science to better address future studies. The present work provides a review of the scientific literature by exploring the different aspects of SA and the goals achieved to date in three main application fields: Detection, monitoring, and promotion of archaeological heritage. The contributions reviewed were divided within each of these three categories by analyzing the type of SA data and methods applied. The results indicate that (1) most studies aim to detect archaeological sites compared to monitoring and promotion; (2) optical images are used more than SAR data; and (3) techniques of image enhancement and visual interpretation are applied more than other data processing methods. This brief overview allows us to consider SA as an evolving discipline, an engine for cross-skills training, as well as a promising future science that can play a key role in the frontier of sustainable development and the new space economy.

Reliable deep-time environmental and climate reconstructions are needed to understand the drivers of Earthâs system evolution over geological time. Palaeozoic temperature estimates, including reconstructions of the climate change through the Ordovician, are based mainly on oxygen isotope (18O/16O; δ18OVSMOW) thermometry derived from carbonate rocks with fossils such as calcitic brachiopods and phosphatic conodonts that are often the best preserved repositories of environmental conditions. Palaeoenvironmental reconstructions are reliable only if the geochemical data is obtained using well-calibrated analytical tools. Most previous research devoted to oxygen isotope composition of conodonts has been conducted using the bulk method (gas source isotope ratio mass spectrometry (GS-IRMS)) that typically requires pooling several dozens of conodont elements for a single isotope ratio measurement. As such, studies of conodont-poor intervals and assessments of taxon-specific δ18O variability require extensive sample preparation and are challenging using the bulk method. Such challenges can be addressed by in situ secondary ion mass spectrometry (SIMS) analyses using only picogram sampling masses. However, several studies have reported inconsistencies between SIMS and GS-IRMS δ18O data for the same research material. We aim to solve this controversy by establishing a robust analytical protocol for conodont isotope analysis by SIMS. Here we present conodont data on Pterospathodus and Amorphognathus specimens extracted from Ordovician strata in Nurme and Mehikoorma-421 boreholes (Estonia). Oxygen isotope composition of conodonts was analysed by both SIMS and GS-IRMS, where we paid particular attention to four inorganic apatite reference materials in order to understand the offset between these two techniques that have been reported in the literature. While the results of GS-IRMS measurements conducted using high-temperature reduction of Ag3PO4 represent exclusively δ18O of phosphate-bound oxygen, SIMS analyses do not discriminate between different oxygen components (e.g., (PO4)3â, (SiO4)4â, (CO3)2â, and (OH)â) in apatite, inherently providing information on pooled isotope compositions. We conducted quantitative chemical analyses of selected conodont elements by electron probe microanalysis to assess to what extent matrix effects cause the offsets between the two isotope techniques. We also used scanning electron microscopy and white light optical profilometry to evaluate sample topography and porosity, which have a major impact on SIMS data quality. We collected oxygen isotope data using a CAMECA 1280-HR large geometry instrument at the Potsdam SIMS user facility over several months to determine reproducibility of the results and to optimise a routine measurement protocol. Our tests included a variety of instrumental settings, e.g., different raster parameters for both pre-sputtering and data collection, which yielded slightly differing results due to different instrumental mass fractionation. SIMS is a comparative method, and as such relies on reference materials that have been previously characterised by bulk methods, ideally provided by multiple laboratories. We noted that the inconsistent offsets between SIMS and GS-IRMS data obtained for a given conodont specimen (with SIMS δ18O values in most cases being higher) are linked to reference material measurements that are necessary for conodont data calibration and are often biased towards lighter δ18O values. Our tests show that such bias is even more significant when calibration is based on a single reference material characterised by a single GS-IRMS laboratory, which has been a common practice in past conodont studies.

In this research, an operative procedure for the evaluation of land use change that occurred in highly reclaimed coastal areas from the middle of the XIX century up to the present day is proposed. The multitemporal analysis envisages the use of historical maps, aerial photographs, and satellite images, whose interpretation is performed in a GIS environment. The proposed methodological approach starts from the interpretation of the symbols used in the legend of the historical maps. Subsequently, in order to compare historical information with the most recent land use classifications (i.e., CORINE land cover), a set of twenty-two macro-categories is proposed to find a compromise between a highly detailed land use classification and its applicability to both historical and present-day data. The study area is located in the coastal sector of the Tavoliere di Puglia (Apulia region, Southern Italy), the second-largest coastal plain in Italy. In this area, environmental changes were mostly driven by extensive reclamation and drainage works, which allowed more than 170 km2 of land to be removed from coastal marshes and wetlands. The results show a strong increase in the surface occupied by arable land, urban areas, and saltwork, which today occupy about 57%, 3%, and 23% of the total investigated area, respectively. In contrast, the total surface occupied by grassland, pasture, meadow, and shrubland decreased from 59.6% in 1869 to 4.6% in the present-day setting. It is worth noting that although fluvial sediments were trapped and used to fill the marshy areas, the coastline prograded up to the first half of the XX century, favouring the formation of wide coastal dune systems. Nevertheless, the natural coastal resilience of the investigated system has been reduced since the second half of the XX century, probably as a consequence of the construction of numerous dams in the Ofanto River catchment, which represents the main river in the investigated area.

Growing evidence has shown that high levels of lipoprotein (a) (Lp(a)) and chronic inflammation may be responsible for the residual risk of cardiovascular events in patients managed with an optimal evidence-based approach. Clinical studies have demonstrated a correlation between higher Lp(a) levels and several atherosclerotic diseases including ischemic heart disease, stroke, and degenerative calcific aortic stenosis. The threshold value of Lp(a) serum concentrations associated with a significantly increased cardiovascular risk is >125 nmol/L (50 mg/dL). Current available lipid-lowering drugs have modest-to-no impact on Lp(a) levels. Chronic inflammation is a further condition potentially implicated in residual cardiovascular risk. Consistent evidence has shown an increased risk of cardiovascular events in patients with high sensitivity C reactive protein (>2 mg/dL), an inflammation biomarker. A number of anti-inflammatory drugs have been investigated in patients with or at risk of cardiovascular disease. Of these, canakinumab and colchicine have been found to be associated with cardiovascular risk reduction. Ongoing research aimed at improving risk stratification on the basis of Lp(a) and vessel inflammation assessment may help refine patient management. Furthermore, the identification of these conditions as cardiovascular risk factors has led to increased investigation into diagnostic and therapeutic strategies targeting them in order to reduce atherosclerotic cardiovascular disease burden.
