Background The aim of this study is to describe muscle strength in pediatric patients with repaired tetralogy of Fallot compared with healthy peers and to analyze the correlation between muscle strength and peak oxygen uptake, exercise capacity (mL/min). Methods and Results A prospective, cross‐sectional study was carried out in the University Medical Center Groningen between March 2016 and December 2019, which included 8 ‐to‐19‐year‐old patients with repaired tetralogy of Fallot. Exclusion criteria comprised the following: Down syndrome, unstable pulmonary disease and severe scoliosis affecting pulmonary function, neuromuscular disease, and mental or physical limitations that prohibit the execution of the functional tests. Muscle strength was compared with 2 healthy pediatric cohorts from the Northern Netherlands. Handgrip strength, maximal voluntary isometric contraction, and dynamic muscle strength in correlation with peak oxygen uptake, exercise capacity (mL/min) were the main outcomes of the study. The 67 patients with repaired tetralogy of Fallot (42% female; aged 12.9 [interquartile range, 10.0–16.3] years old) were compared with healthy children. The patients showed reduced grip strength (z‐score [mean±SD] −1.5±1.2, P

Abstract Muscle strength is decreased in adults with pulmonary arterial hypertension (PAH). We aim to investigate muscle strength in children with PAH in relation to a cohort of healthy children, and investigate correlations with disease severity markers. This prospective study included children with PAH aged 4−18 years, who visited the Dutch National Referral Center for Pulmonary Hypertension in Childhood between October 2015 and March 2016. Muscle strength was assessed using handgrip strength and maximum voluntary isometric contractility (MVIC) of four peripheral muscles. Dynamic muscle function was evaluated with the Bruininks−Oseretsky test of motor proficiency (BOT‐2). These measurements were compared with those in two cohorts of healthy children and correlated with 6‐minute walk distance (6MWD), World Health Organization functional class (WHO‐FC), N‐terminal pro‐brain natriuretic peptide (NT‐proBNP), and time since diagnosis. Eighteen children with PAH aged 14.0 [interquartile range: 9.9−16.0] years showed reduced muscle strength. Handgrip strength z‐score −2.4 ± 1.2, p

Chapter 17 in High-Luminosity Large Hadron Collider (HL-LHC) : Preliminary Design Report. The Large Hadron Collider (LHC) is one of the largest scientific instruments ever built. Since opening up a new energy frontier for exploration in 2010, it has gathered a global user community of about 7,000 scientists working in fundamental particle physics and the physics of hadronic matter at extreme temperature and density. To sustain and extend its discovery potential, the LHC will need a major upgrade in the 2020s. This will increase its luminosity (rate of collisions) by a factor of five beyond the original design value and the integrated luminosity (total collisions created) by a factor ten. The LHC is already a highly complex and exquisitely optimised machine so this upgrade must be carefully conceived and will require about ten years to implement. The new configuration, known as High Luminosity LHC (HL-LHC), will rely on a number of key innovations that push accelerator technology beyond its present limits. Among these are cutting-edge 11-12 tesla superconducting magnets, compact superconducting cavities for beam rotation with ultra-precise phase control, new technology and physical processes for beam collimation and 300 metre-long high-power superconducting links with negligible energy dissipation. The present document describes the technologies and components that will be used to realise the project and is intended to serve as the basis for the detailed engineering design of HL-LHC., Comment: 8 pages, chapter 17 in High-Luminosity Large Hadron Collider (HL-LHC) : Preliminary Design Report

The endoplasmic reticulum (ER) stress response is triggered by any condition that disrupts protein folding and promotes the accumulation of unfolded proteins in the lumen of the organelle. In eukaryotic cells, the evolutionarily conserved unfolded protein response is activated to clear unfolded proteins and restore ER homeostasis. The recovery from ER stress is accomplished by decreasing protein translation and loading into the organelle, increasing the ER protein processing capacity and ER-associated protein degradation activity. However, if the ER stress persists and cannot be reversed, the chronically prolonged stress leads to cellular dysfunction that activates cell death signaling as an ultimate attempt to survive. Accumulating evidence implicates ER stress-induced cell death signaling pathways as significant contributors for stress adaptation in plants, making modulators of ER stress pathways potentially attractive targets for stress tolerance engineering. Here, we summarize recent advances in understanding plant-specific molecular mechanisms that elicit cell death signaling from ER stress. We also highlight the conserved features of ER stress-induced cell death signaling in plants shared by eukaryotic cells.

- Abstract
The HL-LHC IT STRING, an integrated test stand for the major components of the HL-LHC Inner Triplet (IT) zone, is in its construction phase in a surface building at CERN. The main motivation is to study and validate the collective behavior of the different systems: magnets, circuit protection, cryogenics for magnets and superconducting link, magnet powering, vacuum, alignment, and interconnections between magnets and superconducting link. During the past two years, the major focus has been on the technical infrastructure definition and implementation, while preparing the installation sequences and procedures for the major elements. The String Validation Program (SVP) has been agreed with the HL-LHC Work Packages, allowing to set up a joint and optimized test program and to integrate it in a schedule. In this paper we describe the IT String installations and describe the main differences between the HL-LHC IT String and the future HL-LHC machine. The main line of the test program and the motivation of a full thermal cycle with a cost benefit analysis will be presented.

Physics - Accelerator Physics - Abstract
The multi-MW target proposed for the EURISOL facility will be based on fission of uranium (or thorium) compounds to produce rare isotopes far from stability. A two-step process is used for the isotope production. First, neutrons are generated in a liquid mercury target, irradiated by the 1 GeV proton or deuteron beam, provided by the EURISOL linac driver. Then, the neutrons induce fission in a surrounding assembly of uranium carbide. R&D projects on several aspects of the target assembly are ongoing. Key criteria for the target design are a maximum beam power capability of 4 MW, a remote handling system with minimum downtime and maximum reliability, as well as radiation safety, minimization of hazards and the classification of the facility. In the framework of the ongoing radiation characterization and safety studies, radiation transport simulations have been performed to calculate the prompt radiation dose in the target and surrounding materials, as well as to determine shielding material and angle-dependent parameters. In this paper, we report the results of these studies and the proposed radiation shield design for the multi-MW target area. Furthermore, accurate estimates have been performed of the amount of fissile elements being produced in the uranium target assembly, for typical running conditions, in order to understand the implications for the classification of the facility. The results are reported and briefly discussed., Comment: 11 pages,5 figures, Invited talk at the SATIF-8 Workshop, May 22-24,2006, Pohang Accelerator Laboratory, Rep. Of Korea

GmNAC81 ,Glycine max ,leaf senescence ,drought tolerance ,NAC transcription factors ,Genetics ,QH426-470 - Abstract
Glycine max NAC81 (GmNAC81) is a downstream effector of the DCD/NRP-mediated cell death signaling, which interacts with GmNAC30 to fully induce the caspase 1-like vacuolar processing enzyme (VPE) expression, the executioner of the cell death program. GmNAC81 has been previously shown to positively modulate leaf senescence via the NRP/GmNAC81/VPE signaling module. Here, we examined the transcriptome induced by GmNAC81 overexpression and leaf senescence and showed that GmNAC81 further modulates leaf senescence by regulating an extensive repertoire of functionally characterized senescence-associated genes (SAGs). Because the NRP/GmNAC81/VPE signaling circuit also relays stress-induced cell death signals, we examined the effect of GmNAC81 overexpression in drought responses. Enhanced GmNAC81 expression in the transgenic lines increased sensitivity to water deprivation. Under progressive drought, the GmNAC81-overexpressing lines displayed severe leaf wilting, a larger and faster decline in leaf Ψw, relative water content (RWC), photosynthesis rate, stomatal conductance, and transpiration rate, in addition to higher Ci/Ca and lower Fm/Fv ratios compared to the BR16 control line. Collectively, these results indicate that the photosynthetic activity and apparatus were more affected by drought in the transgenic lines. Consistent with hypersensitivity to drought, chlorophyll loss, and lipid peroxidation were higher in the GmNAC81-overexpressing lines than in BR16 under dehydration. In addition to inducing VPE expression, GmNAC81 overexpression uncovered the regulation of typical drought-responsive genes. In particular, key regulators and effectors of ABA signaling were suppressed by GmNAC81 overexpression. These results suggest that GmNAC81 may negatively control drought tolerance not only via VPE activation but also via suppression of ABA signaling.

Abstract Background Research on Juvenile Idiopathic Arthritis (JIA) should support patients, caregivers/parents (carers) and clinicians to make important decisions in the consulting room and eventually to improve the lives of patients with JIA. Thus far these end-users of JIA-research have rarely been involved in the prioritisation of future research. Main body Dutch organisations of patients, carers and clinicians will collaboratively develop a research agenda for JIA, following the James Lind Alliance (JLA) methodology. In a ‘Priority Setting Partnership’ (PSP), they will gradually establish a top 10 list of the most important unanswered research questions for JIA. In this process the input from clinicians, patients and their carers will be equally valued. Additionally, focus groups will be organised to involve young people with JIA. The involvement of all contributors will be monitored and evaluated. In this manner, the project will contribute to the growing body of literature on how to involve young people in agenda setting in a meaningful way. Conclusion A JIA research agenda established through the JLA method and thus co-created by patients, carers and clinicians will inform researchers and research funders about the most important research questions for JIA. This will lead to research that really matters.

- Abstract
BACKGROUND: Genomic regions identified by genome-wide association studies (GWAS) for bladder cancer risk provide new insights into etiology. OBJECTIVE: To identify new susceptibility variants for bladder cancer in a meta-analysis of new and existing genome-wide genotype data. DESIGN, SETTING, AND PARTICIPANTS: Data from 32 studies that includes 13,790 bladder cancer cases and 343,502 controls of European ancestry were used for meta-analysis. OUTCOME MEASUREMENTS AND STATISTICAL ANALYSES: Log-additive associations of genetic variants were assessed using logistic regression models. A fixed-effects model was used for meta-analysis of the results. Stratified analyses were conducted to evaluate effect modification by sex and smoking status. A polygenic risk score (PRS) was generated on the basis of known and novel susceptibility variants and tested for interaction with smoking. RESULTS AND LIMITATIONS: Multiple novel bladder cancer susceptibility loci (6p.22.3, 7q36.3, 8q21.13, 9p21.3, 10q22.1, 19q13.33) as well as improved signals in three known regions (4p16.3, 5p15.33, 11p15.5) were identified, bringing the number of independent markers at genome-wide significance (p < 5 × 10-8) to 24. The 4p16.3 (FGFR3/TACC3) locus was associated with a stronger risk for women than for men (p-interaction = 0.002). Bladder cancer risk was increased by interactions between smoking status and genetic variants at 8p22 (NAT2; multiplicative p value for interaction [pM-I] = 0.004), 8q21.13 (PAG1; pM-I = 0.01), and 9p21.3 (LOC107987026/MTAP/CDKN2A; pM-I = 0.02). The PRS based on the 24 independent GWAS markers (odds ratio per standard deviation increase 1.49, 95% confidence interval 1.44-1.53), which also showed comparable results in two prospective cohorts (UK Biobank, PLCO trial), revealed an approximately fourfold difference in the lifetime risk of bladder cancer according to the PRS (e.g., 1st vs 10th decile) for both smokers and nonsmokers. CONCLUSIONS: We report novel loci associated

The release of the last soybean (Glycine max) genome assembly (Wm82.a2.v1) raised the possibility that new NAC genes would be present in the soybean genome. Here, we interrogated the last version of the soybean genome against a conserved NAC domain structure. Our analysis identified 32 putative novel NAC genes, updating the superfamily to 180 gene members. We also organized the genes in 15 phylogenetic subfamilies, which showed a perfect correlation among sequence conservation, expression profile, and function of orthologous Arabidopsis thaliana genes and NAC soybean genes. To validate our in silico analyses, we monitored the stress-mediated gene expression profiles of eight new NAC-genes by qRT-PCR and monitored the GmNAC senescence-associated genes by RNA-seq. Among ER stress, osmotic stress and salicylic acid treatment, all the novel tested GmNAC genes responded to at least one type of stress, displaying a complex expression profile under different kinetics and extension of the response. Furthermore, we showed that 40% of the GmNACs were differentially regulated by natural leaf senescence, including eight (8) newly identified GmNACs. The developmental and stress-responsive expression profiles of the novel NAC genes fitted perfectly with their phylogenetic subfamily. Finally, we examined two uncharacterized senescence-associated proteins, GmNAC065 and GmNAC085, and a novel, previously unidentified, NAC protein, GmNAC177, and showed that they are nuclear localized, and except for GmNAC065, they display transactivation activity in yeast. Consistent with a role in leaf senescence, transient expression of GmNAC065 and GmNAC085 induces the appearance of hallmarks of leaf senescence, including chlorophyll loss, leaf yellowing, lipid peroxidation and accumulation of H2O2. GmNAC177 was clustered to an uncharacterized subfamily but in close proximity to the TIP subfamily. Accordingly, it was rapidly induced by ER stress and by salicylic acid under late kinetic response and promoted cell death in planta. Collectively, our data further substantiated the notion that the GmNAC genes display functional and expression profiles consistent with their phylogenetic relatedness and established a complete framework of the soybean NAC superfamily as a foundation for future analyses.

- Abstract
Abstract Single-unit recording of layer II-III cells in olfactory (piriform) cortex was performed on awake, unrestrained rats actively engaged in learning novel odors in an olfactory discrimination task. Five of the 67 cells tested had very brief monophasic action potentials and high spontaneous firing rates (30-80 Hz); it is suggested that these units were interneurons. The remainder of the neurons had broader spikes and did not discharge for prolonged periods. Thirty-nine percent of the broad spike cells responded to at least one and usually more of the odors presented to the rats during either of the first two trials on which that odor was present, but, in most cases, these responses occurred only very infrequently over the course of subsequent trials. Six percent of the broad-spike group, how ever, continued firing robustly to a single odor but not to others. From these results it appears that most cells in piriform cortex do not respond to most odors, i.e., coding is exceedingly sparse. A subgroup of the predominant broad-spike cell type does react to several odors but this response drops out with repeated exposure, perhaps because of training. However, a few members of this class (a small fraction of the total cell population) do go on responding to a particular odor, thus exhibiting a form of odor specificity. The results are discussed with regard to predictions from recently developed models of the olfactory cortex.

Toward accurately simulating both hardening and softening effects for metals up to failure, a new finite strain elastoplastic J2-flow model is proposed with the yield strength therein as a function of the plastic work in the explicit form. With no need to identify any adjustable parameters, the uniaxial stress-strain response predicted from this new model is shown to automatically and accurately match any given data from monotonic uniaxial extension tests of bars. As such, the objectives in three respects are achieved for the first time, i.e., (i) both the hardening and softening effects up to failure can be simulated in the sense of matching test data with no errors, (ii) the usual tedious implicit procedures toward identifying numerous unknown parameters need not be involved and can be totally bypassed, and (iii) the model applicability can be ensured in a broad sense for various metallic materials with markedly different transition effects from hardening to softening. With the new model, the complete response features of stretched bars and twisted tubes up to failure are studied, including the failure effects of bars under monotonic extension and tubes under monotonic torsion and, furthermore, the fatigue failure effects of bars under cyclic loading. The results show accurate agreement with the uniaxial data, and the results for both the shear stress and the normal stress at the finite torsion display realistic hardening-to-softening transition effects for the first time.

Cost is one aspect of treatment to be considered, in addition to clinical benefits, when selecting optimal therapy for a patient. The objective of this study was to estimate the average dose usage and real world daily cost of the GLP-1 receptor agonists, exenatide twice daily and liraglutide once daily, in Germany, the Netherlands, and the UK.Methods: Administrative databases were used to source the data from longitudinal records of dispensed prescriptions. Data were extracted from the IMS Longitudinal Prescription database which captures details of prescriptions dispensed in pharmacies. Information on the dispensed quantity of each product was used to estimate average daily usage per patient. Daily dose usage was multiplied by the public price per unit to estimate daily cost. Results: The dispensed volume in Germany corresponded to a mean dispensed daily dose of 16.81 µg for exenatide twice daily and 1.37 mg for liraglutide (mean daily cost €4.02 and €4.54, respectively). In the Netherlands, average dispensed daily doses of 17.07 µg and 1.49 mg were observed for exenatide twice daily and liraglutide (mean daily cost €3.05 and €3.97, respectively). In the UK, the mean dispensed volume corresponded to a daily usage of 20.49 µg for exenatide twice daily and 1.50 mg for liraglutide (mean daily cost £2.53 and £3.28, respectively). Conclusion: Estimates of average daily dispensed doses of GLP-1 receptor agonists derived from pharmacy data in real world settings corresponded to the dosing recommendation of the summaries of product characteristics. Nevertheless, the mean daily cost of exenatide twice daily was lower than that of liraglutide in Germany, the Netherlands, and the UK. Such estimates can be used to inform health care decision-makers on the real world usage and cost of medications effective in achieving glycemic control in patients with type 2 diabetes. Keywords: type 2 diabetes, exenatide twice daily, liraglutide, dosage, cost, real world

Background: To investigate the possible associations between childhood noninfectious uveitis and cardio-respiratory fitness, physical activity, health related quality of life and fatigue. Methods: Cross-sectional analysis of 23 patients with noninfectious uveitis, aged 8-18 years. BMI, exercise capacity, muscle strength and physical activity were measured. Health-related quality of life and fatigue were assessed. The results were compared to standardized values for age matched healthy children. Results: Twenty-three patients were included. Children with uveitis had a higher bodyweight and body mass index. Children with uveitis had lower cardio-respiratory fitness and they were less physically active, but they experienced a normal quality of life and normal fatigue. Parents of children with uveitis reported a lower quality of life and more fatigue for their children than parents of healthy children. Conclusion: Our study indicates that children with noninfectious uveitis are at risk of developing lower physical and psychosocial health.

Background Involving the end-users of scientific research (patients, carers and clinicians) in setting research priorities is important to formulate research questions that truly make a difference and are in tune with the needs of patients. We therefore aimed to generate a national research agenda for Juvenile Idiopathic Arthritis (JIA) together with patients, their caregivers and healthcare professionals through conducting a nationwide survey among these stakeholders. Methods The James Lind Alliance method was used, tailored with additional focus groups held to involve younger patients. First, research questions were gathered through an online and hardcopy survey. The received questions that were in scope were summarised and a literature search was performed to verify that questions were unanswered. Questions were ranked in the interim survey, and the final top 10 was chosen during a prioritisation workshop. Results Two hundred and seventy-eight respondents submitted 604 questions, of which 519 were in scope. Of these 604 questions, 81 were generated in the focus groups with younger children. The questions were summarised into 53 summary questions. An evidence checking process verified that all questions were unanswered. A total of 303 respondents prioritised the questions in the interim survey. Focus groups with children generated a top 5 of their most important questions. Combining this top 5 with the top 10s of patients, carers, and clinicians led to a top 21. Out of these, the top 10 research priorities were chosen during a final workshop. Research into pain and fatigue, personalised treatment strategies and aetiology were ranked high in the Top 10. Conclusions Through this study, the top 10 research priorities for JIA of patients, their caregivers and clinicians were identified to inform researchers and research funders of the research topics that matter most to them. The top priority involves the treatment and mechanisms behind persisting pain and fatigue when the disease is in remission.

International audience; The olive tree was an iconic plant for most of the past Mediterranean civilizations, for which it had important economic value. Here we report the earliest use of fruits and wood from olive trees in Africa so far, around 100,000 years ago. These findings suggest the presence of olive trees on the Atlantic coast of Morocco during most of the last glacial period, and the use of olives by the early Homo sapiens for fuel management and most probably for consumption.

The theory of plasticity as a special field of continuum mechanics deals with the irreversible, i.e. permanent, deformation of solids. Under the action of given loads or deformations, the state of the stresses and strains or the strain rates in these bodies is described. In this way, it complements the theory of elasticity for the reversible behavior of solids. In practice, it has been observed that many materials behave elastically up to a certain load (yield point), beyond that load, however, increasingly plastic or liquid-like. The combination of these two material properties is known as elastoplasticity. The classical elastoplastic material behavior is assumed to be time-independent or rate-independent. In contrast, we call a time- or rate-dependent behavior visco-elastoplastic and visco-plastic—if the elastic part of the deformation is neglected. In plasticity theory, because of the given loads the states of the state variables stress, strain and temperature as well as their changes are described. For this purpose, the observed phenomena are introduced and put into mathematical relationships. The constitutive relations describing the specific material behavior are finally embedded in the fundamental relations of continuum theory and physics. Historically, the theory of plasticity was introduced in order to better estimate the strength of constructions. An analysis based purely on elastic codes is not in a position to do this, and can occasionally even lead to incorrect interpretations. On the other hand, the entire field of forming techniques requires a theory for the description of plastic behavior. Starting from the classical description of plastic behavior with small deformations, the present review is intended to provide an insight into the state of the art when taking into account finite deformations.

Objectives To evaluate the clinical utility of the urinary bladder cancer antigen test UBC (R) Rapid for the diagnosis of bladder cancer (BC) and to develop and validate nomograms to identify patients at high risk of primary BC. Patients and Methods Data from 1787 patients from 13 participating centres, who were tested between 2012 and 2020, including 763 patients with BC, were analysed. Urine samples were analysed with the UBC (R) Rapid test. The nomograms were developed using data from 320 patients and externally validated using data from 274 patients. The diagnostic accuracy of the UBC (R) Rapid test was evaluated using receiver-operating characteristic curve analysis. Brier scores and calibration curves were chosen for the validation. Biopsy-proven BC was predicted using multivariate logistic regression. Results The sensitivity, specificity, and area under the curve for the UBC (R) Rapid test were 46.4%, 75.5% and 0.61 (95% confidence interval [CI] 0.58-0.64) for low-grade (LG) BC, and 70.5%, 75.5% and 0.73 (95% CI 0.70-0.76) for high-grade (HG) BC, respectively. Age, UBC (R) Rapid test results, smoking status and haematuria were identified as independent predictors of primary BC. After external validation, nomograms based on these predictors resulted in areas under the curve of 0.79 (95% CI 0.72-0.87) and 0.95 (95% CI: 0.92-0.98) for predicting LG-BC and HG-BC, respectively, showing excellent calibration associated with a higher net benefit than the UBC (R) Rapid test alone for low and medium risk levels in decision curve analysis. The R Shiny app allows the results to be explored interactively and can be accessed at www.blucab-index. net. Conclusion The UBC (R) Rapid test alone has limited clinical utility for predicting the presence of BC. However, its combined use with BC risk factors including age, smoking status and haematuria provides a fast, highly accurate and non-invasive tool for screening patients for primary LG-BC and especially primary HG-BC.

We propose an enhanced form of thermocoupled J2-flow models of finite deformation elastoplasticity with temperature-dependent yielding and hardening behaviour. The thermomechanical constitutive structure of these models is rendered free and explicit in the rigorous sense of thermodynamic consistency. Namely, with a free energy function explicitly introduced in terms of almost any given form of the thermomechanical constitutive functions, the requirements from the second law are identically fulfilled with positive internal dissipation. We study the case when a dependence of yielding and hardening on temperature is given and demonstrate that thermosensitive yielding with anisotropic hardening may give rise to appreciable plastic flow either in a process of heating or in a cyclic process of heating/cooling, thus leading to the findings of one- and two-way thermoinduced plastic flow. We then show that such theoretical findings turn out to be the effects found in shape memory materials, such as one- and two-way memory effects. Thus, shape memory effects may be explained to be thermoinduced plastic flow resulting from thermosensitive yielding and hardening behaviour. These and other relevant facts may suggest that, from a phenomenological standpoint, thermocoupled elastoplastic J2-flow models with thermosensitive yielding and hardening may furnish natural, straightforward descriptions of thermomechanical behaviour of shape memory materials.

- Abstract
In response to the 2013 Update of the European Strategy for Particle Physics, the Future Circular Collider (FCC) study was launched, as an international collaboration hosted by CERN. This study covers a highest-luminosity high-energy lepton collider (FCC-ee) and an energy-frontier hadron collider (FCC-hh), which could, successively, be installed in the same 100 km tunnel. The scientific capabilities of the integrated FCC programme would serve the worldwide community throughout the 21st century. The FCC study also investigates an LHC energy upgrade, using FCC-hh technology. This document constitutes the second volume of the FCC Conceptual Design Report, devoted to the electron-positron collider FCC-ee. After summarizing the physics discovery opportunities, it presents the accelerator design, performance reach, a staged operation scenario, the underlying technologies, civil engineering, technical infrastructure, and an implementation plan. FCC-ee can be built with today’s technology. Most of the FCC-ee infrastructure could be reused for FCC-hh. Combining concepts from past and present lepton colliders and adding a few novel elements, the FCC-ee design promises outstandingly high luminosity. This will make the FCC-ee a unique precision instrument to study the heaviest known particles (Z, W and H bosons and the top quark), offering great direct and indirect sensitivity to new physics.
