17. Current state of Fusarium wilt of banana in the subtropics

Abstract

Subtropically grown bananas have different interactions with Fusarium oxysporum f. sp. cubense (Foc), causal agent of Fusarium wilt of banana (FWB), when compared with those grown in the tropics. The designation ‘subtropical’ race 4 (SR4) was assigned to Foc strains able to infect Cavendish (AAA) cultivars in the subtropics, but not in the tropics. ‘Tropical’ race 4 (TR4) gained prominence with the appearance and spread of a Foc strain that severely affects Cavendish bananas in both the tropics and subtropics. Recent surveys in Asia revealed TR4 as the predominant strain both in the tropics and subtropics, affecting not only Cavendish, but also many other cultivars. In many subtropical banana zones, Foc races 1 and 2, which complete the racial structure of Foc, are also found and cause severe losses in certain market-preferred dessert bananas. Environmental constraints increase the susceptibility of Cavendish bananas to SR4. However, the role of abiotic stresses in cultivar susceptibility to Foc needs to be better understood. It is not clear why some Foc strains only affect Cavendish in the subtropics, and what other factors, including pathogen virulence and microbial functional diversity, might be driving disease epidemics. Field experience suggests that optimum soil and water management strategies can reduce disease intensity, but targeted studies are needed to understand the virulence of different strains, the impact of fluctuating temperatures, soil pH, organic matter breakdown and soil microbial dynamics. Research findings on the epidemiology and management of FWB in Australia, Brazil, China, India and South Africa are discussed to propose priority areas for the development of improved disease management practices. © 2020 International Society for Horticultural Science. Abstract

An accelerated negative buoyancy method has been developed to assess cell adhesion strength. This method has been used in conjunction with 3-D morphometric analysis to understand the effects of surface topology on cell response. Aligned micro-grooved surface topographies (with a range of groove depths) were produced on stainless steel 316L substrates by laser ablation. An investigation was carried out on the effect of the micro-grooved surface topography on cell adhesion strength, cell and nucleus volumes, cell phenotypic expression and attachment patterns. Increased hydrophobicity and anisotropic wettability was observed on surfaces with deeper grooves. A reduction was noted in cell volume, projected areas and adhesion sites for deeper grooves, linked to lower cell proliferation and differentiation rates and also to reduced adhesion strength. The results suggest that the centrifugation assay combined with three-dimensional cell morphometric analysis has considerable potential for obtaining improved understanding of the cell/substrate interface.

Abstract

The effects of energy pulse packing on material transport during single-pulse laser processing of < 1 | 1 | 1 > silicon is studied using temporarily shaped pulses with durations from 50 to 150 ns. Six regimes of material transport were identified and disambiguated through energy packing considerations over a range of pulse durations. Energy packing has been shown to shift the interaction to energetically costlier regimes without appreciable benefit in either depth, material removal or crater morphology and quality.

Abstract

Three Gros Michel mutants (‘IBP 5-B’, ‘IBP 5–61’ and ‘IBP 12’) from the Cuban Instituto de Biotecnología de las Plantas, two semi-dwarf Gros Michel varieties (‘Highgate’ and ‘Cocos’) and a Thai accession (‘Hom Thong Mokho’) were evaluated in Australia over a five year period. They were screened for their resistance to Fusarium wilt Race 1 (FocR1) caused by the pathogen Fusarium oxysporum f.sp. cubense, as well as resistance to yellow Sigatoka (Pseudocercospora musae Zimm [teleomorph Mycosphaerella musicola Leach]). They were also grown for a plant and ratoon crop in the tropics (17°S) and a plant crop in the subtropics (28°S) with no disease pressure to record their agronomic characteristics. They were compared with Australian industry standards, ‘Williams’ (AAA, Cavendish subgroup) and ‘Lady Finger’ (AAB, Pome subgroup). In the subtropics the Gros Michel mutants and semi-dwarf accessions were sensitive to cold and very susceptible to FocR1 and yellow Sigatoka while their agronomic performance in the tropics was good, with ‘Highgate’ having the best bunch weight on a shorter, more manageable plant. Of the six accessions evaluated, ‘Hom Thong Mokho’ showed the highest level of resistance to FocR1 although it had poor cold tolerance, as did the other Gros Michel selections, and consequently had low productivity compared to ‘Williams’ and even ‘Lady Finger’. However in the warmer, more humid tropics ‘Hom Thong Mokho's performance was much better and it was less susceptible to yellow Sigatoka than the other Gros Michel selections. Subsequent genetic analysis by Christelová et al. (2011) has revealed that although ‘Hom Thong Mokho’ is marketed as a Gros Michel variety in Asia, it is closer to the Rio subgroup (AAA) of dessert bananas.

Abstract

Aim To investigate the putative modifying effect of dual antiplatelet therapy (DAPT) use on the incidence of stent thrombosis at 3 years in patients randomized to Endeavor zotarolimus-eluting stent (E-ZES) or Cypher sirolimus-eluting stent (C-SES). Methods and results Of 8709 patients in PROTECT, 4357 were randomized to E-ZES and 4352 to C-SES. Aspirin was to be given indefinitely, and clopidogrel/ticlopidine for ≥3 months or up to 12 months after implantation. Main outcome measures were definite or probable stent thrombosis at 3 years. Multivariable Cox regression analysis was applied, with stent type, DAPT, and their interaction as the main outcome determinants. Dual antiplatelet therapy adherence remained the same in the E-ZES and C-SES groups (79.6% at 1 year, 32.8% at 2 years, and 21.6% at 3 years). We observed a statistically significant (P = 0.0052) heterogeneity in treatment effect of stent type in relation to DAPT. In the absence of DAPT, stent thrombosis was lower with E-ZES vs. C-SES (adjusted hazard ratio 0.38, 95% confidence interval 0.19, 0.75; P = 0.0056). In the presence of DAPT, no difference was found (1.18; 0.79, 1.77; P = 0.43). Conclusion A strong interaction was observed between drug-eluting stent type and DAPT use, most likely prompted by the vascular healing response induced by the implanted DES system. These results suggest that the incidence of stent thrombosis in DES trials should not be evaluated independently of DAPT use, and the optimal duration of DAPT will likely depend upon stent type (Clinicaltrials.gov number NCT00476957)

Abstract

Anodes of 304 stainless steel have been processed by a continuous wave Yb fiber laser with a wavelength of 1.064 μm and subjected to 50 keV electron bombardment in order to determine the extent to which hydrogen outgassing is reduced by the laser surface melting treatment. The results show a reduction in outgassing, by approximately a factor of four compared to that from untreated stainless steel. This is attributed to a reduction in the number of grain boundaries which serve as trapping sites for hydrogen in stainless steel. Such laser treated anodes do not require post-processing to preserve the benefits of the treatment and are excellent candidates for use in high power source devices.

43. Cerebral blood flow regulation in end-stage kidney disease.

Abstract

Patients with chronic kidney disease (CKD) and end-stage kidney disease (ESKD) experience an increased risk of cerebrovascular disease and cognitive dysfunction. Hemodialysis (HD), a major modality of renal replacement therapy in ESKD, can cause rapid changes in blood pressure, osmolality, and acid-base balance that collectively present a unique stress to the cerebral vasculature. This review presents an update regarding cerebral blood flow (CBF) regulation in CKD and ESKD and how the maintenance of cerebral oxygenation may be compromised during HD. Patients with ESKD exhibit decreased cerebral oxygen delivery due to anemia, despite cerebral hyperperfusion at rest. Cerebral oxygenation further declines during HD due to reductions in CBF, and this may induce cerebral ischemia or “stunning.” Intradialytic reductions in CBF are driven by decreases in cerebral perfusion pressure that may be partially opposed by bicarbonate shifts during dialysis. Intradialytic reductions in CBF have been related to several variables that are routinely measured in clinical practice including ultrafiltration rate and blood pressure. However, the role of compensatory cerebrovascular regulatory mechanisms during HD remains relatively unexplored. In particular, cerebral autoregulation can oppose reductions in CBF driven by reductions in systemic blood pressure, while cerebrovascular reactivity to CO2 may attenuate intradialytic reductions in CBF through promoting cerebral vasodilation. However, whether these mechanisms are effective in ESKD and during HD remain relatively unexplored. Important areas for future work include investigating potential alterations in cerebrovascular regulation in CKD and ESKD and how key regulatory mechanisms are engaged and integrated during HD to modulate intradialytic declines in CBF.
