The specific genes and molecules that drive physiological angiogenesis differ from those involved in pathological angiogenesis, suggesting distinct mechanisms for these seemingly related processes. Unveiling genes and pathways preferentially associated with pathologic angiogenesis is key to understanding its mechanisms, thereby facilitating development of novel approaches to managing angiogenesis-dependent diseases. To better understand these different processes, we elucidated the transcriptome of the mouse retina in the well-accepted oxygen-induced retinopathy (OIR) model of pathological angiogenesis. We identified 153 genes changed between normal and OIR retinas, which represent a molecular signature relevant to other angiogenesis-dependent processes such as cancer. These genes robustly predict the survival of breast cancer patients, which was validated in an independent 1,000-patient test cohort (40% difference in 15-year survival; p = 2.56 x 10-21). These results suggest that the OIR model reveals key genes involved in pathological angiogenesis, and these may find important applications in stratifying tumors for treatment intensification or for angiogenesis-targeted therapies.

9. Development of a carotenoid-rich microalgae colorant by microencapsulation

- Abstract
Producción Científica, Dunaliella salina has been recognized as an excellent biomass source of carotenoid, which can be used as a natural orange coloring agent for food products. The most eco-friendly approach for extracting carotenoid is through supercritical carbon dioxide extraction, as it yields highly concentrated extracts while preventing pigment thermal degradation. However, there are limitations when a lipophilic extract is considered a food ingredient, in particular very difficult handling and low solubility in water-based products. The aim of this study was to develop a hydrosoluble form of a natural carotenoid-rich extract recovered from algae biomass within a biorefinery concept to be incorporated in aqueous-based food products. A two-step process was developed, starting with the emulsification of the supercritical extract into a mixture of maltodextrin and gum arabic, using soy lecithin as an emulsifier. The emulsification was followed by a spray-drying step. The impact of process variables on the encapsulation yield, efficiency, emulsion properties, and particle characteristics was studied. The resulting particles exhibited an intense orange color and good water dispersibility, facilitating uniform yellow coloring when incorporated into an aqueous-based product. Prostate cancer antigen 3 (PCA3) is the most specific prostate cancer biomarker but its function remains unknown. Here we identify PRUNE2, a target protein-coding gene variant, which harbors the PCA3 locus, thereby classifying PCA3 as an antisense intronic long noncoding (lnc)RNA. We show that PCA3 controls PRUNE2 levels via a unique regulatory mechanism involving formation of a PRUNE2/PCA3 double-stranded RNA that undergoes adenosine deaminase acting on RNA (ADAR)-dependent adenosine-to-inosine RNA editing. PRUNE2 expression or silencing in prostate cancer cells decreased and increased cell proliferation, respectively. Moreover, PRUNE2 and PCA3 elicited opposite effects on tumor growth in immunodeficient tumor-bearing mice. Coregulation and RNA editing of PRUNE2 and PCA3 were confirmed in human prostate cancer specimens, supporting the medical relevance of our findings. These results establish PCA3 as a dominant-negative oncogene and PRUNE2 as an unrecognized tumor suppressor gene in human prostate cancer, and their regulatory axis represents a unique molecular target for diagnostic and therapeutic intervention.

MotivationExtracellular vesicles (EVs) are spherical bilayered proteolipids, harboring various bioactive molecules. Due to the complexity of the vesicular nomenclatures and components, online searches for EV-related publications and vesicular components are currently challenging.ResultsWe present an improved version of EVpedia, a public database for EVs research. This community web portal contains a database of publications and vesicular components, identification of orthologous vesicular components, bioinformatic tools and a personalized function. EVpedia includes 6879 publications, 172 080 vesicular components from 263 high-throughput datasets, and has been accessed more than 65 000 times from more than 750 cities. In addition, about 350 members from 73 international research groups have participated in developing EVpedia. This free web-based database might serve as a useful resource to stimulate the emerging field of EV research.Availability and implementationThe web site was implemented in PHP, Java, MySQL and Apache, and is freely available at http://evpedia.info.

In recent decades, wildfires have become common disasters that threaten people's lives and assets, particularly in wildland–urban interfaces (WUIs). Developing an effective evacuation strategy for a WUI presents challenges to emergency planners because of the spatial variations in biophysical hazards and social vulnerability. The aim of this study was to map priority WUIs in terms of evacuation. The factors considered were the seriousness of the risk of wildfire exposure, and the population centres whose greatest constraints on the evacuation process stemmed from the nature of the exposed population and the time required to travel to the nearest shelter/refuge. An integrated framework linking wildfire hazard, social vulnerability, and the time taken to travel by foot or by car to the nearest refuge/shelter was applied. The study area includes two municipalities (Lousã and Sertã) in the mountainous areas of central Portugal that are in high-wildfire-risk areas and have very vulnerable and scattered pockets of exposed population. The combination of wildfire risk and travelling time to the nearest shelters made it possible to identify 20% of the WUIs that were priority areas for evacuation in the case of Sertã. In the case of Lousã, 3.4% were identified, because they were highly exposed to wildfire risk and had a travelling time to the nearest shelter of more than 15 min on foot. These results can assist in designing effective pre-fire planning, based on fuel management strategies and/or managing an effective and safe evacuation. [ABSTRACT FROM AUTHOR]

15. Further Studies on the Effects of Greening on Juice Quality: Do Nutritional Sprays Ameliorate HLB-Induced Off-flavor?

- Abstract
Citrus groves receiving nutritional sprays were compared with groves in the same areas managed with conventional fertilization treatments. Fruit were harvested from healthy and Huanglongbing (HLB)-affected trees. Within HLB-affected trees, fruit were sorted into asymptomatic (HLB-a) and symptomatic (HLB-s) fruit. Sensory tests were performed using the difference-from-control (DFC) method, where juice from HLB-affected trees was compared with juice from healthy trees. Results show that panelists could detect differences between juice from HLB-affected and healthy trees in the 2009-2010 and 2010-2011 seasons, regardless of nutritional treatments, for Hamlin and Valencia. Like in previous years, those differences were perceived as more bitter or metallic for early harvests of HLB-affected Hamlin, but those differences were less and inconsistently perceived as more bitter, sweeter or more sour for late harvests or Valencia HLB-affected fruit. In the 2011-2012 season, there were much less differences between juice from healthy and HLB trees, possibly due to a season with high Brix/TA ratio, or due to later harvests. Results will be discussed in relation to chemical analysis of sugars, acids, and limonoids. Nutritional treatments that mitigate HLB symptoms on trees did not have a consistent effect on the HLB induced off-flavor of the fruit and juice, necessitating more seasons of study.

- Abstract
Forest remnants are hotspots of biodiversity and play an important role in providing services such as regulating the climate, reducing surface runoff, helping erosion control, protecting and contributing to the balance of ecosystems, and other functions. Despite this, natural vegetation is fragmented and limited to a few remnants, which are gradually suffering from anthropogenic pressures. Assessing the environmental quality of these remnants is therefore vital to understanding their current condition and to provide support for their conservation. This study aims to assess the environmental quality of forest remnants in six water basins in the municipality of Campinas/SP, Brazil. Forest remnants were mapped, and their environmental quality was assessed by applying an analytic hierarchy process (AHP), considering a set of structural landscape metrics previously selected from the literature. Of the 2319 forest remnants evaluated, 4.5% and 30%, respectively, registered high and low environmental quality. The Atibaia and Jaguari basins recorded the highest number of environmentally fragile remnants due to their small size and being predominantly elongated, and to the high erodibility of the soil. In the Anhumas, Capivari, Capivari-Mirim, and Quilombo basins, medium-sized forest remnants predominate. There is a greater distance between them, with a high intensity of land use/land cover in their surroundings, related to the prevalence of urbanized areas. The main objectives of this study were to understand the frequency of forest fires, post-fire off-site hydrological response and erosional processes from a social and ecological perspective in two basins located in the central cordillera, Portugal. It also discusses the driving forces that contribute towards increasing the social-ecological vulnerability of systems in the face of hazards and emphasizes the importance of learning from disasters. Based on the historical incidence of wildfires, it is possible to identify several areas affected by two, three or four fires, since 1975. Following the two major fires, in 1987 and 2005, flash floods, intense soil erosion and sedimentation processes were generated, causing severe damage. Significant socioeconomic, political and ecological changes have been affecting mountain regions in the last decades. Approximately 80% of the population and more than 90% of the livestock have disappeared, common lands have been afforested with Pinus pinaster, and several agricultural plots have been abandoned. These factors have all contributed towards creating non- or submanaged landscapes that have led to a dramatic increase in the magnitude and frequency of wildfires and to post-fire hydrological and erosional processes when heavy rainfall occurs. Moreover, the low population density, high level of population ageing and very fire-prone vegetation that now covers large areas of both basins, contribute to a situation of extreme socio-ecological vulnerability, meaning that disasters will continue to occur unless resilience can be restored to improve the capacity to cope with this high susceptibility to hazards.

- Abstract
Whereas targeted and shotgun sequencing approaches are both powerful in allowing the study of tissue-associated microbiota, the human: microorganism abundance ratios in tissues of interest will ultimately determine the most suitable sequencing approach. In addition, it is possible that the knowledge of the relative abundance of bacteria and fungi during a treatment course or in pathological conditions can be relevant in many medical conditions. Here, we present a qPCR-targeted approach to determine the absolute and relative amounts of bacteria and fungi and demonstrate their relative DNA abundance in nine different human tissue types for a total of 87 samples. In these tissues, fungi genomes are more abundant in stool and skin samples but have much lower levels in other tissues. Bacteria genomes prevail in stool, skin, oral swabs, saliva, and gastric fluids. These findings were confirmed by shotgun sequencing for stool and gastric fluids. This approach may contribute to a more comprehensive view of the human microbiota in targeted studies for assessing the abundance levels of microorganisms during disease treatment/progression and to indicate the most informative methods for studying microbial composition (shotgun versus targeted sequencing) for various samples types.

Orientador: Ângela Fátima Soligo Tese (doutorado) - Universidade Estadual de Campinas, Faculdade de Educação Resumo: No discurso socialmente corrente difunde-se a urgência da implementação de alterações na prática pedagógica de modo que a escola possa dar respostas às demandas impostas pelo processo de globalização e internacionalização da economia, seja capaz de criar condições que favoreçam a participação social e o exercício da cidadania, além de garantir o desenvolvimento de aprendizagens significativas. Nesta perspectiva professores e professoras são vistos como catalisadores das principais mudanças a serem efetivadas, atribuindo-se grande relevância ao seu processo de formação, seja inicial ou continuada. O presente trabalho busca contribuir com as reflexões sobre a formação de professores e professoras a partir da teoria das representações sociais, tendo como principal norteador as contribuições de Serge Moscovici (1978; 2001; 2004). A pesquisa realizada teve como objeto de estudo as representações dos professores sobre conhecimento, uma vez que este tem assumido um papel cada vez mais importante tanto no contexto social, quanto escolar, constituindo-se em um fenômeno social que mobiliza os sujeitos a pensarem sobre sua realidade e sobre si. A investigação se deu a partir da estruturação de uma situação de conversação entre professores e professoras em formação inicial e profissionais que atuavam em uma escola pública, por meio da elaboração de cartas. No processo de análise das narrativas docentes foi possível perceber que ao falar sobre conhecimento professores e professoras o fazem a partir do cotidiano escolar, sendo identificadas duas dimensões nas quais se inserem suas representações, a primeira envolve os conhecimentos que eles julgam relevantes para a sua atuação profissional; a segunda, o papel que assumem no processo de apropriação de conhecimentos pelos alunos e alunas que estão sob sua responsabilidade. Defende-se a tese de que a integração à vida cotidiana de conceitos ou termos que fazem parte do discurso pedagógico, transformando-os em temas de conversação, possibilita a desconstrução de representações sociais, o que abre possibilidades para a promoção de mudanças na prática docente. Abstract: In the socially current speach has been spreaded the urgency of the implementation of amendments in the pedagogical practice so that the school can provide answers to the demands imposed by the process of globalization and internationalization of the economy, is able to create conditions that promote social participation and the exercise of citizenship, in addition ensure the development of significant learning. In this perspective the teachers are seen as catalysts of the main changes to be efectives, great importance is attributed to their training process, whether initial or continuing. This work seeks to contribute with the reflections on the training of teachers from the social representations theory, having as the main reference the contributions of Serge Moscovici (1978; 2001; 2004). The research had as its object of study the representations of teachers about knowledge, since this has assumed an increasingly important both as much as in the social context, as in the school, becoming a social phenomenon which mobilizes the subjects to think about their reality and to itself. The research has been estructured from the creation of a situation of conversation between teachers in initial training and professionals who acted in a public school, through letters. In the process of analysis faculty narratives was possible to realize that when the teachers talk about knowledge, they do it from the school everyday life, being possible to identify two dimensions, the first involves the knowledge that they consider relevant to their professional performance; the second, the role in the process of appropriation of knowledge by the students who are under their responsibility. The thesis defends that the integration in the daily life of concepts or terms which are part of the pedagogical discourse, transforming it into topics of conversation, allows the deconstruction of social representations, creating possibilities for the promotion of changes in teaching practice. 46. Evaluation of music therapy to reduce stress in hospitalized cats.

- Abstract
Objectives: This study aimed to evaluate the use of two different types of music – cat-specific music and classical music – compared with no music, to reduce stress in cats during hospitalization. Methods: Thirty-five hospitalized cats were randomly divided into three groups and each group received a different stimulus – cat-specific music, classical music or no music (control) – throughout their hospitalization. Respiratory rate, salivary cortisol and social interaction were documented. A blinded researcher performed the Cat Stress Score (CSS) during the video analysis of recordings at five specific times over 31 h of hospitalization. Results: There was no difference in the mean CSS between cats listening to cat-specific music, classical music and control throughout the five evaluations. Cat-specific music had a higher percentage of positive social interactions than the other groups on the first evaluation (P <0.05). The average respiratory rate was significantly lower in the classical music group vs control on the fourth evaluation (P <0.05). Although statistically insignificant, the average respiratory rate decreased only in the classical music group during the five evaluations. Cortisol quantification did not seem to follow the CSS results. However, owing to the low and unrepresentative number of samples, it was not possible to perform statistical analysis on these results or a group sample comparison. Conclusions and relevance: Both cat-specific music and classical music seem to have some benefit to hospitalized cats. The salivary cortisol analysis was not adequate nor useful to measure stress in hospitalized cats in our study. [ABSTRACT FROM AUTHOR]

Changes in landscape and ecosystem services. Agricultural abandonment and variation in soil organic carbon Soil organic carbon (SOC) contributes to improving the physical, chemical and biological properties of the soil and supply several goods and services of production, regulation and support, which interfere on human health and well-being. This study aim to analyse plant succession processes following the agricultural abandonment and the respective effect on the organic carbon variation in the surface soil layer, in the southern part of Beira Transmontana. The results show that the process of agricultural abandonment promotes an increase in the COS content, which is irrelevant when the vegetation cover is incipient, dominated by herbaceous communities, and more than doubles when there is the establishment of shrub or oak dominated communities (+ 139% and 108%, respectively, compared to soils with arable crops). Despite this increase of carbon in the soil, these communities only contain around half of the SOC obtained for oak woodlands, representative of the native vegetation. Thus, proper soil management, following abandonment, can be a good option to improve its quality and a good strategic measure for carbon sequestration., O carbono orgânico do solo (COS) contribui para melhorar as propriedades físicas, químicas e biológicas do solo e para o fornecimento de bens e serviços vários, de aprovisionamento, regulação ou suporte, que interferem na saúde e bem-estar humano. Com o presente estudo pretende-se analisar os processos sucessão vegetal, na sequência do abandono agrícola, e a respetiva influência na variação do carbono orgânico na camada edáfica superficial, na parte meridional da Beira Transmontana. Os resultados mostram que o processo de abandono agrícola se traduz num aumento do teor de COS, pouco relevante quando coberto vegetal é incipiente, dominado por comunidades herbáceas, mas mais do que duplica quando se verifica a instalação de comunidades arbustivas ou de carvalhal ainda em fase de recuperação (+ 139% e 108%, respetivamente, face aos solos com culturas arvenses de sequeiro). Apesar deste acréscimo de carbono no solo, estas comunidades só contêm aproximadamente metade do valor apurado para o carvalhal representativo da vegetação autóctone. Assim, a gestão adequada do solo, na sequência do abandono, pode constituir, uma boa opção para melhorar a sua qualidade e estratégia para o sequestro do carbono.
