Abstract The Terminal Fusarium Clade (TFC) is a group in the Nectriaceae family with agricultural and clinical relevance. In recent years, various phylogenies have been presented in the literature, showing disagreement in the topologies, but only a few studies have conducted analyses on the divergence time scale of the group. Therefore, the evolutionary history of this group is still being determined. This study aimed to understand the evolutionary history of the TFC from a phylogenomic perspective. To achieve this objective, we performed a phylogenomic analysis using the available genomes in GenBank and ran eight different pipelines. We presented a new robust topology of the TFC that differs at some nodes from previous studies. These new relationships allowed us to formulate new hypotheses about the evolutionary history of the TFC. We also inferred new divergence time estimates, which differ from those of previous studies due to topology discordances and taxon sampling. The results suggested an important diversification process in the Neogene period, likely associated with the diversification and predominance of terrestrial ecosystems by angiosperms. In conclusion, we presented a robust time-scale phylogeny that allowed us to formulate new hypotheses regarding the evolutionary history of the TFC.

Recent publications have argued that there are potentially serious consequences for researchers in recognising distinct genera in the terminal fusarioid clade of the family Nectriaceae. Thus, an alternate hypothesis, namely a very broad concept of the genus Fusarium was proposed. In doing so, however, a significant body of data that supports distinct genera in Nectriaceae based on morphology, biology, and phylogeny is disregarded. A DNA phylogeny based on 19 orthologous protein-coding genes was presented to support a very broad concept of Fusarium at the F1 node in Nectriaceae. Here, we demonstrate that re-analyses of this dataset show that all 19 genes support the F3 node that represents Fusarium sensu stricto as defined by F. sambucinum (sexual morph synonym Gibberella pulicaris). The backbone of the phylogeny is resolved by the concatenated alignment, but only six of the 19 genes fully support the F1 node, representing the broad circumscription of Fusarium. Furthermore, a re-analysis of the concatenated dataset revealed alternate topologies in different phylogenetic algorithms, highlighting the deep divergence and unresolved placement of various Nectriaceae lineages proposed as members of Fusarium. Species of Fusarium s. str. are characterised by Gibberella sexual morphs, asexual morphs with thin- or thick-walled macroconidia that have variously shaped apical and basal cells, and trichothecene mycotoxin production, which separates them from other fusarioid genera. Here we show that the Wollenweber concept of Fusarium presently accounts for 20 segregate genera with clear-cut synapomorphic traits, and that fusarioid macroconidia represent a character that has been gained or lost multiple times throughout Nectriaceae. Thus, the very broad circumscription of Fusarium is blurry and without apparent synapomorphies, and does not include all genera with fusarium-like macroconidia, which are spread throughout Nectriaceae (e.g., Cosmosporella, Macroconia, Microcera). In this study four new genera are introduced, along with 18 new species and 16 new combinations. These names convey information about relationships, morphology, and ecological preference that would otherwise be lost in a broader definition of Fusarium. To assist users to correctly identify fusarioid genera and species, we introduce a new online identification database, Fusarioid-ID, accessible at www.fusarium.org. The database comprises partial sequences from multiple genes commonly used to identify fusarioid taxa (act1, CaM, his3, rpb1, rpb2, tef1, tub2, ITS, and LSU). In this paper, we also present a nomenclator of names that have been introduced in Fusarium up to January 2021 as well as their current status, types, and diagnostic DNA barcode data. In this study, researchers from 46 countries, representing taxonomists, plant pathologists, medical mycologists, quarantine officials, regulatory agencies, and students, strongly support the application and use of a more precisely delimited Fusarium (= Gibberella) concept to accommodate taxa from the robust monophyletic node F3 on the basis of a well-defined and unique combination of morphological and biochemical features. This F3 node includes, among others, species of the F. fujikuroi, F. incarnatum-equiseti, F. oxysporum, and F. sambucinum species complexes, but not species of Bisifusarium [F. dimerum species complex (SC)], Cyanonectria (F. buxicola SC), Geejayessia (F. staphyleae SC), Neocosmospora (F. solani SC) or Rectifusarium (F. ventricosum SC). The present study represents the first step to generating a new online monograph of Fusarium and allied fusarioid genera (www.fusarium.org).

The cliff rose (Armeria maritima), like other halophytes, has a phenolics-based antioxidant system that allows it to grow in saline habitats. Provided that antioxidant properties are usually accompanied by antimicrobial activity, in this study we investigated the phytochemicals present in a hydromethanolic extract of A. maritima flowers and explored its antifungal potential. The main phytocompounds, identified by gas chromatography–mass spectrometry, were: hexadecanoic acid, octadecanoic acid, 9-octadecenoic acid, 3-(3,4-dihydroxy-phenyl)-acrylic acid ethyl ester, and benzeneacetaldehyde. The antifungal activity of the extract and its main constituents—alone and in combination with chitosan oligomers—was tested against six pathogenic taxa associated with soil-borne diseases of plant hosts in the family Cucurbitaceae: Fusarium equiseti, F. oxysporum f. sp. niveum, Macrophomina phaseolina, Neocosmospora falciformis, N. keratoplastica, and Sclerotinia sclerotiorum. In in vitro tests, EC90 effective concentrations in the 166−865 μg·mL−1 range were obtained for the chitosan oligomers–A. maritima extract conjugate complexes, lower than those obtained for fosetyl-Al and azoxystrobin synthetic fungicides tested for comparison purposes, and even outperforming mancozeb against F. equiseti. In ex situ tests against S. sclerotiorum conducted on artificially inoculated cucumber slices, full protection was achieved at a dose of 250 μg·mL−1. Thus, the reported results support the valorization of A. maritima as a source of biorationals for Cucurbitaceae pathogens protection, suitable for both organic and conventional agriculture. Plant-parasitic nematodes (PPNs) are significant pests that result in considerable economic losses in global crop production. Due to the high toxicity of chemical nematicides, there is a need to develop new strategies for nematode control. In this context, nematophagous fungi may offer a viable option for biological control. Two fungal strains (GUCC2212 and GUCC2232) were isolated from cysts of Globodera sp., identified as Neocosmospora stercicola. The fungal filtrates of the strains were evaluated for their nematicidal activity against three species of PPNs: Aphelenchoides besseyi, Bursaphelenchus xylophilus and Ditylenchus destructor. The fermentation filtrates of two strains exhibited substantial toxicity towards the evaluated nematodes, with mortality rates reaching up to 100% within 72 h. Concurrently, N. stercicola also demonstrated predatory and parasitic behavior. The eggs of Globodera sp. were parasitized by the two strains. N. stercicola represents a newly recorded species in China and a novel nematophagous species. In conclusion, the two strains of N. stercicola show promise as biocontrol agents for PPNs management. For decades there have been anecdotal claims of synergistic interactions between plant-parasitic nematodes and soil-borne fungi causing decline of productivity of passion fruit (Passiflora edulis) orchards. An empirical confirmation of these disease complexes would impact disease management and plant breeding for resistance. To test those claims, we subjected passion fruit plants to single or concomitant parasitism by Meloidogyne javanica or M. incognita and Fusarium nirenbergiae or Neocosmospora sp. under controlled conditions. Non-inoculated plants served as control for the assays. The severity of shoot symptoms and variables related to plant growth, the extent of fungal lesions, and nematode reproduction were assessed to characterize the interactions. The shoot symptoms and effect on plant growth induced by the pathogens varied, but no synergy between the pathogens was observed. Moreover, the volume of tissue lesioned by the fungi was not affected by co-parasitism of the nematodes. Conversely, plant resistance to the nematodes was not affected by co-parasitism of the fungi. The interactions M. incognita-F. nirenbergiae, M. incognita-Neocosmospora sp., M. javanica-F. nirenbergiae, and M. javanica-Neocosmospora sp. were not synergistic as previously claimed, but instead neutral.

Sea turtles are presently considered severely endangered species that are historically threatened by many environmental factors. Recently, additional threats to sea turtles from two pathogenic species of fungi in the Fusarium solani species complex (F. falciforme and F. keratoplasticum) have been identified. These species infect marine turtle eggs, causing sea turtle egg fusariosis, and kill their embryos, with recent reports of hatch-failure in seven globally distributed species of endangered sea turtles (Caretta caretta, Chelonia mydas, Dermochelys coriaceae, Eretmochelys imbricata, Lepidochelys olivacea, Lepidochelys kempi and Natator depressus). Mycelia and spores of pathogenic species of Fusarium are produced in disturbed terrestrial soils and are transported to the ocean in coastal run off. We propose that these fungi grow on floating particles of plant tissues (leaves and wood), animal tissues, silt and plastics, which are carried by wind and currents and the turtles themselves to the beaches where the turtles lay their eggs.

Bacterial fruit blotch (BFB) is a bacterial disease that devastates Cucurbitaceae crops worldwide, causing significant economic losses. Currently, there is no means to treat or control the disease. This study focused on exploring the antibacterial properties of endophytic fungi against Acidovorax citrulli (Aac), the causative agent of BFB. Based on disc diffusion, time kill and MIC microdilution broth assays, four endophytes showed promise in controlling Aac. Nonetheless, only one strain, Neocosmospora sp. MFLUCC 17-0253, reduced the severity of disease on watermelon and melon seedlings up to 80%. Structure analysis revealed production of several compounds by the fungus. Three of these secondary metabolites, including mixture of 2-methoxy-6-methyl-7-acetonyl-8-hydroxy-1,4-maphthalenedione and 5,8-dihydroxy-7-acetonyl-1,4-naphthalenedione, anhydrojavanicin, and fusarnaphthoquinones B exhibited antagonistic activity against Aac. The chemical profile data in planta experiment analyzed by LC-Q/TOF-MS suggested successful colonization of endophytic fungi in their host plant and different metabolic profiles between treated and untreated seedling. Biofilm assay also demonstrated that secondary metabolites of Neocosmospora sp. MFLUCC 17-0253 significantly inhibited biofilm development of Aac. To the best of our knowledge, secondary metabolites that provide significant growth inhibition of Aac are reported for the first time. Thus, Neocosmospora sp. In the fungal pathogen Aspergillus fumigatus, resistance to azole antifungals is often linked to mutations in CYP51A, a gene that encodes the azole antifungal drug target lanosterol 14α-demethylase. The aim of this study was to investigate whether similar changes could be associated with azole resistance in a Malaysian Fusarium solani species complex (FSSC) isolate collection. Most (11 of 15) clinical FSSC isolates were Neocosmospora keratoplastica and the majority (6 of 10) of environmental isolates were Neocosmospora suttoniana strains. All 25 FSSC isolates had high minimum inhibitory concentrations (MICs) for itraconazole and posaconazole, low MICs for amphotericin B, and various (1 to >32 mg/l) voriconazole susceptibilities. There was a tight association between a 23 bp CYP51A promoter deletion and high (>32 mg/l) voriconazole MICs; of 19 FSSC strains sequenced, nine isolates had voriconazole MICs > 32 mg/l, and they all contained the 23 bp CYP51A promoter deletion, although it was absent in the ten remaining isolates with low (≤12 mg/l) voriconazole MICs. Surprisingly, this association between voriconazole resistance and the 23 bp CYP51A promoter deletion held true across species boundaries. It was randomly distributed within and across species boundaries and both types of FSSC isolates were found among environmental and clinical isolates. Three randomly selected N. keratoplastica isolates with low (≤8 mg/l) voriconazole MICs had significantly lower (1.3–7.5 times) CYP51A mRNA expression levels than three randomly selected N. keratoplastica isolates with high (>32 mg/l) voriconazole MICs. CYP51A expression levels, however, were equally strongly induced (~6,500-fold) by voriconazole in two representative strains reaching levels, after 80 min of induction, that were comparable to those of CYP51B. Our results suggest that FSSC isolates with high voriconazole MICs have a 23 bp CYP51A promoter deletion that provides a potentially useful marker for voriconazole resistance in FSSC isolates. Early detection of possible voriconazole resistance is critical for choosing the correct treatment option for patients with invasive fusariosis.

در تابستان سال 1396، طی یک بررسی به منظور شناسایی گونه‌های قارچی از خاک‌های بکر استان کرمانشاه، نمونه‌های خاک از عمق 20-0 سانتی‌متری جمع‌آوری و از خاک جداسازی و سپس با روش تهیه رقت با استفاده از محیط کشت سیب زمینی دکستروز آگار صورت گرفت. در این بررسی، یک جدایه از خاک بکر (دامنه کوه) سومار واقع در منطقه قصر شیرین به دست آمد. میسلیوم در این جدایه به رنگ سفید مایل به کرم بود (شکل 1A). پریتسیوم نارنجی رنگ تا قرمز تیره، کروی تا گلابی شکل با گردن کوتاه، روزنه‌دار، دارای پریفیز و به ابعاد 240 تا 350 میکرومتر بود (شکل 1B). آسک‌ها سیلندری، دارای هشت آسکوسپور، دیواره نازک، غیرآمیلوییدی، با انتهای صاف و پدیسل کوتاه و به ابعاد 12–10 × 100–80 میکرومتر بود (شکل 1C). آسکوسپورها یک جداره، دارای دیواره مضرس، کروی تا بیضوی، شفاف و در زمان بلوغ قهوه‌ای و گاهی اوقات دارای قطره روغن و به ابعاد 63/9 × 23/11 میکرومتر بود (شکل 1D).براساس خصوصیات مورفولوژیکی، این جدایه Neocosmospora شناسایی شد.برای شناسایی گونه، اسخراج دی.ان.ای. به روش سی تب صورت گرفت و نواحی نسخه‌برداری شده داخلی دی.ان.ای. ریبوزومی هسته (ITS) و قسمتی از فاکتور طویل‌کننده ترجمه (TEF1) با واکنش زنجیره‌ای پلیمراز و جفت آغازگرهای ITS1/ITS4 و EF1F/EF1R تکثیر شدند. آنالیز فیلوژتیکی به روش بیشینه درست‌نمایی (Maximum likelihood) و با استفاده از نسخه 5 نرم‌افزار مگا صورت گرفت. در توالی‌یابی ناحیه نسخه‌برداری شده داخلی دی.ان.ای. ریبوزومی هسته و قسمتی از فاکتور طویل‌کننده ترجمه برای این جدایهتعداد 550 و 650 جفت باز به ترتیب به دست آمد که ناحیه نسخه‌برداری شده داخلی دی.ان.ای. ریبوزومی هسته 99 درصد با گونه Neocosmosporavasinfecta با شماره دسترسی KM231804 از کشور هند و فاکتور طویل‌کننده ترجمه 100 درصد با گونه‌ای به همین نام با شماره دسترسی JX997934 از فرانسه شباهت داشت. این جدایه با شماره دسترسی MG811579 برای ناحیه نسخه‌برداری شده داخلی دی.ان.ای. ریبوزومی هسته و با شماره دسترسی MH976665 برای فاکتور طویل‌کننده ترجمه در پایگاه اطلاعاتی بانک ژن ثبت گردید.آنالیز فیلوژنتیکی ناحیه نسخه‌برداری شده داخلی دی.ان.ای. ریبوزومی هسته با بیشینه درست‌نمایی صحت شناسایی گونه را تایید کرد و نمونه ما با ضریب اطمینان بالا همراه با نمونه‌های معتبر از کشورهای دیگر در یک گروه قرار گرفت (شکل 2). جدایه قارچ فوق در مجموعه قارچ‌های زنده وزارت جهاد کشاورزی در مؤسسه تحقیقات گیاه‌پزشکی کشور (تهران) نگهداری می‌شود. این گونه برای فلور قارچی ایران برای نخستین بار از ایران گزارش می‌شود. نمونه بررسی شده: استان کرمانشاه، قصر شیرین، سومار، 45° 95΄ E, 33° 96΄ N، 5/4/1396، جداسازی شده از خاک بکر، شیما سعیدی (IRAN 3320 C).

Jacobs-Venter et al. ([8]) separated I F. polyphialidicum i strains into three species, namely I F. concolor, F. babinda i and I F. austroafricanum i , and confirmed that I F. polyphialidicum i is synonymous with I F. concolor. Together with other species previously described from the KNP ( I F. fredkrugeri, F. convalutum, F. transvaalense i and I F. concolor i as I F. polyphialidicum i ), there are thus at least nine I Fusarium i species present in the KNP. I Fusarium nygamai, F. proliferatum, F. oxysporum sensu lato, F. chlamydosporum, N. vasinfectum i and I F. concolor i are species that have a world-wide occurrence, including South Africa (Jacobs et al. [8]; Leslie & Summerell [13]). Previously, I F. nygamai, F. polyphialidicum, F. fredkrugeri, F. convalutum i and I F. transvaalense i were described from the KNP, while possible new species have also been characterised in this study. In this study, endophytic fungi were isolated from Euphorbia larica Boiss. and their antagonistic potential against phytopathogenic fungi viz. Fusarium sp., Rosellinia sanctae-cruciana and Cladosporium sp. was evaluated using an in vitro confrontation assay. A total of four endophytic fungi were isolated from the roots of E. larica. Among them, two fungal isolates (SQUCC-F1-1 and SQUCCF6-2) showed inhibitory activity against Fusarium sp. None of the fungal endophytes restricted the growth of Rosellinia sanctae-cruciana and Cladosporium sp. On the basis of multi-gene phylogenetic analysis, these two endophytic fungi were identified as Neocosmospora sp. (SQUCC-F1-1) and Alternaria alternata (SQUCC-F6-2). Scanning electron microscopic observation of hyphae of the phytopathogenic fungi from the dual culture plate revealed morphological abnormalities such as loss of turgidity, shrinkage and disintegration of the hyphae. Analysis of the metabolites produced by these endophytic fungi by GC-MS revealed the existence of various fatty acids, fatty acid methyl esters, hydrocarbons and alkanes, which have been previously demonstrated to have antimicrobial activity. During surveys conducted in 2010-2014, several disease symptoms were observed on eight ornamental plant species in different nurseries located in Sicily (Southern Italy). Two Neocosmospora and 31 Fusarium isolates were recovered from symptomatic plants. Fungus identification was based on morphological characteristics and phylogenetic analyses of fragments of the intergenic spacer (IGS), internal transcribed spacer (ITS) and large subunit (LSU) regions of the rDNA; partial fragments of the beta-tubulin (TUB), RNA polymerase largest subunit (RPB1), RNA polymerase second largest subunit (RPB2) and translation elongation factor 1-alpha (EF-1a) genes. The Fusarium species recovered from ornamental plants were F. agapanthi (from Agapanthus africanus), F. anthophilum (Dasylirion longissimum), F. fujikuroi (Trachycarpus princeps), F. oxysporum (Bougainvillea glabra, Cordyline australis 'Purpurea', Dasylirion longissimum, Eremophila laanii and Philoteca myoporoides) and F. proliferatum (T. princeps), while N. solani was isolated from crown and root rot of Ficus carica. The pathogenicity of representative isolates collected from each host was tested on seedlings or cuttings grown in a growth chamber. All the Fusarium and Neocosmospora isolates tested were pathogenic and reproduced symptoms identical to those observed in the field, except for F. fujikuroi on T. princeps and F. oxysporum on D. longissimum that were non-pathogenic. The genus Neocosmospora encompasses highly prevalent and aggressive human and animal fungal pathogens. Here we assign formal descriptions and Latin binomials to some of the most clinically relevant phylogenetic species of the genus. Three new species, named Neocosmospora catenata, N. gamsii and N. suttoniana (previously assigned to the informal names 'Fusarium' solani species complex (FSSC) lineages, FSSC 43, FSSC 7 and FSSC 20, respectively) are described on the basis of multilocus phylogenetic analyses (using EF-1α, ITS, LSU and RPB2 loci) and morphological characters. Lineage FSSC 9 is conspecific with the ex-type strain of Cylindrocarpon tonkinense, thus the new combination Neocosmospora tonkinensis is proposed. Chinese white pine, Pinus armandii , is a source of high-quality timber and an afforestation tree in China, which plays an important ecological and social role in water and soil conservation. Recently, a new canker disease has been reported in Longnan City, Gansu Province, where P. armandii is mainly distributed. In this study, the causal agent was isolated from diseased samples and identified as a fungal pathogen, Neocosmospora silvicola , based on morphological characteristics and molecular analyses (internal transcribed spacer, large subunit, RNA polymerase II, and translation elongation factor-1α). Pathogenicity tests on P. armandii revealed that N. silvicola isolates caused a 60% average mortality rate in artificially inoculated 2-year-old seedlings. The pathogenicity of these isolates was also observed on the branches of 10-year-old P. armandii trees with a 100% mortality rate. These results agree with the isolation of N. silvicola from diseased plants, suggesting the possible role of this fungus in the decline of P. armandii plants. Mycelial growth of N. silvicola was fastest on potato dextrose agar medium, and growth occurred at pH values ranging from 4.0 to 11.0 with temperatures between 5 and 40°C. The fungus also grew rapidly in complete darkness compared with other light conditions. Of the eight carbon and seven nitrogen sources tested, starch and sodium nitrate, respectively, were highly efficient in supporting the mycelial growth of N. silvicola . The ability of N. silvicola to grow at low temperatures (5°C) may explain its occurrence in the Longnan area of Gansu Province. The productivity of arid legumes, such as Clusterbean (Cyamopsis tetragonoloba), Cowpea (Vigna unguiculata), Moth bean (Vigna aconitifolia) and Horse gram (Macrotyloma uniflorum), may remain stagnant over decades because of their high susceptibility to root diseases. Besides, there is a limitation on the information about molecular diagnosis and intraspecific genetic variability of root pathogens in arid legumes. To contribute in this field, we assessed a total of 52 isolates from 88 root samples that were found infected with fungal pathogens in Jodhpur, Jaipur and Bikaner Districts of Rajasthan. Diseased roots samples were analyzed following standard microbiological methods for fungus extraction and purification, and for genetic studies. Irrespective of the geographical location from where the diseased samples were collected, all pathogen isolates were clustered in RAPD dendrograms as per their respective genera. Phylogram, based on multiple sequence alignment, revealed that different genera (i.e. Fusarium, Neocosmospora and Syncephalastrum), separated from each other, and species within the same genera, clustered together with their reference sequences with apreciable bootstrap values. Out of 20 representative isolates representing each cluster and all outgroups sequenced, eight were molecularly identified as Neocosmospora vasinfecta, five as Fusarium solani, two as Neocosmospora striata, two as Fusarium acutatum, one as Syncephalastrum monosporum, one as Fusarium oxysporum and one as Fusarium species. The root pathogens of the arid legumes were found neither restricted to a geographical location nor were host specific in nature. Fusarium solani wilt in cowpea and seedling rot in moth bean, F. oxysporum wilt in moth bean, F. acutatum damping off in cowpea and Clusterbean, Fusarium sp. seedling rot in Clusterbean, Neocosmospora striata root rot in cowpea and wilt in Clusterbean and Syncephalastrum monosporum root rot in Clusterbean were molecularly identified as new fungal records as pathogens causing root diseases in arid legumes. A large collection of strains belonging to the Fusarium solani species complex (FSSC) was isolated from soil and perithecia in primap/ forests in Sri Lanka (from fallen tree bark) and tropical Australia (Queensland, from fallen tree fruits and nuts). Portions of the translation elongation factor 1-alpha (tef1) gene, the nuclear large subunit (NLSU) and internal transcribed spacer regions (ITS) of the nuclear ribosomal RNA genes were sequenced in 52 isolates from soil and perithecia. The FSSC was divided previously into three clades with some biogeographic structure, termed Clades 1, 2 and 3. MI Sri Lankan and Australian soil isolates were found to be members of Clade 3, most grouping with the cosmopolitan soil-associated species F. falciforme. All but two Sri Lankan perithecial isolates were associated with a set of five divergent phylogenetic lineages that were associated with Clade 2. Australian perithecial isolates resided in a subclade of Clade 3 where most of the previously defined mating populations of the FSSC reside. Isolates from perithecia and those cultured from soil were always members of different species lineages, even when derived from proximal locations. The previous biogeographic assignment of Clade 2 to South America is now expanded to the worldwide tropics. Sri Lanka appears to be an important center of diversity for the FSSC. Nectria haematococca is epitypified with a collection from the type locality in Sri Lanka; its anamorph is described as a new species, Fusarium haematococcum. Neocosmospora E.F. Smith is adopted as the correct genus for Nectria haematococca. These new species are described: F. kurunegalense/Neo, kurunegalensis, F. rectiphorus/ Neo. rectiphora/, F. mahasenii/Neo. mahasenii/, F. kelerajum/Neo, keleraja. [ABSTRACT FROM AUTHOR]

We isolated a soybean saponin hydrolase from Neocosmospora vasinfecta var. vasinfecta PF1225, a filamentous fungus that can degrade soybean saponin and generate soyasapogenol B. This enzyme was found to be a monomer with a molecular mass of about 77 kDa and a glycoprotein. Nucleotide sequence analysis of the corresponding gene (sdn1) indicated that this enzyme consisted of 612 amino acids and had a molecular mass of 65,724 Da, in close agreement with that of the apoenzyme after the removal of carbohydrates. The sdn1 gene was successfully expressed in Trichoderma viride under the control of the cellobiohydrolase I gene promoter. The molecular mass of the recombinant enzyme, about 69 kDa, was smaller than that of the native enzyme due to fewer carbohydrate modifications. Examination of the degradation products obtained by treatment of soyasaponin I with the recombinant enzyme showed that the enzyme hydrolyzed soyasaponin I to soyasapogenol B and triose [α-L-rhamnopyranosyl (1→2)-β-D-galactopyranosyl (1→2)-D-glucuronopyranoside]. Also, when soyasaponin II and soyasaponin V, which are different from soyasaponin I only in constituent saccharides, were treated with the enzyme, the ratio of the reaction velocities for soyasaponin I, soyasaponin II, and soyasaponin V was 2,680:886:1. These results indicate that this enzyme recognizes the fine structure of the carbohydrate moiety of soyasaponin in its catalytic reaction. The amino acid sequence of this enzyme predicted from the DNA sequence shows no clear homology with those of any of the enzymes involved in the hydrolysis of carbohydrates. [ABSTRACT FROM AUTHOR]

Seven Fusarium species complexes are treated, namely F. aywerte species complex (FASC) (two species), F. buharicum species complex (FBSC) (five species), F. burgessii species complex (FBURSC) (three species), F. camptoceras species complex (FCAMSC) (three species), F. chlamydosporum species complex (FCSC) (eight species), F. citricola species complex (FCCSC) (five species) and the F. concolor species complex (FCOSC) (four species). New species include Fusicolla elongata from soil (Zimbabwe), and Neocosmospora geoasparagicola from soil associated with Asparagus officinalis (Netherlands). New combinations include Neocosmospora akasia, N. awan, N. drepaniformis, N. duplosperma, N. geoasparagicola, N. mekan, N. papillata, N. variasi and N. warna. Newly validated taxa include Longinectria gen. nov. , L. lagenoides , L. verticilliforme , Fusicolla gigas and Fusicolla guangxiensis . Furthermore, Fusarium rosicola is reduced to synonymy under N. brevis. Finally, the genome assemblies of Fusarium secorum (CBS 175.32) , Microcera coccophila (CBS 310.34), Rectifusarium robinianum (CBS 430.91), Rugonectria rugulosa (CBS 126565), and Thelonectria blattea (CBS 952.68) are also announced here. A new chaetochiversin analog, designated chaetochiversin C (1), was discovered from a cultured broth of fungal strain FKI-7792 by physicochemical screening. This strain was identified as a member of genus Neocosmospora based on morphology and DNA barcoding. The partially relative configuration of 1 was determined by 13 C-NMR chemical shifts of the acetonide analog of 1. The absolute configuration was determined using an advanced Mosher's method. Compound 1 was assessed for anti-tumor, anti-microbial, and anti-malarial activities, and its ability to scavenge or quench reactive oxygen species (ROS), such as superoxide anion radicals, hydroxy radicals and singlet oxygen ( 1 O 2 ). Compound 1 showed a quenching effect on 1 O 2 .
