In this book, epigenetics is understood as the science of molecules and mechanisms involved in controlling gene activity without altering their sequence, and which thus contribute to cellular functions. It enables us to decipher the development and differentiation processes of cells in a complex organism, which perform different functions despite sharing the same nuclear genetic information. This science also supports cellular memory and responses to the environment. The book introduces the history and evolution of the concept of "epigenetics" and explains the molecular mechanisms that govern the development of mammals, fish or plants. Knowledge of the diversity of these mechanisms, and our understanding of their importance in biology, is probably just getting started. Some processes, such as X chromosome inactivation, are specific to mammals, while others, such as parental imprinting, are surprisingly common to both mammals and plants with sexual reproduction. Their importance in responses to the biotic or abiotic environment is highlighted to show possible applications in medicine or agronomy. This book is intended for students, teachers in higher education and researchers in the field of biology.

- Abstract
In 2021, both social and economic activities began to return towards the levels preceding the COVID-19 pandemic for some parts of the globe, with others still experiencing restrictions. Meanwhile, the climate has continued to respond to the ongoing increase in greenhouse gases and resulting warming. La Niña, a phenomenon which tends to depress global temperatures while changing rainfall patterns in many regions, prevailed for all but two months of the year. Despite this, 2021 was one of the six-warmest years on record as measured by global mean surface temperature with an anomaly of between +0.21° and +0.28°C above the 1991–2020 climatology.

- Abstract
In 2021, both social and economic activities began to return towards the levels preceding the COVID-19 pandemic for some parts of the globe, with others still experiencing restrictions. Meanwhile, the climate has continued to respond to the ongoing increase in greenhouse gases and resulting warming. La Niña, a phenomenon which tends to depress global temperatures while changing rainfall patterns in many regions, prevailed for all but two months of the year. Despite this, 2021 was one of the six-warmest years on record as measured by global mean surface temperature with an anomaly of between +0.21° and +0.28°C above the 1991–2020 climatology. Lake surface temperatures were their highest on record during 2021. The number of warm days over land also reached a new record high. Exceptional heat waves struck the Pacific Coast of North America, leading to a new Canadian maximum temperature of 49.6°C at Lytton, British Columbia, on 29 June, breaking the previous national record by over 4°C. In Death Valley, California, the peak temperature reached 54.4°C on 9 July, equaling the temperature measured in 2020, and the highest temperature recorded anywhere on the globe since at least the 1930s. Over the Mediterranean, a provisional new European record of 48.8°C was set in Sicily on 11 August. In the atmosphere, the annual mean tropospheric temperature was among the 10 highest on record, while the stratosphere continued to cool. While La Niña was present except for June and July, likely influencing Australia’s coolest year since 2012 and wettest since 2016, other modes of variability played important roles. A negative Indian Ocean dipole event became established during July, associated with a warmer east and cooler west Indian Ocean. Northern Hemisphere winters were affected by a negative phase of the North Atlantic Oscillation at both the beginning and end of 2021. The process development by IFP Energies nouvelles (IFPEN)/ENI/Axens of a Fischer-Tropsch process is described. This development is based on upstream process studies to choose the process scheme, reactor technology and operating conditions, and downstream to summarize all development work in a process guide. A large amount of work was devoted to the catalyst performances on one hand and the scale-up of the slurry bubble reactor with dedicated complementary tools on the other hand. Finally, an original approach was implemented to validate both the process and catalyst on an industrial scale by combining a 20 bpd unit in ENI’s Sannazzaro refinery, with cold mock-ups equivalent to 20 and 1 000 bpd at IFPEN and a special “Large Validation Tool” (LVT) which reproduces the combined effect of chemical reaction condition stress and mechanical stress equivalent to a 15 000 bpd industrial unit. Dedicated analytical techniques and a dedicated model were developed to simulate the whole process (reactor and separation train), integrating a high level of complexity and phenomena coupling to scale-up the process in a robust reliable base on an industrial scale.

The electron paramagnetic resonance (EPR) technique was used to study the behavior of the asphaltenes from the Merey crude oil (ASCM), its vacuum residue (ARVM), its fractions, and trapped compounds (A1-ASCM, A2-ASCM, TC-ASCM, A1-ARVM, A2-ARVM, and TC-ARVM). Asphaltenes were precipitated from Merey heavy crude oil and its vacuum residue. The fractionation of our petroleum asphaltenes was carried out using the para-nitrophenol (PNP) method. Two fractions named A1 and A2 and trapped compounds were obtained. The temperature dependence of the EPR spectra was used to obtain a set of temperatures related to free radical (FR) generation and recombination. From the analysis of the EPR spectrum of the most intense signal of the vanadium, we obtain the temperature ranges of the anisotropic-to-isotropic domains, we found a nonlinear behavior of the B parameter in the temperature range studied, and we propose a functional relationship for this parameter as a function of the temperature. After autologous hematopoetic cell transplantation, (HCT in 1st complete remission (CR1), patients with acute myeloid leukemia (AML) may relapse and undergo allogeneic HCT in CR2. The aim of this study was to analyze outcome of allogeneic HCT performed in CR2 comparing patients with prior consolidation by autologous HCT vs. patients with chemotherapy consolidation. Included were 2619 adults, with allogeneic HCT in CR2, in 2000-2017 with (n=417) or without (n=2202) prior autologous HCT. Patient groups were not entirely comparable; patients with prior autologous HCT were younger, had less often a favorable cytogenetic profile, had more commonly donors other than matched siblings and more often received reduced intensity conditioning (RIC) conditioning. In multivariate analysis non relapse mortality (NRM) risks in patients with prior autologous HCT were 1.34 (1.07-1.67), p=0.01 after adjustment for age, cytogenetic risk, transplant year, donor, conditioning intensity, sex matching, interval diagnosis-relapse and relapse-allogeneic HCT as compared to chemotherapy consolidation. Similarly, risks of events in leukemia free survival and graft versus host disease, relapse free survival were higher with prior autologous HCT, 1.17 (1.01-1.35), p=0.03 and 1.18 (1.03-1.35) p= 0.02, respectively. Risk of death was also higher 1.13 (0.97-1.32) p=0.1 but this was not significant. Post remission consolidation with autologous HCT for AML in CR1 increases toxicity of subsequent allogeneic HCT in CR2.

The electron paramagnetic resonance (EPR) technique is used to evaluate the temperature dependence of the aggregation processes in asphaltenes of heavy and extra-heavy crude oils and their vacuum residues from the Venezuelan Oil Belt. All of the EPR spectra of the samples studied show an intense central signal assigned to free radicals (FRs) and a multiplet as a result of the presence of a V+ 4 porphyrin. From the analyses of the inverse of the normalized area as a function of the temperature for the FR signal and the most intense peak in the vanadium multiplet, we obtain a set of temperatures related to the transition of fast-to-slow motion regions, the temperature of generation-recombination of FRs, and the cracking temperature in all of the samples. On the other hand, applying a Curie-Weiss law, we obtain the order-disorder temperatures and correlated them with the American Petroleum Institute grade of the samples. A computational simulation was performed to propose possible conformers based on the experimental data to confirm our results, which can be used as a probe of the structural and conformational changes in the asphaltene structure. The Judicial Ethics Education Project at the University of Minnesota Law School is part of the Minnesota Judicial Trial Skills Training Program and uses case simulations to show ethical and management problems needing immediate attention during a trial. Although simulation is effective according to the literature and empirical data, using the technique to teach judicial ethics has some problems. A program using the technique to train judges on the ethical problems in ex parte communication, political activity, demeanor and bias is reported on. This program received much positive anecdotal response and could be redone to fit other jurisdictions.

Plants deploy distinct secondary metabolisms to cope with environment pressure and to face bio-aggressors notably through the production of biologically active alkaloids. This metabolism-type is particularly elaborated in Catharanthus roseus that synthesizes more than a hundred different monoterpene indole alkaloids (MIAs). While the characterization of their biosynthetic pathway now reaches completion, still little is known about the role of MIAs during biotic attacks. As a consequence, we developed a new plant/herbivore interaction system by challenging C. roseus leaves with Manduca sexta larvae. Transcriptomic and metabolic analyses demonstrated that C. roseus respond to folivory by both local and systemic processes relying on the activation of specific gene sets and biosynthesis of distinct MIAs following jasmonate production. While a huge local accumulation of strictosidine was monitored in attacked leaves that could repel caterpillars through its protein reticulation properties, newly developed leaves displayed an increased biosynthesis of the toxic strictosidine-derived MIAs, vindoline and catharanthine, produced by up-regulation of MIA biosynthetic genes. In this context, leaf consumption resulted in a rapid death of caterpillars that could be linked to the MIA dimerization observed in intestinal tracts. Furthermore, this study also highlights the overall transcriptomic control of the plant defense processes occurring during herbivory. Unrelated allogeneic transplantation for severe aplastic anemia is a treatment option after immunosuppressive treatment failure in the absence of a matched sibling donor. Age, delay between disease diagnosis and transplantation, and HLA matching are the key factors in transplantation decisions, but their combined impact on patient outcomes remains unclear. Using the French Society of Bone Marrow Transplantation and Cell Therapies registry, we analyzed all consecutive patients (n=139) who underwent a first allogeneic transplantation for idiopathic severe aplastic anemia from an unrelated donor between 2000 and 2012. In an adjusted multivariate model, age over 30 years (Hazard Ratio=2.39; P=0.011), time from diagnosis to transplantation over 12 months (Hazard Ratio=2.18; P=0.027) and the use of a 9/10 mismatched unrelated donor (Hazard Ratio=2.14; P=0.036) were independent risk factors that significantly worsened overall survival. Accordingly, we built a predictive score using these three parameters, considering patients at low (zero or one risk factors, n=94) or high (two or three risk factors, n=45) risk. Paralelamente al rápido aumento de la prevalencia de sobrepeso y obesidad han surgido en el mercado múltiples productos que prometen estimular la pérdida de peso sin modificar la dieta y estilo de vida de los individuos. El objetivo de este trabajo es evaluar la evidencia científica que apoye el uso de tres productos disponibles en Chile para el control ponderal y la reducción de sobrepeso y obesidad: una emulsión lipídica que estimula el freno ileal, un inhibidor de proteasa y un inhibidor de amilasa. Los resultados indican que el inhibidor de amilasa y el inhibidor de proteasa, si bien parecen funcionar in vitro, no muestran ningún efecto sobre el control ponderal en los ensayos clínicos realizados tanto en sujetos normopeso como en obesos. El nivel de evidencia que apoya el uso de estos productos para el control ponderal es por lo tanto insuficiente. En caso de la emulsión lipídica, los resultados son conflictivos y más estudios son necesarios para confirmar su eventual efecto sobre el control del peso corporal.

Key message This review highlights some of the discoveries and applications made possible by “omics” technologies over the last 10 years and provides perspectives for pioneering research to increase our understanding of tree biology. Context A decade after the first forest tree genome sequence was released into the public domain, the rapidly evolving genomics and bioinformatics toolbox has advanced our understanding of the structure, functioning, and evolution of forest tree genomes. Aims and methods This review highlights some of the discoveries and applications that “omics” technologies have made possible for forest trees over the past 10 years. Results In this review, we start by our current understanding of genome evolution and intricacies of gene regulation for reproduction, development, and responses to biotic and abiotic stresses. We then skim over advances in interactome analysis and epigenomics, the knowledge of the extent of genetic variation within and between species, revealing micro- and macro-evolutionary processes and species history, together with the complex architecture of quantitative traits. We finally end with applications in genetic resource conservation and breeding. Conclusion The knowledge gained through the use of these technologies has a huge potential impact for adapting forests to the main challenges they will have to face changing demand from ecosystem services with potentially conflicting strategies in terms of conservation and use, as well as climate changes and associated threats. Genomics will undoubtedly play a major role over the next decade and beyond, not only to further understand the mechanisms underlying adaptation and evolution but also to develop and implement innovative management and policy actions to preserve the adaptability of natural forests and intensively managed plantations. © 2015, INRA and Springer-Verlag France.

Increasing development north of Charlotte, NC, threatens aquatic life in streams by reducing riparian zones and increasing runoff. Runoff, sedi- mentation from erosion, and poor construction practices are principal sources of pollution. We asked how land use and disturbance affected benthic insects. We visited nine streams from May to October 2001, collected data on insect diver- sity, chemistry, and physical habitat. We used a Geographic Information System to delineate watersheds and land use patterns. Watersheds were categorized based on land use, abiotic variables, and disturbance. Insect communities were more diverse in streams draining low disturbance watersheds than in streams draining highly developed watersheds. Sensitive taxa were found in streams with extensively forested watersheds, but were nonexistent in extensively devel- oped watersheds. Disturbances occurring in streams caused declines in diver- sity, often eliminating sensitive taxa. Aquatic insect diversity is related to land use patterns and disturbances, and anthropogenic alteration of habitat has nega- tive consequences to that diversity.
