Item does not contain fulltext, The therapeutic potential of minigastrin (MG) analogs for the treatment of cholecystokinin-2 receptor (CCK2R)-expressing cancers is limited by poor in vivo stability or unfavorable accumulation in non-target tissues. Increased stability against metabolic degradation was achieved by modifying the C-terminal receptor-specific region. This modification led to significantly improved tumor targeting properties. In this study, further N-terminal peptide modifications were investigated. Two novel MG analogs were designed starting from the amino acid sequence of DOTA-MGS5 (DOTA-DGlu-Ala-Tyr-Gly-Trp-(N-Me)Nle-Asp-1Nal-NH(2)). Introduction of a penta-DGlu moiety and replacement of the four N-terminal amino acids by a non-charged hydrophilic linker was investigated. Retained receptor binding was confirmed using two CCK2R-expressing cell lines. The effect on metabolic degradation of the new (177)Lu-labeled peptides was studied in human serum in vitro, as well as in BALB/c mice in vivo. The tumor targeting properties of the radiolabeled peptides were assessed using BALB/c nude mice bearing receptor-positive and receptor-negative tumor xenografts. Both novel MG analogs were found to have strong receptor binding, enhanced stability, and high tumor uptake. Replacement of the four N-terminal amino acids by a non-charged hydrophilic linker lowered the absorption in the dose-limiting organs, whereas introduction of the penta-DGlu moiety increased uptake in renal tissue.

Cytomegalovirus (CMV) is associated with immunosenescence and reduced vaccine responses in the elderly (>70 years). However, the impact of CMV in young adults is less clear. In this study, healthy UK and Senegalese adults aged 18-50 years (average 29 years) were vaccinated with the Ebola vaccine candidate chimpanzee adenovirus type 3 vectored Ebola Zaire vaccine (ChAd3-EBO-Z) and boosted with modified vaccinia Ankara Ebola Zaire vectored (MVA-EBO-Z) vaccine. CMV carriage was associated with an expansion of phenotypically senescent CD4+ and CD8+ T cells expressing CD57 and killer cell lectin-like receptor G1 (KLRG1), which was negatively associated with vaccine responses in both cohorts (P

A hallmark of science is the open exchange of knowledge. At this time of crisis, it is more important than ever for scientists around the world to openly share their knowledge, expertise, tools, and technology. Scientific models are critical tools for anticipating, predicting, and responding to complex biological, social, and environmental crises, including pandemics. They are essential for guiding regional and national governments in designing health, social, and economic policies to manage the spread of disease and lessen its impacts. However, presenting modeling results alone is not enough. Scientists must also openly share their model code so that the results can be replicated and evaluated.

- Abstract
The human respiratory tract hosts a diverse community of cocirculating viruses that are responsible for acute respiratory infections. This shared niche provides the opportunity for virus–virus interactions which have the potential to affect individual infection risks and in turn influence dynamics of infection at population scales. However, quantitative evidence for interactions has lacked suitable data and appropriate analytical tools. Here, we expose and quantify interactions among respiratory viruses using bespoke analyses of infection time series at the population scale and coinfections at the individual host scale. We analyzed diagnostic data from 44,230 cases of respiratory illness that were tested for 11 taxonomically broad groups of respiratory viruses over 9 y. Key to our analyses was accounting for alternative drivers of correlated infection frequency, such as age and seasonal dependencies in infection risk, allowing us to obtain strong support for the existence of negative interactions between influenza and noninfluenza viruses and positive interactions among noninfluenza viruses. In mathematical simulations that mimic 2-pathogen dynamics, we show that transient immune-mediated interference can cause a relatively ubiquitous common cold-like virus to diminish during peak activity of a seasonal virus, supporting the potential role of innate immunity in driving the asynchronous circulation of influenza A and rhinovirus. These findings have important implications for understanding the linked epidemiological dynamics of viral respiratory infections, an important step towards improved accuracy of disease forecasting models and evaluation of disease control interventions.

- Abstract
Seasonal influenza infections have a significant global impact leading to increased health and economic burden. The efficacy of currently available seasonal influenza vaccines targeting polymorphic surface antigens has historically been suboptimal. Cellular immune responses against highly conserved Influenza A virus antigens, such as nucleoprotein (NP) and matrix protein-1 (M1), have previously been shown to be associated with protection from disease, whilst viral-vectored vaccines are an effective strategy to boost cell-mediated immunity. We have previously demonstrated that MVA encoding NP and M1 can induce potent and persistent T cell responses against influenza. In this Phase I study, we evaluated the safety and immunogenicity of MVA-NP+M1, which was newly manufactured on an immortalized cell line, in six healthy adult participants. The vaccine was well-tolerated with only mild to moderate adverse events that resolved spontaneously and were comparable to previous studies with the same vaccine manufactured in chick embryo fibroblasts. A significant increase in vaccine-specific T cell responses was detected seven days after immunization and was directed against both antigens in the vector insert. This small Phase I study supports progression of this vaccine to a Phase IIb study to assess immunogenicity and additional protective efficacy in older adults receiving licensed seasonal influenza vaccines.

The specific effect of initial energy of CH5(super +), C2H4(super +), C2H5(super +), and C2H6(super +) ions on the extent of surface-induced fragmentation is investigated. The initial internal energy content of the projectile ions is entirely preserved in the projectile ion during the ion/surface collision, and thus is available in the subsequent dissociative processes as additional energy to the internal energy acquired by the projectile ion in the surface-excitation process.

- Abstract
This guide provides a basic introduction to the related concepts of psychological literacy and psychologically literate citizenship. Psychological literacy encapsulates the idea that an understanding of basic principles of human behaviour and development gained through study of Psychology will help us to better understand ourselves and others, and will help individuals and organisations to function better. Psychologically literate citizenship extends this idea, to incorporate the benefits that an understanding of Psychology can bring to the individuals who study it, and their local and global communities. The guide will argue that Psychology offers potential solutions to many of the world’s most pressing problems. Its publication is particularly timely following the report of Trapp, Banister, Ellis, Latto, Miell and Upton (2011) and the subsequent inclusion of psychological literacy within the criteria for accreditation of undergraduate Psychology degrees by the British Psychological Society (BPS 2012). The guide commences by introducing the theoretical contexts of psychological literacy and psychologically literate citizenship. It links these concepts to issues around graduate attributes, including employability and global citizenship, suggesting that psychologically literate graduates are also highly employable global citizens. It then goes on to outline some possible areas of the typical undergraduate Psychology curriculum that lend themselves to developing psychological literacy in students. These suggestions are not intended to be exhaustive, but rather to stimulate creative thinking in the reader, and encourage psychologists everywhere to think about the real-life value of their particular areas of expertise in Psychology. We hope that you will find it useful, share it with your colleagu es, and use it to spark enthusiasm for Psychology in your students.

The paper presents a comparison of tag frequencies in two matching one-million word reference corpora of British standard English, the 1961 LOB-corpus and its 1991 “clone” produced at Freiburg. Both corpora were tagged using a version of the CLAWS part-of-speech-tagger developed at Lancaster, and part of the material was post-edited manually in Freiburg to assess the accuracy of the automatic procedure. The comparison of tag frequencies is an essential complement to work on recent linguistic change carried out on the untagged material, because this work has been based on the – so far unverified – assumption that tag frequencies have remained constant over the thirty-year period in question. In addition, the paper discusses some common and partly contradictory claims about the prevalence of a “nominal” style in present-day written English. It is shown that while part-of-speech frequencies have not remained constant over the period investigated, the shifts are usually not big enough to invalidate the results obtained in analyses of the untagged material. With regard to style, the material shows a significant rise in the frequency of nouns, which, however, is not paralleled by a corresponding decrease in verbs.

Reactions induced by the impact of acetonitrile and deuterated acetonitrile molecular ions on a hydrocarbon-covered stainless steel surface were investigated using a recently constructed tandem mass spectrometer (BESTOF). Mass spectra of product ions formed were recorded for projectile collision energies in the range of about 5–70 eV. Both, simple dissociations of the projectile ion and chemical reactions involving H-atom transfer from the surface material (followed by subsequent dissociation of the protonated projectile ion formed) were observed. In particular, formation of CD2H+ and CD3+ via dissociation of the surface-protonated deuterated acetonitrile ion CD3CNH+ and the collision energy dependence of their relative abundance indicate substantial degree of isomerization of the acetonitrile molecular radical cation CD3CN⋅+ to the ketenimine ion CD2&z.dbnd6;C&z.dbnd6;ND⋅+. The isomerization seems to take place upon electron impact ionization of the acetonitrile molecule in the ion source preceding the tandem mass spectrometer. [Copyright &y& Elsevier]

- Abstract
ABSTRACTCandidemia is the fourth most common kind of microbial bloodstream infection, with Candida albicansbeing the most common causative species. Echinocandins are employed as the first-line treatment for invasive candidiasis until the fungal species is determined and confirmed by clinical diagnosis. Echinocandins block the FKSglucan synthases responsible for embedding β-(1,3)-d-glucan in the cell wall. The increasing use of these drugs has led to the emergence of antifungal resistance, and elevated MICs have been associated with single-residue substitutions in specific hot spot regions of FKS1and FKS2. Here, we show for the first time the caspofungin-mediated in vivoselection of a double mutation within one allele of the FKS1hot spot 1 in a clinical isolate. We created a set of isogenic mutants and used a hematogenous murine model to evaluate the in vivooutcomes of echinocandin treatment. Heterozygous and homozygous double mutations significantly enhance the in vivoresistance of C. albicanscompared with the resistance seen with heterozygous single mutations. The various FKS1hot spot mutations differ in the degree of their MIC increase, substance-dependent in vivoresponse, and impact on virulence. Our results demonstrate that echinocandin EUCAST breakpoint definitions correlate with the in vivoresponse when a standard dosing regimen is used but cannot predict the in vivoresponse after a dose escalation. Moreover, patients colonized by a C. albicansstrain with multiple mutations in FKS1have a higher risk for therapeutic failure.
