Cancer is a complex disease caused by combinations of cellular genetic alterations and heterogeneous microenvironments. The use of the robust and programmable CRISPR-Cas systems has greatly improved genome editing for precision cancer modeling and enabled multiplexed genetic manipulation for cancer treatment and mutation detection. In this review, we outline the current CRISPR-Cas toolkit, and discuss the promises and hurdles in translating this revolutionary technology into effective and safe clinical applications for cancer treatment and diagnosis.

The aim of this study was to evaluate the benefit of adding hypoalbuminemia to the American College of Surgeons-National Surgical Quality Improvement Program (ACS-NSQIP) Surgical Risk Calculator when predicting postoperative outcomes in colorectal cancer patients.The ACS-NSQIP Surgical Risk Calculator offers qualified risk evaluation in surgical decision-making and informed patient consent. To date, malnutrition defined as hypoalbuminemia, an important independent surgical risk factor in colorectal cancer, is not included.This is a retrospective, multi-institutional study of ACS-NSQIP patients (n = 18,532) who received colorectal surgery from 2009 to 2012. Models were constructed for predicting postoperative mortality and morbidity using the risk factors of the ACS-NSQIP Surgical Risk Calculator before and after adding hypoalbuminemia as a risk factor. The 2 models' performance was then compared using c-statistics and Brier scores. The ACS-NSQIP database in 2008 was used for validation of the created models.The prevalence of hypoalbuminemia (27.8%) is higher in colorectal cancer, when compared with other most common cancers. In univariate analyses, hypoalbuminemia was significantly associated with postoperative mortality and morbidity in colorectal cancer patients. In multivariate logistic regression analyses, 15 postoperative complications, including mortality and serious morbidities, were significantly predicted by hypoalbuminemia. Most of the models with hypoalbuminemia showed better performance and validation in predicting postoperative complications than those without hypoalbuminemia.In colorectal cancer, hypoalbuminemia, with levels below 3.5 g/dL, serves as an excellent assessment tool and preoperative predictor of postoperative outcomes. When combined with hypoalbuminemia as a risk factor, the ACS-NSQIP Surgical Risk Calculator offers more accurate information and estimation of surgical risks to patients and surgeons when choosing treatment options.

The characteristics of 3-dimensional (3D) cultured periodontal ligament stem cells derived from permanent teeth (pPDLSCs) and supernumerary teeth (sPDLSCs) were compared and evaluated in this study. pPDLSCs and sPDLSCs were cultured in an ultra-low attachment 6-well plate, and the changes in morphology and size were observed under an optical microscope for 14 days. Cell counting kit-8 was used to quantitatively evaluate cell viability, and a live/dead assay was performed for the qualitative assessment. The degrees of osteogenic, chondrogenic, and adipogenic differentiation of pPDLSCs and sPDLSCs were also assessed. The pPDLSCs and sPDLSCs were initially spherical, and sPDLSCs were smaller than pPDLSCs. The viability of pPDLSCs decreased by 47.9% (day 3) and 10.4% (day 7), whereas that of sPDLSCs decreased by 34.3% (day 3) and 2.5% (day 7) compared to the percentage on Day 1. No significant difference in osteogenic differentiation was found (p = 0.94); however, significant differences in adipocyte and cartilage differentiation were observed (p = 0.003 and p = 0.013, respectively). Within the limitations of this study, sPDLSCs and pPDLSCs exhibited multilineage differentiation capacities, including osteogenic, chondrogenic, and adipogenic differentiation, in 3D culture. This study developed a QR-based U-Learning Material Production System (QR-ULMPS) that provides teachers with an education tool to motivate college level students enrolled in a liberal arts course. QR-ULMPS was specifically designed to support the development of u-learning materials and create an engaging context-aware u-learning environment for students. A quasi-experimental research design was used to evaluate the overall effectiveness of QR-ULMPS; the Unified Theory of Acceptance and Use of Technology (UTAUT) assessed the feasibility of using QR-ULMPS to implement teaching activities; while the Instructional Materials Motivation Survey (IMMS) was used to measure the students' learning motivation after using the proposed u-learning system. From the results of the UTAUT questionnaire, we found that teachers rated the system positively and were willing to accept and adopt QR-ULMPS into their course content. Teachers also agreed that QR-ULMPS was a useful tool to motivate students' learning during outdoor teaching activities. Moreover, results of the IMMS questionnaire indicated that students assigned to the proposed u-learning system achieved better results than participants learning via conventional methods. We believe that the proposed u-learning system is advantageous because it enhances student motivation and allows for higher levels of engagement, particularly during outdoor learning activities. Thus, we conclude that the proposed u-learning system can create a learning experience that both interests and engages students. Although QR-ULMPS is not mature enough to be used across a sundry of educational domains, it provides an innovative opportunity for teachers to integrate a novel teaching methodology that challenges traditional educational norms.

Pulpectomy is a common treatment for severe carious lesions in primary molars. Care should be taken during pulpectomy of the primary teeth for successive permanent teeth. This case series report aimed to describe the cases of three patients who experienced excessive extrusion of canal filling materials and file separation during primary molar pulpectomy. The presence of separated files or excessive overfilling materials observed around successive permanent tooth germs leads to underdevelopment and may trigger cystic changes owing to abscess formation. The most important aspect is to preserve the unerupted successive developing permanent tooth. Therefore, clinicians should consider the anatomy of the primary molars prior to endodontic treatment, be careful when manipulating instruments during pulpectomy, provide appropriate treatment if iatrogenic incidents occur during endodontic treatment, and perform close follow-up to ensure the successful development and eruption of subsequent permanent teeth. Abstract Background The hypothalamic–pituitary–adrenal (HPA) and -gonadal (HPG) axes are two major pathways that connect the neural and endocrine systems in vertebrates. Factors such as prenatal stress and maternal exposure to exogenous steroids have been shown to affect these pathways during fetal development. Another less studied factor is the transfer of hormones across fetuses in multifetal pregnancies. This form of transfer has been shown to influence the morphology, anatomy, physiology, and behavior of the offspring in litter-bearing mammals, an influence termed the intrauterine position (IUP) effect. In this study, we sought to delineate how the IUP effects HPA and HPG brain receptors, peptides, and enzymes (hereafter components) in utero and how these influences may differ between males and females. Methods We utilized the unconventional model of culled free-ranging nutria (Myocastor coypus), with its large natural variation. We collected brain tissues from nutria fetuses and quantified the expression of key HPA and HPG components in three brain regions: prefrontal cortex, hypothalamus, and striatum. Results We found an interaction between sex and IUP in the mineralocorticoid receptor (MR), gonadotropin-releasing hormone receptor (GNRHR), androgen receptor (AR), and estrogen receptor alpha (ESR1). IUP was significant in both gonadotropin-releasing hormone (GnRH) and its receptor GNRHR, but in different ways. In the hypothalamus, fetuses adjacent to same-sex neighbors had higher expression of GnRH than fetuses neighboring the opposite sex. Conversely, in the cortex, GNRHR exhibited the inverse pattern, and fetuses that were neighboring the opposite sex had higher expression levels than those neighboring the same sex. Regardless of IUP, in most components that showed significant sex differences, female fetuses had higher mRNA expression levels than male fetuses. We also found that HPA and HPG components were highly related in the early stages of gestation, and that there was an interaction between sex and developmental stage. In the early stages of pregnancy, female component expression levels were more correlated than males’, but in the last trimester of pregnancy, male components were more related to each other than female’s. Conclusions This study suggests that there are sexually different mechanisms to regulate the HPA and HPG axes during fetal development. Higher mRNA expression levels of endocrine axes components may be a mechanism to help females cope with prolonged androgen exposure over a long gestational period. Additionally, these findings suggest different coordination requirements of male and female endocrine axes during stages of fetal development.
