Im Zuge der Untersuchung grundlegender Ionentransportmechanismen über die Haut des Regenwurms stellte sich das Epithel als sehr dicht (durchschnittlich 11.593 Ohm·cm²) und hochselektiv für die Aufnahme von Natrium gegenüber anderen Alkalimetallen heraus. Histologische Untersuchungen zeigten, dass die Haut des Regenwurms ein einschichtiges Epithel aufweist, welches sich aus mindestens drei verschiedenen Zelltypen zusammen-setzt. Das Epithel wird nach außen durch eine kollagene Cuticula begrenzt. Auf der basolateralen Seite folgen Muskelschichten, die vom Epithel durch eine Basallamina getrennt sind. Elektrophysiologische Messungen mit der Current-clamp Methode in modifizierten Ussingkammern zeigten, dass ein Teil des transepithelialen Stroms amiloridsensitiv war. Das deutete auf das Vorhandensein eines Epithelialen Natriumkanals (ENaC) in dem untersuchten Gewebe hin. Die halbmaximale Hemmkonzentration von k1/2 = 1,28 ± 0,08 µM Amilorid lag deutlich unter der bei anderen Anneliden, wie dem limnisch lebenden Blutegel, gefundenen. Weitere Experimente mit amiloridanalogen Substanzen ergaben ein Sensitivitätsprofil von Benzamil > Phenamil > Amilorid > EIPA. Die Anneliden sind somit der niedrigste Tierstamm, in dem ein funktioneller ENaC beschrieben ist, während in niedrigeren Phyla (z.B. beim Nematoden C. elegans) nur eine strukturell ähnliche Vorstufe des Kanals in Form so genannter Degenerine bekannt ist. Das macht das Gewebe interessant für die Grundlagenforschung zur epithelialen Natriumresorption, insbesondere da der Regenwurm ein natürlicher knock-out-Modellorganismus für das Steroidhormon Aldosteron ist. Peptidhorme hingegen kommen in diesem Tier vor, was eine schrittweise Erforschung des Effekts von Hormonen der Oxytocin/Vasopressin-Familie auf die Osmoregulation und speziell den ENaC ohne eine Überlagerung durch den Effekt von Steroidhormonen erlaubt. Eine solche Möglichkeit besteht in keinem Vertebratenmodell. Experimente mit Oxytocin, Vasopressin, Vasotocin, Isotocin, Annetocin, 8-lys-Conopressin und einem neusynthetisierten Peptid (2-tyr-8-lys-Conopressin) auf den amiloridsensitiven Strom (Iamilo) über die Regenwurmhaut ergaben, dass nur die Conopressinderivate den Iamilo steigerten, während alle anderen Peptidhormone diesen gesenkt hatten. Eine dabei aufgedeckte Dosisabhängigkeit des Oxytocineffekts auf Iamilo kann nun erklären, warum in der Literatur dieses Peptid von verschiedenen Autoren entweder als diuretisches oder auch als antidiuretisches Hormon beschrieben wird. Andere Ionentransportmechanismen wie ein apikaler Na/K/2Cl-Cotransporter, eine basolaterale Na/K-ATPase und nach außen gerichtete Kaliumkanäle in der basolateralen Membran konnten in der vorliegenden Arbeit ebenfalls funktionell beschrieben werden. Die Entwicklung eines detaillierten Modells von Ionenbewegungen über die Haut des Regenwurms soll in der Folge Experimente mit dem Regenwurm z.B. als Bioinidikator für ökotoxikologische Fragestellungen ermöglichen. In the course of the investigation of fundamental ion transport mechanisms across earthworm skin epithelium we revealed that this tissue is a very tight (11,593 Ohm·cm² in mean) and highly selective Na+-absorbing epithelium. Earthworm integument consists of a single layer of at least three different types of epithelial cells, covered by a collagenous cuticula and limited at its base by a basal-lamina, followed by muscle layers. Performing Ussing-chamber experiments in current-clamp mode, we could show an amiloride-sensitive current which indicates an epithelial sodium channel (ENaC) in this tissue. The half maximum blocking concentration of amiloride (k1/2 = 1.28 ± 0.08 µM) is considerably lower than in other annelids such as the limnic leech and further experiments with amiloride analogues showed an overall sensitivity profile of benzamil > phenamil > amiloride > EIPA. Annelids are therefore the lowest animal-strain shown to have this ion-channel, whereas in lower phyla (such as the nematode C. elegans) only a pre-stage of the ENaC in the form of a so called degenerin is described. It makes the tissue an interesting model for fundamental research on epithelial sodium absorption, especially because earthworm skin is a natural aldosterone knock-out model, but peptide hormones are present in this phylum. This allowed a stepwise investigation of the effect of hormones belonging to the oxytocin-vasopressin superfamily on the osmoregulation and especially the ENaC without interference of steroid hormones, an approach which is not possible in vertebrate species. The investigation of the effect of oxytocin, vasopressin, vasotocin, isotocin, annetocin, 8-lys-conopressin and a newly synthesized peptide (2-tyr-8-lys-conopressin) on the amiloride-sensitive short-circuit current (Iamilo) across earthworm skin showed that only the conopressin-derivates led to an increase in Iamilo whereas all the other tested peptides led to a decrease. An also revealed dose-dependency of the hormone effect on Iamilo can explain why oxytocin is being described in the literature either as a natriuretic or an anti-natriuretic peptide by different authors. Other ion transport systems like an apical Na/K/2Cl-cotransporter, a basolateral Na/K-ATPase and outward directed basolateral K+-channels could also be functionally described. The establishment of a detailed model of ion movements across earthworm skin will allow further experiments with the earthworm e.g. as a biomonitoring system for soil contaminations.
