Radiological objective response (OR) was assessed using modified response evaluation criteria in solid tumors (mRECIST) guidelines.Results: Baseline demographic parameters were comparable between 126 (69.6%) patients with CP-A disease and 55 patients (30.4%) with CP-B disease. Frequency of lenvatinib-related adverse events, including decreased appetite (P=0.034), diarrhea (P=0.040), elevated serum bilirubin (P=0.016) and vomiting (P=0.009), were higher in CP-B than in CP-A patients. Relative dose intensity (RDI) was significantly higher in CP-A (0.69) than CP-B patients (0.50, P < 0.001). Furthermore, OR rate (44.0%) was markedly higher in CP-A5 patients as compared to CP-A6 (25.5%), CP-B7 (22.2%), and CP-B8 patients (5.3%), respectively (P=0.002). In multivariable analysis, performance status (0 vs 1, 2, P=0.026), CP class (A vs B, P=0.045) and RDI (≥ 0.7 vs < 0.7, P=0.034) were identified as factors associated with response to lenvatinib treatment. Overall survival (OS) at 12 months was significantly different between CP-A (66.3%) and CP-B patients (30.0%, P=0.002), and between CP 5– 7 (59.2%) and CP 8 patients (34.8%, P=0.003). In multivariable analysis, CP class (A vs B, P=0.007) and Barcelona clinic liver cancer (BCLC) stage (B vs C, P=0.002) were associated with OS following lenvatinib treatment.Conclusion: Lenvatinib treatment offers significant benefits in patients with good liver function in real-world practice. The various characteristics identified in this study might be helpful as clinical predictors of response to lenvatinib and survival in clinical practice. Abstract

Major construction and initial-phase operation of a second-generation gravitational-wave detector KAGRA has been completed. The entire 3-km detector is installed underground in a mine in order to be isolated from background seismic vibrations on the surface. This allows us to achieve a good sensitivity at low frequencies and high stability of the detector. Bare-bones equipment for the interferometer operation has been installed and the first test run was accomplished in March and April of 2016 with a rather simple configuration. The initial configuration of KAGRA is named {\it iKAGRA}. In this paper, we summarize the construction of KAGRA, including the study of the advantages and challenges of building an underground detector and the operation of the iKAGRA interferometer together with the geophysics interferometer that has been constructed in the same tunnel., Comment: Resolution of some figures has been decreased from its original version submitted to a journal

Abstract

This paper documents three case studies that involve dynamic centrifuge tests that simulated large-scale shaking table tests on soil-pile-structure systems. The large-scale shaking table tests were performed using the world's largest laminar shear box with depth of 6 m and plan dimensions of 11 m and 3.5 m. Life-size steel and prestressed concrete piles were used in these tests. The large-scale tests involved intense shaking that produced strong nonlinear stress--strain effects and degradation of soil stiffness due to liquefaction in the foundation soil models. The dynamic centrifuge tests treated the large-scale models as their prototypes. Only essential information about the large-scale test models and the testing conditions were available to design and perform the dynamic centrifuge tests. The three case studies showed that carefully designed performed centrifuge tests could reproduce the key features of the responses of the large-scale models. However, some differences were also found in the results from these two types of tests. Abstract

We present possible observing scenarios for the Advanced LIGO, Advanced Virgo and KAGRA gravitational-wave detectors over the next decade, with the intention of providing information to the astronomy community to facilitate planning for multi-messenger astronomy with gravitational waves. We estimate the sensitivity of the network to transient gravitational-wave signals, and study the capability of the network to determine the sky location of the source. We report our findings for gravitational-wave transients, with particular focus on gravitational-wave signals from the inspiral of binary neutron star systems, which are the most promising targets for multi-messenger astronomy. The ability to localize the sources of the detected signals depends on the geographical distribution of the detectors and their relative sensitivity, and 90% credible regions can be as large as thousands of square degrees when only two sensitive detectors are operational. Determining the sky position of a significant fraction of detected signals to areas of 5– 20 deg[superscript 2] requires at least three detectors of sensitivity within a factor of ∼2 of each other and with a broad frequency bandwidth. When all detectors, including KAGRA and the third LIGO detector in India, reach design sensitivity, a significant fraction of gravitational-wave signals will be localized to a few square degrees by gravitational-wave observations alone. Abstract

While several reports have documented immunosuppressant-induced osteoporosis, the exact mechanism of the pathological change of the joint remains to be clarified. In the present study, we have demonstrated the pathological change of the articular cartilage in the mandibular head of five Sprague-Dawley rats administered with the immunosuppressant FK 506 for 28 days. Three-dimensional micro-computed tomography of the mandibular heads in treated rats showed a significant decrease in trabecular bone volume compared to control rats. Histological observation revealed atrophic change of the articular cartilage. Immunohistological observation using anti-proliferative cell nuclear antibody (PCNA), type I, II, and type X collagen antibodies showed significantly decreased proliferation and differentiation of chondrocytes in the articular cartilage compared with the control group (p

Abstract

This paper describes the application of the beam emission spectroscopy (BES) to Heliotron J, having the nonsymmetrical helical-magnetic-axis configuration. The spectral and spatial profile of the beam emission has been estimated by the numerical calculation taking the collisional excitation processes between plasmas (electrons/ions) and beam atoms. Two sets of the sightlines with good spatial resolution are presented. One is the optimized viewing chords which have 20 sightlines and observe the whole plasma region with the spatial resolution Δρ less than ±0.055 using the newly designed viewing port. The other is 15 sightlines from the present viewing port of Heliotron J for the preliminary measurement to discuss the feasibility of the density fluctuation measurement by BES. The beam emission has been measured by a monochromator with a CCD camera. A good consistency has been obtained between the spectral profiles of the beam emission measured by the monochromator and the beam emission spectrum deduced by the model calculation. An avalanche photodiode with an interference filter system was also used to evaluate the signal-to-noise (S/N) ratio of the beam emission in the present experimental setup. The modification of the optical system is being planned to improve the S/N ratio, which will enable us to estimate the density fluctuation in Heliotron J.

Abstract

This paper discusses environmental issues in mining and smelting, the recent technologies for environmental conservation and their application in Japanese companies. The history of non-ferrous mining and smelting activity in Japan is outlined, a list given of the resulting environmental disasters and countermeasures discussed. The absence of any mine disasters since 1970 demonstrates the effectiveness of the collaboration between government and private sector in the successful adherence to strict environmental standards. Abstract

It is important to search the biomarker to predict the development and prognosis of autoimmune thyroid diseases ( AITDs) such as Hashimoto's disease ( HD) and Graves' disease ( GD). MicroRNA (mi R) bind directly to the 3′ untranslated region of specific target m RNAs to suppress the expression of proteins, promote the degradation of target m RNAs and regulate immune response. mi R-125a is known to be a negative regulator of regulated upon activation normal T cell expressed and secreted ( RANTES), interleukin ( IL)-6 and transforming growth factor ( TGF)-β; however, its association with AITDs remains unknown. To clarify the association between AITDs and miR-125a, we genotyped the rs12976445 C/ T, rs10404453 A/ G and rs12975333 G/ T polymorphisms in the MIR125 A gene, which encodes mi R-125a, using direct sequencing and polymerase chain reaction-restriction fragment length polymorphism (PC R-RFLP) methods in 155 patients with GD, 151 patients with HD and 118 healthy volunteers. We also examined the expression of mi R-125a in peripheral blood mononuclear cells ( PBMCs) from 55 patients with GD, 79 patients with HD and 38 healthy volunteers using quantitative real-time PCR methods. We determined that the CC genotype and C allele of the rs12976445 C/ T polymorphism were significantly more frequent in patients with HD compared with control subjects ( P < 0·05) and in intractable GD compared with GD in remission ( P < 0·05). The expression of mi R-125a was correlated negatively with age ( P = 0·0010) and down-regulated in patients with GD compared with control subjects ( P = 0.0249). In conclusion, mi R-125a expression in PBMCs and the rs12976445 C/ T polymorphism were associated with AITD development and prognosis. [ABSTRACT FROM AUTHOR]

Abstract

Vitamin D is a multi-functional immune regulator, and a low serum concentration of vitamin D promotes autoimmune inflammation. In this study, we evaluate the association between the prognosis of autoimmune thyroid disease ( AITD) and the functional polymorphisms of genes that regulate vitamin D metabolism. For 139 Graves' disease ( GD) patients, 116 Hashimoto's disease ( HD) patients and 76 control subjects, we genotyped the following polymorphisms using polymerase chain reaction-restriction fragment length polymorphism ( PCR- RFLP): vitamin D receptor ( VDR): rs731236, rs7975232, rs2228570 and rs1544410; group-specific component ( GC): rs7041 and rs4588; and CYP2 R1: rs10741657. The frequency of the TT genotype for the rs731236 polymorphism was higher in GD patients than in HD patients ( P = 0·0147). The frequency of the C allele for the rs7975232 polymorphism was higher in GD patients than in control subjects ( P = 0·0349). The proportion of GD patients whose anti-thyrotrophin receptor antibody ( TRAb) level was >51% was higher in those with the CC genotype than in those with the CA+ AA genotypes ( P = 0·0065). The frequency of the CC genotype for the rs2228570 polymorphism was higher in HD patients than in control subjects ( P = 0·0174) and GD patients ( P = 0·0149). The frequency of the Gc1 Gc1 genotype for the GC polymorphism and the AG genotype for the CYP2 R1 polymorphism were lower in intractable GD than in GD in remission ( P = 0·0093 and 0·0268, respectively). In conclusion, genetic differences in the VDR gene may be involved in the development of AITD and the activity of GD, whereas the genetic differences in the GC and CYP2 R1 genes may be involved with the intractability of GD.
