International audience; We propose to extend the fix of Roe's approximate Riemann solver developed for the Barotropic Euler equations in [2] to the full Euler equations. This scheme is built mainly to handle low Mach acoustic waves. Moreover, compared to pressure-centered type schemes, this numerical fix has the advantage of improving the numerical solution in the sense that the oscillating modes are reduced. The theoretical study is based on a two-time scales asymptotic analysis. It is proved that the Euler system equipped with a general equation of state is consistent with a first-order wave system in a low Mach number regime. Similar analysis is performed at the discrete level on the Roe scheme to derive the new fix. Numerical tests confirm the results obtained for the Barotropic case about the ability of this fix to deal with both steady and low Mach acoustic computations also in the case of full Euler equations.

Microglia play a pivotal role in neurodegenerative disease pathogenesis, but the mechanisms underlying microglia dysfunction and toxicity remain to be fully elucidated. To investigate the effect of neurodegenerative disease-linked genes on the intrinsic properties of microglia, we studied microglia-like cells derived from human induced pluripotent stem cells (iPSCs), termed iMGs, harboring mutations in profilin-1 (PFN1) that are causative for amyotrophic lateral sclerosis (ALS). ALS-PFN1 iMGs exhibited lipid dysmetabolism and deficits in phagocytosis, a critical microglia function. Our cumulative data implicate an effect of ALS-linked PFN1 on the autophagy pathway, including enhanced binding of mutant PFN1 to the autophagy signaling molecule PI3P, as an underlying cause of defective phagocytosis in ALS-PFN1 iMGs. Indeed, phagocytic processing was restored in ALS-PFN1 iMGs with Rapamycin, an inducer of autophagic flux. These outcomes demonstrate the utility of iMGs for neurodegenerative disease research and highlight microglia vesicular degradation pathways as potential therapeutic targets for these disorders.

This article is dedicated to the long time behavior of a finite volume approximation of general symmetrizable linear hyperbolic system on a bounded domain. In the continuous case this problem is very difficult, and the $\omega $–limit set (namely the set of all the possible long time limits) may be large and complicated to depict if no dissipation is introduced. In this article we prove that in general, with a stable finite volume scheme, the discrete solution converges to a steady state when the time goes to infinity. This property is a direct consequence of the numerical dissipation mechanisms used for stabilizing the discretization. We apply this result for determining the long time limit for several stabilizations of the wave system, and perform a formal link with the low Mach number problem of the nonlinear Euler system. Numerical experiments with the wave system are performed for confirming the theoretical results obtained.

Fossil‐bound organic material holds great potential for the reconstruction of past changes in nitrogen (N) cycling. Here, with a series of laboratory experiments, we assess the potential effect of oxidative degradation, fossil dissolution, and thermal alteration on the fossil‐bound N isotopic composition of different fossil types, including deep and shallow water scleractinian corals, foraminifera, diatoms and tooth enamel. Our experiments show that exposure to different oxidizing reagents does not significantly affect the N isotopic composition or N content of any of the fossil types analyzed, demonstrating that organic matter is well protected from changes in the surrounding environment by the mineral matrix. In addition, we show that partial dissolution (of up to 70%–90%) of fossil aragonite, calcite, opal, or enamel matrixes has a negligible effect on the N isotopic composition and N content of the fossils. These results suggest that the isotopic composition of fossil‐bound organic material is relatively uniform, and also that N exposed during dissolution is lost without significant isotopic discrimination. Finally, our heating experiments show negligible changes in the N isotopic composition and N content of all fossil types at 100°C. At 200°C and hotter, any N loss and associated nitrogen isotope changes appear to be directly linked to the sensitivity of the mineral matrix to thermal stress, which depends on the biomineral type. These results suggest that, so long as high temperature does not compromise the mineral structure, the biomineral matrix acts as a closed system with respect to N, and the N isotopic composition of the fossil remains unchanged., Plain Language Summary: The ratio of the heavy and light isotopes of nitrogen (15N and 14N) in the organic material contained within the mineral structure of fossils can be used to reconstruct past changes in biological and chemical processes. With a series of laboratory experiments, we evaluate the potential effects of chemical conditions, fossil dissolution, and heating on the nitrogen isotopic composition (15N/14N ratio) of corals, foraminifera, diatoms and tooth enamel. Our results indicate that these processes do not have a significant effect on the 15N/14N of fossils, suggesting that the mineral matrix provides a barrier that isolates a fossil's organic nitrogen from the surrounding environment, preventing alteration of its 15N/14N. In addition, we show that if part of the fossil‐bound organic nitrogen is exposed by dissolution or heating, it is lost without affecting the 15N/14N of the organic material that remains in the mineral. These findings imply that the original 15N/14N ratio incorporated by the organism is preserved in the geologic record. Therefore, measurements of the nitrogen isotopes on fossils can provide faithful biological, ecological, and environmental information about the past., Key Points: Fossil‐bound organic matter is well protected by the mineral matrix from chemical changes in the surrounding environment. This thesis deals with the modeling and numerical approximation of compressible gas-liquid flows. The main difficulty lies in modeling and approximation of the liquid-gas interface. The two-fluid model is a system of conservation laws closed with a mixture pressure law. The law has to be chosen carefully, it conditions good properties of the system as hyperbolicity or existence of a Lax entropy. Classic conservative Godunov-type schemes lead to inaccuracies that make them unusable inpractice. The existence of discontinuous solutions makes it difficult to build high order schemes and requires very fine meshes to an acceptable accuracy. It is therefore essential to provide efficient algorithms for the High Performance Computing. In this thesis, we will partially treat each of these issues : construction of a "good" pressure law, building adapted numerical schemes, programming on GPU or GPU cluster., Cette thèse traite de la modélisation et de l'approximation numérique des écoulements liquide-gaz compressibles. La difficulté centrale est la modélisation et l'approximation de l'interface liquide-gaz. Le modèle bifluide est constitué d'un système de lois de conservation fermé par une loi d'état du mélange. La loi d'état conditionne les bonnes propriétés (hyperbolicité, existence d'une entropie de Lax) du système. Les schémas classiques de type Godunov conduisent à des imprécisions les rendant inutilisables en pratique. L'existence de solutions discontinues rend difficile la construction de schémas d'ordre élevé et nécessite des maillages très fins pour une précision acceptable. Il est indispensable de proposer des algorithmes performants pour les calculateurs parallèles les plus récents. Nous aborderons chacune de ces problématiques: construction d'une "bonne" loi de pression, construction de schémas numériques adaptés, programmation sur calculateur massivement multicoeur.

Short-term preoperative methionine restriction (MetR) shows promise as a translatable strategy to modulate the body's response to surgical injury. Its application, however, to improve post-interventional vascular remodeling remains underexplored. Here, we find that MetR protects from arterial intimal hyperplasia in a focal stenosis model and adverse vascular remodeling after vein graft surgery. RNA sequencing reveals that MetR enhances the brown adipose tissue phenotype in arterial perivascular adipose tissue (PVAT) and induces it in venous PVAT. Specifically, PPAR-α was highly upregulated in PVAT-adipocytes. Furthermore, MetR dampens the post-operative pro-inflammatory response to surgery in PVAT-macrophages in vivo and in vitro . This study shows for the first time that the detrimental effects of dysfunctional PVAT on vascular remodeling can be reversed by MetR, and identifies pathways involved in browning of PVAT. Furthermore, we demonstrate the potential of short-term pre-operative MetR as a simple intervention to ameliorate vascular remodeling after vascular surgery.
