Abstract

Grapevine trunk diseases (GTDs) are devastating diseases involving xylem-inhabiting fungi. Among them, Esca and Botryosphaeria dieback affect vineyards in major worldwide grape-producing areas. The causal fungi alter the woody issues and could produce toxins that disturb the plant physiology, leading to chronic or apolectic leaf symptoms. The unique effective fungicide against these diseases, sodium arsenite, is now prohibited because of toxicity. As GTDs are complex and still partly unknown, the identification of protection strategies remain a deadlock. Global and targeted approaches were therefore conducted to get new information on the impact of the diseases and sodium arsenite treatment on grapevine metabolism, development and physiology.

Abstract

Les travaux réalisés sur deux maladies du bois de la vigne (esca, BDA) ont montré leur effet sur la récolte et la croissance de la pousse printanière qui était annulé deux années après l’apparition des symptômes. Ces maladies ont aussi conduit à des modifications des propriétés analytiques des moûts et des vins. Les vins issus de raisins collectés sur des ceps symptomatiques n’étaient pas toujours les plus dépréciés. Les études effectuées sur le cycle biologique ont permis de mettre au point des outils pour différencier les individus de deux espèces de Botryosphaeriaceae et des outils d’hybridation in situ qui permettront de les localiser précisément dans les tissus. Elles ont aussi identifié la période de sensibilité de la plante face à ces champignons et montré l’importance de deux facteurs (stade phénologique, charge) pour leur développement. Les organes porteurs de leurs sources d’inoculum ont été identifiés et une méthode a été mise au point pour les quantifier. Parmi les pratiques agronomiques testées (taille, densité, aménagement de coteaux), aucunes n’influaient sur leur expression dans le vignoble, la situation semblait avoir plus d’importance que les pratiques testées. Enfin, la production de plants totalement exempts de champignons pathogènes ou moins porteurs n’a pas été efficace dans la lutte contre ces maladies.

Abstract

Non-self-recognition of microorganisms partly relies on the perception of microbe-associated molecular patterns (MAMPs) and leads to the activation of an innate immune response. Bacillus subtilis produces three main families of cyclic lipopeptides (LPs), namely surfactins, iturins and fengycins. Although LPs are involved in induced systemic resistance (ISR) activation, little is known about defence responses induced by these molecules and their involvement in local resistance to fungi. Here, we showed that purified surfactin, mycosubtilin (iturin family) and plipastatin (fengycin family) are perceived by grapevine plant cells. Although surfactin and mycosubtilin stimulated grapevine innate immune responses, they differentially activated early signalling pathways and defence gene expression. By contrast, plipastatin perception by grapevine cells only resulted in early signalling activation. Gene expression analysis suggested that mycosubtilin activated salicylic acid (SA) and jasmonic acid (JA) signalling pathways, whereas surfactin mainly induced an SA-regulated response. Although mycosubtilin and plipastatin displayed direct antifungal activity, only surfactin and mycosubtilin treatments resulted in a local long-lasting enhanced tolerance to the necrotrophic fungus Botrytis cinerea in grapevine leaves. Moreover, challenge with specific strains overproducing surfactin and mycosubtilin led to a slightly enhanced stimulation of the defence response compared with the LP-non-producing strain of B. subtilis. Altogether, our results provide the first comprehensive view of the involvement of LPs from B. subtilis in grapevine plant defence and local resistance against the necrotrophic pathogen Bo. cinerea.
