Background Proprotein convertase subtilisin/kexin type 9 (PCSK9) has multiple roles in the development and progression of atherosclerosis. Recent studies have indicated an association of PCSK9 with thrombotic process. Purpose We aimed to examine whether there exists a relationship between plasma PCSK9 concentration and the most commonly coagulation parameters including prothrombin time (PT) and activated partial thromboplastin time (APTT). Methods A total of 2293 consecutive patients with angina-like chest pain who had no treatment of lipid-lowering drugs were enrolled in this study. The baseline clinical and laboratory data were collected. PT and APTT tests were performed. Circulating PCSK9 concentrations were determined by ELISA and classified into three subgroups according to their levels of PCSK9 tertiles. The relation of PCSK9 concentration to PT or APTT were analyzed. Results Firstly, we found that the patients with high PCSK9 levels trended to have lower PT and APTT (p Conclusion Circulating PCSK9 concentration was independently negative associated with PT, suggesting a potential link between PCSK9 and PT that may be involved in atherogenesis and atherothrombosis. Ti-based metallic glass matrix composites (MGMCs) with a composition of Ti50Zr20V10Cu5Be15 (atomic percent, at. %) exhibit excellent tensile ductility and distinct work-hardening capability. A dislocation pile-up model (DPM) has been established to elaborate the dislocation motion near the yield point and to theoretically derive a linear Hall-Petch-like relationship between the yield strength, σ, and the inverse square root of the diameter of dendrite arms, d−1/2. The materials constant, k, in the present Hall-Petch-like relationship can be calculated on the basis of the pile-up model, and is very close to the experimental value. The hardness variation in the dendrites and the strengthening effect from unloading-reloading tests prove the reasonability of the DPM during tension. The Hall-Petch-like relationship is verified for a variety of MGMCs, whose plastic deformation is only dominated by dislocation motion. Mean-field theory (MFT) has been first utilized to build a relationship between the critical diameter of dendrites, dc, and the composition in MGMCs with the similar atomic percentages of low solubility elements. By tuning the composition, one can universally scale the Hall-Petch-like relationship, and accurately predict the yield strength of such in-situ MGMCs.

Objectives For patients with recurrent/metastatic cervical cancer, incorporation of anti-angiogenesis therapy with chemotherapy yields a modest survival benefit of 3.7 months over chemotherapy alone (Tewari et al. NEJM. 2014). The rationale for checkpoint inhibition is supported by programmed death ligand-1 (PD-L1) expression in some cervical cancers (∼70%), with a higher proportion noted in squamous cell carcinoma vs adenocarcinoma. Based on the 14.3% objective response in KEYNOTE-158, the US FDA granted accelerated approval to pembrolizumab for PD-L1–positive cervical cancer for second-line therapy and beyond. Methods KEYNOTE-826 is a phase 3, randomized, double-blind, placebo-controlled, multinational trial of chemotherapy with pembrolizumab or placebo for first-line treatment of recurrent, persistent, or metastatic cervical cancer. Patients not previously treated with chemotherapy for recurrence and not amenable to curative treatment will be randomized 1:1 to chemotherapy + pembrolizumab 200 mg or placebo every 3 weeks. The chemotherapy regimen (paclitaxel 175 mg/m2 + cisplatin 50 mg/m2 or carboplatin AUC 5, with or without bevacizumab 15 mg/kg) will be selected by investigators pre-randomization. Discrete, jerky stress-drop bursts (serrated flows) during plastic deformation of a Zr-based bulk metallic glass exhibit different magnitudes in response to variant strain rates. Through the classical statistical analysis of stress-strain curves during serrated flow, plastic dynamic behavior changes from Gauss to the power-law distribution with increasing the strain rates. In addition, a quantitative liner relation between mean stress drops and the logarithm of strain rates is obtained to describe the influence of strain rates on mean amplitudes of the stress drop. The power-law scaling behavior is illustrated in different views including the strain-rate dependence, energy dynamics, the Poisson's ratio of material, and shear-banding operation. These results are helpful for understanding the serration behavior at room temperature.

To understand the causes of the past water cycle variations and the influence of climate variability on the streamflow, lake storage, and flood potential, we analyze the changes in streamflow and the underlying drivers in four typical watersheds (Gaosha, Meigang, Saitang, and Xiashan) within the Poyang Lake Basin, based on the meteorological observations at 79 weather stations, and datasets of streamflow and river level at four hydrological stations for the period of 1961-2000. The contribution of different climate factors to the change in streamflow in each watershed is estimated quantitatively using the water balance equations. Results show that in each watershed, the annual streamflow exhibits an increasing trend from 1961–2000. The increases in streamflow by 4.80 m3 s−1 yr−1 and 1.29 m3 s−1 yr−1 at Meigang and Gaosha, respectively, are statistically significant at the 5% level. The increase in precipitation is the biggest contributor to the streamflow increment in Meigang (3.79 m3 s−1 yr−1), Gaosha (1.12 m3 s−1 yr−1), and Xiashan (1.34 m3 s−1 yr−1), while the decrease in evapotranspiration is the major factor controlling the streamflow increment in Saitang (0.19 m3 s−1 yr−1). In addition, radiation and wind contribute more than actual vapor pressure and mean temperature to the changes in evapotranspiration and streamflow for the four watersheds. For revealing the possible change of streamflow due to the future climate change, we also investigate the projected precipitation and evapotranspiration from of the Coupled Model Intercomparison Project phase 3 (CMIP3) under three greenhouse gases emission scenarios (SRESA1B, SRESA2 and SRESB1) for the period of 2061–2100. When the future changes in the soil water storage changes are assumed ignorable, the streamflow shows an uptrend with the projected increases in both precipitation and evapotranspiration (except for the SRESB1 scenario in Xiashan watershed) relative to the observed mean during 1961–2000. Furthermore, the largest increase in the streamflow is found at Meigang (+4.31%) and Xiashan (+3.84%) under the SRESA1B scenario, while the increases will occur at Saitang (+6.87%) and Gaosha (+5.15%) under the SRESB1 scenario.

Background Patients with advanced cervical cancer comprise a high-risk, poor prognostic group. Vascular endothelial growth factor (VEGF) has emerged as a therapeutic target for these patients, though incorporation of the anti-VEGF agent bevacizumab to platinum- and taxane-based chemotherapy (CT) is associated with a modest OS benefit vs CT alone (median OS, 16.8 vs 13.3 mo; hazard ratio, 0.77, 95% CI, 0.62-0.95; P = 0.007; Tewari et al. Lancet. 2017). On the basis of an ORR of 14.3% (95% CI, 7.4-24.1) among 77 pretreated women with PD-L1–positive tumors in the cervical cancer cohort of KEYNOTE-158 (Chung et al. J Clin Oncol. 2019), the PD-1 inhibitor pembrolizumab was granted accelerated approval by the US FDA for patients with PD-L1–positive (combined positive score [CPS] of > 1) cervical cancer who had progressed during or after first-line CT. KEYNOTE-826 (NCT03635567) is a phase 3, randomized, double-blind, multinational study designed to evaluate the efficacy and tolerability of CT with or without pembrolizumab and/or bevacizumab in the first-line setting. Trial design Eligible patients with recurrent, persistent, or metastatic cervical cancer not previously treated with CT in a recurrent or metastatic setting who are not amenable to curative treatment will be randomized 1:1 to CT + pembrolizumab 200 mg or placebo every 3 weeks. Background Pembro, a PD-1 inhibitor, confers durable benefit in many tumor types but has limited efficacy in non–microsatellite instability–high (MSI-high)/pMMR mCRC. Chemotherapies that include 5-fluorouracil (5-FU), oxaliplatin and irinotecan, which are commonly used to treat mCRC, may modulate intrinsic tumor immunogenicity and sensitize tumors to immunotherapy agents. In the phase 1b KEYNOTE-651 study (NCT03374254), pembro + mFOLFOX7 (cohort B) or pembro + FOLFIRI (cohort D) was evaluated in patients with non–MSI-high/pMMR mCRC. Methods Patients were ≥18 years with non–MSI-high/pMMR mCRC, Eastern Cooperative Oncology Group performance status 0/1, and no prior systemic chemotherapy for stage IV mCRC (cohort B) or 1 prior therapy including a fluoropyrimidine + oxaliplatin-based regimen (cohort D). Patients received pembro 200 mg every 3 weeks (Q3W) + mFOLFOX7 (oxaliplatin [85 mg/m2]; leucovorin [400 mg/m2]; 5-FU [2400 mg/m2]) Q2W (cohort B) or pembro 200 mg Q3W + FOLFIRI (irinotecan [180 mg/m2]; leucovorin [400 mg/m2]; 5-FU [2400 mg/m2]) Q2W (cohort D). Primary objectives were safety/tolerability and establishment of the recommended phase 2 dose (RP2D); the secondary objective was objective response rate. Results At data cutoff (Feb 18, 2019), 15 patients in cohort B and 16 in cohort D started treatment; median follow-up was 6.8 months (cohort B) and 5.7 months (cohort D). Treatment was discontinued in 4 patients (27%) in cohort B (adverse event [AE], n = 1 [7%]; PD, n = 3 [20%]) and 9 patients (56%) in cohort D (AEs, PD, patient withdrawal; n = 3 [19%] each). There was 1 dose-limiting toxicity in cohort D: grade 3 small-intestinal obstruction. See RP2Ds for cohorts B and D in the Methods section. All patients had ≥1 treatment-related AE (TRAE). Grade 3 TRAEs occurred in 8 patients each in cohort B (53%) and cohort D (50%), most commonly anemia and neutropenia (13% each) in cohort B and neutropenia, diarrhea, fatigue, and leukopenia (13% each) in cohort D. There were no grade 4-5 TRAEs. Efficacy results will be presented. Conclusions Pembro in combination with mFOLFOX7 or FOLFIRI was safe and tolerable in patients with mCRC. This study investigated the effects of angiotensin II (AngII) intervention, using captopril and losartan, on the expression of matrix metalloproteinase-2 (MMP-2), MMP-9, tissue inhibitor of metalloproteinase-1 (TIMP-1), and collagen in rats with pulmonary hypertension, in an effort to understand mechanisms underlying pulmonary vascular remodeling. A total of 40 male Sprague-Dawley rats were randomly divided into normal group, model group, captopril group, and losartan group. After 5 weeks, the mean pulmonary arterial pressure (mPAP), right ventricular index, and neointima formation in each group were determined. Immunohistochemical analysis was performed to determine the degree of pulmonary arterial muscularization as well as MMP-2, MMP-9, and TIMP-1 protein expression in lung tissue. Real-time fluorescent quantitative PCR was used to detect MMP2, MMP9, TIMP1, COL1A1, and COL4A1 mRNA expression. Picro-sirius red staining was performed to detect collagen protein expression. Neointima formation was observed in the model group. Moreover, the mPAP, right ventricular index, degree of arterial muscularization, and collagen deposition, as well as mRNA and protein expression of MMP2, MMP9, and TIMP1 were significantly higher than those in the other groups (P < 0.05). The mPAP, right ventricular index, degree of arterial muscularization, and mRNA and protein expression in the captopril and losartan groups were significantly decreased compared with those of the model group (P < 0.05). AngII regulates MMP-2, MMP-9, and TIMP-1 expression and affects collagen deposition. Thus, this hormone is involved in pulmonary vascular remodeling, indicating a possible mechanism that can be targeted in pulmonary hypertension intervention.

Localized heating during slip avalanches is relevant to the understanding of the transition from shear banding to cracking in bulk metallic glasses (BMGs). Here we focus on the released elastic energies (incorporating the influence of machine stiffness) and avalanche durations for regularly appearing large avalanches to study the maximum temperature increases of a Zr-based BMG compressed at three strain rates of 3 × 10−5, 3 × 10−4, and 3 × 10−3 s−1. The maximum released elastic energies during slip avalanches decrease with the increase of strain rates. The durations of the avalanches are on the scale of tens of milliseconds for each strain rate. As strain rate increases, the maximum temperature rises due to shear-band slips are decreased from 2.6, 2.3 to 1.9 K. Moderate heating during the large slip avalanches for three strain rate implies that thermal softening has only a small effect on the shear-banding instability.

Microcrystalline boron-doped diamond (BDD) films were prepared on silicon substrates by hot-filament chemical vapor deposition method. Different grain sizes of the boron-doped diamond films with nearly the same boron concentration were obtained by changing the nuclei concentration on the silicon substrates. The electrical transport behaviors of as-prepared boron-doped diamond films were measured, and the results revealed that with the increase of the grain sizes on the BDD films, the superconducting transition temperature was evidently increased. This phenomenon indicates that the superconducting transition of microcrystalline BDD films is influenced by the inhomogeneity aroused by the grain boundary effect. Our results could expand our understanding of the superconducting mechanism of microcrystalline BDD films, and are also significant to conventional applications of BDD materials.

Guo's et al. comments on our paper (Z.T. Guo, J.Y, Ge, G.Q. Xiao, Q.Z. Hao, H.B. Wu, T. Zhan, L. Liu, L. Qin, F.M. Zeng, B.Y. Yuan, Comment on “Mudflat/distal fan and shallow lake sedimentation (upper Vallesian–Turolian) in the Tianshui Basin, Central China: Evidence against the late Miocene eolian loess” by A.M. Alonso-Zarza, Z. Zhao, C.H. Song, J.J. Li, J. Zhang, A. Martín-Pérez, R. Martín-García, X.X. Wang, Y. Zhang and M.H. Zhang [Sedimentary Geology 222 (2009) 42–51], Sedimentary Geology, 2010-this issue) mostly stress their previous data and their model of configuration and evolution of the study area; it is not a real discussion of the sedimentological features we describe. In this reply we will discuss some of the key features of the basin configuration, correlations and sedimentology of the Tinshui basin. Our work has followed the common procedures used in stratigraphy and sedimentology and so we can confirm our interpretation on basin configuration and correlations. In all cases we have taken into account previous papers, including those of Guo's group. In addition the sedimentological model we proposed is new due to the lack of previous sedimentological studies, including facies analysis and petrography, in the studied area. Our model of a continental alluvial-lacustrine basin fits well with other well-known examples over the world and explains clearly the lateral facies transitions across the basin. It is not the aim of this reply to discuss all the previous papers by Guo's group, but to reply to their main comments on our paper. © 2010

A 2-D optical scanner was developed for the imaging and quantification of up-converting phosphor (UCP) labels in immunoassays. With resolution better than 500 microm, a scan rate of 0.4 mm/s, and a 1-2% coefficient of variation for repeatability, this scanner achieved a detection limit of fewer than 100 UCP particles in an 8.8. x 10(4) microm(2) area and a dynamic range that covered more than three orders of magnitude. Utilizing this scanner, a microfluidic chip immunoassay for the cytokine interferon-gamma (IFN-gamma) was developed: concentrations as low as 3 pM (50 pg/mL) were detected from 100 microL samples with a total assay time of under an hour, including the 8 min readout. For this UCP-based assay, 2-D images of the capture antibody lines were scanned, image processing techniques were employed to extract the UCP emission signals, a response curve that spanned 3-600 pM IFN-gamma was generated, and a five-parameter logistic mathematical model was fitted to the data for determination of unknown IFN-gamma concentrations. Relative to common single-point or 1-D scanning optical measurements, our results suggest that a simple 2-D imaging system can speed assay development, reduce errors, and improve accuracy by characterizing the spatial distribution and uniformity of surface-captured optical labels as a function of assay conditions and device parameters.

The information of coal-bed distribution, rank and gas-bearing nature of Jurassic coals in eastern Junggar basin were analyzed, and the controlling factors of coal-bed methane (CBM) accumulation were pointed out, which were coal rank, coal seam depth, and preservation conditions. The most important factor was depth of weathered zone of methane and coal rank. Consequently, Badaowan formation in Bogeda piedmont and Jimusaer depression and coal bed of Xishanyao formation in Wutongwozi depression were identified as the enriched areas of CBM in eastern Junggar basin. In a word, the Bogeda piedmont would be the main target for exploration in this area.

This paper aims at the problems and countermeasures of cathodic protection acceptance of the Yili River crossing project by horizontal directional drilling (HDD) method, addressing the performability of the acceptance criteria using polarization potential offset of not less than 0.7V required by the Kazakhstan Owners. Polarization potential and depolarization potential measurements as well as coating resistance measurements have been carried out based on the methods and requirements specified in NACE and ISO standard. The results show that protection coating has been damaged in the process of the HDD implement and cathodic protection system can protect the steel exposed, and that the polarization potential offset of not less than 0.7V required by the Owner cannot be met even the protection potential within the protection criteria specified in the design document. Finally, polarization potential offset of not less than 0.33V has been agreed on by all parties for the acceptance of HDD pipe section. Pipelines in more undulated areas are likely to have crossing points in pipeline transportation as oil throughput varies. This is to say that the potential energy of oils, from the peak to the terminal, is possibly higher than energy required for overcoming friction factor in oil movement, thus producing slack flow in pipes after the peak and accelerating partial flow velocity to consume remaining energy. Existed slack flow will be problem-prone like increased surge pressure, slow response to post-peak accident and vibration incurred by repeated occurrence and collapse of the steam bubble, when there are separation and crash of the liquid column in sudden change of the liquid flow speed. Therefore, targeted measures are necessary for avoiding slack flow. This text, for the first time, develops a parameter calculation model based on mechanism and characteristics of slack flow, proposes technology of predicting characteristics of the slack flow of pipelines in hilly areas with large fall and correctly predicts the defined parameters for the slack flow, including position, length, steam space volume, air space ratio and oil flow speed. Semiconductor nanoparticles (quantum dots) were studied in the context of liquid scintillator development for upcoming neutrino experiments. The unique optical and chemical properties of quantum dots are particularly promising for the use in neutrinoless double beta decay experiments. Liquid scintillators for large scale neutrino detectors have to meet specific requirements which are reviewed, highlighting the peculiarities of quantum-dot-doping. In this paper, we report results on laboratory-scale measurements of the attenuation length and the fluorescence properties of three commercial quantum dot samples. The results include absorbance and emission stability measurements, improvement in transparency due to filtering of the quantum dot samples, precipitation tests to isolate the quantum dots from solution and energy transfer studies with quantum dots and the fluorophore PPO., version 2, minor text updates

The Tianshui Basin in central China contains a thick sedimentary sequence (~1400 m) of continental deposits, Aragonian to Villafranchian (Miocene-Pliocene) in age. Intense Himalayan movements around the Paleogene/Miocene boundary triggered the uplift of mountain ranges around the Tianshui Basin, providing the deposition site for continental sediments. The sedimentary infill of the basin consists of four stratigraphic units (I to IV). This paper focuses on Unit II. Most of the accommodation space was occupied by Unit I, so during the sedimentation of Unit II, the morphology of the basin was relatively flat, promoting the development of wide distal fan/mudflat areas and wide shallow lakes. Deposits include: red mudstones, pedogenic and groundwater calcretes, reworked calcrete deposits, sheet-floods, fluvial channels, rippled sandstones/siltstones, ooidal/peloidal packstones, palustrine limestones, bioturbated marls and intraclastic limestones/marls. The characteristics and organization of the deposits indicate the gradual transition from alluvial to lake environments. Within the distal fan/mudflat, the deposition of reworked calcrete clasts as one of the most striking facies of the basin may be attributed to substantial recycling of calcrete levels and red clays. At the lake margins, the presence of ooids and palustrine limestones suggests the possibility of ramplike margins within different energy settings. The origin of some of the deposits of the basin's QA-I section is under discussion, and their consideration as eolian has been recently proposed. However, we believe this possibility is precluded by the characteristics of the deposits. Our proposal has important implications for revising the interpretation of Miocene paleoclimatic conditions in central Asia.

High aspect ratio diamond nanocone arrays are formed on freestanding diamond film by means of focused ion beam (FIB) milling technology and hot-filament chemical vapor deposition (HFCVD) method. The structure and phase purity of an individual diamond nanocone are characterized by scanning electron microscopy (SEM) and micro-Raman spectroscopy. The result indicates that the diamond cones with high aspect ratio and small tip apex radius can be obtained by optimizing the parameters of FIB milling and diamond growth. The diamond nanocone arrays were also used to study the electron field emission properties and electric field shielding effect, finding high emission current density, low threshold and weak shielding effect, all attributable to the high field enhancement factor and suitable cone density of the diamond nanocone emitter

In the USA, Kaposi's sarcoma associated with the acquired immunodeficiency syndrome (AIDS-KS) is ten times more common in homosexual or bisexual men than in heterosexual men with AIDS. One explanation for this finding is that AIDS-KS may be caused by an infectious agent. Because there is a high incidence of human papillomavirus (HPV) infection, especially HPV-16, in homosexual men, we have sought HPV DNA sequences in Kaposi's sarcoma. We used the polymerase chain reaction with a primer pair specific for the highly conserved E6 region of HPV-16 to detect HPV-16 homologous DNA fragments in tumour tissues from 97 patients with KS and in KS-derived cell cultures. HPV DNA sequences were found in 11 of 69 KS skin tumours from homosexual men with AIDS-KS, in 3 of 11 KS biopsy specimens from homosexual men who had no clinical or laboratory evidence of HIV-infection, and in 5 of 17 KS skin lesions from HIV-1-negative elderly men and women with classic KS. The same primer pair amplified HPV-16 homologous fragments from two different continuous cell cultures derived from pleural effusion fluid of patients with pulmonary AIDS-KS and two continuous cell cultures derived from KS skin lesions. The findings suggest that HPV-16-related DNA sequences are associated with different forms of KS and may have a role in the pathogenesis of this neoplasm.
