2. Investigation of the genetic cause and related phenotypes of rare early onset retinal dystrophies

Early onset retinal dystrophies (EORD) are a group of disorders presenting in childhood with degenerative abnormalities in photoreceptor cells. They are one of the leading causes of sight impairment in the United Kingdom. Since the initial discovery of Rho causing dominant retinitis pigmentosa in 1990, more than 160 genes have been associated with retinal dystrophy. Many, including CRB1, CRX, and RPE65 exhibit phenotypic heterogeneity and have been associated with more than one retinal disorder. Increasingly, with the advent of next generation sequencing, the association of non-syndromic retinal dystrophy with mutations in syndromic genes has been reported including CEP290, CLN3, and BBS1. In this thesis, a large cohort of patients with EORD underwent both detailed phenotyping to characterise their condition and molecular genetic investigations to identify and investigate the underlying causative variants. Many areas of the presented research were driven by novel findings on whole-exome sequencing such as the association of IFT140 with non-syndromic retinal dystrophy or CRX with macular dystrophy. Other areas were driven by unusual groups of patients with limited published data on their condition such as COL18A1 and Knobloch syndrome, with novel phenotypic features of cone-rod dysfunction and pigmentary glaucoma. Sanger sequencing was performed for confirmation and segregation of identified variants but in addition, for investigation of phenotypically similar patient panels for unusual gene associations. This included systemically mild Hermansky-Pudlak syndrome due to HPS6, juvenile macular dystrophy and CDH3, macular dystrophy and CRX and microcephaly with familial exudative vitreoretinopathy due to LRP5. Functional investigation of missense variants in IFT140 related retinal dystrophy was performed with transient cell transfection. This thesis highlights the vast heterogeneity of rare forms of EORD, presents novel clinical and molecular data and describes the key features of conditions to aid diagnosis and opportunities for future research.

Building a new generation of fission reactors in the United States presents many technical and regulatory challenges. One important challenge is the need to share and present results from new high-fidelity, high-performance simulations in an easily usable way. Since modern multiscale, multi-physics simulations can generate petabytes of data, they will require the development of new techniques and methods to reduce the data to familiar quantities of interest (e.g., pin powers, temperatures) with a more reasonable resolution and size. Furthermore, some of the results from these simulations may be new quantities for which visualization and analysis techniques are not immediately available in the community and need to be developed. This paper describes a new system for managing high-performance simulation results in a domain-specific way that naturally exposes quantities of interest for light water and sodium-cooled fast reactors. It describes requirements to build such a system and the technical challenges faced in its development at all levels (simulation, user interface, etc.). AimsTo report the clinical phenotype in a series of four children from three families with the rare association of high myopia, central macular atrophy, and normal full-field electroretinography (ERG).MethodsFour male patients were ascertained with reduced vision, nystagmus, and atrophy of the macula from early childhood. Patients underwent full ophthalmic examination, electrophysiological testing, and retinal imaging.ResultsMinimum duration of follow-up was 8 years. At last review, visual acuity ranged from 0.22 to 1.20 logMAR (6/9.5-6/95 Snellen) at a mean age of 10.5 years (median 9.5 years, range 9-14 years). Refractive error ranged from a spherical equivalent of -7.40 D to -24.00 D. Three had convergent squint. Fundus examination and imaging demonstrated bilateral macular atrophy in all patients that varied from mild atrophy of the retinal pigment epithelium (RPE) to well-demarcated, punched-out atrophic lesions of retina, RPE, and choroid. Flash ERG was normal under photopic and scotopic conditions in all patients. Pattern ERG, performed in three patients, was consistent with mild to severe macular dysfunction. Progression of the area of atrophy was evident in one patient and of the myopia in two patients but all patients had stable visual acuity.ConclusionsPatients with congenital high myopia and macular atrophy present in infancy with reduced visual acuity and nystagmus. The macular atrophic lesions vary in size and severity but electrophysiological testing is consistent with dysfunction confined to the macula. There was no deterioration in visual acuity over 8-10 years of monitoring.

The structure of the pseudo-binary mercury chalcogenide alloy HgSe0.7S0.3 has been studied by means of X-ray and neutron powder diffraction at pressures up to 8.5 GPa. A phase transition from the cubic zinc blende structure to the hexagonal cinnabar structure was observed at P ~ 1 GPa. A phenomenological model of this reconstructive phase transition based on the displacement mechanism is proposed. The analysis of the geometrical relationship between the zinc blende and the cinnabar phases has shown that the possible order parameter for the zinc blende - cinnabar structural transformation is the spontaneous strain e4. This assignment agrees with the previously observed high pressure behaviour of the elastic constants of some mercury chalcogenides., Comment: 13 pages, 6 figs, submitted to J. Phys: Condensed Matter

- Abstract
BACKGROUND: Provision of virtual health care (VHC) home monitoring for patients who are experiencing mild to moderate COVID-19 illness is emerging as a central strategy for reducing pressure on acute health systems. Understanding the enablers and challenges in implementation and delivery of these programs is important for future implementation and re-design. The aim of this study was to explore the perspectives of staff involved with the implementation and delivery, and the experience of patients managed by, a VHC monitoring service in Melbourne, Australia during the COVID-19 pandemic. METHODS: A descriptive qualitative approach informed by naturalist inquiry was used. Staff interviews were analysed using the Consolidated Framework for Implementation Research (CFIR). Patient experience was captured using a survey and descriptive statistics were used to describe categorical responses while content analysis was used to analyse free text responses as they related to the CFIR. Finally, data from the interviews and patient experience were triangulated to see if patient experience validated data from staff interviews. RESULTS: All 15 staff were interviewed, and 271 patients were surveyed (42%). A total of four final overarching themes emerged: service implementation enablers, service delivery benefits for patients, fragmentation of care, and workforce strengths. 19 subthemes aligned with 18 CFIR constructs from staff and patient data. CONCLUSION: Rapid implementation was enabled through shared resources, dividing implementation tasks between senior personnel, engaging furloughed healthcare staff in design and delivery, and having a flexible approach that allowed for ongoing improvements. Benefits for patients included early identification of COVID-19 deterioration, as well as provision of accurate and trustworthy information to isolate safely at home. - Abstract
Transitioning to electrified transport requires improvements in sustainability, energy density, power density, lifetime, and approved the cost of lithium-ion batteries, with significant opportunities remaining in the development of next-generation cathodes. This presents a highly complex, multiparameter optimization challenge, where developments in cathode chemical design and discovery, theoretical and experimental understanding, structural and morphological control, synthetic approaches, and cost reduction strategies can deliver performance enhancements required in the near- and longer-term. This multifaceted challenge requires an interdisciplinary approach to solve, which has seen the establishment of numerous academic and industrial consortia around the world to focus on cathode development. One such example is the Next Generation Lithium-ion Cathode Materials project, FutureCat, established by the UK’s Faraday Institution for electrochemical energy storage research in 2019, aimed at developing our understanding of existing and newly discovered cathode chemistries. Here, we present our perspective on persistent fundamental challenges, including protective coatings and additives to extend lifetime and improve interfacial ion transport, the design of existing and the discovery of new cathode materials where cation and cation-plus-anion redox-activity can be exploited to increase energy density, the application of earth-abundant elements that could ultimately reduce costs, and the delivery of new electrode topologies resistant to fracture which can extend battery lifetime

- Abstract
During its approach to asteroid (101955) Bennu, NASA's Origins, Spectral Interpretation, Resource Identification, and Security-Regolith Explorer (OSIRIS-REx) spacecraft surveyed Bennu's immediate environment, photometric properties, and rotation state. Discovery of a dusty environment, a natural satellite, or unexpected asteroid characteristics would have had consequences for the mission's safety and observation strategy. Here we show that spacecraft observations during this period were highly sensitive to satellites (sub-meter scale) but reveal none, although later navigational images indicate that further investigation is needed. We constrain average dust production in September 2018 from Bennu's surface to an upper limit of 150 g s-1 averaged over 34 min. Bennu's disk-integrated photometric phase function validates measurements from the pre-encounter astronomical campaign. We demonstrate that Bennu's rotation rate is accelerating continuously at 3.63 ± 0.52 × 10-6 degrees day-2, likely due to the Yarkovsky-O'Keefe-Radzievskii-Paddack (YORP) effect, with evolutionary implications.

- Abstract
This report was commissioned under ERAMMP to review monitoring activity at the Land-Sea interface identifying opportunities to better align monitoring activities and improve understanding of pathways and impact, working with Natural Resources Wales (NRW) and Welsh Government (WG) marine policy. The Welsh nation is closely connected to the sea with 60% of the population living on or near the coast, with the furthest settlement only 50 miles from the Irish Sea. The coastal economy makes up a significant percentage of national GDP through tourism, ports and shipping.

45. In-Situ Neutron Studies of Electrodes for Li-Ion Batteries Using a Deuterated Electrolyte: LiCoO2 as a Case Study

- Abstract
An electrochemical cell for in-situ neutron powder diffraction studies of electrode materials for lithium-ion batteries is presented. The device has a coin cell geometry, consisting of 8.4 cm diameter, circular components that can be stacked together and clamped tight using sixteen polyetheretherketone (PEEK) screws. The background issue associated with incoherent scattering from hydrogen within the organic electrolyte was addressed by replacing the normal electrolyte with a deuterated analogue, significantly improving the peak-to-background ratio of the in-situ neutron data. Initial in-situ studies showed clear structural evolution within LixCoO2 during charge in a half-cell with lithium metal as the counter electrode, in agreement with previous studies. In addition, the in-situ cell was shown to provide electrochemical performance comparable to that of equivalent coin cells of the commercial design and, following these demonstration studies, is available for in-situ structural studies of other lithium cathode and anode materials during charge/discharge cycling.
